본 발명은 Cdo (Cell adhesion molecule-related/down-regulated by oncogenes) 단백질 또는 이를 암호화하는 유전자를 이용하여, 운동 신경, 특히 근육 줄기세포 및 신경근 접합부에서 노화, 산화 스트레스 등에 의한 운동 신경 손상에 의해 유발되는 신경 근육 질환의 예방 또는 치료 용도 및 상기 Cdo의 발현을 활성화하는 후보 물질을 스크리닝 하기 위한 방법에 대한 것이다. 운동 신경 또는 신경근 접합부에서 Cdo가 결손된 경우, 노화에 의한 퇴행성 운동 신경의 손상 악화, 산화 스트레스 및 염증에 의한 DNA 손상이 유발되어 신경 근육 질환이 유발될 수 있으므로, 상기 Cdo 단백질 및 이를 암호화하는 유전자의 과발현 및 활성을 통해 상기 질환의 치료를 가능하게 할 수 있다.
Description
본 발명은 운동 신경 및 신경 근육 접합부에서 발현하는 Cdo 단백질 및 이를 암호화하는 유전자의 신경 근육 질환의 예방 또는 치료 용도에 대한 것이다.
골격근은 주로 위성세포(satellite cells, SCs) 라고 하는 근육 줄기 세포에 의해 재생된다. 위성 세포는 평소에는 정지 상태에 있다가 부상 등 자극에 의해 활성화되어 자가 재생 및 분화를 진행한다. 위성 세포 분화 후, 정지 상태로 다시 돌아가는데, 이는 줄기세포의 집단 유지에 중요한 것이다.
노화 또는 영양 장애와 관련된 다양한 근육의 소모 상태는 위와 같은 위성 세포의 수와 증식 능력의 감소와 관련이 있다. 위성 세포는 노화에 따라 증식하고 분화하는 능력이 점차 감소한다. 위성 세포의 노화 및 기능 장애는 DNA 손상을 축적시키며, 세포 주기를 정지시켜 세포 노화를 유발할 수 있다. 노화된 세포는 편평한 외관, 노화와 관련된 β-갈락토시다아제(SA-β-gal) 활성, 사이클린의존성 키나아제 억제제 p16Ink4a (p16) 및 p21Waf1 (p21)의 상향 조절과 같은 특징을 가진다. 또한 노화 세포는 노화 관련 분비 표현형(SASP)을 생성하여 염증성 사이토카인과 인슐린 유사 성장 인자 결합 단백질을 생성한다. 세포 주기 억제제의 조절 장애가 세포 노화의 기본 메커니즘인 것으로 종래에는 알려져 있으나, 위성 세포 노화에 대한 분자 메커니즘은 아직 많은 연구가 이루어지지 않고 있는 실정이다.
세포 표면 단백질인 Cdo (Cdon이라고도 함) (Homo sapiens Gene ID: 50937; Mus musculus Gene ID: 57810)은 근육 분화에 관여하는 면역 글로불린/피브로넥틴 III 형 반복 패밀리의 세포 표면 수용체이다. Cdo는 골격근아세포 사이의 접촉 부위에서 N-cadherin과 복합체를 형성하며, 이러한 상호 작용은 Cdo의 promyogenic 기능에 중요한 영향을 주는 것으로 알려져 있다. 그러나 Cdo가 발현되는 다양한 운동 신경 또는 신경 근육 접합부에서의 기작에 대해서는 알려진 바가 없다.
신경 근육 질환이란, 말초신경과 근육에 생기는 질병으로, 말초신경계 질환은 말초신경 자체의 질병, 말초신경과 인접해 있는 주위 조직의 질병에 의해 말초신경이 손상되어 생기는 말초신경의 기능장애, 말초신경이 근육과 연결되는 신경근 접합부, 근육 자체에 생긴 질환을 포함한다.
또한 상기와 같은 말초 신경계의 이상으로 인하여 유발되는 운동 신경 질환은 근력은 정상이나, 이의 활동을 통제하는 운동 신경이 퇴행적 진행을 일으켜 손상된 신경학적 질환을 의미하는 것으로, 유전적 요인, 환경적 요인, 독극물 중독, 바이러스 감염, 다른 질환의 합병증, 노화 등 다양한 원인에 의해 발생된다. 인구의 고령화에 따라 특히 노화에 의한 퇴행성 운동 신경 질환의 유병률이 높아지고 있으며, 이는 인간의 삶의 질에 큰 영향을 줄 수 있으므로, 근본적인 원인과 치료 방법에 대한 연구가 요구되고 있는 실정이다.
이에, 본 발명자들은 운동 신경 세포 손상과 연관성이 있는 다양한 단백질 및 유전자를 연구하던 중, 근육 줄기세포 (Muscle stem cell, satellite cell)에서 발현하는 Cdo(Cell adhesion molecule-related/down-regulated by oncogenes) 단백질에 대한 연구를 진행하여, 상기 단백질 및 이를 암호화하는 유전자의 발현 조절을 통해 다양한 원인에 의해 유발된 세포 스트레스로 인한 운동 신경 세포 손상을 방지하고, 이로 인해 유발되는 질병에 대한 치료 및 완화 효과를 달성할 수 있음을 발견하고 본 발명을 완성하였다.
따라서, 본 발명의 목적은 Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는, 운동 신경 세포 보호용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는, 신경 근육 질환의 예방, 개선 또는 치료용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은, Cdo의 발현 여부와 관련하여 신경 근육 질환의 예방, 개선 또는 치료 물질을 스크리닝하는 방법을 제공하는 것이다.
상기와 같은 목적을 달성하기 위하여, 본 발명은 Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는 운동 신경 세포 보호용 조성물을 제공한다.
본 발명은 또한, Cdo 단백질 또는 이를 암호화하는 유전자를 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 운동 신경 세포 보호 방법을 제공한다.
본 발명은 또한, Cdo 단백질 또는 이를 암호화하는 유전자의 운동 신경 세포 보호 용도를 제공한다.

본 발명의 또 다른 목적을 달성하기 위하여, 본 발명은 i) in vitro 상에서 운동 신경 세포 또는 위성세포에 후보 물질을 처리하는 단계; ii) 상기 후보 물질을 처리한 운동 신경 세포 또는 위성세포에서 Cdo의 발현 또는 활성을 측정하는 단계; 및 iii) 미처리군과 비교하여 Cdo의 발현 또는 활성을 증진시키는 후보 물질들을 신경 근육 질환 치료제 후보 물질로 선정하는 단계를 포함하는, 신경 근육 질환의 치료 물질을 스크리닝 하는 방법을 제공한다

상기 본 발명의 운동 신경 세포 보호는, 특히 노화로 인한 운동 신경의 퇴행성 손상으로부터 이를 보호하는 것을 포함하며, 이에 제한되는 것은 아니나, 생존율 향상, 체중 유지, 행동능력 유지, 손상된 신경 복구, DNA 손상 복구, 세포 스트레스로부터 운동 신경 세포를 보호하는 것으로, 노화 등 다양한 요인에 의하여 운동 신경 세포에서 발생될 수 있는 스트레스를 완화하고 경감시키는 것을 의미한다.
본 발명의 일 양태로서, 상기 운동 신경 세포의 보호는 노화, 산화 스트레스, 염증성 스트레스로부터 운동 신경 세포의 손상, 이로 인한 근육 위축 및 섬유화를 방지하고, 운동 신경세포의 스트레스 민감성을 감소시키는 것이며, 손상된 운동 신경세포의 재 신경화를 촉진하는 것을 포함한다.
또한 본 발명은 Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는, 신경 근육 질환의 예방, 개선 또는 치료용 조성물에 대한 것이다.
본 발명은 또한, Cdo 단백질 또는 이를 암호화하는 유전자를 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 신경 근육 질환의 예방, 개선 또는 치료방법에 관한 것이다.

신경 근육 질환은 말초 신경계의 이상으로 인하여 유발되며, 근육 운동을 통제하는 운동 신경이 퇴행적 진행을 일으켜 손상된 신경학적 질환으로, 근이영양증, 운동 신경 질환, 근병증, 신경근 접합부 질환 등이 포함된다. 또한, 이에 제한되는 것은 아니나, 상기 신경 근육 질환은 신경근접합부 및 근육의 질환으로 분류(질병분류기호 상 G70-G73)되는 질환으로서, 중증근무력증, 근신경장애, 근디스트로피, 근긴장장애, 근병증, 근육의 원발성 장애, 근무력증후군, 람베르트-이튼 증후군 등이 포함된다. 또한, 상기 운동 신경 질환은 근위축성 측삭경화증(amyotrophic lateral sclerosis, ALS), 척수성 진행성 근위축(infantile progressive spinal muscular atrophy, SMA), 진행성 핵상마비(progressive bulbar palsy), 가연수성마비(pseudobulbar palsy), 진행성 근육위축(progressive muscular atrophy, PMA), 진행성 축삭경화증(progressive lateral sclerosis, PLS), 단일 사지 근위축증(monomelic amyotrophy, MMA) 또는 헌팅턴 병 등이 포함되며, 기타 운동 신경원 질환을 포함한다.
본 발명자들은 운동 신경, 근육 줄기세포인 위성 세포 또는 신경근 접합부에서 Cdo의 발현이 노화 등의 스트레스 자극에 의해 감소되는 것을 확인하였으며, 특히 운동 신경 특이적으로 Cdo가 감소된 동물모델은 생존률과 운동능력이 크게 감소되는 것을 확인하였다. 이를 통해 운동 신경에서의 Cdo 발현 여부가 운동 신경과 관련된 다양한 질환에 영향을 미칠 수 있다는 것을 알 수 있다. 또한 위성 세포 특이적으로 Cdo가 감소된 경우에는 근신경 접합부에서의 핵의 숫자가 감소된 것을 확인할 수 있었다. 이와 같은 결과는 Cdo가 근섬유의 유형을 결정하는 역할을 하는 것을 보여주는 것으로, 특히 단일근섬유 분화에도 영향을 줄 수 있다는 점을 확인하였다.
본 발명의 일 양태로서, 본 발명의 조성물은 의약품, 건강기능식품, 기능성 식품, 동물용 사료, 세포 배양액 조성물 등 다양한 용도로 활용될 수 있으며, 운동 신경 세포의 손상을 방지하여 상기 신경 근육 질환에 대한 예방, 개선 또는 치료 효과를 가진다.
본 명세서에서 용어 "예방"은 본 발명에 따른 상기 약학적 조성물의 투여에 의해 신경 근육 질환을 억제시키거나 발병을 지연시킬 수 있는 모든 행위를 의미한다.
본 명세서에서 용어 "치료"는 본 발명에 따른 상기 약학적 조성물의 투여에 의해 증세가 호전되거나 이롭게 되는 모든 행위를 말한다.
본 명세서에서 용어 “개체”는 본 발명에 따른 운동 신경 세포 보호가 필요하거나, 신경 근육 질환이 발병하였거나 발병할 가능성이 있는 인간을 포함한 모든 동물을 의미할 수 있다. 상기 동물은 인간뿐만 아니라 이와 유사한 증상의 치료를 필요로 하는 소, 말, 양, 돼지, 염소, 낙타, 영양, 개, 고양이 등의 포유동물일 수 있다.
본 발명의 상기 약학적 조성물은 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있으며, 상기 제형화에 필요한 담체 혹은 부형제 등을 추가로 포함할 수 있다. 상기 유효 성분에 추가로 포함될 수 있는 약학적으로 허용 가능한 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 마그네슘 스테아레이트 및 광물유 등이 포함된다. 제제화 할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
예를 들어, 경구 투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 추출물 또는 화합물에 적어도 하나 이상의 부형제 적어도 면, 전분, 칼슘카보네이트 (calcium carbonate), 수크로스 (sucrose) 또는 락토오스 (lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 경구 투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜 (propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔 (witepsol), 마크로골, 트윈 (tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 또는 비경구 투여(정맥주사, 피하, 복강 내 또는 국소에 적용)될 수 있으며, 투여량은 환자의 상태 및 체중, 질병의 정도 및 약물 형태, 투여 경로 및 시간에 따라 다르며, 당업자에 의해 적절한 형태로 선택될 수 있다.
본 발명의 상기 약학적 조성물은, 약학적으로 유효한 양으로 투여한다. 본 발명에서 "약학적으로 유효한 양"이란 의학적 치료에 적용 가능한 합리적인 양으로, 질병을 치료하기에 충분한 양을 의미하며, 그 기준은 환자의 질환, 중증도, 약물 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 병용되는 성분 및 기타 사항에 따라 결정될 수 있다. 본 발명의 상기 약학적 조성물은, 개별 치료제 혹은 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 상기 요소들을 모두 고려하여 부작용을 최소화할 수 있는 수준으로 투여량을 결정할 수 있고, 이는 당업자에 의해 용이한 수준으로 결정될 수 있다. 구체적으로 상기 약학적 조성물의 투여량은 환자의 연령, 체중, 중증도, 성별 등에 따라 달라질 수 있으며, 일반적으로 체중 1kg 당 0.001 내지 150 mg, 보다 바람직하게는 0.01 내지 100mg의 양을 매일 또는 격일, 1일 1 내지 3회 투여할 수 있다. 다만, 이는 예시적인 것으로, 상기 투여량은 필요에 달리 설정할 수 있다.
또한 본 발명의 상기 조성물은 식품 또는 건강기능식품일 수 있으며, 특히 상기 "건강기능식품"은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다
본 발명의 상기 식품 또는 건강기능식품은, 운동 신경 질환의 예방 및 개선을 위한 목적으로 산제, 과립제, 정제, 캡슐제, 환제, 현탁액, 에멀젼, 시럽 등 약학적 투여 형태 또는 티백, 침출자, 음료, 캔디, 젤리, 껌 등의 건강 기능 식품으로 제조 및 가공이 가능하다.
본 발명의 상기 식품 또는 건강기능식품 조성물은, 식품 첨가물로 사용될 수 있으며, 단독으로 혹은 다른 성분과 조합하여 제품화될 수 있다. 또한, 영양제, 비타민, 전해질, 풍미제, 착색제 및 증진제, 펙트산 및 이의 염, 알긴산 및 이의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등이 포함될 수 있다. 상기 성분은 단독 혹은 조합하여 사용될 수 있으며, 적절한 함량으로 조합되어 사용할 수 있다.
또한 본 발명은, i) in vitro 상에서 운동 신경 세포 또는 위성 세 포에 후보 물질을 처리하는 단계; ii) 상기 후보 물질을 처리한 운동 신경 세포 또는 위성 세포에서 Cdo의 발현 또는 활성을 측정하는 단계; 및 iii) 미처리군과 비교하여 Cdo의 발현 또는 활성을 증진시키는 후보 물질들을 운동 신경 질환 치료제 후 보 물질로 선정하는 단계를 포함하는, 운동 신경 질환의 치료 물질을 스크리닝하는 방법을 제공한다.
본 발명에서 상기 Cdo의 발현 또는 활성을 측정하는 단계는 Cdo 유전자의 발현에 의해 생성된 RNA 또는 단백질의 양을 측정할 수 있는 다양한 방법을 통해 실시될 수 있다. 바람직하게, RNA의 발현량은 RT-PCR(실시간 PCR), 마이크로어레이, 노던 블롯팅 등을 이용하여 확인하였으며, 단백질의 활성 및 발현은 면역 침강, 면역염색, 크로마틴 면역염색, 효소면역분석(ELISA), 웨스턴 블롯팅 등의 방법으로 확인할 수 있으며, 그 방법에 제한되는 것은 아니다.
본 발명의 조성물은 Cdo 단백질 또는 이를 암호화하는 유전자를 포함하며, 상기 Cdo 단백질은 운동 신경 세포의 손상 방지와 관련된 중요한 역할을 담당하고 있는 인자로서, 다양한 요인에 의한 스트레스로부터 운동 신경의 손상 방지, 상기 운동 신경 손상 및 퇴행성 요인에 의한 운동 신경 질환 등에 대한 예방, 개선 및 치료 효과를 가진다.

실시예 1. 재료 및 실험 방법
1-1) 실험 모델 제작
실험동물 사육실은 일정한 조건 (온도 22±2℃, 습도 55±5%)을 유지하였다. 1일 12시간의 광주기와 12시간의 암주기를 적응하였다.
운동신경 특이적 Cdo 결핍 마우스는 Cdo-floxed 대립 유전자 (EUCOMM에서 입수한 Cdo f/f)를 보유한 마우스와 Hb9 프로모터 (Jackson Laboratory에서 제공하는 Hb9-Cre)의 제어 하에 Cre 재조합 효소를 발현하는 형질전환 마우스와 교배시켜 제작하였다. 근육 줄기세포 특이 Cdo 결핍 마우스는 Cdo-floxed 대립 유전자 (EUCOMM에서 입수한 Cdo f/f)를 보유한 마우스와 Pax7 프로모터 (Jackson Laboratory에서 제공하는 Pax7ERT2Cre)에 Cre 재조합 효소를 발현하는 형질 전환 마우스를 교배시켜 Cdofl/fl;Pax7ERT2Cre+/- 마우스를 제작하였다. 특히, Cdofl/fl;Pax7ERT2Cre+/- 마우스는 이틀 간격으로 5회 Tamoxifen (100mg/kg body weight)을 복강 내 주입하여 근육 줄기세포 특이적 Cdo의 결핍을 유도하였다.
노화 마우스 (Geriatric model)는 형질 전환시키지 않은 야생형 마우스를 27달간 사육시설에서 유지했다. 대조군으로는 야생형 마우스 중 5달 자란 마우스를 사용했다.
1-2) 행동분석
Cdo f/f와 Cdo hb9cre 마우스의 행동 차이를 분석하기 위해 Open field, Gait-test, Grip-test를 수행했다.
Open field 테스트는 개방형 플라스틱 용기 (30x30x30cm)에서 수행되었다. 각 실험이 시작될 때 마우스를 플라스틱 용기의 모서리에 놓고 20 분 동안 움직임을 기록했다. Noldus EthoVision 13.0 추적 소프트웨어는 이동 거리와 속도를 분석하는데 사용되었다.
Gait 테스트는 벽이 투명한 treadmill 장치(Columbus Exer-6 M treadmill)의 바닥에 A4 용지를 놓고 마우스를 한 방향으로 똑바로 걷게 하는 방식으로 수행되었다. 마우스의 뒷발에 보라색 잉크를 충분히 바른 후 A4 용지에 기록된 마우스의 발자국으로 stride, sway, stance를 측정했다.
Grip 테스트는 테스트 장비(Bioseb)를 사용해 측정했다. 마우스의 앞발이 장비의 grid를 완전히 잡으면 일정한 힘으로 마우스를 앞발이 grid에서 떨어질 때까지 잡아당기고 이때 측정되는 힘을 기록했다.
각 마우스의 test는 3번씩 반복했으며 blind test로 진행되었다.
1-3) 세포 배양 및 형질감염
C2C12 세포는 DMEM media를 기반으로 15%의 FBS, 10 units/mL 페니실린, 10μg/mL 스트렙토마이신의 조성에서 배양한다.
C2C12 cell에 Cdo 발현을 결손 시키기 위하여 Lentiviral pLKO. 1-puro, Cdo-shRNA를 cell에 infection시킨다.
1-4) 단백질 및 RNA 분석
세포 및 조직 수확 후 세포에 Lysis buffer(20mM Tris-HCl, pH8, 150mM NaCl, 1% Triton X, Proteinase Inhibitor)를 첨가하여 30분간 용해한 뒤, 13000rmp에서 30분간 원심 분리 후, 세포 용해 샘플을 정량 하여, 동일양의 단백질을 SDS-PAGE 전기 영동을 하고 PVDF membrane으로 transfer했다. 해당하는 일차항체, 이차항체 반응 후 ECL reagent를 이용하여, X-ray 필름에 노출시켜 단백질 발현량을 확인하였다.
C2C12 세포 및 근육조직은 FastPrepR-24 (MP Biomedicals)로 균질화하였고 easy-spin Total RNA Extraction Kit (iNtRON Biotechnology)를 사용하여 RNA 추출을 하였다. 유전자 발현의 배수 변화는 리보솜 유전자 L32의 발현에 대비해 정규화되었다.
1-5) 조직학 및 면역 형광 분석
실험에 사용된 조직은 전경골근 및 장지신근(Tibialis anterior, extensor digitorum longus muscle) 그리고 요추 (Lumbar spine)를 사용하였으며, 전경골근과 요추는 분리 즉시 액화질소를 이용하여 OCT compound로 동결고정을 한 후, -80℃의 초저온 냉동고에 보관하였다. 조직절편은 영하 20℃로 유지되는 동결절편기을 이용하여 각각 10 um, 7 um의 두께로 절단하였다.
상기 동물 조직에서 분리한 전경골근 및 요추를 사용하여 면역 조직 화학 염색을 실시하였다. -80℃의 초저온 냉동고에 보관된 동결절편을 1xPBS에 2번 세척하였다. 4% 파라포름알데하이드(paraformaldehyde)을 사용하여 15분간 상온에서 고정을 시키고, 1xPBS에 2번 세척하였다. 조직과 투과용 버퍼 (0.5% Triton-X/PBS)를 5분간 상온에서 반응시키고 1xPBS에 2번 세척하였다. 조직과 TE buffer를 사용하여 100℃에서 10분간 반응시켜 항원성부활 (Antigen retrieval)을 하고, 1xPBS에 2번 세척하였다. Blocking buffer (5% Goat serum, 0.1% Triton-X)로 1시간 동안 blocking을 한 후에 일차항체 (Neurofilament, Synaptophysin)을 Blocking buffer에 1:300으로 희석하여 4℃에서 24시간 동안 반응시킨 후, 1xPBS에 3번 세척하였다. 2차항체와 2차 항체 conjugate된 BTX antibody를 실온에서 2시간 반응시킨 후, 1xPBS에 2번 세척하였다. 후에 봉입하여 공초점 현광 현미경 (LSM710)으로 분석하였다.
상기 동물 조직에서 분리한 장지신근을 사용하여 신경근 접합부 염 색(NMJ staining)을 실시하였다. 장지신근 근육을 분리 즉시 4% 파라포름알데하이드 (paraformaldehyde)을 사용하여 15분간 상온에서 고정을 시키고, 1xPBS에 3번 세척하였다. Blocking buffer (3% BSA, 0.5% Triton-X)로 2시간 동안 blocking을 한 후에 일차항체 (Neurofilament, Synaptophysin)을 Blocking buffer에 1:300으로 희석하여 4℃에서 24시간 동안 반응시킨 후, 1xPBS에 3번 세척하였다. 2차항체와 2차 항체 conjugate된 BTX antibody를 실온에서 2시간 반응시킨 후, 1xPBS에 2번 세척하였다. 후에 봉입하여 공초점 형광 현미경(LSM710)으로 분석하였다.
단일 근섬유 분리를 통한 면역조직화학적 염색에는 마우스 신근(Extensor digitorum longus, EDL)을 사용하였다. 각 단일 근섬유는 근손상이 유도되거나 되지 않은 마우스에서 분리되었으며, 근육 손상은 PBS에 희석한 10μM Cardiotoxin을 근육 내 주입하여 유도하였다. 마우스에서 EDL 근육 분리 후, 200U/ml의 CollagenaseⅡ를 37℃에서 90분간 처리하여 단일 근섬유 분리를 실시하였고, 이어서 BTX와 Cdo 항체 (1:300)를 사용하여 각 단일 근섬유의 조직면역화학을 수행하였다. 후에, 공초점 형광현미경 (LSM710)으로 신경근 접합부에서의 Cdo의 발현을 관찰하였다.
1-6) 통계분석
모든 값은 평균±SEM 또는 SD로 표시되었다. 통계적 유의성은 쌍 또는 짝이 없는 양측 스튜던트 t 검정 또는 분산 분석(ANOVA) 검정에 이어 Tukey의 검정에 의해 계산되었다. 차이는 P<0.05에서 유의한 것으로 간주되었다.
실험예 1. 운동신경 특이적 Cdo 결손에 의한 영향
1-1) Cdo 결손 마우스의 생존율 감소 효과
운동 신경 (motor neuron)특이적으로 Cdo이 결핍된 마우스(Cdo
Hb9cre)의 생존률 및 체중을 각 월령별로 측정하였다. 그 결과, 야생형(Cdo
f/f)에 비하여 노화가 진행될수록 마우스의 생존률이 급격히 감소됨을 확인하였다 (도 1). 즉, 운동 신경에서 발현되는 Cdo 은 특히 노화 마우스의 생존율과 체중 유지에 중요한 역할을 담당한다는 사실을 확인하였다.
1-2) Cdo 결손 마우스의 노화에 의한 행동 능력 변화
운동 신경 (motor neuron)특이적으로 Cdo가 결핍된 마우스(Cdo
Hb9cre mice)의 경우, 도 2에서 보듯이, 족적평가에서 Hb9cre의 경우, 야생형에 비하여 보폭이 감소되는 등, 걸음걸이에 문제가 생긴 것이 관찰되어, 운동신경이 퇴화되었음을 확인할 수 있었다. 또한, 행동 평가로서 Open field test를 시행하여 확인한 결과, Cdo
Hb9cre 마우스에서 움직인 거리 및 보행속도가 생존군 및 사망군에서 모두 정상 마우스(Cdo
f/f)에 비해 감소된 것을 확인하였다. 아울러, 근력 평가 결과에서도 Cdo
Hb9cre 마우스의 생존군 및 사망군 모두 정상 대조군에 비하여 근력이 저하된 것이 확인되었다(도 3).
위와 같은 실험 결과로부터, 운동 신경에서 Cdo이 노화된 마우스의 행동 능력을 유지하는데 중요한 역할을 담당하는 것을 알 수 있다.
1-3) 노화 마우스의 Cdo 발현량 확인
노화에 의하여 Cdo 발현에 영향이 있는지 여부를 확인하기 위하여 5 개월령 마우스와 27개월령 마우스에서 각각 근신경 접합부 마커인 α-BTX(alpha-bungarotoxin)의 발현량을 확인한 결과, Cdo의 발현이 감소하는 것을 확인하였다(도 4 및 도 5).
즉, 노화에 의해 Cdo 발현이 감소되며, 특히 근신경 접합부에서 특이적으로 발현 감소가 일어남을 확인하였다.
실험예 2. 근육 줄기세포(위성 세포)에서의 Cdo 결핍에 의한 효과
2-1) Cdo 결핍 마우스의 근신경접합부 주변 변화
타목시펜을 투입함으로써 근육 줄기세포에 특이적으로 Cdo를 결핍시킬 수 있는 유전자 조작 마우스를 사용하였다. Cdo를 근육 줄기세포 특이적으로 결핍시킨 후 재생 과정에서 일어나는 현상을 관찰하기 위해 뱀독의 일종인 카디오톡신(CTX)이라는 독을 TA(Tibialis anterior)근육 및 EDL(Extensor digitorum longus) 근육에 주입해 손상시킨 후, 두 달 후에 재생이 완료된 근육을 분석하였다.
그 결과, 도 6에서 보듯이, EDL 근육에서 분리한 단일근 섬유에서의 근신경 접합부 분석결과, 타목시펜을 주입한 마우스의 NMJ에서 핵의 숫자가 감소되어 있는 것이 확인되었다 (n=20; Vehicle, n=22; tmx).
또한, 도 7에서 보듯이 핵막 구조 단백 유전자인 Syne2의 mRNA 발현이 Cdo를 결핍시킨 그룹에서 크게 감소해 있음을 확인할 수 있었다 (TA muscle, n=3; Vehicle, n=4; tmx).
특히 글리코리틱 근섬유에서 발현이 증가되는 유전자인 Hdac4의 발현이 크게 감소해 있으며, 이에 따라 Hdac4의 타겟 유전자가 전반적으로 감소해 있는 것을 확인되었다. 특히, Hdac4의 타겟 유전자인 Myh4 (MyHC-IIB)의 발현 감소가 확인되었다. 이 결과는 Cdo가 근섬유의 유형을 결정하는 역할을 할 가능성을 제시하는 결과이다 (TA muscle, n=3; Vehicle, n=4; tmx). 특히 tmx그룹에서 나타나는 근신경 접합부의 핵수의 감소는 근섬유 유형에 따른 특징일 가능성이 있음을 확인하였다.
다시 말해서 상기 실험 결과는, 근육 줄기세포 특이적 Cdo 결핍 모델의 경우 근신경 접합부의 형성에는 큰 차이를 보이지 않았으나 근섬유 유형에 따라 NMJ에서 특징을 나타낼 가능성이 있다는 것을 보여주는 것이다. 근 위축성 질환인 노화성 근 감소증 (Sarcopenia) 또는 당뇨와 같은 대사질환은 근 위축성 표현형을 유발하며, 가장 큰 특징중에 하나는 당성 근섬유인 MyHC-Ⅱb의 감소이다. 즉, Cdo는 근섬유의 유형을 결정하는 역할을 하는 것으로, 근 위축성 표현형을 대상으로 한 치료 목적의 단백질이 될 가능성을 제시한다.
2-2) 단일근 섬유 근신경접합부의 Cdo 발현 확인
Cdo의 특이적 발현 부위를 확인하기 위하여 유전자 조작을 하지 않은 야생형 마우스의 EDL 근육으로부터 단일근 섬유를 분리하여 Cdo 및 근신경 접합부 마커인 α-BTX를 형광 염색을 통하여 확인하였다. 그 결과 도 9에서 보듯이, 근신경 접합부 마커와 Cdo의 발현 부위가 거의 일치되는 것으로 확인하였으며, 이를 통하여 근신경 접합부에 특이적으로 Cdo가 발현하는 것을 알 수 있다.
2-3) 운동 신경에서 Cdo 발현 확인
운동신경에서 Cdo의 발현여부를 확인하기 위하여, 야생형 마우스의 척수 요추 부위에서 Cdo 및 운동 신경 마커인 ChAT(Choline acetyltransferase)를 형광 염색을 통하여 확인하였다. 그 결과 도 10에서 보듯이, 운동신경 마커와 Cdo의 발현 부위가 거의 일치되는 것으로 확인되었다. 따라서, 운동 신경에서 Cdo가 발현된다는 점을 알 수 있다.
2-4) 운동 신경 특이적 Cdo 결핍 마우스의 근신경 접합부 이상 확인
운동 신경 특이적으로 Cdo를 결핍시킨 마우스의 EDL 근육에서 운동 신경의 축색 돌기(motor neuron exon) 마커로서 사용되는 NF(neurofilament)와, 근신경 접합부에서 활성을 띄도록 도와주는 물질을 염색하는 마커인 시냅토파이신 (synaptophysin) 및 α-BTX를 형광 염색을 통해 확인하였다. 도 11과 같이, 운동신경 특이적으로 Cdo를 결핍시킨 마우스의 근신경 접합부는 그렇지 않은 쥐에 비하여 모양을 유지하지 못하고(Fragmentation) 있으며, 상대적으로 얉은 축색 돌기에 형성되고 있는 것이 관찰되었다.
또한, 도 11에서 화살표로 표시된 부분과 같이 시냅토파이신과 co-staning이 감소된 것으로 보아, 근신경 접합부에서의 활성 또한 감소한 것을 확인할 수 있었다.
2-5) 근육 줄기세포 특이적인 Cdo 결핍 마우스 및 단일근섬유 근신경 접합부에서의 Cdo 발현 변화
Cdo fl/fl;Pax7 ERT2Cre+/- 마우스는 타목시펜을 주입하여 근육 줄기세포(위성세포) 특이적으로 Cdo를 제거할 수 있는 마우스 모델이다. 도 12에 제시된 모식도와 같이, 마우스에 타목시펜 (tmx)을 주입하여 Cdo를 결핍시킨 후, TA-EDL 근육에 CTX 손상을 수행한 후 두 달 뒤, TA 및 EDL 근육을 분리하여 분석하였다. 이후, 타목시펜을 주입한 마우스(실험군)와 Vehicle(대조군)에서의 체중, 근육 무게 및 근육 무게/체중 값을 각각 비교하였다 (n=4 per each group). 그 결과, 도 12에서 보듯이, 타목시펜을 주입한 마우스의 TA 근육에서 Cdo의 mRNA 발현이 크게 감소함을 확인하였다 (n=3; Vehicle, n=4; tmx).
또한, 야생형의 성체 마우스(5 months male)의 EDL 단일 근섬유에서는 Cdo과 BTX이 발현되었으나, Cardiotoxin(CTX) 손상 7일 후와 14 일 후 관찰한 결과, 도 13에서 보듯이 각각 근신경 접합부에서 Cdo의 발현이 비 손상군의 근육과 비교해서 약해져 있음이 확인하였으며, 14일 후에는 7일째보다 더욱 선명하게 NMJ의 발현이 관찰되었다.
상기와 같은 결과로부터, 손상된 근섬유에서 근신경 접합부가 형성될 때, Cdo의 발현이 중요하다는 점을 알 수 있다.
2-6) 근육 줄기세포 특이적인 Cdo 결핍 시 근육 타입 변화
ShCdo를 도입한 렌티바이러스를 형질감염하여 Cdo가 결핍된 근육 줄기세포(C2C12)에서 발현되는 mRNA를 분석하여, Cdo 결핍 시, 근육 줄기세포 분화에 미치는 영향을 확인하였다.
그 결과, 도 14와 같이, Hdac4와 Myh4의 mRNA 발현이 크게 감소하였음을 확인하였으며, 이는 in vivo와 동일한 결과임을 알 수 있다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
Claims (10)
- Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는 운동 신경 세포 보호용 조성물.
- 제1항에 있어서,상기 운동 신경 세포 보호는 노화, 산화 스트레스 및 염증에 의한 운동 신경 세포의 손상을 방지하는 것임을 특징으로 하는, 운동 신경 세포 보호용 조성물.
- 제1항에 있어서,상기 조성물은 손상된 운동 신경 세포의 재 신경화를 촉진하는 것임을 특징으로 하는, 운동 신경 세포 보호용 조성물.
- Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는, 신경 근육 질환의 예방 또는 치료용 약학적 조성물.
- 제4항에 있어서,상기 신경 근육 질환은 근이영양증, 운동 신경 질환, 근병증 및 신경근 접합부 질환을 포함하는 군으로부터 선택되는 것을 특징으로 하는 조성물.
- 제5항에 있어서,상기 운동 신경 질환은 근위축성 측삭경화증(amyotrophic lateral sclerosis, ALS), 척수성 진행성 근위축(infantile progressive spinal muscular atrophy, SMA), 진행성 핵상마비(progressive bulbar palsy), 가연수성마비 (pseudobulbar palsy), 진행성 근육위축(progressive muscular atrophy, PMA), 진행성 축삭경화증(progressive lateral sclerosis, PLS), 단일 사지 근위축증 (monomelic amyotrophy, MMA) 및 헌팅턴 병을 포함하는 군으로부터 선택되는 것을 특징으로 하는 조성물.
- Cdo 단백질 또는 이를 암호화하는 유전자를 포함하는, 신경 근육 질환의 예방 또는 개선용 건강 기능 식품 조성물.
- 제7항에 있어서,상기 신경 근육 질환은 근이영양증, 운동 신경 질환, 근병증 및 신경근 접합부 질환을 포함하는 군으로부터 선택되는 것을 특징으로 하는 조성물.
- 제8항에 있어서,상기 운동 신경 질환은 근위축성 측삭경화증(amyotrophic lateral sclerosis, ALS), 척수성 진행성 근위축(infantile progressive spinal muscular atrophy, SMA), 진행성 핵상마비(progressive bulbar palsy), 가연수성마비 (pseudobulbar palsy), 진행성 근육위축(progressive muscular atrophy, PMA), 진행성 축삭경화증(progressive lateral sclerosis, PLS), 단일 사지 근위축증 (monomelic amyotrophy, MMA) 및 헌팅턴 병을 포함하는 군으로부터 선택되는 것을 특징으로 하는 조성물.
- i) 운동 신경 세포 또는 신경근 접합부에 후보 물질을 처리하는 단계;ii) 상기 후보 물질을 처리한 운동 신경 세포 또는 신경근 접합부에서 CDO의발현 또는 활성을 측정하는 단계; 및iii) 비 처리군과 비교하여 Cdo의 발현 또는 활성을 증진시키는 후보 물질들을 선정하는 단계를 포함하는 신경 근육 질환의 예방, 개선 또는 치료 물질의 스크리닝 방법.
