Abstract
본 발명은 3'-시알릴락토스 및 6'-시알릴락토스를 유효성분으로 포함하는 연골 보호 및 보존용 조성물에 관한 것으로, 보다 상세하게는 3'-시알릴락토스 및 6'-시알릴락토스를 포함하는 연골 보호용 식품 조성물; 및 노화로 야기되는 연골소실 예방 또는 보존용 조성물;에 관한 것이다. 본 발명에 따른 3'-시알릴락토스 및 6'-시알릴락토스는 연골 보호 및 연골 소실 예방 효과가 우수한바, 연골 보호 또는 퇴행성 관절염 치료를 위한 식품 및 약학적 조성물 등에 다양하게 활용될 수 있다.
Description
본 발명은 모유올리고당을 유효성분으로 포함하는 연골 보호용 조성물에 관한 것으로, 보다 상세하게는 3‘-시알릴락토스 및 6’-시알릴락토스를 포함하는 연골 보호용 식품 조성물; 및 노화로 야기되는 연골소실 예방 또는 보존용 조성물;에 관한 것이다.
대부분의 포유동물의 젖에 포함되어 있는 올리고당들은 3탄당 이상의 다양한 탄수화물로 구성되어 있으며, 모유와 동물 우유의 올리고당 구성비에는 많은 차이를 보이 있어 모유에 존재하는 올리고당을 특히 모유올리고당(Human milk oligosaccharide, HMO)이라 한다. 모유올리고당은 면역체계와 관련된 중요한 역할을 가지고 있으며 특히, 유아의 면역체계가 완성되기 전, 다양한 바이러스로부터 그 들을 방어하는 역할과 항염증 및 항균 등의 생물학적 기능이 보고되어있다. 현재까지 200여종의 모유올리고당이 보고되고 있는데 대부분은 장내 미생물에 의해 소화되어 프리바이오틱스의 기능을 보이는 반면 시알산이 붙어있는 시알릴올리고당들은 일부 장내 미생물에 의해 분해되지만 대부분은 혈중에 노출되어 다양한 장기에서 당단백질, 당지질, 기타 탄수화물 등의 비환원 말단에 시알화를 증대시키는 기능이 있는 것으로 알려져 있다.
시알산은 동물에만 특이하게 존재하는 단당류로 식물과 동물을 구분짓는 중요한 물질로 2종류가 있다. 사람은 N-아세틸 5 뉴라미닌산 (N-acetylneuraminic acid, Neu5AC, 시알산)으로만 구성되어 있으며, 모든 당화물질의 마지막에 시알산으로 캐핑이 되어 있어 사람과 다른 종 유래의 단백질 등을 구분할수 있다. 영장류를 포함한 다른 포유류 들은 Neu5AC 외에 N-글리코실 5 뉴라민산 (N-glycosylneuraminic acid, Neu5GC)이 주류를 이루고 있어 시알산은 사람과 동물을 구분짓는 중요한 물질이다. 시알산은 인체 내에서 생성되기도 하고 외부에서 섭취하기도 하는데 시알산 단독 형태보다 시알산이 다른 곳에 붙어 있는 형태가 흡수 및 이용에 용이하다.
모유올리고당에 포함되어 있는 대표적인 시알릴올리고당인 3‘-시알릴락토스와 6’-시알릴락토스는 락토스의 갈락토스 위치에 알파-2,3 또는 알파-2,6 연결구조로 연결되어있다. 3‘-시알릴락토스는 대부분의 포유류 젖에 공통적으로 포함되어 있으며 우유올리고당 (Bovine milk oligosaccharide, BMO)의 70%는 3‘-시알릴락토스로 구성되어 있다. 6’-시알릴락토스는 모유올리고당(HMO)에 특이적으로 많이 나타나는 물질로 모유올리고당과 우유올리고당을 구분 짖는 중요한 요소이기도 하다. 이와 같은 2종의 시알릴락토스는 각기 다른 구조와 함량을 보여주며, 특이한 시알산 구조를 활용하여 바이러스가 세포 내 침투하는 것을 막고, 마크로파지를 비롯한 면역반응에 관계된 면역세포 활성에 관여하며, 항염증 효능을 나타내기도 한다. 특히 3‘-시알릴락토스는 항염증 및 면역조절 효능 등을 통해 류마티스 관절염 치료에 이용될 수 있으며 타입II 콜라겐, MMP3, MMP13 등의 발현억제를 통하여 퇴행성관절염에도 효과가 있음이 보고 되었다.
이에 본 발명자들은 설치류보다는 사람과 소화장기 및 대사과정이 유사한 미니돼지에서 3‘-시알릴락토스와 6’-시알릴락토스를 복합함으로써 증대되는 연골 보호 효과; 및 노화로 야기되는 자연적 연골소실 예방 또는 보존효과;가 있음을 확인함으로써 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은, 모유올리고당 중 시알릴올리고당을 유효성분으로 포함하는 연골 보호용 조성물을 제공하는 것이다.
본 발명의 다른 목적은, 모유올리고당 중 시알릴올리고당을 유효성분으로 포함하는 노화로 야기되는 자연적 연골의 소실 예방, 보존, 개선 또는 치료용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은, 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 퇴행성 관절염 치료방법을 제공하는 것이다.
본 발명의 또 다른 목적은, 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 연골 보호 및 보존방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 모유올리고당을 유효성분으로 포함하는 연골 보호용 건강기능식품 조성물을 제공한다.
또한 본 발명은 모유올리고당을 유효성분으로 포함하는 연골 보호용 식품 조성물을 제공한다.
또한 본 발명은 모유올리고당 중 시알릴올리고당을 유효성분으로 포함하는, 연골 보호용 식품 보충제 조성물을 제공한다.
또한 본 발명은 모유 유래 시알릴올리고당을 유효성분으로 포함하는, 퇴행성 관절염 예방 또는 치료용 약학적 조성물을 제공한다.
또한 본 발명은 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 퇴행성 관절염 치료방법을 제공한다.
또한 본 발명은 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 연골 보호 및 보존방법을 제공한다.
본 발명에 따른 3‘-시알릴락토스 및 6’-시알릴락토스는 노화로 야기되는 자연적 연골의 소실 예방 뿐아니라 이전에 남아있는 연골을 보존하는 효과가 우수한바, 연골 보호 또는 관절건강을 위한 식품 및 약학적 조성물 등에 다양하게 활용될 수 있다.

이하, 본 발명을 상세히 설명한다.
본 발명의 양태에 따르면, 본 발명은 모유올리고당 중 시알릴올리고당을 유효성분으로 포함하는 연골 보호용 조성물을 제공한다.
본 발명에 따른 연골 보호용 조성물은 건강기능식품 조성물, 식품 조성물 또는 식품 보충제 조성물일 수 있다.
본 발명의 구체 예에서, 상기 모유올리고당은 3′-Sialyllactose(3-SL), 6′-sialyllactose(6-SL), 3′-sialyl-N-acetyllactosamine(3-SLN), 6′-sialyl-N-acetyllactosamine(6-SLN), sialyllacto-N-tetraose a(LSTa), sialyllacto-N-tetraose d(LSTd), sialyllacto-N-tetraose c(LSTc), sialyllacto-N-tetraose b(LSTb), 3′-sialyl-Lewis A tetrasaccharide(SLeA), 3′-sialyl-Lewis×tetrasaccharide(SLeX), sialylated tetraose type 1(STetra1) 및 sialylated tetraose type 2(STetra2)로 이루어진 군에서 선택되는 1 종 이상인 것이 바람직하며, 더 바람직하게는 3’-SL 또는 6’-SL이며, 더욱 바람직하게는 3’-SL 및 6’-SL의 혼합물일 수 있다.
본 발명의 조성물의 유효성분인 상기 3′-Sialyllactose(3-SL), 6′-sialyllactose(6-SL), 3′-sialyl-N-acetyllactosamine(3-SLN), 6′-sialyl-N-acetyllactosamine(6-SLN), sialyllacto-N-tetraose a(LSTa), sialyllacto-N-tetraose d(LSTd), sialyllacto-N-tetraose c(LSTc), sialyllacto-N-tetraose b(LSTb), 3′-sialyl-Lewis A tetrasaccharide(SLeA), Disialyllacto-N-Tetraose (DSLNT) 모유로부터 분리한 것과 동일한 구조를 가지고 있어야 하며, 통상적인 생합성 방법으로 제조하거나 시판되는 것을 사용할 수 있다.
본 발명의 구체예에서, 상기 모유올리고당은 3′-Sialyllactose(3-SL) 및 6′-sialyllactose(6-SL)가 1:0.01-100의 비율로 혼합된 것이 바람직하다.
본 발명의 모유올리고당 중 시알릴올리고당은 약학적 또는 식품학적으로 허용되 는 염의 형태로 사용될 수 있으며, 통상의 방법에 의해 제조되는 모든 염, 수화물 및 용매화물이 포함된다.
본 발명의 구체예에서, 상기 모유올리고당 중 시알릴올리고당은 농도가 10 내지 1000 mg/kg인 것이 바람직하며, 더 바람직하게는 100 내지 500 mg/kg일 수 있다.
본 발명의 바람직한 구체예에서, 모유올리고당 중 시알릴올리고당이 3’-SL 또는 6’-SL인 경우 200 mg/kg일 수 있고, 3’-SL 및 6’-SL의 혼합물인 경우 각각 200 mg/kg을 포함하는 것일 수 있다.
본 발명의 구체 예에서, 상기 모유올리고당 중 시알릴올리고당은 전염증성 사이토카인 IL-1β 및 TNF-α; 및 항염증성 사이토카인 IL-10 및 TGF-β;의 발현을 억제하는 것이 바람직하다.
본 발명의 구체예에서, 상기 연골 보호용은 시알릴올리고당은 경골(tibia) 및 대퇴골(femur)의 연골 보호용인 것이 바람직하나, 이에 제한되지 않는다.
상기 경골은 정강이뼈라고도 하며, 인체 골격 중 대퇴골 다음으로 크다. 또한 대퇴골은 넓적다리뼈라고도하며, 인체 골격 중 제일 크다. 경골 및 대퇴골은 개체를 지탱하는 데에 많은 하중이 실리며, 보행 등의 행동에 있어 큰 역할을 한다. 관절염은 노화 또는 퇴행성 변화 등으로 발생하며, 주로 경골 및 대퇴골의 사이(즉, 무릎)의 연골이 손상되거나 닳아서 발생하는바, 경골 및 대퇴골 연골의 보호 및 재생이 매우 중요하다.
본 발명의 조성물은 모유올리고당과 함께 연골 보호 효과를 갖는 공지의 유효 성분을 1 종 이상 더 함유할 수 있다.
본 발명에 따른 모유올리고당은 일반 식품, 건강기능식품, 식품 첨가제 또는 식이보조제로 사용될 수 있다. 상기 모유올리고당이 식품 첨가제로 사용할 경우, 상기 모유올리고당을 그대로 첨가하거나, 다른 식품 또는 식품 성분과 함께 혼합하여 사용되는 등 통상적인 방법에 따라 적절하게 사용될 수 있다.
또한 상기 모유올리고당 및 공지의 효과를 갖는 유효성분의 혼합은 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 변경될 수 있음은 물론이며, 식품 조성물 총 중량에 대하여 0.001 내지 99.9 중량%로 포함되는 것이 바람직하고, 더욱 바람직하게는 1 내지 80중량%로 포함되는 것이다. 그 함량이 0.001중량% 미만일 경우에는 섭취의 효율성이 떨어질 수 있으며, 99.9중량%를 초과할 경우에는 제형화에 어려움이 있다.
구체적인 예로, 식품 또는 음료의 제조 시에는 본 발명의 모유올리고당은 원료에 대하여 15 중량% 이하, 바람직하게는 10 중량% 이하의 양으로 첨가된다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하여 장기간 섭취할 경우에는 상기 범위 이하의 양으로 첨가될 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다. 상기 식품의 종류에는 특별한 제한은 없으나, 본 발명의 모유올리고당을 첨가할 수 있는 식품의 예로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 수프, 음료수, 차, 드링크제, 알코올 음료, 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
본 발명의 식품 조성물이 음료로 제조될 경우 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등의 추가 성분을 포함할 수 있다. 상기 천연 탄수화물로는 포도당, 과당 등의 모노사카라이드; 말토오스, 수크로오스 등의 디사카라이드; 덱스트린, 사이클로덱스트린 등의 천연 감미제; 사카린, 아스파르탐 등의 합성 감미제 등이 사용될 수 있다. 상기 천연 탄수화물은 본 발명의 식품 조성물 총 중량에 대하여 0.01 내지 10중량%, 바람직하게는 0.01 내지 0.1중량%로 포함된다.
본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 포함할 수 있으며, 천연 과일주스, 과일주스 음료 및 야채 음료의 제조를 위한 과육을 포함할 수 있으나 이에 제한되지 않는다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 상기의 첨가제 비율은 크게 제한되지는 않으나, 본 발명의 식품 조성물 총 중량에 대하여 0.01 내지 0.1중량% 범위내로 포함되는 것이 바람직하다.
건강 및 위생을 목적으로 하거나 건강 조절을 목적으로 하는 장기간의 섭취인 경우, 본 발명의 식품 조성물은 안전성 면에서 아무런 문제가 없기 때문에 장기간 복용이 가능하다.
본 발명의 다른 양태에 따르면, 본 발명은 모유올리고당을 유효성분으로 포함하는 퇴행성 관절염 예방, 개선 또는 치료용 조성물을 제공한다.
본 발명에 따른 퇴행성 관절염 예방, 개선 또는 치료용 조성물은 건강기능식품 조성물, 식품 조성물 또는 약학적 조성물일 수 있다.
본 발명의 조성물이 퇴행성 관절염 예방 또는 개선용 식품 조성물; 또는 건강기능식품 조성물;인 경우, 퇴행성 관절염에 대해 예방 또는 개선 효과를 갖는 공지의 유효 성분을 1 종 이상 더 함유할 수 있다.
본 발명의 구체예에서, 본 발명의 조성물이 퇴행성 관절염 예방 또는 치료용 약학적 조성물인 경우, 약학적 조성물은 비경구 투여 또는 경구 투여용인 것이 바람직하며, 더 바람직하게는 경구 투여용이나, 이에 제한되지 않는다.
본 발명의 약학적 조성물은 투여를 위하여 약학적으로 허용 가능한 담체를 1종 이상 포함할 수 있다. 약학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로오스 용액, 말토덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 이상의 성분을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화 할 수 있다. 더 나아가 당 분야의 적정한 방법으로 각 질환에 따라 또는 성분에 따라 제제화 할 수 있다.
상기 약학적 조성물은 추가적인 성분을 더 포함할 수 있다. 상기 추가적인 성분의 예로는 유당, 탈크, 전분, 스테아린산 마그네슘, 결정성 셀룰로오스, 락토오스, 젤라틴, 만니톨, 과옥소산, 이성화당, 폴리비닐알코올, 붕산 및 탄산나트륨이 있다.
상기 약학적 조성물은 경구 투여 또는 비경구 투여할 수 있으며, 바람직하게는 경구 투여하는 것일 수 있다.
비경구 투여를 하는 경우, 정맥내 주입, 근육내 주입, 관절내(intra-articular) 주입, 활액내(intra-synovial) 주입, 수망강내 주입, 간내(intrahepatic) 주입, 병변내(intralesional) 주입 또는 두 개강내(intracranial) 주입 등으로 투여할 수 있다. 상기 약학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다.
본 발명의 조성물은 실제 임상투여 시에 경구 또는 비경구의 여러 가지 제형으로 투여될 수 있는데, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제할 수 있으며, 당해 기술 분야에 알려진 적합한 제제는 문헌(Remington's Pharmaceutical Science, 최근, Mack Publishing Company, Easton PA)에 개시되어 있는 것을 이용하는 것이 바람직하다.
상기 경구 투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘 카보네이트(Calcium carbonate), 수크로스(Sucrose) 또는 락토오스(Lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 또한, 상기 경구 투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
상기 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성용제, 현탁용제로는 프로필렌글리콜(Propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 퇴행성 관절염의 예방 및 치료를 위하여 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명의 또 다른 양태에 따르면, 본 발명은 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 퇴행성 관절염 치료방법; 및 연골 보호 및 보존방법;을 제공한다.
본 발명의 구체예에서, 상기 개체는 퇴행성 관절염이 발병할 것으로 예상되는 개체; 발병한 개체; 또는 완치판정을 받은 개체일 수 있다. 또한 상기 개체는 연골 보호 및 보존이 필요한 개체일 수 있으나, 이에 제한 되지 않는다.
중복되는 내용은 본 명세서의 복잡성을 고려하여 생략하며, 본 명세서에서 달리 정의되지 않은 용어들은 본 발명이 속하는 기술분야에서 통상적으로 사용되는 의미를 갖는 것이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지는 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실험예 1. 골관절염 미니돼지 제작
옵티팜(OPTIPHARM Co., Ltd., Korea)으로부터 12-18 개월령 40-70kg의 암컷 유카탄 미니돼지 9 마리를 입수했다. 골관절염은 미니돼지의 왼쪽 무릎에 전방 십자인대 절개 수술을 하여 유도하였다. 상기 절개 수술을 위하여, 케타민(30 mg/kg) 및 염산자일라진(3 mg/kg)을 근육 내 주사하여 전신 마취하였고, 마취를 유지하기 위해 이소플루란(2~3% v/v)을 흡입하게 하였다. 절개 수술 중 미니돼지의 체온과 호흡률을 지속적으로 모니터링하였다.
실험예 2. 3‘-시알릴락토스 및 6’-시알릴락토스의 경구투여
정제 형태인 위약, 200 mg 3’-시알릴락토스 및 200 mg 6’-시알릴락토스를 GeneChem Inc.(GeneChem Inc., 대전, 한국)에서 제작하였다. 상기 위약은 84.6% 미정질 셀룰로오스를 포함한다.
대조군 및 실험군은 표 1에 나타내었다.

실험예 3. 보행 분석
트레드밀을 걷는 모든 실험용 미니돼지는 설명한 방법을 사용하여 녹화하였다.
미니돼지는 러닝머신에서 5-10분 동안 걷기 전에 적응시켰고, 적응 후에는 러닝머신에서 2 km/h로 걷게 하였다. 런닝머신을 걷는 동안 미니돼지의 왼쪽은 비디오 카메라(HDR-CX240, Sony)를 사용하여 촬영하였다. 각 동물의 무릎 각도, 보폭 시간(왼쪽 뒷다리의 두 연속 첫 번째 접촉 사이의 시간) 및 스윙 시간(첫 번째 접촉과 발가락 사이의 시간)은 Kinovea program(Kinovea 0.9.1, Charmant, Joan)을 사용하여 20초 짜리 비디오(10 걸음 이상)를 분석하였다.
실험예 4. 연골 파괴 및 조직화학염색 점수
미니돼지 무릎 관절을 10% 중성 완충 포르말린에 고정하고, 4주 동안 10 % 포름산으로 석회를 제거하였다. 석회 제거 후 무릎 관절 연골을 파라핀에 매립하고 4 μm 두께로 절삭하였다. 경골 및 대퇴골 절편을 H&E 염색 및 Safranin-O 염색하였고, Osteoarthritis Research Society International(OARSI) 의 지침을 사용하여 육안 관찰 점수를 매겼다.
실시예 1. 3'-SL 및 6'-SL 투여가 미니돼지 걸음걸이에 미치는 영향 조사
골관절염에 대한 보호 효과를 알아보기 위해 9마리의 유카탄 미니돼지를 세 그룹(대조군, 200 mg 3'-SL 그룹, 3'-SL+6'-SL 그룹)으로 나누었다. 각 그룹의 동물은 0; 200 mg/head/day 3'-SL; 또는 200 mg/head/day 3'-SL+200 mg/head/day 6'-SL 조합;으로 12주 동안 경구 투여하였다. 투여 4~5주 후, 모든 그룹의 왼쪽 뒷다리에 전방십자인대 절개수술을 시행하였다. 부검 전, 전방십자인대 절개수술 후 8주 동안 3'-SL 및 6'-SL이 동물의 보행 행동에 미치는 영향을 조사하였다. 구체적인 실험 스케줄은 도 1A에 나타내었다. 상기 보행 행동 조사는 미니돼지가 러닝머신에서 걷는 동안 왼쪽 뒷다리의 이동성을 분석하였으며, 보다 자세한 보행 행동들은 도 1B에 나타내었다.
먼저 한주기 동안 왼쪽 무릎의 운동범위를 분석하였으며, 그 결과는 도 1C 내지 1G에 나타내었다.
도 1C 내지 1G에 나타낸 바와 같이, 각 시점의 무릎 각도는 대조군과 비교하여 모든 그룹에서 달랐다(도 1C 및 D). 또한 대조군에 비해 모든 그룹에서 보폭 시간이 단축된 것을 확인하였다(도 1E). 특히, 200 mg/head/day 3'-SL 투여에 비교하여 200 mg/head/day 3'-SL+200 mg/head/day 6'-SL 처리 그룹에서 더 많이 감소하였다. 그러나 스윙 시간(도 1F)과 스탠스 시간(도 1G)은 모든 그룹이 유사하였다. 상기 결과는 3'-SL 및 6'-SL의 혼합 경구 투여가 골관절염 통증에 의한 보행 이상의 완화 효과를 가질 수 있음을 시사한다.
실시예 2. 3'-SL 및 6'-SL의 연골 보호 효과 조사
전방십자인대 절개 수술 8주 후 부검 후 미니돼지 무릎 관절의 연골 파괴를 조사하였다. 부검 후 미니돼지 무릎 관절을 관찰한 결과는 도 2A에 나타내었다.
도 2A에 나타낸 바와 같이, 대조군에서는 관절 연골에서 심각한 침식과 석회화 영역의 균열이 관찰되었지만, 이러한 구조적 파괴는 SL 처리 후 감소하였다.
특히 대조군의 침식은 연골 하골까지 확장되는 것으로 나타났으나, 3’-SL 처리 그룹은 관절 연골까지 확장되는 것을 확인하였다. 흥미롭게도, 3'-SL 및 6'-SL의 동시 투여는 관절 연골의 큰 침식보다는 표면 거칠어짐, 세동 및 균열을 보였다.
연골의 육안 관측 결과를 OARSI 스코어로 정량화한 결과는 도 2B에 나타내었다.
도 2B에 나타낸 바와 같이, 200 mg/head/day 3'-SL+200 mg/head/day 6'-SL 처리 그룹은 OARSI 점수가 현저히 낮았으며, 골관절염 보호 효과를 보였다. 상기 결과는 3'-SL 및 6'-SL의 혼합 경구 투여가 골관절염에 대한 보호 효과를 가질 수 있음을 시사한다.
실시예 3. 조직병리검사를 이용한 3’-SL 및 6’-SL의 연골 보호 효과 조사
본 실시예에서는 연골의 조직병리검사 점수를 통해 연골 파괴를 평가하였다. 구체적으로, 미니돼지 경골 및 대퇴골 연골을 safranin-O 염색하여, 연골 파괴를 관찰하였다. safranin-O 염색 결과는 도 3에 나타내었다.
도 3에 나타난 바와 같이, safranin-O 염색 결과는 육안 관찰 결과와 유사하게 대조군 경골 및 대퇴골 연골에서 연골 파괴가 관찰되었다. 대조군에서는 관절 연골에서 심각한 침식과 석회화 영역의 균열이 관찰되었지만, 이러한 구조적 파괴는 SL 처리 후 감소하였다.
상기 결과는 3'-SL 및 6'-SL의 혼합 투여가 미니돼지 골관절염 모델링에서 연골 보호에 강력한 역할을 한다는 것을 의미한다.
이하, 제제예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 제제예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 제제예에 의해 제한되는 것으로 해석되지 않는다.
제제예 1. 식품 제제의 제조
1-1. 건강식품의 제조
3’-시알릴락토스 200 mg 혹은 6’-시알릴락토스 200 mg
비타민 혼합물 적량
비타민 A 아세테이트 70 g
비타민 E 1.0 mg
비타민 B1 0.13 mg
비타민 B2 0.15 mg
비타민 B6 0.5 mg
비타민 B12 0.2 g
비타민 C 10 mg
비오틴 10 g
니코틴산아미드 1.7 mg
엽산 50 g
판토텐산 칼슘 0.5 mg
무기질 혼합물 적량
황산제1철 1.75 mg
산화아연 0.82 mg
탄산마그네슘 25.3 mg
제1인산칼륨 15 mg
제2인산칼슘 55 mg
구연산칼륨 90 mg
탄산칼슘 100 mg
염화마그네슘 24.8 mg
상기의 비타민 및 미네랄 혼합물의 조성비는 비교적 건강식품에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만, 그 배합비를 임의로 변형 실시하여도 무방하며, 통상의 건강식품 제조방법에 따라 상기의 성분을 혼합한 다음, 과립을 제조하고, 통상의 방법에 따라 건강식품 조성물 제조에 사용할 수 있다.
1-2. 건강음료의 제조
3’-시알릴락토스 200 mg
6’-시알릴락토스 200 mg
비타민 C 15 g
비타민 E(분말) 100 g
젖산철 19.75 g
산화아연 3.5 g
니코틴산아미드 3.5 g
비타민 A 0.2 g
비타민 B1 0.25 g
비타민 B2 0.3 g
물 정량
통상의 건강음료 제조방법에 따라 상기의 성분을 혼합한 다음, 약 1 시간 동안 85 ℃ 에서 교반 가열한 후, 만들어진 용액을 여과하여 멸균된 2 L 용기에 취득하여 밀봉 멸균한 뒤 냉장 보관한 다음 본 발명의 건강음료 조성물 제조에 사용한다.
상기 조성비는 비교적 기호음료에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만 수요계층이나, 수요국가, 사용용도 등 지역적, 민족적 기호도에 따라서 그 배합비를 임의로 변형 실시하여도 무방하다.
제제예 2. 약학적 조성물의 제조
2-1. 산제의 제조
3’-시알릴락토스 20 mg 혹은 6’-시알릴락토스 20 mg
유당 100 mg
탈크 10 mg
상기의 성분들을 혼합하고 기밀포에 충진하여 산제를 제조한다.
2-2. 정제의 제조
3’-시알릴락토스 10 mg 혹은 6’-시알릴락토스 10 mg
옥수수전분 100 mg
유당 100 mg
스테아린산 마그네슘 2 mg
상기의 성분들을 혼합한 후 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조한다.
2-3. 캡슐제의 제조
3’-시알릴락토스 10 mg 혹은 6’-시알릴락토스 10 mg
결정성 셀룰로오스 3 mg
락토오스 14.8 mg
마그네슘 스테아레이트 0.2 mg
통상의 캡슐제 제조방법에 따라 상기의 성분을 혼합하고 젤라틴 캡슐에 충전하여 캡슐제를 제조한다.
2-4. 주사제의 제조
3’-시알릴락토스 10 mg 혹은 6’-시알릴락토스 10 mg
만니톨 180 mg
주사용 멸균 증류수 2974 mg
인산이나트륨 이수화물 26 mg
통상의 주사제의 제조방법에 따라 1 앰플당 (2 ml) 상기의 성분 함량으로 제조한다.
2-5. 액제의 제조
3’-시알릴락토스 20 mg 혹은 6’-시알릴락토스 20 mg
이성화당 10 g
만니톨 5 g
정제수 적량
통상의 액제의 제조방법에 따라 정제수에 각각의 성분을 가하여 용해시키고 레몬향을 적량 가한 다음 상기의 성분을 혼합한 다음 정제수를 가하여 전체를 정제수를 가하여 전체 100ml로 조절한 후 갈색병에 충진하여 멸균시켜 액제를 제조한다.
이상, 본 발명내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적인 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의해 정의된다고 할 것이다.
Claims (12)
- 모유올리고당을 유효성분으로 포함하는, 연골 보호 및 보존용 조성물.
- 제1항에 있어서,상기 모유올리고당은 3′-Sialyllactose (3-SL), 6′-sialyllactose (6-SL), 3′-sialyl-N-acetyllactosamine (3-SLN), 6′-sialyl-N-acetyllactosamine (6-SLN), sialyllacto-N-tetraose a (LSTa), sialyllacto-N-tetraose d (LSTd), sialyllacto-N-tetraose c (LSTc) 및 sialyllacto-N-tetraose b (LSTb)로 이루어진 군에서 선택되는 1 종 이상인, 연골 보호 및 보존용 조성물.
- 제1항에 있어서,상기 모유올리고당은 농도가 10 내지 1000 mg/kg인, 연골 보호 및 보존용 조성물.
- 제1항에 있어서,상기 연골 보호용은 시알릴올리고당은 경골(tibia) 및 대퇴골(femur)의 연골 보호용인, 연골 보호 및 보존용 조성물.
- 제1항에 있어서,상기 조성물은 건강기능식품 또는 식품 조성물인, 연골 보호 및 보존용 조성물.
- 모유올리고당을 유효성분으로 포함하는, 연골 보호용 식품 보충제 조성물.
- 모유올리고당을 유효성분으로 포함하는, 퇴행성 관절염 예방 또는 치료용 약학적 조성물.
- 제7항에 있어서,상기 약학적 조성물은 경구 투여용인, 퇴행성 관절염 예방 또는 치료용 약학적 조성물.
- 모유올리고당을 유효성분으로 포함하는, 퇴행성 관절염 예방 또는 개선용 건강기능식품 조성물.
- 모유올리고당을 유효성분으로 포함하는, 퇴행성 관절염 예방 또는 개선용 식품 조성물.
- 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 퇴행성 관절염 치료방법.
- 모유올리고당을 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 연골 보호 및 보존방법.
