Description
したがって、本発明は、生理活性物質を内包させた超分子構造を有するタンパク質の新規な製造方法を提供することを課題とする。
1.生理活性物質を内包する超分子構造を有するタンパク質の製造方法であって、
(I)超分子構造を構成するタンパク質のサブユニットと、生理活性物質と、前記サブユニットから超分子構造を有するタンパク質を形成させるための溶液とを、フローマイクロミキサー中で接触させることを含む、
前記製造方法。
2.超分子構造を有するタンパク質がフェリチンである、前記1に記載の製造方法。
3.工程(I)において、前記サブユニットの流速と前記溶液の流速との合計が、約10mL/min以上である、前記1又は2に記載の製造方法。
4.前記サブユニットが、(II-1)超分子構造を有するタンパク質を含む溶液のpHを酸性又は塩基性に変化させることにより得られる、前記1~3のいずれかに記載の製造方法。
5.工程(II-1)において用いる溶液が、pH約1.5~約3.0又はpH約10~12の溶液である、前記4に記載の製造方法。
6.工程(I)において用いるサブユニットから超分子構造を有するタンパク質を形成させるための溶液が、pH約5.0~約9.0の溶液である、前記1~5のいずれか1に記載の製造方法。
7.前記サブユニットが、(II-2)超分子構造を有するタンパク質を含む溶液に溶剤を添加することにより得られる、前記1~3のいずれかに記載の製造方法。
8.前記工程(II-2)において用いる溶剤が、ジメチルスルホキシド(DMSO)、N,N-ジメチルホルムアミド(DMF)、アセトニトリル、及びエタノールからなる群から選ばれる、前記7に記載の製造方法。
9.工程(I)において用いる溶液が、Tris緩衝液、HEPES緩衝液、リン酸緩衝液、ホウ酸緩衝液、クエン酸緩衝液、炭酸緩衝液、及びグリシン緩衝液等からなる群から選ばれる、前記7又は8に記載の製造方法。
10.工程(II-1)又は(II-2)を、フローマイクロミキサーを用いて行う、前記4~9のいずれか1に記載の製造方法。
11.前記生理活性物質が、質量平均分子量約1,000~約20,000のsiRNA、DNA、オリゴペプチドまたはこれらの組合せからなる群から選ばれる、前記1~10のいずれかに記載の製造方法。
本明細書において使用する用語を、以下のとおり定義する。
「超分子」とは、複数の分子やイオンが非共有結合的な相互作用で(例えば自己組織化により)分子会合した、高次構造を有する会合体である。
「脱会合」とは、超分子構造を取っているタンパク質を、個々のサブユニットに解離させることをいう。
「再会合」とは、脱会合したタンパク質に、再び超分子構造を取らせることをいう。
「pH」は、25℃において、ガラス電極を用いて測定した値を指す。
「ミス会合体」とは、フェリチンを例に説明すると、正しく会合したフェリチンは、サブユニット24個の集合体であるが、2つの24量体が2価金属イオンなどを介して結合したものや、サブユニットの数が24に満たない会合体、サブユニットの数が24を超える会合体、会合できずに凝集してしまったタンパク質の塊など、24個のサブユニットが会合体を形成していないタンパク質を総称して「ミス会合体」と言う。
本発明は、超分子構造をとるタンパク質のサブユニットを、生理活性物質の存在下、FMM中で会合させることにより、その中心に存在する空洞に生理活性物質を内包させた、超分子構造をとるタンパク質を製造する方法である。
サブユニットから生理活性物質内包会合体を得る工程、すなわち再会合工程を、工程(I)と称する。
FMMは、一般的に、数十から数百ミクロンの空間において2種以上の液体を混合する装置である。本発明の方法で用いるFMMは、連続フロー式である。
本発明において用いるFMMは、例えば、サブユニットが流通する第一流路と、生理活性物質が流通する第二流路と、サブユニットから会合体を形成させるための溶液(「再会合用溶液」)が流通する第三流路とを有し、サブユニットと、生理活性物質と、前記溶液とが合流して混合される場を備える。サブユニット流と生理活性物質流とを予め混合し、そこに再会合用溶液流を合流させて混合してもよく、サブユニット流と、生理活性物質流と、前記溶液流とを混合場で同時に合流させて混合してもよい。内包させる生理活性物質が、脱会合に用いる酸や塩基や溶剤に弱い場合、後者の方が、生理活性物質が酸等に触れる時間を短くできるので好ましい。
サブユニットから超分子構造を有するタンパク質を再会合させる手段は特に限定されない。例えば、(I-1)サブユニット流と生理活性物質流の混合流のpHを中性、例えば約5~約9に調整することにより行うことができる。この工程は、脱会合を、後述する工程(II-1)により行ったときに適している。工程(I-1)におけるpH調整は、再会合用溶液を前記混合流に加えることにより行うことができる。再会合用溶液のpHは、脱会合用溶液のpHに応じて適宜設定することができるが、一般的には、5.0~10.0程度であるとよい。pHを中性にする物質としては、Tris緩衝液、HEPES緩衝液、リン酸緩衝液、ホウ酸緩衝液、クエン酸緩衝液、炭酸緩衝液、グリシン緩衝液等が挙げられる。このうち、幅広いpH緩衝能をもつためTris緩衝液またはリン酸緩衝液などが好ましい。pHを中性にする物質を含む溶液としては、pH約5.0~約9.0の、リン酸またはTris塩酸塩からなる群から選ばれる溶液が、望ましい。
タンパク質がフェリチンの場合、pHを5.0とすると再会合することが知られている(Biochemistry 1987, 26, 1831-1837)ことから、再会合用溶液のpHは、約5.0~約9.0とするのが好ましく、約7.0~約9.0とするのがより好ましい。pHを酸性にする物質としては、内包させたい物質との競合を避けるため低分子であることが望ましいため塩酸が好ましい。
再会合手段は、当業者であれば、再会合させた超分子構造の濃度および液量に応じて適宜選択することができるが、安定的な超分子構造を効率よく得るため、工程(I-1)が好ましい。
流路の断面形状は特に限定されないが、V字型を用いるとミス会合体を低減させることができる。
流路の内径は特に限定されず、例えば、約0.1~約1.0mmのものを好適に用いることができる。内径を約0.2~約0.5mmとすると、さらにミス会合体比率を低減に有利となる。
V字型で内径約0.2~約0.5μmの流路を備えたフローマイクロミキサーを用いると、さらにミス会合体が低減する。。
各流路の長さは適宜設定することができるが、約10~約20000μmとするのが好ましく、約100~約5000μmとするのがさらに好ましく、約200~約3000μmであると、充分な混合性能が得られるため好ましい。なお、各流路の長さは、互いに同一でも異なっていても良いが、同一であるのが好ましい。
流路の材質としては、例えば、SUS等の無機材料や、ポリテトラフルオロエチレン(PTFE)等の有機材料を用いることができる。
本発明で用いるフローマイクロミキサーとしては、例えばFraunhofer IMM社やYMC社、三幸精機工業社から市販されているのを利用することができる。
フローマイクロミキサーを出た混合液は、レシーバータンクに送液される。超分子構造を再び形成し、その中心部に生理活性物質を含むタンパク質は、限外ろ過やカラムクロマトグラフィー技術により、レシーバータンクから回収することができる。
工程(I)において用いるサブユニットは、このような会合体タンパク質を構成する1本のポリペプチド鎖からなる。サブユニットは、適当な溶剤に溶解させて溶液の状態で使用する。このときの溶剤としては、ジメチルスルホキシド(DMSO)、N,N-ジメチルホルムアミド(DMF)、アセトニトリル、エタノール等が挙げられる。このうち、両親媒性をもつDMSO、DMFが好ましい。このときのサブユニットの濃度を約0.1~約50g/Lとすると、粘度が低い溶液の取り扱いが容易なため好ましい。
本発明の製造方法は、工程(I)の前に、超分子構造を有するタンパク質を脱会合させる工程を含んでもよい。すなわち、会合体の状態のタンパク質を、個々のサブユニットに解離させる工程を含んでも良い。この工程を、工程(II)と称する。この工程において得られたサブユニットを、工程(I)において使用することができる。本発明者らの知見によれば、脱会合に要する時間が再会合後の会合体の収率に与える影響は、再会合に要する時間が再会合後の会合体の収率に与える影響よりも大きくないと考えられる。したがって、脱会合は、バッチでもフローでもよい。
タンパク質がフェリチンの場合、pHを2.5とすると脱会合することが知られている(Biochemistry 1987, 26, 1831-1837)ことから、溶液のpHは、約1.5~約3.0とするのが好ましく、約1.5~約2.5とするのがより好ましい。pHを酸性にする物質としては、pHの制御に優れるグリシン塩酸塩が好ましい。
フェリチンは、pH約10~12で可逆的に分解できる、との報告がある(Yi Gou, et. al., Frontiers in Pharmacology, 2018 Apr 27; 9:421)。したがって、タンパク質がフェリチンの場合、工程(II-1)において、生理活性物質を内包させる前のタンパク質会合体を含む溶液のpHを約10~12にすることによっても、フェリチンをそのサブユニットに脱会合することができる。
経験的にアルカリ性よりも酸性のほうが脱会合に有利であることから、pH約1.5~3.0の溶液を用いて脱会合する方が好ましい。
脱会合手段は、当業者であれば、フェリチンの濃度、脱会合を行うスケールに応じて適宜選択することができるが、溶剤除去の手間や、溶剤残留による影響を考慮すると、工程(II-1)の方が好ましく、pHを酸性にする方法のほうがより好ましい。
内包させる生理活性物質としては、siRNA(small interfering RNA)、DNA、オリゴペプチド等の、質量平均分子量が例えば1,000~20,000程度の物質が挙げられる。このうち、フェリチン内部の空洞が7nm程度であるため、生理活性物質の質量平均分子量は1,000~15,000程度がより好ましく、1,000~10,000程度が更に好ましい。
会合体により空洞の大きさが異なるため、内包させるのに適した生理活性物質が異なる。例えば、フェリチンの空洞の大きさは約7nmであるため、ペプチドおよびDNA, siRNAが適している。
生理活性物質は、溶液の状態でタンパク質に接触させ、内包させる。生理活性物質溶液を構成する溶剤としては、トリス緩衝液等が挙げられる。このうち、酸による加水分解を抑制するため、中性であることが好ましい。生理活性物質溶液の濃度が約1~約100μMであると、生理活性物質の凝集を抑制するので好ましい。
脱会合をFMMにより行う場合、回収した生理活性物質を含む溶液を、工程(I)におけるサブユニット溶液の流れ、あるいは、工程(II-1)におけるpH約1.5~約3.0程度の溶液又はpH10~12程度の溶液の流れ、工程(II-2)における溶剤の流れ、又は生理活性物質を含まないタンパク質会合体の溶液の流れに合流させることにより、生理活性物質のタンパク質会合体への内包を連続的に行うこともできる。内包させる生理活性物質が、脱会合に用いる酸や塩基や溶剤に弱い場合、工程(I)におけるサブユニット溶液の流れか、又は生理活性物質を含まないタンパク質会合体の溶液の流れに合流させると、生理活性物質が酸等に触れる時間を短くできるので好ましい。
フェリチン内部に含まれるDNAの内包量は、HPLC-ICP-MSを用いて、フェリチン内部に含まれるDNA量中のP濃度を定量することにより測定した。なお、HPLCは高速液体クロマトグラフィー、ICP-MSは誘導結合プラズマ質量分析計の略語である。各分析条件は以下に示す。
ミス会合体比率分析方法と同じ方法で回収したフェリチンに対して0.2N 塩酸(HCl)溶液を滴下しpHを2以下にして再度フェリチンを脱会合した。脱会合した溶液を再度ゲル濾過クロマトグラフィーに供して内包されていた成分を定量した。
溶液dは、1000mMトリス塩酸塩バッファー(pH9.0)を、超純水にて希釈して調製した350mMトリス塩酸塩バッファーである。
なお、フェリチンは特許文献1に開示されている方法にしたがって自社で発酵精製したフェリチン(ロットA)を用いた。
参考例1~7(フェリチン1g/Lで、FMMの再会合流速検討)
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンを含有する10mMトリスバッファー溶液aを、溶液bと混合してpHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。各溶液の流速は、5mL/minとした。これにより、フェリチン濃度が1g/L、グリシン塩酸塩濃度が50mMの脱会合溶液cを得た。
内径500μmのV字強制接触型マイクロミキサーを用いて、脱会合溶液cを、溶液dと所定の流速で混合してpHを約7.3とすることにより、フェリチンの再会合を行った。得られた溶液を上記方法及び条件により分析し、ミス会合体比率を測定した。溶液c及び溶液dの流速、並びにミス会合体比率を表1に示す。
溶液aのフェリチン濃度を6g/Lに変更したこと以外は参考例1~7と同様にしてフェリチンの脱会合及び再会合を行った。なお、脱会合溶液c中のフェリチン濃度は3g/Lである。溶液c及び溶液dの流速、並びにミス会合体比率を表2に示す。
溶液aのフェリチン濃度を10g/Lに変更したこと以外は参考例1~7と同様にしてフェリチンの脱会合及び再会合を行った。なお、脱会合溶液c中のフェリチン濃度は5g/Lである。溶液c及び溶液dの流速、並びにミス会合体比率を表3に示す。
2g/Lのフェリチンを含有する10mMトリスバッファー溶液aの100μLをエッペンチューブにとり、100μLの溶液bを加えて混合し、pHを約2.3~2.5にすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/Lの脱会合溶液cを得た。なお、特に記載の無い限り、実施例の欄において、最終的な脱会合溶液cの濃度は、バッチ法とフローマイクロミキサー法とで同じになるようにした。
200μLの脱会合溶液cをエッペンチューブにとり、そこに、50μLの溶液dを加えて混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。その結果、ミス会合体比率は、10.1%となった。
10g/Lのフェリチンを含有する10mMトリスバッファー溶液aの1mLをエッペンチューブにとり、1mLの溶液bを加えて混合し、pHを約2.3~2.5にすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が5g/Lの脱会合溶液cを得た。
2mLの脱会合溶液cをファルコンチューブにとり、500μLの溶液dを加えて混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。この時のミス会合体比率は31.0%であった。
10g/Lのフェリチンを含有する10mMトリスバッファー溶液aの5mLをビーカーにとり、5mLの溶液bを加え、マグネチックスターラーで撹拌してpHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が5g/Lの脱会合溶液cを得た。
10mLの脱会合溶液cが入ったビーカーに、2mLの溶液dを加え、マグネチックスターラーで撹拌してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて実施した。この時のミス会合体比率は33.9%であった。
一方で、FMMを用いた参考例1~14について、横軸に再会合時の流速の総和、縦軸にミス会合体比率をプロットすると図2のようになる。流速の総和が小さい範囲では、ミス会合体比率が多いが、流速の増加にしたがいミス会合体比率が減少し、流速の総和が10mL/min以上になると、ミス会合体比率は一定となる。すなわち、バッチ法においては液量増加に伴いミス会合体比率が増加するのに対し、FMMを用いた場合は、流速の総和を上げてもミス会合体比率が増加しないばかりか、実施例15及び16の様な小スケールで実施した場合と同等のミス会合体比率を得ることができる。
参考例5と同様の実験をさらに二度行い、そのミス会合体比率を測定したところ、それぞれ11.3%、9.2%であった。このことから、FMMは再現性にも優れており、生産プロセスにおいても、バッチ法と比較して優位である。
2g/Lのフェリチンを含有する10mMトリスバッファー溶液aの1mLをエッペンチューブにとり、1mLの溶液bを加えて混合し、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/Lの脱会合溶液cを得た。
2mLの脱会合溶液cをファルコンチューブにとり、500μLの溶液dを静かに加えることで、緩やかにpHを約7.3に調整した。これにより、フェリチンの再会合をバッチ法にて行った。ミス会合体比率を測定すると67.1%であり、緩やかなpH調整、すなわち混合の速さがミス会合体比率増加に影響することが明らかである。
以下に開示する参考例21~24はあらたに特許文献1に記載の方法に従って準備した参考例1~20とは異なるロット(ロットB)のフェリチンを用いた。
参考例21(比較用バッチ、フェリチン1g/L、200μL);参考例15のLot違い
2g/Lのフェリチンを含有する10mMトリスバッファー溶液aの100μLをエッペンチューブにとり、100μLの溶液bを加え、混合して、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。フェリチン濃度が1g/Lの脱会合溶液cを得た。
200μLの脱会合溶液をエッペンチューブにとり、50μLの溶液dを加えてpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。その結果、ミス会合体比率は、3.6%となった。
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンを含有する10mMトリスバッファー溶液aを、溶液bと混合してpHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。各溶液の流速は、5mL/minとした。これにより、フェリチンが1g/L、グリシン塩酸塩が50mMの脱会合溶液cを得た。
内径500μmのV字強制接触型マイクロミキサーを用いて、脱会合溶液cを、溶液dと所定の流速で混合してpHを約7.3とすることにより、フェリチンの再会合を行った。溶液c及び溶液dの流速、並びにミス会合体比率を表4に示す。
参考例25(比較用バッチ、フェリチン1g/L、2mL)
2g/Lのフェリチンを含有する10mMトリスバッファー溶液aの1000μLをエッペンチューブにとり、1000μLの溶液bを加えて混合することにより、pHを約2.3~2.5とした。その結果、フェリチン濃度が1g/Lの脱会合溶液cを得た。このようにして、フェリチンの脱会合をバッチ法にて行った。
2000μLの脱会合溶液cをエッペンチューブにとり、500μLの溶液dを加えてpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて実施した。その結果、ミス会合体比率は、17.0%となった。
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンを含有する10mMトリスバッファー溶液aと、溶液bを混合してpHを約2.3~2.5とすることにより、フェリチンの脱会合を行った。各溶液の流速は、5mL/minとした。これにより、フェリチンが1g/L、グリシン塩酸塩が50mMの脱会合溶液cを得た。
2000μLの脱会合溶液cをエッペンチューブにとり、500μLの溶液dを加えてpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて実施した。その結果、ミス会合体比率は、11.2%となった。
2g/Lのフェリチンを含有する10mMトリスバッファー溶液aの2000μLをエッペンチューブにとり、2000μLの溶液bを混合してpHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。フェリチン濃度が1g/Lの脱会合溶液cを得た。
脱会合溶液cと、溶液dを、V字強制接触型マイクロミキサーを用いて混合することで、pHを約7.3とし、フェリチンの再会合をおこなった。この時、脱会合溶液cの流速は8mL/min、溶液dの流速は2mL/minとした。その結果、ミス会合体比率は、8.0%となった。
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンを含有する10mMトリスバッファー溶液aと、溶液bを混合してpHを約2.3~2.5とすることにより、フェリチンの脱会合を行った。各溶液の流速は、5mL/minとした。これにより、フェリチンが1g/L、グリシン塩酸塩が50mMの脱会合溶液cを得た。
脱会合溶液cと、溶液dを、V字強制接触型マイクロミキサーを用いて混合することで、pHを約7.3とし、フェリチンの再会合をおこなった。この時、脱会合溶液cの流速は8mL/min、溶液dの流速は2mL/minとした。その結果、ミス会合体比率は、6.3%となった。
参考例25~28の結果を抜粋してまとめると、図3の様な関係になる。
参考例1~28の条件一覧を表5に示した。
実施例1~2(DNA内包FMM実施例、フェリチン1g/L)
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンおよび10μMのssDNA(商品名「K3 Et-Free」、株式会社ジーンデザイン製)を含有する10mMトリスバッファー溶液aを、溶液bと混合して、pHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。これにより、フェリチン濃度が1g/L、dsDNA濃度が5μM、グリシン塩酸塩濃度が50mMの脱会合溶液cを得た。
内径500μmのV字強制接触型マイクロミキサーを用いて、脱会合溶液cと、溶液dとを所定の流速で混合してpHを約7.3とすることにより、フェリチンの再会合を行った。溶液c及び溶液dの流速、並びに再会合体中のdsDNA濃度を表6に示す。
2g/Lのフェリチンおよび10μMのssDNA(商品名「K3 Et-Free」、株式会社ジーンデザイン製)を含有する10mMトリスバッファー溶液aの200μLをエッペンチューブにとり、200μLの溶液bを加えて混合し、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/L、dsNDA濃度が5μMの脱会合溶液cを得た。
400μLの脱会合溶液cをエッペンチューブにとり、100μLの溶液dを混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。フェリチンに内包されたDNA濃度を測定したところ、定量限界(0.06μM)未満であった。
2g/Lのフェリチンおよび10μMのssDNA(商品名「K3 Et-Free」、株式会社ジーンデザイン製)を含有する10mMトリスバッファー溶液aの1mLをファルコンチューブにとり、1mLの溶液bを加えて混合し、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/L、dsNDA濃度が5μMの脱会合溶液cを得た。
400μLの脱会合溶液cをエッペンチューブにとり、500μLの溶液dを混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。フェリチンに内包されたDNA濃度を測定したところ、定量限界(0.06μM)未満であった。
実施例3(siRNA内包FMM実施例、フェリチン1g/L)
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンおよび50μMのsiRNA(EUROFINSGENOMICS製、配列番号1の塩基配列(GGCGCUGCCAAGGCUGUGGGCAAGGUC)を有する。)を含有する10mMトリスバッファー溶液aを、溶液bと混合して、pHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。これにより、フェリチン濃度が1g/L、siRNA濃度が10μM、グリシン塩酸塩濃度が50mMの脱会合溶液cを得た。
内径500μmのV字強制接触型マイクロミキサーを用いて、脱会合溶液cと、溶液dとを所定の流速で混合してpHを約7.3とすることにより、フェリチンの再会合を行った。溶液c及び溶液dの流速、並びに上記の分析によって得られた再会合体中のsiRNA濃度を表7に示す。なお、本明細書において、「FTH1個」はFTHのサブユニットの24量体を指し、FTH1モルと同義である。
2g/Lのフェリチンおよび50μMのsiRNA(EUROFINSGENOMICS製、配列番号1の塩基配列を有する。)を含有する10mMトリスバッファー溶液aの400μLをエッペンチューブにとり、400μLの溶液bを加えて混合し、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/L、siRNA濃度が10μMの脱会合溶液cを得た。
800μLの脱会合溶液cをエッペンチューブにとり、200μLの溶液dを混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。フェリチンに内包されたsiRNA濃度を測定した。結果を表7に示す。
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンおよび50μMのsiRNA(Thermo Fisher Scientific製、配列番号2の塩基配列(GACCUUGCCCACAGCCUUGGCAGCGUC)を有する。)を含有する10mMトリスバッファー溶液aを、溶液bと混合して、pHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。これにより、フェリチン濃度が1g/L、siRNA濃度が10μM、グリシン塩酸塩濃度が50mMの脱会合溶液cを得た。

実施例5(FAM-venepeptide内包FMM実施例、フェリチン1g/L)
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンおよび0.2mMのFAM-venepeptide(EUROFINSGENOMICS製、配列番号3のアミノ酸配列をフルオレセインで蛍光標識したもの(FAM-MNVITNLLAGVVHFLGWLV)。蛍光標識は「FAM」で表わす。以下同じ。)を含有する10mMトリスバッファー溶液aを、溶液bと混合して、pHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。これにより、フェリチン濃度が1g/L、FAM-venepeptide濃度が62.5μM、グリシン塩酸塩濃度が50mMの脱会合溶液cを得た。
内径500μmのV字強制接触型マイクロミキサーを用いて、脱会合溶液cと、溶液dとを所定の流速で混合してpHを約7.3とすることにより、フェリチンの再会合を行った。溶液c及び溶液dの流速、並びに上記の分析によって得られた再会合体中のFAM-venepeptide濃度を表9に示す。
2g/Lのフェリチンおよび0.2mMのFAM-venepeptide(EUROFINSGENOMICS製、配列番号3のアミノ酸配列をフルオレセインで蛍光標識したもの。)を含有する10mMトリスバッファー溶液aの400μLをエッペンチューブにとり、400μLの溶液bを加えて混合し、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/L、FAM-venepeptide濃度が125μMの脱会合溶液cを得た。
800μLの脱会合溶液cをエッペンチューブにとり、200μLの溶液dを混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。フェリチンに内包されたFAM-venepeptide濃度を測定した。結果を表9に示す。
実施例6(FAM-GFIL8内包FMM実施例、フェリチン1g/L)
内径500μmのV字強制接触型マイクロミキサーを用いて、2g/Lのフェリチンおよび1.0mMのFAM-GFIL8(EUROFINSGENOMICS製、配列番号4のアミノ酸配列(GFILGFIL)をフルオレセインで蛍光標識したもの。)を含有する10mMトリスバッファー溶液aを、溶液bと混合して、pHを約2.3~2.5にすることにより、フェリチンの脱会合を行った。これにより、フェリチン濃度が1g/L、FAM-GFIL8濃度が62.5μM、グリシン塩酸塩濃度が50mMの脱会合溶液cを得た。
内径500μmのV字強制接触型マイクロミキサーを用いて、脱会合溶液cと、溶液dとを所定の流速で混合してpHを約7.3とすることにより、フェリチンの再会合を行った。溶液c及び溶液dの流速、並びに上記の分析によって得られた再会合体中のFAM-GFIL8濃度を表10に示す。
2g/Lのフェリチンおよび1.0mMのFAM-GFIL8(EUROFINSGENOMICS製、配列番号4のアミノ酸配列をフルオレセインで蛍光標識したもの。)を含有する10mMトリスバッファー溶液aの400μLをエッペンチューブにとり、400μLの溶液bを加えて混合し、pHを約2.3~2.5とすることにより、フェリチンの脱会合をバッチ法にて行った。これにより、フェリチン濃度が1g/L、FAM-GFIL8濃度が125μMの脱会合溶液cを得た。
800μLの脱会合溶液cをエッペンチューブにとり、200μLの溶液dを混合してpHを約7.3とすることにより、フェリチンの再会合をバッチ法にて行った。フェリチンに内包されたFAM-GFIL8濃度を測定した。結果を表10に示す。

Claims (11)
- 生理活性物質を内包する超分子構造を有するタンパク質の製造方法であって、
(I)超分子構造を構成するタンパク質のサブユニットと、生理活性物質と、前記サブユニットから超分子構造を有するタンパク質を形成させるための溶液とを、フローマイクロミキサー中で接触させることを含む、
前記製造方法。 - 超分子構造を有するタンパク質がフェリチンである、請求項1記載の製造方法。
- 工程(I)において、前記サブユニットの流速と前記溶液の流速との合計が、約10mL/min以上である、請求項1又は2記載の製造方法。
- 前記サブユニットが、(II-1)超分子構造を有するタンパク質を含む溶液のpHを酸性又は塩基性に変化させることにより得られる、請求項1~3のいずれか1項記載の製造方法。
- 工程(II-1)において用いる溶液が、pH約1.5~約3.0又はpH約10~12の溶液である、請求項4に記載の製造方法。
- 工程(I)において用いるサブユニットから超分子構造を有するタンパク質を形成させるための溶液が、pH約5.0~約9.0の溶液である、請求項1~5のいずれか1項に記載の製造方法。
- 前記サブユニットが、(II-2)超分子構造を有するタンパク質を含む溶液に溶剤を添加することにより得られる、請求項1~3のいずれか1項記載の製造方法。
- 前記工程(II-2)において用いる溶剤が、ジメチルスルホキシド(DMSO)、N,N-ジメチルホルムアミド(DMF)、アセトニトリル、及びエタノールからなる群から選ばれる、請求項7に記載の製造方法。
- 工程(I)において用いる溶液が、Tris緩衝液、HEPES緩衝液、リン酸緩衝液、ホウ酸緩衝液、クエン酸緩衝液、炭酸緩衝液、及びグリシン緩衝液からなる群から選ばれる、請求項7又は8に記載の製造方法。
- 工程(II-1)又は(II-2)を、フローマイクロミキサーを用いて行う、請求項4~9のいずれか1項記載の製造方法。
- 前記生理活性物質が、質量平均分子量約1,000~約20,000のsiRNA、DNA、オリゴペプチドまたはこれらの組合せからなる群から選ばれる、請求項1~10のいずれか1項記載の製造方法。
