Abstract
본 발명은 피로갈롤 (PG) 모이어티 (pyrogallol moiety)로 컨쥬게이션 된 히알루론산 (HA, hyaluronic acid)을 기재로 하는 하이드로젤 및 이를 포함하는 면역증강용 조성물에 관한 것이다. 본 발명은 상기 면역증강용 조성물과 항원을 포함하는 백신 조성물에 관한 것이다. 또한 본 발명은 상기 면역증강용 조성물과 단백질, 항체 또는 항원을 포함하는 숙주의 증대된 면역반응을 유도하는 약물전달용 조성물에 관한 것이다.
Description
본 발명은 히알루론산 유도체를 기재로 하는 면역증강제 및 이의 용도에 관한 것이다.
백신 또는 면역항암 치료제 등을 사용함에 있어서, 이들 백신 또는 치료 약물의 체내 효력을 유지하고 증강시키기 위한 방법이 요구된다. 이러한 목적으로, 하이드로젤 (hydrogel), 에멀젼 (emulsion), 리포좀 (liposome), 무기 나노입자 (inorganic nanoparticle-based adjuvant) 등과 같은 면역증강제(애주번트, 보체, 또는 면역보조제)가 이용되고 있다. 면역증강제는 체액성 면역반응, 세포매개성 면역반응, 또는 이들의 조합에 의한 다양한 메카니즘으로 면역계를 향상시킨다. 면역증강제는 이의 작용 메커니즘에 따라 항원전달계열의 면역증강제와 면역자극성 면역증강제로 구분될 수 있다. 구체적으로, 항원전달체로 작용하는 면역증강제는 주로 non-immunostimulatory 입자로 구성되고 항원을 적절한 면역세포에 전달한다. 알루미늄 염, 리포좀 (liposome), 에멀션 (emulsion), 사포닌, 나노입자 면역증강제, 점막 면역증강제, 면역증강제 시스템 등이 이에 포함된다. 면역조절형 면역증강제(Immunostimulatory adjuvant)는 면역 활성이 있는 물질로 구성되고 면역 세포를 직접 자극하여 내재 면역을 활성화 하여 항원에 대한 적응 면역을 유도한다. TLRs, NOD-like receptors (NLRs), C-type lectins, 및 RIG-I-like receptors와 같은 패턴인식수용체(Pattern recognition receptors; PRRs)를 효과적으로 자극하는 작용제(agonist)가 이에 포함된다. 따라서, 면역증강제에 의한 활성은 예를 들어 다음을 포함한다: 항원제시세포 (APC, antigen presenting cell) 및 호중구 증가, 백신 항원의 APC에 의한 섭취 및 제시, APC로부터의 단백질 분비, 면역세포 침윤 (recruitment), 표적화 및 활성화, 유도된 면역반응 조절작용, 및 항원의 분해 또는 제거 억제. 또한, 면역증강제는 항암 치료 효과를 높이기 위해 면역관문(체크 포인트)(예를 들어, PD-1, PD-L1, CTLA-4와 같은 면역관문 단백질) 억제제와 함께 병용 투여될 수 있다.
현재 상용화된 면역증강제는 알루미늄 염, MF59, AS03, AF03 및 AS04 등이 있다. MF59는 스쿠알렌과 계면활성제 Tween80 및 Span85로 구성된 수중유 (oil-in-wtaer) 에멀젼이다. AS03는 스쿠알렌, 폴리소르베이트80 및 α-토코페롤(비타민 E)로 구성된 수중유 에멀젼이다. AS04는 2개의 알려진 보조제, 3-O-데사실-4'-모노포스포릴 지질 A (3-O-desacyl-4′-monophosphoryl lipid A)(MPL) 및 불용성 알루미늄 염으로 구성된다. 알루미늄 염인 알럼 (alum) 면역증강제는, 국소 항원 농도를 증가시키고 항원을 대식세포와 같은 항원제시세포(APC; antigen-presenting cell)의 항원 흡수 능력(phagocytosis)을 증가시키며, 항체 생성과 같은 체액성 면역반응을 강화하는 것을 특징으로 한다. 그러나, 알루미늄 염 면역증강제는 항체-의존적 세포독성(antibody dependent cellular cytotoxicity; ADCC)이나, CTL(Cytotoxic T lymphocytes)에 의한 세포성 면역 반응을 거의 유도하지 못하여, 백신 보호 효능을 충분히 달성하지 못하는 한계가 있다.
생체적합소재로서 히알루론산에 대한 다양한 연구 및 개발이 수행되어 왔다. 친수성의 히알루론산은 생체유래 고분자로서 부작용이 거의 발생하지 않고, 히알루론산 효소(hyaluronidase)에 의해 일정 기간 경과 후 분해된다. 따라서, 의료용 생체재료, 조직공학용 지지체 및 약물전달용 소재로서 분자로 폭넓게 활용하고 있다.
본 발명자는 상기 종래기술의 문제점을 개선하고자 예의 노력한 결과, 특정 화합물로 수식한 히알루론산 유도체가 면역증강제로서 우수한 특성이 있음을 확인하고 본 발명을 완성하였다.
이와 같이, 본 발명의 목적은, 히알루론산을 피로갈롤기로 수식하여 제조된 히알루론산 유도체를 포함하는 면역증강제 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 히알루론산 유도체를 가교시켜 제조한 히알루론산 유도체 하이드로젤을 포함하는 면역증강제 조성물을 제공하는 것이다.
본 발명의 다른 목적은 히알루론산 유도체가 가교된 구조를 갖는 히알루론산 유도체 하이드로젤 미세입자를 포함하는 면역증강제 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 히알루론산을 피로갈롤기로 수식하여 제조된 히알루론산 유도체와 항원을 포함하는 백신 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 히알루론산 유도체를 가교시켜 제조한 히알루론산 유도체 하이드로젤과 항원을 포함하는 백신 조성물을 제공하는 것이다.
본 발명의 다른 목적은 히알루론산 유도체가 가교된 구조를 갖는 히알루론산 유도체 하이드로젤 미세입자와 항원을 포함하는 백신 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 히알루론산 유도체 및/또는 하이드로젤, 및 항원, 단백질 또는 항체를 포함하는 증대된 면역반응을 유도하는 약물전달용 조성물을 제공하는 것이다.
본 발명이 이루고자 하는 기술적 과제는 이에 제한되지 않으며, 상기 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기와 같은 본 발명의 목적을 달성하기 위하여, 본 발명은 히알루론산 (hyaluronic acids)과 5'-하이드록시도파민(5'-hydroxydopamine)을 반응시켜 제조한 갈롤기로 수식된 히알루론산 유도체를 포함하는 면역증강제 조성물을 제공한다. 또한, 상기 히알루론산 유도체가 가교된 히알루론산 하이드로젤을 포함하는 면역증강제 조성물을 제공한다. 또한, 상기 히알루론산 유도체 및/또는 히알루론산 하이드로젤, 및 항원을 포함하는 백신 조성물을 제공한다. 또한, 상기 히알루론산 유도체 및/또는 하이드로젤; 및 항원, 단백질 또는 항체를 포함하는 증대된 면역반응을 유도하는 약물전달용 조성물을 제공한다.
본 발명의 일 구현예에서, 상기 히알루론산 유도체는 하기 화학식 1일 수 있다. 여기서, 상기 히알루론산 유도체의 분자량은 1,000 Da 내지 2,000,000 Da일 수 있고, 상기 히알루론산 유도체의 갈롤기 치환율은 0.1% 내지 50%일 수 있다.

본 발명의 다른 일 구현예에서, 상기 가교는 산화제 또는 pH 조절제를 첨가하여 수행될 수 있다. 상기 산화제는 과요오드산나트륨, 과산화수소, 겨자무과산화효소 또는 타이로시나아제일 수 있고, 상기 pH 조절제는 수산화나트륨, 수산화리튬, 수산화칼륨, 수산화루비듐, 수산화세슘, 수산화마그네슘, 수산화칼슘, 수산화스트론튬 또는 수산화바륨일 수 있으나, 이에 제한되는 것은 아니다. 이와 관려된 일 구현예에서, 상기 가교는 체내 활성 산소에 의해 수행될 수 있다.
본 발명의 다른 일 구현예에서, 상기 산화제를 첨가하여 가교시키는 단계는, 히알루론산 유도체간 하기 화학식 2의 가교 결합을 형성할 수 있다.

또한, 본 발명은 상기 화학식 1의 히알루론산 유도체를 가교시켜 제조된 히알루론산 하이드로젤로서, 히알루론산 유도체가 상기 화학식 2 또는 화학식 3의 가교 결합을 포함하는 것인 히알루론산 하이드로젤을 포함하는 면역증강제 조성물을 제공한다.
또한, 본 발명은 상기 화학식 1의 히알루론산 유도체를 가교시켜 제조된 히알루론산 하이드로젤 및 항원을 포함하는 백신 조성물로서, 상기 히알루론산 유도체가 상기 화학식 2 또는 화학식 3의 가교 결합을 포함하는 것인 백신 조성물을 제공한다.
또한, 본 발명은 상기 화학식 1의 히알루론산 유도체를 가교시켜 제조된 히알루론산 하이드로젤, 및 항원, 단백질 또는 항체를 포함하여 증대된 면역반응을 유도하는 약물전달용 조성물을 제공한다.
본 발명의 일 실시예에 따르면, 상기 본 발명의 면역증강제는 체액성 면역반응 및/또는 세포성 면역반응을 증대시키며, 항원 또는 약물의 서방출(sustained release)을 제공한다.
본 발명의 다른 일 실시예에서, 상기 히알루론산 유도체의 분자량은 1,000 Da 내지 20,000,000 Da일 수 있고, 상기 히알루론산 유도체의 피로갈롤기 치환율은 0.01% 내지 70%, 바람직하게는 1% 내지 30%이며, 더욱 바람직하게는 2% 내지 20%일 수 있다. 본 발명의 다른 실시예에서, 상기 히알루론산 유도체는, 전체 조성물에 대하여 0.01%(w/v) 내지 80%(w/v)로 함유될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 또 다른 실시예에서, 상기 면역증강제 조성물은 체외에서 액체 상태이고, 체내 투여 후 겔을 형성할 수 있다.
본 발명의 또 다른 실시예에서, 상기 하이드로겔은 직경 수 ㎚ ~ 수십 ㎛를 갖도록 건조된 미세입자 일 수 있으며, 상기 미세입자는 항원 및/또는 약물을 포함할 수 있다.
본 발명의 다른 일 실시예에서, 항원 또는 면역치료를 위한 약물 외에 약제학적으로 허용되는 부형제, 담체, 항산화제, 비타민, 및 이들의 조합으로 이루어진 군으로부터 선택되는 어느 하나 이상의 성분을 추가로 포함할 수 있다.
본 발명에 따른 히알루론산 유도체 또는 이를 가교시켜 제조한 히알루론산 하이드로젤은 사람 또는 동물의 면역화를 위한 백신 및/또는 약물과 함께 사용되어 세포성 면역 및/또는 체액성 면역반응의 증대를 위해 사용될 수 있다.

본 발명의 히알루론산 유도체 및 상기 히알루론산 유도체가 가교된 하이드로젤은, 본 출원인의 선행특허(한국특허등록공보 제10-2099981B1호)에 기술된 방법에 따라 제조하였다.
본 발명에서 사용되는 용어, "히알루론산(Hyaluronic acid, HA)"은 D-글루쿠론산 (D-glucuronic acid, GlcA) 및 N-아세틸-D-글루코사민 (GlcNAc) 기 β-1,3-글라이코시드 결합(β 1,3-glycosidic bond)에 의해 연결되어 있는 다이사카라이드를 반복단위로 포함하는 고분자량의 선형 폴리사카라이드로서, 히알루론산과 그의 염을 모두 지칭하며, 이에 제한되는 것은 아니나, 상기 염으로는 나트륨염, 칼륨염, 마그네슘염, 칼슘염, 알루미늄염 등이 예시된다.
본 발명에서 사용된 용어, "면역증강제는"는 애주번트 (adjuvant), 보체, 면역보조제와 혼용하여 같은 의미로 사용되었다. 면역증강제는 항원 또는 면역치료제, 예컨대, 면역항암제와 같이 면역 치료를 목적으로 사용되는 약물과 함께 사용되어, 개체의 체액성 및/또는 세포성 면역반응을 증대시키는 작용을 하는 것을 의미한다.
히알루론산의 다이사카라이드 반복단위(repeating unit)는 하기 화학식 4와 같으며, 상기 반복단위는 1 내지 100,000개일 수 있으나, 이에 제한되는 것은 아니다.

본 발명에서 사용되는 용어, "히알루론산 유도체" 또는 "히알루론산-피로갈롤 컨쥬게이트"는 히알루론산 또는 그의 염의 글루쿠론산의 골격에 갈롤기가 도입되어 있는 히알루론산 또는 그의 염을 모두 포함하는 것으로 해석된다.
일 구체예로서, 상기 화학식 4의 다이사카라이드 반복단위의 말단, 구체적으로, 이의 카르복시기와 5'-하이드록시도파민간 반응에 의해 제조될 수 있다. 상기의 반응에 의해 제조된 히알루론산 유도체는 하기 화학식 5의 반복단위를 하나 이상 포함하며, 하기 화학식 1과 같이 나타낼 수 있다.

또한, 상기 히알루론산 유도체의 분자량은 1,000 Da 내지 10,000,000 Da 일 수 있고, 상기 히알루론산 유도체의 갈롤기 치환율은 0.1% 내지 50%일 수 있으나, 이에 제한되는 것은 아니다. 상기 분자량은 중량평균분자량 또는 수평균분자량일 수 있다.
상기 "치환율"은 히알루론산 또는 그의 염의 특정 기능기가 갈롤기로 대체되거나 수식되는 것을 의미한다. 갈롤기로의 치환율은 전체 히알루론산 반복단위 중 갈롤기가 도입된 반복단위의 비율로 정의되며, 정의상 0 초과 1 이하의 수치, 또는 0% 초과 100% 이하의 수치, 또는 0몰% 초과 100몰% 이하의 수치로 표현될 수 있다.
본 발명에서 사용되는 용어, "하이드로젤"은 충분한 양의 수분을 보유하고 있는 친수성 고분자의 3차원적 구조를 의미하며, 본 발명의 목적상, 상기 하이드로젤은 갈롤기로 수식된 히알루론산 유도체 간에 형성된 하이드로젤을 나타낸다. 또한, 본 발명에서 "하이드로젤"은 건조되어 파우더 또는 미세입자 형태로 사용되는 것으로 이해될 수 있다.
또한, 본 발명에서, "개체"는 면역증강을 필요로 하는 대상으로서, 사람을 포함한 동물로 정의 된다.
본 발명의 용어 "약학적으로 허용가능한"이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 조성물을 말한다.
본 발명의 조성물에 함유될 수 있는 상기 담체, 부형제 및 희석제는, 예를 들어 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘실리케이트, 셀룰로즈, 메틸 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 포함하며 이에 제한되는 것은 아니다. 약학적으로 허용되는 담체는, 예를 들어 물, 적합한 오일, 식염수, 수성 글루코스 및 글리콜 등과 같은 비경구 투여용 담체 등이 있다. 또한, 항산화제, 보존제, 충진제, 안정화제, 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등이 더 포함될 수 있을 것이다. 항산화제는 아황산수소나트륨, 아황산나트륨, 아스코르브산, 폴리올, 만니톨, 및 소르비톨로 이루어진 군으로부터 선택될 수 있으며, 상기 항산화제의 양은 전체 필러 조성물 중량에 대하여 바람직하게 0.01 중량% 내지 5.0 중량%, 보다 바람직하게, 0.2 중량% 내지 1.0% 중량%로 함유될 수 있으나, 이에 제한되는 것은 아니다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸- 또는 프로필-파라벤 및 클로로부탄올이 있다.
본 발명의 조성물은 통상적으로 사용되는 면역보조제를 더 포함할 수 있다. 또한, 본 발명의 조성물은 분말, 과립, 정제, 에멀젼, 시럽, 에어로졸, 연질 또는 경질 젤라틴 캅셀, 멸균 주사용액 또는 멸균 분말 형태로 제형화 될 수 있다.
본 발명의 조성물은 피하, 경피, 근육 또는 복강 내로 주입되거나, 마이크로니들을 이용한 경피용 패치 형태로 제작되어 체내로 투여될 수 있을 것이다. 또한, 본 발명의 조성물은 경구 또는 비경구로 투여될 수 있다. 비경우는 예를 들어, 좌제, 경피, 정맥, 복강, 근육내, 병변내, 비강, 척추관내 투여를 포함한다.
본 발명의 면역증강용 조성물은 그 투여방법이나 제형에 따라 필요한 경우, 현탁제, 용해보조제, 등장화제, 흡착방지제, 계면활성화제, 희석제, 부형제, pH 조정제, 무통화제, 완충제 등을 더 포함할 수 있다. 약학적으로 허용되는 담체 및 제제는 문헌(Remington's Pharmaceutical Sciences)에서 확인 할 수 있을 것이다.
본 발명의 조성물은 체중을 기준으로 100 ng/kg ~ 1,000 ㎎/㎏, 1 ~ 500 ㎍/kg, 또는 5 ~ 50 ㎍/kg으로 투여 될 수 있으며, 투여를 필요로 하는 개체의 성별, 연령, 질환의 중증도 등에 따라 조절될 수 있다.
본 발명의 용어 "약학적으로 유효한 양"이란, 의학적 치료에 적용가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
본 발명에서 "항원"은 바이러스 항원, 종양 항원 (tumor antigen) 또는 이들의 조합이 될 수 있다. 구체적으로, 코로나 바이러스, 인플루엔자 바이러스 (influenza virus), 대상포진 바이러스, EBV (Epstein-Barr virus), HAV (hepatitis A virus), HBV (hepatitis B virus), HCV (hepatitis C virus), HDV (hepatitis D virus), HEV (hepatitis E virus), 한탄바이러스 (Hantaan virus), CMV (cytomegalovirus), HIV (human immunodeficiency virus), HPV (human papilloma virus), 소아마비 바이러스 (poliovirus), 에볼라 바이러스 (ebola virus), 로타바이러스 (rotavirus), 댕기열바이러스 (dengue virus), 웨스트나일 바이러스 (West Nile virus), 황열바이러스 (yellow fever virus), 아데노바이러스 (adenovirus), 일본뇌염 바이러스 (Japanese encephalitis virus), BK 바이러스 (BK virus), 천연두 바이러스 (smallpox virus), 지카 바이러스 (Zika virus), 중증열성혈소판감소증후근 바이러스 (SFTS virus), HSV (herpes simplex virus) 등일 수 있으며, 이에 제한되는 것은 아니다.
본 발명의 면역증강제 조성물은 면역반응을 유도하는 항원 또는 상기 항원을 포함하는 백신 조성물과 동시에 투여될 수도 있고, 상기 백신 조성물을 투여하기 이전 또는 이후에 투여될 수도 있다.
상기 히알루론산 하이드로젤의 제조 과정은, 상기 히알루론산 유도체의 가교 반응에 의해 진행될 수 있으며, 상기 가교 반응을 위하여 상기 히알루론산 유도체와 인산완충생리식염수 (PBS; phosphate-buffered saline) 등과 혼합하여 히알루론산 하이드로젤 전구체 용액을 제조하는 단계를 추가로 포함할 수 있다. 이러한 가교는 UV 조사에 의한 화학적 가교, 물리학적 가교 또는 생물학적 가교에 의해 하이드로젤로 형성될 수 있다. 여기서, UV 조사에 의한 화학적 가교로는 광가교 (photo-crosslinking) 또는 반응성 가교제 (reactive crosslinker)를 활용한 가교 등이 있고, 생물학적 가교로는 헤파린과 성장인자의 결합력을 활용한 가교 또는 DNA 등의 상보적 결합을 이용한 가교 등이 있으며, 물리적 가교로는 수소결합에 의한 가교, 소수성(hydrophobic) 상호작용에 의한 가교 또는 정전기적 상호작용을 활용한 가교 등이 있으나, 바람직하게는, 산화제 또는 pH 조절제를 첨가하여 가교시킬 수 있다. 상기 산화제는 과요오드산나트륨, 과산화수소, 겨자무과산화효소 또는 타이로시나아제일 수 있고, 상기 pH 조절제는 수산화나트륨, 수산화리튬, 수산화칼륨, 수산화루비듐, 수산화세슘, 수산화마그네슘, 수산화칼슘, 수산화스트론튬 또는 수산화바륨일 수 있으나, 이에 제한되는 것은 아니다. 체내에서는 생체내 활성 산소에 의해 산화되어 겔을 형성할 수 있다.
일 구체예로서, 상기 산화제를 첨가하여 가교시키는 경우, 히알루론산 유도체간 하기 화학식 2의 가교 결합을 형성한다. 화학식 2에서, HA'는 카르복시기가 아마이드기로 치환된 히알루론산을 나타낸다.

다른 구체예로서, 상기 pH 조절제를 첨가하여 가교시키는 경우, 히알루론산 유도체간 하기 화학식 3의 가교 결합을 형성한다. 화학식 3에서, HA'는 카르복시기가 아마이드기로 치환된 히알루론산을 나타낸다.

본 발명의 일 실시예에서는, 갈롤기로 수식된 히알루론산 유도체의 가교 단계에서 각각 산화제인 과요오드산나트륨 또는 pH 조절제인 수산화나트륨을 이용하여 히알루론산 하이드로젤을 제조하였으며 (제조예 2 참조), 그 결과, 각각의 가교 방식에 따라 제조된 히알루론산 하이드로젤은 우수한 생체 적합성과 함께 가교 속도, 강도, 탄성, 접착력, 분해 양상 등 물리적 특성의 현격한 차이를 확인할 수 있었다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[제조예]
제조예 1: 갈롤기로 수식된 히알루론산 유도체의 제조
도 1에 나타낸 바와 같이, 본 발명의 갈롤기로 수식된 히알루론산 유도체를 제조하였다. 구체적으로, 히알루론산 (MW 200K, Lifecore Biomedical, IL, USA)을 물 (TDW)에 완전히 용해시키고, 1 당량의 NHS (N-hydroxysuccinimide, Sigma, St. Louis, MO, USA)와 1.5 당량의 EDC (1-(3-dimethylaminopropyl)-3-ehtylcarbodiimide hydrochloride, Thermo Scientific, Rockford, IL, USA)를 첨가하였으며, 이로부터 30분 후, PG 모이어티로서 1 당량의 5'-하이드록시도파민 (5-hydroxydopamine, Sigma)을 첨가하여 pH 4 ~ 4.5에서 24시간 동안 반응시켰다. 2개의 다른 몰비의 반응물을 HA-PG 컨쥬게이트 제조에 사용하였다 (HA : EDC : NHS : PG = 1 : 1.5 : 1 : 1 or 2 : 1.5 : 1 : 1). 이후, EDC, NHS, 5'-하이드록시도파민을 PBS 및 물 기반의 투석 (Cellu/Sep (상표명), dialysis membrane (6.8 kDa cut-off, Membrane Filtration Products Inc., Seguin, TX, USA))으로 제거하고, 동결 건조를 통해 용매를 증발시켜 본 발명의 히알루론산 유도체를 제조하였다. 상기 히알루론산 유도체의 합성을 확인하고자, FT-IR (Fourier transform-infrared spectroscopy) (Vertex 70, Bruker, Billerica, MA, USA)및 H-NMR (proton Nuclear magnetic Resonance) (Bruker 400 MHz, Bruker)로 분석한 결과, 도 2a에 나타낸 바와 같이, 약 1,580 cm-1에서 1,700 cm-1 파수 영역에서의 강한 피크를 통해 새롭게 형성된 아마이드 결합을 확인할 수 있었으며, 도 2b에 나타낸 바와 같이, 6.5 ppm 및 3 ppm 부근의 피크를 통해 각각 5'-하이드록시도파민의 방향족의 벤젠고리 및 -CH2CH2-의 구조를 확인할 수 있었다. 상기 결과로부터 본 발명의 히알루론산 유도체는, 히알루론산의 카르복시기와 5'-하이드록시도파민의 아민기간 형성된 아마이드 결합으로 5'-하이드록시도파민이 도입됨을 알 수 있었다. HA 골격에 대한 갈롤기의 치환 정도를 계산하기 위해, HA-PG 컨쥬게이트를 PBS (pH 5)에 1 ㎎/㎖에서 용해시키고 용액의 흡광도를 283 ㎚ (UV-visible spectrophotometer (Cary 100 UV-vis, Varian Inc., Palo Alto, CA, USA)에서 측정하였다. PG로 치환된 HA내의 카르복실시의 %를 5-hydroxydopamine solution (from 1 mg/㎖ concentration)의 연속 희석을 통해 수득한 표준 곡선을 이용하여 계산하였다.
제조예 2: 히알루론산 하이드로젤의 제조
상기 제조예 1의 히알루론산 유도체를 가교 (cross-linking)시켜 히알루론산 하이드로젤을 제조하였으며, 이때, 가교 방식은 산화제인 과요오드산나트륨 (NaIO4) 또는 pH 조절제 (pH 8 ~ 9)인 수산화나트륨 (NaOH)을 각각 이용하였다. 구체적으로, 상기 히알루론산 유도체를 PBS에 용해시킨 후 (1% (w/v), 2% (w/v)), 4.5 mg/㎖의 NaIO4 또는 0.08 M의 NaOH를 히알루론산 유도체 용액 부피 대비 1.5:1 ~ 4:1의 비율로 혼합시키면서 가교를 진행하였으며, 도 3에 나타낸 바와 같이, 이들 각각을 통해 밝은 갈색 또는 푸른색을 띄는 히알루론산 하이드로젤을 제조하였다.
상기 히알루론산 하이드로젤의 가교를 구체적으로 확인하고자, UV-vis (Ultraviolet-visible spectroscopy)으로 분석하였다. NaIO4를 이용한 경우, 도 4에 나타낸 바와 같이, 시간의 경과에 따라 350 ~ 400 nm 파장 영역이 변화됨을 확인할 수 있었으며 (도 4a), 이는 산화 과정에서 중간체인 라디컬의 순간적인 형성과 감소를 의미하는 것으로, 이후 라디컬 중합반응에 의해 바이페놀 (biphenol)이 형성됨을 알 수 있었다 (도 4b). 또한, NaOH를 이용한 경우, 도 5에 나타낸 바와 같이, 시간의 경과에 따라 600 nm 파장 영역이 변화됨을 확인할 수 있었으며 (도 5a), 이는 산화 과정에서 [5+2] tautomerization에 의해 전하 이동 복합체 및 벤조트로폴론 (benzotropolone)이 형성됨을 알 수 있었다 (도 5b).
[실시예]
실시예 1. 마우스 체액성 면역반응의 유도
(1) OVA 항원을 이용한 면역반응 유도
6주령의 암컷, Balb/c 마우스(입수처: 코아텍, 한국)를 그룹당 4마리 씩 5그룹으로 구분하였다. 마우스에 면역을 유도하기 위해, 본 발명의 면역증강제 및 항원 (Ovalbumin (OVA; Sigma-Aldrich))을 혼합한 혼합액을 각 마우스의 양쪽 대퇴부 근육에 50 ㎕씩 총 100 ㎕ 주입하였다. 각 실험균 별 항원과 면역증강제의 주입량은 아래와 같았다.
G1 : PBS 100 ㎕
G2 : OVA (20 ㎍/mouse) in 100 ㎕ PBS
G3 : OVA (20 ㎍/mouse), alum (100 ㎍/mouse) in 100 ㎕ PBS
G4 : OVA (20 ㎍/mouse), HA-PG (1 MDa, 100 ㎍/mouse) in 100 ㎕ PBS
G5 : OVA (20 ㎍/mouse), HA-PG (200 kDa, 100 ㎍/mouse) in 100 ㎕ PBS
여기서 1 MDa (1,000,000) 및 200 KDa (200,000)는 히알루론산의 분자량(Da)을 나타낸 것이며, 양성대조군으로서 Alum(Sigma-Aldrich)을 사용하였다.
0일째에 본 발명의 시료를 마우스에 근육내 주사하여 1차 면역화하고, 14일째에 혈액 샘플을 수집하고 2차 면역화 하였다. 28일째에 마우스의 안와정맥총에서 전혈을 약 200 ㎕ 채취하고 Serum separating tube에 혈액을 넣어 4,000 rpm, 10분 원심분리 하여 혈청을 분리하였다. 분리된 혈청을 -70 ℃에서 보관하였다.
(2) SARS-CoV-2 S protein 항원을 이용한 면역반응 유도
6주령의 암컷, Balb/c 마우스(입수처: 일본 에스엘시 주식회사)를 G1 및 G2는 각각 4마리, G3 ~ G5는 각각 5마리씩 5그룹으로 구분하였다. 마우스에 면역을 유도하기 위해, 본 발명의 면역증강제 및 항원 (Recombinant SARS-COV-2 S1+S2 ECD(S-ECD) Protein with His tag; ABclonal)을 혼합한 혼합액을 각 마우스의 양쪽 대퇴부 근육에 50 ㎕씩 총 100 ㎕ 주입 하였다. 각 실험균 별 항원과 면역증강제의 주입량은 아래와 같았다.
G1 : PBS 100 ㎕
G2 : SARS-COV-2 Spike S1-S2 (10 ug/mouse) in 100 ㎕ of PBS
G3 : SARS-COV-2 Spike S1-S2 (10 ug/mouse) + ALUM (100 ug/mouse) in 100 ㎕ of PBS
G4 : SARS-COV-2 Spike S1-S2 (10 ug/mouse) + HA-PG (200 kDa, 100 ug/mouse) in 100 ㎕ of PBS
G5: SARS-COV-2 Spike S1-S2 (10 ug/mouse) + HA-PG (1 MDa, 100 ug/mouse) in 100 ㎕ of PBS
양성대조군으로서 알룸을 사용하였다. 0일째에 본 발명의 시료를 마우스에 근육내 주사하여 1차 면역화하고, 14일째에 안와정맥총에서 혈액 샘플을 수집하고 2차 면역화 하였다. 28일째에 마우스의 심장에서 혈액 약 200 ㎕ 채취하고 혈청분리큐브(Serum separating tube)에 혈액을 넣고 4,000 rpm, 10분 원심분리하여 혈청을 분리하였다. 분리된 혈청을 -70 ℃에서 보관하였다.
실시예 2. 혈청 내 항원 특이적 항체 분석을 위한 ELISA
(1) OVA 항원 특이적 항체분석
OVA 특이적 ELISA 수행을 위해 플레이트(immunoplate)의 각 웰에 1 ㎍/㎖ OVA를 포함하는 PBS 용액을 100 ㎕씩 넣고 4 ℃ 조건에서 16시간 동안 코팅하였다. 코팅된 플레이트의 각 웰을 0.05% Tween80을 포함하는 PBS (PBST) 200 ㎕ 이상을 넣어 3회 세척하였다. 각 웰의 블로킹(blocking)을 위해 1% BSA를 포함하는 PBS를 well 당 300 ㎕씩 넣고 37 ℃에서 2시간 인큐베이션 하였다. 이후 PBST를 이용하여 3회 세척하였다. 샘플의 제작 및 연속 희석(serial dilution)을 위해 혈청을 얼음 위에서 녹이고, 녹은 혈청을 0.1% BSA를 포함하는 PBS에 64배부터 시작하여 2배씩 11회 연속 희석하였다. 희석된 혈청을 블로킹 및 세척된 플레이트에 넣고 37 ℃에서 2시간 동안 인큐베이션 하였다. 이후 PBST로 3회 세척하고 검출 과정을 수행하였다. 검출을 위해 horseradish peroxidase (HRP) conjugated anti-mouse IgG, IgG1 또는 IgG2b Ab (SouthernBiotech)를 5000:1의 비율로 PBST에 희석하고 well 당 100 ㎕씩 넣은 후 37 ℃ 1시간 배양하였다. 배양 후 PBST를 이용하여 5회 세척 후 발색 과정을 수행하였다. 발색 과정을 위해 TMB (Tetramethylbenzidine; Surmodics) solution (Surmodics)을 세척된 well 당 100 ㎕씩 첨가하고 약 5 분 후 0.5 M 의 HCl 용액을 50 ㎕ 첨가하여 발색 과정을 완료하였다. 발색이 완료된 플레이트를 450 nm 파장의 흡광도를 분광광도계를 통해 측정하여 OD로 나타내며, 블랭크 웰 흡광도 측정값의 2배를 초과하는 웰의 희석 배수를 이용하여 titer를 결정하였다.
(2) SARS-COV-2 단백질 항원 특이적 항체분석
96웰 ELISA 플레이트(ThermoFisher Scientic)의 각 웰에 1 ㎍/㎖ SARS-CoV-2-S protein을 포함하는 PBS 용액을 100 ㎕씩 넣고 4 ℃ 조건에서 16시간 동안 코팅하였다. 다음 플레이트를 4 ℃에서 하룻밤 인큐베이션 하였다. 코팅된 플레이트(immunoplate)의 각 웰을 0.05% Tween 80을 포함하는 PBS 200 ㎕ 이상을 넣어 3회 세척하였다. 샘플의 제작 및 연속 희석을 위해 혈청을 얼음 위에서 녹이고, 녹은 혈청을 0.1% BSA를 포함하는 PBS에 64배부터 시작하여 2배씩 11회 연속 희석하였다. 플레이트를 150 ㎕/웰의 블로킹 완충액 (56 ℃에서 60분 동안 0.05% 열 불활성화를 함유하는 PBS;FBS(fetal bovine serum); Sigma-Aldrich)으로 실온에서 1시간 동안 블로킹하였다. 혈청 샘플을 블로킹 완충액을 사용하여 1:100으로 연속 희석하고 100 ㎕/웰로 플레이트에 가하였다. 플레이트를 실온에서 30분 동안 인큐베이션한 다음 세척 완충액으로 5회 세척하였다. 다음 특정 알칼리 포스파타제 결합 이차 항체인 염소 안티-인간 IgG(Southern Biotechnology)(블로킹 완충액에 1:1000)을 100 ㎕/웰로 첨가하고 실온에서 30분 동안 인큐베이션 하였다. 발색 과정을 위해 각 웰에 100 ㎕/well의 p-nitrophenyl phosphate (p-NPP)(Sigma-Aldrich)를 첨가하고, 30분간 암실에서 보관한 후, 정지 용액(1.2N 수산화나트륨, Reagecon, UK) 100㎕/웰을 첨가하여 반응을 종료시켰다. 450 nm에서의 광학 밀도(OD)를 측정하였다. 도 9에 그 결과를 나타냈다.
실시예 3: 생체 내 세포독성 T 림프구 반응 측정 시험
건강한 마우스의 비장 및 림프절을 적출하여 나일론 매쉬 위에서 분쇄하고 적혈구를 제거하여 세포 혼합액을 준비하였다. 세포 혼합액을 같은 세포 수를 가지는 2 그룹(A액, B액)으로 나누어 A액에는 1 ㎍/㎖의 농도를 가지는 SIINFEKL (OVA의 H2-Kb 결합 서열) 펩티드를 첨가하고 B액은 그대로 두었다. 37 ℃에서 1시간 배양 후 PBS로 세척하고 A액은 5 μM의 Cell Trace violet (CTV)를 이용해 핵을 염색하고, B액은 0.5μM의 CTV로 핵을 염색하였다. 이 후 A액, B액의 세포를 PBS로 세척 후 같은 비율로 혼합하여 타겟 세포를 제작하였다. 타겟 세포를 2 x 107 cell/마우스의 농도로 실험을 수행한 마우스의 각 그룹(G1, G2, G3, G4, G5)에 꼬리 정맥을 통해 주사하였다. 24시간 후 타겟 세포의 제거(A세포가 만드는 피크의 감소)를 FACS를 이용하여 spleen, inguinal lymph node에서 확인하였다.
그런 다음 플레이트를 150㎕/웰의 차단 완충액(56°C에서 60분 동안 0.05% 열 불활성화를 함유하는 PBS; FBS(fetal bovine serum); Sigma-Aldrich)으로 실온에서 1시간 동안 차단했습니다. 그런 다음, 혈청 샘플을 차단 완충액을 사용하여 1:100으로 연속 희석하고 100㎕/웰로 3중으로 첨가했습니다. 플레이트를 실온에서 30분 동안 인큐베이션한 다음 세척 완충액으로 5회 세척하였다.
결과
도 7 및 도 8은 상기 수집한 혈액 샘플로부터 OVA 특이적 면역글로불린 IgG, IgG1, IgG2b 및 IgM을 ELISA를 사용하여 측정한 것이다. 도 9는 SARS-CoV-2-S 단백질 특이적 IgG를 ELISA로 분석한 결과이다. 상기 도 7 및 8에서 확인되는 것과 같이, 본 발명의 면역증강제를 함유하는 G4 및 G5가 양성대조군인 G3와 비교하여 현저히 우수한 면역글로불린 생성을 보인 것을 확인하였다. 또한, 도 9에서, 이는 본 발명의 면역증강제를 함유하는 시료 G3 및 G5가 항원만 사용한 시험군 G2에 비해 현저히 우수하였으며, G5는 양성대조군과 대응한 수준의 항체 생성을 보인 것을 확인하였다. 이러한 결과는 본 발명의 면역증강제를 조성물이 면역반응 증대에 현저히 우수한 효과가 있음을 뒷받침 한다.
Claims (12)
- 제1항에 있어서, 상기 히알루론산 유도체의 중량평균 분자량 (Mw)은 1,000 Da 내지 20,000,000 Da인 것을 특징으로 하는, 면역증강용 조성물.
- 제1항 또는 제2항에 있어서, 상기 히알루론산 유도체는, 0.1% 내지 50%의 갈롤기 치환율을 갖는 것을 특징으로 하는, 면역증강용 조성물.
- 제4항에 있어서, 상기 히알루론산 유도체의 중량평균 분자량 (Mw)이 1,000 Da 내지 20,000,000 Da인 것인, 면역증강용 조성물.
- 제4항 또는 제5항에 있어서, 상기 히알루론산 유도체가, 0.1% 내지 50%의 갈롤기 치환율을 갖는 것인, 면역증강용 조성물.
- 제4항에 있어서, 상기 히알루론산 유도체는, 전체 면역증강제 조성물에 대하여 0.01%(w/v) 내지 30%(w/v)로 함유되는 것을 특징으로 하는, 면역증강용 조성물.
- 제1항 또는 제4항에 있어서, 상기 조성물은 생체 외에서 액체 상태이고, 생체 내에서 가교제 없이 겔화 상태를 형성하는 것을 특징으로 하는, 면역증강용 조성물.
- 제1항에 있어서, 상기 하이드로겔이 수 ㎚ ~ 수십 ㎛의 입자크기를 갖는 미세입자인 것인, 면역증강용 조성물.
- 제1항 또는 제4항의 면역증강용 조성물 및 항원을 포함하는 백신 조성물.
- 제1항 또는 제4항의 면역증강용 조성물, 및 항원, 단백질 또는 항체를 포함하는 증대된 면역반응을 유도하는 약물전달용 조성물.
