- Plasma was separated from the blood by centrifugation, and after deproteinization with acetonitrile, the plasma concentration of Compound 1 was measured using an LC/MS/MS apparatus (XEVO TQ-XS, manufactured by WATERS).
- Figure 1 shows the changes in blood concentration of each formulation.
- pharmacokinetic parameters were calculated by non-compartmental analysis using analysis software Phoenix WinNonlin 8.2 (manufactured by Certara LP). Table 10 shows the results.
- AUC plasma concentration-area under the time curve
- Cmax maximum plasma concentration after oral administration
- BA bioavailability
- rBA relative Bioavailability
- BA was calculated as the ratio (AUCsol/AUCiv or AUCLC/AUCiv) of the AUC (AUCsol) of the solution formulation or the AUC (AUCLC) of the absorption enhancing formulation to the AUC (AUCiv) of the iv formulation when the same compound was administered.
- rBA was calculated as the ratio (AUCLC/AUCsol) of the AUC (AUCLC) of the absorption-promoting preparation to the AUC (AUCsol) of the solution preparation when the same compound was administered.
- the AUCs of the groups administered with the absorption-enhancing preparations (1)-(3) of Examples 1-3 were all greater than the AUC of the group administered with the solution preparation (1) of Comparative Example 1.

- the AUC of the absorption-enhancing formulation (13)-administered group of Example 13 was greater than the AUC of the solution formulation (7)-administered group of Comparative Example 7, and an increase in Cmax was also observed ( Table 16). From this, it was confirmed that by using lauroyl-L-carnitine, a compound with low membrane permeability exhibits higher absorbability than when lauroyl-L-carnitine is not used.
- Rat PK test (30 mg/kg) The blood kinetics after oral administration of the solution formulation (39) prepared in Comparative Example 39 and the absorption enhancement formulations (49) to (54) prepared in Examples 49 to 54 in rats were evaluated in the same manner as in Evaluation Example 4. evaluated.
- Table 48 shows the results of pharmacokinetic parameters calculated by the same analysis as in Evaluation Example 4 from the obtained plasma concentration transition. As a result, it was confirmed that the AUC of the absorption enhancer formulations (49) to (54) administration groups of Examples 49 to 54 was greater than the AUC of the solution formulation (39) administration group of Comparative Example 39. An increase was also observed (Table 48).
- the CV value which indicates the variation in AUC for each individual, was also reduced. From this, it is possible to exhibit sufficiently high absorbency by using a surfactant having a linear alkylene structure and having 5 or more and 13 or less carbon atoms in the alkylene structure as a surfactant. was confirmed. Furthermore, by using lauroyl-L-carnitine, it was confirmed that the compound with low membrane permeability exhibits higher absorbability and suppression of variation in absorbed amount compared to the case where lauroyl-L-carnitine is not used. rice field.
- lauroyl-L-carnitine suppresses the expression of claudin protein and widens tight junctions (Drug Metab. Pharmacokinet., 26(2): 162170 (2011)), and peptide compounds pass through tight junctions. It is thought that it assists in the uptake into cells as a result.
- lauroyl-L-carnitine since lauroyl-L-carnitine has a function as a surfactant, it pulls out phospholipids, which are fat-soluble components present in the cell membrane, making the cell membrane surface "sparse” and entering the phospholipid membrane. It is thought that this makes it easier for the peptide compound to pass through the cell membrane.
