Abstract

本発明は、生体条件に近い環境下で凝集性タンパク質の凝集体を可視化する方法を提供することを目的とし、具体的には、タンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を通液することで、凝集性タンパク質含有溶液を回収する方法;当該回収方法により回収した凝集性タンパク質含有溶液において、該凝集性タンパク質の凝集体を可視化することを含む、凝集性タンパク質の凝集体の可視化方法;並びに40kDa以上の凝集阻害物質を除去した培地又は培養上清中で標識された目的の凝集性タンパク質の凝集体を形成させることで、凝集を可視化する方法に関する。

Description

凝集性タンパク質の測定方法及び凝集可視化方法、並びにこれらの方法に使用するためのキット
 本発明は、例えば凝集性タンパク質含有溶液の回収方法、凝集性タンパク質の凝集量の測定方法及び凝集性タンパク質の凝集体の可視化方法、並びにこれらの方法に使用するためのキットに関する。
 アルツハイマー病(Alzheimer's disease:本明細書では、しばしば「AD」と略称する)は、不可逆的な進行性中枢神経疾患の一種であり、認知機能障害(認知症)、行動障害、又は人格変化等の症状を伴う。全世界における認知症の患者数は、2019年時点で5,000万人以上と推定されているが、約70%がAD患者と考えられており、その発症率は増加傾向にある。AD患者の増加に伴う医療費の増大や介護問題は、国や患者関係者達にとって経済的又は精神的負担になる等、近年大きな社会的問題となっている。
 ADは、患者脳内で疎水性のペプチドであるアミロイドβタンパク質(本明細書では、しばしば「Aβ」と表記する)の凝集及び蓄積に始まり、その後、微小管結合タンパク質であるタウタンパク質が過リン酸化されて線維化した後に、神経細胞が破壊され、脳が萎縮することによって発症する(非特許文献1~3)。
 このADの発症機序に基づき、in vitroで添加した被験物質のAβ凝集阻害効果を評価し、AD治療の候補化合物をスクリーニングする技術が開発されている。例えば、量子ドットナノプローブを用いたアミロイドβタンパク質凝集阻害物質の微量ハイスループットスクリーニング(Microliter-scale High-throughput Screening:本明細書では、「MSHTS」と表記する)法は、PBS溶媒中でアミロイドβタンパク質の凝集阻害効果を有する候補化合物を探索できるCell-free Assays (無細胞試験)系を基盤技術としたスクリーニング技術である(非特許文献4)。
 MSHTSの例として、例えば、特許文献1には、アミロイド形成を評価する方法、装置及びプログラムが開示されており、具体的には、水又はPBS等の適当な緩衝液中で、被験物質の存在下又は非存在下で、アミロイドβタンパク質等のアミロイド形成性タンパク質と、当該アミロイド形成性タンパク質の重合により形成されるアミロイドへの結合能を有する蛍光プローブ(量子ドットを蛍光色素として含む、又は量子ドット等)とを反応させる凝集反応ステップと、凝集反応ステップにおいて得た凝集反応生成物の蛍光を撮像する撮像ステップと、撮像ステップにおいて撮像した蛍光画像内の関心領域に含まれる各画素の輝度値から標準偏差を算出する標準偏差算出ステップと、標準偏差算出ステップにおいて算出した被験物質存在下での輝度値標準偏差の値と被験物質非存在下での輝度値標準偏差の値との比較から、被験物質存在下での輝度値標準偏差の値が被験物質非存在下での輝度値標準偏差の値より小さい場合に、被験物質がアミロイド形成阻害活性を有すると判定する活性判定ステップとを含む、被験物質のアミロイド形成阻害活性を判定する方法が開示されている。
 また、特許文献2には、タンパク質やペプチドのアミロイド凝集性を評価するための汎用性量子ドットナノプローブ、及び当該量子ドットナノプローブを用いたアミロイド形成阻害剤の評価方法が開示され、具体的には、アミロイド形成ペプチドのN末端又はC末端にシステインを介して量子ドットが結合した量子ドットナノプローブが開示されている。
 しかしながら、Cell-free Assays系で得られた候補化合物はCell-Based Assays(細胞を使用した試験)系では、その阻害効果が認められない場合が多い。これはCell-free Assays系が生体環境を必ずしも反映していないことが原因と考えられる。生体環境により近い培地中でMSHTSを実施することができれば、生体内でも阻害効果が期待できる候補化合物や候補素材を精度高くかつ効率的にスクリーニングすることが可能となる。ところが、量子ドットを培地等に添加した場合、Aβが凝集しないという新たな問題が生じていた。それ故に、これまでは、生体環境により近い環境下では候補化合物を探索するスクリーニング系として活用できなかった。

 本発明は、上述の実情に鑑み、生体条件に近い環境下であってもAβ等の凝集性タンパク質の凝集体を可視化する方法を提供することを目的とする。
 PBSより生体条件に近い環境下(例えば、細胞が培養可能な培地等)で凝集性タンパク質(例えば、アミロイドβタンパク質)の凝集体を可視化する技術を構築する一つのツールを見出すことで、本技術開発にて、より精度高く生体内での効果が期待出来る化合物を見出すことが期待できる。
 生体環境下に近い条件(例えば、培地、又は培養上清)において、凝集性タンパク質(例えば、アミロイドβタンパク質)の凝集体は、PBSのような環境下とは異なり、様々な夾雑物の共存下で形成される。本発明者等は、培地又は培養上清中にはアミロイドβタンパク質をはじめとする凝集性タンパク質の凝集を阻害する何らかの夾雑物が混在すると仮定し、その探索を行った。その結果、培地又は培養上清中に40kDa以上の夾雑物(例えば、アクチンタンパク質(本明細書では、しばしば「アクチン」と表記する)、アルブミンタンパク質(本明細書では、しばしば「アルブミン」と表記する)及びグロブリン等)が含まれている場合、アミロイドβタンパク質の凝集が阻害され、凝集を可視化できないことが判明した。そこで、限外濾過フィルター等を用いて凝集を阻害する40kDa以上の夾雑物を培地中から除去した後、その培地中で再度アミロイドβタンパク質を凝集させた場合には、凝集体を可視化できることが明らかとなった。一態様において、本発明は、当該新たな知見に基づくものである。
 一方、培地中に含まれる夾雑物を除去すると培地中に含まれる凝集性タンパク質が夾雑物除去工程で消失し、回収できる目的タンパク総量が低減してしまう。
 そこで、上記課題を解決するため鋭意研究を行った結果、タンパク凝集促進可視化液を使用することにより、上述の夾雑物除去工程において、タンパク質の吸着低減処理を施し、効率的に凝集性タンパク質を回収でき、さらに回収液(濾液)中で凝集性タンパク質の凝集体を可視化できることを見出し、本発明を完成するに至った。
 すなわち、本発明は、以下を包含する。
 (1)濾過フィルターをタンパク凝集促進可視化液で通液する第1工程と、タンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を通液する第2工程とを含む、凝集性タンパク質含有溶液の回収方法。
 (2)第1工程前に、濾過フィルターを洗浄液で通液する前処理工程をさらに含む、(1)記載の方法。
 (3)第2工程前に、凝集性タンパク質含有溶液中の凝集性タンパク質を定量する工程をさらに含む、(1)又は(2)記載の方法。
 (4)第2工程後、濾液中の凝集性タンパク質を定量する工程をさらに含む、(1)~(3)のいずれか1記載の方法。
 (5)濾過フィルターが、50kDa以下の公称分画分子量(NMWL)を有する限外濾過フィルターである、(1)~(4)のいずれか1記載の方法。
 (6)タンパク凝集促進可視化液が、50kDa以下の界面活性作用のある物質を含む、(1)~(5)のいずれか1記載の方法。
 (7)界面活性作用のある物質が界面活性剤である、(6)記載の方法。

 本発明によれば、凝集性タンパク質を含む溶液から夾雑物を除去しつつ、凝集性タンパク質を効率的に回収し、回収液中にて凝集性タンパク質の凝集体を可視化することができる。また、本発明によれば、培地又は培養上清中で、凝集性タンパク質を凝集させ、その凝集を可視化することができる。

 以下、本発明を詳細に説明する。
1.凝集性タンパク質含有溶液の回収方法
1-1.概要
 本発明の第1の態様は、凝集性タンパク質含有溶液の回収方法である。本態様の方法は、濾過フィルターをタンパク凝集促進可視化液で通液する第1工程と、タンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を通液する第2工程とを含むことを特徴とする。本態様の方法によれば、凝集性タンパク質を含む溶液から夾雑物(凝集阻害物質)を除去しつつ、凝集性タンパク質を効率的に回収することができる。
1-2.用語の定義
 本明細書で使用する以下の用語について定義する。なお、特に断りのない限り、本項に記載の以下の定義は、本発明の他の態様においても共通する。
(1)濾過フィルター
 「濾過フィルター」とは、濾過によって所定の分子量の物質を分画できるフィルターをいう。例えば、限外濾過フィルターが挙げられる。限外濾過フィルターの種類は限定しないが、本発明では、凝集性タンパク質の凝集を阻害する40kDa以上の凝集阻害物質を除去可能な公称分画分子量(NMWL)を有する限外濾過フィルターが好ましい。一般に、多くの限外濾過フィルターでは、NMWLよりも若干低い分子量も濾過できることが知られている。例えば、50 kDa(UFC505096, Merck)の濾過フィルターであれば、分子量67kDaのBSA溶液(1mg/mL)を約90%分画できるが、分子量45kDaのオボアルブミン溶液(1mg/mL)であっても、約半量程度(スイングバスケットローターで~65%、固定角ローターで~55%)は分画できることが知られている(Amicon(登録商標)Ultra-15 Centrifugal Filter Devices User Guide)。つまり、NMWLが50kDaであっても、実質的には40kDa以上のタンパク質を除去することが可能である。従って、限定はしないが、NMWLが50kDa以下の限外濾過フィルターは本発明で使用する濾過フィルターとして好適である。50kDa以下のNMWLを有する限外濾過フィルターとしては、例えばアミコンウルトラ(Amicon Ultra)-0.5 ウルトラセル(Ultracel)-50メンブレン、50 kDa(UFC505096, Millipore)が挙げられる。
(2)凝集阻害物質
 「凝集阻害物質」とは、凝集性タンパク質の凝集を阻害する物質をいう。本発明では、40kDa以上の凝集阻害物質が該当する。したがって、本明細書では特段の断りのない限り、単に「凝集阻害物質」と表記した場合、40kDa以上の凝集阻害物質を意味するものとする。40kDa以上の凝集阻害物質は、40kDa以上、41kDa以上、又は42kDa以上の物質であれば特に限定はしない。また、40kDa以上の凝集阻害物質は、例えば250kDa以下、240kDa以下、230kDa以下、220kDa以下、又は210kDa以下の分子量を有する物質である。40kDa以上の凝集阻害物質の具体的な例として、アクチンタンパク質、アルブミン及びグロブリンから成る群から選択されるいずれか一以上のポリペプチド又はその断片から成る凝集阻害物質が挙げられる。
 凝集性タンパク質含有溶液中に凝集阻害物質が特定量以上含まれている場合、凝集性タンパク質は凝集体を形成することができない。したがって、凝集阻害物質の存在が凝集性タンパク質による凝集体の可視化の阻害原因となる。本発明では、凝集阻害物質が除去された凝集性タンパク質含有溶液を用いて凝集性タンパク質を凝集させることを最大の特徴とする。なお、本明細書において「凝集阻害物質が除去された」とは、後述する除去ステップに記載の方法等を用いて凝集阻害物質を凝集性タンパク質含有溶液(又は培地若しくは培養上清)から除去すること、又は除去工程を経ずとも既に除去された状態、すなわち最初から包含されていないことをいう。除去工程を経る場合、除去後の凝集性タンパク質含有溶液において凝集阻害物質は100%除去されていることが好ましいが、凝集阻害物質が凝集性タンパク質の凝集を阻害できない程度の量であれば、残存していてもよい。例えば、凝集性タンパク質含有溶液において0.5% v/v以下、0.4% v/v以下、0.3% v/v以下、0.2% v/v以下、又は0.1% v/v以下であれば、凝集阻害物質の残存は許容範囲である。
(3)タンパク凝集促進可視化液
 タンパク凝集促進可視化液は、濾過フィルターを用いた凝集阻害物質除去の過程において、濾過フィルターに通液し、タンパク質の吸着を低減し、回収液中にて凝集性タンパク質の凝集を促進し、該凝集を可視化することができる溶液である。
 タンパク凝集促進可視化液としては、例えば50kDa以下の界面活性作用のある物質を含むものが挙げられる。界面活性作用のある物質としては、例えば界面活性剤が挙げられ、好ましくは、界面活性剤は非イオン界面活性剤である。非イオン界面活性剤としては、例えばエステル型非イオン界面活性剤、エーテル型非イオン界面活性剤、エステルエーテル型非イオン界面活性剤、アルキルグリコシド型非イオン界面活性剤が挙げられる。
 エステル型非イオン界面活性剤としては、例えば、アルキル硫酸エステル塩、ポリオキシエチレンアルキル硫酸エステル塩、グリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、糖脂肪酸エステル、脂肪酸メチルエステルエトキシレートなどが挙げられる。
 エーテル型非イオン界面活性剤としては、例えば、ポリオキシエチレンアルキルエーテル、脂肪酸メチルエステルエトキシレート、ポリオキシエチレンアルキルフェニルエーテル、ポリオキシエチレンポリオキシプロピレングリコールなどが挙げられる。
 エステルエーテル型非イオン界面活性剤としては、例えばTween-20(Polysorbate 20又はポリエチレングリコールソルビタンモノラウラートとも称される)、Tween-40、Tween-60、Tween-80、Tween-85が挙げられる。
 アルキルグリコシド型非イオン界面活性剤としては、例えばn-オクチル-β-D-グルコシド、n-オクチル-β-D-マルトシド、n-デシル-β-D-グルコシド(デシルグルコシド)、n-デシル-β-D-マルトシド、n-ドデシル-β-D-グルコシド(ラウリルグルコシド)、n-ヘプチル-β-D-チオグルコシド、n-オクチル-β-D-チオグルコシド、n-ノニル-β-D-チオマルトシドなどが挙げられる。
 好ましくは、非イオン界面活性剤は、エステルエーテル型非イオン界面活性剤であり、特に好ましくは、エステルエーテル型非イオン界面活性剤は、Tween-20である。例えば、タンパク凝集促進可視化液として、Tween-20を含む水溶液が挙げられる。当該水溶液中におけるTween-20の濃度は、例えば0.1~10%、好ましくは0.01~1%、特に好ましくは0.05~0.5%である。
(4)凝集性タンパク質
 凝集性タンパク質とは、集合して、凝集体を形成するタンパク質をいう。凝集性タンパク質の種類は限定しない。例えば、ポリグルタミン酸、疾患に関連するタンパク質、又はオートファジー関連タンパク質が挙げられる。
 疾患に関連する凝集性タンパク質には、例えば、アルツハイマー病の原因タンパク質であるアミロイドβタンパク質及びタウタンパク質(リン酸化タウタンパク質を含む)、パーキンソン病の原因タンパク質であるα-シヌクレインタンパク質、伝達性海綿脳症(クロイツフェルトヤコブ病、狂牛病、又はプリオン病を含む)の原因タンパク質であるプリオンタンパク質、ハンチントン病の原因タンパク質であるハンチンチンタンパク質、II型糖尿病の原因タンパク質であるアミリンタンパク質、動脈硬化症(脳梗塞、肺梗塞、心筋梗塞を含む)の原因タンパク質であるアポリポタンパク質A1(APOA1タンパク質)、間接リウマチの原因タンパク質である血清アミロイドAタンパク質、全身性ALアミロイドーシスの原因タンパク質である免疫グロブリン軽鎖、透析アミロイドーシスの原因タンパク質であるβ2ミクログロブリン、又は筋萎縮性側索硬化症の原因タンパク質とされるTDP-43タンパク質が挙げられる。特に、アミロイドβ42タンパク質、アミロイドβ43タンパク質、アミロイドβ38タンパク質等のアミロイドβタンパク質は好適である。
 オートファジー関連タンパク質には、例えば、ユビキチン様タンパク質であるAtg-8、Atg-12が挙げられる。
 あるいは、凝集性タンパク質としては、例えばタウタンパク質(例えば、リン酸化タウタンパク質)、α-シヌクレイン、アミロイドβタンパク質、プリオンタンパク質、TDP-43、ポリグルタミン酸、Atg-8、Atg-15が挙げられ、好ましくはアミロイドβ42タンパク質、アミロイドβ43タンパク質、アミロイドβ38タンパク質等のアミロイドβタンパク質である。
 凝集性タンパク質は、自然界に存在する天然タンパク質であってもよいし、天然タンパク質に人工的に変異や修飾を導入した改変タンパク質であってもよいし、又は人工的に設計されたアミノ酸配列に基づく人工タンパク質であってもよい。
 本明細書における凝集性タンパク質及び後述する凝集性断片は、その全部又は一部が標識されている。ここでいう「全部」とは、全ての凝集性タンパク質又は凝集性断片をいい、また「一部」とは、凝集性タンパク質群又は凝集性断片群において、一以上全部未満の凝集性タンパク質又は凝集性断片が標識されていることをいう。
 本明細書において「凝集性断片」とは、前記凝集性タンパク質における凝集に寄与し得る領域又はドメインを有し、それのみで凝集性タンパク質と同様の凝集活性を有するペプチドをいう。
(5)凝集体
 本明細書において「凝集体」とは、二以上の凝集性タンパク質及び/又はその凝集性断片の集合体をいう。本明細書では、いわゆるタンパク質複合体も凝集体に包含される。
 凝集体は、同一種類のタンパク質又はその凝集性断片からなるホモ凝集体であってもよいし、異なる種類のタンパク質又はその凝集性断片からなるヘテロ凝集体であってもよい。
 凝集体は、それ自身が生物学的機能を有する複合体であってもよいし、単なる集合体であってもよい。
 凝集体の直径(大きさ)は、限定はしないが、通常は10 nm以上~10 mm以下の範囲にあればよい。好ましくは100 nm以上~5 mm以下、より好ましくは500 nm以上~1mm以下である。
(6)凝集性タンパク質含有溶液
 凝集性タンパク質含有溶液は、凝集性タンパク質を含有する限り、いずれの溶液であってよく、例えば、凝集性タンパク質を含有する生理食塩水、緩衝液、培地又は培養上清が挙げられる。好ましくは生体内の環境に近い培地又は培養上清である。例えば、凝集性タンパク質を分泌する細胞を培養後、得られた培養物から培養上清を回収し、回収した培養上清を、凝集性タンパク質含有溶液として使用することができる。
 凝集性タンパク質含有溶液に含有される凝集性タンパク質は、溶媒中に元から含まれていたもの、添加したもの、又はその両方のいずれであってもよい。
 凝集性タンパク質含有溶液には、凝集性タンパク質以外にも、細胞代謝物質、細胞分泌因子、無機物(金属無機塩類)、有機酸が含まれていてもよい。細胞代謝物質としては、例えばグルタミン酸、アスパラギン酸などのアミノ酸類、5'グアニル酸などの核酸類、イソアスコルビン酸などの抗酸化物質類、酢酸、酪酸などの有機酸類、グリセロール等のポリオール類、ビタミンB2、ビタミンAなどのビタミン類が挙げられる。また、細胞分泌因子としては、例えば、Neuronal growth regulator 1 (NEGR1)、Neurogenin-3、Kallikrein-8/Neuropsin (Cleaved-Val33)、Neurofibromin (NF1)、Neuroplastin (NPTN)、Neurotrimin (NT)が挙げられる。無機物(金属無機塩類)としては、金属塩(亜鉛、銅、鉄、セレン等)、無機塩類(Na、K、Ca等)、無機イオン(カチオン:Li+、Na+、Mg2+、K+、Ca2+;アニオン:Cl-、Br-、NO2 -、SO4 2-など)が挙げられる。有機酸としては、コハク酸、ピルビン酸、低級アミン類などが挙げられる。なお、無機物や有機酸の濃度は、限定はしない。例えば、0.001ng/mL~1.00ng/mL、又は0.01ng/mL~0.10ng/mLの範囲であればよい。
(7)細胞
 本発明において対象となる「細胞」は、凝集性タンパク質を産生及び/又は分泌する細胞である。
 細胞は、多細胞生物に由来する細胞であればよい。好ましくは動物由来細胞、より好ましくは哺乳動物由来細胞である。例えば、マウス、ラット、ハムスター、モルモット等のげっ歯類、イヌ、ネコ、ウサギ、ウシ、ウマ、ヒツジ、ヤギ等の家畜又は愛玩動物、そしてヒト、アカゲザル、ゴリラ、チンパンジー等の霊長類が挙げられる。特に好ましくは、ヒト由来細胞である。
 細胞の種類は、限定はしない。例えば、生体組織に由来する細胞、生体組織に由来する細胞から派生した細胞、幹細胞、又は幹細胞から分化した細胞やその前駆細胞等が挙げられる。
 「生体組織」とは、生物の生体を構成する各種組織をいう。例えば、上皮組織、結合組織、筋組織、及び神経組織等が挙げられる。
 「幹細胞」とは、様々な細胞への分化能、及び自己複製能を持つ細胞をいう。例えば、成体幹細胞、及び多能性幹細胞等が挙げられる。
 「成体幹細胞」とは、成体の各組織中に存在し、最終分化が未完了で、ある程度の多分化能を有する幹細胞であって、体性幹細胞又は組織性幹細胞とも呼ばれる。例えば、間葉系幹細胞、神経幹細胞、腸管上皮幹細胞、造血幹細胞、毛包幹細胞、色素幹細胞等が挙げられる。
 「多能性幹細胞」とは、生体を構成する全ての種類の細胞に分化することができる多分化能(多能性)を有し、適切な条件下のインビトロ(in vitro)での培養において多能性を維持したまま無限に増殖を続けることができる細胞をいう。例えば、胚性幹細胞(ES細胞:embryonic stem cell)、胚性生殖幹細胞、生殖系幹細胞、そして人工多能性幹細胞(iPS細胞:induced pluripotent stem cells)等が挙げられる。
 例えば、アルツハイマー病(AD)患者由来iPS細胞から神経前駆細胞を経て分化誘導した神経細胞を、アミロイドβ42タンパク質等のアミロイドβタンパク質を産生及び分泌する細胞として使用することができる。
(8)緩衝液
 本明細書において「緩衝液」とは、細胞やタンパク質等の混合する目的の物質の性質に合わせてpHが所定の範囲に維持されるように調整された溶液をいう。例えば、リン酸緩衝液、HEPESバッファ、NaHCO3/CO2バッファ、トリス塩酸バッファ、グリシンバッファ等が挙げられる。
(9)培地
 本発明において「培地」とは、細胞(例えば、凝集性タンパク質を産生及び/又は分泌する細胞)を培養するために調製された液状又は固形状の物質をいう。原則として、細胞の増殖及び/又は維持に不可欠の成分を必要最小限以上含有する。ただし、本発明では、培地は細胞の培養以外にも凝集性タンパク質の凝集の場として機能し得る。培地は、基礎培地又は特殊細胞培養用培地のいずれであってもよい。
 「基礎培地」とは、様々な動物細胞用培地の基礎となる培地をいう。単体でも培養は可能であり、また様々な培養添加物を加えて、目的に応じた各種細胞に特異的な培地(特殊細胞培養用培地)に調製することもできる。基礎培地としては、Neurobasal(登録商標)培地、BME培地、BGJb培地、CMRL1066培地、Glasgow MEM培地、Improved MEM Zinc Option培地、IMDM培地(Iscove's Modified Dulbecco's Medium)、Medium 199培地、Eagle MEM培地、αMEM培地、DMEM培地(Dulbecco's Modified Eagle's Medium)、ハムF10培地、ハムF12培地、RPMI 1640培地、Fischer's培地、及びこれらの混合培地(例えば、DMEM/F12培地)等の培地を使用することができるが、特に限定されない。その他、ヒトiPS細胞やヒトES細胞の培養に使用されている培地も好適に使用することができる。
 「特殊細胞培養用培地」とは、前述のように基礎培地に各種補助剤を添加することによって、特定の細胞の培養に最適となるように調製された培地、又は特定の細胞に分化誘導できるように調製された培地をいう。例えば、各メーカーで市販されている神経細胞培養用培地が挙げられる。具体的には、例えば、DMEM/F12(5:5)に、インシュリンとトランスフェリンを添加した培地を基礎培地として、栄養培地で培養した初代アストログリア細胞の培養上清と血清アルブミンを加えて調製された住友ベークライト(Sumitomo bakelite)の神経細胞培養用培地にさらにNGF2.5S及びBDNFを補助剤として加えた「M培地」が該当する。ヒトiPS細胞やヒトES細胞等の多能性幹細胞培養用培地も該当する。
 培地は、血清を含む培地、血清を含まない培地(すなわち、無血清培地)のいずれであってもよい。
(10)培養上清
 本明細書において「培養上清」とは、前記培地で細胞を所定の期間培養した後、遠心又は濾過等により細胞を除去して得られる培養液をいう。ここでいう「所定の期間」とは、例えば、一般的な培養条件(例えば5%CO2、37℃)下で、6時間~4日、12時間~3日、18時間~2日、又は1日であればよい。通常は細胞がコンフルエントに達するまでの期間でよい。
 培養上清には、一般的に細胞が代謝、産生、及び/又は分泌した様々な物質(核酸、ペプチド、低分子化合物等)を含む。
(11)標識
 本明細書において「標識する」とは、目的物質を識別するため、その目的物質を修飾することをいう。標識によって目的物質の検出又は選別等が容易、かつ確実となる。
 標識は、目的物質の種類に応じて行われる。本明細書では、凝集性タンパク質又はその凝集性断片の凝集体が検出すべき目的物質であることから、その種類はペプチドである。したがって、標識は、ペプチドを直接的又は間接的に標識することのできるあらゆる標識手段が対象となる。直接的な標識には、凝集性タンパク質又はその凝集性断片に標識物質を結合させる標識方法、又は凝集性タンパク質又はその凝集性断片を標識ペプチドとの融合タンパク質として発現させる標識方法が挙げられる。間接的な標識には、凝集性タンパク質又はその凝集性断片を特異的に認識し、結合する抗体又はその活性断片に直接的に、又は二次抗体を介して間接的に標識する方法が挙げられる。好ましくは標識物質を結合させる方法である。
 ペプチドの標識に用いられる標識物質には、限定はしないが、例えば、光学的標識、酵素、放射性同位元素、又は結合性低分子化合物が挙げられる。
 本明細書において「光学的標識」とは、蛍光物質又は発光物質等のように可視光や近赤外線又は近紫外線の光を発する物質による標識をいう。
 「蛍光物質」とは、特定波長の励起光を吸収することで励起状態となり、元の基底状態に戻る際に蛍光を発する性質を有する物質である。蛍光物質は、蛍光色素、及び蛍光タンパク質のいずれも包含する。
 「蛍光色素」には、例えば、量子ドット、FITC、Texas、Texas Red(登録商標)、Alexa Flour 405、Alexa Flour 488、Alexa Flour 647、Alexa Flour 700、Pacific Blue、DyLight 405、DyLight 550、DyLight 650、PE-Cy5(phycoerythrin-cyanin5)、PE-Cy7 (phycoerythrin -cyanin 7)、PE(phycoerythrin)、PerCP(peridinin chlorophyll protein)、PerCP-Cy5.5(peridinin chlorophyll protein-cyanin5.5)、cy3、cy5、cy7、FAM、HEX、VIC(登録商標)、JOE、ROX、TET、Bodipy493、NBD、TAMRA、Quasar(登録商標)670、Quasar(登録商標)705、APC(Allophycocyanin)、コンゴーレッド、チオフラビンT、チオフラビンS、フルオレサミン若しくはその誘導体、フルオレセイン若しくはその誘導体、アゾ類、又はローダミン若しくはその誘導体、クマリン若しくはその誘導体、ピレン若しくはその誘導体、シアニン若しくはその誘導体等が挙げられる。好ましくは量子ドットである。
 「量子ドット」(Quantum Dot:本明細書では、しばしば「QD」と表記する)とは、量子力学に従う光学特性を有し、可視光及び近赤外領域において蛍光を発するナノスケールの半導体結晶をいう。通常、直径2nm~10nmで、10~50個ほどの原子で構成され、粒径に依存した多数の蛍光色が得られる他、蛍光退色が起こりにくい等の優れた特徴を有していることからバイオセンシング材料、及び細胞若しくは動物のイメージング材料として応用が進んでいる。
 「蛍光タンパク質」には、例えば、GFPが挙げられる。
 「酵素」には、例えば、西洋ワサビペルオキシダーゼ(Horseradish peroxidase:HRP)、アルカリフォスファターゼ(Alkaline Phosphatase: ALP)、グルコースオキシダーゼ(Glucose oxidase:GOx)が挙げられる。
 「放射性同位元素」とは、質量数が異なる同位元素のうち、放射線を放出する元素をいう。例えば、35S等が挙げられる。
 本明細書において「結合性低分子化合物」とは、特定の結合性タンパク質と親和性に基づいて結合する低分子化合物をいう。具体例として、ビタミンB7として知られるビオチンが挙げられる。ビオチンは、卵白由来のタンパク質であるアビジン、その誘導体であるストレプトアビジン又はニュートラアビジンと極めて高い親和性を有し、互いに強固に結合する。この性質を利用して、抗体をビオチン標識しておくことで、アビジン標識した他の標識物質(蛍光色素等)が結合し、抗体標識することができる。
(12)可視化
 本明細書において「可視化」とは、凝集性タンパク質及び/又はその凝集性断片の凝集を肉眼又は光学顕微鏡(蛍光顕微鏡、微分干渉顕微鏡を含む)で認識できる状態にすることをいう。通常は凝集体の形成によって可視化できることから、凝集性タンパク質及び/又はその凝集性断片による凝集の可視化と、それらの凝集体の形成は同義である。前記認識は、形成された凝集体の視覚による確認の他、凝集性タンパク質が標識されている場合、その標識(蛍光又は発光等)の強度や状態(塊状又は斑点状等)等に基づく認識が挙げられる。
1-3.方法
 本態様の方法は、濾過フィルターをタンパク凝集促進可視化液で通液する第1工程と、タンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を通液する第2工程とを必須の工程として、第1工程前に、濾過フィルターを洗浄液で通液する前処理工程;第2工程前に、凝集性タンパク質含有溶液中の凝集性タンパク質を定量する工程;第2工程後、濾液中の凝集性タンパク質を定量する工程、を選択工程として含む。
 以下、それぞれの工程について、説明をする。
1-3-1.第1工程前に、濾過フィルターを洗浄液で通液する前処理工程
 濾過フィルターを洗浄液で通液し、遠心分離に供することで洗浄する。この操作を1回以上、好ましくは2回以上繰り返す。洗浄液として水酸化ナトリウムを使用すると、濾過操作時に交雑物がゲル化する虞があり、また、濾過処理で得られた濾液中で凝集性タンパク質が凝集体を形成しなくなる虞がある。従って、洗浄液としては、例えば純水、PBS、生理食塩水などが挙げられる。遠心分離は、例えば15~25℃(例えば室温)の温度条件下、3,000~25,000g(例えば14000g)で3~15分間(例えば10分間)行う。
1-3-2.濾過フィルターをタンパク凝集促進可視化液で通液する第1工程
 濾過フィルターをタンパク凝集促進可視化液で通液することで、その後、通液する凝集性タンパク質含有溶液中のタンパク質の吸着を低減する。濾過フィルターにタンパク凝集促進可視化液を滴下し、例えば15~25℃(例えば室温)の温度条件下、0.5~4時間(例えば2時間)インキュベーションを行う。インキュベーション後、遠心分離を、例えば15~25℃(例えば室温)の温度条件下、3,000~25,000g(例えば14000g)で3~15分間(例えば10分間)行う。さらに、第1-3-1節に記載の前処理工程同様、濾過フィルターを洗浄液で通液し、遠心分離に供することで洗浄してもよい。
1-3-3.第2工程前に、凝集性タンパク質含有溶液中の凝集性タンパク質を定量する工程
 第2工程前に、事前に凝集性タンパク質含有溶液中の凝集性タンパク質を定量する。凝集性タンパク質の定量方法としては、例えば凝集性タンパク質に対する抗体を用いた免疫学的測定法が挙げられる。このような免疫学的測定方法としては、例えば酵素免疫測定法(ELISA)、イムノクロマトグラフィー法が挙げられる。
1-3-4.タンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を通液する第2工程
 第1工程後のタンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を滴下し、濾過処理を行う。滴下後、遠心分離を、例えば10~25℃(例えば室温)の温度条件下、3,000~25,000g(例えば14000g)で3~60分間(例えば30分間)行い、濾液を回収する。回収した濾液には、凝集性タンパク質が含まれ、タンパク凝集促進可視化液の使用によって、濾液において凝集性タンパク質の凝集体を可視化することができる。
1-3-5.第2工程後、濾液中の凝集性タンパク質を定量する工程
 第1-3-3節同様に、得られた濾液中の凝集性タンパク質を定量する。この定量により、第2工程前に、事前に定量した凝集性タンパク質含有溶液中の凝集性タンパク質量と比較し、凝集性タンパク質の回収率を得ることができる。
 具体的には、下記1)式により、凝集性タンパク質の回収率を算出することができる:
 凝集性タンパク質の回収率(%)=第2工程後の凝集性タンパク質の定量値/第2工程前の凝集性タンパク質の定量値×100  ‐‐‐‐‐式1)
 1)式で求められた凝集性タンパク質の回収率が、例えば60%を超える、好ましくは65%以上、70%以上、特に好ましくは75%以上である場合に、本態様の方法により凝集性タンパク質を有意に回収できたと判断することができる。
1-4.効果
 本態様の方法によれば、凝集性タンパク質を効率的に回収することができる。また、タンパク凝集促進可視化液の使用により、回収液中にて凝集性タンパク質の凝集体を可視化することができる。
2.凝集性タンパク質の凝集量の測定方法、又は凝集性タンパク質の凝集可視化方法
2-1.概要
 本発明の第2の態様は、凝集性タンパク質の凝集量の測定方法(本明細書では、しばしば「凝集量測定方法」と略記する)、又は凝集性タンパク質の凝集可視化方法(本明細書では、しばしば「凝集可視化方法」と略記する)である。本態様の凝集量測定方法又は凝集可視化方法は、凝集性タンパク質及び/又はその凝集性断片の凝集を阻害する凝集阻害物質を除いた凝集性タンパク質含有溶液を用いることによって可視可能な凝集体を形成させ、それを検出することを特徴とする。本態様の凝集量測定方法又は凝集可視化方法によれば、培地や細胞上清のような生体環境により近い環境下での、凝集阻害物質を探索するスクリーニング技術を提供することができる。
 例えば、本発明の第2の態様は、第1の態様の方法により回収した凝集性タンパク質を含有する溶液(濾液)において、該凝集性タンパク質の凝集量を測定することを含む、凝集性タンパク質の凝集量の測定方法、又は第1の態様の方法により回収した凝集性タンパク質を含有する溶液(濾液)において、該凝集性タンパク質の凝集体を可視化することを含む、凝集性タンパク質の凝集体の可視化方法である。第1の態様の方法により回収した凝集性タンパク質を含有する溶液(濾液)においては、タンパク凝集促進可視化液の使用により、凝集性タンパク質の凝集体を可視化することができ、その結果、回収した凝集性タンパク質を含有する溶液(濾液)において、凝集性タンパク質の凝集量を測定することができる。
2-2.方法
 本態様の凝集量測定方法、又は凝集可視化方法は、凝集工程、及び検出工程を必須の工程として含む。以下、それぞれの工程について説明をする。
2-2-1.凝集工程
 「凝集工程」は、全部又は一部が標識された凝集性タンパク質及び/又はその凝集性断片を凝集性タンパク質含有溶液中でインキュベートし、前記凝集性タンパク質及び/又はその凝集性断片を凝集させる工程である。本工程は、凝集阻害物質が除去された凝集性タンパク質含有溶液中で、凝集性タンパク質及び/又はその凝集性断片を凝集させることを特徴とする。
 本工程で、凝集性タンパク質及び/又はその凝集性断片を凝集性タンパク質含有溶液に添加することができる。または、凝集性タンパク質含有溶液が予めそれらを含んでいてもよい。
 インキュベートの条件は、凝集性タンパク質及び/又はその凝集性断片が凝集可能であれば特に限定はしない。例えば、温度は、凝集性タンパク質含有溶液の融点以上、そして凝集性タンパク質及び/又はその凝集性断片の変性温度未満であればよい。好適には5~50℃、10~45℃、15~42℃、20~40℃、25~39℃、30~38℃、又は35~37℃である。また、時間は、限定はしないが、30分~240時間(10日間)の範囲であればよい。例えば、30分~24時間、1時間~20時間、2時間~18時間、4時間~16時間、6時間~14時間、又は8時間~12時間、あるいは24時間(1日間)~240時間(10日間)、48時間(2日間)~216時間(9日間)、72時間(3日間)~192時間(8日間)、96時間(4日間)~168時間(7日間)、又は120時間(5日間)~144時間(6日間)であればよい。
 本工程は、必要に応じて除去ステップを含むことができる。
(1)除去ステップ
 「除去ステップ」は、本工程で使用する凝集性タンパク質含有溶液から凝集阻害物質、すなわち40kDa以上の凝集阻害物質、又はアクチン、アルブミン及びグロブリンからなる群から選択されるいずれか一以上のポリペプチド又はその断片を除去するステップで、凝集工程における選択的ステップである。
 当該除去ステップは、第1の態様の方法であっても良い。
 凝集性タンパク質含有溶液中に凝集阻害物質が含まれる場合、本工程は達成し得ない。したがって、少なくとも凝集開始段階において、本工程で使用する凝集性タンパク質含有溶液中に凝集阻害物質が含まれないことが本工程の前提となる。しかしながら、凝集性タンパク質含有溶液の溶媒が培地又は培養上清等の場合、その溶媒中に凝集阻害物質が混在している可能性がある。本ステップでは、そのような凝集阻害物質を予め除去することを特徴とする。
 凝集阻害物質の除去方法は限定しない。当該分野で公知の方法を用いることができる。例えば、濾過法、吸着法、遠心法、又はそれらの二以上の組み合わせが挙げられる。
 「濾過法」は、フィルターを介した除去方法である。濾過方法では、凝集性タンパク質含有溶液又はその溶媒をフィルターに通液し、凝集阻害物質をその分子量に基づいて前記フィルターで捕捉して除去する。前述のように、除去対象の凝集阻害物質が40kDa以上の凝集阻害物質の場合、40kDa未満で分子量分画が可能な限外濾過フィルターを用いることで、本ステップを達成することができる。また40kDaを超える凝集阻害物質の場合、40kDa以上で分子量分画が可能な限外濾過フィルターを用いることで、本ステップを達成することができる。凝集性タンパク質の分子量が凝集阻害物質のそれと大きく異なる場合、本方法は好適である。なお、濾過に際しては、自然落下による濾過であってもよいが、遠心法と組み合わせた遠心濾過が短時間で処理できるため便利である。試料から特定の分子量の物質を遠心除去する遠心濾過フィルターには、各種ライフサイエンスメーカーから限外濾過フィルターキット等が市販されており、それらを利用してもよい。例えば、Amicon(登録商標) Ultra 50K Kit(Merck社)等が挙げられる。
 「吸着法」は、凝集阻害物質との親和性が高い担体を用いて、その担体の表面及び/又は内部に凝集阻害物質を吸着させて除去する方法である。凝集性タンパク質含有溶液又はその溶媒と担体を混合し、凝集阻害物質を担体に吸着させた後、担体と凝集性タンパク質含有溶液又はその溶媒を分離することで、本ステップを達成することができる。担体の種類は、吸着すべき凝集阻害物質の種類に応じて適宜選択すればよい。例えば、凝集阻害物質がアルブミンの場合、アルブミン吸着性を有するシリカを担体として利用することができる。また、イオン交換の原理を用いてアルブミンを担体表面上に吸着させることもできる。さらに、表面修飾した担体を用いてアルブミンを担体表面上に吸着させることもできる。具体的には、例えば、ミリストイル化試薬を用いて担体表面を修飾する方法、パルミトイル化試薬を用いて担体表面を修飾する方法、ジフェニルシクロヘキサン化合物を用いて担体表面を修飾する方法、又は凝集阻害物質に特異的に結合する、抗体若しくはその活性断片又は核酸アプタマーを用いて担体表面を修飾する方法が挙げられる。
 ミリストイル化試薬を用いた表面修飾は、例えば、N-Boc-N-tetradecanoyl-L-lysine [B5366]のBoc基を除去後、シリカ、アルミナ、ポリスチレン、量子ドット等の担体に[B5366]を修飾することで達成し得る。
 パルミトイル化試薬を用いた表面修飾は、例えば、1-tert-Butyl 5-(N-Succinimidyl)-N-Palmitoyl-L-Glutamateをシリカ、アルミナ、ポリスチレン、量子ドット等の担体に修飾することで達成し得る。
 ジフェニルシクロヘキサン化合物を用いた表面修飾は、例えば、ジフェニルシクロヘキサン化合物をシリカ、アルミナ、ポリスチレン、量子ドット等の担体に修飾することで達成し得る。
 抗体若しくはその活性断片又は核酸アプタマーを用いた表面修飾は、例えば、抗アクチン抗体、抗アルブミン抗体又は抗グロブリン抗体やそれらの活性断片、あるいはアクチン結合性核酸アプタマー、アルブミン結合性核酸アプタマー又はグロブリン結合性核酸アプタマー等を各種担体(樹脂、ガラス、磁性体等)に結合することで達成し得る。
 抗体は、モノクローナル抗体、ポリクローナル抗体、組換え抗体、合成抗体、及び抗体フラグメントを含む。抗体がポリクローナル抗体又はモノクローナル抗体の場合、免疫グロブリン分子は、任意のクラス(例えば、IgG、IgE、IgM、IgA、IgD及びIgY)でよい。ポリクローナル抗体又はモノクローナル抗体は、限定はしないが、通常は哺乳動物又は鳥類等の動物由来の抗体であればよい。例えば、マウス、ラット、モルモット、ウサギ、ヤギ、ロバ、ヒツジ、ラクダ、ウマ、ニワトリ又はヒト等に由来する抗体が挙げられる。
 「組換え抗体」とは、キメラ抗体、ヒト化抗体、又は多重特異性抗体をいう。「キメラ抗体」とは、異なる動物由来の抗体のアミノ酸配列を組み合わせて作製される抗体で、ある抗体の可変領域(V領域)を他の抗体のV領域で置換した抗体である。例えば、マウスモノクローナル抗体のV領域をヒト抗体のV領域と置き換え、可変領域(V領域)がマウス由来、C領域がヒト由来となった抗体が該当する。「ヒト化抗体」とは、ヒト以外の哺乳動物、例えば、適当なマウス抗体の可変領域(V領域)における相補性決定領域(CDR;CDR1、CDR2及びCDR3)をヒトモノクローナル抗体のCDRとを置換したグラフト抗体である。キメラ抗体とヒト化抗体は、重鎖及び軽鎖のV領域、又は重鎖及び軽鎖のV領域における相補性決定領域がマウス等の非ヒト動物抗体由来ではあるが、C領域、又はV領域におけるフレームワーク領域(FR;FR1、FR2、FR3及びFR4)と重鎖及び軽鎖のC領域がヒト抗体由来であることから、ヒト体内における当該抗体に対する免疫反応を軽減し得る。「多重特異性抗体」とは、多価抗体、すなわち抗原結合部位を一分子内に複数有する抗体において、それぞれの抗原結合部位が異なるエピトープと結合する抗体をいう。例えば、IgGのように2つの抗原結合部位を有する抗体で、それぞれの抗原結合部位が異なるエピトープと結合する二重特異性抗体(Bispecific抗体)が挙げられる。
 「合成抗体」とは、化学的に又は組換えDNA法を用いることによって合成した抗体をいう。例えば、組換えDNA法を用いて新たに合成された抗体が挙げられる。具体的には、例えば、単鎖抗体(scFv:single chain Fragment of variable region)、ダイアボディ(diabody)、トリアボディ(triabody)又はテトラボディ(tetrabody)等が挙げられる。
 「抗体フラグメント」とは、例えば、Fab、F(ab’2)、Fv等が該当する。
 抗体は、修飾されていてもよい。抗体の修飾には、機能上の修飾又は標識上の修飾を含む。機能上の修飾には、例えば、グリコシル化、アセチル化、ホルミル化、アミド化、リン酸化、又はPEG化が挙げられる。標識上の修飾には、蛍光色素(フルオレセイン、FITC、ローダミン、テキサスレッド、Cy3、Cy5)、蛍光タンパク質(例えば、PE、APC、GFP)、酵素(例えば、西洋ワサビペルオキシダーゼ、アルカリフォスファターゼ、グルコースオキシダーゼ)、放射性同位元素(例えば、3H、14C、35S)又はビオチン若しくは(ストレプト)アビジンによる標識が挙げられる。
 「アプタマー」とは、立体構造によって標的物質と強固、かつ特異的に結合し、標的物質の機能を特異的に抑制する能力を持つリガンド分子である。「核酸アプタマー」とは、核酸で構成されるアプタマーをいう。核酸アプタマーを構成する核酸は、DNA、RNA又はそれらの組合せのいずれであってもよい。必要に応じて、PNA、LNA/BNA、メチルホスホネート型DNA、ホスホロチオエート型DNA、2'-O-メチル型RNA等の化学修飾核酸を含むこともできる。
 核酸アプタマーは、必要に応じて標識されていてもよい。標識は当該分野で公知のあらゆる核酸用標識物質を利用することができる。例えば、放射性同位元素(例えば、32P、3H、14C)、DIG、ビオチン、蛍光色素(例えば、FITC、Texas、cy3、cy5、cy7、FAM、HEX、VIC、JOE、Rox、TET、Bodipy493、NBD、TAMRA)、又は発光物質(例えば、アクリジニウムエスター)が挙げられる。
 その他、試料からのアルブミンやグロブリンの除去には、各種ライフサイエンスメーカーから除去キットが市販されており、それらを利用してもよい。例えば、AlbuVoidTM(Biotech Support Group社)、Albumin & IgG Depletion SpintrapTM(Cytiva社)、アクチン結合タンパク質スピンダウンBiochem KitTM(Cytoskeleton社)及びProMax Albumin Removal Kit(Polysciences社)等が挙げられる。
 「遠心法」は、遠心分離によって分子量分画を行う方法である。本ステップでは、凝集性タンパク質含有溶液又はその溶媒を遠心後、凝集阻害物質を含む40kDa以上の分画を除去することで達成し得る。必要に応じてショ糖等による密度勾配を用いた遠心による分画を行ってもよい。濾過法と同様に、凝集性タンパク質の分子量が凝集阻害物質のそれと大きく異なる場合に好適な方法である。なお、遠心法は、前述のように、濾過法や吸着法の補助方法として、それらと組み合わせて使用することもできる。
 除去ステップは複数回行ってもよい。一度のステップで凝集性タンパク質含有溶液又はその溶媒から凝集阻害物質を十分に除去できない場合は、特に複数回行うことが好ましい。その場合、各除去ステップの除去方法は同一であっても、又は異なっていてもよい。
2-2-2.検出工程
 「検出工程」は、凝集工程後の凝集体を検出する工程である。本工程では、凝集性タンパク質及び/又はその凝集性断片の凝集によって形成される凝集体を視覚的、及び/又は定量的に検出する。
 凝集体の検出方法は、当該分野で公知のペプチド凝集体の検出方法を用いることができる。一般に凝集体は検出可能なことから目視又は光学顕微鏡を用いた観察によって、その形状から凝集体の有無を確認し、検出することができる。また、凝集性タンパク質及び/又はその凝集性断片が標識されている場合、その標識に基づいた検出方法により凝集体を検出することができる。例えば、凝集性タンパク質及び/又はその凝集性断片が量子ドットのような光学的標識物質で標識されている場合、蛍光顕微鏡を用いた蛍光観察により凝集体を検出すればよい。斑点状の蛍光塊が観察された場合には、凝集体が形成されていると認定することができる。または、蛍光強度を定量化し、陰性対照との蛍光強度の値の比較によって、凝集体の形成の有無を検出してもよい。さらに、ゲル電気泳動のような分子量に基づいた分子篩によって、凝集体を検出することも可能である。
 凝集性タンパク質の凝集量の測定方法又は凝集性タンパク質の凝集体の可視化方法、凝集性タンパク質の凝集抑制活性を算出方法としては、例えば量子ドットナノプローブを用いたアミロイドβタンパク質凝集阻害物質の微量ハイスループットスクリーニング(「MSHTS」)法に準じた方法(特許文献1)が挙げられる。当該方法において可視化することができる凝集性タンパク質の凝集のサイズとしては、例えば0.01~200μm2、好ましくは1~200μm2、特に好ましくは10~150μm2が挙げられる。
 例えば、凝集性タンパク質がアミロイドβタンパク質(例えばアミロイドβ42タンパク質)である場合には、MSHTS法に準じて、第1の態様の方法により回収したアミロイドβタンパク質を含有する溶液に、量子ドット修飾アミロイドβタンパク質(例えば、アミロイドβ40タンパク質)を添加し、該量子ドットを指標に該アミロイドβタンパク質と該量子ドット修飾アミロイドβタンパク質との凝集体を測定するか、又は該量子ドットにより該アミロイドβタンパク質と該量子ドット修飾アミロイドβタンパク質との凝集体を可視化する。
 特許文献1に記載されるように、量子ドットは、三次元的な量子閉じ込め構造を持つナノ材料であり、半導体量子ドット、炭素系量子ドット等が知られている。量子ドットとしては、例えば半導体量子ドット、特にコア・シェル型CdSe/ZnS量子ドットであるQdot(登録商標)525、Qdot545、Qdot565、Qdot585、Qdot605、Qdot655、Qdot705及びQdot800(いずれもThermo Fisher Scientific)等が挙げられる。
 第1の態様の方法により回収したアミロイドβタンパク質を含有する溶液中のアミロイドβタンパク質と量子ドット修飾アミロイドβタンパク質との間の凝集反応は、アミロイドβタンパク質が重合して凝集体を形成することができる反応条件で行われる。凝集反応開始時のアミロイドβタンパク質の濃度は、反応液中の最終濃度で1~100μM程度、好ましくは10~50μMである。また、量子ドット修飾アミロイドβタンパク質の濃度は、アミロイドβタンパク質濃度の0.005~0.5%程度、好ましくは0.01~0.1%である。
 凝集反応は、蛍光観察に一般的に使用されるマイクロプレートのウェル内で行うことができる。アミロイドβタンパク質の凝集量の正確な測定のためには凝集体の厚みを均一にすることが好ましく、その観点で、凝集反応液の液面と凝集体が堆積するウェル底部が水平であること、すなわちウェルの底部形状は平坦であることが好ましい。特に、マイクロリットルオーダーの液量の反応液でアミロイドβタンパク質の凝集量の測定が可能であることから、1536ウェルの平底マイクロプレート等の使用が好ましい。
 凝集反応の温度は、およそ室温~37℃、好ましくは37℃であり、反応時間はおよそ4~36時間、好ましくは12~24時間である。振盪や攪拌は凝集体の形成を過度に促してアミロイドβタンパク質の凝集量の判定に悪影響を及ぼすことがあるため、凝集反応は静置状態で行うことが好ましい。
 次いで、凝集反応において得た凝集反応生成物の蛍光を、撮像される蛍光画像内の関心領域に含まれる各画素の輝度値から算出されるSum Intensityが15~85%となる露出条件で撮像する。
 撮像において、量子ドットに基づく凝集反応生成物(凝集体)の蛍光は、撮像条件の設定及び制御、撮像データの画像化及び表示等の動作を制御するコンピュータと接続された撮像装置を用いて撮像することができる。撮像装置は典型的にはデジタルカメラ(CCDやCMOS)を備えた落射蛍光顕微鏡であり、これに凝集反応生成物を収納した容器、例としてマイクロウェルプレートをセットして励起光を照射し、凝集反応生成物から蛍光を発光させて、CCDカメラで撮像を行う。励起光は、用いられる量子ドットの特性に応じて適宜決定することができる。例えば量子ドットとしてQdot605を用いる場合、励起光の波長は580nmより短波長側にあればよく、例として532~552nmである。生じた蛍光を選択的に透過させて蛍光撮像するためのバンドパスフィルタは、用いられる量子ドットの蛍光波長を含む波長帯域の光のみを選択的に透過することができるものを選択すればよく、例えば量子ドットとしてQdot605を用いる場合、例として594~646nmの波長帯域の光のみを選択的に透過することができるバンドパスフィルタを用いることができる。
 凝集反応生成物(凝集体)の蛍光の撮像は、撮像される蛍光画像内の関心領域に含まれる各画素の輝度値から以下の式2):
 Sum Intensity(%)=(関心領域内の各画素の輝度値の合計/関心領域内の各画素の理論上の最大輝度値の合計)×100
によって算出されるSum Intensityが15~85%となるように露出を制御することにより行われる。
 Sum Intensityを基にした露出制御下での撮像は、具体的には以下のように行われる。最初に、適当な露出条件で予備的に撮像した凝集反応生成物の蛍光画像内に任意の関心領域を設定し、その中に含まれる各画素の輝度値情報を取得する。蛍光画像はカラーであってもモノクロであってもよい。カラー画像の場合は、各画素のRGB値から以下の式によってグレースケール変換を行い、輝度値情報を取得することができる。
 輝度値Y=0.299×R + 0.587×G + 0.114×B
 8ビット画像において、輝度値の範囲は0~255である。従って、例えば関心領域サイズが432ピクセル×432ピクセルの8ビット画像の場合、Sum Intensityは以下の式によって算出される。
 Sum Intensity(%)=(関心領域内に含まれる全186,624個の画素の輝度値の合計/(186,624×255))×100
 なお、蛍光画像のビット数は、8ビットであることが好ましい。8ビットより大きいビット数の画像を用いることも可能であるが、画像処理の負荷が大きいため、8ビット画像に変換した後にSum Intensityを算出することが望ましい。
 このように算出された予備的蛍光画像からのSum Intensityを参照して、Sum Intensityが所定の設定値となるように露出を制御することで、撮像が行われる。露出は、露光時間及びカメラゲイン(ISO感度)の調節並びに減光フィルターの使用により、特に露光時間及びカメラゲイン(ISO感度)を適宜調節して制御することができ、市販CCDカメラに備わる自動露出機能を利用することが好ましい。例えば、予備的蛍光画像から算出されたSum Intensityが所定の設定値を下回る場合、露光時間を長くする及び/又はカメラゲインを上げることにより、Sum Intensityを上昇させることができる。また予備的蛍光画像から算出されたSum Intensityが所定の設定値を上回る場合、露光時間を短くする及び/又はカメラゲインを下げることにより、Sum Intensityを低下させることができる。Sum Intensityの設定値は15~85%の範囲内であれば任意の値をとることができ、好適には45~65%の範囲内に設定される。
 上記の予備的蛍光画像は、判定に用いる凝集反応生成物の蛍光を撮像する直前に撮像することができ、これによりそのSum Intensityの値を参照してSum Intensityが所定の設定値となるように露出をフィードバック制御することができる。あるいは、判定方法を開始する前に予備的蛍光画像を撮像し、Sum Intensityが所定の設定値となる露出条件を決定してから判定方法を開始してもよい。
 好ましい実施形態において、Sum Intensityが15~85%となる露出条件は、ISO感度200相当(ISO感度200相当のカメラの場合、ゲイン1倍)で150ms~1.8s、好適には300ms~1.6s露光する露出条件である。また、さらに好ましい実施形態において、Sum Intensityが45~65%となる露出条件は、ISO感度200相当で500~900ms露光する露出条件である。又はCCDカメラの自動露出機能を利用して、目標最大輝度を50%に設定してISO感度200相当で最大160msの露光時間での露光を行い、当該露光時間内に目標最大輝度が50%に達しない場合はこれに達するまでISO感度を6400相当(ISO感度200相当のカメラの場合、ゲイン32倍)まで段階的に上げる露出条件も、好適に用いられる。ここで目標最大輝度(target maximum light intensity)とは、撮像範囲内に含まれる画素のうち最も明るい画素の輝度値の、カメラ階調に対するパーセンテージである。例えば8ビット画像を撮像可能な256階調のカメラにおいて目標最大輝度を50%に設定した場合、撮像範囲内に含まれる画素のうち最も明るい画素の輝度値が128になるように撮像が行われる。この際、オーバーイルミネーションの許容量を適宜設定してもよい。
 次いで、撮像した蛍光画像内の関心領域に含まれる各画素の輝度値からSDを算出する。ここで用いる蛍光画像は、上で述べたように、カラーであってもモノクロであってもよく、カラー画像の場合は各画素のRGB値からグレースケール変換を行い、また8ビット画像であることが好ましい。
 ある実施形態のSD算出においては、各画素の輝度値に代えて以下の式3):
 補正後輝度値=輝度値×(目標Sum Intensity/前記式2によって算出されるSum Intensity)
によって算出される各画素の補正後輝度値を用いてSDを算出してもよい。この補正により、異なる露出条件で撮像された蛍光画像間でSum Intensityの値を揃えることができる。目標Sum Intensityは、45~65%の範囲から適宜設定することができる。
 この補正は、全ての蛍光画像に対して行ってもよいが、輝度値SDの値を大きくしてより高精度にアミロイドβタンパク質の凝集量を測定するという観点では、前記式2によって算出されるSum Intensityが目標Sum Intensityより小さい値であった蛍光画像に対して補正を行ってからSDを算出し、Sum Intensityが目標Sum Intensity以上の値であった蛍光画像は補正を行わずにそのままの輝度値を用いてSDを算出することが好ましい。
 アミロイドβタンパク質の凝集量の測定は、アミロイドβタンパク質の凝集体の蛍光画像から算出される輝度値SDがアミロイドβタンパク質の凝集量と正に相関することを利用して、輝度値SDの値を指標として行われる。
2-3.効果
 本発明の方法によれば、生体環境により近い培地や培養上清等において、従来不可能であった凝集性タンパク質を凝集させ、その凝集を可視化及び凝集量を測定できるようになる。これによって、培地や培養上清等でもMSHTS等の高い精度が期待できるスクリーニング技術の構築が可能となる。また、そのスクリーニング技術は、ADやパーキンソン病等のアミロイドーシスをはじめとした凝集性タンパク質に起因する様々な疾病の治療や予防に有用な薬の探索ツールとしての有効性が期待できる。
 また、本態様の方法によれば、第1の態様の方法で得られた回収液(濾液)中にて凝集性タンパク質の凝集体を可視化することができ、また、凝集性タンパク質の凝集量を測定することができる。特に、第1の態様の方法で得られた回収液(濾液)中の凝集性タンパク質は、生体条件に近い環境下にある凝集性タンパク質であり、本態様の方法によれば、生体条件に近い環境下にある凝集性タンパク質の凝集体を可視化することができる。
 従って、本態様の方法によれば、タンパク凝集促進可視化液を使用し、より生体内に近い状態(細胞分泌因子等)の溶液中で、様々な凝集性タンパク質に起因する表1に示した疾病に対する予防医薬品、機能性加工物の探索が可能になる。

3.凝集可視化剤
3-1.概念
 本発明の第3の態様は、凝集可視化剤である。本態様の凝集可視化剤は、生体に類似する環境下で凝集性タンパク質の凝集の場を提供し得るものであり、凝集阻害物質が除去された培地又は培養上清からなる。本発明の凝集可視化剤によれば、その溶液中の凝集性タンパク質及び/又はその凝集性断片を凝集させ、凝集体として可視化することができる。それ故に、凝集性タンパク質の凝集阻害効果を有する候補化合物を探索するスクリーニング系において、凝集性タンパク質含有溶液の溶媒又は溶剤として利用することができる。
3-2.構成
 本態様の凝集可視化剤は、凝集阻害物質が除去された培地又は培養上清からなる。本態様の凝集可視化剤は、液体状態又は固体状態(粉末状態、顆粒状態、粒状態を含む)のいずれであってもよい。固体状態の場合、使用時は、適当な溶液(水、又はバッファ等)に溶解する。
 本態様の凝集可視化剤の成分は、前記培地又は培養上清中に包含され得る様々な物質を含む。ただし、凝集阻害物質は予め除去されているため含まれない。
 本態様の凝集可視化剤は、凝集性タンパク質及び/又はその凝集性断片は含んでいてもよいし、含まなくてもよい。通常はそれらを含まず、必要に応じて、任意の凝集性タンパク質及び/又はその凝集性断片を本発明の凝集可視化剤と混合し、凝集体を形成させることによって、それらの凝集を可視化することができる。
3-3.調製方法
 本態様の凝集可視化剤は、前記凝集阻害物質が混在しないように培地又は培養上清を調製する方法、調製した培地又は培養上清から凝集阻害物質を除去する方法、又はそれらの組み合わせによって調製することができる。
 凝集阻害物質が混在しないように培地又は培養上清を調製する方法としては、凝集阻害物質を含まない構成成分、例えば、アルブミンフリー及び/又はグロブリンフリーの試薬を調合する方法が挙げられる。
 調製した培地又は培養上清から凝集阻害物質を除去する方法としては、第2態様の凝集工程における「(1)除去ステップ」に記載の方法に準じて行う方法が挙げられる。
 液体として調製された凝集可視化剤は、使用時まで液体状態のまま保存してもよいし、固体状態に変換して保存してもよい。液体状態で保存する場合、凝集可視化剤中に含まれるタンパク質等の各種成分の失活や分解を防ぐため、氷点下、好ましくは-20℃以下、-80℃以下、又は液体窒素中にて保存することが好ましい。
 液体として調製された凝集可視化剤を固体状態に変換する方法は、限定はしないが、例えば凍結乾燥法が挙げられる。凍結乾燥は公知技術であり、従来方法に従って行えばよい。固体状態として調製された凝集可視化剤は、10℃以下、4℃以下、又は0℃以下で保存することが好ましい。
3-4.効果
 本発明の凝集可視化剤は、例えば、生体環境により近い環境下でMSHTS等を実施し、疾患原因となる任意の凝集性タンパク質の凝集阻害効果を有する候補化合物を探索するスクリーニング系において、凝集性タンパク質の凝集を阻害せず、凝集を可視化できる凝集性タンパク質含有溶液の溶媒又は溶剤として提供することができる。
4.第1又は第2の態様の方法に使用するためのキット
4-1.概要
 本発明の第4の態様は、第1又は第2の態様の方法に使用するためのキットである。
4-2.構成
 本発明の第4の態様は、凝集性タンパク質を回収するためのキット(本明細書では、しばしば「回収キット」と略記する)、凝集性タンパク質の凝集量を測定するためのキット(本明細書では、しばしば「凝集量測定キット」と略記する)又は凝集性タンパク質の凝集体を可視化するためのキット(本明細書では、しばしば「凝集可視化キット」と略記する)である。本態様の凝集量測定キット又は凝集可視化キットは、凝集性タンパク質及び/又はその凝集性断片を培地又は培養上清等の生体環境により近い環境下で凝集させ、可視化するための構成要素を包含する。本発明の回収キットによれば、凝集性タンパク質を含む溶液から夾雑物を除去しつつ、凝集性タンパク質を効率的に回収することができ、本発明の凝集量測定キット又は凝集可視化キットによれば、凝集阻害物質が除去された培地又は培養上清で、凝集性タンパク質及び/又はその凝集性断片による凝集体を容易に検出することができる。
 本態様のキットは、必須の構成要素として濾過フィルターとタンパク凝集促進可視化液とを含み、また選択的構成要素として洗浄液、凝集性タンパク質の定量化手段(例えば、免疫学的測定方法に使用する抗凝集性タンパク質抗体等)、(第2の態様の方法に使用するためのキットにおいて)凝集性タンパク質の標識手段、標識された凝集性タンパク質(例えば、量子ドット修飾アミロイドβタンパク質)、プロトコルを包含する。プロトコルには、本態様のキットの使用方法が記載されている。
(1)凝集阻害物質の除去手段
 「凝集阻害物質の除去手段」は、凝集性タンパク質含有溶液の溶媒である培地又は培養上清等から、40kDa以上の凝集阻害物質、又はアクチン、アルブミン及びグロブリンからなる群から選択されるいずれか一以上のポリペプチド又はその断片を除去する手段である。本手段は、第2態様の凝集可視化方法における凝集工程の「(1)除去ステップ」に記載の方法を具現化する構成を有していればよい。例えば、40kDaでの分子量分画が可能な限外濾過フィルターを備えたスピンカラム、及び/又は凝集阻害物質を吸着する担体等が挙げられる。
(2)凝集性タンパク質の標識手段
 「凝集性タンパク質の標識手段」は、目的の凝集性タンパク質及び/又はその凝集性断片の凝集を検出し、可視化するために凝集性タンパク質及び/又はその凝集性断片を標識する手段である。本手段は、第1態様の凝集性タンパク質含有溶液の回収方法における用語の定義の「(11)標識」に記載の標識物質、及びその標識物質を修飾可能な試薬やデバイス等が挙げられる。例えば、標識物質が量子ドットのような光学的標識であれば、量子ドット及びそれをペプチドに付加し、ペプチドを修飾するための試薬等が挙げられる。具体的な構成については、使用する標識物質の種類に応じて、各種適切な修飾試薬やデバイスを備えればよい。
(3)標識された凝集性タンパク質
 ここでいう「標識された凝集性タンパク質」は、既に標識された凝集性タンパク質及び/又はその凝集性断片をいう。凝集阻害物質を除去した培地や培養上清を用いて凝集性タンパク質含有溶液を調製する際に、目的の凝集性タンパク質及び/又はその凝集性断片が定まっている場合、そのタンパク質等が標識された状態で本態様のキットに包含され得る。標識については、第1態様の凝集性タンパク質含有溶液の回収方法における用語の定義の「(11)標識」に記載に準ずる。
(4)その他の構成要素
 本態様の凝集可視化キットは、前記構成要素に加えて、その他の構成要素を選択的に含むことができる。その他の構成要素には、凝集性タンパク質の凝集を可視化する上で必要又は有用な様々なものを包含し得、特に限定はしない。具体的な構成要素については、例えば、凝集性タンパク質の種類等に応じて適宜定めればよい。一例として、凝集阻害物質を未除去若しくは除去済みの培地又は培養上清、標識二次抗体、標識の検出に必要な酵素基質、洗浄バッファ、試料希釈液、使用説明書等を含むことができる。
5.凝集性タンパク質による凝集体を形成することを特徴とする製造方法
5-1.概念
 本発明の第5の態様は、凝集性タンパク質による凝集体を形成することを特徴とする製造方法(本明細書では、しばしば「凝集体製造方法」と略記する)である。本態様の凝集体製造方法は、凝集阻害性物質が予め除かれた凝集性タンパク質含有溶液中で全部又は一部が標識された目的とする凝集性タンパク質及び/又はその凝集性断片をインキュベートして、その凝集体を形成することを特徴とする製造方法である。本態様の凝集体製造方法によれば、凝集性タンパク質及び/又はその凝集性断片からなる凝集体を製造し、入手することができる。得られた凝集体は、それを解離することのできる凝集解離効果を有する候補化合物を探索するスクリーニング系において利用することができる。
5-2.方法
 本態様の凝集体製造方法は、凝集工程及び確認工程を必須工程として、また回収工程を選択工程として含む。以下、それぞれの工程について説明をする。
5-2-1.凝集工程
 本態様における「凝集工程」は、基本的には第2態様の凝集可視化方法における凝集工程に準ずる。第2態様の凝集工程と同様に、凝集阻害性物質が除去された凝集性タンパク質含有溶液中で、目的とする凝集体を構成する凝集性タンパク質及び/又はその凝集性断片を所定の条件下でインキュベートして凝集体を形成させることを特徴とする。この時、凝集体形成に使用する凝集性タンパク質及び/又はその凝集性断片は、全部又は一部が標識されている。標識方法についても、第1態様の凝集性タンパク質含有溶液の回収方法における用語の定義の「(11)標識」に準じ、特に限定はしないが、標識物質による修飾が好ましい。標識物質としては、限定はしないが、光学的標識、特に量子ドットによる標識が好適である。
 凝集性タンパク質の種類も制限はされないが、タンパク質凝集を原因とする疾患に関連タンパク質、例えばADにおけるAβやタウタンパク質が好ましい。
5-2-2.確認工程
 「確認工程」は、凝集工程後に溶液中で凝集した前記凝集性タンパク質を検出し、凝集体の生成を確認する工程である。生産物である凝集体の形成を確認する工程であって、その確認のため凝集体を検出することから基本的には第2態様の凝集可視化方法における検出工程に準ずる。
5-2-3.回収工程
 「回収工程」は、前記確認工程で凝集体が確認された場合、その凝集体を回収する工程で、本態様の凝集体製造方法における選択工程である。
 凝集体の回収は、タンパク質の凝集体を回収し得る当該分野で公知の方法を用いることができる。例えば、濾過法、吸着法、遠心法、又はそれらの二以上の組み合わせが挙げられる。いずれも具体的な方法については、第2態様の凝集可視化方法における凝集工程の「(1)除去ステップ」に記載の方法に準ずるため、本工程での説明は省略する。
 回収した凝集体は、必要に応じて溶媒に再懸濁することができる。使用する溶媒として、例えば、生理食塩水、緩衝液、培地、又は培養上清等が挙げられる。好ましくは生体内の環境に近い培地又は培養上清である。この場合使用する培地又は培養上清は、凝集阻害性物質が除去された培地又は培養上清であることが好ましい。
 以下、実施例を用いて本発明をより詳細に説明するが、本発明の技術的範囲はこれら実施例に限定されるものではない。
1.材料及び方法
〔実施例1〕
 B27(Life Technologies社)をNeurobasal(フェノールレッド不含)培地(Life Technologies社)で50倍希釈したNb培地(NbM)に終濃度50pMアミロイドβ42タンパク質(Aβ42)溶液(DMSO)に調製した混合溶液(以下、模擬液)を準備した。濾過フィルターデバイス(Amicon Ultra - 0.5 Ultracel - 50メンブレン)を1.5mLチューブに取り付け、常温(15~25℃)のMilliQ水および模擬液を用いて、濾過フィルターにMilliQ水を400μL滴下した。1.5mLチューブを室温(15~25℃)で14,000×g 10分間遠心し、濾液を捨て、再度フィルターにMilliQ水400μlを滴下した。さらに、室温(15~25℃)で14,000×g 10分間遠心し、濾液を棄て室温(15~25℃)で14,000×gで2分間空遠心し、濾過フィルターデバイスを新たな1.5mLチューブに取り付け、模擬液500μLを濾過フィルターに滴下後、室温(15~25℃),14,000×g 30分間遠心し、濾液を回収した。前記、模擬液と濾液に含まれるAβ42濃度をヒトβアミロイドELISAキット(富士フィルム和光純薬)で測定した。模擬液中のAβ42濃度を100%として、濾液中のAβ42濃度をAβ42含有模擬液のAβ42濃度で割りパーセント換算し、Aβ42残存率として算出し53.8%であることが確認された(図1)。なお、精度評価の為、50pM、25pM、10pM、5pM、2.5pM、1pMの人工Aβ42含有NbMで標準曲線を作成し、ヒトAβELISAキット内の標準液とのリカバリー率を比較したところ、92.3%であることが確認できた。
〔実施例2〕
 以下、ブロッキング1~6を調製した。ブロッキング1は、5% BSA含有PBS溶液を示し、ブロッキング2は、5% BSA含有PBS溶液にtween-20を最終濃度0.1%に調製した溶液を示し、ブロッキング3は、5% BSA含有PBS溶液を示し、ブロッキング4は、5% BSA含有PBS溶液にtween-20を最終濃度0.1%に調製した溶液を示し、ブロッキング5は、0.4% BSA含有PBS溶液を示し、ブロッキング6は、0.4% BSA含有PBS溶液にtween-20を最終濃度0.1%に調製した溶液を示す。
 実施例1と同様、濾過フィルターデバイス(Amicon Ultra - 0.5 Ultracel - 50メンブレン)を1.5mLチューブに取り付け、MilliQ水を400μl濾過フィルターデバイスに滴下し、室温(15~25℃)で14,000×g、10分間遠心した。
 濾液を捨て、濾過フィルターにブロッキング1~6を500μL滴下し、インキュベートした。なお、インキュベート条件は、ブロッキング1, ブロッキング2, ブロッキング3が室温(15~25℃)で2時間静置、ブロッキング4、ブロッキング5、ブロッキング6は、冷蔵(4±2℃)で18時間静置した。ブロッキング操作後、1.5mLチューブを室温(15~25℃)、14,000×gで2分間遠心し、濾液を棄て室温(15~25℃)で14,000×gで10分間遠心した。濾液を捨て、フィルターにMilliQ水400μLを滴下し、室温(15~25℃),14,000×g 10分間遠心し、再度、濾液を捨て、フィルターにMilliQ水400μlを滴下し、室温,14,000×gで10分間遠心し、濾液を捨て、室温(15~25℃),14,000×gで2分間空遠心した。濾過フィルターデバイスを新たな1.5mLチューブに取り付け、模擬液500μLを濾過フィルターに滴下し、室温(15~25℃),14,000×gで30分間遠心し濾液を回収した。前述と同様、模擬液と濾液に含まれるAβ42濃度をヒトβアミロイドELISAキット(富士フィルム和光純薬)で測定した。各ブロッキング1~6条件で回収した濾液中のAβ42濃度からAβ42残存率を算出したところ、ブロッキング2の組成、及び条件が最もAβ42残存率が高い(93.8%)ことが分かった(図2)。
〔実施例3〕
 上述した実施例2でブロッキング2の条件が最もAβ42残存率が高かったことから、実際に細胞が分泌したアミロイドβタンパク質(Aβ)(Aβ42やアミロイドβ38タンパク質(Aβ38)等のAβを含む)含有培養上清を用いた場合も、同様にAβ回収率は、高く維持できるかを確認した。まず、分泌Aβ(細胞が分泌したAβ)含有培養上清を、以下の手順で調製した。
 マトリゲル(コーニング社)をDMEM(Life Technologies社)で25~50倍に希釈したコート溶液、又はSureBond-XF溶液(Axol社)をPBS(Mg-,Ca-)で200倍に希釈した溶液を6ウェルの培養プレートに1mL/ウェルずつ添加し、1~3時間インキュベートした。恒温槽で解凍した生存率70%以上のiPS細胞由来神経前駆細胞をNbMに5μM Y-27632(ナカライテスク社)を添加した培養液又は、Neural Maintenance Medium(Axol社)にNeural Maintenance Medium Supplement(Axol社)を添加したNMMs培地に5μM Y-27632を添加した培養液に懸濁し、コート溶液を除去した培養プレートに8~16×10^5cells/well(0.84~1.7×10^5/cm2)の密度で細胞を播種し、37℃、5%CO2条件で2日間培養した。その後、培養上清を吸引除去し、室温に戻した新鮮培養液を2mL/well加え、37℃、5%CO2条件で、さらに2日間培養した。解凍から5日目のiPS細胞由来神経前駆細胞をAccutage(Stem Cell technologies社)で3~5分処理後、ピペッティングによりシングルセルへと解離し、DMEM/F-12を5mL加え、遠心管に細胞を移し、遠心分離により上澄みを除去した。沈殿した細胞にNbMに5μM Y-27632を添加した培養液又はNMMs培地に10μM Y-27632を添加した培養液を加え、ピペッティングで細胞を分散させ、細胞懸濁液を調製した。コート溶液を除去後の培養プレートに8.0~16×10^5cells/well(0.84~1.7×10^5/cm2)の密度で細胞を播種し、37℃、5%CO2条件で2日間培養した。その後、培養上清を吸引除去し、室温に戻した新鮮NbMを2mL/well加え、37℃、5%CO2条件で2日間培養した。9日目にiPS細胞由来神経前駆細胞をAccutage(Stem Cell technologies社)で3~5分処理後、ピペッティングによりシングルセルへと解離し、DMEM/F-12を5mL加え、遠心管に細胞を移し、遠心分離により上澄みを除去した。沈殿した細胞にNbMに5μM Y-27632を添加した培養液又はNMMs培地に10μM Y-27632を添加した培養液を加え、ピペッティングにて分散させ、細胞懸濁液を調製し、8~16×10^5cells/well(0.84~1.7×10^5/cm2)の密度でコート溶液を除去後の培養プレートに播種し、37℃、5%CO2条件で培養した。翌日、培養プレート内の上清を除去後、終濃度100ng/mLのSonic Hedgehog(Shh)タンパク質を添加したNbM+Shh培地2mL/wellに交換し、37℃、5%CO2条件で3.5日間培養した。さらに、培養プレートから半量培養上清を回収し、NbM+Shh培地を2mL/well添加し、37℃、5%CO2条件で3.5日間培養した。培養プレートから回収した培養上清は、遠心管に回収し、300×gで3分間遠心後、底から1mLを残した状態で培養上清を新しい遠心管に移し、‐20℃で凍結保存した(培養上清Day7)。その後、3.5日間に1度の頻度で培地を半量回収後、新鮮NbMを2mL/well加え、37℃、5%CO2条件で培養した。
 Shh添加から14日以上経過した細胞の培養上清を回収した。これら回収した培養上清中に含まれるAβ濃度をヒトβアミロイドELISAキット(富士フィルム和光純薬)で測定したところ、43.6pMであった(図3)。本培養上清を使用し、実施例2で最もAβ42残存率が高いブロッキング2の条件において、実際の細胞が分泌したAβ含有培養上清でもAβ除去の軽減ができるかを確認した。また、比較対象として、実施例1と同様の手順でブロッキングなしでのAβ残存率を算出した。その結果、ブロッキング2の条件であれば、細胞が分泌したAβ含有培養上清でも高いAβ残存率(79.3%)が保持できることが分かった(図3)。
〔実施例4〕
 上述した実施例3から、ブロッキング2の条件であれば、実際の細胞が分泌したAβ含有培養上清から濾液を回収しても高いAβ残存率を示すことが確認できた。そこで、本濾液中でアミロイドβタンパク質の凝集が可視化できるか否かを確認した。量子ドット修飾アミロイドβ40タンパク質(QD-Aβ40)を実施例3で回収した濾液で溶解し、この溶液を1mM Aβ42溶液に加えて混合し、50μM Aβ42及び50nM QD-Aβ40を混合した、実施例3で回収した濾液を調製した。150mMロスマリン酸(RA)溶液を実施例3で回収した濾液で5段階の濃度(3000μM、600μM、120μM、24μM、4.8μM)に希釈した。Aβ42及びQD-Aβ40を実施例3で回収した濾液で希釈し、50μM Aβ42と50nM QD-Aβ40混合溶液を調整した。Aβ42及びQD-Aβ40混合溶液とロスマリン酸(RA)溶液を1:1比で混合し、RA終濃度が1500μM、300μM、60μM、12μM、2.4μMとなる反応溶液を準備した。なお、RAをDMSOに代替した溶液は陰性対照として準備した。これらの反応溶液を24時間、37℃でインキュベートし、蛍光顕微鏡で凝集Aβを観察したところ、いずれの溶液でも凝集Aβを定量解析できるほど、十分なSD値の感度を満たすには至らなかった(図4)。
〔実施例5〕
 上述した実施例4から、再度ブロッキング液の組成及び処理条件について検討し、新たに以下ブロッキング(Buffer)8~12を調製した。ブロッキング8は、0.4% BSA含有PBS溶液にTween-20を最終濃度0.1%に調製した溶液を示し、ブロッキング9は、0.4% BSA含有PBS溶液を示し、ブロッキング10は、0.4% B27含有PBS溶液にtween-20を最終濃度0.1%に調製した溶液を示し、ブロッキング11は、0.4% B27含有PBS溶液を示し、ブロッキング12は、tween-20をPBSで最終濃度0.1%に調製した溶液を示す。実施例2と同様、濾過フィルターデバイス(Amicon Ultra - 0.5 Ultracel - 50メンブレン)を1.5mLチューブに取り付け、400μl MilliQ水を濾過フィルターデバイスに滴下し、室温(15~25℃)で14,000×g、10分間遠心した。
 濾液を捨て、再度フィルターにMilliQ水400μlを滴下後、室温(15~25℃)で14,000×gで10分間遠心した。濾液を捨て、濾過フィルターに各ブロッキング(PBS,8~12)液を500μL滴下し、インキュベートする。インキュベート条件は、ブロッキングPBS,8~12では室温(15~25℃)で2時間静置した。ブロッキング操作後、遠心し、濾液を棄て室温(15~25℃)で14,000×gで10分間遠心した。濾液を捨て、フィルターにMilliQ水400μlを滴下し、室温(15~25℃),14,000×gで10分間遠心し、再度、濾液を捨て、フィルターにMilliQ水400μlを滴下し、室温,14,000×gで10分間遠心後、濾液を捨て、室温(15~25℃),14,000×gで2分間空遠心した。濾過フィルターデバイスを新たな1.5mLチューブに取り付け、50pMのAβ42含有模擬液500μLを濾過フィルターに滴下し、室温(15~25℃),14,000×gで30分間遠心し濾液を回収した。前述と同様、模擬液と濾液に含まれるAβ42濃度をヒトβアミロイドELISAキット(富士フィルム和光純薬)で測定した。模擬液中のAβ42濃度を100%として、各濾液中のAβ42濃度を算出したところ、ブロッキング12の組成、及び条件が最もAβ42残存率が高くなることが分かった(83.9%:図5)。また、各ブロッキング(PBS,8~12)操作により回収した濾液で50μM Aβ42溶液を調製し、さらに、11.1μM QD-Aβ40を回収した濾液で50nM QD-Aβ40溶液を調製した。これらAβ42溶液とQD-Aβ40溶液を1:1で混合し、24時間、37℃でインキュベートし、蛍光顕微鏡で凝集Aβを観察したところ、PBS, ブロッキング8, ブロッキング10, ブロッキング11, ブロッキング12では、凝集Aβが可視化できることが分かった(図5)。
 中でも、ブロッキング12の0.1% Tween-20溶液はタンパク凝集促進可視化液として、最も高感度(SD値)であることが確認できた。
 そこで、ブロッキング12を用いて、実施例2と同様操作で回収した濾液から50μM Aβ42溶液を調製し、さらに回収した濾液にQD-Aβ40を希釈し、50nM QD-Aβ40に調製することで溶液Bを準備した。この溶液Bを使用し、実施例4と同様操作で、RA終濃度が1500μM、300μM、60μM、12μM、2.4μMとなる反応溶液を準備した。なお、RAをDMSOに代替した反応溶液も陰性対照として準備した。これら反応液を24時間、37℃でインキュベートし、蛍光顕微鏡で凝集Aβを観察し、凝集Aβを定量化したところ、RA濃度が46.0±7.4μM以下であれば、全体の50%程度に相当するAβが凝集しなくなることが確認できた。従って、本タンパク凝集促進可視化液を使用することで濾液中でも、Aβ凝集抑制活性を算出できることが分かった。なお、凝集Aβの定量化には、少なくともSD値の感度が、PBSを100%とした場合と比較し、60%以上必要であること分かった。
2.結果及び考察
 凝集性タンパク質含有培地(人為的な添加、細胞/組織からの分泌を問わない)から夾雑物を除去すると、培地中の凝集性タンパク質も同時に除去されてしまう問題があった(図1)。
 そこで、様々なタンパク質の非特異吸着を軽減する非吸着溶液(BSA/PBS, BSA/PBS plus 0.1% Tween-20等)での処理を検討し、非吸着溶液/洗浄液の種類等を最適化することで、回収液中の凝集性タンパク質の除去軽減が可能となった(図2)。しかしながら、前記で最適化した方法で神経細胞が分泌するAβを含む培養上清から夾雑物を除去しても、同時に消失する分泌Aβ量を軽減できることが分かった(図3)。
 一方、前記回収液中にQD-Aβ40と人工Aβ42を共存させても、蛍光観察にて凝集Aβを可視化することができなかった(図4)。
 そこで、鋭意精査し、凝集性タンパク質を含む溶液から夾雑物を効率よく除去しつつ、回収液中でQD-Aβ40と人工Aβ42を共存させても蛍光観察により凝集Aβを可視化できる、タンパク凝集促進可視化液(Buffer12:0.1% Tween-20溶液)を見出した(図5)。
〔実施例6〕溶媒種による凝集体形成の確認
(目的)
 微量ハイスループットスクリーニング(MSHTS)において、各種溶媒使用時におけるアミロイドβタンパク質の凝集体の形成を確認する。
(方法)
 (1)アミロイドβタンパク質溶液の調製
 凝集性タンパク質にはアミロイドβタンパク質(Aβ)を用いた。以下の手順により、1mM Aβ溶液を調製した。5 mgのAβ(Human,1-42)(ペプチド研究所社)に5mLのHFIP(1,1,1,3,3,3-Hexafluoro-2-propanol)(富士フィルム和光純薬社)を添加し、懸濁した後、懸濁液を1時間室温静置した。その後、25℃、43 kHzで10分間、超音波処理してモノマー化した後、クリーンベンチ内に24時間放置してHFIPを揮発させた。揮発後、1071μLのDMSOで溶解し、得られた溶液を1mM Aβ溶液とした。1.5 mLチューブに256μLずつ分注し、使用まで-80℃にて保存した。
 なお、ここで調製したAβ溶液を、以下「Aβ42」と表記する。
 (2)量子ドット修飾化アミロイドβタンパク質(QDAβ)の調製
 以下の手順によりAβを量子ドット(QD)で修飾し、量子ドット修飾化アミロイドβタンパク質(QDAβ)を調製した。修飾方法は以下の手順で行った。
 125μLの8μM QdotTM605 ITKTM amino(PEG) Quantum Dotsを2本の1.5 mLチューブに入れ、10000×gで4℃にて1分間遠心した。それぞれの上清を遠心濃縮チューブ(VIVASPIN6)に移し、4500μLのPBSを加えた。2本の合計量がおよそ50μL以下になるまで、4℃、3800×gで遠心し、濾液は廃棄した。PBSを補充した後、再度50μLになるまで遠心した。得られたQD溶液を全量が約180μLくらいになるように1本にまとめた。10mM sulfo-EMCSを20μL追加し、1時間常温で静置してQD-EMCSを調製した。
 脱塩カラムを2本用意し、それぞれに樹脂を約800μL入れて、1000×gで4℃にて1分間遠心した。各カラムに300μLのPBSEを積載し、1000×gで4℃にて1分間遠心を行う工程を2セット行い、脱塩カラムを調製した。
 QD-EMCS調製後に、液中に含まれる未反応N-ヒドロキシスクシンイシド基を不活性化させるために100mM K-グルタミン酸を20μL加えて、常温で10分間静置した。
 110μLの前記QD-EMCSを2本の脱塩カラムの中心に染み込ませた後、スタッカーとしてPBSEを15μL加えた。1000×gで4℃にて2分間遠心した後、2本の脱塩カラムの濾液(脱塩QD-EMCS)をまとめた。得られた脱塩QD-EMCSに1.0 mM Cys-Aβを20μL/DMSOを加え、混合後、1時間常温で静置した。
 QD-EMCS調製後に、液中に含まれる未反応マレイミド基を不活性化させるために20μLの100mMの2-メルカプトエタノールを加えて、常温で10分間静置した。
 サンプル溶液を2本のVIVASPIN6へ145μLずつ移し、水を4500μL加えた後、3800×gで4℃にて17分間遠心した。濾液は廃棄し、得られた溶液の全量が約140μLになるようにして1本にまとめた。
 脱塩カラムに水を300μL入れて1000×gで4℃にて1分間遠心を行い、この工程を2回繰り返した。サンプル溶液を70μLずつ脱塩カラムに染み込ませて、スタッカーの水を15μL加えた。1000×gで4℃にて2分間遠心し、目的のQDAβを得た。完成したQDAβと未修飾のQD(QdotTM605 ITKTMamino(PEG) Quantum Dots)をそれぞれ、QD/QDAβ:超純水=0.4μL:2.8μLで8倍希釈し、nanodropを用いて濃度を測定した。
 (3)評価溶液の調製
 凝集体の形成を確認する以下の評価溶液を、溶媒毎に調製した。
 ・BSA/PBS溶液
   BSA(Wako)をPBSに溶解し、50mg/mLに調製した溶液である。
 ・NBM溶液
   Neurobasal培地にB-27(登録商標)(Gibco社)を添加し、0.5% v/vに調製した溶液である。
 ・DFBM溶液
   DMEM/F12培地(Thermo Fisher Scientific社)にB-27(登録商標)(Gibco社)を添加し、0.5% v/vに調製した溶液である。
 (4)凝集体の形成
 最終濃度の2倍量に相当する50nM QDAβ及び50μM Aβ42を含むAβ溶液を調製した。各評価溶液3μLにAβ溶液を3μLずつ添加してサンプル溶液を調製した。各サンプル溶液を10000×gで2分間遠心した後、ウェルプレートに5μLずつ分注した。続いて、プレート遠心機で1530×gで5分間遠心した。凝集前をO時間として、凝集の有無を確認した後、37℃で24時間インキュベートした。24時間後、各サンプル溶液における凝集体の有無を倒立顕微鏡(ニコン社製TE2000/オリンパス社製DP72)を用いて画像を撮影した。
(結果)
 図6に結果を示す。従来法で用いるPBSを溶媒としたサンプル溶液では、bに示すようにインキュベート24時間後にAβの凝集を確認することができた。一方、PBSにBSAを添加したサンプル溶液(c)や、NBM溶液(d)又はDFBM溶液(e)を溶媒としたサンプル培地では24時間後も凝集は確認できなかった。この結果から、培地中には、Aβの凝集を阻害する凝集阻害物質の存在が推定された。
〔実施例7〕凝集阻害物質の除去によるAβの凝集体形成の確認(1)
(目的)
 実施例6より、培地中にはAβの凝集を阻害する物質の存在が推定された為、前記サンプル液中のAβ凝集阻害物質を除去することで、凝集Aβが可視化できるかを確認した。
(方法)
 分画サイズが異なる4種の限外濾過フィルター(アミコンウルトラ;3kDa:UFC500324、10kDa:UFC501024、50kDa:UFC505096、及び100kDa:UFC510024;Merck社)をキット付属の1.5mLチューブに取り付けた。500μLのNBM溶液を上記各限外濾過フィルターに滴下し、室温(20~25℃) 14,000×gで10分間、遠心した。濾液を回収し、50nM QD-Aβ溶液(量子ドット修飾アミロイドβ)と50μM Aβとの混合溶液を1対1で混合させ、24時間、37℃でインキュベート後、倒立顕微鏡を用いて凝集Aβの様子を観察した。
(結果)
 図7に結果を示す。観察の結果、100kDaの限外濾過で回収した濾液中では、Aβの凝集が不均一に起こり、Aβ凝集の十分な定量が認められずに、EC50算出ができないことが明らかとなった。その一方で、3kDa、10kDa、及び50kDaの限外濾過フィルターで回収した濾液中では、Aβの凝集が均一に起こることが分かった。この結果から、Aβの凝集阻害物質は、50kDaの限外濾過フィルターを通過できない分子量の物質、すなわち50kDaの限外濾過フィルターで除去される物質が含まれることが明らかとなった。
〔実施例8〕凝集阻害物質の評価
(目的)
 実施例7より、Aβの凝集阻害物質は50kDaの限外濾過フィルターで除去されることが明らかとなった。前述のように公称分画分子量(NMWL)が50kDaであっても、実際には50kDa以下の分子量の物質も除去され得る。そこで、NBM溶液及びDFBM溶液内で除去された成分を確認するために、SDS-PAGEによる分子量分画とCBBによる染色を行った。
(方法)
 (1)Aβ凝集阻害物質除去溶液の調製
 限外濾過フィルター(アミコンウルトラ;50kDa:UFC505096;Merck社)をキット付属の1.5mLチューブに取り付けた。500μLのNBM溶液、及びDFBM溶液を上記外濾過フィルターに滴下し、室温(20~25℃) 14,000×gで10分間遠心した。濾液を回収し、それぞれをAβの凝集阻害物質が除去されたNBM溶液(除去)、及びDFBM溶液(除去)として調製した。
 (2)SDS-PAGEゲルの調製
 30% アクリルアミドストック溶液(29% アクリルアミド、1% N,N’-メチレンビスアクリルアミド)、1.5 M Tris-HCl(pH 8.8)溶液、10% SDS(ドデシル硫酸ナトリウム)溶液、10% APS(過硫酸アンモニウム)溶液、TEMED(N,N,N′,N′-テトラメチルエチレンジアミン)を用いて調製した。10%アクリルアミドランニングゲル溶液(最終濃度:9.7% アクリルアミド、0.3% N,N’-メチレンビスアクリルアミド、0.38 M Tris-HCl (pH 8.8)、0.1% SDS、0.033% APS、0.05% TEMED)を2枚のガラス板の間に注入し、続けて水を注入し、1時間静置して10%アクリルアミドランニングゲルを作製した。上部の水を除去後、30% アクリルアミドストック溶液、0.5 M Tris-HCl(pH6.8)溶液、10% SDS溶液、10% APS溶液、TEMEDを用いて調製したスタッキングゲル溶液(最終濃度:4.6% アクリルアミド、0.15% N,N’-メチレンビスアクリルアミド、0.125 M Tris-HCl (pH 6.8)、0.1% SDS、0.033% APS、0.05% TEMED)を注入し、コームを差込み、1時間静置し、10% アクリルアミドゲルを調製した。
 (3)泳動用サンプルの調製
 25μLのNBM溶液、NBM溶液(除去)、DFBM溶媒、DFBM溶液(除去)、及び50mg/ml BSA/PBSを、それぞれ25μLの2×サンプルバッファー(100 mM Tris-HCl(pH 6.8)、4% SDS、12% 2メルカプトエタノール、20% グリセロール、0.01% ブロモフェノールブルー)と混合し、95℃に熱したヒートブロック(FG-01N,Fast Gene)上で、3分間加熱し、泳動用サンプル溶液を調製した。
 (4)SDS-PAGE
 NBM溶液、NBM溶液(除去)、DFBM溶媒、及びDFBM溶液(除去)の各泳動用サンプル溶液は10μLずつ、50mg/ml BSA/PBSの泳動用サンプル溶液は1μLを、それぞれ(2)で調整した10%アクリルアミドゲルのレーンに注入し、電源装置(BP-9, BIO CRAFT)を用いて、20 mA, 75 分間の条件で電気泳動を行った。分子量マーカーには10μLのCLEARLY Stained Protein Ladder(Code No. 3454A, Takara Bio)を用いた。
 泳動後、ゲルを回収し、CBB溶液(nacalai tesque)で20分間浸漬した後、7.5%酢酸で30分間の洗浄を行い、電子レンジで3回ほど加熱して脱染色したゲルを撮影した。
(結果)
 図8に結果を示す。観察の結果、NBM溶液(除去)(レーン3)及びDFBM溶液(除去)(レーン5)には、NBM溶液(レーン2)及びDFBM溶液(レーン4)に見られる矢印で示した42kDa付近、50kD付近と150kDa付近のタンパク質が除去されていることが明らかとなった。ウシ血清アルブミン(BSA)(レーン6)で50kDa付近に主要バンドが存在することや、図6cの結果を鑑み、Aβ凝集阻害物質の成分は分子量が約50kDaのアルブミンであることが推察された。
 また、NBM溶液やDFBM溶液は、グロブリンやアクチンが多く含まれるところ、グロブリンの分子量は約150kDaであり、またアクチンの分子量は42kDaである。図8のSDS-PAGEにおいて約150kDa付近で認められるバンドはグロブリンであり、また約42kDa付近で認められるバンドはアクチンであると考えられる。したがって、グロブリン及びアクチンもAβ凝集阻害物質の成分であると推察された。以上の結果から、40kDa以上250kDa以下、40kDa以上240kDa以下、40kDa以上230kDa以下、40kDa以上220kDa以下、又は40kDa以上210kDa以下の分子量を有する物質を除去することで凝集阻害物質を除去できることが明らかになった。
〔実施例9〕凝集阻害物質の除去によるAβの凝集体形成の確認(2)
(方法)
 実施例7より、各フィルターで通液後に回収した濾液中で、形成した凝集Aβの画像を画像解析ソフトImage Jを使用し、自動で凝集体の形状を検出(Threshold機能の利用を含む)した。その後、単独の粒子と判断できた凝集体を各条件に無作為に10個ずつ選び、凝集体の面積、周囲長、フェレー径を算出した。Image Jの解析方法は、撮影画像をImage Jに取り込んだ後、Imageタブ内のAdjust、Color Thresholdモードで凝集体の形状を検出した。この凝集体の形状をWand toolを使用し、単独の粒子を無作為に10個ずつ選び、Analyzeタブ内のTools、ROI managerへ加えた。ROI manager内のMeasureタブを押し、各粒子の「Area(面積)」、「Perimeter(周囲長)」、「Feret's Diameter(フェレー径)」を算出し、この値を平均化してグラフ化した。なお、フェレー径とは、選択範囲の外周上にある任意の2点を結ぶ直線のうち、最も長い距離をいう。
(結果)
 結果を図9に示す。A(面積)、B(周囲長)、及びC(フェレー径)のいずれも100kDaの限外濾過フィルターで通液し回収した濾液のみが、他の限外濾過フィルターの濾液と比べてAβ凝集体が、巨大、かつ不均一であることが明らかとなった。したがって、100kDaの限外濾過フィルターで通液し、回収した濾液は、EC50算出には使用できないことが判明した。
 これらの結果を利用したスクリーニング技術のコンセプトを図10に示す。疾患モデル細胞を培養した培養上清を回収し、当該培養上清中の凝集性タンパク質を本発明の方法により凝集、可視化する。前記可視化の際に様々な化合物や抽出エキスなどの凝集抑制活性を示す候補物質を所定濃度で添加することでEC50が算出できる為、実際に凝集抑制活性を示す候補物質を選択することができる。本スクリーニング技術は、自動化することが可能である為、ハイスループットで実施できる。

Claims (24)

  1.  濾過フィルターをタンパク凝集促進可視化液で通液する第1工程と、
     タンパク凝集促進可視化液で通液した濾過フィルターに、凝集性タンパク質含有溶液を通液する第2工程と、
    を含む、凝集性タンパク質含有溶液の回収方法。
  2.  第1工程前に、濾過フィルターを洗浄液で通液する前処理工程をさらに含む、請求項1記載の方法。
  3.  第2工程前に、凝集性タンパク質含有溶液中の凝集性タンパク質を定量する工程をさらに含む、請求項1又は2記載の方法。
  4.  第2工程後、濾液中の凝集性タンパク質を定量する工程をさらに含む、請求項1~3のいずれか1項記載の方法。
  5.  濾過フィルターが、50kDa以下の公称分画分子量(NMWL)を有する限外濾過フィルターである、請求項1~4のいずれか1項記載の方法。
  6.  タンパク凝集促進可視化液が、50kDa以下の界面活性作用のある物質を含む、請求項1~5のいずれか1項記載の方法。
  7.  タンパク凝集促進可視化液が、Tween-20を含む水溶液である、請求項1~6のいずれか1項記載の方法。
  8.  水溶液中におけるTween-20の濃度が0.1~10%である、請求項7記載の方法。
  9.  凝集性タンパク質含有溶液が緩衝液、培地、又は培養上清を含む、請求項1~8のいずれか1項記載の方法。
  10.  凝集性タンパク質含有溶液が、細胞代謝物質、細胞分泌因子、無機物、又は有機酸を含む、請求項1~9のいずれか1項記載の方法。
  11.  前記凝集性タンパク質含有溶液が、疾患に関連するタンパク質を含む、請求項1~10のいずれか1項記載の方法。
  12.  前記疾患がアルツハイマー病であり、それに関連するタンパク質がアミロイドβタンパク質又はタウタンパク質である、請求項11記載の方法。
  13.  下記1)式で求められる凝集性タンパク質の回収率が、60%を超える、請求項1~12のいずれか1項記載の方法。
     凝集性タンパク質の回収率(%)=第2工程後の凝集性タンパク質の定量値/第2工程前の凝集性タンパク質の定量値×100  ‐‐‐‐‐式1)
  14.  請求項1~13のいずれか1項記載の方法により回収した凝集性タンパク質含有溶液において、該凝集性タンパク質の凝集量を測定することを含む、凝集性タンパク質の凝集量の測定方法。
  15.  請求項1~13のいずれか1項記載の方法により回収した凝集性タンパク質含有溶液において、該凝集性タンパク質の凝集体を可視化することを含む、凝集性タンパク質の凝集体の可視化方法。
  16.  凝集性タンパク質の凝集体の可視化は、
     全部又は一部が標識された凝集性タンパク質及び/又はその凝集性断片を前記回収した凝集性タンパク質含有溶液中でインキュベートし、前記凝集性タンパク質及び/又はその凝集性断片を凝集させる凝集工程;及び、
     前記凝集性タンパク質及び/又はその凝集性断片の凝集体を検出する検出工程を含み、
     前記回収した凝集性タンパク質含有溶液は、前記凝集性タンパク質及び/又はその凝集性断片の凝集を阻害する、40kDa以上の凝集阻害物質が除去されていることを特徴とする、
    請求項15記載の方法。
  17.  前記40kDa以上の凝集阻害物質の分子量が250kDa以下である、請求項16記載の方法。
  18.  前記40kDa以上の凝集阻害物質がアクチン、アルブミン及びグロブリンから成る群から選択されるいずれか一以上のポリペプチド又はその断片である、請求項16又は17記載の方法。
  19.  前記標識が光学的標識である、請求項16~18のいずれか1項記載の方法。
  20.  前記光学的標識が量子ドットである、請求項19記載の方法。
  21.  濾過フィルターとタンパク凝集促進可視化液とを含む、請求項1~13のいずれか1項記載の方法により凝集性タンパク質含有溶液から凝集性タンパク質を回収するためのキット。
  22.  濾過フィルターとタンパク凝集促進可視化液とを含む、請求項14記載の方法により凝集性タンパク質含有溶液において該凝集性タンパク質の凝集量を測定するためのキット。
  23.  濾過フィルターとタンパク凝集促進可視化液とを含む、請求項15~20のいずれか1項記載の方法により凝集性タンパク質含有溶液において該凝集性タンパク質の凝集体を可視化するためのキット。
  24.  凝集性タンパク質の標識手段若しくは標識された凝集性タンパク質をさらに含む、請求項23記載のキット。
