Abstract
생리활성물질을 포함하는 생분해성 고분자 지지체 및 이의 제조방법에 관한 것으로, 상기 생분해성 고분자 지지체는 분해 과정에서 생성되는 산성 물질에 의한 염증 반응을 억제하고, 기계적 강도 조절이 용이하며, 표적 조직의 세포 유래 생체활성소재 및 생리활성물질을 포함함으로써 보다 효과적으로 조직 재생을 유도할 수 있다.
Description
본 출원은 2021년 3월 8일 출원된 대한민국 특허출원 제 10-2021-0030041호 를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다.
생리활성물질을 포함하는 생분해성 고분자 지지체 및 이의 제조방법에 관한 것이다.
조직공학(tissue engineering)이란 과학의 발달과 함께 등장한 새로운 분야의 하나로서 생명 과학과 공학, 의학 등의 기본 개념과 과학 기술을 통합 응용하는 다학제간 학문으로 생체 조직의 구조와 기능 사이의 상관 관계를 이해하고, 더 나아가 손상된 조직이나 장기를 정상적인 조직으로 대체하거나 재생시키기 위해 체 내에 이식이 가능한 인공 조직을 만들어 우리 몸의 기능을 유지, 향상 또는 복원하는 것을 목적으로 하는 응용학문이다.
대표적인 조직 공학 기법을 요약하면 다음과 같다. 우선, 환자의 몸에서 필요한 조직을 채취하고, 그 조직편으로부터 세포를 분리한 후 분리된 세포를 배양을 통해 필요한 양만큼 증식시킨다. 증식된 세포를 다공성 생분해성 고분자 지지체에 심어 일정기간 동안 체외 배양하여 얻어지는 하이브리드형 세포/고분자 구조물을 다시 인체내에 이식한다. 이식된 세포들은 대부분의 조직이나 장기에서 신생 혈관이 형성될 때까지는 체액의 확산에 의해 산소와 영양분을 공급받다가 체내에 혈관이 자라서 혈액의 공급이 이루어지면 세포들이 증식, 분화하여 새로운 조직 및 장기를 형성하고 고분자 지지체는 그동안 분해되어 소실되는 기법을 응용하는 것이다.
이러한 조직 공학 연구를 위해서는 우선 생체 조직과 유사한 생분해성 고분자 지지체를 제조하는 일이 중요하다. 인체 조직의 재생을 위해 사용되는 지지체 재료의 주된 요건은 조직 세포가 재료 표면에 유착하여 3차원적 구조를 가진 조직을 형성할 수 있도록 세포 친화성의 역할을 충분히 해내야 하고, 또한 이식된 세포와 숙주 세포 사이에 위치하는 중간 장벽으로서의 역할도 할 수 있어야 한다. 이는 이식 후 혈액 응고나 염증 반응이 일어나지 않는 무독성의 생체 적합성을 가져야 함을 의미한다. 또한, 새로운 조직이 형성되면서 분해되어, 궁극적으로 신체에 외래 물질을 남기지 않는 생분해성 특성을 가져야 한다. 그러나, 일반적으로 생분해성 합성 고분자들은 물성이 떨어지고 생분해시 산성 물질이 생성되어 인체 내에서 염증 반응 및 세포 독성을 일으킨다는 문제점이 있다. 또한, 천연 고분자의 경우, 물리적 및 기계적 물성이 부족하고 분해 기간의 조절이 어려울뿐만 아니라 유기 용매에 대한 낮은 용해도로 인해 화학적 개질이 제한적이고 다양한 응용이 이루어지지 못하는 문제점이 있다.
따라서, 생분해성 고분자의 분해 산물인 산성 물질을 중화하여 세포 및 조직의 염증 반응을 억제함과 동시에 생체 적합성 및 조직재생능을 강화하여 목표 기관의 재생을 효과적으로 유도할 수 있는 고기능성 복합 지지체를 개발할 필요가 있다.
일 양상은 염기성 나노 세라믹 입자, 세포외기질, 생리활성물질 및 생분해성 고분자를 포함하는 생분해성 고분자 지지체를 제공하는 것이다.
다른 양상은 상기 생분해성 고분자 지지체를 포함하는 생체 이식물을 제공하는 것이다.
다른 양상은 염기성 나노 세라믹 입자를 제조하는 단계; 상기 염기성 나노 세라믹 입자, 세포외기질, 생리활성물질 및 생분해성 고분자를 포함하는 제1 고분자 용액을 제조하는 단계; 상기 제1 고분자 용액에 100 내지 500 ㎛의 기공 유도체를, 상기 제1 고분자 용액 100 중량부에 대하여 100 내지 2000 중량부로 혼합하여 제2 고분자 용액을 제조하는 단계; 및 상기 제2 고분자 용액을 동결 건조하여 다공성 고분자 지지체를 제조하는 단계를 포함하는 생분해성 고분자 지지체의 제조방법을 제공하는 것이다.
일 양상은 염기성 나노 세라믹 입자, 세포외기질, 생리활성물질 및 생분해성 고분자를 포함하는 생분해성 고분자 지지체를 제공한다. 상기 생분해성 고분자 지지체는 다공성 고분자 지지체의 기공 크기 및 밀도, 다공성 등을 조절할 수 있을 뿐만 아니라 세포외기질 및 생리활성물질(DNA 단편 혼합물 및 세포외소포체)을 포함하는 고분자 지지체의 형태 및 크기를 다양하게 조절할 수 있다. 또한, 상기 염기성 나노 세라믹 입자, 세포외기질 및 생리활성물질을 다양한 농도로 함유함으로써 생분해성 고분자 지지체의 염증 억제능, 조직재생능 및 친수성이 향상되고, 기계적 강도 및 분해 기간을 제어할 수 있다.
상기 염기성 나노 세라믹 입자는 알칼리 금속 또는 이의 산화물, 또는 수산화물, 알칼리 토금속 또는 이의 산화물, 또는 수산화물로 구성된 군에서 선택되는 것일 수 있다. 상기 알칼리 금속 또는 알칼리 토금속은 예를 들어, 리튬(Li), 베릴늄(Be), 나트륨(Na), 마그네슘(Mg), 칼륨(K), 칼슘(Ca), 루비듐(Rb), 스트론튬(Sr), 바륨(Ba), 세슘(Cs), 프란슘(Fr), 라듐(Ra) 등인 것일 수 있다. 또한, 상기 알칼리 금속 또는 알칼리 토금속의 산화물 또는 수산화물은 예를 들어, 수산화리튬, 수산화베릴륨, 수산화나트륨, 수산화마그네슘, 수산화칼륨, 수산화칼슘, 수산화루비듐, 수산화스트론튬, 수산화바륨, 수산화세슘, 수산화프란슘, 수산화라듐, 산화마그네슘, 산화나트륨, 산화리튬, 산화나트륨, 산화망간, 산화칼륨, 산화칼슘, 산화바륨, 산화세슘, 산화라듐 등인 것일 수 있다.
일 구체예에 있어서, 상기 염기성 나노 세라믹 입자는 지방산, 또는 고분자로 표면 개질된 것일 수 있다. 상기 지방산은 예를 들어, 카프릴릭산, 카프릭산, 라우릭산, 미리스트올레산, 팔미톨레익산, 사피에닉산, 올레익산, 엘라이딘산, 박센산, 리놀레익산, 리신올레익산, 린올레아딕산, a-린올레익산, 아라키돈산, 에이코사펜타에노산, 에루식산, 스테아린산, DHA, 옥타테카노익산, 코코넛오일, 팜오일, 목화오일, 말씨눈오일, 콩오일, 올리브오일, 옥수수오일, 해바라기오일, 홍화오일, 대마오일, 카놀라오일 등인 것일 수 있다. 또한, 상기 고분자는 예를 들어, L-락타이드, D-락타이드, D,L-락타이드, 글라이콜라이드, 카프로락톤, 다이옥산온, 트리메틸렌카보네이트, 수산화알카노에이트, 펩티드, 시아노아크릴레이트, 락트산, 글라이콜산, 수산화카프로산, 말레산, 포스파젠, 아미노산, 수산화부틸릭산, 세바식산, 수산화에톡시아세트산 및 트리메틸렌글라이콜로 구성된 군에서 선택되는 1종 이상의 단량체로 폴리-L-락타이드, 폴리-D-락타이드, 폴리-D,L-락타이드, 폴리글라이콜라이드, 폴리카프로락톤, 폴리-L-락타이드-co-글라이콜라이드, 폴리-D-락타이드-co-글라이콜라이드, 폴리-D,L-락타이드-co-글라이콜라이드, 폴리-L-락타이드-co-카프로락톤, 폴리-D-락타이드-co-카프로락톤, 폴리-D,L-락타이드-co-카프로락톤, 폴리글라이콜라이드-co-카프로락톤, 폴리다이옥산온, 폴리트리메틸렌카보네이트, 폴리글라이콜라이드-co-다이옥산온, 폴리아미드에스터, 폴리펩티드, 폴리오르쏘에스터, 폴리말레산, 폴리포스파젠, 폴리안하이드라이드, 폴리세바식안하이드라이드, 폴리수산화알카노에이트, 폴리수산화부틸레이트, 폴리시아노아크릴레이트 등인 것일 수 있다. 일 구체예에 따른 지방산 또는 고분자로 표면이 개질된 염기성 나노 세라믹 입자를 포함하는 생분해성 고분자 지지체는 유기 용매 상에서 분산 안정성이 향상되었을 뿐만 아니라, 기계적 물성을 향상될 수 있으며, 산성 물질을 염기성 세라믹 입자로 중화시킴으로써 체내 염증 반응 및 세포 독성을 개선할 수 있다.
다른 구체예에 있어서, 상기 염기성 나노 세라믹 입자 또는 표면이 개질된 염기성 나노 세라믹 입자의 직경은 1 ㎚ 내지 1 ㎜인 것일 수 있다. 염기성 나노 세라믹 입자의 직경이 상기 범위를 초과하는 경우, 염기성 나노 세라믹 입자의 무게에 의해 침전이 발생되어 유기 용매 내에서 상 분리가 일어나는 문제가 있다.
또 다른 구체예에 있어서, 상기 염기성 나노 세라믹 입자는 고분자 지지체 100 중량부에 대하여 5 내지 50 중량부로 포함될 수 있다. 상기 염기성 나노 세라믹 입자는 고분자 지지체 100 중량부에 대하여 예를 들어, 5 내지 50 중량부, 5 내지 45 중량부, 5 내지 40중량부, 5 내지 30중량부, 10 내지 50 중량부¸10 내지 40 중량부, 10 내지 30 중량부, 20 내지 40 중량부 또는 10 내지 25 중량부로 포함될 수 있다. 이때, 염기성 나노 세라믹 입자의 함량이 상기 범위 미만인 경우, 고분자 지지체의 분해 산물인 산성 물질을 충분히 중화시킬 수 없는 문제점이 있으며, 상기 범위를 초과하는 경우, 고분자 지지체 주변 환경의 알칼리화가 유도될 수 있다는 문제점이 있다.
상기 세포외기질은 인체 또는 동물의 조직이나 세포로부터 유래된 기질 단백질로서, 복합적으로 혼합된 상태 또는 인위적으로 분리된 단일 분자 상태일 수 있으며, 단백질은 변성된 구조를 갖는 것일 수 있다. 상기 세포외기질은 예를 들어, 인체, 돼지, 소, 쥐, 양, 말, 개 또는 고양이 등의 척수동물에서 유래할 수 있으며, 목적에 따라 신장, 양막, 피부, 소장점막하조직, 근막 또는 척수막으로부터 분리된 것일 수 있다. 상기 세포외기질은 구조 또는 기능에 따라 적절하게 선택될 수 있다. 일 구체예에 있어서, 상기 세포외기질은 섬유 구조를 갖는 그룹, 골분화 및 골형성과 관련된 그룹, 글루코스아미노글리칸 그룹 또는 프로테오글리칸 그룹 등인 것일 수 있다. 상기 섬유 구조를 갖는 그룹은 예를 들어, 콜라겐 섬유, 엘라스틴 섬유, 라미닌, 피브리노겐, 피브로넥틴, 젤라틴 등인 것일 수 있다. 이때, 상기 콜라겐은 타입별로 I, II, III, IV, V, VI, VII, VIII, IX, X, XI, XII, XIV, XV, XVI, XVIII, XIX, XX, XXI, XXII, XXIII, XXIV, XXV, XXVI, XXVII, XXVIII 콜라겐 등인 것일 수 있다. 또한, 상기 골분화 및 골형성과 관련된 그룹은 예를 들어, 오스테오넥틴, 오스테오폰틴, 비트로넥틴, 비멘틴 등인 것일 수 있다. 또한, 상기 글루코스아미노글리칸 그룹은 예를 들어, 헤파란 황산, 케라탄 황산, 콘드로이틴 황산, 더마탄 황산, 헤파린, 저분자 헤파린, 히알루론산 등인 것일 수 있다. 또한, 상기 프로테오글리칸 그룹은 예를 들어, 데코린, 바이클라이칸, 베르시칸, 테르티칸, 퍼레칸, 비큐닌, 뉴로칸, 어그리칸, 피브로모듈린, 루미칸 등인 것일 수 있다. 일 구체예에 있어서, 상기 세포외기질은 고분자 지지체 100 중량부에 대하여 0.1 내지 20 중량부로 포함될 수 있다. 상기 세포외기질은 고분자 지지체 100 중량부에 대하여 예를 들어, 0.1 내지 20 중량부, 0.1 내지 15 중량부, 0.1 내지 15 중량부, 1 내지 20 중량부, 1 내지 15 중량부¸ 5 내지 20 중량부, 5 내지 15 중량부, 5 내지 10 중량부, 또는 10 내지 20 중량부로 포함될 수 있다. 이때, 세포외기질의 함량이 상기 범위 미만인 경우, 세포 적합성 개선 효과를 충분히 발휘할 수 없다는 문제점이 있으며, 상기 범위를 초과하는 경우, 고분자 지지체의 기계적 물성이 감소하는 문제점이 있다.
다른 구체예에 있어서, 상기 세포외기질은 탈세포화된 것일 수 있으며, 조직 또는 세포를 배양한 후, 물리적, 또는 화학적 방법에 의해 탈세포화된 것일 수 있다. 물리적 탈세포화 방법은 예를 들어, 동결-해동법, 초음파 처리, 물리적 교반 등이 있으며, 화학적 탈세포화 방법은 예를 들어, 동물 유래 조직의 분말을 물을 포함한 저장액, 음이온성 계면활성제, 비이온성 계면활성제 또는 양이온성 계면활성제, DNase, RNase 또는 트립신 등으로 처리하는 것일 수 있다. 상기 화학적 탈세포 방법에 있어서, 상기 저장액으로는 트리스-HCl(Tris-HCl)(pH 8.0) 용액이 사용될 수 있고, 상기 음이온성 계면활성제로는 소디움도데실설페이트(SDS), 소디움데옥시초레이트(sodium deoxycholate), 트리톤 X-200(Triton X-200) 등이 사용될 수 있다. 또한, 상기 비이온성 계면활성제로서는 트리톤 X-100(Triton X-100), 트윈 20(Tween 20) 또는 트윈 80(Tween 80)이 사용될 수 있고, 상기 양이온성 계면활성제로는 CHAPS, 술포베타인-10(Sulfobetaine-10, SB-10), 술포베타인-16(SB-16), 또는 트리-n-부틸포스페이트(Tri-n-butyl phosphate), N-라우로일-사르코시네이트(N-lauroyl-sarcosinate), IGEPAL CA-630 등이 사용될 수 있다. 또한, 상기 탈세포화는 동물의 조직, 예를 들어 신장 조직을 채취한 후 분말화 과정을 수행하기 이전 또는 분말화 과정을 수행한 이후에, 또는 분말화와 동시에 수행될 수 있다.
상기 생리활성물질은 DNA 단편 혼합물 또는 세포외소포체인 것일 수 있다. 일 실시예에서는 나노 세라믹 입자, 세포외기질, DNA 단편 혼합물, 세포외소포체 및 생분해성 고분자를 포함하는 생분해성 고분자 지지체를 신장 손상 모델 마우스에 이식한 결과, 조직의 재생능이 개선되고 재생과 관련된 인자의 발현이 증가하여 손상된 신장 조직이 재생된 것을 확인하였다. 또한, 상기 마우스의 사구체 재생이 증가하고, 사구체 경화 현상이 감소하였으며, 정상 마우스와 유사한 수준으로 사구체 여과율이 회복되는 것을 확인하였다. 따라서, 일 양상에 따른 생분해성 고분자 지지체는 DNA 단편 혼합물, 및/또는 세포외소포체 등과 같은 생리활성 물질을 포함함으로써, 손상된 조직을 재생시킬 뿐만 아니라 기능 회복에 탁월한 효과가 있다.
상기 DNA 단편 혼합물은 고분자 지지체 100 중량부에 대하여 3 내지 30 중량부로 포함될 수 있다. 상기 DNA 단편 혼합물은 고분자 지지체 100 중량부에 대하여 예를 들어, 3 내지 30 중량부, 3 내지 25 중량부, 3 내지 20 중량부, 5 내지 30 중량부, 5 내지 25 중량부¸5 내지 20 중량부, 10 내지 30 중량부 또는 15 내지 20 중량부로 포함될 수 있다. 이때, DNA 단편 혼합물의 함량이 상기 범위 미만인 경우, 생리활성 효과를 충분히 발휘 할 수 없다는 문제점이 있으며, 상기 범위를 초과하는 경우, 세포 독성을 유발할 수 있다는 문제점이 있다. 또한, 상기 DNA 단편 혼합물은 예를 들어, 폴리뉴클레오타이드(polynucleotide, PN), 폴리디옥시리보뉴클레오타이드(Polydeoxyribonucleotide, PDRN), Hydrolyzed DNA를 포함하는 뉴클레오타이드 중합체 등인 것일 수 있다.
상기 세포외소포체는 세포 간 정보 교환을 위해 모든 세포들이 외부 환경으로 분비하는 나노 크기의 소포체로서, 단백질, 지질, 핵산, 대사 물질 등 생물학적 활성을 보이는 다양한 물질을 포함하고 있다. 상기 세포외소포체는 엑소좀, 마이크로베지클을 포함하는 것일 수 있다. 상기 엑소좀은 제대, 제대혈, 골수, 지방, 근육, 피부, 양막 및 태반 유래 줄기세포에서 분리된 것일 수 있다.
상기 생분해성 고분자는 폴리-L-락타이드, 폴리-D-락타이드, 폴리-D,L-락타이드, 폴리글라이콜라이드, 폴리카프로락톤, 폴리-L-락타이드-co-글라이콜라이드, 폴리-D-락타이드-co-글라이콜라이드, 폴리-D,L-락타이드-co-글라이콜라이드, 폴리-L-락타이드-co-카프로락톤, 폴리-D-락타이드-co-카프로락톤, 폴리-D,L-락타이드-co-카프로락톤, 폴리글라이콜라이드-co-카프로락톤, 폴리다이옥산온, 폴리트리메틸렌카보네이트, 폴리글라이콜라이드-co-다이옥산온, 폴리아미드에스터, 폴리펩티드, 폴리올쏘에스터계, 폴리말레산, 폴리포스파젠, 폴리안하이드라이드, 폴리세바식안하이드라이드, 폴리수산화알카노에이트, 폴리수산화부틸레이트 및 폴리시아노아크릴레이트로 구성된 군에서 1종 이상 선택되는 것일 수 있다.
다른 양상은 상기 생분해성 고분자 지지체를 포함하는 생체 이식물을 제공한다. 상기 생분해성 고분자 지지체의 구체적인 내용은 전술한 바와 같다.
일 구체예에 있어서, 상기 생체 이식물은 상기 생분해성 고분자 지지체로 표면이 개질되거나 코팅된 것일 수 있다. 상기 생체 이식물은 예를 들어, 조직재생용 지지체, 스텐트, 수술용 봉합사, 바이오 나노 섬유, 하이드로젤, 바이오스폰지, 핀, 나사, 막대, 임플란트 등인 것일 수 있다.
다른 양상은 염기성 나노 세라믹 입자를 제조하는 단계를 포함하는, 생분해성 고분자 지지체의 제조방법을 제공한다.

상기 기공 유도체는 얼음 입자인 것일 수 있다. 상기 얼음 입자의 크기 및 함량을 조절함으로써 다공성 고분자 지지체의 기공 크기 및 기공도를 조절할 수 있다. 상기 얼음 입자의 크기는 10 내지 500 ㎛인 것일 수 있다. 상기 얼음 입자의 크기는 예를 들어, 10 내지 500 ㎛, 10 내지 450 ㎛, 10 내지 400 ㎛, 10 내지 300 ㎛, 30 내지 500 ㎛, 30 내지 300 ㎛, 30 내지 250 ㎛, 50 내지 500 ㎛, 50 내지 400 ㎛, 50 내지 300 ㎛, 50 내지 200 ㎛, 100 내지 500 ㎛, 100 내지 300 ㎛ 또는 150 내지 200 ㎛인 것일 수 있다. 이때, 얼음 입자의 크기가 상기 범위 미만인 경우, 세포의 침윤도가 낮아지며 신생 혈관 생성 효과를 충분히 발휘할 수 없다는 문제점이 있으며, 상기 범위를 초과하는 경우, 고분자 지지체의 기계적 물성이 감소하는 문제점이 있다. 또한, 상기 얼음 입자는 염기성 제1 고분자 용액 100 중량부에 대하여 100 내지 2000 중량부로 포함될 수 있다. 상기 얼음 입자는 제1 고분자 용액 100 중량부에 대하여 100 내지 2000 중량부, 100 내지 1500 중량부, 100 내지 1300 중량부, 100 내지 1000 중량부, 100 내지 500 중량부, 500 내지 2000 중량부, 500 내지 1500 중량부, 500 내지 1000 중량부, 1000 내지 2000 중량부 또는 1500 내지 2000 중량부로 포함된 것일 수 있다. 이때, 얼음 입자의 함량이 상기 범위 미만인 경우, 고분자 지지체 내부에 기공이 충분히 형성되지 않는다는 문제가 있으며, 상기 범위를 초과하는 경우, 고분자 지지체의 기계적 물성이 감소하는 문제가 있다. 또한, 상기 얼음 입자를 이용하여 동결전조법으로 고분자 지지체를 제조함으로써 통상의 염 발포법(salt forming)에서 발생할 수 있는 염기성 나노 세라믹 입자 및 생체활성물질(세포외기질 및 DNA 단편 혼합물)의 이탈을 방지할 수 있다.
상기 방법은 다공성 고분자 지지체에, 세포외소포체를 담지하는 단계를 추가로 포함할 수 있다. 상기 세포외소포체의 구체적인 내용은 전술한 바와 같다. 상기 세포외소포체는 다공성 고분자 지지체에 10 내지 500 ㎍의 용량으로 단순 로딩법에 의해 담지되는 것일 수 있다. 상기 다공성 고분자 지지체에 담지되는 세포외소포체의 용량은 예를 들어, 10 내지 500 ㎍, 10 내지 400 ㎍, 10 내지 300 ㎍, 10 내지 200 ㎍, 50 내지 500 ㎍, 50 내지 450 ㎍, 50 내지 350 ㎍, 50 내지 200 ㎍, 100 내지 500 ㎍, 100 내지 300 ㎍ 또는 200 내지 400 ㎍인 것일 수 있다. 이때, 세포외소포체의 함량이 상기 범위 미만인 경우, 조직 재생 효과가 충분히 발휘되지 않는다는 문제점이 있으며 상기 범위를 초과하는 경우, 로딩 가능한 최대 용량을 초과하는 바, 다공성 고분자 지지체 내에 세포외소포체의 담지가 불가능하다는 문제점이 있다.
일 양상에 따른 생분해성 고분자 지지체는 분해 과정에서 생성되는 산성 물질에 의한 염증 반응을 억제하고, 기계적 강도 조절이 용이하며, 표적 조직의 세포외 기질 유래 생체활성소재 및 생리활성물질을 포함함으로써 보다 효과적으로 조직 재생을 유도할 수 있다.

이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[제조예]
제조예 1. 염기성 나노 세라믹 입자의 제조
1-1. 수산화마그네슘 입자의 제조
수산화나트륨 10.8 g을 3차 증류수 300 ㎖에 용해하여 수산화나트륨 용액을 제조하였다. 이후, 3차 증류수 150 ㎖에 질산 마그네슘 20 g이 용해된 질산 마그네슘 용액을 적하 깔대기를 이용하여, 상기 수산화나트륨 용액에 분당 40 드롭의 속도로 적하 하였다. 반응 용액에 침전된 나노 수산화마그네슘입자를 증류수를 흘려주며 정제한 뒤, 여과하여 수득하였다. 수득한 나노 수산화마그네슘 입자는 진공건조하여 보관하였다.
1-2. 산화마그네슘 입자의 제조
상기 제조예 1-1에서 제조한 수산화마그네슘 입자를 전기로(electric furnace)를 이용하여 500~1500℃의 온도에서 하소시켜 산화마그네슘 입자를 제조하였다.
1-3. 고분자로 표면 개질된 수산화마그네슘 입자의 제조
상기 제조예 1-1에서 제조한 염기성 나노 세라믹 입자를 L-락타이드로 표면 개질하였다. 구체적으로, 전체 혼합물의 총 중량을 기준으로 상기 제조예 1-1의 수산화마그네슘 80 중량부 및 L-락타이드 20 중량부를 혼합하였다. 이후, 반응물 (수산화마그네슘 및 L-락타이드)의 총 중량에 대해 0.05 중량%의 옥토산주석(촉매)을 톨루엔에 희석하여 투입하였다. 반응물이 담긴 유리 반응기를 교반하면서 70 ℃에서 진공 상태로 6시간 동안 유지시켜 톨루엔과 수분을 완전히 제거하였다. 봉인된 유리 반응기를 150 ℃로 조절된 기름 중탕에서 교반하면서 48시간 동안 개환 중합반응을 수행하였다. 회수된 중합체를 충분한 양의 클로로포름에 넣어 1시간 이상 호모 폴리머 및 미반응 잔여물을 제거하여 고분자로 개질된 염기성 나노 세라믹 입자를 제조하였다.
1-4. 고분자로 표면 개질된 산화마그네슘 입자의 제조
상기 제조예 1-2에서 제조한 산화마그네슘 입자를 사용한 점을 제외하고는 상기 제조예 1-3과 동일한 방법으로 고분자로 개질된 염기성 나노 세라믹 입자를 제조하였다.
제조예 2. 탈세포화 및 분말화된 세포외기질의 제조
2-1. 사람 지방 유래
개체의 지방 조직을 채취한 후 생리식염수를 이용하여 10분간 3회 세척하였다. 세척된 조직을 에탄올로 탈수시킨 후, 조직에 존재하는 지방 세포 및 유전자 성분을 제거하고 순수한 세포외기질을 탈세포화 과정을 수행하여 수득하였다. 구체적으로, 탈수된 조직을 10 g 당 0.1%의 SDS(sodium dodecyl sulfate) 1ℓ용액에 넣고 100 rpm에서 24시간 동안 교반하였다. 이후, 3차 증류수로 100 rpm에서 30분간 5회 세척한 다음, 200 U/㎖ 농도의 DNase 200 ㎖을 넣고, 37 ℃에서 100 rpm으로 24시간 교반하였다. 이후, 3차 증류수로 100 rpm에서 30분간 5회 세척한 후, 건조하였다. 탈세포된 세포외기질을 동결분쇄기를 사용하여 약 50 ㎛ 크기로 동결 분쇄하여 분말화 하였다.
2-2. 사람 피부 유래
개체의 피부 조직을 사용하였다는 점을 제외하고는 상기 제조예 2-1과 동일한 방법으로 세포외기질을 제조하였다.
2-3. 돼지 신장 유래
돼지의 신장 조직을 사용하였다는 점을 제외하고는 상기 제조예 2-1과 동일한 방법으로 세포외기질을 제조하였다.
2-4. 쥐 신장 유래
쥐의 신장 조직을 사용하였다는 점을 제외하고는 상기 제조예 2-1과 동일한 방법으로 세포외기실을 제조하였다.
제조예 3. 줄기세포 유래 엑소좀의 분리
인간 탯줄 유래 줄기세포를 70% 정도 증식시킨 후, 인산염 완충용액으로 2회 세척하였다. 이후, 엑소좀이 제거된 우태아혈청 10%를 함유하는 무페놀레드 배지로 교체한 뒤, 상기 줄기세포를 배양하여 12시간마다 4회에 걸쳐 상층액을 회수하였다. 배양액을 0.22 ㎛ 필터로 여과하여 불순물을 제거하고, MWCO 300 또는 500 kDa 필터와 접선흐름여과장치(Tangential Flow Filtration, TFF)를 이용하여 선별적으로 엑소좀을 분리 및 농축하였다.
[실시예]
실시예 1. 생리활성물질이 함유된 고분자 지지체 (1)
염기성 나노 세라믹 입자, 세포외기질, 세포외소포체 및 생분해성 고분자를 포함하는 다공성 고분자 지지체에, 엑소좀을 로딩하여 엑소좀이 로딩된 고분자 지지체를 제조하였다. 구체적으로, 상기 제조예 1-1의 염기성 나노 세라믹 입자를 고분자 용액 총 중량부에 대하여 20 중량부, 제조예 2-1의 세포외기질 분말 10 중량부, 폴리데옥시리보뉴클레오티드(PolyDeoxyRiboNucleotide, PDRN) 20 중량부 및 분자량 40K의 폴리락타이드-co-글라이콜라이드(50:50) 생분해성 고분자 50 중량부를 유기 용매에 혼합하여 고분자 용액을 제조하였다. 이후, 상기 용액에 100~300 ㎛ 크기의 얼음 입자를 상기 고분자 용액 총 중량부에 대하여 1600 중량부로 투입하여 균일하게 혼합한 후, 실리콘 몰드를 이용하여 액체 질소 내에서 지지체의 크기 및 형태를 고정한 뒤 0℃, 5 mTorr 조건에서 동결건조하여 다공성 고분자 지지체를 제조하였다. 이후, 상기 다공성 지지체를 에탄올에 침지하여 멸균한 뒤, 멸균 증류수로 세척하여 에탄올을 제거하고, 생리식염수에 침지하여 수화하였다. 상기 제조예 3에서 분리된 엑소좀의 단백질 양을 BCA 발색법으로 측정한 뒤, 엑소좀 100 ㎍을 단순 로딩법으로 상기 다공성 고분자 지지체에 로딩하여 생리활성물질이 함유된 고분자 지지체를 제조하였다.
실시예 2. 생리활성물질이 함유된 고분자 지지체 (2)

[실험예]
실험예 1. 염기성 나노 세라믹 입자의 크기 확인
상기 제조예 1에서 제조한 염기성 나노 세라믹 입자의 크기를 나노 입자 분석기 또는 입도 분석기(Malvern Zen 3600 Zatasizer, Zetasizer Ivano, UK)를 이용하여 확인하였다.
도 2는 일 구체예에 따른 염기성 세라믹 입자의 표면 개질 전, 및 개질 후의 입자 크기를 나타낸 그래프이다.
그 결과, 도 2에 나타낸 바와 같이, 제조예 1-1의 염기성 나노 세라믹 입자의 경우, 입자의 크기가 평균 2.382 ㎛ 인 것을 확인하였고 고분자로 표면이 개질된 제조예 1-3의 경우, 0.48㎛인 것을 확인할 수 있었다. 즉, 일 구체예에 따른 고분자로 표면이 개질된 염기성 나노 세라믹 입자는 표면 개질에 의해 분산성이 개선될 수 있다.
실험예 2. 고분자 지지체의 기계적 인장 강도 측정
상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체의 기계적 인장 강도를 ASTM D638의 방법에 따라 Instron으로 측정하였다.
도 3a는 일 구체예에 따른 생분해성 고분자 지지체의 압축 응력-변형 곡선을 나타낸 그래프이다.
도 3b는 일 구체예에 따른 생분해성 고분자 지지체의 압축 모듈러스를 나타낸 그래프이다.
그 결과, 도 3a 및 3b에 나타낸 바와 같이, 실시예 1의 고분자 지지체는 비교예 1과 비교하여 기계적 물성이 개선된 것을 확인할 수 있었다. 즉, 일 양상에 따른 고분자 지지체는 염기성 세라믹 입자, 세포외기질, 생리활성물질을 포함함으로써 기계적 물성이 개선될 수 있다.
실험예 3. 고분자 지지체의 물 접촉각 변화 확인
상기 실시예 1 및 비교예 1~3에서 제조한 고분자 지지체의 물 접촉각을 측정하여 그 결과를 하기 표 1에 나타내었다.

그 결과, 표 1에 나타낸 바와 같이, 실시예 1의 고분자 지지체는 비교예 1~3과 비교하여 물 접촉각이 현저하게 감소하는 것을 확인할 수 있었으며, 엑소좀을 담지함으로써 친수성이 증가한 것을 확인할 수 있었다.
실험예 4. 고분자 지지체의 생분해 기간 및 pH 변화 확인
상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체의 생분해 기간 및 pH 변화를 확인하였다.
도 3c는 일 구체예에 따른 생분해성 고분자 지지체의 분해 거동을 나타낸 그래프이다.
도 3d는 일 구체예에 따른 생분해성 고분자 지지체의 pH 변화를 나타낸 그래프이다.
그 결과, 도 3c에 나타낸 바와 같이, 실시예 1의 고분자 지지체는 비교예 1~4와 비교하여 초기(0~15일) 생분해 속도가 유의적으로 높은 것을 확인할 수 있었다. 또한, 도 3d에 나타낸 바와 같이, 실시예1의 고분자 지지체는 비교예 1과 비교하여 시간에 따른 pH 변화가 크지 않은 것을 확인할 수 잇었다.
실험예 5. 고분자 지지체의 기공 크기, 및 구조 확인
상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체의 기공 크기 및 구조를 주사전자현미경을 이용하여 분석하였다.
도 3e는 일 구체예에 따른 생분해성 고분자 지지체의 기공 크기, 구조를 주사전자현미경을 이용하여 분석한 사진이다.
그 결과, 도 3e에 나타낸 바와 같이, 실시예 1의 경우, 지지체 내 기공의 평균 크기가 200 ㎛로 측정되었다. 즉, 일 양상에 따른 고분자 지지체는 기공 유도체의 크기 및 함량 사용함으로써, 기공 내 크기를 조절할 수 있다.
실험예 6. 고분자 지지체의 방출 거동 확인
상기 실시예 1에서 제조한 고분자 지지체의 DNA 단편 혼합물의 방출 거동을 확인하였다. 구체적으로, 상기 실시예 1의 고분자 지지체를 nuclease free water 1 mL에 침지하여 28일 동안 보관하였다. 침지 후 1일째, 2일째, 3일째, 5일째, 7일째 및 28일째에 상층액을 수득한 후, DNA intercalator와 반응시켜 형광값을 측정 및 분석하였다.
도 4a는 일 구체예에 따른 생분해성 고분자 지지체의 DNA 단편 혼합물의 방출 거동을 나타낸 그래프이다.
그 결과, 도 4a에 나타낸 바와 같이, 침지 후 24시간이 경과하였을 때 실시예 1의 고분자 지지체에 담지된 PDRN의 약 47%가 방출되었으며, 실험이 진행되는 기간(28일) 동안 약 95%의 PDRN이 방출되는 것을 확인할 수 있었다. 즉, 일 양상에 따른 생분해성 고분자 지지체는 지지체 내부에 담지된 DNA 단편 혼합물이 효과적으로 방출됨으로써 표적 세포 또는 조직에 운반할 수 있다.
실험예 7. 고분자 지지체 내부에 담지된 엑소좀 확인
상기 실시예 1 및 비교예 3의 고분자 지지체 내부에 담지된 엑소좀이 지지체 기공의 표면에 도포되어 있는지 여부를 전계방출형 주사전자현미경을 이용하여 확인하였다. 또한, 상기 엑소좀을 친유성 형광물질로 염색하여 지지체에 단순 로딩한 후, 공초점 레이저 주사현미경을 이용하여 확인하였다.
도 4b는 일 구체예에 따른 생분해성 고분자 지지체에 담지된 엑소좀을 주사전자현미경으로 분석한 그림이다.
도 4c는 일 구체예에 따른 생분해성 고분자 지지체에 담지된 형광 염색된 엑소좀을 공초점 레이저 주사현미경으로 분석한 그림이다.
그 결과, 도 4b 및 도 4c에 나타낸 바와 같이, 비교예 3에서는 엑소좀이 관찰되지 않았으나, 실시예 1의 고분자 지지체의 기공 외부 및 내부 표면에서 엑소좀이 고르게 분포되어 있는 것을 확인할 수 있었다.
실험예 8. 고분자 지지체의 세포 독성 확인
상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체의 세포 독성을 평가하기 위하여, 신장 유래 세포를 상기 실시예 1 및 비교예 1~3, 5의 지지체에서 배양한 후, 세포 성장률 및 독성을 각각 CCK-8 및 Calcein AM/EthD-1으로 염색하여 분석하였다.
도 5a는 일 구체예에 따른 생분해성 고분자 지지체 내부에서 세포의 성장률을 나타낸 그래프이다.
도 5b는 일 구체예에 따른 생분해성 고분자 지지체 내부에서 세포 독성을 분석한 그림이다.
그 결과, 도 5a에 나타낸 바와 같이, 실시예 1의 고분자 지지체는 비교예 1~4와 비교하여 상대적 세포 성장률이 현저하게 높은 것을 확인할 수 있었다. 특히, 1일 째에 비하여 7일 째에 세포 성장률이 약 2~3배 상승한 것을 확인할 수 있었다. 또한, 도 5b에 나타낸 바와 같이, 실시예 1의 고분자 지지체는 비교예 1~4와 비교하여 형광의 강도가 더 높았으며 형광이 비교적 고르게 분포되어 있는 것을 확인할 수 있었다. 즉, 일 양상에 따른 고분자 지지체는 세포 독성이 없어 지지체 내부에서 세포 생장을 효과적으로 촉진할 수 있다.
실험예 9. 고분자 지지체의 조직재생능 평가
상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체의 조직재생능을 평가하기 위하여, 신장 손상 모델에 상기 지지체를 이식하였다. 구체적으로, 상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체를 에틸렌 옥사이드 가스와 에탄올로 멸균한 뒤, 생리 식염수에 침지하여 준비하였다. 신장 손상 모델 마우스는 부분 신절제 방법으로 제조하였다. 상기 신장 손상 모델 마우스에 신장 절제술을 실시하고, 신장 피질의 부분적 손상 부위에 5x2x2 ㎥ 크기의 상기 고분자 지지체를 이식하여 8주 동안 유지하였다. 이식 후, 2주, 및 8주 후에 상기 마우스 신장의 조직을 적출한 후, 면역조직화학염색 및 중합효소연쇄반응법을 이용하여 조직학적 분석을 수행하였다.
도 6a는 일 구체예에 따른 생분해성 고분자 지지체를 부분 신절제 쥐 모델에 이식하고 신장을 적출하여 조직학적 염색법으로 분석한 그림이다.
도 6b는 일 구체예에 따른 생분해성 고분자 지지체를 부분 신절제 쥐 모델에 이식하고 신장을 적출하여 재생 관련 인자의 발현을 실시간 중합효소 연쇄반응으로 분석한 그래프이다.
그 결과, 도 6a에 나타낸 바와 같이, 비교예 1~4는 낮은 조직 재생능을 보였을 뿐만 아니라, 조직 주변의 섬유화가 관찰되었다. 그러나, 실시예 1의 고분자 지지체는 조직 재생능이 개선되었고, 주변 조직의 섬유화 또한 현저히 감소됨을 확인할 수 있었다. 또한, 도 6b에 나타낸 바와 같이, 실시예 1의 경우, 비교예 1~4와 비교하여 신장 재생 관련 인자인 Pax2 mRNA의 발현 수준이 유의적으로 높은 것을 확인할 수 있었다. 특히, 비교예 1에 비하여 실시예 1의 지지체를 이식한 마우스에서 Pax2 mRNA의 발현 수준이 약 6배 이상 높은 것을 확인할 수 있었다. 즉, 일 양상에 따른 생분해성 고분자 지지체는 세포외소포체, DNA 단편 혼합물과 같은 생리활성물질을 포함함으로써 조직 재생 기능이 우수한 것을 알 수 있다.
실험예 10. 고분자 지지체의 신장 기능 개선 효능 평가
상기 실시예 1 및 비교예 1~4에서 제조한 고분자 지지체의 신장 기능 개선 효능을 평가하였다. 구체적으로, 상기 실험예 8의 고분자 지지체를 이식한 마우스의 재생된 사구체 수 및 사구체 경화증 정도를 조직학적 염색법으로 분석하고, 사구체 여과율을 FITC-이눌린을 사용하여 분석하였다.
도 7a는 일 구체예에 따른 생분해성 고분자 지지체를 부분 신절제 쥐 모델에 이식한 후 재생된 사구체의 수를 나타낸 그래프이다.
도 7b는 일 구체예에 따른 생분해성 고분자 지지체를 부분 신절제 쥐 모델에 이식한 후 재생된 사구체 경화증을 나타낸 그래프이다.
도 7c는 일 구체예에 따른 생분해성 고분자 지지체를 부분 신절제 쥐 모델에 이식하고 사구체 여과율을 분석한 그래프이다.
그 결과, 도 7a에 나타낸 바와 같이, 비교예 1~4와 비교하여 실시예 1의 지지체를 이식한 마우스에서 재생된 사구체의 수가 유의적으로 증가한 것을 확인할 수 있었다. 또한, 도 7b에 나타낸 바와 같이, 실시예 1의 지지체를 이식한 마우스의 경우, 비교예 1~4의 지지체를 이식한 마우스에 비해 사구체 경화증이 현저하게 낮았으며, 특히 비교예 1과 비교하여 약 50% 이상 감소한 것을 확인할 수 있었다. 또한, 도 7c에 나타낸 바와 같이, 실시예 1의 지지체를 이식한 마우스는 정상 대조군(Native)와 유사한 수준으로 사구체 여과율이 회복되는 것을 확인할 수 있었다. 즉, 일 양상에 따른 생분해성 고분자 지지체는 생리활성물질을 포함함으로써 손상된 신장의 기능의 회복을 촉진하는 바, 조직재생용 재료로서 유용하게 사용될 수 있다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
