Abstract
본 출원은 아밀로이드 전구체 단백질에 대한 저해 활성을 갖는 펩타이드 및 이의 용도 관한 것으로서, 서열번호 1 내지 서열번호 8, 및 서열번호 20 내지 서열번호 23 중 어느 하나의 아미노산 서열로 이루어진 펩타이드, 상기 펩타이드를 유효성분으로 포함하는 퇴행성 신경 질환의 예방 또는 치료용 약제학적 조성물을 제공한다.
Description
본 출원은 아밀로이드 전구체 단백질에 대한 저해 활성을 갖는 펩타이드 및 이의 용도에 관한 것이다. 본 특허출원은 2021년 11월 24일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2021-0163629호, 2021년 11월 24일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2021-0163630호에 대하여 우선권을 주장하며, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.
알츠하이머병(Alzheimer’s disease: AD)은 점진적으로 기억력이 감퇴되고 인지력이 상실되는 증상을 나타내는 병이며 인간의 평균수명이 길어짐에 따라 그 발병률도 현저히 증가하고 있다. 알츠하이머 병의 병리학적 특징 중 하나는 신경세포의 외부에 축적되는 노인성 반점(senile plaques)을 들 수 있는데 이의 원인 물질로는 베타-아밀로이드(β-amyloid, Aβ)를 들 수 있다.
베타-아밀로이드(Aβ)는 아밀로이드 전구체 단백질 (amyloid precursor protein: APP)에서 잘려져 나온 단백질 파편이고, 베타-아밀로이드 생성에 관여하는 효소 중 가장 중요한 것이 베타위치 APP 절단 효소(β-site APP-cleaving enzyme: BACE1)이라 명명된 베타-시크리테이즈(β-secretase)이다. 베타-시크리테이즈(BACE1)에 의해 잘려진 APP는 90 kDa 내외의 sAPPβ라 불리는 N 말단 도메인(N-terminal domain)과 CTFβ(C99)라 불리는 세포질 도메인(cytoplasmic domain)으로 나눠지는데 이렇게 생성된 sAPPβ는 세포 밖으로 분비되고, C99는 다시 감마-시크리테이즈(γ-secretase)에 의해 잘려 4 kDa의 베타-아밀로이드가 생성된다.
아밀로이드 전구체 단백질의 비정상적인 대사로 인하여 베타-아밀로이드(Aβ)가 다량 생성되어 뇌세포 독성을 일으키고 알츠하이머병이 발병한다. 따라서 아밀로이드 전구체 단백질의 활성을 억제하는 물질은 베타-아밀로이드의 생성을 저해하므로 알츠하이머병의 예방 또는 치료 효과를 기대할 수 있다.
현재 시판 중이거나 개발 중인 약물의 대부분은 알츠하이머 환자의 증상을 개선 시켜 삶의 질을 높이는 약물이며, 콜린성 신경계 가설에 근거하여 개발된 아세틸콜린가수분해효소 (acetylcholinesterase) 억제제인 타크린(tacrine), 도네페질 (donepezil), 리바스티그민(rivastigmine), 갈란타민(galantamine)과, NMDA-글루타메이트(NMDA-glutamate) 수용체 억제 기능을 가지고 있는 메만틴 (memantine)등이 있다. 그러나, 이러한 치료제들은 발병 초기에 일시적인 임상증상개선 효과에 그치는 것들이 대부분이고 부작용까지 나타나고 있어 치료에 어려움이 있다 (한국 등록 특허 제10-2085358호).
본 발명자들은 알츠하이머병 발명 원인에 근거한 근본적인 치료에 목표를 두고 베타-아밀로이드 단백질의 생성을 억제하거나 분해할 수 있는 치료제를 개발하고자 연구한 끝에 아밀로이드 전구체 단백질의 발현 및/또는 활성을 억제하는 물질을 개발하고, 이에 기초하여 본 발명을 완성하였다.
일 양상은 서열번호 1 내지 서열번호 8 및 서열번호 20 내지 서열번호 23 중 어느 하나의 아미노산 서열로 이루어진 펩타이드를 제공하는 것이다.
다른 양상는 상기 펩타이드를 유효성분으로 포함하는 퇴행성 신경 질환의 예방 또는 치료용 약제학적 조성물을 제공하는 것이다.
또 다른 양상은 퇴행성 신경 질환의 예방 또는 치료를 위한 상기 펩타이드의 의약적 용도, 또는 개체에 치료학적 유효량의 상기 펩타이드를 투여하는 단계를 포함하는 퇴행성 신경 질환을 치료하는 방법을 제공하는 것이다.
본 출원의 다른 목적 및 이점은 첨부한 청구범위 및 도면과 함께 하기의 상세한 설명에 의해 보다 명확해질 것이다. 본 명세서에 기재되지 않은 내용은 본 출원의 기술 분야 또는 유사한 기술 분야 내 숙련된 자이면 충분히 인식하고 유추할 수 있는 것이므로 그 설명을 생략한다.
본 출원에서 개시된 각각의 설명 및 실시형태는 각각의 다른 설명 및 실시 형태에도 적용될 수 있다. 즉, 본 출원에서 개시된 다양한 요소들의 모든 조합이 본 출원의 범주에 속한다. 또한, 하기 기술된 구체적인 서술에 의하여 본 출원의 범주가 제한된다고 볼 수 없다.
일 양상은 서열번호 1 내지 서열번호 8 및 서열번호 20 내지 서열번호 23 중 어느 하나의 아미노산 서열로 이루어진 펩타이드를 제공한다.
본 명세서에서 사용되는 용어, "펩타이드"는 펩타이드 결합에 의해 아미노산 잔기들이 서로 결합되어 형성된 선형의 분자를 의미할 수 있다. 상기 펩타이드는 당업계에 공지된 화학적 합성 방법, 특히 고상 합성 기술 또는 액상 합성 기술 (US 등록특허 제5,516,891호)에 따라 제조될 수 있다. 본 발명자들은 생물학적으로 유효한 활성을 갖는 펩타이드를 개발하고자 예의 노력한 결과, 서열번호 1 내지 서열번호 8 중 어느 하나의 아미노산 서열로 이루어진 펩타이드를 규명하였다. 여기서, 상기 생물학적으로 유효한 활성은 (a) 아밀로이드 전구체 단백질 (amyloid precursor protein: APP) 발현 저해; (b) ADAM 10 (A Disintegrin And Metalloproteinase-containing protein 10) 또는 ADAM 17 (A Disintegrin And Metalloproteinase-containing protein 17) 단백질의 발현 증가; (c) IDE (insulin degrading enzyme) 단백질의 발현 증가; (d) 항염증성 사이토카인의 발현 증가; (e) 신경돌기의 성장 촉진; 및 (f) 뇌유래신경생장인자 (Brain Derived Neurotrophic Factor: BDNF)의 발현 증가와 같은 특성으로부터 선택되는 어느 하나 이상을 나타내는 것일 수 있다.
따라서, 상기 펩타이드는 퇴행성 신경 질환을 예방 또는 치료를 위한 용도로 활용될 수 있다.
상기 펩타이드는 화학적 안정성, 강화된 약리 특성 (반감기, 흡수성, 역가, 효능 등), 변경된 특이성 (예를 들어, 광범위한 생물학적 활성 스펙트럼), 감소된 항원성을 획득하기 위하여, 펩타이드의 N- 또는 C-말단에 보호기가 결합되어 있을 수 있다. 일 구체예에 있어서, 상기 펩타이드의 N-말단은 아세틸기(acetyl group), 플루오레닐메톡시카르보닐기(fluoreonylmethoxycarbonyl group), 포르밀기(formyl group), 팔미토일기(palmitoyl group), 미리스틸기(myristyl group), 스테아릴기(stearyl group) 및 폴리에틸렌글리콜(polyethylene glycol; PEG)로 이루어진 군으로부터 선택되는 어느 하나의 보호기와 결합될 수 있고; 및/또는 상기 펩타이드의 C-말단은 아미노기(amino group, -NH2), 및 아자이드(azide, -NHNH2)로 이루어진 군으로부터 선택되는 어느 하나의 보호기와 결합될 수 있다. 또한, 상기 펩타이드는 선택적으로, 표적화 서열, 태그 (tag), 표지된 잔기, 반감기 또는 펩타이드 안정성을 증가시키기 위한 특정 목적으로 제조된 아미노산 서열도 추가적으로 포함할 수 있다.
상기 펩타이드는 인위적으로 합성되거나 (Artificially synthesized), 또는 비-천연 발생 또는 조작 (Non-naturally occurring or engineered)된 것이며, 상기 “비-천연 발생 또는 조작”은 자연 상태에서 일어나는 존재 그대로의 상태가 아닌, 인위적인 변형을 가하여 생성된 상태를 의미한다. 여기서, 상기 인위적인 변형은 복수 개의 아미노산 구조를 모방하여 인위적으로 아미노산 서열을 합성하는 것 또는 전술한 바와 같이 화학적 안정성, 강화된 약리 특성, 변경된 특이성, 또는 감소된 항원성을 획득하기 위하여 조작된 것을 포함할 수 있다.
본 명세서에서 사용되는 용어, "안정성"은 생체 내 단백질 절단효소의 공격으로부터 상기 펩타이드를 보호하는 생체 내 안정성뿐만 아니라, 저장 안정성 (예컨대, 상온 저장 안정성)도 의미할 수 있다.
다른 양상은 서열번호 1 내지 서열번호 8 및 서열번호 20 내지 서열번호 23 중 어느 하나의 아미노산 서열로 이루어진 펩타이드를 유효성분으로 포함하는 퇴행성 신경 질환 예방 또는 치료용 약제학적 조성물을 제공한다.
상기 펩타이드에 대한 설명에서 언급된 용어 또는 요소 중 이미 언급된 것과 동일한 것은 상술한 바와 같다.
본 명세서에서 용어, "예방"은 상기 조성물의 투여로 질병의 발병을 억제 또는 지연시키는 모든 행위를 의미한다.
본 명세서에서 용어, "치료"는 질병을 앓거나 또는 질병이 발병할 위험이 있는 개체에게, 상기 개체의 상태(예를 들면, 하나 이상의 증상)의 개선, 질병 진행의 지연, 증상 발생의 지연 또는 증상 진행의 둔화 등을 포함한 효과를 제공하는 임의의 형태의 치료를 의미한다. 따라서, 상기 "치료" 및 "예방"은 증상의 치유 또는 완전한 제거를 의미하도록 의도되지 않는다.
상기 "개체"는 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는, 인간 또는 비-인간인 영장류, 생쥐(mouse), 개, 고양이, 말, 및 소 등의 포유류를 의미한다.
상기 약제학적 조성물에 의한 예방 또는 치료 대상 질병인, "퇴행성 신경 질환"은 신경계의 신경세포에 점진적이고 꾸준한 사멸로 인한 퇴행성 변화가 나타나면서 여러 가지 증상을 유발하는 질환을 지칭할 수 있다. 상기 퇴행성 질환으로는 예를 들어, 알츠하이머병(Alzheimer's disease), 파킨슨 병, 헌팅턴 병, 다발성경화증(multiple sclerosis), 경도인지장애(mild cognitive impairment), 대뇌 아밀로이드 맥관병증, 아밀로이드성 뇌졸증(stroke), 전신성 아밀로이드병, 더취(Dutch) 형 아밀로이드증, 니만-픽병, 노인성 치매, 근위축성 측삭 경화증 (amyotrophic lateral sclerosis), 척수소뇌성 운동실조증(Spinocerebellar Atrophy), 뚜렛 증후군(Tourette`s Syndrome), 프리드리히 보행실조(Friedrich`s Ataxia), 마차도-조셉 병 (Machado-Joseph`s disease), 루이 소체 치매(Lewy Body Dementia), 근육긴장이상(Dystonia), 진행핵위마비(Progressive Supranuclear Palsy) 전두측두엽 치매(Frontotemporal Dementia), 말초신경손상(peripheral neuropathy), 및 다발성신경병증(polyneuropathy)로 이루어진 군에서 선택되는 것일 수 있으며, 바람직하게는 알츠하이머병일 수 있으나, 이에 제한되는 것은 아니다.
일 실시예에 따르면, 상기 펩타이드 (peptide-1 내지 peptide-8)는 알츠하이머병 등 퇴행성 신경 질환의 주요 원인 물질인 APP의 발현을 억제함과 동시에, 기 생성된 세포막 표면의 APP를 절단하여 sAPPa의 형태로 유리시키는 ADAM 10 및 ADAM 17의 발현, 그리고, 기 생성된 노인반을 분해하는 IDE를 증가시킬 수 있다. 또한, 상기 펩타이드는 퇴행성 신경 질환의 주요 병인 중 하나인 신경 염증 반응을 완화하고, 손상된 신경계를 재생시킬 수 있었다.
일 실시예에 따르면, 상기 펩타이드 (peptide-9 내지 peptide-12)는 알츠하이머병 등 퇴행성 신경 질환의 주요 원인 물질인 APP의 발현을 억제함과 동시에, 기 생성된 세포막 표면의 APP를 절단하여 sAPPa의 형태로 유리시키는 ADAM 17의 발현을 증가시킬 수 있다. 또한, 상기 펩타이드는 퇴행성 신경 질환의 주요 병인 중 하나인 신경 염증 반응을 완화하고, 손상된 신경계를 재생시킬 수 있었다.
따라서, 상기 펩타이드는 퇴행성 신경 질환 예방 또는 치료용 약제학적 조성물의 유효성분으로 활용될 수 있다.
상기 약제학적 조성물은 상기 펩타이드의 약제학적 유효량 (pharmaceutically effective amount); 및/또는 약제학적으로 허용되는 담체를 포함하는 것일 수 있으나, 이에 제한되는 것은 아니다.
본 명세서에서 사용되는 용어 "약제학적 유효량"은 상기 약제학적 조성물의 퇴행성 신경 질환 예방 또는 치료 효능을 달성하는 데 충분한 양을 의미한다.
상기 약제학적으로 허용되는 담체는 제제 시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
상기 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있으나, 이에 제한되는 것은 아니다.
상기 약제학적 조성물은 경구 또는 비경구, 바람직하게는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 근육 주입, 정맥 내 주입, 피하 주입, 복강 주입, 국소 투여, 경피 투여 등으로 투여할 수 있으나, 이에 한정되는 것은 아니다.
상기 약제학적 조성물의 투여량은 1일당 0.0001 내지 1000 ug(마이크로그램, 0.001 내지 1000 ug, 0.01 내지 1000 ug, 0.1 내지 1000 ug, 또는 1.0 내지 1000 ug일 수 있으나, 이에 제한되는 것은 아니며, 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다.
상기 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다.
상기 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나, 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 및/또는 안정화제를 추가적으로 포함할 수 있다.
또 다른 양상은 치료학적 유효량의 서열번호 1 내지 서열번호 8 및 서열번호 20 내지 서열번호 23 중 어느 하나의 아미노산 서열로 이루어진 펩타이드를 유효성분으로 포함하는 약제학적 조성물을 개체에 투여하는 단계를 포함하는, 퇴행성 신경 질환을 예방 또는 치료하는 방법을 제공한다.
상기 펩타이드 또는 약제학적 조성물에 대한 설명에서 언급된 용어 또는 요소 중 이미 언급된 것과 동일한 것은 상술한 바와 같다.
일 양상에 따른 펩타이드에 의하면, 아밀로이드 전구체 단백질의 발현을 저해하고, 상기 아밀로이드 전구체 단백질의 분해와 연관 유전자인 ADAM 10, ADAM 17 및 IDE의 발현을 증가시킬 수 있다.
또한, 일 양상에 따른 펩타이드에 의하면, 항염증성 사이토카인의 생성을 증가시켜 신경 염증 반응을 완화 또는 개선할 수 있고, 신경돌기의 성장과 같은 신경세포 재생을 유도할 수 있다.
따라서, 일 양상에 따른 펩타이드는 퇴행성 신경 질환 예방 또는 치료용 약제학적 조성물의 유효성분으로 활용될 수 있다.
도 1은 일 실시예에 따른 펩타이드를 인간 신경 세포주에 첨가한 후, 아밀로이드 전구체 단백질 (APP)의 발현 저해를 확인한 결과로서, 도 1의 A는 Peptide-1, 도 1의 B는 Peptide-4, 도 1의 C는 Peptide-5, 도 1의 D는 Peptide-7, 도 1의 E는 Peptide-8, 및 도 1의 F는 비교군 펩타이드를 첨가한 결과를 나타낸 것이다.
도 2는 일 실시예에 따른 펩타이드를 인간 신경 세포주에 첨가한 후, ADAM 10의 발현 증가를 확인한 결과로서, 도 2의 A는 Peptide-3, 도 2의 B는 Peptide-4, 도 2의 C는 Peptide-5, 도 2의 D는 Peptide-7, 및 도 2의 E는 Peptide-8을 첨가한 결과를 나타낸 것이다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
Ⅰ. 펩타이드-1 내지 펩타이드-8
실시예 1: 펩타이드의 합성
자동 펩타이드 합성기 (Milligen 9050, Millipore, 미국)를 이용하여 하기 표 1에 기재된 서열번호 1 내지 서열번호 8의 아미노산 서열을 갖는 펩타이드를 합성하고, C18 역상 고성능액체크로마토그래피(HPLC)(Waters Associates, 미국)를 이용하여 이들 합성된 펩타이드를 순수 분리하였다. 컬럼은 ACQUITY UPLC BEH300 C18 (2.1 ㎜ Х 100 ㎜, 1.7㎛, Waters Co, 미국)을 이용하였다.

실시예 2: 아밀로이드 전구체 단백질 발현 저해 효과 확인
본 실시예에서는 펩타이드의 처리에 의한 아밀로이드 전구체 단백질의 발현 저해 효과를 확인하고자 하였다. 이를 위하여, 신경모세포종(neuroblastoma)을 가진 인간의 골수에서 분리 및 증식시켜 수득된 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 하기 표 2의 APP 프라이머를 이용한 real-time qPCR 을 수행하여, 아밀로이드 전구체 단백질 (APP)의 발현 수준을 측정하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군, 비교군으로 임의의 펩타이드를 첨가한 군 (NRP1: AFMVDNEAIYDICR(서열번호 9))을 사용하였다.

그 결과, 도 1에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-1, Peptide-4, Peptide-5, Peptide-7, 또는 Peptide-8)의 첨가에 의해, 알츠하이머병을 포함하는 퇴행성 신경 질환의 주요 원인 단백질인 APP의 발현이 저해되었다. 특히, 상기 APP의 발현 저해 효과는 대조군 대비 80-90%의 낮은 발현 수준을 나타낸 것으로 매우 현저한 것이었다.
실시예 3: 아밀로이드 전구체 단백질의 분해와 연관된 유전자의 발현 증가 효과 확인
3-1. ADAM 10 (A Disintegrin And Metalloproteinase-containing protein 10) 유전자의 발현
ADAM 10은 APP를 비독성 단편으로 절단하여 베타-아밀로이드(b-amyloid, Ab1-42, 1-40)의 생성을 억제하는 alpha-secretase이다. 따라서, 상기 ADAM 10의 발현 증가는 퇴행성 신경 질환의 치료 효과를 나타내는 지표로 사용될 수 있다. 구체적으로, 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 하기 표 3의 ADAM 10 프라이머를 이용한 real-time qPCR을 수행하여, ADAM 10의 발현 수준을 측정하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군을 사용하였다.
그 결과, 도 2에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-3, Peptide-4, Peptide-5, Peptide-7, 또는 Peptide-8)의 첨가에 의해, ADAM 10의 발현이 증가되었다.

3-2. ADAM 17 (A Disintegrin And Metalloproteinase-containing protein 17) 유전자의 발현
ADAM 17은 APP를 비독성 단편인 sAPPa로 절단하여 세포막 표면에서 APP를 제거하는 효소이다. 따라서, 상기 ADAM 17의 발현 증가는 퇴행성 신경 질환의 치료 효과를 나타내는 지표로 사용될 수 있다. 구체적으로, 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 하기 표 4의 ADAM 17 프라이머를 이용한 real-time qPCR을 수행하여, ADAM 10의 발현 수준을 측정하였다. 또한, 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 ADAM 17 단백질의 발현과 상기 ADAM 17 및 ADAM 10에 의해 APP로부터 절단되어 유리된 sAPPa의 발현을 각 단백질에 대한 항체를 이용한 웨스턴-블로팅을 통하여 확인하였다. 이와 함께, 유세포분석법 (Flow-cytometric analysis)를 이용하여 신경세포막 표면의 APP의 분포를 분석하고자 하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군을 사용하였다.

그 결과, 도 3 및 도 4에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-1, Peptide-4, 및 Peptide-7)의 첨가에 의해 sAPPa 및 ADAM 17의 발현이 증가되었고, 도 5에 나타낸 바와 같이, 상기 발현 변화는 신경세포막 표면에 존재하는 APP의 수준을 감소시킴에 기여함을 알 수 있었다.
3-3. IDE (Insulin degrading enzyme) 유전자의 발현
IDE는 베타-아밀로이드 (b-amyloid, Ab1-42, 1-40)에 의해 생성 및 응집된 노인반 (senile plaque)을 분해하는 효소이다. 따라서, 상기 IDE의 발현 증가는 퇴행성 신경 질환의 치료 효과를 나타내는 지표로 사용될 수 있다. 구체적으로, 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 하기 표 5의 IDE 프라이머를 이용한 real-time qPCR을 수행하여, IDE의 발현 수준을 측정하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군을 사용하였다.

그 결과, 도 6 및 도 7에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-1, Peptide-2, Peptide-3, Peptide-4, Peptide-5, Peptide-6, Peptide-7, 및 Peptide-8)의 첨가에 의해, IDE의 발현이 증가되었다.
상기의 실험 결과를 종합해 보면, 일 실시예에 따른 펩타이드는 세포막 표면의 APP를 절단하여 비독성 단편인 sAPPa의 형태로 유리시키는 ADAM 10 및 ADAM 17의 발현을 증가시키고, 병리학적 요인으로부터 기인된 뇌 조직 내 노인반을 분해시키는 IDE의 발현을 감소시킴으로써, 퇴행성 신경 질환의 발병을 방지하거나 관련 증상을 개선시킬 수 있다.
실시예 4: 항염증성 사이토카인의 발현 증가 효과 확인
본 실시예에서는 펩타이드의 처리에 의한 퇴행성 신경 질환에서 주요 병인 중 하나인 신경 염증 반응의 억제 효과를 확인하기 위하여, 항염증성 사이토카인 중 하나인 IL-10 발현 증가 효과를 확인하고자 하였다. 구체적으로, 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 하기 표 6의 IL-10 프라이머를 이용한 real-time qPCR을 수행하여, IL-10의 발현 수준을 측정하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군을 사용하였다.

그 결과, 도 8에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-1, Peptide-3, Peptide-4, Peptide-5, 또는 Peptide-7)의 첨가에 의해, IL-10의 발현이 증가되었다.
실시예 5: 신경돌기의 성장 촉진 효과 확인
본 실시예에서는 펩타이드의 처리에 의한 신경세포의 재생을 확인하기 위하여, 신경돌기의 성장을 확인하고자 하였다. 구체적으로, 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 배양하였다. 이후, 성장된 신경돌기를 현미경을 통해 확인하고, 이의 길이를 측정하여 평가하였다.
그 결과, 도 9에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-1, Peptide-2, 또는 Peptide-6)의 첨가에 의해, 신경돌기의 길이가 증가되었다.
상기 결과를 통해, 일 실시예에 따른 펩타이드는 알츠하이머병, 파킨슨병, 헌팅튼병, 다발성경화증 등을 포함하는 퇴행성 신경 질환의 발생과 진행을 억제하는데 기여할 수 있음을 알 수 있었다.
Ⅱ. 펩타이드-9 내지 펩타이드-12
실시예 1: 펩타이드의 합성

신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 상기 표 4의 ADAM 17 프라이머를 이용한 real-time qPCR을 수행하여, ADAM 17의 발현 수준을 측정하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군을 사용하였다.
그 결과, 도 10에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-9, Peptide-10, Peptide-11, 또는 Peptide-12)의 첨가에 의해 ADAM 17의 발현이 증가되었고, 이러한 경향성은 농도에 의존하는 경향을 보여주었다.
상기의 실험 결과를 종합해 보면, 일 실시예에 따른 펩타이드는 세포막 표면의 APP를 절단하여 비독성 단편인 sAPPa의 형태로 유리시키는 ADAM 17의 발현을 증가시킴으로써, 퇴행성 신경 질환의 발병을 방지하거나 관련 증상을 개선시킬 수 있다.
실시예 3: 아밀로이드 전구체 단백질 발현 저해 효과 확인

그 결과, 도 12에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide- 9, Peptide-11, 또는 Peptide-12)의 첨가에 의해, IL-10의 발현이 증가되었다. 특히, 이러한 항염증 효과는 펩타이드-9를 첨가한 군에서 보다 현저하였다.
상기의 실험 결과를 종합해 보면, 일 실시예에 따른 펩타이드는 아밀로이드 전구체 단백질의 분해를 억제함과 동시에 병변 부위의 항염증 반응을 유도할 수 있다.
실시예 5. 뇌신경생장인자의 발현 증가 효과 확인
본 실시예에서는 펩타이드의 처리에 의한 신경계의 재생을 통한 퇴행성 신경 질환의 개선 또는 치료 효과를 확인하기 위하여, 뇌유래신경생장인자 (Brain Derived Neurotrophic Factor: BDNF)의 발현 증가 효과를 확인하고자 하였다. 구체적으로, 인간 신경 세포주인 SH-SY5Y에 상기 제조된 펩타이드를 0.001, 0.01, 0.1, 또는 1 mM 농도로 첨가하고, 이를 24시간 동안 37℃, 5% CO2 조건에서 배양하였다. 이후, 상기 배양물로부터 total RNA를 추출하여 cDNA합성 한 뒤, 하기 표 8의 BDNF 프라이머를 이용한 real-time qPCR을 수행하여, BDNF의 발현 수준을 측정하였다. 한편, 대조군으로 펩타이드를 첨가하지 않은 군을 사용하였다.

그 결과, 도 13에 나타낸 바와 같이, 일 실시예에 따른 펩타이드 (Peptide-9, Peptide-10, Peptide-11, 또는 Peptide-12)의 첨가에 의해, BDNF의 발현이 증가되었다. 특히, 이러한 발현 증대 효과는 펩타이드-11 또는 펩타이드-12를 첨가한 군에서 보다 현저하였다.
상기 결과를 통해, 일 실시예에 따른 펩타이드는 알츠하이머병, 파킨슨병, 헌팅튼병, 다발성경화증 등을 포함하는 퇴행성 신경 질환의 발생과 진행을 억제하는데 기여할 수 있음을 알 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.

- 청구항 1에 있어서, 상기 펩타이드는 하기와 같은 특성으로부터 선택되는 어느 하나 이상을 나타내는 것인, 펩타이드:(a) 아밀로이드 전구체 단백질 (amyloid precursor protein: APP) 발현 저해;(b) ADAM 10 (A Disintegrin And Metalloproteinase-containing protein 10) 또는 ADAM 17 (A Disintegrin And Metalloproteinase-containing protein 17) 단백질의 발현 증가;(c) IDE(insulin degrading enzyme) 단백질의 발현 증가;(d) 항염증성 사이토카인의 발현 증가;(e) 신경돌기의 성장 촉진; 및(f) 뇌유래신경생장인자 (Brain Derived Neurotrophic Factor: BDNF)의 발현 증가.
- 청구항 1 내지 4 중 어느 한 항의 펩타이드를 유효성분으로 포함하는 퇴행성 신경 질환의 예방 또는 치료용 약제학적 조성물.
- 청구항 5에 있어서, 상기 퇴행성 신경 질환은 알츠하이머병(Alzheimer's disease), 파킨슨 병, 헌팅턴 병, 다발성경화증(multiple sclerosis), 경도인지장애 (mild cognitive impairment), 대뇌 아밀로이드 맥관병증, 아밀로이드성 뇌졸증 (stroke), 전신성 아밀로이드병, 더취(Dutch) 형 아밀로이드증, 니만-픽병, 노인성 치매, 근위축성 측삭 경화증(amyotrophic lateral sclerosis), 척수소뇌성 운동실조증(Spinocerebellar Atrophy), 뚜렛 증후군(Tourette`s Syndrome), 프리드리히 보행실조 (Friedrich`s Ataxia), 마차도-조셉 병(Machado-Joseph`s disease), 루이 소체 치매(Lewy Body Dementia), 근육긴장이상(Dystonia), 진행핵위마비(Progressive Supranuclear Palsy) 전두측두엽 치매(Frontotemporal Dementia), 말초신경손상(peripheral neuropathy), 및 다발성신경병증(polyneuropathy)로 이루어진 군에서 선택되는 것인, 약제학적 조성물.
- 청구항 5에 있어서, 상기 퇴행성 신경 질환은 알츠하이머병(Alzheimer's disease)인 것인, 약제학적 조성물.
