Abstract
植物由来の抽出物からなり、凝集性タンパク質に対して凝集抑制効果を有する新規凝集抑制剤を開発し、提供する。 キク科、バラ亜科、ユキノシタ科、セリ科、ユリ科、キキョウ科、ツツジ科、シロネ属、フウロソウ属、オオバコ属、オトギリソウ属、ハコベ属、クサノオウ属、フッキソウ属、及びクサソテツ属に属する植物からなる群から選択される植物の抽出物からなる凝集抑制剤を提供する。
Description
本発明は、凝集性タンパク質の凝集抑制剤に関する。
アルツハイマー病(Alzheimer's disease:本明細書では、しばしば「AD」と略称する)は、不可逆的な進行性中枢神経疾患の一種であり、認知機能障害(認知症)、行動障害、又は人格変化等の症状を伴う。全世界における認知症の患者数は、2019年時点で5,000万人以上と推定されているが、約70%がAD患者と考えられており、その発症率は増加傾向にある。AD患者の増加に伴う医療費の増大や介護問題は、国や患者関係者達にとって経済的又は精神的負担になる等、近年大きな社会的問題となっている。
ADは、患者脳内で疎水性のペプチドであるアミロイドβタンパク質(本明細書では、しばしば「Aβ」と表記する)の凝集及び蓄積に始まり、その後、微小管結合タンパク質であるタウタンパク質が過リン酸化されて線維化した後に、神経細胞が破壊され、脳が萎縮することによって発症する(非特許文献1~3)。
このADの発症機序に基づき、in vitroで添加した被験物質のAβ凝集阻害効果を評価し、AD治療の候補化合物をスクリーニングする技術が開発されている。例えば、量子ドットナノプローブを用いたアミロイドβタンパク質凝集阻害物質の微量ハイスループットスクリーニング(Microliter-scale High-throughput Screening:本明細書では、「MSHTS」と表記する)法は、PBS溶媒中でアミロイドβタンパク質の凝集阻害効果を有する候補化合物を探索できるCell-free Assays (無細胞試験)系を基盤技術としたスクリーニング技術である(非特許文献4)。
MSHTSの例として、例えば、特許文献1には、アミロイド形成を評価する方法、装置及びプログラムが開示されており、具体的には、水又はPBS等の適当な緩衝液中で、被験物質の存在下又は非存在下で、アミロイドβタンパク質等のアミロイド形成性タンパク質と、当該アミロイド形成性タンパク質の重合により形成されるアミロイドへの結合能を有する蛍光プローブ(量子ドットを蛍光色素として含む、又は量子ドット等)とを反応させる凝集反応ステップと、凝集反応ステップにおいて得た凝集反応生成物の蛍光を撮像する撮像ステップと、撮像ステップにおいて撮像した蛍光画像内の関心領域に含まれる各画素の輝度値から標準偏差を算出する標準偏差算出ステップと、標準偏差算出ステップにおいて算出した被験物質存在下での輝度値標準偏差の値と被験物質非存在下での輝度値標準偏差の値との比較から、被験物質存在下での輝度値標準偏差の値が被験物質非存在下での輝度値標準偏差の値より小さい場合に、被験物質がアミロイド形成阻害活性を有すると判定する活性判定ステップとを含む、被験物質のアミロイド形成阻害活性を判定する方法が開示されている。
また、特許文献2には、タンパク質やペプチドのアミロイド凝集性を評価するための汎用性量子ドットナノプローブ、及び当該量子ドットナノプローブを用いたアミロイド形成阻害剤の評価方法が開示され、具体的には、アミロイド形成ペプチドのN末端又はC末端にシステインを介して量子ドットが結合した量子ドットナノプローブが開示されている。
しかしながら、Cell-free Assays系で得られた候補化合物はCell-Based Assays(細胞を使用した試験)系では、その阻害効果が認められない場合が多い。これはCell-free Assays系が生体環境を必ずしも反映していないことが原因と考えられる。

本発明は、上述の実情に鑑み、Cell-Based Assays(細胞を使用した試験)系において、Aβ等の凝集性タンパク質に対する凝集抑制効果又は凝集促進効果を有する候補化合物を探索できるスクリーニング方法を開発し、その方法に基づいて凝集性タンパク質に対する新規の凝集抑制剤を単離し、提供することを目的とする。
上記課題を解決するために、本発明者らはCell-Based Assays(細胞を使用した試験)系として、標識された凝集性タンパク質及び被験物質の存在下で細胞を培養し、当該細胞培養物において、細胞表面上や細胞中の凝集した及び/又は沈着した凝集性タンパク質を、標識を指標として定量することで、被験物質が凝集性タンパク質に対する凝集抑制活性又は凝集促進活性を有するか否かを評価する方法を開発した。
当該評価方法を用いて、天然物由来の様々な被験物質について凝集抑制活性又は凝集促進活性を検証した結果、特定の種類の植物抽出物に凝集性タンパク質に対する強い凝集抑制作用があることを見出した。本発明は、前記研究結果として得られた新規知見に基づくものであって、以下を提供する。
(1)キク科(Asteraceae)、バラ亜科(Rosoideae)、ユキノシタ科(Saxifragaceae)、セリ科(Apiaceae)、ユリ科(Liliaceae)、キキョウ科(Campanulaceae)、ツツジ科(Ericaceae)、シロネ属(Lycopus)、フウロソウ属(Geranium)、オオバコ属(Plantago)、オトギリソウ属(Hipericum)、ハコベ属(Stellaria)、クサノオウ属(Chelidonium)、フッキソウ属(Pachysandra)、及びクサソテツ属(Matteuccia)に属する植物からなる群から選択される植物の抽出物からなる凝集性タンパク質の凝集抑制剤。
(2)前記キク科に属する植物がコウモリソウ属(Parasenecio)、アザミ属(Cirsium)、ヨモギ属(Artemisia)、又はノブキ属(Adenocaulon)に属する植物である、(1)に記載の凝集抑制剤。

本発明の凝集抑制剤によれば、植物由来の抽出物からなり、凝集性タンパク質に対して凝集抑制効果を有する新規凝集抑制剤を提供することができる。
1.凝集抑制剤
1-1.概要
本発明の第1の態様は、凝集性タンパク質の凝集抑制剤である。本発明の凝集抑制剤は、本発明の出願人らが開発した被験物質の凝集性タンパク質に対する凝集抑制活性又は凝集促進活性評価方法によって見出された特定の種類の植物抽出物からなる。本発明の凝集抑制剤によれば、凝集性タンパク質と混合することにより、凝集性タンパク質による凝集体の形成を抑制することができる。
1-1.概要
本発明の第1の態様は、凝集性タンパク質の凝集抑制剤である。本発明の凝集抑制剤は、本発明の出願人らが開発した被験物質の凝集性タンパク質に対する凝集抑制活性又は凝集促進活性評価方法によって見出された特定の種類の植物抽出物からなる。本発明の凝集抑制剤によれば、凝集性タンパク質と混合することにより、凝集性タンパク質による凝集体の形成を抑制することができる。

疾患に関連する凝集性タンパク質には、例えば、アルツハイマー病の原因タンパク質であるアミロイドβタンパク質及びタウタンパク質(リン酸化タウタンパク質を含む)、パーキンソン病の原因タンパク質であるα-シヌクレインタンパク質、伝達性海綿脳症(クロイツフェルトヤコブ病、狂牛病、又はプリオン病を含む)の原因タンパク質であるプリオンタンパク質、ハンチントン病の原因タンパク質であるハンチンチンタンパク質、II型糖尿病の原因タンパク質であるアミリンタンパク質、動脈硬化症(脳梗塞、肺梗塞、心筋梗塞を含む)の原因タンパク質であるアポリポタンパク質A1(APOA1タンパク質)、間接リウマチの原因タンパク質である血清アミロイドAタンパク質、全身性ALアミロイドーシスの原因タンパク質である免疫グロブリン軽鎖、透析アミロイドーシスの原因タンパク質であるβ2ミクログロブリン、又は筋萎縮性側索硬化症の原因タンパク質とされるTDP-43タンパク質が挙げられる。限定はしないが、アミロイドβ40タンパク質、アミロイドβ42タンパク質、アミロイドβ43タンパク質、アミロイドβ38タンパク質等のアミロイドβタンパク質は、本明細書の凝集性タンパク質として特に好適である。
オートファジー関連タンパク質には、例えば、ユビキチン様タンパク質であるAtg-8、Atg-12が挙げられる。
凝集性タンパク質は、自然界に存在する天然タンパク質であってもよいし、天然タンパク質に人工的に変異や修飾を導入した改変タンパク質であってもよいし、又は人工的に設計されたアミノ酸配列に基づく人工タンパク質であってもよい。
(2)凝集体
本明細書において「凝集体」とは、二以上の凝集性タンパク質の集合体をいう。本明細書では、いわゆるタンパク質複合体も凝集体に包含される。
凝集体は、同一種類のタンパク質から成るホモ凝集体であってもよいし、異なる種類のタンパク質から成るヘテロ凝集体であってもよい。
本明細書において「凝集体」とは、二以上の凝集性タンパク質の集合体をいう。本明細書では、いわゆるタンパク質複合体も凝集体に包含される。
凝集体は、同一種類のタンパク質から成るホモ凝集体であってもよいし、異なる種類のタンパク質から成るヘテロ凝集体であってもよい。
(3)沈着
本明細書において「沈着」とは、細胞表面上及び/又は細胞中に凝集性タンパク質が固着及び/又は蓄積することをいう。

(4)治療及び予防
本明細書において「治療」とは、疾患を治癒すること、疾患の症状の進行若しくは慢性化を抑制又は防止すること、及び疾患の症状を軽減、軽快、寛解することをいう。
また本明細書において「予防」とは、特定の疾患の発症を抑制、又は阻止することをいう。

(5)植物抽出物
本明細書において「植物(の)抽出物」とは、植物体から得られる複数種の成分からなる混合物をいう。通常は、植物体から溶媒中に抽出された低分子化合物等の植物成分、又は植物体の搾汁液中に包含されるタンパク質、核酸、脂質、糖、及び低分子化合物等を含む植物成分が該当する。植物抽出物に包含される各成分の全部又は一部は未同定成分であってもよい。植物抽出物の状態は問わない。液体状態であってもよいし、固体状態(粉末状態、顆粒状態を含む)であってもよい。
本明細書において「搾汁液」とは、植物体を圧搾して得られる液体成分をいう。植物体の小片が固体成分として含まれていてもよいし、濾過して液体成分のみにしたものであってもよい。
本明細書において「植物(の)抽出物」とは、植物体から得られる複数種の成分からなる混合物をいう。通常は、植物体から溶媒中に抽出された低分子化合物等の植物成分、又は植物体の搾汁液中に包含されるタンパク質、核酸、脂質、糖、及び低分子化合物等を含む植物成分が該当する。植物抽出物に包含される各成分の全部又は一部は未同定成分であってもよい。植物抽出物の状態は問わない。液体状態であってもよいし、固体状態(粉末状態、顆粒状態を含む)であってもよい。

(6)植物体

1-3.構成
(1)植物抽出物
本発明の凝集性タンパク質の凝集抑制剤は、植物の抽出物からなる。本発明の植物抽出物は、植物体から得られた抽出物であれば、その抽出方法や状態は問わない。例えば、植物体を水、又は有機溶媒(エタノール等)等の溶媒中に所定の温度下で、所定の時間浸漬した後、植物体を遠心や濾過によって除去することで得られる液体成分(いわゆる抽出液)、前記抽出液から溶媒を減圧、蒸発・乾燥等によって除去した後に残存する固形物(顆粒、粉末を含む)、又はその中間状態のペーストが挙げられる。また、植物体を圧搾して得られる搾汁液、それを濾過して固体成分を除去したもの、又はそれらの水分を蒸発・乾燥等によって得られる固形物が挙げられる。
(1)植物抽出物
本発明の凝集性タンパク質の凝集抑制剤は、植物の抽出物からなる。本発明の植物抽出物は、植物体から得られた抽出物であれば、その抽出方法や状態は問わない。例えば、植物体を水、又は有機溶媒(エタノール等)等の溶媒中に所定の温度下で、所定の時間浸漬した後、植物体を遠心や濾過によって除去することで得られる液体成分(いわゆる抽出液)、前記抽出液から溶媒を減圧、蒸発・乾燥等によって除去した後に残存する固形物(顆粒、粉末を含む)、又はその中間状態のペーストが挙げられる。また、植物体を圧搾して得られる搾汁液、それを濾過して固体成分を除去したもの、又はそれらの水分を蒸発・乾燥等によって得られる固形物が挙げられる。
溶媒を用いた抽出方法は、公知の抽出方法で良い。抽出時の溶媒温度は、溶媒が水の時は、1気圧下であれば10~100℃、好ましくは40~99℃、より好ましくは70~99℃の範囲の温度下で、またオートクレーブ等の耐熱耐圧密閉器を用いた1~2気圧下であれば100~121℃の範囲であればよい。また溶媒がエタノールの時は、1気圧下であれば0~78.3℃、好ましくは15~77℃、より好ましくは20~77℃の範囲であればよい。さらに、溶媒が水とエタノールの混合液の時は、エタノールの濃度に応じて、上記水の場合又はエタノールの場合の条件を適宜応用すればよい。抽出時間は、溶媒の温度によって異なる。一般的には、溶媒温度が40℃未満の低温であれば、例えば、数日~1年以上の長時間を要し、また1気圧下40℃以上沸点未満の高温であれば、例えば、2分~数時間程度の短時間でよい。さらに、溶媒が水の時、1~2気圧下100~121℃の温度であれば、2分~30分、好ましくは10~20分程度の時間でよい。
なお、植物抽出物を抽出又は搾汁等によって調製する際、使用する植物体は、植物体そのままの状態であってもよいし、植物体を断片化、又は粉末化したものであってもよい。「植物体そのままの状態」とは、全草使用の場合、全草の状態、花、葉、根部等の一部を使用の場合、その一部器官のままの状態をいう。一方、「断片化」とは、植物体を切裁、刷りおろし等によって小片化したものをいう。断片のサイズは特に限定はしないが、溶媒中への抽出効率や搾汁効率を向上するには小さいサイズにすることが好ましい。例えば、1~5mmの範囲であればよい。「粉末化」は、植物体をそのままの状態で、又は凍結、乾燥若しくは凍結乾燥した後、粉砕して粉状化したものをいう。粉末を構成する粒子のサイズも1mm以下であればよく、特に限定はしない。例えば、顆粒サイズ、微粉サイズ、又はそれらの混合物であってもよい。
溶媒を用いた抽出法は、搾汁液と異なり、植物体の細胞片等の夾雑物の混入率が低く、抽出物中の実質的な有効成分の濃度も高いため少量で薬効を得ることができることから特に好ましい。
(2)由来植物
本発明の凝集性タンパク質の凝集抑制剤を構成する植物抽出物の由来植物については、その種類がキク科(Asteraceae)に属する種、ユキノシタ科(Saxifragaceae)に属する種、バラ亜科(Rosoideae)に属する種、セリ科(Apiaceae)に属する種、ユリ科(Liliaceae)に属する種、キキョウ科(Campanulaceae)に属する種、ツツジ科(Ericaceae)に属する種、シロネ属(Lycopus)に属する種、フウロソウ属(Geranium)に属する種、オオバコ属(Plantago)に属する種、オトギリソウ属(Hipericum)に属する種、ハコベ属(Stellaria)に属する種、クサノオウ属(Chelidonium)の属する種、フッキソウ属(Pachysandra)に属する種、及びクサソテツ属(Matteuccia)に属する種から選択されるいずれかの植物である。以下、各植物について具体的に説明をする。
本発明の凝集性タンパク質の凝集抑制剤を構成する植物抽出物の由来植物については、その種類がキク科(Asteraceae)に属する種、ユキノシタ科(Saxifragaceae)に属する種、バラ亜科(Rosoideae)に属する種、セリ科(Apiaceae)に属する種、ユリ科(Liliaceae)に属する種、キキョウ科(Campanulaceae)に属する種、ツツジ科(Ericaceae)に属する種、シロネ属(Lycopus)に属する種、フウロソウ属(Geranium)に属する種、オオバコ属(Plantago)に属する種、オトギリソウ属(Hipericum)に属する種、ハコベ属(Stellaria)に属する種、クサノオウ属(Chelidonium)の属する種、フッキソウ属(Pachysandra)に属する種、及びクサソテツ属(Matteuccia)に属する種から選択されるいずれかの植物である。以下、各植物について具体的に説明をする。
(キク科植物)
キク科植物については、後述する実施例で示すように、同科に属する属の異なる様々な種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれの種であってもよい。例えばキク科植物であれば、コウモリソウ属(Parasenecio)、アザミ属(Cirsium)、ヨモギ属(Artemisia)、又はノブキ属(Adenocaulon)に属する種が挙げられる。
キク科植物については、後述する実施例で示すように、同科に属する属の異なる様々な種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれの種であってもよい。例えばキク科植物であれば、コウモリソウ属(Parasenecio)、アザミ属(Cirsium)、ヨモギ属(Artemisia)、又はノブキ属(Adenocaulon)に属する種が挙げられる。
コウモリソウ属植物の具体例としては、ミミコウモリ(P. kamtschaticus)、タイミンガサ(P. peltifolius)、ヤマタイミンンガサ(P. yatabei)、カニコウモリ(P. adenostyloides)、イズカニコウモリ(P. amagiensis)、コウモリソウ(P. maximowiczianus)、モミジガサ(P. delphiniifolius)、及びウスゲタマブキ(P. farfarifolius)等が挙げられる。
アザミ属植物の具体例としては、ノアザミ(C. japonicum)、フジアザミ(C. purpratum)、チシマアザミ(C. nipponicum)、タイアザミ(C. kamtschaticum)、ハマアザミ(C. martitimum)、モリアザミ(C. dipsacolepis)、及びオニアザミ(C. borealinipponense)等が挙げられる。
ヨモギ属植物の具体例としては、ヨモギ(A. indica)、オオヨモギ(A. montana)、エトロフヨモギ(A. inslaris)、カワラヨモギ(A. capillaris)、オトコヨモギ(A. japonica)、エゾヨモギ(A. montana)、及びシロヨモギ(A. stelleriana)、ニシヨモギ等が挙げられる。
ノブキ属植物の具体例としては、ノブキ(A. himalaicum)、A. bicolor、A. lyratum、A. chilense等が挙げられる。
(ユキノシタ科植物)
ユキノシタ科植物については、同科に属する植物種であれば、いずれの種であってもよい。例えばユキノシタ属(Saxifraga)に属する種、チシマイワブキ属(Micranthes)に属する種、及びヤグルマソウ属(Rodgersia)に属する種等が挙げられる。
ユキノシタ科植物については、同科に属する植物種であれば、いずれの種であってもよい。例えばユキノシタ属(Saxifraga)に属する種、チシマイワブキ属(Micranthes)に属する種、及びヤグルマソウ属(Rodgersia)に属する種等が挙げられる。
ユキノシタ属植物の具体例としては、ユキノシタ(S. stolonifera)、及びダイモンジソウ(S. fortunei)等が挙げられる。
チシマイワブキ属植物の具体例としては、チシマイワブキ(M. punctata)、クロクモソウ(M. fusca)、及びフユウキノシタ(M. Japonica)等が挙げられる。
ヤグルマソウ属植物の具体例としては、ヤグルマソウ(R. podophylla)が挙げられる。
(バラ亜科植物)
バラ亜科植物については、実施例で示すように同亜科に属する属の異なる様々な種において、それぞれの抽出物に凝集抑制効果が認められたことから、同亜科に属する植物種であれば、いずれであってもよい。例えばダイコンソウ属(Geum)、バラ属(Rosa)、ワレモコウ属(Sanguisorba)、又はエゾツルキンバイ属(Argentina)に属する種が挙げられる。
バラ亜科植物については、実施例で示すように同亜科に属する属の異なる様々な種において、それぞれの抽出物に凝集抑制効果が認められたことから、同亜科に属する植物種であれば、いずれであってもよい。例えばダイコンソウ属(Geum)、バラ属(Rosa)、ワレモコウ属(Sanguisorba)、又はエゾツルキンバイ属(Argentina)に属する種が挙げられる。
ダイコンソウ属植物の具体例としては、ダイコンソウ(G. japonicum)、大ダイコンソウ(G. aleppicum)、チングルマ(G. pentapetalum)、ベニバナダイコンソウ(G. coccineum)、及びコキンバイ(G. ternatum)等が挙げられる。
バラ属植物の具体例としては、ノイバラ(R. multiflora)、ヤブイバラ(R. sambucina)、ヤマイバラ(R. onoei)、ハナマス(R. rugosa)、カラフトイバラ(R. amblyotis)、及びタカネバラ(R. nipponensis)等が挙げられる。
ワレモコウ属植物の具体例としては、ワレモコウ(S. officinalis)、ミヤマワレモコウ(S. longifolia)、ナガレボシノワレモコウ(S. tenuifolia)カライトソウ(S. hakusanensis)、タカネトウウチソウ(S. stipulata)、及びシロバナトウウチソウ(S. albiflora)等が挙げられる。
エゾツルキンバイ属植物の具体例としては、エゾツルキンバイ(A. anserina)が挙げられる。
(セリ科植物)
セリ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、ウマノミツバ属(Sanicula)に属する種、又はミツバ属(Cryptotaenia)に属する種等が挙げられる。
ウマノミツバ属植物の具体例としては、ウマノミツバ(S. chinensis)、及びクロバナウマノミツバ(S. rubriflora)等が挙げられる。
ミツバ属植物の具体例としてはミツバ(C. japonica)等が挙げられる。
セリ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、ウマノミツバ属(Sanicula)に属する種、又はミツバ属(Cryptotaenia)に属する種等が挙げられる。
ウマノミツバ属植物の具体例としては、ウマノミツバ(S. chinensis)、及びクロバナウマノミツバ(S. rubriflora)等が挙げられる。
ミツバ属植物の具体例としてはミツバ(C. japonica)等が挙げられる。
(ユリ科植物)
ユリ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、マイヅルソウ属(Maianthemum)に属する種、又はネギ属(Allium)に属する種等が挙げられる。
ユリ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、マイヅルソウ属(Maianthemum)に属する種、又はネギ属(Allium)に属する種等が挙げられる。
マイヅルソウ属植物の具体例としては、マイヅルソウ(M. dilatatum)、ヒメマイヅルソウ(M. bifolium)、エビチャザサ(M. bicolor)、カラフトマイヅルソウ(M. dahuricum)、カナダマイヅルソウ(M. canadense)、ユキザサ(M. japonicum)、トナカイソウ(M. trifolium)、M. stellatum、及びM. racemosum等が挙げられる。
ネギ属植物の具体例としては、セイヨウネギ(A. porrum)、ニラ(A. tuberosum)、ニンニク(A. sativum)、及びギョウジャニンニク(A. victtorialis)等が挙げられる。
(キキョウ科植物)
キキョウ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、ミゾカクシ属(Lobelia)に属する種、又はツリガネニンジン属(Adenophora)に属する種等が挙げられる。
ミゾカクシ属植物の具体例としては、サワギキョウ(L. sessilifolia)、及びミゾカクシ(L. chinensis)等が挙げられる。
ツリガネニンジン属植物の具体例としては、ツリガネニンジン(A. triphylla)、及びソバナ(A. remotiflora)等が挙げられる。
キキョウ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、ミゾカクシ属(Lobelia)に属する種、又はツリガネニンジン属(Adenophora)に属する種等が挙げられる。
ミゾカクシ属植物の具体例としては、サワギキョウ(L. sessilifolia)、及びミゾカクシ(L. chinensis)等が挙げられる。
ツリガネニンジン属植物の具体例としては、ツリガネニンジン(A. triphylla)、及びソバナ(A. remotiflora)等が挙げられる。
(ツツジ科植物)
ツツジ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、イソツツジ属(Ledum)に属する種、又はスノキ属(Vaccinium)に属する種等が挙げられる。
イソツツジ属植物の具体例としては、イソツツジ(L. palustre)等が挙げられる。
スノキ属植物の具体例としては、コケモモ(V. vitis)、及びスノキ(V. smallii)等が挙げられる。
ツツジ科植物については、実施例で示すように同科に属する属の異なる種において、それぞれの抽出物に凝集抑制効果が認められたことから、同科に属する植物種であれば、いずれであってもよい。例えば、イソツツジ属(Ledum)に属する種、又はスノキ属(Vaccinium)に属する種等が挙げられる。
イソツツジ属植物の具体例としては、イソツツジ(L. palustre)等が挙げられる。
スノキ属植物の具体例としては、コケモモ(V. vitis)、及びスノキ(V. smallii)等が挙げられる。
(シロネ属植物)
シロネ属(Lycopus)植物の具体例としては、シロネ(L. lucidus)、コシロネ(L. cavaleriei)、ヒメシロネ(L. maackianus)、及びエゾシロネ(L. uniflorus)が挙げられる。
シロネ属(Lycopus)植物の具体例としては、シロネ(L. lucidus)、コシロネ(L. cavaleriei)、ヒメシロネ(L. maackianus)、及びエゾシロネ(L. uniflorus)が挙げられる。
(フウロソウ属植物)

(クサノオウ属植物)

2.凝集抑制組成物
2-1.概要
本発明の第2の態様は、凝集性タンパク質の凝集抑制組成物である。本発明の組成物は、第1態様に記載の凝集抑制剤を1種以上、有効成分として包含する。本発明の凝集抑制組成物によれば、凝集抑制剤を扱いやすく、また安定した状態で適用することが可能となる。それによって、例えば医薬組成物として、生体投与時の負荷や侵襲性を低減し、より投与しやすい形態で提供することができる。
2-1.概要
本発明の第2の態様は、凝集性タンパク質の凝集抑制組成物である。本発明の組成物は、第1態様に記載の凝集抑制剤を1種以上、有効成分として包含する。本発明の凝集抑制組成物によれば、凝集抑制剤を扱いやすく、また安定した状態で適用することが可能となる。それによって、例えば医薬組成物として、生体投与時の負荷や侵襲性を低減し、より投与しやすい形態で提供することができる。

本態様の凝集抑制組成物は(本明細書では、以下、しばしば「本組成物」と略称する)、必須の構成因子として有効成分を、また選択的構成因子として溶媒及び/又は担体を含む。以下、それぞれの構成因子について具体的に説明をする。

(1)有効成分
本組成物は、第1態様に記載の凝集抑制剤を有効成分として含む。本組成物は、1種、又は異なる2種以上の凝集抑制剤を含むことができる。また、第1態様に記載の凝集抑制剤以外で、凝集性タンパク質に対して、同様の凝集抑制作用を有する公知の物質(核酸、ポリペプチド、低分子化合物等)を選択的に含んでいてもよい。
本組成物は、第1態様に記載の凝集抑制剤を有効成分として含む。本組成物は、1種、又は異なる2種以上の凝集抑制剤を含むことができる。また、第1態様に記載の凝集抑制剤以外で、凝集性タンパク質に対して、同様の凝集抑制作用を有する公知の物質(核酸、ポリペプチド、低分子化合物等)を選択的に含んでいてもよい。
本組成物に含まれる凝集抑制剤の含有量は、包含される凝集抑制剤の種類及び/又はその有効量、本組成物の剤形、後述する担体又は添加物の種類、並びに疾患の種類によって異なるため、それぞれの条件を勘案して適宜定めればよい。
本明細書において「有効量」とは、本組成物において凝集抑制剤が有効成分として、その機能を発揮する上で必要な量で、かつそれを適用する生体に対して有害な副作用を、ほとんど又は全く生じない量をいう。この有効量は、被験体の情報、投与経路、及び投与回数等の様々な条件によって変化し得る。
本明細書において「被験体」とは、凝集抑制剤又は本組成物の適用対象となる生体をいう。例えば、ヒト、家畜(ウシ、ウマ、ヒツジ、ヤギ、ブタ、ニワトリ、ダチョウ等)、競走馬、愛玩動物(イヌ、ネコ、ウサギ等)、実験動物(マウス、ラット、モルモット、サル等)等が該当する。好ましくはヒトである(この場合、特に「被験者」と称する)。また、「被験体の情報」とは、適用する生体の様々な個体情報であって、例えば、被験者の場合であれば、全身の健康状態、疾患・病害に罹患している場合にはその進行度や重症度、年齢、体重、性別、食生活、薬剤感受性、併用薬物の有無及び治療に対する耐性等を含む。本組成物における凝集抑制剤の有効量、及びそれに基づいて算出される適用量は、個々の被験体の情報等に応じて、最終的には医師、又は獣医師等の判断によって決定される。
(2)溶媒
本組成物は、薬学的に許容可能な溶媒を含むことができる。溶媒は、本組成物における選択的構成因子であり、必要に応じて添加すればよい。「薬学的に許容可能な」とは、生体に無害又は低毒性で、製剤技術分野において通常使用可能な、好ましくは医薬組成物に使用可能なことをいう。
溶媒には、例えば、水若しくは水溶液、緩衝液、又は乳化剤、有機溶剤が挙げられる。

上記の他にも、必要であれば医薬において通常用いられる賦形剤、充填剤、結合剤、崩壊剤、吸収促進剤、増量剤、付湿剤、保湿剤、湿潤剤、吸着剤、崩壊抑制剤、コーティング剤、着色剤等を適宜含むこともできる。
このような担体は、主として剤形形成を容易にし、また剤形及び薬剤効果を維持する他、有効成分である凝集抑制剤が生体内での分解を受け難くするために用いられるものであって、必要に応じて適宜使用すればよい。
2-2-2.剤形と投与方法
本組成物の剤形は、凝集抑制剤及び他の公知の凝集性タンパク質の凝集抑制剤等の有効成分を不活化させず、投与後に生体内で有効成分の薬理効果を発揮し得る形態であれば特に限定しない。また、本組成物の具体的な剤形は、投与法及び/又は処方条件に応じて適宜選択すればよい。一般に投与法は、経口投与と非経口投与に大別することができるが、本組成物はそれぞれの投与法に適した剤形にすればよい。
本組成物の剤形は、凝集抑制剤及び他の公知の凝集性タンパク質の凝集抑制剤等の有効成分を不活化させず、投与後に生体内で有効成分の薬理効果を発揮し得る形態であれば特に限定しない。また、本組成物の具体的な剤形は、投与法及び/又は処方条件に応じて適宜選択すればよい。一般に投与法は、経口投与と非経口投与に大別することができるが、本組成物はそれぞれの投与法に適した剤形にすればよい。
経口投与の場合、剤形としては、固形剤(錠剤、丸剤、舌下剤、カプセル剤、ドロップ剤を含む)、顆粒剤、粉剤、散剤、液剤(内用水剤、懸濁剤、乳剤、シロップ剤を含む)等が挙げられる。固形剤は、必要に応じて当該技術分野で公知の剤皮を施した剤形、例えば、糖衣錠、ゼラチン被包錠、腸溶錠、フィルムコーティング錠、二重錠、多層錠にすることができる。経口投与は侵襲性が極めて低く、投与が容易なため、好ましい投与法である。
非経口投与の場合、全身投与及び局所投与に分けることができ、また局所投与は、さらに組織内投与、経表皮投与、経粘膜投与及び経直腸的投与にさらに細分することもできる。本組成物もそれぞれの投与法に適した剤形にすればよい。例えば、全身投与又は組織内投与に適した剤形としては、液剤である注射剤を挙げることができる。経表皮投与又は経粘膜投与に適した剤形としては、液剤(塗布剤、点眼剤、点鼻剤、吸引剤を含む)、懸濁剤(乳剤、クリーム剤を含む)、粉剤(点鼻剤、吸引剤を含む)、ペースト剤、ゲル剤、軟膏剤、硬膏剤等を挙げることができる。経直腸的投与に適した剤形としては、坐剤等を挙げることができる。
本組成物の投与法は、後述する適用対象疾患の種類や適用対象部位に応じて変動する。最終的に本組成物の有効成分である凝集抑制剤が適用対象部位に到達し、その効果を発揮することができれば、投与方法は限定されない。例えば、適用対象疾患が神経変性疾患であれば、適用対象部位は主に中枢神経系である脳脊髄となる。この場合、中枢神経内に直接投与する組織内投与であってもよいし、循環系を介した全身投与であってもよい。具体的には、例えば、脳脊髄内注射による組織内投与、又は血管内注射(静脈内注射、動脈内注射を含む)若しくはリンパ管内注射等の循環器内投与が挙げられる。一般的に、組織内投与のような直接投与は、適用対象部位に有効成分を確実に、かつ高濃度で送達できる。一方、循環系を介した全身投与は、適用対象部位に低侵襲性で、漏れなく送達できる。いずれを選択するかは、状況や病態に応じて適宜定めればよい。
注射剤には、前記乳化剤、懸濁剤、界面活性剤、安定剤、pH調節剤等と適宜組み合わせて、一般に認められた製薬実施で要求される単位用量形態で混和することによって製剤化すればよく、単位用量アンプル又は多用量容器の状態で提供される。
なお、上記各剤形の具体的な形状、大きさについては、いずれもそれぞれの剤形において当該分野で公知の剤形の範囲内にあればよく、特に限定はしない。
2-2-3.適用対象疾患
本組成物は、凝集性タンパク質の凝集を原因とする各種疾患の治療又は予防に適用できる。適用対象となる疾患の種類は、前述の通り、疾患原因となる凝集性タンパク質が凝集し、それによって形成された凝集体が細胞内、又は細胞外に沈着することによって発症する疾患であれば限定しない。例えば、疾患の種類として、アルツハイマー病、パーキンソン病、ハンチントン病、動脈硬化症、全身性ALアミロイドーシス、及び筋萎縮性側索硬化症が挙げられる。また、凝集性タンパク質の種類についても、それぞれの疾患の原因となる凝集性タンパク質であればよく、特に限定はしない。本組成物の適用対象疾患とその原因となる凝集性タンパク質との関連について、以下でいくつかの例を挙げて説明をする。

アルツハイマー病は、アミロイドβタンパク質の脳内の神経細胞に凝集蓄積に始まり、微小管タンパク質であるタウタンパク質が過リン酸化されて線維化した後に、神経細胞が破壊され脳が萎縮することにより発症する。したがって、アルツハイマー病の疾患原因となる凝集性タンパク質としては、βアミロイド、又はタウタンパク質の他、α-シヌクレイン等が該当し得る。本組成物に含まれる凝集抑制剤は、これらのタンパク質の凝集を抑制することにより、アルツハイマー病の進行抑制又は予防に対する効果を奏し得る。
パーキンソン病(Perkinson's disease:PD)は、レビー小体(Lewy body)と呼ばれるタンパク質凝集体を構成するミスフォールドされたα-シヌクレイン(α-Synuclein)の多量化と蓄積による神経細胞死が発症に関与するとされている。したがって、パーキンソン病の疾患原因となる凝集性タンパク質としては、ミスフォールドした変異型α-シヌクレイン等が該当し得る。本組成物に含まれる凝集抑制剤は、このタンパク質の凝集を抑制することにより、パーキンソン病の進行抑制又は予防に対する効果を奏し得る。
ハンチントン病(Huntington's disease:HD)は、CAGリピート数が伸長した変異ハンチンチン(mutated Huntingtin:mHTT)遺伝子の発現により生じる変異ハンチンチン(mHTT)が神経細胞の機能障害と細胞死を誘導し、発症に関与することが知られている。したがって、ハンチントン病の疾患原因となる凝集性タンパク質としては、変異型ハンチンチンが該当し得る。本組成物に含まれる凝集抑制剤は、変異ハンチンチンによる凝集を抑制することにより、ハンチントン病の進行抑制又は予防に対する効果を奏し得る。
伝達性海綿脳症(クロイツフェルトヤコブ病、狂牛病、又はプリオン病を含む)は、異常プリオンタンパク質が中枢神経系で増加し、細胞外凝集することによって正常な神経組織が破壊され、引き起こされる神経変性疾患である。したがって伝達性海綿脳症の疾患原因となる凝集性タンパク質としては、異常プリオンタンパク質が該当し得る。本組成物に含まれる凝集抑制剤は、異常プリオンタンパク質による凝集を抑制することにより、伝達性海綿脳症の進行抑制又は予防に対する効果を奏し得る。
<実施例1:植物抽出物の調製>
(目的)
本発明の凝集抑制剤である各種植物抽出物を調製する。

(方法)
(1)シロネ(シロネ属植物)の植物抽出物の調製
北海道白糠町で採取したシロネ(L. lucidus)の全草(葉、茎、及び根を含む)を溶媒に95%エタノールを用いて1週間浸漬した。その後、溶媒を濾過し,濾液をロータリーエバポレーター(東京理化器械株式会社)に付して減圧下溶媒留去した。得られた残渣を油回転ポンプ(株式会社アルバック)で、さらに乾燥させてエタノール抽出物を得た。
(1)シロネ(シロネ属植物)の植物抽出物の調製
北海道白糠町で採取したシロネ(L. lucidus)の全草(葉、茎、及び根を含む)を溶媒に95%エタノールを用いて1週間浸漬した。その後、溶媒を濾過し,濾液をロータリーエバポレーター(東京理化器械株式会社)に付して減圧下溶媒留去した。得られた残渣を油回転ポンプ(株式会社アルバック)で、さらに乾燥させてエタノール抽出物を得た。
次に、得られたエタノール抽出物を水中で分散させ、分液ロートを用いて、t-ブチルメチルエーテルで抽出した。このt-ブチルメチルエーテル抽出層を合わせ、ロータリーエバポレーターに付して減圧下溶媒留去した。得られた残渣を油回転ポンプでさらに乾燥させてt-ブチルメチルエーテル画分を得た。
続いて、水層を、分液ロートを用いて、水飽和n-ブタノールで抽出した。このt-ブチルメチルエーテル抽出層を合わせ、再度ロータリーエバポレーターに付し、減圧下溶媒留去した。得られた残渣を油回転ポンプでさらに乾燥させ、n-ブタノール画分を得た。
(2)ゲンノショウコ(フウロソウ属)植物の植物抽出物の調製(No.1)
北海道白糠町で採取、又は栽培したゲンノショウコ(G. thunbergii)の全草(葉、茎、及び根を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でゲンノショウコ抽出物を含む、エタノール画分、t-ブチルメチルエーテル画分、及びn-ブタノール画分を得た。

北海道白糠町で採取、又は栽培したエゾフウロ(Geranium yesoense)の全草(葉、茎、及び根を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でエゾフウロ抽出物を含む、エタノール画分、t-ブチルメチルエーテル画分、及びn-ブタノール画分得た。

北海道白糠町で採取、又は栽培したミミコウモリ(Parasenecio adenostyloides)の全草(葉、茎、及び根を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でミミコウモリ抽出物を含む、エタノール画分、t-ブチルメチルエーテル画分、及びn-ブタノール画分を得た。

北海道白糠町で採取、又は栽培したオオヨモギ(Artemisia montana)の葉、及び茎を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でオオヨモギ抽出物を含むエタノール画分を得た。

北海道白糠町で採取、又は栽培したトモエソウ(Hypericum ascyron)の全草(葉、茎、及び根を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でトモウエソウ抽出物を含む、エタノール画分、t-ブチルメチルエーテル画分、及びn-ブタノール画分を得た。

北海道白糠町で採取、又は栽培したウマノミツバ(Sanicula chinensis)の全草(葉、茎、及び根を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でウマノミツバ抽出物を含む、エタノール画分、t-ブチルメチルエーテル画分、及びn-ブタノール画分を得た。

北海道白糠町で採取、又は栽培したフキユキノシタ(Micranthes japonica)の全草(葉、茎、及び根を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でフユウキノシタ抽出物を含むエタノール画分を得た。

北海道白糠町で採取、又は栽培したエゾツルキンバイ(Potentilla egedei var. grandis)の地上部(葉、茎、及び花を含む)を、室温にて95%エタノールに1週間浸漬した後、前記(1)と同様の手順でエゾツルキンバイ抽出物を含む、エタノール画分、t-ブチルメチルエーテル画分、及びn-ブタノール画分を得た。

(15)甘茶(アジサイ科アジサイ属植物)の植物抽出物の調製(対照用)
甘茶(Hydrangea macrophylla)の乾燥葉(オーピーバイオ社)をミルで粉砕後、粉砕物に90%エタノールを溶媒として添加し、28℃にて20時間振盪抽出した。溶媒を3,000rpmで20分間遠心後、上清を綿栓濾過し、再度遠心濃縮をした後、凍結乾燥して、甘茶抽出物を得た。
甘茶(Hydrangea macrophylla)の乾燥葉(オーピーバイオ社)をミルで粉砕後、粉砕物に90%エタノールを溶媒として添加し、28℃にて20時間振盪抽出した。溶媒を3,000rpmで20分間遠心後、上清を綿栓濾過し、再度遠心濃縮をした後、凍結乾燥して、甘茶抽出物を得た。

<実施例2:無細胞系における凝集抑制剤の凝集抑制活性の検証>
(目的)
本発明の凝集抑制剤の無細胞系における凝集抑制活性について検証する。

(方法)
(1)凝集タンパク質溶液の調製
凝集タンパク質にはアミロイドβタンパク質(Aβ)を用いた。5mgのAβ(Human,1-42:ペプチド研究所社)に5mLのHFIP(1,1,1,3,3,3-Hexafluoro-2-propanol:富士フィルム和光純薬社)を添加し、懸濁した後、懸濁液を1時間室温静置した。その後、25℃、43 kHzで10分間、超音波処理してモノマー化した後、クリーンベンチ内に24時間放置してHFIPを揮発させた。揮発後、1071μLのDMSOで溶解し、1mM Aβ溶液とした。ここで調製したAβ溶液を、以下「Aβ42」と表記する。Aβ42は、1.5mLチューブに256μLずつ分注した後、使用まで-80℃にて保存した。
(1)凝集タンパク質溶液の調製

(2)量子ドット修飾化アミロイドβタンパク質(QDAβ)の調製
Aβを量子ドット(QD)で修飾し、量子ドット修飾化アミロイドβタンパク質(QDAβ)を調製した。125μLの8μM QdotTM605 ITKTM amino(PEG) Quantum Dotsを2本の1.5mLチューブに入れ、10,000×gで4℃にて1分間遠心した。それぞれの上清を遠心濃縮チューブに移し、4500μLのPBSを加えた。2本の合計量が50μL以下になるまで、4℃、3800×gで遠心し、濾液は廃棄した。PBSを補充した後、再度50μLになるまで遠心した。得られたQD溶液を全量が約180μLになるように1本にまとめた。10mM sulfo-EMCSを20μL追加し、1時間常温で静置してQD-EMCSを調製した。
Aβを量子ドット(QD)で修飾し、量子ドット修飾化アミロイドβタンパク質(QDAβ)を調製した。125μLの8μM QdotTM605 ITKTM amino(PEG) Quantum Dotsを2本の1.5mLチューブに入れ、10,000×gで4℃にて1分間遠心した。それぞれの上清を遠心濃縮チューブに移し、4500μLのPBSを加えた。2本の合計量が50μL以下になるまで、4℃、3800×gで遠心し、濾液は廃棄した。PBSを補充した後、再度50μLになるまで遠心した。得られたQD溶液を全量が約180μLになるように1本にまとめた。10mM sulfo-EMCSを20μL追加し、1時間常温で静置してQD-EMCSを調製した。
QD-EMCS調製後に、液中に含まれる未反応N-ヒドロキシスクシンイシド基を不活性化させるために100mM K-グルタミン酸を20μL加えて、常温で10分間静置した。
脱塩カラムを2本用意し、それぞれにレジンを約800μL入れて、1,000×gで4℃にて1分間遠心した。各カラムに300μLのPBSEを積載し、1,000×gで4℃にて1分間遠心を行う工程を2セット行い、脱塩カラムを調製した。
2本の脱塩カラムの中心に前記QD-EMCSを110μL染み込ませた後、スタッカーとしてPBSEを15μL加えた。1,000×gで4℃にて2分間遠心した後、2本の脱塩カラムの濾液(脱塩QD-EMCS)をまとめた。得られた脱塩QD-EMCSに1.0mM Cys-Aβを20μL/DMSOを加え、混合後、1時間常温で静置した。その後、液中に含まれる未反応マレイミド基を不活性化させるために20μLの100mMの2-メルカプトエタノールを加えて、常温で10分間静置した。濾液を2本の遠心濃縮チューブへ145μLずつ移し、水を4500μL加えた後、3,800×gで4℃にて17分間遠心した。濾液は廃棄し、得られた溶液の全量が約140μLになるようにして1本にまとめた。
脱塩カラムに水を300μL入れて1,000×gで4℃にて1分間遠心を行い、この工程を2回繰り返した。濾液を70μLずつ脱塩カラムに染み込ませて、スタッカーの水を15μL加えた。1000×gで4℃にて2分間遠心し、目的のQDAβを得た。
(3)各植物抽出物のAβ凝集抑制活性評価
濾過フィルターデバイス(Amicon Ultra-0.5 Ultracel-50メンブレン)を1.5mLチューブに取り付け、MilliQ水を濾過フィルターデバイスに滴下し、室温(15~25℃)にて14,000×gで10分間遠心した。濾液を除き、再度フィルターにMilliQ水400μlを滴下後、室温(15~25℃)にて14,000×gで10分間遠心した。濾液を捨て、濾過フィルターに0.1% Tween-20水溶液を滴下し、室温(15~25℃)で2時間静置した。
濾過フィルターデバイス(Amicon Ultra-0.5 Ultracel-50メンブレン)を1.5mLチューブに取り付け、MilliQ水を濾過フィルターデバイスに滴下し、室温(15~25℃)にて14,000×gで10分間遠心した。濾液を除き、再度フィルターにMilliQ水400μlを滴下後、室温(15~25℃)にて14,000×gで10分間遠心した。濾液を捨て、濾過フィルターに0.1% Tween-20水溶液を滴下し、室温(15~25℃)で2時間静置した。
ブロッキング操作後、遠心し、濾液を棄て、室温(15~25℃)にて14,000×gで10分間遠心した。濾液を除き、フィルターにMilliQ水を滴下し、室温(15~25℃)にて14,000×gで10分間遠心し、再度、濾液を除き、フィルターにMilliQ水を滴下し、室温(15~25℃)にて14,000×gで10分間遠心後、濾液を除き、室温(15~25℃)にて14,000×gで2分間空遠心した。
濾過フィルターデバイスを新たな1.5mLチューブに取り付け、AD-iPS由来神経系細胞をNbM 培地(実施例3参照)で培養した後、培養上清を濾過フィルターに滴下し、室温(15~25℃)にて14,000×gで30分間遠心し、濾液を回収した。この時、培養上清に含まれるAβ42濃度を予めヒトβアミロイドELISAキット(富士フィルム和光純薬)で測定した。濾液(PBS又は培養上清)にQD-Aβ40溶液(終濃度50nM)、Aβ42溶液(終濃度10μM)、及び実施例1の(1)~(15)で調製した各植物抽出物(凝集抑制剤)を添加して反応液を作製した。陽性対照には、植物抽出物の代替液としてロスマリン酸(RA)溶液を使用した。陰性対照には、植物抽出物の代替液としてDMSOを使用した反応液を使用した。植物抽出物は、終濃度1500μM、300μM、60μM、12μM、及び2.4μMとなる複数反応液を調製した。これら反応液を37℃で24時間インキュベートし、各植物抽出物の各濃度におけるAβ凝集量を定量した。
Aβ凝集量の測定は、凝集体の蛍光画像から算出される輝度値SDがAβ凝集量と正に相関することを利用して、輝度値SDの値を指標として行った。蛍光画像は、OLYMPUS DP72(CCD)を装備したNikon ECLIPSE TE2000-S、又はNikon DS-Ri2(CMOS)カメラを備えたNikon ECLIPSE Tiの蛍光顕微鏡に反応液が保持されたマイクロウェルプレートをセットし、励起光を照射して反応生成物から蛍光を発光させ、これをCCDカメラで撮影して取得した。励起光は、用いられる量子ドットに応じて適宜設定した。例えば、使用する量子ドットがQdot605の場合、励起光の波長は580nmより短波長側、例えば532~552nmの波長とした。発生した蛍光に応じたバンドパスフィルタを使用して蛍光撮像を行った。例えば、使用する量子ドットがQdot605の場合、594~646nmの波長帯域の光のみを選択的に透過することができるバンドパスフィルタを用いた。
(結果)
陽性対照のロスマリン酸(RA)溶液では、Aβの凝集が確認できたが、陰性対照のDMSO溶液では確認できなかった。得られた輝度値SDに基づいて、各植物抽出物について全体の50%に相当するAβが凝集しなくなる濃度(EC50)を算出した。陽性対照における凝集Aβを定量化したところ、RA濃度が46.0±7.4μM以下であれば、全体の50%程度に相当するAβが凝集しなくなることが確認できた。また、EC50を逆数とすると、凝集抑制活性を有する場合は数値が0以上となる。さらに、その数値が大きいほど抑制活性が高いことを示す。これらの情報を基準とした、各植物抽出物の1/EC50を表1に示す。
陽性対照のロスマリン酸(RA)溶液では、Aβの凝集が確認できたが、陰性対照のDMSO溶液では確認できなかった。得られた輝度値SDに基づいて、各植物抽出物について全体の50%に相当するAβが凝集しなくなる濃度(EC50)を算出した。陽性対照における凝集Aβを定量化したところ、RA濃度が46.0±7.4μM以下であれば、全体の50%程度に相当するAβが凝集しなくなることが確認できた。また、EC50を逆数とすると、凝集抑制活性を有する場合は数値が0以上となる。さらに、その数値が大きいほど抑制活性が高いことを示す。これらの情報を基準とした、各植物抽出物の1/EC50を表1に示す。
表1に示すように、本発明の植物のエタノール抽出物は、全てAβ凝集抑制活性(1/EC50)を有することが確認された。またエタノール抽出物をt-ブチルメチルエーテル又はn-ブタノールで抽出した場合にも、1/EC50は高い数値を示す傾向が見られた。さらに、表1の結果からt-ブチルメチルエーテルのような疎水性抽出物はAβ凝集抑制に特に有効と考えられた。一方、対照植物である甘茶のエタノール抽出物では、Aβ凝集抑制活性は全く確認できなかった。
<実施例3:細胞系でのアミロイドβ凝集抑制活性の検証>

細胞表面又は内部でのAβの沈着又は凝集抑制活性を評価するため、下記の手順により、評価用の神経系細胞を調製した。

(1)培地及び試薬の調製
(a)NbM培地:Neuro basal Medium minus phenol red(Thermo Fisher Scientific社)50mLに、B-27 supplement(Thermo Fisher Scientific社)1mL、及びPenicillin-Streptomycin Mixed Solution(ナカライテスク社)を添加し、NbM培地を調製した。

(c)matrigel-PBS:25 mLのPBSに500μLのhESC-Qualified Matrigel(Corning社)を添加・懸濁し、matrigel-PBSを調製した。
(d)SHH添加NbM培地:ヒト組み換えソニックヘッジホッグ(ベリタス社)10μgに100μLのDMSOを添加し、SHH溶液とした。前記NbM培地10mLに前記SHH溶液10μLを添加し、SHH添加NbM培地を調製した。
(2)評価用神経系細胞の調製
予め培養したAD-iPS由来神経前駆細胞を剥離、遠心、洗浄し、Y-27632添加NbM培地に懸濁して神経前駆懸濁液を調製した。この懸濁液を、予めmatrigel-PBSでコーティング処理を施した96 well平底プレートに100μL/wellで分注し細胞を播種した。プレートは静置し、37℃で1日間インキュベートした。
予め培養したAD-iPS由来神経前駆細胞を剥離、遠心、洗浄し、Y-27632添加NbM培地に懸濁して神経前駆懸濁液を調製した。この懸濁液を、予めmatrigel-PBSでコーティング処理を施した96 well平底プレートに100μL/wellで分注し細胞を播種した。プレートは静置し、37℃で1日間インキュベートした。
(Day0)翌日、培地を全てSHH添加NbM培地に交換してインキュベートを継続した。当該操作を実施した日をDay0とし、以降のインキュベート日数を経時した。
(Day2)新たなNbM培地を50μL/wellで添加し、インキュベートを継続した。
(Day6)プレートの細胞を剥がさないよう培地の半量を除去した後、新たなNbM培地を添加し、インキュベートを継続した。
(Day9)新たなNbM培地を50μL/wellで添加し、インキュベートを継続した。

実施例2でAβ凝集抑制活性が確認された植物抽出物のうち、トモエソウ、エゾフウロ、ウマノミツバ、フキユキノシタ、ナガレボシワレモコウと、新たに実施例1の(16)~(19)で調製したサワギキョウ(Lobelia sessilifolia:キキョウ科ミゾカクシ属)、ノブキ(Adenocaulon himalaicum:キク科ノブキ属)、マイヅルソウ(Maianthemum dilatatum:マイヅルソウ属)、エゾオオヤマハコベ(Stellaria radians:ハコベ属)の各植物抽出物について、細胞系にてAβ凝集抑制活性を確認した。細胞系での評価は下記の方法で実施した。
実施例2でAβ凝集抑制活性が確認された植物抽出物のうち、トモエソウ、エゾフウロ、ウマノミツバ、フキユキノシタ、ナガレボシワレモコウと、新たに実施例1の(16)~(19)で調製したサワギキョウ(Lobelia sessilifolia:キキョウ科ミゾカクシ属)、ノブキ(Adenocaulon himalaicum:キク科ノブキ属)、マイヅルソウ(Maianthemum dilatatum:マイヅルソウ属)、エゾオオヤマハコベ(Stellaria radians:ハコベ属)の各植物抽出物について、細胞系にてAβ凝集抑制活性を確認した。細胞系での評価は下記の方法で実施した。
なお、トモエソウはn-ブタノール画分を、エゾフウロはt-ブチルメチルエーテル画分を使用した以外、他は全てエタノール画分を使用した。
前記実施例2(1)及び(2)で調製したAβ42溶液、及びQD-Aβ溶液をそれぞれ新しいNbM培地で希釈し、調製済みAβ42溶液、及び調製済みQD-Aβ溶液を調製した。また、実施例1で調製した植物抽出物を新鮮NbM培地で希釈し、植物抽出物含有液(300 ng/μL、又は150 ng/μL)を調製した。さらに、DMSOのみを植物抽出物の場合と同倍率で希釈したものを陰性対照とし、調製した評価用神経系細胞の培養上清をピペットで除去した後、前記調製済みQD-Aβ溶液を、細胞を剥離させないように70μL/wellずつ分注した。次いで、前記植物抽出物含有液10μL/well、及び前記調製済みAβ42溶液70μL/wellを注入した(終濃度: QD-Aβ溶液30nM,Aβ42溶液10μM)。プレートにガス透過性シールを貼り、37℃, 5% CO2インキュベーター内に設置したIncucyte(登録商標)ライブセル解析システム(ザルトリウス社)にセットした。セット後、測定条件の安定する2時間後から計測を開始し、その後、22時間の経時的なAβの凝集量を計測した。Incusyteの設定は以下の通りとした。

(結果)
結果を図1及び図2に示す。
図1は、9種の植物抽出物について、細胞系にてAβの凝集/沈着効果を示すプロット図である。陰性対照であるDMSOよりもY軸プロットが下がる時には、植物抽出物に神経系細胞に対するAβ凝集/沈着抑制活性を有することを示す。
結果を図1及び図2に示す。
図1は、9種の植物抽出物について、細胞系にてAβの凝集/沈着効果を示すプロット図である。陰性対照であるDMSOよりもY軸プロットが下がる時には、植物抽出物に神経系細胞に対するAβ凝集/沈着抑制活性を有することを示す。
その結果、無細胞系評価のみならず、細胞系評価においても、検証した前記9種の植物抽出物の全てにAβ凝集/沈着抑制活性が確認された。
以上の結果から、本発明で得られた植物抽出物は、凝集性タンパク質の凝集を抑制する凝集抑制活性を有し、新規凝集抑制剤となることが立証された。
図2は、フキユキノシタ抽出物を添加した系とDMSOを添加した系の、22時間時点の細胞観察画像である。この図からも、フキユキノシタ添加系において細胞へのAβ凝集/沈着が抑制されていることが確認された。
<実施例4:各素材エキスのAβ凝集抑制活性評価>
新たに実施例1(20)~(28)で調製したミツバ、ギョウジャニンニク、ツリガネニンジン、イソツツジ、コケモモ、ハコベ、クサソテツ、クサノオウ、及びフッキソウの9種の植物抽出物について、実施例2と同様にして、細胞へのAβ凝集/沈着に対する効果を調べた。
その結果、用いた9種の植物抽出物全てで、細胞へのAβ凝集/沈着をDMSOよりも抑制することが確認された。
新たに実施例1(20)~(28)で調製したミツバ、ギョウジャニンニク、ツリガネニンジン、イソツツジ、コケモモ、ハコベ、クサソテツ、クサノオウ、及びフッキソウの9種の植物抽出物について、実施例2と同様にして、細胞へのAβ凝集/沈着に対する効果を調べた。
その結果、用いた9種の植物抽出物全てで、細胞へのAβ凝集/沈着をDMSOよりも抑制することが確認された。
表3に、実施例4で用いたAβ凝集/沈着抑制活性を有する前記9種の植物が属する科、及び属をまとめた。
Claims (14)
- キク科(Asteraceae)、バラ亜科(Rosoideae)、ユキノシタ科(Saxifragaceae)、セリ科(Apiaceae)、ユリ科(Liliaceae)、キキョウ科(Campanulaceae)、ツツジ科(Ericaceae)、シロネ属(Lycopus)、フウロソウ属(Geranium)、オオバコ属(Plantago)、オトギリソウ属(Hipericum)、ハコベ属(Stellaria)、クサノオウ属(Chelidonium)、フッキソウ属(Pachysandra)、及びクサソテツ属(Matteuccia)に属する植物からなる群から選択される植物の抽出物からなる凝集性タンパク質の凝集抑制剤。
- 前記キク科に属する植物がコウモリソウ属(Parasenecio)、アザミ属(Cirsium)、ヨモギ属(Artemisia)、又はノブキ属(Adenocaulon)に属する植物である、請求項1に記載の凝集抑制剤。
- 前記バラ亜科に属する植物がダイコンソウ属(Geum)、バラ属(Rosa)、ワレモコウ属(Sanguisorba)、又はエゾツルキンバイ属(Argentina)に属する植物である、請求項1に記載の凝集抑制剤。
- 前記セリ科に属する植物が、ウマノミツバ属(Sanicula)又はミツバ属(Cryptotaenia)に属する植物である、請求項1に記載の凝集抑制剤。
- 前記ユリ科に属する植物が、マイヅルソウ属(Maianthemum)又はネギ属(Allium)に属する植物である、請求項1に記載の凝集抑制剤。
- 前記キキョウ科に属する植物が、ミゾカクシ属(Lobelia)又はツリガネニンジン属(Adenophora)に属する植物である、請求項1に記載の凝集抑制剤。
- 前記ツツジ科に属する植物が、イソツツジ属(Ledum)又はスノキ属(Vaccinium)に属する植物である、請求項1に記載の凝集抑制剤。
- 前記植物がキク科、ユキノシタ科、フウロソウ属、オトギリソウ属、ウマノミツバ属、ワレモコウ属、マイヅルソウ属、及びハコベ属に属する植物である、請求項1~7のいずれか1項に記載の凝集抑制剤。
- 前記抽出物が全草由来である、請求項1に記載の凝集抑制剤。
- 前記凝集性タンパク質がアミロイドβである、請求項1に記載の凝集抑制剤。
- 請求項1に記載の凝集抑制剤を有効成分として含む凝集性タンパク質の凝集抑制組成物。
- 請求項1に記載の異なる二以上の凝集抑制剤を有効成分として含む、請求項11に記載の凝集抑制組成物。
- 凝集性タンパク質の凝集を原因とする疾患の治療又は予防用である、請求項11又は12のいずれか1項に記載の凝集抑制組成物。
- 前記疾患がアルツハイマー病である、請求項13に記載の凝集抑制組成物。
