Abstract
Description
本明細書の記述は典型的な例示に過ぎず、本開示の請求の範囲又は適用例を如何なる意味に於いても限定するものではないことを理解する必要がある。
上記した以外の課題、構成及び効果は、以下の実施形態の説明により明らかにされる。
本開示は、主に、水の屈折率1.33と同等か、あるいは屈折率が1.36未満の低屈折率を有する分離媒体を用いることができるようにする技術を提案する。このような低屈折率の分離媒体を用いた場合、いずれの公知例(特許文献1及び2、および非特許文献1)に開示の技術を用いてもマルチフォーカスが機能せず、複数のキャピラリのレーザビームによる同時照射が困難である。
<キャピラリアレイ電気泳動装置の構成例>
図1は、キャピラリアレイ電気泳動装置の構成例を示す図である。本キャピラリアレイ電気泳動装置では、従来のキャピラリアレイ電気泳動装置で行われているDNAシーケンスや1本鎖DNAフラグメント解析に加えて、2本鎖DNAフラグメント解析を実施する。本実施形態では、24本のキャピラリを用い(ただし、図1では4本のキャピラリのみを示している)、まず、各キャピラリで異なるサンプルのDNAシーケンスを実施し、次に、各キャピラリで異なるサンプルの2本鎖DNAフラグメント解析を実施した。DNAシーケンスのサンプルには、4種類の塩基に対応した4種類の蛍光体で標識された、種々の長さの1本鎖DNA断片が含まれる。また、DNAシーケンスを行う際に各キャピラリに充填する電気泳動分離媒体は、変性剤として8 Mのウレアが含まれたポリマ溶液であり、その屈折率はn3=1.41である。一方、2本鎖DNAフラグメント解析のサンプルには、2種類の蛍光体で標識された、種々の長さの2本鎖DNA断片が含まれる。片方の蛍光体で標識された2本鎖DNA断片はPCR産物であり、もう片方の蛍光体で標識された2本鎖DNA断片はサイズマーカーである。2本鎖DNAフラグメント解析を行う際に各キャピラリに充填する電気泳動分離媒体は、変性剤であるウレアが含まれていないポリマ溶液であり、その屈折率はn3=1.33である。以下の(i)~(vi)の工程によって、1回の分析セッションを実行した。
図2は、キャピラリアレイ電気泳動装置の蛍光検出を行う光学系の構成例を示す断面図である。本光学系は、図1のレーザ照射部14の奥側に位置している。図1と同様に、図2では4本のキャピラリアレイの片側照射が描かれているが、実際には24本キャピラリアレイの両側照射である。レーザビーム13のマルチフォーカスによる照射により、配列平面上に配列する各キャピラリ1を同時照射した。各キャピラリ1のレーザ照射部14はそれぞれ蛍光の発光点20となる。各発光点20からの発光蛍光21を、一括して、集光レンズ15によってコリメートし、レーザカットフィルタ16でレーザ光を遮断し、透過型回折格子17を透過させることで各キャピラリの中心軸方向に波長分散させ、結像レンズ18でセンサ19上に結像点22をそれぞれ結像させた。センサ19は、CCDやCMOS等のエリアセンサ、あるいはフォトダイオードアレイ等、複数の結像点22を同時計測できるものであれば良い。各結像点22は実際には図2の奥行方向に波長分散されているが、図2では各結像点22の単一波長部分が模式的に描かれている。
図3は、センサと計算機の連携を示す構成例を示す図である。光学系はキャピラリアレイ電気泳動装置の一部であり、センサは光学系の一部である。計算機はキャピラリアレイ電気泳動装置と接続されている。計算機は、データ解析だけでなく、キャピラリアレイ電気泳動装置の制御も行う。入力部であるタッチパネル、キーボード、マウス等を通じて、データ解析の条件設定やキャピラリアレイ電気泳動装置制御の条件設定を行う。センサから出力される信号の時系列生データが順次メモリに格納される。また、HDDの内部にあるデータベースに格納されている解析パラメータ情報がメモリに格納される。CPUは、メモリに格納された解析パラメータ情報を用いて、メモリに格納された時系列生データを解析し、時系列解析データを導出し、順次メモリに格納すると同時に、表示部であるモニタに表示する。また、ネットワークインターフェースNIFを通じて解析結果をネットワーク上の情報と照合することができる。
図4(a)は、特許文献1に基づく3500シリーズジェネティックアナライザのキャピラリアレイの構成を示す断面図である。外径2R=323 μm、内径2r=50 μmの24本のキャピラリのレーザ照射部が間隔370 μmで同一平面上に配列している。配列誤差はゼロである(ΔZ=0 μm)。キャピラリ外部は空気でありn1=1.00、キャピラリ素材は石英ガラスでありn2=1.46である。
図5(a)は、図4(b)および(c)に示される片側照射の結果を両側照射の場合に焼き直した場合についての、各キャピラリの相対蛍光強度を示す図である。キャピラリ番号は、図4(a)~(c)の一番左側のキャピラリを1とし、右側に向かって順番に付けた番号である。相対蛍光強度は、各キャピラリのレーザ照射部に一定濃度の蛍光体が存在すると仮定し、レーザビーム反射ロスを加味した各キャピラリの照射強度から計算される蛍光強度である。レーザ光源から発振したレーザビームの全強度が1本のキャピラリの内部に照射された場合に期待される蛍光強度を1としている。両側照射の計算では、レーザビームの全強度の半分がキャピラリアレイの両側から照射されるとした。n3=1.41の場合、24本のキャピラリについて相対蛍光強度の最小値がMIN=0.42、変動係数(=相対蛍光強度の標準偏差/相対蛍光強度の平均値)がCV=11%という結果が得られ、実用性能であるMIN≧0.2およびCV≦20%を満たすことが分かる。キャピラリ番号に対する相対蛍光強度が下に凸の分布になっているのは、マルチフォーカスが機能しているにも関わらず、レーザビームがキャピラリアレイ内を進行するのに伴って、反射ロスによってレーザビームの強度が減衰するためである。これに対して、n3=1.33の場合、MIN=0.068、およびCV=74%が得られ、実用性能を満たさないことが分かる。
図6(a)は、特許文献2に基づく3730シリーズジェネティックアナライザのキャピラリアレイの構成を示す断面図である。外径2R=126 μm、内径2r=50 μmの96本のキャピラリのレーザ照射部が間隔155 μmで同一平面上に配列している。配列誤差はゼロである(ΔZ=0 μm)。キャピラリ外部はフッ素溶液でありn1=1.29、キャピラリ素材は石英ガラスでありn2=1.46である。
図7(a)は、図6(b)および(c)に示される片側照射の結果を両側照射の場合に焼き直した場合についての、各キャピラリの相対蛍光強度を示す図である。n3=1.41の場合、96本のキャピラリについて相対蛍光強度の最小値がMIN=0.63、変動係数がCV=3.2%という結果が得られ、実用性能であるMIN≧0.2およびCV≦20%を満たすことが分かる。これに対して、n3=1.33の場合、MIN=0.00067、およびCV=192%となり、実用性能を満たさないことが分かる。
図8(a)は、第1の実施形態に基づくキャピラリアレイの構成例を示す断面図である。外径2R=126 μm、内径2r=50 μmの24本のキャピラリのレーザ照射部が間隔155 μmで同一平面上に配列している。配列誤差はゼロである(ΔZ=0 μm)。キャピラリ外部は空気でありn1=1.00、キャピラリ素材は石英ガラスでありn2=1.46である。
図9(a)は、図8(b)および(c)に示される片側照射の結果を両側照射の場合に焼き直した場合についての、各キャピラリの相対蛍光強度を示す図である。n3=1.41の場合、24本のキャピラリについて相対蛍光強度の最小値がMIN=0.42、変動係数がCV=11%という結果が得られ、実用性能であるMIN≧0.2およびCV≦20%を満たすことが分かる。一方、n3=1.33の場合も、MIN=0.40、およびCV=12%となり、実用性能を満たすことが分かる。
以上より、第1の実施形態の構成は、n3=1.41を含めて、n3≧1.33の任意の屈折率を有する分離媒体を用いた場合に、各キャピラリが凸レンズ作用を示し、マルチフォーカスが機能することが明らかになった。また、本構成の変形例として、R/r≦4.4の任意のキャピラリ、例えば、内径2r=50 μmを固定した場合、外径2R≦220 μmの任意のキャピラリを用いた場合にも、n3≧1.33の条件下で各キャピラリが凸レンズ作用を示すため、マルチフォーカスを機能させることが可能である。
第1の実施形態では、キャピラリアレイの配列誤差がゼロ(ΔZ=0 μm)の場合についての検討結果を示した。しかし、現実にΔZ=0 μmとなることはない。そこで、第2の実施形態では、配列誤差とマルチフォーカス性能および各キャピラリの相対蛍光強度との関係を系統的に検討する。さらに、配列誤差とキャピラリアレイの製造コストとの関係も検討する。これらの検討は、本開示の技術によって初めてなされるものである。
図10は、配列誤差ΔZの定義を説明するための図である。図10(a)は、配列誤差がゼロの場合(ΔZ=0 μm)の24本のキャピラリアレイを示す断面図である。この構成は図8(a)と同じである。配列平面に沿ってX軸を設定し、配列平面と垂直方向にZ軸を設定する。また、各キャピラリの中心軸と平行方向にY軸を設定する。各キャピラリの中心軸はX軸上にあり、Z座標はゼロである。
図11は、キャピラリアレイの配列誤差ΔZと製造コストとの関係を検討した結果を示す図である。キャピラリアレイの構造は、図8(a)と同じであり、外径2R=126 μm、内径2r=50 μmの24本のキャピラリのレーザ照射部を間隔155 μmで同一平面上に配列させた。キャピラリ外部は空気でありn1=1.00、キャピラリ素材は石英ガラスでありn2=1.46とする。以上の条件下で、配列誤差を可能な限りゼロに近づけるようにして、多数のキャピラリアレイを製造した。製造された個々のキャピラリアレイの配列誤差ΔZを、レーザ顕微鏡を用いて測定した。
図12および図13は、図8(a)に示す24本のキャピラリアレイでn3=1.41とする構成を基準として、ΔZを変更した場合の両側照射によって得られる各キャピラリの相対蛍光強度を示す図である。図12(a)はΔZ=0 μm、図12(b)はΔZ=3 μm、図12(c)はΔZ=6 μm、図13(a)はΔZ=9 μm、図13(b)はΔZ=12 μmとした場合を示している。ΔZ=0 μmについては1組のキャピラリアレイ、ΔZ=0 μm以外のΔZについてはそれぞれ10組のランダムな配列のキャピラリアレイについて相対蛍光強度を求めた結果を重ねて示している。図12(a)(ΔZ=0 μm)は、図9(a)のn3=1.41の場合と同じ結果を示している。図13(c)は、ΔZ=0 μmについては1組の各キャピラリの相対蛍光強度を示し、ΔZ=0 μm以外のΔZについては上記10組の各キャピラリの相対蛍光強度の平均を重ねて示している。ΔZが増大するに従って、相対蛍光強度の平均値および最小値が低下するとともに、相対蛍光強度のばらつきが大きくなることが分かる。相対蛍光強度の最小値はそれぞれ、ΔZ=0 μmのときMIN=0.42、ΔZ=3 μmのときMIN=0.40、ΔZ=6 μmのときMIN=0.33、ΔZ=9 μmのときMIN=0.22、およびΔZ=12 μmのときMIN=0.066であった。また、相対蛍光強度の変動係数はそれぞれ、ΔZ=0 μmのときCV=11%、ΔZ=3 μmのときCV=11%、ΔZ=6 μmのときCV=12%、ΔZ=9 μmのときCV=17%、およびΔZ=12 μmのときCV=28%であった。ここで、ΔZ=0 μmについては1組の各キャピラリの相対蛍光強度の最小値および変動係数を示し、ΔZ=0 μm以外のΔZについては上記10組の各キャピラリの相対蛍光強度の最小値および変動係数を示した。
以上より、実用性能であるMIN≧0.2を満たすためには、ΔZ≦9 μmであれば良いことが分かった。また、実用性能であるCV≦20%またはCV≦15%を満たすためには、ΔZ≦9 μmまたはΔZ≦6 μmであれば良いことが分かった。これらの結果は、上記の図12および図13で示したn3=1.41の場合の結果と同じである。
第3の実施形態では、第2の実施形態で検討した配列誤差とマルチフォーカス性能および各キャピラリの相対蛍光強度との関係をさらに詳細に検討する。
図16(a)は、図8(a)に示す24本のキャピラリアレイでn3=1.41とする構成を基準として、ΔZ=0.0、1.5、3.0、4.5、6.0、7.5、9.0、10.5、12.0、13.5、および15.0 μmと変化させた場合の、両側照射によって得られる各キャピラリの相対蛍光強度を示す図である。図16(c)は、同条件下における、相対蛍光強度の変動係数を示す図である。図16(b)および(d)は、それぞれ図16(a)および(c)の拡大図である。ΔZ=0.0 μmについては1組のキャピラリアレイを用い、ΔZ≠0.0 μmの各ΔZについては100組のランダムな配列で構成されるキャピラリアレイを用いている。図16(a)および(b)では、ΔZ=0.0 μmについては24個の相対蛍光強度データを用い、ΔZ≠0.0 μmの各ΔZについては2400個の相対蛍光強度データを用いた。黒丸プロットは平均値、エラーバーは±標準偏差、黒三角プロットは最大値、黒四角プロットは最小値を示す。ΔZの増加に伴い、平均値、最大値、最小値のいずれもが低下し、標準偏差が増大している。図16(c)および(d)では、ΔZ=0.0 μmについては1組の相対蛍光強度の変動係数を用い、ΔZ≠0.0 μmの各ΔZについては100組の相対蛍光強度の変動係数を用いた。黒丸プロットは平均値、エラーバーは±標準偏差、黒三角プロットは最大値、黒四角プロットは最小値を示す。ΔZの増加に伴い、平均値、最大値、最小値のいずれもが増加し、標準偏差が増大している。
図18は、図17の条件において、キャピラリ外径2Rを75 μm≦2R≦250 μmの範囲内で変化させた際に、図17と同様に実用性能であるCV≦20%あるいはCV≦15%を満足する最大の配列誤差ΔZを求めた結果を示す図である。各2Rについてのキャピラリアレイの配列間隔は2R+29 μmで統一している。上述の通り、式(1)によって2R>220 μmのときは各キャピラリが凹レンズ作用を示すため、マルチフォーカスが機能せず、CV≦20%あるいはCV≦15%は満足されない。このとき、図18の縦軸の配列誤差ΔZをゼロで示す。図18の結果より、キャピラリ外径2Rを100 μm≦2R≦200 μmの範囲内で適宜選択し、あるいはキャピラリの外径と内径の比率R/rを2≦R/r≦4の範囲内で適宜選択するとき、実用性能であるCV≦20%を満たすためには、ΔZ≦9 μmであれば良い。また、キャピラリ外径2Rを100 μm≦2R≦175 μmの範囲内で適宜選択し、あるいはキャピラリの外径と内径の比率R/rを2≦R/r≦3.5の範囲内で適宜選択するとき、実用性能であるCV≦15%を満たすためには、ΔZ≦6 μmであれば良い。製造コスト基準比は、ΔZ=9 μmのとき1.04であり、ΔZ=6 μmのとき2.10である。これらの製造コストは許容範囲内であり、以上の条件を実施可能である。

前記複数のキャピラリからの発光を一括計測する光学系と、を備え、
前記レーザ照射部における前記複数のキャピラリの、外半径をR、内半径をr、外部の媒体の屈折率をn1、素材の屈折率をn2、および内部の媒体の屈折率をn3とするとき、
n1=1.00、n2=1.46±0.01、n3<1.36、およびR/r<5.9
