Abstract
本発明は、熱安定性に優れた高活性アルカリホスファターゼを提供する。より具体的には、本発明は、以下(A)、(B)、または(C)のタンパク質である改変アルカリホスファターゼを提供する: (A)配列番号3、5、7、もしくは9のアミノ酸配列を含むタンパク質; (B)配列番号3、5、7、もしくは9のアミノ酸配列に対して95%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または (C)配列番号3、5、7、もしくは9のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~23個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
Description
本発明は、改変アルカリホスファターゼなどに関する。
アルカリホスファターゼ(EC 3.1.3.1)は、アルカリ性に至適pHを有する、基質選択性の低いホスホモノエステラーゼである。アルカリホスファターゼは、実験試薬および臨床検査試薬において汎用されている。例えば、実験では、アルカリホスファターゼは、DNAまたはRNAの5’末端の脱リン酸化処理、およびレポーターアッセイにおけるレポーターに利用されている。臨床検査試薬では、アルカリホスファターゼは、アルカリホスファターゼで標識された物質(例、抗体等の親和性物質)の形態において、標的分子の検出または定量に利用されている。
アルカリホスファターゼとしては、ウシ小腸由来アルカリホスファターゼ(Bovine Intestinal Alkaline Phosphatase:bIAP)の複数のアイソマーが知られている。このうち、アイソマーIIであるbIAPIIは、活性が高いものの、熱安定性は低く、これらの特性の両立には困難を伴うこと、およびbIAPIIの322位がアスパラギン酸、323位がリジンもしくはアスパラギン、385位がセリンもしくはグリシンもしくはアスパラギン、430位がアラニンであるbIAPII変異体が、天然型bIAPIIと同等の活性を維持しつつ高い熱安定性を示すことが知られている(特許文献1)。
本発明の目的は、熱安定性に優れた高活性アルカリホスファターゼを提供することである。
本発明者らは、鋭意検討した結果、bIAPIIの活性および熱安定性の双方を改善できる特定の変異を有する改変アルカリホスファターゼを作製することに成功し、本発明を完成させるに至った。
すなわち本発明は、以下のとおりである。
〔1〕以下(A)、(B)、または(C)のタンパク質である、改変アルカリホスファターゼ:
(A)配列番号3、5、7、もしくは9のアミノ酸配列を含むタンパク質;
(B)配列番号3、5、7、もしくは9のアミノ酸配列に対して95%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号3、5、7、もしくは9のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~23個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
〔2〕(B)または(C)のアミノ酸配列が、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異を維持する、〔1〕の改変アルカリホスファターゼ。
〔3〕(B)または(C)のアミノ酸配列が、I332V、E343Q、G402S、T413E、E416D、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異を維持する、〔2〕の改変アルカリホスファターゼ。
〔4〕(B)または(C)のアミノ酸配列が、E343Q、I352T、G402S、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異を維持する、〔1〕の改変アルカリホスファターゼ。
〔5〕ウシ小腸由来アルカリホスファターゼをコードするアミノ酸配列において、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有する改変アルカリホスファターゼ。
〔6〕前記1個以上のアミノ酸残基の変異が、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異である、〔5〕の改変アルカリホスファターゼ。
〔7〕前記1個以上のアミノ酸残基の変異が、E343、I352、G402、およびE455からなる群より選ばれる1個以上のアミノ酸残基の変異である、〔5〕の改変アルカリホスファターゼ。
〔8〕ウシ小腸由来アルカリホスファターゼが以下(A)、(B)、または(C)のタンパク質である、〔5〕~〔7〕のいずれかの改変アルカリホスファターゼ:
(A)配列番号1のアミノ酸配列を含むタンパク質;
(B)配列番号1のアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号1のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~47個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
〔9〕改変アルカリホスファターゼが、改変アルカリホスファターゼをコードする部分に加えて、他の機能性ペプチドをコードする部分をさらに含むタンパク質である、〔1〕~〔8〕のいずれかの改変アルカリホスファターゼ。
〔10〕以下を含む、改変アルカリホスファターゼの製造方法:
ウシ小腸由来アルカリホスファターゼに対して、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を導入して、

〔19〕〔1〕~〔9〕のいずれかの改変アルカリホスファターゼ、または〔10〕の方法により製造された改変アルカリホスファターゼをコードするポリヌクレオチドの発現単位を含む形質転換細胞。
本発明の改変アルカリホスファターゼ変異体は、活性および熱安定性の双方に優れる。例えば、本発明の改変アルカリホスファターゼ変異体は、試薬(例、臨床検査試薬、実験試薬)として有用である。
1.改変アルカリホスファターゼ、または改変アルカリホスファターゼで標識された物質
(1)改変アルカリホスファターゼ
本発明は、以下(A)、(B)、または(C)のタンパク質である、改変アルカリホスファターゼを提供する:
(A)配列番号3、5、7、もしくは9のアミノ酸配列を含むタンパク質;
(B)配列番号3、5、7、もしくは9のアミノ酸配列に対して95%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号3、5、7、もしくは9のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~23個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
(1)改変アルカリホスファターゼ
本発明は、以下(A)、(B)、または(C)のタンパク質である、改変アルカリホスファターゼを提供する:
(A)配列番号3、5、7、もしくは9のアミノ酸配列を含むタンパク質;
(B)配列番号3、5、7、もしくは9のアミノ酸配列に対して95%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号3、5、7、もしくは9のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~23個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
配列番号3、5、7、および9のアミノ酸配列は、それぞれ、実施例に開示されるbIAPII変異体であるMut1、Mut2、Mut3、およびMut4のアミノ酸配列である。
上記アミノ酸配列についての同一性%は、95%以上である。好ましくは、同一性は、96%以上、97%以上、98%以上または99%以上であってもよい。このような同一性%の算出は、アルゴリズムblastpにより行うことができる。より具体的には、このような同一性の算定%は、National Center for Biotechnology Information(NCBI)において提供されているアルゴリズムblastpにおいて、デフォルト設定のScoring Parameters(Matrix:BLOSUM62;Gap Costs:Existence=11 Extension=1;Compositional Adjustments:Conditional compositional score matrix adjustment)を用いて行うことができる。
上記アミノ酸残基の変異は、アミノ酸残基の欠失、置換、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる。変異の個数は、改変アルカリホスファターゼの特性(例、活性、熱安定性)を損なわない程度の個数であり、1~23個である。変異の個数は、好ましくは1~20個、より好ましくは1~15個、さらにより好ましくは1~10個、特に好ましくは1~5個(例、1個、2個、3個、4個、または5個)である。
本発明の改変アルカリホスファターゼは、所望の同一性%および特性が維持される範囲内において1以上のアミノ酸残基の変異を有していてもよい。変異が導入されてもよいアミノ酸残基の位置は、当業者に明らかである。例えば、当業者は、1)同種の特性を有する複数のタンパク質のアミノ酸配列(例、配列番号3、5、7、および9)を比較し、2)相対的に保存されている領域、および相対的に保存されていない領域を明らかにし、次いで、3)相対的に保存されている領域および相対的に保存されていない領域から、それぞれ、機能に重要な役割を果たし得る領域および機能に重要な役割を果たし得ない領域を予測できるので、構造・機能の相関性を認識できる。したがって、当業者は、本発明の改変アルカリホスファターゼのアミノ酸配列において変異が導入されてもよいアミノ酸残基の位置を特定できる。このような位置における変異後のアミノ酸残基は、変異前のアミノ酸残基と異なる所望の天然α-アミノ酸残基である。このような所望の天然α-アミノ酸残基は、L-アラニン(A)、L-アスパラギン(N)、L-システイン(C)、L-グルタミン(Q)、L-イソロイシン(I)、L-ロイシン(L)、L-メチオニン(M)、L-フェニルアラニン(F)、L-プロリン(P)、L-セリン(S)、L-スレオニン(T)、L-トリプトファン(W)、L-チロシン(Y)、L-バリン(V)、L-アスパラギン酸(D)、L-グルタミン酸(E)、L-アルギニン(R)、L-ヒスチジン(H)、L-リジン(K)、またはグリシン(G)の残基である。
本発明の改変アルカリホスファターゼにおいて、アミノ酸残基の変異は、保存的置換であってもよい。用語「保存的置換」とは、所定のアミノ酸残基を、類似の側鎖を有するアミノ酸残基で置換することをいう。類似の側鎖を有するアミノ酸残基のファミリーは、当該分野で周知である。例えば、このようなファミリーとしては、塩基性側鎖を有するアミノ酸(例、リジン、アルギニン、ヒスチジン)、酸性側鎖を有するアミノ酸(例、アスパラギン酸、グルタミン酸)、非荷電性極性側鎖を有するアミノ酸(例、アスパラギン、グルタミン、セリン、スレオニン、チロシン、システイン)、非極性側鎖を有するアミノ酸(例、グリシン、アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファン)、β位分岐側鎖を有するアミノ酸(例、スレオニン、バリン、イソロイシン)、芳香族側鎖を有するアミノ酸(例、チロシン、フェニルアラニン、トリプトファン、ヒスチジン)、ヒドロキシル基(例、アルコール性、フェノール性)含有側鎖を有するアミノ酸(例、セリン、スレオニン、チロシン)、および硫黄含有側鎖を有するアミノ酸(例、システイン、メチオニン)が挙げられる。非荷電性極性側鎖を有するアミノ酸および非極性側鎖を有するアミノ酸を包括的に、中性アミノ酸と呼称することがある。好ましくは、アミノ酸の保存的置換は、アスパラギン酸とグルタミン酸との間での置換、アルギニンとリジンとヒスチジンとの間での置換、トリプトファンとフェニルアラニンとの間での置換、フェニルアラニンとバリンとの間での置換、ロイシンとイソロイシンとアラニンとの間での置換、およびグリシンとアラニンとの間での置換であってもよい。
脱リン酸化活性は、p-ニトロフェノールリン酸からp-ニトロフェノールへの変換活性を測定することにより決定することができる。例えば、(B)および(C)のアミノ酸配列を含む改変アルカリホスファターゼは、同一の条件下で活性の測定が行われた場合、上記(A)における対応配列番号のアミノ酸配列を含む改変アルカリホスファターゼを基準として20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上または95%以上の活性を有することが好ましい。このような条件として、アルカリホスファターゼの無細胞抽出液、ならびにジエタノールアミン、塩化マグネシウム、およびp-ニトロフェノールリン酸を含む反応液(例、pH8~11)を37℃で反応させた場合における1分あたりの吸光度(405nm)の変化を測定する条件を採用することができる(実施例3を参照)。
本発明の改変アルカリホスファターゼはまた、優れた熱安定性を有することができる。例えば、(B)および(C)のアミノ酸配列を含む改変アルカリホスファターゼは、同一の条件下で熱処理およびそれに引き続き活性の測定が行われた場合、上記(A)における対応配列番号のアミノ酸配列を含む改変アルカリホスファターゼを基準として10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上または95%以上の残存活性を有することが好ましい。このような条件として、アルカリホスファターゼの無細胞抽出液を70℃で10分間加熱処理する条件、およびその後に、加熱処理されたアルカリホスファターゼの無細胞抽出液、ならびにジエタノールアミン、塩化マグネシウム、およびp-ニトロフェノールリン酸を含む反応液(例、pH8~11)を37℃で反応させた場合における1分あたりの吸光度(405nm)の変化を測定する条件を採用することができる(実施例4を参照)。残存活性は、熱処理していないアルカリホスファターゼの無細胞抽出液について上記のとおり測定された変化を、熱処理していないアルカリホスファターゼの無細胞抽出液について同じく測定された変化に対する百分率として算出することにより決定することができる(実施例4を参照)。酵素の熱安定性と液状安定性および/または長期保存安定性は一般に相関し得るので、優れた熱安定性を有する本発明の改変アルカリホスファターゼは、液状安定性および/または長期保存安定性に優れるということができる。したがって、本発明の改変アルカリホスファターゼは、試薬として有用である。
特定の実施形態では、(B)または(C)のアミノ酸配列は、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異を維持していてもよい。維持されるアミノ酸残基の変異の個数は、1個であってもよく、2個以上、3個以上、4個以上、5個以上、6個以上、7個以上、または8個であってもよい。
好ましい実施形態では、(B)または(C)のアミノ酸配列は、上記変異のなかでも、I332V、E343Q、G402S、T413E、E416D、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異を維持していてもよい。維持されるアミノ酸残基の変異の個数は、1個であってもよく、また、2個以上、3個以上、4個以上、5個以上、または6個であってもよい。
より好ましい実施形態では、(B)または(C)のアミノ酸配列は、上記変異のなかでも、I332V、E343Q、G402S、T413E、およびE416Dからなる群より選ばれる1個以上のアミノ酸残基の変異を維持していてもよい。維持されるアミノ酸残基の変異の個数は、1個であってもよく、また、2個以上、3個以上、4個以上、または5個であってもよい。
さらにより好ましい実施形態では、(B)または(C)のアミノ酸配列は、上記変異のなかでも、I332V、G402S、T413E、およびE416Dからなる群より選ばれる1個以上のアミノ酸残基の変異を維持していてもよい。維持されるアミノ酸残基の変異の個数は、1個であってもよく、また、2個以上、3個以上、または4個であってもよい。
好ましい実施形態では、(B)または(C)のアミノ酸配列は、上記変異のなかでも、E343Q、I352T、G402S、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異を維持していてもよい。維持されるアミノ酸残基の変異の個数は、1個であってもよく、また、2個以上、3個以上、または4個であってもよい。
本発明はまた、ウシ小腸由来アルカリホスファターゼをコードするアミノ酸配列において、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有する改変アルカリホスファターゼを提供する。これらの部位における変異後のアミノ酸残基は、変異前のアミノ酸残基と異なる所望の天然α-アミノ酸残基である。このような所望の天然α-アミノ酸残基は、グリシン、L-アラニン(A)、L-アスパラギン(N)、L-システイン(C)、L-グルタミン(Q)、L-イソロイシン(I)、L-ロイシン(L)、L-メチオニン(M)、L-フェニルアラニン(F)、L-プロリン(P)、L-セリン(S)、L-スレオニン(T)、L-トリプトファン(W)、L-チロシン(Y)、L-バリン(V)、L-アスパラギン酸(D)、L-グルタミン酸(E)、L-アルギニン(R)、L-ヒスチジン(H)、およびL-リジン(K)からなる群より選ばれるアミノ酸残基である。好ましくは、上記変異は、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。このような改変アルカリホスファターゼのアミノ酸配列に含まれるアミノ酸残基の変異の個数は、1個であってもよく、また、2個以上、3個以上、4個以上、5個以上、6個以上、7個以上、または8個であってもよい。
好ましい実施形態では、上記変異は、I332、E343、G402、T413、E416、およびE455からなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。好ましくは、上記変異は、I332V、E343Q、G402S、T413E、E416D、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。変異されるアミノ酸残基の個数は、1個であってもよく、また、2個以上、3個以上、4個以上、5個以上、または6個であってもよい。

さらにより好ましい実施形態では、上記変異は、I332、G402、T413、およびE416からなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。好ましくは、上記変異は、I332V、G402S、T413E、およびE416Dからなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。変異されるアミノ酸残基の個数は、1個であってもよく、また、2個以上、3個以上、または4個であってもよい。
好ましい実施形態では、上記変異は、E343、I352、G402、およびE455からなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。好ましくは、上記変異は、I332V、G402S、T413E、およびE416Dからなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。変異されるアミノ酸残基の個数は、1個であってもよく、また、2個以上、3個以上、または4個であってもよい。
特定の実施形態では、ウシ小腸由来アルカリホスファターゼは、以下(A)、(B)、または(C)のタンパク質であってもよい:
(A)配列番号1のアミノ酸配列を含むタンパク質;
(B)配列番号1のアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号1のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~47個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
(A)配列番号1のアミノ酸配列を含むタンパク質;

上記アミノ酸配列についての同一性は、90%以上である。好ましくは、同一性は、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上または99%以上であってもよい。このような同一性%の算出は、上述したものと同様にして行うことができる。
上記アミノ酸残基の変異は、アミノ酸残基の欠失、置換、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる。変異の個数は、改変アルカリホスファターゼの特性(例、活性、熱安定性)を損なわない程度の個数であり、1~47個である。変異の個数は、好ましくは1~45個、より好ましくは1~40個、さらにより好ましくは1~30個、特に好ましくは1~20個、1~15個、1~10個、または1~5個(例、1個、2個、3個、4個、または5個)である。
本発明の改変アルカリホスファターゼは、所望の同一性%および特性が維持される範囲内において1以上のアミノ酸残基の変異を有していてもよい。変異が導入されてもよいアミノ酸残基の位置は、当業者に明らかである。例えば、当業者は、1)同種の特性を有する複数のタンパク質のアミノ酸配列を比較し〔例えば、配列番号1のアミノ酸配列(ウシ小腸由来アルカリホスファターゼ)と他のウシ小腸由来アルカリホスファターゼ(例、ウシ小腸由来アルカリホスファターゼI、III、IV、V、VI、および/またはVII)をコードするアミノ酸配列との比較、あるいは配列番号1、3、5、7、および9のアミノ酸配列の比較〕、2)相対的に保存されている領域、および相対的に保存されていない領域を明らかにし、次いで、3)相対的に保存されている領域および相対的に保存されていない領域から、それぞれ、機能に重要な役割を果たし得る領域および機能に重要な役割を果たし得ない領域を予測できるので、構造・機能の相関性を認識できる。したがって、当業者は、本発明の改変アルカリホスファターゼのアミノ酸配列において変異が導入されてもよいアミノ酸残基の位置を特定できる。このような位置における変異後のアミノ酸残基は、変異前のアミノ酸残基と異なる所望の天然α-アミノ酸残基である。このような所望の天然α-アミノ酸残基は、上述したものと同様である。アミノ酸残基の変異は、上述したような保存的置換であってもよい。また、脱リン酸化活性、および熱安定性についても、上述したものと同様である。
本発明の改変アルカリホスファターゼは、改変アルカリホスファターゼをコードする部分に加えて、他の機能性ペプチドをコードする部分をさらに含む融合タンパク質であってもよい。例えば、本発明の改変アルカリホスファターゼは、形質転換細胞からの細胞外への分泌を促進するため、N末端にシグナル配列を有していてもよい。このようなシグナル配列としては、例えば、MglBシグナル配列、PelBシグナル配列、OmpAシグナル配列、PhoAシグナル配列、OmpFシグナル配列、PhoEシグナル配列、MalEシグナル配列、OmpCシグナル配列、Lppシグナル配列およびLamBシグナル配列が挙げられる。本発明の改変アルカリホスファターゼはまた、C末端もしくはN末端、またはその双方(好ましくはC末端)に、他のペプチド成分(例、タグ部分)を有していてもよい。本発明の改変アルカリホスファターゼに付加され得る他のペプチド成分としては、例えば、目的タンパク質の精製を容易にするペプチド成分(例、ヒスチジンタグ、Strep-tag II等のタグ部分;グルタチオン-S-トランスフェラーゼ、マルトース結合タンパク質等の目的タンパク質の精製に汎用されるタンパク質)、目的タンパク質の可溶性を向上させるペプチド成分(例、Nus-tag)、シャペロンとして働くペプチド成分(例、トリガーファクター)、他の機能をもつタンパク質あるいはタンパク質のドメインあるいはそれらとをつなぐリンカーとしてのペプチド成分が挙げられる。
特定の実施形態では、本発明の融合タンパク質は、他の機能性ペプチドをコードする部分として、親和性ペプチドまたは抗原性ペプチドをコードする部分を含む融合タンパク質であってもよい。このような親和性ペプチドの標的分子としては、オリゴペプチドもしくはポリペプチド(すなわち、タンパク質)、オリゴ糖もしくは多糖、核酸または(例、DNA、RNA、または人工核酸)、脂質、低分子化合物(例、アミノ酸、単糖、ヌクレオチド、ヌクレオシド、血液等の体液中に存在するその他の生体成分、医薬)が挙げられる。親和性ペプチドとしては、例えば、後述するような抗体またはその断片(例、単鎖抗体)、抗体ミメティックが挙げられる。抗原性ペプチドとしては、例えば、ウイルス抗原性ペプチド、アレルゲン性ペプチド、腫瘍特異抗原ペプチド、腫瘍関連抗原ペプチド、細菌抗原性ペプチド、真菌抗原性ペプチド、寄生虫抗原性ペプチドが挙げられる。
本発明はまた、以下を含む、改変アルカリホスファターゼの製造方法を提供する:
ウシ小腸由来アルカリホスファターゼに対して、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を導入して、
ウシ小腸由来アルカリホスファターゼをコードするアミノ酸配列において、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有する改変アルカリホスファターゼを生成すること。
ウシ小腸由来アルカリホスファターゼに対して、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を導入して、
ウシ小腸由来アルカリホスファターゼをコードするアミノ酸配列において、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有する改変アルカリホスファターゼを生成すること。
ウシ小腸由来アルカリホスファターゼは、天然型アルカリホスファターゼであってもよく、また、変異型アルカリホスファターゼであってもよい。本発明で利用されるウシ小腸由来アルカリホスファターゼは、上記のような1個以上の部位において目的の変異を有しないアルカリホスファターゼである。好ましくは、本発明で利用されるウシ小腸由来アルカリホスファターゼは、上記のような2個以上(例、2個、3個、4個、5個、6個、7個、または8個)の部位において目的の変異を有しないアルカリホスファターゼであってもよい。
これらの部位に導入されるアミノ酸残基の変異は、変異前のアミノ酸残基と異なる所望の天然α-アミノ酸残基の変異である。このような所望の天然α-アミノ酸残基は、上述したものと同様である。好ましくは、導入される変異は、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異であってもよい。導入される変異の個数は、1個であってもよく、また、2個以上、3個以上、4個以上、5個以上、6個以上、7個以上、または8個であってもよい。変異の導入は、所望の部位に変異を導入できる任意の方法(例、部位特異的変異誘発法)により行うことができる。

(C)配列番号1のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~47個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。
配列番号1のアミノ酸配列に対する同一性%の程度、および配列番号1のアミノ酸配列におけるアミノ酸残基の変異の個数は、上述したものと同様である。
本発明では、上述したような改変アルカリホスファターゼの製造方法により得られる改変アルカリホスファターゼもまた提供される。
(2)改変アルカリホスファターゼで標識された物質
本発明はまた、改変アルカリホスファターゼで標識された物質を提供する。

改変アルカリホスファターゼで標識されるべき物質は、任意の標的分子に対する親和性物質、または抗原である。このような親和性物質の標的分子としては、オリゴペプチドもしくはポリペプチド(すなわち、タンパク質)、オリゴ糖もしくは多糖、核酸または(例、DNA、RNA、または人工核酸)、脂質、低分子化合物(例、アミノ酸、単糖、ヌクレオチド、ヌクレオシド、血液等の体液中に存在するその他の生体成分、医薬)が挙げられる。親和性物質としては、例えば、抗体、アプタマー、レクチン、核酸の相補鎖、抗体ミメティックが挙げられる。アルカリホスファターゼで標識された親和性物質としては、種々のものが報告されている(例、Berg et al.,Labeling Antibodies,http://cshprotocols.cshlp.org/content/2020/7/pdb.top099242.full、Suzuki et al., Open sandwich ELISA with VH-/VL-alkaline phosphatase fusion proteins, https://doi.org/10.1016/S0022-1759(99)00020-4を参照)。抗原としては、例えば、ウイルス抗原、アレルゲン、糖鎖抗原、腫瘍特異抗原、腫瘍関連抗原、細菌抗原、真菌抗原、寄生虫抗原が挙げられる。アルカリホスファターゼで標識された抗原としては、種々のものが報告されている(例、特許第2525195号公報を参照)。
特定の実施形態では、親和性物質は、抗体であってもよい。抗体は、ポリクローナル抗体、またはモノクローナル抗体であってもよい。抗体のアイソタイプとしては、例えば、IgG(例、IgG1、IgG2、IgG3、IgG4)、IgM、IgA、IgD、IgE、およびIgYが挙げられる。抗体は、全長抗体、または抗体断片(例、F(ab’)2、Fab’、Fab、Fv、単鎖抗体)であってもよい。改変アルカリホスファターゼで標識された抗体としては、種々のものが報告されている(例、Berg et al.,Labeling Antibodies,http://cshprotocols.cshlp.org/content/2020/7/pdb.top099242.full、およびSuzuki et al., Open sandwich ELISA with VH-/VL-alkaline phosphatase fusion proteins, https://doi.org/10.1016/S0022-1759(99)00020-4)を参照)。
本発明の改変アルカリホスファターゼによる物質の標識は、アルカリホスファターゼによる物質の標識と同様にして行うことができる。アルカリホスファターゼによる物質の標識は慣用されているため、当業者は、本発明の改変アルカリホスファターゼにより物質を適宜標識することができる。アルカリホスファターゼによる物質の標識としては、例えば、化学標識および酵素標識が挙げられる。アルカリホスファターゼによる物質の化学標識としては、例えば、グルタルアルデヒドを架橋剤として使用する方法(例、Winston et al.,Conjugation of Enzymes to Antibodies,https://currentprotocols.onlinelibrary.wiley.com/doi/full/10.1002/0471142727.mb1101s50)、マレイミドを架橋剤として使用する方法(例、Watabe et al.,Ultrasensitive enzyme-linked immunosorbent assay (ELISA) of proteins by combination with the thio-NAD cycling method,Biophysics(Nagoya-shi),2014,10,49-54;特許第5265816号公報)を利用することができる。アルカリホスファターゼによる物質の酵素標識としては、例えば、ソルターゼAを結合酵素として使用する方法(例、Sakamoto et al.,Enzyme-Mediated Site-Specific Antibody-Protein Modification Using a ZZ Domain as a Linker,https://pubs.acs.org/doi/10.1021/bc100206z)、リポ酸リガーゼを結合酵素として使用する方法(例、Cohen et al.,Site-specific protein modification using lipoic acid ligase and bis-aryl hydrazone formation, https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4758125/)が挙げられる。
本発明の改変アルカリホスファターゼで標識された物質は、測定対象物質の検出または定量に有用である。臨床検査では、物質として親和性物質を用いる標的分子を検出または定量する免疫学的方法が汎用されている。例えば、親和性物質が抗体であり、かつ当該抗体の標的分子(抗原)が測定対象物質である場合、本発明の改変アルカリホスファターゼで標識された抗体は、測定対象物質に結合できる検出抗体(例、測定対象物質に対する固相抗体および当該測定対象物質に対する標識抗体を含む少なくとも2種の抗体を用いるサンドイッチアッセイにおける標識抗体、および非サンドイッチアッセイにおける標識抗体)として使用することができる。一方、親和性物質が抗体であり、かつ当該抗体の標的分子が測定対象物質に対する別の抗体である場合、本発明の改変アルカリホスファターゼで標識された抗体は、測定対象物質に対する別の抗体に結合できる二次抗体として使用することができる。物質が抗体の場合も同様に標的分子を検出または定量する免疫学的方法が汎用されている。
2.改変アルカリホスファターゼの製造に関連し得る発明
本発明の改変アルカリホスファターゼは、本発明の改変アルカリホスファターゼをコードするポリヌクレオチド、およびそれに作動可能に連結されたプロモーターを含む発現単位を含む形質転換細胞を用いて、または無細胞系等を用いて、調製することができる。本発明はまた、このようなポリヌクレオチドおよび形質転換細胞、ならびに形質転換体の作製に利用できる発現ベクターを提供する。
本発明の改変アルカリホスファターゼは、本発明の改変アルカリホスファターゼをコードするポリヌクレオチド、およびそれに作動可能に連結されたプロモーターを含む発現単位を含む形質転換細胞を用いて、または無細胞系等を用いて、調製することができる。本発明はまた、このようなポリヌクレオチドおよび形質転換細胞、ならびに形質転換体の作製に利用できる発現ベクターを提供する。
本発明のポリヌクレオチドは、本発明の改変アルカリホスファターゼをコードするポリヌクレオチドである。本発明のポリヌクレオチドは、DNAまたはRNAであってもよいが、DNAが好ましい。
本発明の形質転換細胞は、例えば、本発明のポリヌクレオチドを含む発現ベクターを用いる方法(例、コンピテント細胞法、エレクトロポレーション法)、またはゲノム改変技術により作製することができる。発現ベクターが宿主細胞のゲノムDNAと相同組換えを生じる組込み型(integrative)ベクターである場合、発現単位は、形質転換により、宿主細胞のゲノムDNAに組み込まれることができる。一方、発現ベクターが宿主細胞のゲノムDNAと相同組換えを生じない非組込み型ベクターである場合、発現単位は、形質転換により、宿主細胞のゲノムDNAに組み込まれず、宿主細胞内において、発現ベクターの状態のまま、ゲノムDNAから独立して存在できる。あるいは、ゲノム編集技術(例、CRISPR/Casシステム、Transcription Activator-Like Effector Nucleases(TALEN))によれば、発現単位を宿主細胞のゲノムDNAに組み込むこと、および宿主細胞が固有に備える発現単位を改変することが可能である。
本発明はまた、本発明のポリヌクレオチドを含む発現ベクターを提供する。本発明の発現ベクターは、宿主細胞で機能するターミネーター、リボゾーム結合部位、および薬剤耐性遺伝子等のエレメントをさらに含んでいてもよい。薬剤耐性遺伝子としては、例えば、テトラサイクリン、アンピシリン、カナマイシン、ハイグロマイシン、ホスフィノスリシン等の薬剤に対する耐性遺伝子が挙げられる。
発現ベクターはまた、宿主細胞のゲノムDNAとの相同組換えのために、宿主細胞のゲノムとの相同組換えを可能にする領域をさらに含んでいてもよい。例えば、発現ベクターは、それに含まれる発現単位が一対の相同領域(例、宿主細胞のゲノム中の特定配列に対して相同なホモロジーアーム、loxP、FRT)間に位置するように設計されてもよい。発現単位が導入されるべき宿主細胞のゲノム領域(相同領域の標的)としては、特に限定されないが、宿主細胞において発現量が多い遺伝子のローカスであってもよい。
発現ベクターは、プラスミド、ウイルスベクター、ファージ、または人工染色体であってもよい。発現ベクターはまた、組込み型(integrative)ベクターであっても非組込み型ベクターであってもよい。組込み型ベクターは、その全体が宿主細胞のゲノムに組み込まれるタイプのベクターであってもよい。あるいは、組込み型ベクターは、その一部(例、発現単位)のみが宿主細胞のゲノムに組み込まれるタイプのベクターであってもよい。発現ベクターはさらに、DNAベクター、またはRNAベクター(例、レトロウイルス)であってもよい。発現ベクターはまた、汎用されている発現ベクターであってもよい。このような発現ベクターとしては、例えば、pUC(例、pUC19、pUC18)、pSTV、pBR(例、pBR322)、pHSG(例、pHSG299、pHSG298、pHSG399、pHSG398)、RSF(例、RSF1010)、pACYC(例、pACYC177、pACYC184)、pMW(例、pMW119、pMW118、pMW219、pMW218)、pQE(例、pQE30)、およびその誘導体が挙げられる。
本発明の改変アルカリホスファターゼを発現させるための宿主としては、例えばエシェリヒア・コリ(Escherichia coli)等のエシェリヒア属細菌、コリネバクテリウム属細菌〔例、コリネバクテリウム・グルタミカム(Corynebacterium glutamicum)〕、およびバチルス属細菌〔例、バチルス・ズブチリス(Bacillus subtilis)〕をはじめとする種々の原核細胞、サッカロマイセス属細菌〔例、サッカロマイセス・セレビシエ(Saccharomyces cerevisiae)〕、ピヒア属細菌〔例、ピヒア・スティピティス(Pichia stipitis)〕、アスペルギルス属細菌〔例、アスペルギルス・オリゼ(Aspergillus oryzae)〕をはじめとする種々の真核細胞を用いることができる。あるいは、宿主としては、昆虫細胞、植物細胞、動物細胞(例、チャイニーズハムスター卵巣(CHO)細胞等の哺乳動物細胞)を用いることができる。宿主としては、所定の遺伝子を欠損する株を用いてもよい。形質転換細胞としては、例えば、細胞質中に発現ベクターを保有する形質転換細胞、およびゲノム上に目的遺伝子が導入された形質転換細胞が挙げられる。
本発明の形質転換細胞は、所定の培養装置(例、試験管、フラスコ、ジャーファーメンター)を用いて、例えば後述の組成を有する培地において培養することができる。培養条件は適宜設定することができる。具体的には、培養温度は10℃~37℃であってもよく、pHは6.5~7.5であってもよく、培養時間は1h~100hであってもよい。また、溶存酸素濃度を管理しつつ培養を行っても良い。この場合、培養液中の溶存酸素濃度(DO値)を制御の指標として用いることがある。大気中の酸素濃度を21%とした場合の相対的な溶存酸素濃度DO値が、例えば1~10%を、好ましくは3%~8%を下回らない様に、通気・攪拌条件を制御することが出来る。また、培養はバッチ培養であっても、フェドバッチ培養であっても良い。フェドバッチ培養の場合は糖源となる溶液やリン酸を含む溶液を培養液に連続的あるいは不連続的に逐次添加して、培養を継続することもできる。
形質転換される宿主は、上述したとおりであるが、大腸菌について詳述すると、大腸菌K12株亜種のエシェリヒア コリ JM109株、DH5α株、HB101株、BL21(DE3)株などから選択することが出来る。形質転換を行う方法、および形質転換細胞を選別する方法は、Molecular Cloning: A Laboratory Manual,3rd edition,Cold Spring Harbor press(2001/01/15)などにも記載されている。以下、形質転換された大腸菌を作製し、これを用いて所定の酵素を製造する方法を、一例としてより具体的に説明する。
本発明のポリヌクレオチドを発現させるプロモーターとしては、通常E.coliにおける異種タンパク質生産に用いられるプロモーターを使用することができ、例えば、PhoA、PhoC、T7プロモーター、lacプロモーター、trpプロモーター、trcプロモーター、tacプロモーター、ラムダファージのPRプロモーター、PLプロモーター、T5プロモーター等の強力なプロモーターが挙げられ、PhoA、PhoC、lacが好ましい。また、ベクターとしては、例えば、pUC(例、pUC19、pUC18)、pSTV、pBR(例、pBR322)、pHSG(例、pHSG299、pHSG298、pHSG399、pHSG398)、RSF(例、RSF1010)、pACYC(例、pACYC177、pACYC184)、pMW(例、pMW119、pMW118、pMW219、pMW218)、pQE(例、pQE30)、およびその誘導体等を用いてもよい。他のベクターとしては、ファージDNAのベクターを利用してもよい。さらに、プロモーターを含み、挿入DNA配列を発現させることができる発現ベクターを使用してもよい。好ましくは、ベクターは、pUC、pSTV、pMWであってもよい。
また、本発明のポリヌクレオチドの下流に転写終結配列であるターミネーターを連結してもよい。このようなターミネーターとしては、例えば、T7ターミネーター、fdファージターミネーター、T4ターミネーター、テトラサイクリン耐性遺伝子のターミネーター、大腸菌trpA遺伝子のターミネーターが挙げられる。
本発明のポリヌクレオチドを大腸菌に導入するためのベクターとしては、いわゆるマルチコピー型のものが好ましく、ColE1由来の複製開始点を有するプラスミド、例えばpUC系のプラスミドやpBR322系のプラスミドあるいはその誘導体が挙げられる。ここで、「誘導体」とは、塩基の置換、欠失、挿入および/または付加などによってプラスミドに改変を施したものを意味する。
また、形質転換細胞を選別するために、ベクターがアンピシリン耐性遺伝子等のマーカーを有することが好ましい。このようなプラスミドとして、強力なプロモーターを持つ発現ベクターが市販されている〔例、pUC系(タカラバイオ社製)、pPROK系(クローンテック製)、pKK233-2(クローンテック製)〕。
得られた本発明の発現ベクターを用いて大腸菌を形質転換し、この大腸菌を培養することにより、本発明の改変アルカリホスファターゼを得ることができる。
培地としては、M9-カザミノ酸培地、LB培地など、大腸菌を培養するために通常用いる培地を用いてもよい。培地は、所定の炭素源、窒素源、補酵素(例、塩酸ピリドキシン)を含有していてもよい。具体的には、ペプトン、酵母エキス、NaCl、グルコース、MgSO4、硫酸アンモニウム、リン酸2水素カリウム、硫酸第二鉄、硫酸マンガン、などを用いても良い。また、培養条件、生産誘導条件は、用いたベクターのマーカー、プロモーター、宿主菌等の種類に応じて適宜選択される。
本発明の改変アルカリホスファターゼを回収するには、以下の方法などがある。本発明の改変アルカリホスファターゼは、本発明の形質転換細胞を回収した後、菌体を破砕(例、ソニケーション、ホモジナイゼーション)あるいは溶解(例、リゾチーム処理)することにより、破砕物および溶解物として得ることができる。アルカリホスファターゼが細胞外へ分泌・漏出した場合は、培養液から遠心分離や膜ろ過により、アルカリホスファターゼを含む除菌液を取得することができる。このような破砕物、溶解物および除菌液を、抽出、沈澱、濾過、カラムクロマトグラフィー等の手法に供することにより、本発明の改変アルカリホスファターゼを得ることができる。
3.改変アルカリホスファターゼ、または改変アルカリホスファターゼで標識された物質を用いる方法
(1)アルカリホスファターゼ活性の測定
本発明は、本発明の改変アルカリホスファターゼまたは改変アルカリホスファターゼで標識された物質、および基質を用いて、アルカリホスファターゼ活性を測定することを含む、アルカリホスファターゼ活性の測定方法を提供する。

本発明は、本発明の改変アルカリホスファターゼまたは改変アルカリホスファターゼで標識された物質、および基質を用いて、アルカリホスファターゼ活性を測定することを含む、アルカリホスファターゼ活性の測定方法を提供する。
基質としては、アルカリホスファターゼ活性の測定に利用できる任意の基質を用いることができる。このような基質としては、例えば、リン酸化物質〔例、p-ニトロフェノールリン酸、5-ブロモ-4-クロロ-3-インドリルリン酸、2-クロロ-5-(4-メトキシスピロ[1,2-ジオキセタン-3,2′-(5-クロロトリシクロ[3.3.1.13.7]デカン])-4-イル]-1-フェニルリン酸、リン酸ナフトールAS-TR、フェノールフタレインリン酸、インドキシルリン酸、ナフトールAS-BIリン酸、ナフトールAS-Eリン酸、ナフトールAS-MX リン酸、ビス(p-ニトロフェニル)リン酸、フェノールフタレインビスリン酸、5-ブロモ-4-クロロ-3-インドリルリン酸、1-ナフチルリン酸、2-ナフチルリン酸、4-メチルウンベリフェリルリン酸、1,2-ジオキセタンもくしはアクリダンを基本骨格に含んでなる発光基質(例、CDP-star、APS-5、Lumi-Phos530、AMPPD、Lumigen PPD)、核酸(例、DNA、RNA、人工核酸)〕が挙げられる。基質は、例えば0.1~100mM、好ましくは1~30mMの濃度で使用することができる。例えば、アルカリホスファターゼ活性の測定条件として、アルカリ条件下(例、pH8~11)、適温(例、4~40℃)で脱リン酸化物質の指標値(例、吸光度)を測定する条件を採用することができる。アルカリホスファターゼ活性の測定は、例えば、親和性物質の標的分子を測定するアッセイ(例、親和性物質として抗体を用いる免疫学的アッセイ)、レポーターアッセイなどの生物学的アッセイに有用である。
特定の実施形態では、アルカリホスファターゼ活性の測定方法は、本発明の改変アルカリホスファターゼで標識された物質を用いて標的分子を検出または定量する方法であってもよい。改変アルカリホスファターゼにより標識された物質およびその標的分子は、上述したものと同様である。例えば、本発明の改変アルカリホスファターゼにより標識された物質は、上述したように、測定対象物質に結合できる検出抗体、または測定対象物質に対する別の抗体に結合できる二次抗体として使用することができる。したがって、アルカリホスファターゼ活性の測定により、測定対象物質を検出または定量することができる。
(2)リン酸化物質の脱リン酸化
本発明は、本発明の改変アルカリホスファターゼまたは改変アルカリホスファターゼに標識された物質を用いて、リン酸化物質を脱リン酸化して脱リン酸化物質を生成することを含む、脱リン酸化物質の製造方法を提供する。
本発明は、本発明の改変アルカリホスファターゼまたは改変アルカリホスファターゼに標識された物質を用いて、リン酸化物質を脱リン酸化して脱リン酸化物質を生成することを含む、脱リン酸化物質の製造方法を提供する。
リン酸化物質の脱リン酸化では、上述したようなアルカリホスファターゼ活性の測定条件下で上述のリン酸化物質を改変アルカリホスファターゼまたは改変アルカリホスファターゼに標識された物質と反応させることにより、リン酸化物質から脱リン酸化物質を生成することができる。
特定の実施形態では、リン酸化物質の脱リン酸化は、核酸の5’または3’末端リン酸の脱リン酸化であってもよい。核酸は、一本鎖核酸であっても二本鎖核酸(突出末端もしくは平滑末端)であってもよい。このような方法によれば、所望されない核酸の連結(例、ベクターのセルフライゲーション)を防止することができる。
以下の実施例により本発明をより詳細に説明するが、本発明は以下の実施例に限定されるものではない。
実施例1:アルカリホスファターゼの設計及び発現プラスミドの作製
本検討に用いたプラスミド(発現ベクター)を、それぞれ以下に示す方法で作製した。

bIAPII(AAN19391)を鋳型とし、Blastpから類似配列を5000個取得した。なおBlastpの解析条件として、Max target sequencesを5000,Expected thresholdの値を1.0E-6に設定した。取得した類似配列を解析し、鋳型配列と類似した配列等を除去し、1161配列まで選抜した。これを配列ライブラリーとした。その後文献1(Nakano, S., Motoyama, T., Miyashita, Y., Ishizuka, Y., Matsuo, N., Tokiwa, H., Shinoda, S., Asano, Y., and Ito, S. (2018) Benchmark Analysis of Native and Artificial NAD+-Dependent Enzymes Generated by a Sequence-Based Design Method with or without Phylogenetic Data, Biochemistry 57, 3722-3732.) 及び2 (Nakano, S., Niwa, M., Asano, Y., and Ito, S. (2019) Following the Evolutionary Track of a Highly Specific l-Arginine Oxidase by Reconstruction and Biochemical Analysis of Ancestral and Native Enzymes, Appl Environ Microbiol 85, e00459-00419.)で用いた手法を適用することで、bIAPII(AAN19391)のモチーフ様配列(I54、D86、A93、S113、T163、P303、A324、L351)を同定した。配列ライブラリーにおいて、これらモチーフ様配列を有する配列のみを選抜、さらにClustalWで解析し、相同性の低い配列やギャップを含む配列を除去し、最終的に10配列を得た。10配列を用いて人工設計を行い、Mut1、Mut2、Mut3、およびMut4と名づけた配列番号3、5、7、9のアミノ酸配列で示される人工アルカリホスファターゼ配列を設計した(図2~5)。
(A)bIAPII発現プラスミド
ウシ小腸由来アルカリホスファターゼ(bIAPII)からN末端の分泌シグナル配列およびC末端のGPIアンカー領域を除いたアミノ酸配列(配列番号1)をE.coliのコドンに最適化して塩基配列に変換し、本塩基配列(配列番号2)を含むプラスミドをユーロフィンジェノミクス社で合成した。本プラスミドを鋳型として、bIAPIIをコードする領域を配列番号11および配列番号12のプライマーを用いたPCRにより増幅し、得られた断片をIn-fusion HD cloning kit(タカラバイオ)により、NcoIおよびXhoIで消化したpET-28a(+)ベクターにクローニングした。
ウシ小腸由来アルカリホスファターゼ(bIAPII)からN末端の分泌シグナル配列およびC末端のGPIアンカー領域を除いたアミノ酸配列(配列番号1)をE.coliのコドンに最適化して塩基配列に変換し、本塩基配列(配列番号2)を含むプラスミドをユーロフィンジェノミクス社で合成した。本プラスミドを鋳型として、bIAPIIをコードする領域を配列番号11および配列番号12のプライマーを用いたPCRにより増幅し、得られた断片をIn-fusion HD cloning kit(タカラバイオ)により、NcoIおよびXhoIで消化したpET-28a(+)ベクターにクローニングした。
(B)MglBシグナル配列を付加したbIAPII発現プラスミド
配列番号13および配列番号14のプライマーを用いて、(A)で作製したプラスミドを鋳型として、PCRによりbIAPIIをコードする領域を増幅し、インサート断片を調製した。また、配列番号15および配列番号16のプライマーを用いてpET-28a(+)ベクターを鋳型として、PCRによりベクター断片を調製した。配列番号16にはMglBシグナル配列(配列番号17)に相当する塩基配列を含んでいる。調製したインサートおよびベクター断片をIn-fusion HD cloning kitにより連結した。得られたプラスミドにより、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたbIAPIIが発現する。
配列番号13および配列番号14のプライマーを用いて、(A)で作製したプラスミドを鋳型として、PCRによりbIAPIIをコードする領域を増幅し、インサート断片を調製した。また、配列番号15および配列番号16のプライマーを用いてpET-28a(+)ベクターを鋳型として、PCRによりベクター断片を調製した。配列番号16にはMglBシグナル配列(配列番号17)に相当する塩基配列を含んでいる。調製したインサートおよびベクター断片をIn-fusion HD cloning kitにより連結した。得られたプラスミドにより、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたbIAPIIが発現する。
(C)MglBシグナル配列を付加した人工設計ALP発現プラスミド
Mut1(配列番号3)をコードする塩基配列(配列番号4)をpET-28a(+)ベクターのNcoI/XhoIサイトに挿入したプラスミドをユーロフィンジェノミクス社で合成した。本プラスミドを鋳型として配列番号18および配列番号16のプライマーを用いて、PCRにより直鎖DNAを増幅し、In-fusion HD cloning kitにより環状化した。本プラスミドでは、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたMut1が発現する。

同様に、配列番号18のプライマーの代わりに配列番号19~21のプライマーを用いて、Mut2(配列番号5)、Mut3(配列番号7)およびMut4(配列番号9)を発現するプラスミドを構築した。それぞれをコードする塩基配列は、配列番号6、配列番号8および配列番号10である。
天然アルカリホスファターゼであるbIAPII(配列番号1)、ならびに人工設計アルカリホスファターゼであるMut1(配列番号3)、Mut2(配列番号5)、Mut3(配列番号7)、およびMut4(配列番号9)のアミノ酸配列同一性の関係は、以下のとおりである。
次に、天然アルカリホスファターゼであるbIAPII(配列番号1)では認められず、人工設計アルカリホスファターゼであるMut1(配列番号3)、Mut2(配列番号5)、Mut3(配列番号7)、Mut4(配列番号9)間で保存されているアミノ酸残基の共通変異をアミノ酸配列のアライメントにより解析した。その結果、このような共通変異は、G105A、R108K、I133M、A142S、A159T、L174M、N181E、A188T、V191I、M205K、V223Q、Q231R、A257S、S260P、N277E、Q280R、T287S、Q297R、I332V、N342S、E343Q、I352T、G402S、T413E、E416D、E455Q、I461V、およびG466Aであった(表2)。
実施例2:無細胞抽出液の調製
実施例1の(B)および(C)で構築したプラスミドをE.coli BL21(DE3)を形質転換し、各種ALP発現株を得た。各発現株をカナマイシン50mg/Lを含むLB寒天培地で37℃、一晩生育させた。得られた菌体をカナマイシン50mg/L、硫酸マグネシウム5mMおよび塩化亜鉛10μMを含む4×TB培地100mLに植菌し、坂口フラスコを用いて37℃で振とう培養を行った。OD660が0.6~0.7となった時に終濃度で0.1mMとなるようにIPTGを添加して発現を誘導し、25℃で18時間培養した。
実施例1の(B)および(C)で構築したプラスミドをE.coli BL21(DE3)を形質転換し、各種ALP発現株を得た。各発現株をカナマイシン50mg/Lを含むLB寒天培地で37℃、一晩生育させた。得られた菌体をカナマイシン50mg/L、硫酸マグネシウム5mMおよび塩化亜鉛10μMを含む4×TB培地100mLに植菌し、坂口フラスコを用いて37℃で振とう培養を行った。OD660が0.6~0.7となった時に終濃度で0.1mMとなるようにIPTGを添加して発現を誘導し、25℃で18時間培養した。
培養終了後、得られた培養液1mLから菌体を遠心分離により集めた。次に100μLのxTractor buffer(タカラバイオ)に懸濁し、終濃度1mMとなるように硫酸マグネシウムを、100μMとなるように塩化亜鉛を添加し、室温で20分間インキュベートすることで溶菌した。遠心分離により破砕液から菌体残渣を除き、得られた上清を無細胞抽出液とした。
実施例3:無細胞抽出液の活性測定
無細胞抽出液のALP活性をp-ニトロフェノールリン酸からp-ニトロフェノールへの変換活性で測定した。まず、希釈用緩衝液(100mM Tris-HCl、150mM 塩化ナトリウム、1mM硫酸マグネシウム、0.05mM塩化亜鉛、0.1%ウシ血清アルブミン、pH7.0)で無細胞抽出液の希釈系列を調製した。反応液として、A液(ジエタノールアミン1000mM、0.5mM塩化マグネシウム、pH9.8)およびB液(p-ニトロフェノールリン酸670mM)を調製しておき、測定前にA:B=48:1の比率で混合した。反応液を37℃で10分以上保温したのち、96穴プレートに196μLずつ分注した。無細胞抽出液の希釈系列を4μLずつ反応液に添加し、混合後、37℃でマイクロプレートリーダーSpectraMax 190(モレキュラーデバイス)で405nmの吸光度を測定し、1分あたりの変化を算出した(ΔAtest)。酵素ブランクとして、無細胞抽出液の代わりにA液を添加して同様に算出した(ΔAblank)。
無細胞抽出液のALP活性をp-ニトロフェノールリン酸からp-ニトロフェノールへの変換活性で測定した。まず、希釈用緩衝液(100mM Tris-HCl、150mM 塩化ナトリウム、1mM硫酸マグネシウム、0.05mM塩化亜鉛、0.1%ウシ血清アルブミン、pH7.0)で無細胞抽出液の希釈系列を調製した。反応液として、A液(ジエタノールアミン1000mM、0.5mM塩化マグネシウム、pH9.8)およびB液(p-ニトロフェノールリン酸670mM)を調製しておき、測定前にA:B=48:1の比率で混合した。反応液を37℃で10分以上保温したのち、96穴プレートに196μLずつ分注した。無細胞抽出液の希釈系列を4μLずつ反応液に添加し、混合後、37℃でマイクロプレートリーダーSpectraMax 190(モレキュラーデバイス)で405nmの吸光度を測定し、1分あたりの変化を算出した(ΔAtest)。酵素ブランクとして、無細胞抽出液の代わりにA液を添加して同様に算出した(ΔAblank)。
1分間に1μmolのp-ニトロフェノールを生成する酵素量を1Uと定義し、以下の式で無細胞抽出液の活性を測定した。

各測定は3回ずつ実施した。得られた無細胞抽出液のALP活性を表3に示す。人工設計ALP発現株の無細胞抽出液は、bIAPII発現株の場合に比べて、高いALP活性を示した(表3)。
実施例4:熱安定性の評価
実施例3で測定した測定値に基づき、実施例2で調製した各無細胞抽出液の活性が300U/Lとなるように実施例3の希釈用緩衝液で希釈した。次に酵素希釈液を20μLずつマイクロチューブに分注し、TaKaRa PCR Thermal Cycler Dice(タカラバイオ)を用いて70℃で10分間加熱処理し、氷上に移して冷却した。熱処理していない酵素希釈液、および熱処理後の酵素希釈液のそれぞれを実施例3と同様に3回ずつALP活性を測定した。
実施例3で測定した測定値に基づき、実施例2で調製した各無細胞抽出液の活性が300U/Lとなるように実施例3の希釈用緩衝液で希釈した。次に酵素希釈液を20μLずつマイクロチューブに分注し、TaKaRa PCR Thermal Cycler Dice(タカラバイオ)を用いて70℃で10分間加熱処理し、氷上に移して冷却した。熱処理していない酵素希釈液、および熱処理後の酵素希釈液のそれぞれを実施例3と同様に3回ずつALP活性を測定した。
以下の式で残存活性を算出した。

その結果、人工設計ALPはbIAPIIと比較して、高い熱安定性を示した(表4)。
〔比較例1〕無細胞抽出液の調製および熱安定性の評価
比較のためにMut1、Mut2、Mut3およびMut4に含まれる公知の変異(385G)を導入したbIAPII(特許第6732368号の図5~7に記載される、385Gの有無が相違する「322D/323N/430A」と「322D/323N/385G/430A」の比較を参照)、および公知の3重変異(322D/385N/430A)を導入したbIAPII(特許第6732368号の図5~7における「322D/385N/430A」を参照)の熱安定性を評価した。
比較のためにMut1、Mut2、Mut3およびMut4に含まれる公知の変異(385G)を導入したbIAPII(特許第6732368号の図5~7に記載される、385Gの有無が相違する「322D/323N/430A」と「322D/323N/385G/430A」の比較を参照)、および公知の3重変異(322D/385N/430A)を導入したbIAPII(特許第6732368号の図5~7における「322D/385N/430A」を参照)の熱安定性を評価した。
(A)MglBシグナル配列を付加した385G変異体発現プラスミドの構築
385G変異体は、実施例1の(A)で作成したbIAPII発現プラスミドにPCRにより塩基配列を置換して導入した。まず、bIAPII発現プラスミドを鋳型として、置換する塩基配列を含むプライマーセット(配列番号22および配列番号23)を用いて、PCRにより直鎖DNAを増幅し、In-fusion HD cloning kitにより環状化した。このプラスミドを鋳型として、配列番号13および配列番号14のプライマーを用いて、PCRによりbIAPIIをコードする領域を増幅し、インサート断片を調製した。また、配列番号15および配列番号16のプライマーを用いてpET-28a(+)ベクターを鋳型として、PCRによりベクター断片を調製した。調製したインサートおよびベクター断片をIn-fusion HD cloning kitにより連結した。得られたプラスミドにより、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたbIAPIIの385G変異体が発現する。
385G変異体は、実施例1の(A)で作成したbIAPII発現プラスミドにPCRにより塩基配列を置換して導入した。まず、bIAPII発現プラスミドを鋳型として、置換する塩基配列を含むプライマーセット(配列番号22および配列番号23)を用いて、PCRにより直鎖DNAを増幅し、In-fusion HD cloning kitにより環状化した。このプラスミドを鋳型として、配列番号13および配列番号14のプライマーを用いて、PCRによりbIAPIIをコードする領域を増幅し、インサート断片を調製した。また、配列番号15および配列番号16のプライマーを用いてpET-28a(+)ベクターを鋳型として、PCRによりベクター断片を調製した。調製したインサートおよびベクター断片をIn-fusion HD cloning kitにより連結した。得られたプラスミドにより、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたbIAPIIの385G変異体が発現する。
(B)MglBシグナル配列を付加した322D/385N/430A 変異体発現プラスミドの構築
bIAPIIの322D/385N/430A変異体をコードする塩基配列を(配列番号24)をpET-28a(+)ベクターのNdeI/XhoIサイトに挿入したプラスミドをユーロフィンジェノミクス社で合成した。本プラスミドを鋳型として、配列番号25および配列番号26のプライマーを用いてPCRによりbIAPIIをコードする領域を増幅し、インサート断片を調製した。また、配列番号27および配列番号16のプライマーを用いてpET-28a(+)ベクターを鋳型として、PCRによりベクター断片を調製した。調製したインサートおよびベクター断片をIn-fusion HD cloning kitにより連結した。得られたプラスミドにより、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたbIAPIIの322D/385N/430A変異体が発現する。
bIAPIIの322D/385N/430A変異体をコードする塩基配列を(配列番号24)をpET-28a(+)ベクターのNdeI/XhoIサイトに挿入したプラスミドをユーロフィンジェノミクス社で合成した。本プラスミドを鋳型として、配列番号25および配列番号26のプライマーを用いてPCRによりbIAPIIをコードする領域を増幅し、インサート断片を調製した。また、配列番号27および配列番号16のプライマーを用いてpET-28a(+)ベクターを鋳型として、PCRによりベクター断片を調製した。調製したインサートおよびベクター断片をIn-fusion HD cloning kitにより連結した。得られたプラスミドにより、N末端側にMglBシグナル配列、C末端側にHisタグが付加されたbIAPIIの322D/385N/430A変異体が発現する。
構築したプラスミドを用いて実施例2の方法で、無細胞抽出液を調製し、実施例3の方法でALP活性を測定した。無細胞抽出液の活性はいずれも人工設計ALPと比較して低かった。
次に実施例4の方法で、70℃で10分加熱後に残存するALP活性を測定した。その結果、いずれの酵素も活性が失われていた(表5)。
実施例5:発現量の比較
(A)野生型および人工設計ALP発現菌の構築
実施例1の(A)および(C)で遺伝子合成により作成したプラスミドを鋳型として、各ALPをコードする領域をPCRにより増幅した。また、pET-21b(+)ベクターを鋳型として、直鎖状のベクターをPCRにより増幅した。用いたプライマーの配列の組み合わせを表6に示す。得られた両断片をIn-fusion HD cloning kit(タカラバイオ)により連結して環状化し、pET-21b(+)ベクターのNdeIおよびXhoIサイトに各ALPが挿入された発現プラスミドを得た。本プラスミドをE.coli Tuner(DE3)株、およびE.coli Origami B(DE3)に形質転換した。
(A)野生型および人工設計ALP発現菌の構築
実施例1の(A)および(C)で遺伝子合成により作成したプラスミドを鋳型として、各ALPをコードする領域をPCRにより増幅した。また、pET-21b(+)ベクターを鋳型として、直鎖状のベクターをPCRにより増幅した。用いたプライマーの配列の組み合わせを表6に示す。得られた両断片をIn-fusion HD cloning kit(タカラバイオ)により連結して環状化し、pET-21b(+)ベクターのNdeIおよびXhoIサイトに各ALPが挿入された発現プラスミドを得た。本プラスミドをE.coli Tuner(DE3)株、およびE.coli Origami B(DE3)に形質転換した。
(B)無細胞抽出液の調製および活性評価
(A)で構築した株をアンピシリン100mg/Lを添加したLB液体培地で37℃にて一晩生育させた。得られた菌体をアンピシリン100mg/Lを添加した3mLの本培養培地(組成:20g/L Bacto tryptone、15g/L 酵母エキス、1g/L 塩化ナトリウム、50mM リン酸カリウム緩衝液(pH7.15)、5mM 硫酸マグネシウムおよび10μM 塩化亜鉛)に30μL植菌し、試験管を用いて37℃で振とう培養を行った。OD660が0.5~0.7となった時に終濃度で1mMとなるようにIPTGを添加して発現を誘導し、18℃で培養した。
(A)で構築した株をアンピシリン100mg/Lを添加したLB液体培地で37℃にて一晩生育させた。得られた菌体をアンピシリン100mg/Lを添加した3mLの本培養培地(組成:20g/L Bacto tryptone、15g/L 酵母エキス、1g/L 塩化ナトリウム、50mM リン酸カリウム緩衝液(pH7.15)、5mM 硫酸マグネシウムおよび10μM 塩化亜鉛)に30μL植菌し、試験管を用いて37℃で振とう培養を行った。OD660が0.5~0.7となった時に終濃度で1mMとなるようにIPTGを添加して発現を誘導し、18℃で培養した。
培養24時間後、培養液1mLを回収し菌体を遠心分離により集めた。終濃度1mMとなるように硫酸マグネシウムを、100μMとなるように塩化亜鉛を添加した100μLのxTractor buffer(タカラバイオ)に懸濁し、室温で20分間インキュベートすることで溶菌した。遠心分離により破砕液から菌体残渣を除き、得られた上清を無細胞抽出液とした。
人工設計ALPにおける宿主の差異および硫酸塩の添加の効果を総合的に評価するため、無細胞抽出液に終濃度が1Mとなるように硫安(すなわち、硫酸アンモニウム)溶液を添加し、4℃で1時間保管した。硫安添加後の酵素液を適宜希釈して、実施例3に従ってALP活性を測定し、培養液1Lあたりの活性値に換算した。
その結果、Tuner(DE3)およびOrigami B(DE3)のいずれを宿主とした場合にも人工設計ALPはbIAPIIより高い活性を示した(表7)。
実施例6:精製酵素の比活性と熱安定性評価
(A)野生型および人工設計ALP発現菌の構築
実施例1の(C)で遺伝子合成により作成したプラスミドは、ALPのC末端にHisタグが付加して発現する設計となっていた。Hisタグが付加しないALPを発現するプラスミドを構築するため、ALP配列とHisタグの間に終止コドンを挿入した。まず、実施例1の(C)で遺伝子合成により作成したプラスミドを鋳型として、表8に示されるプライマーを用いてPCRにより増幅した。得られた直鎖状の2本鎖DNAをIn-fusion HD cloning kit(タカラバイオ)により環状化し、pET-28a(+)ベクターのNcoI/XhoIサイトに各ALPが挿入された発現プラスミドを得た。本プラスミドおよび実施例1の(A)で構築したbIAPII発現プラスミドをE.coli Origami 2(DE3)に形質転換した。
(A)野生型および人工設計ALP発現菌の構築
実施例1の(C)で遺伝子合成により作成したプラスミドは、ALPのC末端にHisタグが付加して発現する設計となっていた。Hisタグが付加しないALPを発現するプラスミドを構築するため、ALP配列とHisタグの間に終止コドンを挿入した。まず、実施例1の(C)で遺伝子合成により作成したプラスミドを鋳型として、表8に示されるプライマーを用いてPCRにより増幅した。得られた直鎖状の2本鎖DNAをIn-fusion HD cloning kit(タカラバイオ)により環状化し、pET-28a(+)ベクターのNcoI/XhoIサイトに各ALPが挿入された発現プラスミドを得た。本プラスミドおよび実施例1の(A)で構築したbIAPII発現プラスミドをE.coli Origami 2(DE3)に形質転換した。
(B)無細胞抽出液の調製
(A)で構築した株をカナマイシン50mg/Lを添加したLB寒天培地で37℃にて一晩生育させた。得られた菌体をカナマイシン50mg/Lを添加した100mLの本培養培地(組成:20g/L Bacto tryptone、15g/L 酵母エキス、1g/L 塩化ナトリウム、50mM リン酸カリウム緩衝液(pH7.15)、5mM 硫酸マグネシウムおよび10μM 塩化亜鉛)に植菌し、坂口フラスコを用いて37℃で振とう培養を行った。OD660が0.5~0.7となった時に終濃度で1mMとなるようにIPTGを添加して発現を誘導し、18℃で培養した。24時間後、培養液より菌体を遠心分離により集め、1mM MgCl2および 0.1mM ZnCl2を含む20mM Tris-HCl緩衝液(pH8.0)に懸濁し、超音波破砕を行った。遠心分離により破砕液から菌体残渣を除き、得られた上清を無細胞抽出液とした。

(C)陰イオン交換クロマトグラフィーによる分画
1mM MgCl2および0.1mM ZnCl2を含む20mM Tris-HCl緩衝液(pH8.0)を用いて平衡化したHiTrap Q HP,5mLカラム(Cytiva)に、無細胞抽出液をアプライした。カラムを同緩衝液で洗浄した後、0~0.7MのNaCl直線濃度勾配により、吸着タンパク質を分画した。得られた画分のALP活性を実施例2に従い測定し、活性画分を回収した。
1mM MgCl2および0.1mM ZnCl2を含む20mM Tris-HCl緩衝液(pH8.0)を用いて平衡化したHiTrap Q HP,5mLカラム(Cytiva)に、無細胞抽出液をアプライした。カラムを同緩衝液で洗浄した後、0~0.7MのNaCl直線濃度勾配により、吸着タンパク質を分画した。得られた画分のALP活性を実施例2に従い測定し、活性画分を回収した。
(D)硫安添加による処理
回収した画分に終濃度1Mとなるように硫安溶液を添加し、4℃で保管した。18時間後、遠心分離を行った。bIAPII、Mut1、およびMut2では上清を回収し、(D)の疎水性相互作用クロマトグラフィーにより分画した。Mut3およびMut4では上清を除去し、沈殿を150mM NaCl、1mM MgSO4、0.05mM ZnCl2を含む100mM Tris-HCl緩衝液(pH7.0)0.5mLに溶解し、(F)のゲルろ過クロマトグラフィーにより分画した。
回収した画分に終濃度1Mとなるように硫安溶液を添加し、4℃で保管した。18時間後、遠心分離を行った。bIAPII、Mut1、およびMut2では上清を回収し、(D)の疎水性相互作用クロマトグラフィーにより分画した。Mut3およびMut4では上清を除去し、沈殿を150mM NaCl、1mM MgSO4、0.05mM ZnCl2を含む100mM Tris-HCl緩衝液(pH7.0)0.5mLに溶解し、(F)のゲルろ過クロマトグラフィーにより分画した。
(E)疎水性相互作用クロマトグラフィーによる分画(bIAPII、Mut1、およびMut2のみ)
回収した試料を、1.0M 硫安、1mM MgCl2および0.1mM ZnCl2を含む20mM Tris-HCl緩衝液(pH8.0)を用いて平衡化したHiTrap butyl,5mlカラム(Cytiva)にアプライした。カラムを同緩衝液で洗浄した後、1.0~0 Mの硫酸アンモニウム直線濃度勾配により、吸着タンパク質を溶出した。得られた画分のALP活性を実施例2に従い測定し、活性画分を回収した。
回収した試料を、1.0M 硫安、1mM MgCl2および0.1mM ZnCl2を含む20mM Tris-HCl緩衝液(pH8.0)を用いて平衡化したHiTrap butyl,5mlカラム(Cytiva)にアプライした。カラムを同緩衝液で洗浄した後、1.0~0 Mの硫酸アンモニウム直線濃度勾配により、吸着タンパク質を溶出した。得られた画分のALP活性を実施例2に従い測定し、活性画分を回収した。
(F)硫安沈殿による濃縮(bIAPII、Mut1、およびMut2のみ)
回収した試料に等量の4M硫安溶液を添加し、4℃で一晩保管した。遠心分離したのち、沈殿を150mM NaCl、1mM MgSO4、0.05mM ZnCl2を含む100mM Tris-HCl緩衝液(pH7.0)0.5mLに溶解した。

150mM NaCl、1mM MgSO4、0.05mM ZnCl2を含む100mM Tris-HCl緩衝液(pH7.0)を用いて平衡化したSuperdex 200 Increase 10/300 GLカラム(Cytiva)にアプライし、同緩衝液により溶出した。

ALPの活性をQuantiChrom Alkaline Phosph atase Assay Kit(BioAssay Systems)を用いて、p-ニトロフェノールリン酸からp-ニトロフェノールへの変換活性で測定した。まず、希釈用緩衝液(100mM Tris-HCl、150mM塩化ナトリウム、1mM硫酸マグネシウム、0.05mM塩化亜鉛、0.1%ウシ血清アルブミン、pH7.0)で各ALP溶液の希釈系列を調製した。ALP活性をキットの説明書に従ってアッセイ溶液を96穴プレートに分注し、酵素の希釈液を添加して混合し、37℃でマイクロプレートリーダーSpectraMax 190(モレキュラーデバイス)で405nmの吸光度を測定し、1分あたりの変化を算出した(ΔAtest)。酵素ブランクとして、酵素溶液の代わりに希釈用緩衝液を添加して同様に算出した(ΔAblank)。各測定は2回ずつ実施した。1分間に1μmolのp-ニトロフェノールを生成する酵素量を1Uと定義し、以下の式でALP活性を算出した。

また、それぞれの酵素液を適宜希釈し、280nmの吸光度を超微量分光計ナノドロップ(サーモフィッシャーサイエンティフィック)で測定した。1mg/mLのときのALPの吸光度を1として、各ALP溶液の濃度を算出した。
ALP活性およびALP濃度からそれぞれのALPの比活性を算出した(表9)。
(J)熱安定性の評価
精製した各ALPを希釈用緩衝液で希釈後、マイクロチューブに分注した。サーマルサイクラーTaKaRa PCR Thermal Cycler Dice(タカラバイオ)で55℃から85℃の各温度で10分加熱したのちに氷冷した。これらの試料のALP活性を実施例3に従って3回ずつ測定し、55℃で加熱時の活性値を100%として、それぞれの温度で加熱後の残活性を算出した(図6)。さらに残活性が50%となる温度(T50)を算出した(表9)。
精製した各ALPを希釈用緩衝液で希釈後、マイクロチューブに分注した。サーマルサイクラーTaKaRa PCR Thermal Cycler Dice(タカラバイオ)で55℃から85℃の各温度で10分加熱したのちに氷冷した。これらの試料のALP活性を実施例3に従って3回ずつ測定し、55℃で加熱時の活性値を100%として、それぞれの温度で加熱後の残活性を算出した(図6)。さらに残活性が50%となる温度(T50)を算出した(表9)。
その結果、人工設計ALPはbIAPIIと比べて、T50として13℃以上の向上が認められた。
実施例7:単変異体の評価
4つの人工設計ALPで共通の変異点28か所(表2)について、単独でbIAPIIに導入し、それぞれの影響を評価した。まず、ウシ小腸由来アルカリホスファターゼ(bIAPII)からN末端の分泌シグナル配列およびC末端のGPIアンカー領域を除いたアミノ酸配列(配列番号1)をE.coliのコドンに最適化して塩基配列に変換した。本塩基配列(配列番号2)に開始コドンATGおよび終止コドンTAAを付加した配列をpET28b(+)のNcoI/XhoIサイトに挿入したプラスミドを、ジェンスクリプト社で合成した。本プラスミドをもとにジェンスクリプト社で各アミノ酸置換を導入したプラスミド28種を調製した。本プラスミドをE.coli Origami 2(DE3)に形質転換した。
4つの人工設計ALPで共通の変異点28か所(表2)について、単独でbIAPIIに導入し、それぞれの影響を評価した。まず、ウシ小腸由来アルカリホスファターゼ(bIAPII)からN末端の分泌シグナル配列およびC末端のGPIアンカー領域を除いたアミノ酸配列(配列番号1)をE.coliのコドンに最適化して塩基配列に変換した。本塩基配列(配列番号2)に開始コドンATGおよび終止コドンTAAを付加した配列をpET28b(+)のNcoI/XhoIサイトに挿入したプラスミドを、ジェンスクリプト社で合成した。本プラスミドをもとにジェンスクリプト社で各アミノ酸置換を導入したプラスミド28種を調製した。本プラスミドをE.coli Origami 2(DE3)に形質転換した。
bIAPIIおよび28種の単変異を導入したALPの発現株、および実施例6の(B)で作成したMut2の発現株を、カナマイシン50mg/Lを添加したLB液体培地で37℃にて一晩生育させた。得られた菌体をカナマイシン50mg/Lを添加した3mLの本培養培地(組成:20g/L Bacto tryptone、15g/L 酵母エキス、1g/L 塩化ナトリウム、50mM リン酸カリウム緩衝液(pH7.15)、5mM 硫酸マグネシウムおよび10μM 塩化亜鉛)に30μL植菌し、試験管を用いて37℃で振とう培養を行った。OD660が0.5~0.7となった時に終濃度で1mMとなるようにIPTGを添加して発現を誘導し、18℃で培養した。
培養24時間後、培養液1mLを回収し菌体を遠心分離により集めた。終濃度1mMとなるように硫酸マグネシウムを、100μMとなるように塩化亜鉛を添加した100μLのxTractor buffer(タカラバイオ)に懸濁し、室温で20分間インキュベートすることで溶菌した。遠心分離により破砕液から菌体残渣を除き、得られた上清を無細胞抽出液とした。これらの試料のALP活性を実施例3に従って3回ずつ測定し、培養液あたりの発現量を算出した(表10)。
次に、これらの無細胞抽出液に終濃度1Mとなるように硫安を添加後、適宜希釈して、実施例4の方法で、60℃で10分加熱後に残存するALP活性を測定した。
その結果、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461V変異は単独での導入で残存活性が特に向上していた(表10)。
実施例8:無細胞抽出液を用いた発現量および熱安定性の比較
実施例6の(B)で作成したbIAPII、Mut1、Mut2、Mut3、およびMut4の発現株を用いて、実施例7の方法に従い、培養液あたりの発現量及び60℃で10分加熱後に残存するALP活性を測定した。

その結果、人工設計ALPはいずれもbIAPIIに比べて高い発現量と熱安定性を示した(表11)。
Claims (19)
- 以下(A)、(B)、または(C)のタンパク質である、改変アルカリホスファターゼ:
(A)配列番号3、5、7、もしくは9のアミノ酸配列を含むタンパク質;
(B)配列番号3、5、7、もしくは9のアミノ酸配列に対して95%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号3、5、7、もしくは9のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~23個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。 - (B)または(C)のアミノ酸配列が、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異を維持する、請求項1記載の改変アルカリホスファターゼ。
- (B)または(C)のアミノ酸配列が、I332V、E343Q、G402S、T413E、E416D、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異を維持する、請求項2記載の改変アルカリホスファターゼ。
- (B)または(C)のアミノ酸配列が、E343Q、I352T、G402S、およびE455Qからなる群より選ばれる1個以上のアミノ酸残基の変異を維持する、請求項1記載の改変アルカリホスファターゼ。
- ウシ小腸由来アルカリホスファターゼをコードするアミノ酸配列において、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有する改変アルカリホスファターゼ。
- 前記1個以上のアミノ酸残基の変異が、I332V、E343Q、I352T、G402S、T413E、E416D、E455Q、およびI461Vからなる群より選ばれる1個以上のアミノ酸残基の変異である、請求項5記載の改変アルカリホスファターゼ。
- 前記1個以上のアミノ酸残基の変異が、E343、I352、G402、およびE455からなる群より選ばれる1個以上のアミノ酸残基の変異である、請求項5記載の改変アルカリホスファターゼ。
- ウシ小腸由来アルカリホスファターゼが以下(A)、(B)、または(C)のタンパク質である、請求項5~7のいずれか一項記載の改変アルカリホスファターゼ:
(A)配列番号1のアミノ酸配列を含むタンパク質;
(B)配列番号1のアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質;または
(C)配列番号1のアミノ酸配列において、置換、欠失、挿入、および付加、ならびにそれらの組合せからなる群より選ばれる1~47個のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有するタンパク質。 - 改変アルカリホスファターゼが、改変アルカリホスファターゼをコードする部分に加えて、他の機能性ペプチドをコードする部分をさらに含むタンパク質である、請求項1~8のいずれか一項記載の改変アルカリホスファターゼ。
- 以下を含む、改変アルカリホスファターゼの製造方法:
ウシ小腸由来アルカリホスファターゼに対して、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を導入して、
ウシ小腸由来アルカリホスファターゼをコードするアミノ酸配列において、I332、E343、I352、G402、T413、E416、E455、およびI461からなる群より選ばれる1個以上のアミノ酸残基の変異を含むアミノ酸配列を含み、かつ脱リン酸化活性を有する改変アルカリホスファターゼを生成すること。 - 請求項1~9のいずれか一項記載の改変アルカリホスファターゼ、または請求項10記載の方法により製造された改変アルカリホスファターゼで標識された物質。
- 物質が親和性物質または抗原である、請求項11記載の物質。
- 親和性物質が抗体である、請求項12記載の物質。
- 請求項1~9のいずれか一項記載の改変アルカリホスファターゼ、請求項10記載の方法により製造された改変アルカリホスファターゼ、または請求項11~13のいずれか一項記載の物質、および基質を用いて、アルカリホスファターゼ活性を測定することを含む、アルカリホスファターゼ活性の測定方法。
- アルカリホスファターゼ活性の測定方法が、請求項1~9のいずれか一項記載の改変アルカリホスファターゼ、請求項10記載の方法により製造された改変アルカリホスファターゼ、または請求項11~13のいずれか一項記載の物質、および基質を用いる、測定対象物質を検出または定量する方法である、請求項14記載の方法。
- 請求項1~9のいずれか一項記載の改変アルカリホスファターゼ、請求項10記載の方法により製造された改変アルカリホスファターゼ、または請求項11~13のいずれか一項記載の物質を用いて、リン酸化物質を脱リン酸化して脱リン酸化物質を生成することを含む、脱リン酸化物質の製造方法。
- 請求項1~9のいずれか一項記載の改変アルカリホスファターゼ、または請求項10記載の方法により製造された改変アルカリホスファターゼをコードするポリヌクレオチド。
- 請求項17記載のポリヌクレオチドを含む発現ベクター。
- 請求項1~9のいずれか一項記載の改変アルカリホスファターゼ、または請求項10記載の方法により製造された改変アルカリホスファターゼをコードするポリヌクレオチドの発現単位を含む形質転換細胞。
