Description
本発明は、脳内のγ-アミノ酪酸(GABA)を増加させるための剤に関する。
γ-アミノ酪酸(GABA)は、生体内タンパク質を構成するアミノ酸とは異なる特殊アミノ酸の一つであり、哺乳類の脳における主要な抑制性神経伝達物質である。GABAは、ストレスを軽減する効果や睡眠に対する効果を期待して、食品への配合が試みられている。
一方、イミダゾールジペプチドは、側鎖にイミダゾール基を有するアミノ酸と他のアミノ酸とからなるジペプチドの総称であり、抗酸化作用や抗ストレス作用などの生理機能が期待されている。動物では、カルノシン(β-アラニル-L-ヒスチジン)、アンセリン(β-アラニル-1-メチル-L-ヒスチジン)、およびホモカルノシン(γ-アミノブチリル-L-ヒスチジン)が代表的なイミダゾールジペプチドであり、骨格筋にはカルノシンおよびアンセリンが高濃度に存在するのに対し、脳にはホモカルノシンが存在する。イミダゾールジペプチドに関し、特許文献1には、カルノシン、ホモカルノシン、アンセリンからなる群から選ばれた少なくとも1つのジペプチドを有効成分とする睡眠補助剤、およびそれを含む睡眠補助食品が記載されている。
GABAは、通常、血液脳関門を通過しにくいと考えられており、GABAを経口摂取したときに脳内のGABA濃度が上昇するかどうかは不明である。脳内のGABAを効果的に増やすことができる成分があれば望ましい。
本発明者らは、脳内でのGABA量の増加を目標として、ホモカルノシンの一か月間のマウスへの経口投与を実施した。また同時に、GABA、およびカルノシンについても経口投与を実施し、脳内や血中のGABA量に与える影響について比較解析を行った。その結果、ホモカルノシンの経口投与によって、大脳皮質および海馬においてGABA量が上昇するとの知見を得た。このような知見に基づき、本発明を完成した。

GABAは、機能性表示食品において、機能性関与成分として利用されている。GABAを機能性関与成分として含む機能性食品に表示される機能は、高めの血圧が気になる者に適した機能、血圧が高めの者の血圧を下げる機能、事務的作業に伴う一時的な精神的ストレスや疲労感を緩和する機能、健康な者の一時的な精神的ストレスの緩和機能、仕事や勉強による一時的な精神的ストレスや疲労感を緩和する機能、日常生活で生じる一時的に落ち込んだ気分を前向きにする(活気・活力感(積極的な気分、いきいきとした気分、やる気など)の低下を軽減する)機能、継続的な摂取により、日常生活で生じる一時的な活気・活力感の低下を軽減する機能、睡眠の質(寝つき、眠りの深さ、すっきりとした目覚め)の向上に役立つ機能、健やかな眠り(寝つきの向上)に役立つ機能を含む。本発明の剤は、これらのGABAの機能を発揮させるために用いることができる。
また本発明者らの検討によると、ホモカルノシンを含む飼料を1カ月間投与したマウスの大脳皮質において、pro-melanin-concentrating hormone (Pmch)、arginine vasopressin (Avp)、およびhypocretin (Hcrt)などの発現が2倍程度上昇し、Interleukin 1 β遺伝子の発現量の低下が認められた。従って本発明の剤は、pro-melanin-concentrating hormone遺伝子、arginine vasopressin遺伝子、hypocretin遺伝子、およびinterleukin 1 β遺伝子からなる群より選択されるいずれかの発現制御のため、より特定すると、Pro-melanin-concentrating hormone遺伝子、arginine vasopressin遺伝子、およびhypocretin遺伝子からなる群より選択されるいずれかの発現上昇のため、Interleukin 1 β遺伝子の発現抑制のために用いることができる。このような剤は、食欲制御剤または抗炎症剤、または睡眠剤として使用できると期待できる。
本発明で疾患または状態について「処置」というときは、発症リスクの低減、発症の遅延、予防、治療、進行の停止、遅延を含む。処置は、医師が行う、病気の治療を目的とした医療行為と、医師以外の者、例えば栄養士(管理栄養士、保健師、助産師、看護師、臨床検査技師、美容部員、エステティシャン、食品製造者、食品販売者等が行う、非医療的行為とを含む。また処置は、特定の食品の投与または摂取の推奨、食餌方法指導、保健指導、栄養指導(傷病者に対する療養のため必要な栄養の指導、および健康の保持増進のための栄養の指導を含む。)、給食管理、給食に関する栄養改善上必要な指導を含む。
本発明の剤は、ヒト(個体)、およびヒト以外の動物に投与することができ、また健常なもの、および疾患または状態にあるものに投与することができる。ヒト以外の動物には、実験動物(例えば、マウス、ラット、ハムスター)、およびコンパニオンアニマル(例えば、イヌ、ネコ)が含まれる。対象は、ヒトであることが好ましく、より好ましくは、上述したいずれかの処置を施すことが望ましいか、または上述したいずれかの処置を施す必要のあるヒトである。
〔剤〕
本発明に関し、剤というときは、特に記載した場合を除き、有効成分とそれ以外の成分とを含む組成物を指す。剤は、食品組成物であることもあり、医薬組成物であることもある。本発明に関し、剤を、投与するということがあるが、投与は、医薬品以外の食品等を対象に摂取させる意味でも用いている。

(用法・用量)
本発明の剤は、経口投与、静脈注射、経鼻投与によって投与することができるが、経口投与が好ましい。

本発明の剤の投与量は、当業者であれば、対象の、年齢、体重、性別、適用される疾患または状態等に応じ、適宜設計することができる。剤の一日当たりの投与量は、有効成分量として、例えば、10 mg/dayとすることができ、100 mg/dayとすることが好ましく、200 mg/day以上とすることがより好ましく、500 mg/day以上とすることがさらに好ましい。また、1,000 mg/day以上としてもよく、2,000 mg/day以上としてもよく、5,000 mg/day以上としてもよく、7,500 mg/day以上としてもよい。いずれの場合であっても、10,000 mg/day以下とすることができる。また下限値がいずれの場合であっても、50,000 mg/day以下とすることができ、30,000 mg/day以下とすることが好ましく、20,000 mg/day以下とすることがより好ましく、10,000 mg/day以下とすることがさらに好ましい。
投与回数としては、上記の一日当たりの投与量を一回で投与してもよいし、複数回に分けて投与してもよい。
本発明の剤における有効成分の含有量は、当業者であれば適宜設計できるが、例えば1,000 mg/100 g以上とすることができ、1,500 mg/100 g以上とすることが好ましく、2,000 mg/100 g以上とすることがより好ましく、2,500 mg/100 g以上とすることがより好ましく、3,000 mg/100 g以上とすることがより好ましく、3,500 mg/100 g以上とすることがさらに好ましい。いずれの場合であっても、50,000 mg/100 g以下とすることができ、40,000 mg/100 g以下とすることが好ましく、30,000mg/100 g以下とすることがより好ましく、20,000 mg/100 g以下とすることがさらに好ましい。
本発明の剤は、繰り返し対象に投与することができ、また長期間にわたり連続して対象に投与することができる。期間は特に限定されず、例えば3日以上、1週間以上、1カ月以上、3か月以上、6カ月以上であってもよい。効果が十分に認められるためには、1週間以上連続して投与してもよく、2週間以上連続して投与してもよく、1カ月以上連続して投与してもよい。
本発明の剤を治療食(治療の目的を果たすもの。医師が食事箋を出し、それに従い栄養士等が作成した献立に基づいて調理されたもの。)、食事療法食、成分調整食、減塩食、介護食、減カロリー食、ダイエット食、またはスポーツ食(有酸素運動における能力増強を目的とした食、有酸素運動における持久力の増強を目的とした食、競技当日までの栄養を身体に蓄積するための食、競技中に栄養補給するための食、および競技終了後の疲労の回復を目的とする食が含まれる。)とする場合、有効成分の含量は、一食としての投与量を勘案して設計することができる。
(他の成分)
本発明の剤は、目的の効果を発揮しうる限り、有効成分以外の他の成分を含有することができる。他の成分は、医薬または食品として許容される種々の添加剤であり得る。医薬または食品として許容される種々の添加剤は、賦形剤、酸化防止剤(抗酸化剤)、香料、調味料、甘味料、着色料、増粘安定剤、発色剤、漂白剤、防かび剤、ガムベース、苦味料等、酵素、光沢剤、酸味料、乳化剤、強化剤、製造用剤、結合剤、緊張化剤(等張化剤)、緩衝剤、溶解補助剤、防腐剤、安定化剤、凝固剤を含む。
本発明の剤は、目的の効果を発揮しうる限り、有効成分以外の他の成分を含有することができる。他の成分は、医薬または食品として許容される種々の添加剤であり得る。医薬または食品として許容される種々の添加剤は、賦形剤、酸化防止剤(抗酸化剤)、香料、調味料、甘味料、着色料、増粘安定剤、発色剤、漂白剤、防かび剤、ガムベース、苦味料等、酵素、光沢剤、酸味料、乳化剤、強化剤、製造用剤、結合剤、緊張化剤(等張化剤)、緩衝剤、溶解補助剤、防腐剤、安定化剤、凝固剤を含む。
他の成分は、有効成分以外の機能性成分、栄養成分であってもよい。他の成分の例としては、有効成分以外のイミダゾールジペプチド、アミノ酸類(例えば、分岐鎖アミノ酸類、オルニチン、β-アラニン)、不飽和脂肪酸類(例えば、EPA、DHA)、ビタミン類(例えば、ビタミンA、ビタミンB1、ビタミンB2、ビタミンC、ビタミンD)、ミネラル類(例えば、カルシウム、鉄、マグネシウム、亜鉛)、グルコサミン、コンドロイチン類、コラーゲン、2型コラーゲンペプチド、はちみつ、黒酢、タンパク質、糖質、脂質、食物繊維等が挙げられる。
(形態)
本発明の剤の形態の例として、散剤、細粒剤、顆粒剤、錠剤、カプセル剤、液状製剤(エリキシル剤、リモナーデ剤、シロップ剤、乳剤、懸濁剤、溶液剤、ドリンク剤を含む。)、ゲル状製剤を挙げることができる。
本発明の剤の形態の例として、散剤、細粒剤、顆粒剤、錠剤、カプセル剤、液状製剤(エリキシル剤、リモナーデ剤、シロップ剤、乳剤、懸濁剤、溶液剤、ドリンク剤を含む。)、ゲル状製剤を挙げることができる。
好ましい態様の一つにおいて、本発明の剤は食品組成物である。本発明に関し、食品というときは、固形物のみならず、液状のもの、例えば飲料を含む。また、食品は、特定保健用食品(トクホ)、栄養機能食品、機能性表示食品、それ以外の食品(一般食品、健康食品、サプリメント、等ということもある。)を含む。食品はまた、治療食、食事療法食、成分調整食、減塩食、介護食、減カロリー食、およびダイエット食、スポーツ食、並びにそれらのための素材を含む。
食品組成物としてのより具体的な形態の例は、サプリメント(錠剤、顆粒剤、ドリンク剤)、飲料、ゼリー飲料、菓子、食肉加工品、魚介加工品、野菜加工品、惣菜、調味料組成物、食品添加物である。
(その他)
本発明の剤には、用途、有効成分の機能、特定の対象に対して投与を薦める旨を表示することができる。表示は、直接的にまたは間接的にすることができる。直接的な表示の例は、製品自体、製品のパッケージ、容器、ラベル、タグ等への記載を含み、間接的な表示の例は、ウェブサイト、店頭、展示会、看板、掲示板、新聞、雑誌、テレビ、ラジオ、郵送物、電子メール等の場所または手段による、広告・宣伝活動を含む。
本発明の剤には、用途、有効成分の機能、特定の対象に対して投与を薦める旨を表示することができる。表示は、直接的にまたは間接的にすることができる。直接的な表示の例は、製品自体、製品のパッケージ、容器、ラベル、タグ等への記載を含み、間接的な表示の例は、ウェブサイト、店頭、展示会、看板、掲示板、新聞、雑誌、テレビ、ラジオ、郵送物、電子メール等の場所または手段による、広告・宣伝活動を含む。
〔製造方法〕
本発明の剤は、種々の公知の技術を用いて製造することができる。有効成分を所定の濃度になるように調整する工程は、製造工程の種々の段階で適用できる。当業者であれば、有効成分の溶解性、安定性、揮発性等を考慮して、本発明の剤のための製造工程を、適宜設計しうる。

〔開発のための利用〕
本発明はまた、式Iで表される化合物の構造に基づき、脳内のGABAを増加させるための有効成分等を探索する方法(スクリーニング方法)を提供する。式Iで表される化合物は、脳内のGABAを増加するために用いることができるが、これをさらに化学的に改変することで目的の活性の向上、望ましくない作用の軽減、体内での安定性の向上等が期待できる。
本発明はまた、式Iで表される化合物の構造に基づき、脳内のGABAを増加させるための有効成分等を探索する方法(スクリーニング方法)を提供する。式Iで表される化合物は、脳内のGABAを増加するために用いることができるが、これをさらに化学的に改変することで目的の活性の向上、望ましくない作用の軽減、体内での安定性の向上等が期待できる。
化学的に改変するとは、例えば、化合物を化学修飾することで最適化を行うことを指す。化学修飾は、例えば、式Iの化合物のアミノ酸の側鎖部分における置換、除去、修飾を含み、また式Iの化合物への少なくとも1つのアミノ酸の付加、挿入を含む。
このようなスクリーニング方法により、式Iの化合物に基づき最適化を行うことで、活性、薬物動態等の点でより優れた有効成分を創製できる。
以下、本発明を実施例を用いて説明するが、本発明の範囲は実施例に記載したものに限られない。
〔実験方法〕
1)本研究内容は広島大学動物実験委員会において実験の手順、および方法について承諾を得た後、関連する法令等を遵守しながら実施した(承認番号:C18-15-4)。

5週齢のICR雄性マウス26匹を日本チャールス・リバー株式会社より購入した。12時間明暗サイクル (8:00~21:00は明、21:00~8:00は暗) 、恒温 (24±1℃) で飼育を行った。馴化期間として、購入後1週間は脱イオン水と固形正常食飼料を自由摂取させ、体重を記録し、2週目以降も正常食飼料を与えた。コントロール(生理食塩水)群(n=6) 、カルノシン群(n=7)、GABA群(n=6)、およびホモカルノシン群(n=7)の4群に分け、カルノシン群には2g/kg体重、GABA群には0.92g/kg体重、ホモカルノシン群には2.12g/kg体重の経口投与を1か月間行った。なお、カルノシンと等モルになるようにGABAおよびホモカルノシンの投与量を決定した。マウスは麻酔下において採血後、組織を摘出した。血清中、海馬、および大脳皮質中の、ホモカルノシン、GABA、他イミダゾールジペプチド含量をLCMSMS法により測定した。
〔結果および考察〕
ホモカルノシン経口投与時の、大脳皮質および海馬におけるGABA量は、GABAの経口投与時と比較して増加した(図1、図2)。またホモカルノシンの経口投与時の血液中GABA濃度は、GABAの経口投与時と比較して低値であったことから、ホモカルノシンは血液中で分解されずに血液脳関門を通過し、脳内でホモカルノシンが分解されてGABA量が増加した可能性が考えられた。ホモカルノシンの経口投与時のホモカルノシンの血中濃度は15μM近くまで達している(図3)ことから、ホモカルノシンの血液中の安定性はカルノシンと比較して高いことが考えられた。ホモカルノシンの血中安定性の高さ、および血液脳関門を通過しGABAに分解されることを考慮すると、ホモカルノシンは脳内のGABA量を増加させるジペプチドとして極めて有用であると考えられた。
ホモカルノシン経口投与時の、大脳皮質および海馬におけるGABA量は、GABAの経口投与時と比較して増加した(図1、図2)。またホモカルノシンの経口投与時の血液中GABA濃度は、GABAの経口投与時と比較して低値であったことから、ホモカルノシンは血液中で分解されずに血液脳関門を通過し、脳内でホモカルノシンが分解されてGABA量が増加した可能性が考えられた。ホモカルノシンの経口投与時のホモカルノシンの血中濃度は15μM近くまで達している(図3)ことから、ホモカルノシンの血液中の安定性はカルノシンと比較して高いことが考えられた。ホモカルノシンの血中安定性の高さ、および血液脳関門を通過しGABAに分解されることを考慮すると、ホモカルノシンは脳内のGABA量を増加させるジペプチドとして極めて有用であると考えられた。
〔製造例〕
(1)カプセル剤
ホモカルノシン1.0重量部、プラセンタエキス(粉末)0.2重量部および乳糖1.3重量部を混合して均一化した後、常法に準じてハードカプセルに充填し、内容量250 mgのカプセル剤(ホモカルノシン100 mg/カプセル)を製造する。
(1)カプセル剤
ホモカルノシン1.0重量部、プラセンタエキス(粉末)0.2重量部および乳糖1.3重量部を混合して均一化した後、常法に準じてハードカプセルに充填し、内容量250 mgのカプセル剤(ホモカルノシン100 mg/カプセル)を製造する。
(2)錠剤
マルトース、デキストリン、デンプン、ビタミンE含有植物油、イソマルトオリゴ糖、難消化性デキストリン、貝カルシウム、トレハロース、ショ糖エステル、ビタミンC、クエン酸、リン酸カルシウム、香料、シェラック、ナイアシン、ビタミンK、甘味料、塩化カリウム、ビタミンA、パントテン酸カルシウム、ビオチン、ピロリン酸鉄、ビタミンB類、ビタミンD、炭酸マグネシウム、葉酸とともに、1錠(300 mg)当たりホモアンセリン150 mgを含む錠剤を製造する。
マルトース、デキストリン、デンプン、ビタミンE含有植物油、イソマルトオリゴ糖、難消化性デキストリン、貝カルシウム、トレハロース、ショ糖エステル、ビタミンC、クエン酸、リン酸カルシウム、香料、シェラック、ナイアシン、ビタミンK、甘味料、塩化カリウム、ビタミンA、パントテン酸カルシウム、ビオチン、ピロリン酸鉄、ビタミンB類、ビタミンD、炭酸マグネシウム、葉酸とともに、1錠(300 mg)当たりホモアンセリン150 mgを含む錠剤を製造する。
(3)ドリンク
カルノシン、アンセリン、果糖、ぶどう糖、クエン酸、安定剤、着色料、香料、甘味料とともに、1瓶(50mL)当たりホモカルノシン500 mgを含むドリンク剤を製造する。

Claims (6)
- 式Iで表される化合物を含む、脳内のγ-アミノ酪酸(GABA)増加剤。
- 医薬または食品として許容されるアミノ酸が、ヒスチジン、1-メチルヒスチジン、3-メチルヒスチジン、アルギニン、およびリジンからなる群より選択されるいずれかである、請求項1に記載の剤。
- 式Iで表される化合物が、下記からなる群より選択されるいずれかである、請求項1または2に記載の剤。
- 経口剤である、請求項1から3のいずれか1項に記載の剤。
- 1週間以上連続して用いるための、請求項1から4のいずれか1項に記載の剤。
- 脳内のGABAの増加により改善される疾患または状態に用いるための、請求項1から5のいずれか1項に記載の剤。
