Abstract
본 발명은 2종의 단백질 아미노산 서열에 존재하는 하나씩의 라이신 잔기를 치환 하는 것을 포함하여 단백질의 반감기를 증가시키는 방법 또는 반감기가 증가 된 단백질에 관한 것으로서, 본 발명의 라이신 잔기가 치환된 단백질은 인체 내에 서 오랜 시간 동안 잔류하며 치료효과가 우수하다. 또한 한 번의 생산 과정으로 2종의 단백질을 생산 제공할 수 있다.
Description
본 발명은 기능성 화장품 원료인 FGF-1과 EGF를 생물학적 연결부위(linker)를 이용하여 하나의 결합체로 제조하는 방법에 관한 것이다. 또한 상기 결합체의 반감기를 포함하는 2종 기능성 화장품 원료 및 이를 포함하는 화장료 조성물에 관한 것이다.
기능성 화장품은 일반적으로 세정과 미용 목적 외의 특수한 기능이 부여된 화장품을 의미하며, 피부의 미백에 도움을 주는 제품, 피부의 주름개선에 도움을 주는 제품, 피부를 곱게 태워주거나 자외선으로부터 피부를 보호하는 데에 도움을 주는 제품, 및 특정 기능을 가진 헤어 케어 제품 등을 포함한다. 이러한 기능성 화장품의 제조에 사용되는 원료 물질로서 EGF (Epidermal Growth Factor; 상피세포 성장인자) 또는 FGF (Fibroblast Growth Factor; 섬유아 세포 성장인자)가 개발되어 이용되고 있다. EGF는 상처 회복 촉진 역할을 하는 것으로 알려져 있으며, "피부재생" 효과를 주된 기능으로 하는 화장품 원료로서 널리 사용되고 있다. FGF는 혈관계 세포의 증식을 촉진하거나 신경세포의 성장을 촉진하는 펩티드로서 알려져 있으며, "주름개선 효과"를 주된 기능으로 하는 화장품 원료로서 사용되고 있다. 이와 같이 이들 원료는 노화방지용 (anti-aging) 기능성 소재로서 화장품에 사용되고 있으나, 고가이고 안정성이 낮으며 반감기가 짧아 실질적인 기능을 제공하기 보다는 마케팅에 활용하기 위한 컨셉 원료로서 사용되고 있다. 따라서 가격 경쟁력을 가지면서도 안정성이 높고 반감기를 증가된 소재 개발이 요구되고 있다.
이에 본 발명자들은 대표적인 기능성 화장품 원료인 EGF와 FGF-1이 링커에 의해 결합된 결합체를 제조하고 이들 단백질의 반감기 및 안정성을 높이기 위한 연구를 수행 하였다. 그 결과 이들 기능성 성분의 생산성과 효율성을 높이고, 각 소재의 장점이 결합된 안정형 이중 기능성 화장품 원료를 하나의 공정으로 생산하는 방법을 개발함으로써 본 발명을 완성하였다.
본 발명은 기능성 화장품 원료인 FGF-1과 EGF를 생물학적 연결 부위 (linker, 링커)를 이용하여 하나의 결합체로 제조하는 방법을 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 2종의 단백질 결합체를 포함하는 이중 기능성 화장료 조성물을 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 2종의 단백질 결합체의 반감기 및 안정성을 증가시키는 방법을 제공하는 것을 목적으로 한다.
또한, 본 발명은 반감기 및 안정성이 증가된 상기 2종의 단백질 결합체를 포함하는 화장료 조성물을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위해, 본 발명은 단백질, 펩타이드 또는 폴리펩타이드에서 유비퀴틴(ubiquitin)의 C-말단의 글리신(glycine)과 결합하는 라이신 (lysine) 중 하나 이상을 아르기닌(arginine)으로 치환하는 것을 포함하는, 단백질, 펩타이드 또는 폴리펩타이드의 반감기(half-life)를 증가시키는 방법을 제공한다. 이와 관련된 일 실시예에서, 상기 단백질이 AUT-FGF-1와 AUT-EGF 결합체인 것인, 단백질, 펩타이드 또는 폴리펩타이드의 반감기를 증가시키는 방법을 제공한다. 이와 관련된 다른 일 실시예에서, 상기 AUT-FGF-1와 AUT-EGF 결합체가 서열번호: 3의 아미노산 서열 또는 서열번호: 4의 염기서열을 가지며, 이의 FGF-1의 N-말단으로부터 106째 위치 및 EGF의 N-말단으로부터 28째 위치의 라이신 잔기가 각각 아르기닌으로 치환된 것인, 단백질, 펩타이드 또는폴리펩타이드의 반감기를 증가시키는 방법을 제공한다.
본 발명은 또한 FGF-1-5linkers-EGF 및 AUT-FGF-1-5linkers-AUT-EGF를 숙주세포에 발현 및 정제하는 방법을 제공한다. 이와 관련된 일 실시예에서, 본 발명은 상기 단백질을 엔코딩하는 염기서열; 및 임의의 링커를 포함하는 발현벡터로서, 상기 프로모터와 염기서열이 작동적으로 연결된 것인, 발현벡터를 제공한다. 이와 관련된 또 다른 일 실시예에서, 본 발명은 상기 발현 벡터를 포함하는 숙주세포를 제공한다.
본 발명에서 상기 단백질의 체내 반감기 및 안정성을 증가시키는 것은 단백질의 유비퀴틴화를 억제하는 것에 의해 구현된다. 유비퀴틴화(ubiquitination)는 유비퀴틴이 표적단백질과 결합하는 현상으로, 단백질 분해조절 기전에 작용하여 세포의 항상성에 큰 영향을 끼친다. 특정 단백질의 유비퀴틴화가 진행되면, 단백질은 세포질 내의 프로테아좀(proteasome)에 의해 분해되며 이를 유비퀴틴-프로테아좀 경로(ubiquitin-proteasome pathway)라 한다. 체내에 유입되거나 발현되는 대부분의 단백질들은 유비퀴틴화 과정을 거치게 되며, 이 과정은 E1 (ubiquitin-activating), E2 (ubiquitin-conjugating), E3 (ubiquitin- ligase) 효소에 의해 순차적으로 촉진되는 것으로 알려졌다. 본 발명자들은 선행연구에서 이와 같은 유비퀴틴-프로테아좀 경로(ubiquitin-proteasome pathway)를 차단함으로써 단백질의 분해를 억제하는 기술을 개발하여, AUTTM 기술로 명명한 바 있다. 본 발명자들은 상기 AUTTM 기술을 EGF (Epidermal Growth Factor; 상피세포 성장인자)와 FGF-1 (Fibroblast Growth Factor; 섬유아 세포 성장인자-1)을 적용하였다. 구체적으로, 유비퀴틴화를 차단함으로써 단백질 분해를 억제하기 위해 상기 AUTTM 기술을 이용하여 상기 단백질에 대한 아미노산 치환을 수행하였으며, 세포 내에서 이들 단백질 분해가 감소되는 것을 확인하였고, 표면 전하(Surface Charge)가 변화되는 것을 확인하였다. 그 결과, 반감기 및 안정성이 향상된 AUTTM-FGF-1과 AUTTM-EGF를 수득하였으며, 이들 단백질이 결합된 안정형 기능성 화장품 원료를 제조하였다.
본 발명에서, 단백질의 라이신 잔기는 보존적 아미노산으로 치환될 수 있다. 본 발명에서, "보존적 아미노산 치환"은 아미노산 잔기가 유사한, 예를 들어, 전하 또는 소수성을 갖는 화학적 특성을 갖는 측쇄를 가지는 다른 아미노산 잔기에 의해 치환되는 것을 의미한다. 일반적으로 보존적 아미노산 치환에 의해 단백질의 기능적 특성은 실질적으로 변화하지 않는다. 유사한 화학적 특성을 갖는 측쇄를 갖는 아미노산 그룹의 예는 1) 지방족 측쇄: 글리신, 알라닌, 발린, 류신 및 이소류신; 2) 지방족-하이드록실 측쇄: 세린 및 트레오닌; 3) 아미드-함유 측쇄: 아스파라긴 및 글루타민; 4) 방향족 측쇄: 페닐알라닌, 티로신 및 트립토판; 5) 염기성 측쇄: 라이신, 아르기닌 및 히스티딘; 6) 산성 측쇄: 아스파르 테이트 및 글루타메이트 7) 황-함유 측쇄: 시스테인 및 메티오닌을 포함한다.
본 발명에서 단백질의 라이신 잔기는 염기성 측쇄를 포함하는 아르기닌 또는 히스티딘으로 치환될 수 있으며, 바람직하게는 아르기닌 잔기로 치환된다.
본 발명은 2종의 단백질이 링커에 의해 결합된 결합체를 제공한다. 또한 상기 2종의 단백질의 결합체는 이들 단백질의 아미노산 서열에 존재하는 하나의 라이신 잔기가 아르기닌으로 치환되어 증가된 반감기 및 안정성을 제공할 수 있다. 따라서 본 발명에 따른 2종의 단백질 결합체는 이중 기능성을 갖는 화장품 원료로서 널리 사용될 수 있다.
도 1은 생물학적 연결 부위를 이용하여 결합되어 제공되는 FGF-1과 EGF 결합체의 도식화 및 뉴클레오티드와 염기 서열을 나타낸 것이다. 도 1에서 링거 서열 GGGGS 또는 GGG GGT GGA GGC TCT을 중심으로 좌측은 FGF-1 서열을 우측은 EGF 서열을 나타낸다.

도 9는 NIH3T3 세포를 이용하여 AUT-FGF-1과 AUT-EGF 결합체의 세포 생장 효능을 확인한 것이다. 비교 처리는 기존 FGF-1을 사용하였고 처리 농도는 50 ng/ml로 진행하였다.
본 발명의 일 실시예에서, 단백질은 FGF-1과 EGF의 결합체이다. 서열번호 1은 생물학적 연결 부위를 이용하여 결합되어 제공되는 FGF-1과 EGF의 결합체의 아미노산 서열이며, 서열번호: 2는 이의 염기서열이다. 또한 서열번호 3은 생물학적 연결 부위를 이용하여 결합되어 제공되는 AUT-FGF-1과 AUT-EGF의 결합체의 아미노산 서열이이며, 서열번호: 4는 이의 염기서열이다. 실시예 1에 따르면 본 발명의 AUT-FGF-1과 AUT-EGF의 결합체는 한 번의 제조 공정으로 2종의 단백질을 생산할 수 있는 장점을 제공한다. 실시예 2에서 FGF-1 아미노산 서열의 N-말단으로부터 106번째 라이신 잔기가 아르기닌 잔기로 치환되고, EGF의 아미노산 서열의 N-말단으로부터 28번째 라이신 잔기 하나가 아르기닌 잔기로 각각 치환하였다. 상기 실시예 2의 치환된 아미노산 서열을 갖는 단백질들은 증가된 반감기 및 안정성을 제공한다.
본 발명의 실시예에서, 반감기 증가 확인을 위한 플라스미드 DNA를 제작하기 위하여 내부 제한효소 BamHI 부위를 제거하였다. 이를 위해 먼저 FGF-1의 N-말단에서부터 39번째 아르기닌 잔기를 암호화하는 뉴클레오티드 서열을 AGG에서 AGA로 치환하되 아르기닌 잔기에는 변화가 없도록 하였다. 특정 돌연변이를 유도할 DNA 서열을 이용하여 프라이머를 제작한 후, PCR을 수행하여 특정 뉴클레이티드 서열을 치환시킨 플라스미드 DNA를 제작하였다. 본 발명의 다른 실시예에서, 2종의 단백질을 하나로 결합하기 위하여 생물학적 연결 부위, 링커로서 GGGGS (서열번호: 5)를 사용하였다. E. coli에서 발현시킨 상기 단백질 결합체는 5개의 상기 링커서열 (GGGGSGGGGSGGGGSGGGGSGGGGS)를 반복적으로 사용하였다. 본 발명의 다른 일 실시예에서, 단백질의 아미노산 서열에 존재하는 라이신 잔기를 아르기닌 (arginine, R) 잔기로 치환시키기 위해 부위특이적 돌연변이유도 (site-directed mutagenesis)를 이용하였다. 이 방법은 특정 돌연변이를 유도할 DNA 서열을 이용하여 프라이머를 제작한 후, 특정조건에서 PCR을 진행함으로써 특정 아미노산 잔기를 치환시킨 플라스미드 DNA를 제작한다.
본 발명에서 약학조성물은 경구 (oral), 경피 (transcutaneous), 피하 (subcutaneous), 정맥내 (intravenous) 또는 근육 내 투여를 포함하는 다양한 경로로 체내 전달될 수 있으며, 주사형 제제로 투여될 수 있다. 또한, 본 발명의 약학 조성물은 상기 방법에 따라 투여된 후에 신속한 방출, 지연된 방출 또는 천천히 방출 되도록 당업자에게 잘 알려진 방법에 따라 제형화 될 수 있다. 상기 제형은 정제 (tablet), 알약 (pill), 분말 (powder), 사셰 (sachet), 엘렉시르제 (elixir), 현탁 (suspension), 에멀션 (emulsion), 용액 (solution), 시럽 (syrup), 에어로졸 (aerosol), 소프트 또는 단단한 젤라틴 캡슐 (soft and hard gelatin capsule), 멸균주사용액 (sterile injectable solution), 멸균 팩키지된 분말 등을 포함한다. 적합한 담체, 부형제 및 희석제로는, 락토오스 (lactose), 덱스트로오스 (dextrose), 수크로오스 (sucrose), 만니톨 (mannitol), 자일리톨 (xylitol), 에리스리톨 (erythritol), 말티톨 (maltitol), 탄수화물 (starches), 검 아카시아 (gum acacia), 알지네이트 (alginates), 젤라틴 (gelatin), 인산칼슘 (calcium phosphate), 규산칼슘 (calcium silicate), 셀룰로오스 (cellulose), 메틸셀룰로오스 (methyl cellulose), 마이크로크리스탈린셀룰로오스(microcrystalline cellulose), 폴리비닐 피롤리돈 (polyvinyl pyrrolidone), 물, 메틸히드록시벤조에이트 (methylhydroxybenzoates), 프로필히드록시벤조에이트 (propylhydroxybenzoates), 활석 (talc), 스테아린산마그네슘 (magnesium stearate) 및 미네랄 오일을 포함한다. 또한, 제형은 충진제, 항교착제 (anti-agglutinating agents), 윤활제 (lubricating agents), 습윤제 (wetting agents), 향미료 (flavoring agents), 유화제 (emulsifiers), 보존제 (preservative) 등을 추가로 포함할 수 있다.
본 발명에서, 단수형태는 달리 명확하게 기술하지 않는 한 복수형태를 포함한다. 또한, 본 발명에서 구성된, 갖는 이루어진 과 같은 용어는 “포함하는”과 유사한 의미를 갖는 것으로 해석된다. 본 발명에서, “생리활성 (폴리)펩타이드 또는 단백질”은 인간을 포함하는 포유동물에 투여되었을 때 유용한 생물학적 활성을 나타내는 (폴리)펩타이드 또는 단백질을 의미한다.
이하, 실시예에 의거하여 본 발명을 보다 더 상세히 설명한다. 하기 실시예는 본 발명을 예시하기 위한 것을 뿐, 본 발명이 하기 실시예에 의해 제한되는 것은 아니다.
실시예
실시예1: 2종의 기능성 화장품 원료 결합체의 반감기 증가 확인
1. 라이신 (Lysine, K) 잔기의 치환
부위 특이적 돌연변이유도 (site-directed mutagenesis)를 이용하여 라이신 잔기를 아르기닌(Arginine, R)으로 치환하였다. 이를 위해. 프라이머 (FGF-1 K106R FP 5'-CATGCAGAGAGGAATTGGTTT-3' (서열번호: 6), RP 5'-AAACCAATTCCTCTCTGCATG-3' (서열번호: 7) 및 EGF K28R FP 5'-GAAGCATTGGACAGGTATGCATGCAAC-3' (서열번호: 8), RP 5'-GTTGCATGCATACC TGTCCAATGCTTC-3' (서열번호: 9)를 제작한 후, PCR을 수행하여 특정 아미노산 잔기를 치환시킨 플라스미드 DNA를 제작하였다. pcDNA3-myc-FGF-1-linker-EGF을 템플릿으로 사용하여, 라이신 잔기가 아르기닌으로 치환 (K→R)된 플라스미드 DNA를 제작하였다.
2. 반감기 증가 확인
(1) 인간 세포 내에서 반감기 증가 확인을 위한 발현 벡터 클로닝
2종의 기능성 화장품 원료 결합체 개발을 위하여 FGF-1과 EGF 연결하기 위해서 생물학적 연결 부위 (GGGGS)를 이용하였으며, FGF-1 내의 BamHI 제한효소 자리를 없앤 후, pcDNA3-myc (5.6kb) (Thermo Fisher Scientific)를 제한효소인 BamHI과 XhoI로 절편을 만든 후 접합하여 클로닝하였다 (도 1과 2).
(2) 반감기 증가 확인
pcDNA3-myc-FGF-1-linker-EGF 및 pcDNA3-myc-AUT-FGF-1-linker-AUT-EGF를 각각 8 ㎍씩 HEK293T 세포(Sigma-Aldrich)에 형질감염 (transfection)시켰다. 형질감염 24시간 후, 단백질생성 저해제 시클로헥시미드 (cycloheximide) (CHX) (Sigma-Aldrich) (100 ㎍/㎖)을 처리하고 2시간, 4시간 및 6시간에 걸쳐서 반감기를 측정하였다. 그 결과, 인간 AUT-FGF-1와 AUT-EGF 결합체의 분해가 억제되는 것을 확인하였다 (도 3). 인간 FGF-1과 EGF 결합체의 반감기는 1시간 이내인 반면 인간 AUT-FGF-1와 AUT-EGF 결합체의 반감기는 2시간 이상이었으며, 2시간 이후부터 6시간 사이에는 분해가 더욱 억제되는 것을 확인하였다 (도 3).
실시예2: 2종의 기능성 화장품 원료 결합체의 발현 정제 조건 확립
1. 결합체 발현 및 정제
(1) E.coli에서 결합체 발현 및 정제를 위한 발현 벡터 클로닝
FGF-1과 EGF 연결하기 위해서 생물학적 연결 부위(GGGGS)를 5번 반복적으로 이용하였으며, pET28a (5.6kb) (Sigma-Aldrich)를 제한효소인 NcoI과 XhoI로 절편을 만든 후 접합하여 클로닝하였다 (도 4와 5).
(2) 발현 정제 조건 확립
AUT-FGF-1-5linkers-AUT-EGF의 발현 유도
FGF-1-5linkers-EGF 및 AUT-FGF-1-5linkers-AUT-EGF의 아미노산 서열이 암호화된 재조합 DNA가 transformation된 BL21 (DE3) (Thermo Fisher Scientific) 균주를 Kanamycin이 포함된 선택 LB media에 접종하여 37℃, 300 rpm 조건에서 OD600값이 0.57에 도달할 때까지 진탕 배양하였다. 다음, 0.5 mM isopropyl-b-D-thiogalactopyranoside(IPTG)를 첨가하고 여러 배양 조건에 따른 발현 양상을 확인하였다 (도 6). 가장 적정한 발현 조건으로서, 25℃에서 16시간 진탕 배양을 진행하였다. FGF-1-5linkers-EGF도 위와 동일하게 수행하였다.
AUT-FGF-1-5linkers-AUT-EGF의 세포 획득
배양액을 3000 rpm, 4℃ 조건에서 20분간 원심분리를 수행하여 세포 펠릿(cell pellet)을 획득하였다. FGF-1-5linkers-EGF의 경우도 동일하게 수행하였다.
AUT-FGF-1-5linkers-AUT-EGF의 용해
세포 펠릿을 용해 완충액 (lysis buffer) (50 mM Tris-HCl (pH8.0), 0.5 mM EDTA, 1 mM PMSF, 10% Glycerol)을 배양액 1 L 기준으로 100 ml을 첨가하고 4℃ 조건에서 소니케이션 (sonication) 하여 용해시켰다. 용해물의 불용성 펠릿 (insoluble pellets)을 리폴딩 (refolding) 하기 위해 13000 rpm, 4℃ 조건에서 원심분리한 후, 수득한 불용성 펠릿을 변성 완충액 (denaturation buffer) (50 mM Tris-HCl (pH8.0), 0.5 mM EDTA, 1 mM PMSF, 10% Glycerol, 8 M Urea)를 이용하여 1시간 동안 녹인 후, 13000 rpm, 4℃ 조건에서 15분간 원심 분리하였다. 이어서, 용해된 펠릿을 리폴딩 완충액 (50 mM Tris-HC (pH8.0), 1 mM EDTA, 1 mM PMSF, 10% Glycerol)에서 4℃ 하에 16시간 방치한 후 13000 rpm, 4℃ 조건에서 15분간 원심 분리하여 상층액을 수득하였다. FGF-1-5linkers-EGF에 대해서도 상기와 같이 수행하였다.
AUT-FGF-1-5linkers-AUT-EGF의 affinity chromatography를 이용한 정제
AKTA에 Q column을 장착하고 이퀼리브레이션 완충액 (equilibration buffer) (50 mM Tris-HCl (pH8.0), 0.5 mM EDTA, 1 mM PMSF, 10% Glycerol)로 이퀼리브레이션 한 후, AUT FGF-1-5 linkers-EGF를 걸어주었다. 이후 6배 양의 세척 완충액 (wash buffer) (50 mM Tris-HCl (pH8.5), 0.5 mM EDTA, 1 mM PMS, 10% Glycerol, 50 mM NaCl)로 컬럼을 세척한 후, AUT FGF-1-5 linkers-EGF는 300 mM NaCl를 포함하는 세척 완충액과 동일한 완충액으로 용리하였다 (도 7). FGF-1-5linkers-EGF에 대해서도 상기와 동일하게 수행하였다.
AUT-FGF-1-5linkers-AUT-EGF의 크기 배제 크로마토그래피 (Size exclusion chromatography)를 이용한 정제
용리액은 최종 정제를 위해 50 mM Tris-HCl (pH8.0)와 400 mM NaCl로 구성된 버퍼로 이퀼리브레이션된 HiPrep 16/60 Sepahacryl S-100 컬럼에 흡착시켰다. 다음, 정제된 AUT-FGF-1-linker-AUT-EGF를 인산완충식염수 (phosphate buffered saline) (pH7.4) 투석하였다 (도 7). FGF-1-5linkers-EGF에 대해서도 상기와 동일하게 수행하였다.
AUT-FGF-1-5linkers-AUT-EGF의 순도 확인
최종 정제된 AUT-FGF-1-linker-AUT-EGF 5 ㎍을 SDS-PAGE로 확인하였다 (도 8). FGF-1-5linkers-EGF에 대해서도 위와 동일하게 수행하였다.
실시예3: 2종의 기능성 화장품 원료 결합체의 세포 생장 효과 확인
최종 정제된 AUT-FGF-1-linker-AUT-EGF을 50 ng/ml로 NIH3T3 (Sigma-Aldrich) 섬유아세포에 처리하여 세포 생장율을 비교 분석하였다 (도 9). 양성 대조군으로는 기존 FGF-1을 단독 처리하였다.
Claims (7)
- 단백질, 펩타이드 또는 폴리펩타이드에서 유비퀴틴(ubiquitin)의 C-말단의 글리신(glycine)과 결합하는 라이신 (lysine) 중 하나 이상을 아르기닌(arginine)으로 치환하는 것을 포함하는, 단백질, 펩타이드 또는 폴리펩타이드의 반감기(half-life)를 증가시키는 방법.
- 제 1항에 있어서, 상기 단백질이 AUT-FGF-1와 AUT-EGF 결합체인 것인, 단백질, 펩타이드 또는 폴리펩타이드의 반감기를 증가시키는 방법.
- 제 2항에 있어서, 상기 AUT-FGF-1와 AUT-EGF 결합체가 서열번호: 1의 아미노산 서열을 가지며, 이의 FGF-1의 N-말단으로부터 106째 위치 및 EGF의 N-말단으로부터 28째 위치의 라이신 잔기가 각각 아르기닌으로 치환된 것인, 단백질, 펩타이드 또는폴리펩타이드의 반감기를 증가시키는 방법.
- FGF-1-5linkers-EGF 및 AUT-FGF-1-5linkers-AUT-EGF를 숙주세포에서 발현 및 정제하는 방법.
- 프로모터, AUT-FGF-1 및 AUT-EGF 단백질을 엔코딩하는 염기서열, 및 임의의 링커를 포함하는 발현벡터로서, 상기 프로모터와 염기서열이 작동적으로 연결된 것인, 발현벡터.
- 제5항의 발현벡터를 포함하는 숙주세포.
- 제4항의 방법에 따라 수득한 단백질 결합체를 포함하는 화장료 조성물.
