지질 이중층으로 코팅된 울퉁불퉁한 표면을 갖는 기판을 사용하는, 표적입자의 분석, 확인, 검출, 및/또는 가시화, 및/또는 표적입자의 존재 또는 부재를 결정하기 위한 어레이, 키트, 및/또는 장치 및 이를 이용한 방법이 제공된다.
전염병, 특히 바이러스 감염은 인간의 건강을 위협하고 전 세계적으로 막대한 경제적 손실을 초래하므로, 신속하고 민감하게 선택적으로 표적 바이러스를 탐지할 수 있는 플랫폼의 개발 요구가 증대되고 있다.
바이러스는 숙주 면역 체계를 조작하고 억제하기 때문에, 코로나 바이러스, 아프리카 돼지 열병, 에볼라, 인플루엔자와 같은 바이러스성 전염병은 인간과 가축의 생명을 위협한다. 바이러스성 전염병은 주로 감염이 진단되지 않은 감염된 숙주에 의해 유발된다. 감염된 개체를 적시에 정확하게 치료하고 심각한 전염을 예방하기 위하여, 간단한 방법으로 신속하면서도 높은 민감도로 선택적으로 표적 바이러스를 검출할 수 있는 기술이 필요하다.
현재 널리 사용되는 바이러스 검출 방법은 전체 바이러스 (whole viruses)를 검출하는 신속 인플루엔자 진단검사 (rapid influenza diagnostic test; RIDT), 표적 유전자를 증폭하는 중합효소연쇄반응(PCR), ELISA (Enzyme-linked immunosorbent assay) 등을 기반으로 한다. 전체 바이러스 검출 방법은 간단하고 추가 샘플 처리가 필요 없지만 많은 시간과 노동을 필요로 하는 방법이다. ELISA (Enzyme-linked immunosorbent assay)는 분석 프로토콜의 간편성과 단순성 때문에 전체 바이러스 검출에 주로 사용되고 있다. ELISA와 같은 바이러스 검출을 위한 기존의 칩-기반 바이오센서 분석법은 표적 검출을 위해 기판에 포획분자(예: 항체)를 고정하고 포획된 표적의 농도를 광신호로 변환하는 어려운 downstream labelling을 필요로 하기 때문에, 빠른 시간 내에 높은 감도 및 높은 특이성으로 신뢰할 수 있는 바이러스 검출 및 정량화를 제공하지 못하는 한계를 가진다.
따라서, 보다 간편하고 신속하게 높은 감도로 표적입자를 검출 및/또는 정량할 수 있는 기술의 개발이 요구된다.
본 명세서는 신속하고 민감한 정량적 바이러스 검출을 위한 지질 나노필러 어레이-기반 면역흡착분석 (lipid nanopillar array-based immunosorbent assay; LNAIA) 기술을 제공한다. LNAIA 플랫폼상의 3차원 구조 (3D) 나노필러 어레이 및 형광물질로 표지된 유동성 항체(fluid-fluorophore labeled antibody)는, 통상의 형광현미경 상에서도 매우 높은 감도로 표적입자를 검출할 수 있도록 표적입자 표면으로 형광-항체의 밀집을 유도하여 효율적이고 신속한 표적 결합을 용이하게 한다.
일 예는 다음을 포함하는, 표적입자의 분석, 확인, 검출, 및/또는 가시화용, 및/또는 표적입자의 존재 또는 부재를 결정하기 위한, 어레이, 키트, 및/또는 장치를 제공한다:
울퉁불퉁한 표면(uneven surface)을 갖는 기판 (substrate),
상기 울퉁불퉁한 표면을 코팅하는 지질이중층 (lipid bilayer), 및

본 명세서에서는 형광 표지된 항체가 유동성을 가지고 포함된 지지 지질 이중층 (supported lipid bilayer)가 표면을 덮고 있는 나노필러 패턴을 갖는 기판을 사용하여, 보다 신속하고 민감한 바이러스 검출 및/또는 정량적 분석이 가능한 지질 나노필러 어레이-기반 면역흡착분석 (lipid nanopillar array-based immunosorbent assay; LNAIA) 기술이 제공된다. 본 명세서에서 제공되는 형광 표지된 유동 가능한 항체 및 3차구조 나노필러 어레이 구조체를 이용한 표적입자의 검출 방법은, 3차구조 표면을 유동적으로 움직이는 표지된 다수의 항체가 표적입자와 다중 결합하여 표적입자 주변에 형광 신호를 밀집시킴으로써 고가의 고성능 장비를 사용할 필요 없이, 일반적 사양의 형광현미경 셋업에 의해서도 신속하고 안정적이고 효율적인 표적입자의 분석이 가능한 것을 특징으로 한다. 본 명세서에서 제안되는 LNAIA는 기존 ELISA 플랫폼으로는 달성할 수 없었던 높은 수준의 민감도로 보다 빠른 시간 내에 표적입자 (예컨대, 바이러스)을 검출 및/또는 정량화할 수 있다.

이하, 본 발명을 보다 상세히 설명한다:
본 명세서에서 사용된 바로서, 용어 “어레이”는 복수의 3차원 구조체의 사방 (상하좌우) 반복 배열에 의한 소정의 패턴을 가지는 구조체를 의미하는 것으로, 칩 등의 유사 용어와 동등한 의미로 호환될 수 있다.
상기 기판은 고체 기판으로서, 표면에 지질이중층 코팅이 가능하도록 친수성이거나 표면이 친수성을 가지도록 개질된 것일 수 있다. 일 예에서, 상기 기판은 유리, 실리카, 실리콘 (예, 실리콘 웨이퍼), 운모(mica), 친수성 분자(예컨대, 말단에 -OH, -COOH 등의 작용기를 가짐)의 자기조립단분자막(Self-Assembled Monolayer, SAM)으로 개질된 표면을 가지는 모든 고체 기판들 등으로 이루어진 군에서 선택된 재질의 것일 수 있고, 임의로 친수성을 가지도록 개질된 것일 수 있으나, 이에 제한되는 것은 아니다.
상기 울퉁불퉁한 표면(uneven surface)은 평평하지 않은 표면을 의미하는 것으로, 돌출부 및/또는 함몰부를 포함하는 3차원 구조체가 형성된 (또는 요철이 형성된) 표면을 의미할 수 있다.
일 예에서 상기 울퉁불퉁한 표면은 하나 이상 (예컨대, 2개 이상, 또는 복수)의 3차원 구조체 (돌출부, 함몰부, 또는 이들의 조합)에 의하여 형성된 것일 수 있다. 일 예에서, 상기 울퉁불퉁한 표면은 하나 이상 (예컨대, 2개 이상, 또는 복수)의 필러 (pillars), 하나 이상 (예컨대, 2개 이상, 또는 복수)의 홈 (grooves), 또는 이들의 조합을 포함하는 것일 수 있다. 상기 3차원 구조체의 크기 (지름, 너비, 높이, 깊이 등), 인접한 3차원 구조체 간 간격 등은 검출하고자 하는 표적입자의 크기에 따라서 결정될 수 있다.

상기 돌출부 (예, 나노필러) 및/또는 함몰부 (홈)의 형태는 별다른 제한이 없으며, 수평 단면 모양 기준으로, 다각형 (예, 육각형, 오각형, 사각형, 삼각형, 팔각형 등), 원형, 타원형 등일 수 있으며, 일 구체예에서 육각형일 수 있으나, 이에 제한되는 것은 아니다.

본 명세서에서 제공되는 어레이, 키트, 및/또는 장치에서, 상기 기판의 울퉁불퉁한 표면은 지질이중층으로 코팅되어 있으며, 상기 지질이중층의 유동성에 의하여 지질이중층 내부에 위치하는 2개 이상 (복수)의 포획물질에 이동성을 부여하여, 표적입자 주변으로 밀집될 수 있도록 한다.
일 구체예에서, 상기 표적입자는 포획물질에 의하여 포획 (검출, 인식, 및/또는 결합)될 수 있는 하나 이상(예컨대, 2 이상 또는 복수)의 물질 (예컨대, 단백질, 핵산 분자 등)을 표면에 가지는 모든 입자들 중에서 선택될 수 있다. 예를 들어, 상기 표적입자는 인플루엔자 바이러스 (e.g., 인플루엔자 A 바이러스 서브타입 H1N1, 인플루엔자 A 바이러스 서브타입 H3N2, 조류인플루엔자 바이러스 (인플루엔자 A 바이러스 서브타입 H5N1, 인플루엔자 A 바이러스 서브타입 H7N9 등)), 코로나바이러스 (e.g., Middle East respiratory syndrome coronavirus (MERS-CoV), Severe acute respiratory syndrome coronavirus (SARS-CoV), SARS-CoV-2, 등) 등을 포함하는 바이러스, 박테리아, 박테리오파지, 세포, 단백질 입자 (e.g., aggregates 등), 핵산 분자 (예컨대, DNA, RNA 등) 등으로 이루어진 군에서 선택된 1종 이상 (예컨대, 2종 이상 또는 복수)일 수 있다.
상기 포획물질은 상기 표적입자의 표면 상의 포획 가능한 물질 (예컨대, 단백질, 핵산 분자 등)과 (특이적) 결합 가능한 물질들, 예컨대, 항체, 압타머 등으로 이루어진 군에서 선택된 1종 이상일 수 있으며, 상기 포획물질은 형광물질, 플라즈몬 금속 나노입자 등과 같은 검출 가능한 적절한 표지물질(신호물질)로 표지된 것일 수 있다. 본 명세서에서 제공되는 어레이/키트에 있어서, 1종의 표적입자에 대한 포획물질은 2개 이상 (복수개) 포함될 수 있다. 일 구체예에서, 표적입자가 바이러스 또는 세포인 경우, 상기 포획물질은 상기 바이러스 또는 세포의 표면에 노출된 단백질 (예컨대, 각종 수용체, 항원 등)에 특이적으로 결합하는 항체, 압타머 등으로 이루어진 군에서 선택된 1종 이상일 수 있다.
상기 표지된 포획물질은 상기 울퉁불퉁한 표면을 도포하는 지질이중층 내부에 위치하며, 상기 지질이중층 내부에서 표면을 따라서 이동 가능하여 (유동성을 가짐), 2개 이상의 표지된 포획물질 울퉁불퉁한 표면의 바닥부와 3차원 구조체(돌출부; 예, 나노필러) 측벽에 의하여 형성된 3차원 공간에 위치하는 표적입자의 부위로 이동하여 상기 표적입자를 둘러싸고 표면의 복수의 포획 가능한 물질과 결합함으로써, 표지된 포획물질이 표적입자 주위로 밀집되어, 강한 신호를 발생하게 된다. 이로 인하여, 고가의 고성능 장비 없이도 육안 또는 일반적 검출 장비 (예, 형광현미경)에 의해서도 표적입자의 검출 및/또는 가시화가 가능하다.
본 명세서에서 제공되는 상기 키트 및/또는 장치에 있어서, 상기 어레이에 더하여, 임의로 포함 가능한 신호 (형광신호) 검출 수단은 상기 신호 검출을 위하여 사용 가능한 모든 검출수단일 수 있고, 일반적 수준 (예컨대, 40~400x 또는 40~100x 배율)의 현미경 (예컨대, 형광현미경 등)일 수 있으나, 이에 제한되는 것은 아니다. 이는 본 명세서에서 제공되는 검출 기술이 신호의 입체적 밀집화에 의한 높은 신호강도에 의한 우수한 가시화 효과 및 검출 감도를 설명하기 위함이며, 신호 검출을 위하여 고성능 검출수단을 사용하는 배제하고자 하는 의미로 해석되어서는 아니 된다.
본 명세서에서 제공되는 방법에서, 상기 시료는 개체로부터 얻어지거나 분리된 생물학적 시료 및/또는 환경(수질, 토양, 대기 등) 시료일 수 있으며, 상기 생물학적 시료는 세포, 혈액, 림프, 타액, 가래, 콧물, 소변, 대변, 기타 체액 등으로 이루어진 군에서 선택된 1종 이상일 수 있으나 이에 제한되지 않는다. 상기 개체는 포유동물, 예컨대, 인간, 가축 (예: 소, 돼지, 말, 양, 염소, 개, 고양이 등), 조류 (예: 닭, 오리, 거위, 칠면조, 타조, 메추라기 등) 등일 수 있으나, 이에 한정되는 것은 아니다.
본 명세서에서 제공되는 키트, 장치, 및/또는 방법에 의하여 표적입자 검출시의 검출감도는, 평평한 기판을 사용하는 경우 및/또는 기존의 유사 측정방법 (예컨대, ELISA 등)과 비교하여, 약 2배 이상, 약 5배 이상, 약 10배 이상, 약 20배 이상, 약 30배 이상, 약 40배 이상, 또는 약 50배 이상(예컨대, 약 100배 이상, 약 1,000배 이상, 약 5,000배 이상, 또는 약 10,000배 이상) 높은 것을 특징으로 한다 (도 2, 도 3b, 도3c 등 참조). 일 구체예에서, 상기 표적입자가 바이러스인 경우, 본 명세서에 따른 이질이중층 코팅된 나노필러 기판을 사용하여 검출시의 검출한계는 약 150개 바이러스 입자로, 기존의 ELISA 대비 약 40배 이상의 높은 검출 감도를 얻을 수 있다.
본 명세서에서 제공되는 키트, 장치, 및/또는 방법에 의하여 표적입자 검출시의 검출 시간은 약 3시간 미만, 예컨대, 약 5분 내지 약 300분, 약 5분 내지 약 270분, 약 5분 내지 약 240분, 약 5분 내지 약 210분, 약 5분 내지 약 180분, 약 5분 내지 약 150분, 약 5분 내지 약 120분, 약 5분 내지 약 90분, 약 5분 내지 약 60분, 약 5분 내지 약 50분, 약 5분 내지 약 40분, 약 5분 내지 약 30분, 또는 약 25분 정도로, 기존의 유사 측정 방법 (예컨대 ELISA) 대비 검출 시간이 현저히 짧다는 이점을 갖는다.
또한, 본 명세서에서 제공되는 키트, 장치, 및/또는 방법은 기판 표면에 코팅된 지질이중층 내에 도입된 복수의 표지된 포획물질을 이용하여 표적입자 특이적으로 검출신호를 발생시키므로, 별도의 염료 (형광염료 등)의 첨가를 필요로 하지 않을 수 있다. 이러한 특징으로 인하여, 명세서에서 제공되는 키트, 장치, 및/또는 방법은 별도의 미반응 염료의 세척과정 없이 정확한 분석이 가능하다는 이점이 있으며, 이는 기존의 염료를 사용하는 경우에 필수적으로 필요한 미결합 표적 (세포 등), 염료 등을 제거하기 위한 세척과정이 필요한 것을 고려할 때, 분석에 소요되는 시간 및/또는 노력을 단축하고 분석 정확도를 높이는데 유리하게 작용할 수 있다.
본 명세서에서 제공되는 지질 나노필러 어레이-기반 면역흡착분석 (lipid nanopillar array-based immunosorbent assay; LNAIA) 기술의 일 예를 도 1a 및 1b를 참조하여 바이러스 검출 용도를 대표적인 예로 들어 설명하면 다음과 같다:
지지 지질 이중층 (Supported lipid bilayer; SLB)은 고체 기판 상의 유동성 합성 지질 이중층(synthetic fluid lipid bilayer)으로, 세포막과 유사한 지질의 동적 움직임 (dynamic movement)이 가능하여, 나노 구조체와 혼성화되어 다양한 생물공학적 응용에 사용될 수 있다. 또한 플라즈몬 나노입자(plasmonic nanoparticles)는, 단일 입자 수준에서 나노입자 간의 상호 작용을 이미징하고 분석하기 위해, SLB로 변조될 수 있다. 본 명세서에서는, 엔도사이토시스를 통해 바이러스가 세포내로 포식(engulf)될 때의 숙주 세포막에 있는 수용체의 클러스터링과 동적 움직임과 유사한, 지질 나노필러 어레이-기반 면역흡착분석 (lipid nanopillar array-based immunosorbent assay; LNAIA)이라고 명명된, 미세제작된(nanofabricated) SLB 상에서의 면역흡착성 전체 바이러스 분석법이 제공된다 (도 1a 및 1b 참조). 스트렙타빈 (streptavin) 링커에 의해 지질 나노필러 어레이 (LNA)에 연결된 Cy3 형광 염료 (Cy3-항체)를 가지는 바이오틴화된 항체는 LNA 표면을 따라서 동적 이동이 가능하다 (도 1a의 b-c 참조). LNAIA에서, 표적 바이러스가 LNA에 도입되면 자유롭게 확산하는(diffusing) Cy3-항체에 의해 포획되어 결국 바닥의 표면과 필러의 측벽에 상의 다수의 Cy3-항체로 둘러싸여, 항체와의 3차원 다가 상호작용(three-dimensional polyvalent interaction)이 생성된다 (도 1a의 d-f 참조). 이러한 3차원 다가 상호작용은 일반적으로 사용되는 epifluorescence 현미경으로도 형광 배경과 명확하게 구별되는 국소화 및 고정된 형광 반점(localized and immobilized fluorescent spots)을 생성할 수 있다. 바이러스 표면 상의 hemgglutinin(HA) 분자의 neuraminidase (NA)보다 매우 (수십배 이상) 많기 때문에, 다수의 HA-특이적 Cy3-항체의 효율적인 국소화를 통해 강력한 형광 신호를 얻을 수 있다.
SLB의 기초가 되는 나노필러 어레이는 나노구조 돌출부를 형성하도록 설계되어, 표면에 고정된 형광 표지된 항체 (예컨대, Cy3-항체)의 동적 이동이 가능한 3차원 미로 유사 환경 (3D labyrinth-like environment)과 보다 넓은 표면적을 제공함으로써, 표적 바이러스가 상기 형광 표지된 항체와 충분한 빈도 및/또는 횟수로 충돌하도록 하도록 하여, 표적 바이러스 검출의 신속성과 민감도를 강화시킬 수 있다. resist-free direct UV/thermal nanoimprint lithography로 제작된 실리카 나노필러 어레이는 기판 상의 SLB를 안정적으로 형성하는 데 충분한 친수성을 제공한다. 나노필러 어레이는 지름과 높이가 200nm이고 필러(pillars) 사이에 200nm 간격이 있어서 육각형 어레이를 생성하여 (도 1a의 a, 도 7 및 도 8), 바이러스와 나노필러 사이의 충돌을 최대화하고, 3 차원 다가 항원-항체 상호작용에 의해 다수의 항체가 80-120 nm 크기의 인플루엔자 바이러스를 신속하고 효율적으로 포획할 수 있도록 한다.
일반적인 실험으로도, LNAIA는 분석물을 로딩한 후 약 25분 이하의 매우 단시간에 분석이 가능하며, 단계별 분석 절차는 도 1a, 1b 및 도 6에 예시적으로 설명되어 있다. 친수성 실리카 나노필러 어레이 상에 부착된 8-㎕ 실리콘 챔버에, 0.05%(w/v) biotinylated-DOPE (in 150 mM PBS)를 가지는 작은 단층 소포 (small unilamellar vesicles; SUV)를 도입하였다. 챔버 내의 파열된 소포는 지지 나노필러 어레이 상의 지질 이중층을 형성한다. 과량의 SUV를 세척한 후, Cy3- 표지된 스트렙타비티딘 및 비오틴화 폴리클로날 HA 특이적 항체를 챔버에 도입하였다. 분석물을 로딩하기 전에 일반적으로 사용되는 형광현미경을 사용하여 기판을 이미징하였다. 분석물 8uL를 로딩하고 20 분 후에 automated imageJ software를 사용하여 챔버에서 새로 형성된 형광 스팟의 수를 이미지화 및 정량화 하였다.
요약하면, 본 명세서는 표적입자 (예, 바이러스)를 탁월한 동적 범위 (at least 5 orders of magnitude in concentration)로 매우 낮은 농도 (약 150 개의 바이러스)에서도 신속하고 (~ 25 분) 안정적으로 검출할 수 있는 LNA 기반의 새로운 유형의 면역흡착분석을 제공하며, 일반적으로 사용되는 형광현미경으로도 표적입자의 고감도 검출이 가능한 것을 특징으로 한다. 본 명세서에서는 표적입자 (예, 바이러스)와 유사한 크기를 가진 나노필러 구조가 표적입자와 유동적 항체 사이의 동적이고 협력적인 3 차원 상호작용의 성능을 크게 향상시킬 수 있음을 입증하였다. 지질 이중층 코팅된 나노필러 기판은, 기존의 분석 플랫폼 (예: ELISA)을 사용한 항체의 일반적인 물리적 분자 결합상수를 넘어서, 표적입자 검출 감지에서 있어서의 결합 동력학 및 특이성을 높일 수 있다. 표적입자와 항체 사이의 특이적 다중 결합에 의하여 생성된 국소화되고 집중된 형광신호에 의하여 기존에 일반적으로 사용되는 형광현미경으로 300ms의 연장된 노출시간에서 안정적으로 검출할 수 있다. 배경 신호를 평균화하여 위양성 신호를 효율적으로 제거하고, 진양성 신호를 선택적으로 수집할 수 있다. LNAIA에서의 다중 형광채널을 사용하는 표적입자의 다중 검출은 기존의 다른 다중 면역 분석에서 교차 반응 항체에서 발생하는 많은 어려움을 해결할 수 있다. 표적과 포획 분자 간의 충돌을 극대화하는 3차원 미로-유사 구조의 기판을 채택하고 바이오센서의 변환기와 검출기를 단일 장치에 통합하고 자동화된 신호 분석을 적용하여 검출 시간을 크게 단축하고 분석 단계를 최소화 할 수다. 널리 사용되는 검출 장비에 의해서도 분석(검출) 가능함으로써, 분석 시간이 단축되고 분석 프로세스가 최소화되므로, 신속하고 간편한 고감도 진단 테스트가 용이하게 수행될 수 있다.
LNAIA는 최소한의 sampling handling로 표적의 빠르고 민감하며 선택적이고 신뢰할 수 있는 검출을 위한 새로운 검출 전략 및 플랫폼을 제공하며, 높은 감도와 넓은 동적 범위를 가지고 단백질, DNA 등의 다양한 종류 서로 다른 타겟의 신속한 반응 키트 개발을 포함하여 다양한 실제 용도에 쉽게 적용할 수 있다.
도 1a는 일 실시예에 따른 나노필러-지지 지질 이중층 면역흡착분석(LNAIA)의 과정을 개략적으로 보여주고, 나노필러-지지 지질 이중층 기판의 특징을 보여주는 모식도이다.

이하, 본 발명을 실시예에 의해 보다 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
실시예 1. LNAIA 키트 제작
1.1. small unilamellar vesicles (SUVs)의 제작
50 mL 둥근 바닥 플라스크에서, 97.45 mol% DOPC(1,2-Dioleoyl-sn-glycero-3-phosphocholine), 0.05 mol% 비오틴화 DOPE(1,2-Dioleoyl-sn-glycero-3-phosphoethanolamine), 및 2.5 mol% PEG-DOPE (Molecular Probes, USA)를 혼합하여 (in chloroform), 지질 용액을 준비하였다. 상기 준비된 지질용액을 회전 증발기로 증발시켰다. pillar-to-flat 경계 영역에서의 SLB(Supported lipid bilayer)의 형광 이미징 및 FRAP 실험(Fluorescence recovery after photo bleaching)을 위해, 1 mol% FITC-DHPE(1,2-dihexadecanoyl-sn-glycero-3-phosphoethanolamine) 및 1 mol% TexasRed-DHPE를 각각 둥근 바닥 플라스크에 넣었다. N2 기체의 스트림을 사용하여 지질막을 완전히 건조시켰다. 완전히 건조시킨 후, DI water (Nanopure water with a minimum resistance >18 MΩ cm1)에 재현탁하고, cryo-tube로 옮긴 후, 3 회의 동결-해동 사이클(freeze-thaw cycles)을 수행하였다. 최종 지질 농도는 4mg/mL였다. 상기 지질 용액을 25℃에서 100nm의 기공 지름을 갖는 폴리카보네이트 (PC) 멤브레인 (Whatman, Fisher Scientific)을 통해 21 회 압출시켰다. 약 100 nm의 리포좀이 얻어졌고, 이를 사용시까지 4℃에서 보관하였다.
1.2. Antibody biotinylation.
0.2 mg/mL H1N1 항체 (Influenza A H1N1 (A/Puerto Rico/8/1934) Hemagglutinin / HA Antibody, Rabbit Polyclonal Antibody, Cat.# 11684-T56; Sinobiological; in 150 mM PBS (50 uL (microliter))) 및 20 uM (micromole) NHS-biotin (N-hydrocysuccinimidobiotin Thermo Scientific, USA; in DMSO (5 uL))를 혼합하고, 실온에서 2 시간동안 배양하였다. 미반응 비오틴을 desalting columns (Zeba Spin desalting columns 7K MWCO, Thermo Scientific, Rockford, IL, Rockford)으로 제거하였다. 상기 얻어진 반응물을 세척된 탈염 컬럼(desalting columns)에 로딩하고, 스태커(stacker)로서 150mM PBS를 첨가하였다. 상기 컬럼을 eppendorf tube 안에서 2분동안 1500 g 에서 원심분리하여 tube 아래에 모인 용액에 있는 바이오틴으로 개질된 항체를 지질 지중층 상에 있는 스트렙타비딘 (STV)와 결합 시켰다.
1.3. spin-on-glass 및 nanoimprint lithography를 사용한 Nanopillar pattern 제작
4-inch 유리 웨이퍼(glass wafer)를 plasma asher로 10 분 동안 세척하였다. 이와 같이 준비된 유리 기판을 IC3-200 spin-on-glass (SOG, Futurrex Inc. USA)로 3000rpm에서 30초 동안 스핀 코팅하였다(spin-coated). hole-patterned polydimethylsiloxane (PDMS; SYLGARD 184, Dow Corning, USA) stamp는 직경 200nm, 깊이 300nm, 피치(pitch) 400nm였으며, 이를 상기 SOG-코팅된 유리 웨이퍼 상으로 ANT-6HO2 UV/thermal nanoimprint lithography (KIMM, Korea)를 사용하여 150℃에서 10분동안 2000kgf cm-2로 압착시켰다. 그 후, 홀 스탬프(hole stamp)를 상기 유리 웨이퍼로부터 분리하여, 표면에 나노필러가 형성된 기판 (이하, ‘나노필러 기판, 나노필러 패턴 기판, 또는 나노필러 어레이’라고 칭함)을 얻었다 (도 1a의 a 참조).
1.4. LNAIA 키트 제작
바이러스 분석 키트를 제작하기 위해, 상기 실시예 1.3에서 준비된 실리카 나노필러 패턴 (nanopillar-patterned) 기판을 에탄올과 아세톤으로 각각 10 회 세척한 후 plasma asher로 처리하여, 상기 기판에 친수성을 부여하였다. 상기 기판 위에 스티커 챔버 (sticker chamber; 8uL)를 부착하고, 상기 실시예 1.1에서 준비한 small unilamellar vesicle(SUV)을 150mM 인산염완충식염수(PBS)와 1:1 (v/v) 혼합하고 30 분 동안 상기 챔버에 도입하였다. 일회용 니들로 각 챔버의 바닥에 십자선을 표시하고, 150mM PBS로 과량의 SUV를 제거한 후, 20uM 소 알부민 혈청 (BSA, Sigma Aldrich; in 150 mM PBS) 8uL를 도입하여 상기 nanopillar-patterned 기판 상의 non-covered 영역을 비활성화시켰다. 150mM PBS로 3 회 세척하여 과량의 BSA를 제거한 후, 20nM Cy3-변형 스트렙타비딘 (Cy3-STV, Molecular Probes, USA; in 150 mM PBS) 8uL를 30 분 동안도입 한 후, 150mM PBS로 추가로 세척하였다. 이어서, 280 nm에서의 O.D.값이 0.02인 바이오틴화 항체(실시예 1.2)를 30 분 동안 도입하였다. 분석 전에, 상기 챔버를 150mM PBS로 3 회 세척하였다.
실시예 2. LNAIA
상기 실시예 1.4에서 준비된 LNAIA 키트 (표적 바이러스: H1N1)를 사용하여 LNAIA를 수행하였다. 단계별 실험 과정과 LNAIA 챔버 설계는 도 5 및 도 6에 각각 예시적으로 설명되어 있다. 1.8*104~1.8*109 viral particles/mL virus (Influenza A/Puerto Rico/8/1934 H1N1 (positive target) 및 Adenovirus type 5 (negative target)) 샘플 8uL를 1 %(v/v) 인간 혈청 (from human male AB plasma, USA origin, sterile-filtered, Sigma-Aldrich; in PBS)과 함께 상기 LNAIA 챔버에 도입하기 전 및 20분 후에, 488 nm 레이저 노출 및 60x 렌즈 (via TE-2000; Nikon, Tokyo, Japan)를 이용하여 각 챔버의 4 개 사분면의 Epifluorescence 이미지를 얻었다. 80x80 um2 (512x512 pixel2) 시야(field of view)와 100ms 노출 시간으로 3 개의 이미지를 쌓았다. 상기 형광 이미지를 분석하여 ΔN (4 개의 사분면에서의 증가된 형광 반점 수)를 계산하였으며, 이 때, Image J software 용 MOSAIC plugin을 사용였다. 단일 웰에서 얻어진 4 개의 ΔNq (각 사분면에서 증가 된 형광 반점 수)는 ΔN로서 합산되었다.
참고예 1. ELISA (비교예)
1.8*104~1.8*109 viral particles/mL virus (Influenza A/Puerto Rico/8/1934 H1N1 (positive target) 및 Adenovirus type 5 (negative target)) 샘플 50uL를 1% 인간 혈청 (in coating buffer (0.2 M Na2CO3/NaHCO3, pH 9.6))과 함께 각각의 웰에 첨가하고, 실온에서 2 시간 동안 배양하였다. 웰을 PBST (Phosphate Buffered Saline with Tween; 137mM NaCl, 2.7mM KCl, 10mM Na2HPO4, 0.1w/v% Tween 20) 200uL로 3 회 세척하고 핸드 타올에 두드려 건조시켰다. 각 웰의 나머지 표면은 1 % BSA (in PBS (137mM NaCl, 2.7mM KCl, 10mM Na2HPO4, 1.8mM KH2PO4)) 200uL를 사용하여 실온에서 2 시간 동안 차단시켰다. 그 후, PBST 200 uL로 2 회 세척하여 과량의 BSA를 제거하였다. 일차항체 (in PBS with 1% BSA) 2ug/mL를 총 부피 100uL로 실온에서 2 시간 동안 각 웰에 도입하였다. 그 후, PBST 200 uL로 4 회 세척하여 과량의 항체를 제거하였다. 이차 항체 (with 1 % BSA in PBS) 100uL를 첨가하고 실온에서 2 시간 동안 배양하였다. 웰을 200 uL PBST로 4 회 세척하고 3,3',5,5'-Tetramethylbenzidine (TMB) 50 uL를 첨가하였다. 15 분 후, 각 웰을 2M H2SO4 50uL로 처리하고 microplate reader (Multiple Plate Reader, Victor 3, Perkin Elmer, USA)로 분석하였다.
참고예 2. 투과전자현미경 (transmission electron microscopy; TEM) 분석
나노필러 패턴 상의 SLB 구조를 이미징하기 위해, 나노필러 labyrinth SLB를 실온에서 30분 동안 2% 글루타르알데하이드 및 4% 파라포름알데하이드 (in 0.1M 카코딜산나트륨 버퍼(sodium cacodylate buffer; SCB) (pH 7.3))에서 고정시킨 후, 밤새 고정하였다. 이어지는 단계들은 graded ethanol series까지 4℃에서 수행하였다. 샘플을 0.2 M 수크로스를 포함하는 0.1 M SCB에서 3 회 세척하고 환원된 오스뮴 (1% osmium tetroxide with 0.8 % potassium ferricyanide in 0.1 M SCB)으로 후-고정 시켰다. 증류수로 3 회 세척한 후, 2% 우라닐 아세테이트로 en-bloc 염색을 수행하였다. 샘플을 증류수로 3 회 세척한 후, 30%, 50%, 70%, 80%, 및 100% 에탄올로 순차적으로 탈수시켰다. 그 후, 에탄올을 아세토니트릴로 교환했다. 샘플 침투(Sample infiltration)는 아세토니트릴과 Spurrs' resin의 1:1 혼합물로 2 시간동안, 아세토 니트릴과 상기 수지의 1:2 혼합물로 2 시간동안, 퓨어(pure) 수지로 실온에서 밤새 수행하였다. 수지를 새로운 수지로 교환하고 70℃에서 밤새 베이킹하였다. 바닥 실리카 나노필러 패턴(bottom silica nanopillar pattern)을 흄 후드(fume hood)에서 49% hydrofluoric acid (Sigma Aldrich)으로 식각(etched away)하고, 증류수로 완전히 세척한 후, 70℃에서 30 분간 건조시켰다. 샘플을 퓨어 수지로 재-매립하고 70℃에서 밤새 베이킹하였다. 이어서, 상기 수지 블록을 ultramicrotome (EM UC7, Leica, Wetzlar, Germany)으로 70nm 두께의 절편으로 분할한 다음, TEM copper grid (Veco Center Reference Square Grid, 200-mesh, Cu, Tedpella, USA) 상에 수집하고, 2% 우라닐 아세테이트로 후-염색하였다. 상기 준비된 그리드를 120kV 투과전자현미경 (Talos L120C, FEI, Hillsboro, United States)으로 이미지화하였다.
참고예 3. 주사전자현미경 (Scanning electron microscope; SEM) 분석
LNAIA에 의해 검출된 바이러스가 nanopillar labyrinth의 벽과 바닥 사이에 부착되었는지 여부를 확인하기 위해, 기판을 후-분석 동결 건조시킨 다음, 백금 증착 및 SEM (JSM-7800F Prime, JEOL Ltd, Akishima, Japan)를 사용한 이미징을 수행하였다.
실시예 3. 나노필러 어레이 표면의 SLB(supported lipid bilayer) 형성 확인
상기 실시예 1.4에서 실리카 나노필러 패턴 (nanopillar-patterned) 기판(실시예 1.3)에 small unilamellar vesicle(SUV) (실시예 1.1)가 도입된 기판을 투과전자현미경 (transmission electron microscopy; TEM) (120 kV, Talos L120C, FEI, Hillsboro, United States) 및 형광 현미경 (TE-2000, Nikon, Tokyo, Japan)으로 관찰하여, 도 1b의 h-n에 나타내었다. 그 결과, 나노필러 어레이 상의 SLB의 형성을 확인하였다.
SLB는, 필러의 벽과 바닥의 표면 사이의 효율적인 바이러스 포획을 위해, 나노필러의 측벽과 상부 및 기판의 바닥의 표면을 완전히 덮도록 설계되었다.
나노필러 기판 표면에서의 SLB 커버리지(coverage)의 특징 분석을 위하여, SLB의 phosphate head groups을 우라닐 아세테이트로 염색하고 Spurr’s resin에 함입시키고, ultramicrotome으로 미세 절단하고, TEM으로 관찰하였다 (도 1b의 h 및 i, 도 2, 및 도 9 참조). 도 2의 i의 어두운 선(화살표 표시)은 염색된 SLB가 나노필러 기판 표면을 밀접하게 following함을 보여준다.
평평한 기판에 비해 나노패턴 어레이의 표면적이 넓기 때문에, 나노패턴 기판의 형광 강도는 평평한 표면의 기판보다 훨씬 더 강하다. 형광 강도 비율과 비교하기 위해, 실시예 1.4에서 제작된 LNAIA 키트에서의 필러패턴 영역과 평평한 영역 사이의 이론적 표면적 비율을 다음의 수식 1을 이용하여 계산하였여 도 1b의 k-m에 나타내었다.
상기 수식 1에서, Acell은 하나의 원통형 필러를 포함하는 단위 셀의 총 표면적, A는 필러가 없는 단위 셀의 표면적, h는 필러의 높이 (200nm), p는 필러의 표면 커버리지, R은 필러의 반지름(100nm)이다.
상기 수식 1에 의하여 계산된 값은 2.05로서, 측정된 형광비 1.93과 거의 일치하는 결과를 보였으며, 이는 나노필러 패턴을 따라 SLB가 형성됨을 추가로 보여준다 (도 1b의 k-m 참조).
실시예 4. LNAIA에 의한 바이러스 분석
나노필러 기판 표면에서의 SLB의 유동성을 조사하기 위해, 광표백(photobleaching; FRAP) 실험 후, 형광복구(fluorescence recovery)(https://doi.org/10.1007/978-1-61779-207-6_26 참조)를 수행하여, 얻어진 결과를 도 1b의 n에 나타내었다. 비교를 위하여, 나노필러가 형성되지 않은 4-inch 유리 웨이퍼(glass wafer)를 기판을 사용하여 실시예 1.4를 참조하여 제작된 키트 (Flat SLB)를 사용하였다.
표백 후 형광복구 정도는 SLB가 유동성을 잃지 않고 나노필러 기판을 덮고 있음을 나타낸다. 기존의 나노패턴 기판과 SLB를 결합한 연구에서 SLB는 소수성 표면 또는 부적절한 리포솜 크기로 인해 나노패턴을 완전히 덮지 않거나 부유 구조를 형성하였다. 본 실시예에서 제공되는 LNAIA에서, 나노필러 기판을 실리카로 제작함으로써, 높은 친수성과 적절한 크기(dimensions)를 가지도록 하며, 나노필러 기판 상의 패턴을 따라 지질 이중층의 균일한 형성이 가능하다 (도 2의 a 및 b 참조).
또한, 기존의 epifluorescence 현미경을 통해 표적 바이러스 주변의 유동성 형광표지 항체의 국소화를 모니터링하였다. 시릿예 2에 기재된 LNAIA 분석 중에, 20 분간 샘플 로딩 전후의 형광 이미지를 캡처하여 도 2의 c 및 d에 나타내었다. 도 2의 c 및 d에 나타난 바와 같이, H1N1 비리온이 항체-변형(antibody-modified) LNA(lipid nanopillar array)에 접근함에 따라, 여러 개의 폴리클로날 항체가 바이러스에 결합하고, 상기 형광표지 항체를 단일 바이러스 크기와 비슷한 영역에 위치시킨다. 농축된 항체는 신호 대 배경 노이즈 비율이 높은 단일 바이러스에 해당하는 즉시 감지 가능한(readily-detectable) 형광 스팟을 생성한다.
정량적 바이러스 검출을 위해, particle-counting software (MosaicSuite particle tracker plugin of image J)를 사용하여 증가된 형광 스팟의 수 (ΔN)를 자동으로 계수하였다. 나노필러 어레이 구조가 분석 감도 및 표적 포획 효율성을 증가시키는 데 중요한 역할을 한다는 것을 확인하였다. Cy3-항체의 빠르고 강력한 공동위치화(colocalization) 및 3 차원 항원-항체 상호 작용으로 인한 표적 바이러스의 고정화는 일반적으로 사용되는 기존의 형광 현미경으로도 효율적인 표적 검출을 가능하게 하는 높은 형광 신호 대 배경 신호 비율을 생성한다 (도 2의 a 및 c 참조). 반면, 평평한 SLB에서의 바이러스의 높은 측면 이동성과 Cy3-항체의 비효율적인 공동위치화는 background에서의 단일 바이러스의 가시화를 방해하여, 샘플 로딩 전후의 형광신호 차이가 거의 관찰되지 않았으며 (도 2의 b 및 d 참조), 평평한 SLB이 형성된 기판상에 있는 형광-항체는 응집되지 않으므로 일반적인 형광현미경으로 형광 스팟을 관찰할 수 없으며, 단일 형광-항체를 관찰하기 위해서는 고급형 total internal reflection fluorescence microscope (TIRFm, TE-2000, Nikon, Tokyo, Japan) 이 필요하다.
또한, 실시예 2의 LNAIA 분석 수행 후 동결 건조된 기판을 SEM으로 관찰하여 (참고예 3), 얻어진 이미지를 도 2의 e에 나타내었다. 도 2의 e에 나타난 바와 같이, 분석 후의 LNA 상에서의 바이러스의 위치는 대체적으로 필러의 측벽과 바닥 표면 사이이며, 이는 효율적인 바이러스 포획을 위해 육각형 배열로 돌출된 나노필러 구조가 중요함을 추가로 보여준다.
항체를 포함하는 SLB (Ab-O) 및 항체가 없는 SLB (Ab-X)를 이용하여 LNAIA를 수행하여 얻어진 ΔN 값을 도 2의 f에 나타내었다. 도 2의 f에 나타난 바와 같이, LNA (지질 나노필러 어레이)를 사용한 경우의 항체를 포함하는 시험군의 ΔN 값은 항체가 없는 대조군에서의 수치보다 현저하게 높은 반면 (p-value of 0.0286), 평평한 SLB(flat SLB)는 시험군과 대조군이 유사한 ΔN 값을 나타내었다 (p-value of 0.028).
epifluorescence microscope를 통하여 관찰한 나노필러 SLB상의 형광 반점의 궤적을 도 2의 g에 나타내고, TIRF 현미경(TE-2000, Nikon, Tokyo, Japan)을 통하여 관찰한 평면 SLB의 형광 반점의 궤적을 도 2의 h에 나타내었다. 도 2의 g 및 h에 나타난 형광 반점의 궤적은 각각 LNA 및 평평한 SLB 표면에 대한 바이러스 입자 신호의 밀집화 (LNA; 도 2의 g) 및 자유 확산 (평평한 SLB; 도 2의 h)을 보여준다. LNA의 국소적인 형광 반점의 궤적은 기존의 epifluorescence 현미경을 통해서도 관찰 가능하지만, 평평한 SLB의 형광 반점의 궤적은 epifluorescence 현미경으로 감지할 수 없으며 TIRF 현미경을 통해서만 관찰할 수있다 (도 2의 g-j). LNA는, 표면과 바이러스 사이의 충돌을 강화하는 다수의 돌출구조 및 확장된 3차원 표면적으로 인해, 표면 항체에 대한 바이러스의 binding kinetics를 향상시킨다.
실시예 4. LNAIA와 ELISA의 비교
실시예 2 및 참고예 1과 같이 LNAIA와 ELISA를 수행하여 얻어진 결과를 도 3b 및 3c에 나타내었다. ELISA 사용하는 경우, 비특이적 바이러스인 H3N2 및 아데노 바이러스 (AdnV)는 낮은 농도(약 10^5 particels/chamber 이하)에서 표적 바이러스(H1N1)와 구별되지 않았다 (도 3c). LNAIA와 ELISA 모두에 동일한 항체를 사용했지만, LNAIA는 ELISA에 비해 훨씬 더 높은 표적 선택성을 보여주었다 (도 3b).
이러한 LNAIA의 높은 선택성을 다음과 같은 LNAIA의 특징에 기인한다: LNAIA에서 신호 생성은 Cy3-항체와 바이러스 표적 사이의 협력적 상호작용에 의해 수행된다. LNAIA에서, 진양성 신호(true positive signals; 특이적 결합)는 Cy3-항체의 농도가 형광현미경의 임계값을 초과할 때만 생성되기 때문에, LNAIA를 사용하여 위양성 신호(False positive signals; 비특이적 결합)를 효율적으로 제거할 수 있고, 각각의 개별적 클러스터된 스팟은 바이러스 표적이 형광 표지된 항체에 의하여 다중 포획된 경우에만 확실하게 이미지화 및 분석될 수 있다 (도 3a). 또한, 분석물 내의 세포외 분자는 막 단백질이 없는 인공 세포막인 SLB에 결합할 수 없다 (SLB는 비특이적 결합이 최소화된 우수한 표면임). LNAIA 음성 대조군에서의 배경 신호가 ELISA 기반 음성 대조군을 사용한 경우와 비교하여 훨씬 낮다는 점이 상기 결과를 추가로 지지한다 (도 3c).
중요한 점은, LNAIA 를 사용하면 10^5 구간에서 dN(ΔN) 값이 선형관계를 나타낸다는 것이다. LNAIA의 검출 한계는 약 150 개의 바이러스 입자로, 시판되는 H1N1 ELISA보다 약 10,000배 더 높은 감도를 나타낸다. 또한 ELISA는 어려운 항원 결합, 라벨링, 및 세척 절차로 인해 적어도 3 시간보다 많은 시간이 소요되는 반면, LNAIA는 단지 25분 정도로 분석이 가능하다 (도 3c).
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시예일뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
Claims (18)
- 울퉁불퉁한 표면을 갖는 기판,상기 울퉁불퉁한 표면을 코팅하는 지질이중층, 및상기 지질이중층 내에 유동성을 가지고 위치하고 표적입자에 결합하며 신호물질로 표지된, 복수의 포획물질을 포함하는, 표적입자 검출용 어레이.
- 제1항에 있어서, 상기 울퉁불퉁한 표면은 복수의 필러, 홈, 또는 이들의 조합을 포함하는 것인, 표적입자 검출용 어레이.
- 제2항에 있어서, 상기 필러 또는 홈의 지름, 높이, 또는 간격은 상기 표적 입자의 평균 직경의 1.2배 이상인, 표적입자 검출용 어레이.
- 제1항에 있어서, 상기 기판은 친수성 표면을 가지거나 표면이 친수성으로 개질된 고체 기판인, 표적입자 검출용 어레이.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 표적입자는 바이러스, 세포, 단백질, 및 핵산 분자로 이루어진 군에서 선택된 1종 이상인, 표적입자 검출용 어레이.
- 제5항에 있어서, 상기 표적입자는 바이러스인, 표적입자 검출용 어레이.
- 제6항에 있어서, 상기 표적입자는 코로나바이러스, 인플루엔자바이러스, 또는 이들의 조합인, 표적입자 검출용 어레이.
- 제1항 내지 제4항 중 어느 한 항의 표적입자 검출용 어레이를 2개 이상 포함하는, 표적입자 검출용 키트.
- 제8항에 있어서, 상기 2개 이상의 표적입자 검출용 어레이는 동일한 표적입자를 검출하기 위한 것인, 표적입자 검출용 키트.
- 제8항에 있어서, 상기 2개 이상의 표적입자 검출용 어레이는 상이한 표적입자를 검출하기 위한 것인, 표적입자 검출용 키트.
- 제8항에 있어서, 상기 표적입자는 바이러스인, 표적입자 검출용 키트.
- 제11항에 있어서, 상기 표적입자는 코로나바이러스, 인플루엔자바이러스, 또는 이들의 조합인, 표적입자 검출용 키트.
- 시료를 제1항 내지 제4항 중 어느 한 항의 표적입자 검출용 어레이 또는 상기 어레이를 2개 이상 포함하는 표적입자 검출용 키트와 접촉시키는 단계; 및상기 어레이 또는 키트로부터 발생하는 신호를 측정하는 단계를 포함하는, 표적입자 검출 방법.
- 제8항에 있어서, 상기 시료는 포유동물로부터 얻어진 세포, 혈액, 림프, 타액, 가래, 콧물, 소변, 및 대변으로 이루어진 군에서 선택된 1종 이상인, 표적입자 검출 방법.
- 제13항에 있어서, 상기 표적입자는 바이러스인, 표적입자 검출 방법.
- 제15항에 있어서, 상기 표적입자는 코로나바이러스, 인플루엔자바이러스, 또는 이들의 조합인, 표적입자 검출 방법.
- 제1항 내지 제4항 중 어느 한 항의 표적입자 검출용 어레이를 1개 이상 포함하고, 상기 표적입자는 바이러스인, 바이러스 감염 진단용 키트.
- 제17항에 있어서, 상기 바이러스는 인플루엔자바이러스, 코로나바이러스, 또는 이들의 조합인, 바이러스 감염 진단용 키트.
