Abstract
본 발명은 물과 오일에 용해되지 않는 루테인(Lutein)을 유효성분으로 포함하는 액상용 조성물, 및 상기 액상용 조성물의 사료 용도에 관한 것으로 루테인은 난용성의 성질로 인해 현재 알약이나 캡슐등에 넣은 제품으로만 시판되고 있으나, 본 발명은 루테인을 액상으로 안정화시키는 최적 조성을 확인하였고, 상기 최적 조성으로 제조된 액상용 조성물은 다양한 온도 조건에서 성상이 안정적이고, 침전이 발생하지 않으므로, 눈 건강을 위한 건강식품 또는 애완동물 등의 사료로 유용하게 사용될 수 있다.
Description
본 발명은 물과 오일에 용해되지 않는 루테인(Lutein)을 유효성분으로 함유하는 액상용 조성물, 및 상기 액상용 조성물의 사료 용도에 관한 것이다.
최근 가족 구성이 핵가족화되어갈 뿐만 아니라 애완동물 사육은 정서발달에 도움을 준다는 이유 등으로 우리나라에서도 애완동물을 사육하는 가구수가 상당히 많아지고 있다. 애완동물은 밖에서 자라는 동물이 아닌 가족의 일원으로 보호받으면서 자라고 있지만, 인간들이 겪고 있는 대기나 수질 오염 등과 같은 오염된 자연 환경이나 가공식품 및 스트레스에 빈번히 노출된다. 이러한 이유 때문에 애완동물에게도 사람에 흔히 발병되는 안과질환, 피부질환, 호흡기 질환, 소화기장애 등과 같은 여러 가지 질병이 발병되고 있다. 따라서, 최근 애완동물이 건강하게 삶을 영위할 수 있도록 다양한 사료가 개발 중에 있다.
한편, 루테인(lutein)은 잔토필(xanthophyll)의 하나로서, 현재까지 알려진 600개의 자연 발생 카로티노이드(Carotenoids) 가운데 하나이다. 루테인은 식물, 기타 잔토필에서만 합성되며 시금치, 케일, 노란당근 등의 잎채소에서 대량으로 발견된다. yellow를 의미하는 라틴어 luteus에서 명칭이 유래되었다.
또한, 루테인은 피로한 눈의 건강을 도와주는 영양소로, 태양 및 산화에 의한 손상으로부터 눈을 보호해 주는 역할을 한다. 인간을 포함하는 포유류의 경우 대부분 카로티노이드를 합성하지 못하므로 루테인이 포함된 음식 또는 건강보조식품을 통해 루테인을 섭취할 수 있다. 루테인을 섭취하면 황반색소의 광 밀도가 증가하여 눈 질환이 있는 환자들에서 시각 기능이 개선되며, 눈의 유해활성산소에 의한 시세포 손상에 따른 노화과정을 효과적으로 억제함으로써, 백내장이나 노인성 황반변성(Age-related macular degeneration)과 같은 안구질환의 발생율을 감소시킬 수 있다([Bernstein P.S et al., Ophthalmology, 2002(109), 1780-1787] 및 [Kvansakul J et al., Ophthalmic and Physiological Optics, 2006(26), 362-371]). 또한, 루테인은 1994년 미국의 보조식품건강교육법안(DSHEA) 및 2008년 KFDA에서 각각 안전한 보조식품 및 건강기능성원료(기능성원료 인정 제2008-28호, 59호)로 인정받음에 따라, 최근 루테인을 이용한 건강음료, 기능성 식품 및 영양보조식품 등이 활발히 개발되고 있다.
다만, 루테인은 물에 난용성의 성질을 가지며, 지방 및 계면활성제 등에도 용해도가 낮아 약물을 용해시킨 액상 제형으로의 설계에 어려움이 있고, 용해시키기 위해 필요한 지방과 계면활성제 양이 많아 장기 복용 시 위장관 부작용이 발생할 수 있는 문제점이 있다고 보고된다([Hentschel R1, Jorch G. Acute side effects of surfactant treatment. J Perinat Med. 2002;30(2):143-8] 및 [Aulton's Pharmaceutics E-Book: The Design and Manufacture of Medicines, Michael E. Aulton, Kevin M.G. Taylor, Elsevier Health Sciences, 2013. 7. 29] 등).
이에, 본 발명자들은 루테인을 포함하는 액상용 조성물을 제조하기 위해 노력한 결과, 루테인을 액상으로 안정화시키는 최적 조성을 확인하였고, 상기 최적 조성으로 제조된 액상용 조성물은 다양한 온도 조건에서 성상이 안정적이고, 침전이 발생하지 않는 것을 확인함으로써, 본 발명의 최적 조성을 통해 제조된 루테인을 포함하는 액상용 조성물을 건강식품 또는 애완동물 등의 사료로 유용하게 사용될 수 있음을 밝힘으로써 본 발명을 완성하였다.

본 발명의 목적은 물에 난용성의 성질을 가지는 루테인(Lutein)을 유효성분으로 함유하는 액상용 조성물을 제공하기 위한 것이다.
상기 목적을 달성하기 위하여, 본 발명은 루테인(Lutein), 글리세린(glycerin), 유화제, 결착제, 올리고당 및 물을 유효성분으로 함유하는 액상용 조성물을 제공한다.
또한, 본 발명은 상기 액상용 조성물을 유효성분으로 함유하는 눈 건강 개선용 건강식품을 제공한다.
아울러, 본 발명은 상기 액상용 조성물을 유효성분으로 함유하는 사료 조성물을 제공한다.
루테인(Lutein)은 난용성의 성질로 인해 현재 알약이나 캡슐 등에 넣은 제품으로만 시판되고 있으나, 본 발명은 루테인을 액상으로 안정화시키는 최적 조성을 확인하였고, 상기 최적 조성으로 제조된 액상용 조성물은 다양한 온도 조건에서 성상이 안정적이고, 침전이 발생하지 않으므로, 눈 건강을 위한 건강식품 또는 애완동물 등의 사료로 유용하게 사용될 수 있다.
도 1a는 본 발명의 <실시예 1> 및 <실시예 2>에서 제조된 루테인을 함유하고, 풀루란을 포함 또는 미포함하는 액상 사료 조성물의 4℃에서 온도 안정도를 확인한 도이다.

이하, 본 발명을 상세히 설명한다.
본 발명은 루테인(Lutein), 글리세린(glycerin), 유화제, 결착제, 올리고당 및 물을 유효성분으로 함유하는 액상용 조성물을 제공한다.
상기 루테인은 하기 화학식 1로 기재되는 화합물로 피로한 눈의 건강을 도와주는 영양소로 알려져 있으며, 태양 및 산화에 의한 손상으로부터 눈을 보호해 주는 역할을 수행하며, 다양한 건강식품에 이용되고 있다.

상기 유화제는 글리세린 지방산 에스테르, 레시틴, 메타인산나트륨, 소르비탄 지방산 에스테르, 스테아릴젖산나트륨, 스테아릴젖산칼슘, 자당 지방산 에스테르, 폴리옥시에틸렌 글리세롤 지방산 에스테르, 폴리옥시에틸렌 소르비탄 지방산 에스테르, 폴리인산나트륨 및 스테아릴젖산나트륨으로 구성된 군으로 부터 선택되는 어느 하나 또는 둘 이상인 것이 바람직하나 이에 한정되지 않는다.
또한, 상기 결착제는 난용성의 성질의 루테인을 액상에서 안정화시키는 주요 성분으로 천연 결착제, 합성 결착제 및 합제로 구성된 군으로 부터 선택되는 어느 하나인 것이 바람직하고, 상기 결착제를 포함하지 않을 경우, 루테인의 액상 안정성이 현저히 저해되므로, 루테인을 포함하는 안정화된 액상용 조성물을 제조하기 위해서는 결착제, 특히 풀루란이 포함되어야 한다. 한편, 상기 천연 결착제는 풀루란, 구아검, 리그닌술폰산염(리그닌설폰산염), 셀룰로오스, 송진, 젤라틴, 천연검, 카제인 및 콜라겐으로 구성된 군으로 부터 선택되는 어느 하나인 것이 바람직하고, 합성 결착제는 알긴산나트륨, 알긴산암모늄, 알긴산칼륨, 카르복시메틸셀룰로오스나트륨, 카제인나트륨, 폴리메틸로카바마이드, 폴리아크릴산나트륨 및 히드록시프로필메틸 셀룰로오스로 구성된 군으로 부터 선택되는 어느 하나인 것이 바람직하며, 상기 합제는 천연결착제 합제, 합성결착제 합제 및 결착제 합제인 것이 바람직하나 이에 한정되지 않는다.
또한, 상기 올리고당은 갈락토올리고당, 겐티오올리고당, 말토올리고당, 이소말토올리고당, 자일로올리고당, 키토올리고당, 프락토올리고당 및 올리고당 합제로 구성된 군으로 부터 선택되는 어느 하나인 것이 바람직하나 이에 한정되지 않는다.
한편, 본 발명의 액상용 조성물은 식향(flavor), 아미노산계 첨가제, 비타민계 첨가제 및 기능성 첨가제로 구성된 군으로 부터 선택되는 어느 하나 이상을 추가적으로 포함할 수 있으며, 상기 식향은 기호를 증진시켜 섭취의 용이성을 증가시키는 것이므로, 향신료로부터 추출한 정유, 감귤류 과피에서 추출되는 정유, 커피나 바닐라 콩의 추출물 등 알려진 천연 착향료, 합성착향료 다양하게 사용될 수 있다.
또한, 성장과 발육에 도움을 줄 수 있는 아미노산계 첨가제, 비타민계 첨가제 및 기능성 첨가제도 함께 포함할 수 있으며, 아미노산계 첨가제는 DL-메치오닌(DL-메티오닌), DL-메치오닌수산화유도체(MHA), DL-메치오닌수산화유도체 칼슘염, DL-알라닌, DL-트레이닌철, DL-트립토판, L-글라타민산, L-라이신, L-라이신염산염, L-라이신황산염, L-메치오닌(L-메티오닌), L-발린, L-시스틴, L-알기닌(L-아지닌, L-아르기닌), L-이소로이신, L-트레오닌, L-트립토판, L-히스티딘, L-히스티딘염산염, L-시스테인염산염, 아미노초산(글라이신), 타우린, 아미노산제 합제 및 L-로이신으로 구성된 군으로 부터 선택되는 어느 하나 또는 둘 이상인 것이 바람직하다.
또한, 상기 비타민계 첨가제로는, L-카르니틴, 나이아신(니아신-니코틴산), 니코틴산아미드, 비오틴(비타민H), 비타민A분말(레티놀분말), 비타민A유(레티놀오일), 비타민B1, 비타민B1나프탈린-1,5-디설폰산염(치아민나프탈린-1,5-디설폰산염), 비타민B1라우릴황산염(치아민 라우릴황산염), 비타민B1로단산염(치아민 티오시안산염), 비타민B1염산염(치아민 염산염), 비타민B1질산염(치아민 질산염), 비타민B2(리보플라빈), 비타민B2인산에스테르나트륨(리보플라빈 5’-인산에스테르나트륨), 비타민B6염산염(피리독신 염산염), 비타민B12(시아노코발라민), 비타민C(L-아스콜빈산), 비타민C칼슘염(L-아스콜빈산 칼슘염), 비타민C-2인산마그네슘염(L-아스콜빈산-2-인산에스테르 마그네슘), L-아스콜빈산-2-폴리에틸렌 글리콜 에테르, 비타민C나트륨염(L-아스콜빈산 나느륨염), 비타민D분말(칼시페롤분말), 비타민D2(칼시페롤), 비타민D3(콜레칼시페롤), 비타민E(dl-알파-토코페롤), 비타민E아세테이트(dl-알파-토코페롤아세테이트), d-알파-비타민E아세테이트, 비타민K1(파일로퀴논), 비타민K3(메나디온), 비타민K3아황산나트륨염(메나디온아황산나느륨염), 비타민K3아황산니코틴아마니드(메나디온 아황산 니코틴아마이드), 비타민K4(아세토메나프톤), 염화콜린, 엽산, 이노시톨,주석산수소콜린, 판토텐산 및 프로비타민A(베타-카로틴)로 구성된 군으로 부터 선택되는 어느 하나 또는 둘 이상인 것이 바람직하나 이에 한정되지 않는다.
또한, 상기 기능성 첨가제는 코엔자임큐텐, 빌베리 추출물, 가시오갈피추출물, 감초추출물, 금잔화추출물(루테인, 마리골드 색소 포함), 라즈베리추출물, 목초액, 밀크씨슬추출물, 아티초크추출물, 우엉추출물, 유카추출물, 차추출물, 천연비테인(베타인), 치커리추출물, 칡뿌리추출물, 퀼라야(퀼레아), 타히보추출물, 황금뿌리추출물, 달맞이꽃종자유, 자몽종자추출물, 포도종자추출물, 해바라기씨추출물, 호로파추출물, 참깨유추출물, 글루칸, 만난, 누에추출물, 키토산, 키틴으로 구성된 군으로 부터 선택되는 어느 하나인 것이 바람직하나, 인체 또는 포유류의 기능성이 알려져 있는 물질은 어느 것이라도 사용될 수 있다.
한편, 본 발명의 액상용 조성물은 액상용 조성물 100 중량부에 대하여 루테인 0.04 내지 0.2 중량부, 글리세린 65 내지 75 중량부, 유화제 3 내지 5 중량부, 결착제 4 내지 8 중량부, 올리고당 4 내지 8 중량부 및 물 10 내지 15 중량부인 것이 바람직하고, 상기 액상용 조성물 100 중량부에 대하여 루테인 0.04 내지 0.3 중량부, 글리세린 65 내지 75 중량부, 유화제 3 내지 5 중량부, 결착제 4 내지 8 중량부, 올리고당 4 내지 8 중량부, 식향 0.1 내지 0.3 중량부, 비타민계 첨가제 0.01 내지 0.05 중량부, 기능성 첨가제 0.05 내지 0.2 중량부 및 물 10 내지 15 중량부인 것이 보다 바람직하다.
본 발명의 구체적인 실시예에서, 본 발명자들은 루테인을 함유하고, 풀루란이 함유 또는 미함유된 액상 사료를 제조하였다(표 1 및 표 2 참조).
또한, 상기 제조된 액상 사료에 대한 온도 안정성을 평가한 결과, 루테인을 함유하고, 풀루란을 포함하는 액상 사료 조성물은 60일 동안 4℃, 25℃, 40℃, 45℃ 모든 온도 조건에서 액상 안정성을 나타낸 반면, 루테인을 함유하고, 풀루란을 미포함하는 액상 사료 조성물은 성상이 분리되는 것을 확인하였다(표 3, 도 1 및 도 2 참조).
따라서, 본 발명의 루테인을 함유하고, 풀루란을 포함하는 액상 사료 조성물은 다양한 온도 조건에서 성상이 안정적이고, 침전이 발생하지 않으므로, 건강식품 또는 애완동물 등의 사료로 유용하게 사용될 수 있음을 확인하였다.
아울러, 본 발명은 본 발명의 액상용 조성물을 포함하는 눈 건강 개선용 건강식품 또는 사료 조성물을 제공한다.
본 발명의 '사료'는 애완동물을 포함하는 가축, 동물, 어류 등에 영양이 되거나 건강유지 또는 성장에 필요한 것을 의미하며, 본 발명의 사료는 '배합사료' 또는 '보조사료'일 수 있으나, 이에 한정되지 않는다.
구체적으로, 상기 '보조사료'는 사료의 효능을 높이기 위하여 사료에 첨가하는 것을 의미하고, 상기 '배합사료'는 보조사료를 기존 사료 등에 적정한 비율로 배합 또는 가공한 것을 의미하므로, 본 발명의 사료는 작접 사용되거나, 배합사료 또는 보조사료로 사용될 수 있다.
본 발명의 액상용 조성물은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 그의 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 일반적으로, 건강식품 중의 액상용 조성물은 전체 식품 중량의 0.01 내지 15 중량%로 가할 수 있으며, 건강 음료 조성물은 100 ㎖를 기준으로 0.02 내지 5 g, 바람직하게는 0.3 내지 1 g의 비율로 가할 수 있다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
이하, 본 발명을 실시예 및 실험예에 의하여 상세히 설명한다.
단, 하기 실시예 및 실험예는 본 발명을 구체적으로 예시하는 것이며, 본 발명의 내용이 실시예 및 실험예에 의해 한정되는 것은 아니다.
<실시예 1> 루테인을 함유하는 액상 사료의 제조
하기 [표 1]에 개시된 조성으로 루테인을 함유하는 액상 사료 조성물을 제조하였다.
구체적으로, 글리세린 지방산 에스테르, 글리세린, 정제수, 루테인, 토코페롤을 80℃까지 가온하여 용해하였다. 그 후, 유화기를 사용하여 유화한 후, 프락토올리고당, 플루란, 코엔자임큐텐, 빌베리추출물, 착향료를 투입한 후 30분간 교반한다. 그런 다음, 70℃에서 60분간 멸균처리한 후 저장통에 저장하여 루테인을 함유하는 액상 사료를 제조하였다.
<실시예 2> 루테인을 함유하고, 풀루란이 미함유된 액상 사료의 제조
하기 [표 2]에 개시된 조성으로 루테인을 함유하고, 풀루란이 미함유된 액상 사료 조성물을 제조하였다.
또한 액상 사료 조성물 제조방법은 상기 <실시예 1>과 동일한 방법으로 제조하였다.
<실험예 1> 온도 안정성 평가
상기 <실시예 1> 및 <실시예 2>에서 제조된 루테인을 함유하고, 풀루란을 포함 또는 미포함하는 액상 사료 조성물의 온도 안정도를 평가하였다.
구체적으로, 상기 <실시예 1> 및 <실시예 2>에서 제조된 조성물을 온도별 조건 4℃, 25℃, 40℃, 45℃ 인큐베이터에 넣고 일주일 간격으로 60일동안 성상, 색상, 분리 및 침전되는 현상을 관찰하였다.
그 결과, 하기 표 3, 도 1 및 도 2에 나타낸 바와 같이 루테인을 함유하고, 풀루란을 포함하는 액상 사료 조성물은 60일 동안 4℃, 25℃, 40℃, 45℃ 모든 온도 조건에서 액상 안정성을 나타낸 반면, 루테인을 함유하고, 풀루란을 미포함하는 액상 사료 조성물은 성상이 분리되는 것을 확인하였다(표 3, 도 1 및 도 2).
따라서, 본 발명의 루테인을 함유하고, 풀루란을 포함하는 액상 사료 조성물은 다양한 온도 조건에서 성상이 안정적이고, 침전이 발생하지 않으므로, 건강식품 또는 애완동물 등의 사료로 유용하게 사용될 수 있음을 확인하였다.

- 루테인(Lutein), 글리세린(glycerin), 유화제, 결착제, 올리고당 및 물을 유효성분으로 함유하는 액상용 조성물.
- 제 1항에 있어서, 상기 조성물은 식향(flavor), 아미노산계 첨가제, 비타민계 첨가제 및 기능성 첨가제로 구성된 군으로 부터 선택되는 어느 하나 이상을 추가적으로 포함하는 것을 특징으로 하는 액상용 조성물.
- 제 1항에 있어서, 상기 유화제는 글리세린 지방산 에스테르, 레시틴, 메타인산나트륨, 소르비탄 지방산 에스테르, 스테아릴젖산나트륨, 스테아릴젖산칼슘, 자당 지방산 에스테르, 폴리옥시에틸렌 글리세롤 지방산 에스테르, 폴리옥시에틸렌 소르비탄 지방산 에스테르, 폴리인산나트륨 및 스테아릴젖산나트륨으로 구성된 군으로 부터 선택되는 어느 하나 또는 둘 이상인 것을 특징으로 하는 액상용 조성물.
- 제 1항에 있어서, 상기 결착제는 천연 결착제, 합성 결착제 및 합제로 구성된 군으로 부터 선택되는 어느 하나인 것을 특징으로 하는 액상용 조성물.
- 제 4항에 있어서, 상기 천연 결착제는 풀루란, 구아검, 리그닌술폰산염(리그닌설폰산염), 셀룰로오스, 송진, 젤라틴, 천연검, 카제인 및 콜라겐으로 구성된 군으로 부터 선택되는 어느 하나인 것을 특징으로 하는 액상용 조성물.
- 제 4항에 있어서, 상기 합성 결착제는 알긴산나트륨, 알긴산암모늄, 알긴산칼륨, 카르복시메틸셀룰로오스나트륨, 카제인나트륨, 폴리메틸로카바마이드, 폴리아크릴산나트륨 및 히드록시프로필메틸 셀룰로오스로 구성된 군으로 부터 선택되는 어느 하나인 것을 특징으로 하는 액상용 조성물.
- 제 4항에 있어서, 상기 합제는 천연결착제 합제, 합성결착제 합제 및 결착제 합제 로 구성된 군으로 부터 선택되는 어느 하나인 것을 특징으로 하는 액상용 조성물.
- 제 1항에 있어서, 상기 올리고당은 갈락토올리고당, 겐티오올리고당, 말토올리고당, 이소말토올리고당, 자일로올리고당, 키토올리고당, 프락토올리고당 및 올리고당 합제로 구성된 군으로 부터 선택되는 어느 하나인 것을 특징으로 하는 액상용 조성물.
- 제 1항에 있어서, 상기 액상용 조성물 100 중량부에 대하여 루테인 0.04 내지 0.3 중량부, 글리세린 65 내지 75 중량부, 유화제 3 내지 5 중량부, 결착제 4 내지 8 중량부, 올리고당 4 내지 8 중량부 및 물 10 내지 15 중량부인 것을 특징으로 하는 액상용 조성물.
- 제 1항에 있어서, 상기 액상용 조성물 100 중량부에 대하여 루테인 0.04 내지 0.3 중량부, 글리세린 65 내지 75 중량부, 유화제 3 내지 5 중량부, 결착제 4 내지 8 중량부, 올리고당 4 내지 8 중량부, 식향 0.1 내지 0.3 중량부, 비타민계 첨가제 0.01 내지 0.05 중량부, 기능성 첨가제 0.05 내지 0.2 중량부 및 물 10 내지 15 중량부인 것을 특징으로 하는 액상용 조성물.
- 제 1항의 액상용 조성물을 유효성분으로 함유하는 눈 건강 개선용 건강식품.
- 제 1항의 액상용 조성물을 유효성분으로 함유하는 사료 조성물.
- 제 12항에 있어서, 상기 사료는 보조 사료 또는 배합사료인 것을 특징으로 하는 사료 조성물.
