Abstract
본 발명은 펩타이드 또는 이의 혼합물을 유효성분으로 함유하는 자가면역반응에 의해 유도되는 염증질환 예방 또는 치료용 조성물에 관한 것으로, 보다 상세하게는 P1 내지 P5 펩타이드 또는 이들의 혼합물을 자가면역반응으로 유발된 염증성 질환인 건선 및 염증성 장질환 마우스 동물모델에 복강주사하거나 피부외용제로 도포한 결과, 건선 마우스 모델의 피부 궤양과 피부 염증이 개선된 것이 확인되었으며, 장염 모델의 염증 증상이 개선됨에 따라, 상기 P1 내지 P5 펩타이드 또는 이들의 혼합물을 유효성분으로 함유하는 조성물은 자가면역반응에 의해 유발된 염증성 질환 치료제로 제공될 수 있다.
Description
본 발명은 펩타이드 또는 이의 혼합물을 유효성분으로 함유하며, 자가면역반응에 의해 건선, 염증성장질환, 크론병, 알러지성 천식, 아토피성 피부염 및 전신성염증반응 증후군으로 이루어진 군에서 선택된 질환을 예방하거나 치료하기 위한 조성물에 관한 것이다.
인간의 면역계는 인체에 침입한 외부 항원으로부터 신체를 보호하는 역할을 하나, 자기 관용성이 있어서 자기 조직을 공격하지 않는다. 그러나 면역계의 자기 관용성이 파괴되어 자신의 유전자에 의해 정상적으로 발현되는 단백질을 면역세포가 공격대상으로 인식하여 항체를 만들거나 T 세포 반응을 일으켜서 정상조직을 파괴하는 경우를 자가면역이라 부르며 구체적인 증상이 나타나면 자가면역질환이라고 한다.
염증반응은 생체 혹은 조직에 기질적 변화를 가져오는 화학적 물질, 세균 감염 및 물리적 작용에 의해 발생된 손상 부위를 재생하려는 복구기전이다. 유해한 자극, 감염 및 외상 등에 의해 염증이 발현되면 국소적으로 염증성 성분과 같은 혈관 활성 물질이 유리되어 염증을 유발하는데, 이와 같은 염증반응이 지속되면 오히려 점막손상을 촉진함에 따라 발적, 발열, 종창, 동통, 기능장애 등으로 인한 류마티스 관절염, 동맥경화증, 위염, 천식 등의 염증성질환을 유발할 수 있다.
이러한 염증반응은 사이토카인(cytokines), 프로스타글란딘E2(prostagrandins E2, PGE2), 유리라디칼(free radicals) 등 다양한 매개물질이 관여하고 있다. 특히, 대식세포에서는 사이토카인, 종양괴사인자-a(tumor necrosis factor-α, TNF-α), 리포폴리사카라이드(lipopolysaccharide, LPS)와 같은 자극에 의해 염증반응의 전사인자인 NF-κB(nclear factor-κB)를 활성화시키며, 그 결과 유도성 산화질소(inducible nitiric oxide synthase, iNOS), 사이클로옥시제나아제-2(cyclooxygenase-2, COX-2)를 발현시켜 과량의 산화질소(NO)와 프로스타글라란딘 E2를 생성하여 염증을 일으키게 된다.
건선 (psoriasis)은 과증식성 각질세포 및 비늘 반의 증가로 특징지어지는 비전염성의 만성적인 염증성 피부질환으로, 아직까지 발병 원인이 명확하게 밝혀지지는 않았지만 피부에서 염증성 세포 침윤의 만성적 활성화와 표피 각질 세포의 조절 장애에 의한 면역학적 이상에 의해 발생되는 피부 질환이다. 연구 결과들에 의하여 건선은 T 림프 세포가 개입하여 발생한 자가면역 질환 중 하나라고 알려져 있으며, T 림프 세포의 과도한 활성화가 건선 발병의 주요 원인이 되는 것으로 보고되어 있다.
염증성 장질환(inflammatory bowel disease)은 장에 만성적인 원인 불명의 염증을 일으키는 질환으로 궤양성 대장염과 크론병으로 나눌 수 있다. 궤양성 대장염 및 크론병 모두 일시적으로 증상이 좋아지다가 재발이 반복되는 만성 난치성 질환이다. 염증성 장 질환의 발생 원인이나 병태생리에 대해서는 아직까지 명확히 알려져 있지 않지만, 유전적 요인, 장내 세균이나 음식물 등의 환경적 요인, 면역학적 요인 등이 복합적으로 발생기전에 관여하리라 추정되고 있다. 궤양성 대장염 및 크론병의 발생율이 급증하고 있음에도 불구하고 원인이 불분명한 이유 등으로 근본적 치료법은 아직 확립되어 있지 않아 근본적 치료가 아닌 증상의 진행을 지연 및 완화시키는 약제가 사용되고 있는 실정이다.
펩타이드 또는 이들의 혼합물을 유효성분으로 함유하는 조성물을 제공하여 자가면역반응에 의해 유도되는 건선, 염증성장질환, 크론병, 알러지성 천식, 아토피성 피부염 및 전신성염증반응 증후군으로 이루어진 군에서 선택된 질환을 예방 또는 치료하기 위한 약학조성물을 제공하고자 한다.
본 발명은 서열번호 1로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 2로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 3으로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 4로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 5로 표시되는 아미노산으로 이루어진 펩타이드 또는 이들의 혼합물을 유효성분으로 함유하며, 건선, 염증성장질환, 크론병, 알러지성 천식, 아토피성 피부염 및 전신성염증반응 증후군으로 이루어진 군에서 선택된 질환 예방 또는 치료용 약학조성물을 제공한다.
또한, 본 발명은 서열번호 1로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 2로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 3으로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 4로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 5로 표시되는 아미노산으로 이루어진 펩타이드 또는 이들의 혼합물을 유효성분으로 함유하며, 건선, 염증성장질환, 크론병, 알러지성 천식, 아토피성 피부염 및 전신성염증반응 증후군으로 이루어진 군에서 선택된 질환 예방 또는 개선용 건강식품을 제공한다.
본 발명에 따르면, P1 내지 P5 펩타이드 또는 이들의 혼합물을 자가면역반응으로 유발된 염증성 질환인 건선 및 염증성 장질환 마우스 동물모델에 복강주사하거나 피부외용제로 도포한 결과, 건선 마우스 모델의 피부 궤양과 피부 염증이 개선된 것이 확인되었으며, 장염 모델의 염증 증상이 개선됨에 따라, 상기 P1 내지 P5 펩타이드 또는 이들의 혼합물을 유효성분으로 함유하는 조성물은 자가면역반응에 의해 유발된 질환의 치료제 또는 건강식품으로 제공될 수 있다.
도 1은 건선 마우스 동물모델에 펩타이드 혼합물 투여 후 말초혈액세포에서 수지상 세포 활성화 마커인 CD40+ 세포와 CD83+ 세포의 빈도를 확인한 FACS 분석 결과이다.
도 2는 건선 마우스 동물모델에 펩타이드 혼합물 투여 후 말초혈액세포에서 RORγt 발현 세포 빈도를 확인한 FACS 분석 결과이다.
도 3은 건선 마우스 동물모델에 펩타이드 혼합물 투여 후 말초혈액세포에서 조절 T 세포 (regulatory T cell)의 빈도를 확인한 FACS 분석 결과이다.
도 4는 건선 마우스 동물모델에 펩타이드 혼합물 투여 후 복강 대식세포에서 Ly6G를 발현하는 호중구 세포 (Neutrophil)의 빈도를 확인한 FACS 분석 결과이다.
도 5는 건선 마우스 동물모델에 펩타이드 혼합물 투여 후 림프절에서 분리한 림프구(lymphocytes)에서 CD8+NK1.1+ 세포의 빈도를 확인한 FACS 분석 결과이다.
도 6은 건선 마우스 동물모델의 피부에 대조군 (Control), Control cream 및 펩타이드 P124를 피부외용제로 5일간 투여한 후 염증 증상을 확인한 결과이다.
도 7은 Dextran sulfate sodium (DSS)으로 유도된 장염 마우스 동물모델에 펩타이드 혼합물을 경구 투여 후 비장 크기 변화를 확인한 결과이다.
도 8은 장염 마우스 동물모델에 펩타이드 혼합물을 경구 투여 후 말초혈액세포에서 수지상 세포 활성화 마커인 CD40+ 세포와 CD83+ 세포의 빈도와 복강내 대식세포에서 CD83+ 세포 및 CD86+ 세포의 빈도를 확인한 FACS 분석 결과이다.
도 9는 장염 마우스 동물모델에 펩타이드 혼합물을 경구 투여 후 복강 내 대식세포에서 대식세포 마커인 CD11b+ 세포의 빈도, 비장세포에서 자연살해세포 (natural killer cell) 마커인 NK1.1+ 세포의 빈도, 호중구 마커인 Ly6G+를 발현하는 세포의 빈도 및 Th17+ 세포 분화에 관여하는 유전자인 ROR gammaT 발현세포의 빈도를 확인한 FACS 분석 결과이다.
도 10은 펩타이드 1, 2 및 4를 각각 다른 형광을 결합(conjugation)한 후 정상 마우스에 경구로 투여하고, 48 및 72 시간 경과 후 복강 내 대식세포에서 형광 표지 세포의 빈도를 확인한 결과이다.
이하, 본 발명을 보다 상세하게 설명한다.
본 발명은 서열번호 1로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 2로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 3으로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 4로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 5로 표시되는 아미노산으로 이루어진 펩타이드 또는 이들의 혼합물을 유효성분으로 함유하며, 건선, 염증성장질환, 크론병, 알러지성 천식, 아토피성 피부염 및 전신성염증반응 증후군으로 이루어진 군에서 선택된 질환 예방 또는 치료용 약학조성물을 제공할 수 있다.
상기 펩타이드는 자가면역반응에 의해 유도되는 염증을 예방하거나 감소시키는 것일 수 있다.
상기 약학조성물은 경구제, 외용제, 좌제 및 주사제로 이루어진 군에서 선택되는 제형일 수 있으며, 이에 제한되지 않는다.
본 발명의 한 구체예에서, 약학조성물은 약학조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제, 붕해제, 감미제, 피복제, 팽창제, 활택제, 향미제, 항산화제, 완충액, 정균제, 희석제, 분산제, 계면활성제, 결합제 및 윤활제로 이루어진 군에서 선택되는 하나 이상의 첨가제를 추가로 포함할 수 있다.
구체적으로 담체, 부형제 및 희석제는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 사용할 수 있으며, 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 조성물에 적어도 하나 이상의 부형제, 예를 들면, 전분, 칼슘카보네이트, 수크로스 또는 락토오스, 젤라틴 등을 섞어 조제할 수 있다. 또한 단순한 부형제 이외에 마그네슘 스티레이트, 탈크 같은 윤활제들도 사용할 수 있다. 경구를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 있으며 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제 등이 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기재로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 일실시예에 따르면 상기 약학 조성물은 정맥내, 동맥내, 복강내, 근육내, 흉골내, 경피, 비측내, 흡입, 국소, 직장, 경구, 안구내 또는 피내 경로를 통해 통상적인 방식으로 대상체로 투여할 수 있다.
상기 펩타이드의 바람직한 투여량은 대상체의 상태 및 체중, 질환의 종류 및 정도, 약물 형태, 투여경로 및 기간에 따라 달라질 수 있으며 당업자에 의해 적절하게 선택될 수 있다. 본 발명의 일실시예에 따르면 이에 제한되는 것은 아니지만 1일 투여량이 0.01 μg/kg 내지 200 mg/kg, 구체적으로는 0.1 내지 200 mg/kg, 보다 구체적으로는 0.1 내지 100 mg/kg 일 수 있다. 투여는 하루에 한 번 투여할 수도 있고 수회로 나누어 투여할 수도 있으며, 이에 의해 본 발명의 범위가 제한되는 것은 아니다.
본 발명에 있어서, 상기 '대상체'는 인간을 포함하는 포유동물일 수 있으나, 이들 예에 한정되는 것은 아니다.
본 발명은 서열번호 1로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 2로 표시되는 아미노산 서열로 이루어진 펩타이드, 서열번호 3으로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 4로 표시되는 아미노산으로 이루어진 펩타이드, 서열번호 5로 표시되는 아미노산으로 이루어진 펩타이드 또는 이들의 혼합물을 유효성분으로 함유하며, 건선, 염증성장질환, 크론병, 알러지성 천식, 아토피성 피부염 및 전신성염증반응 증후군으로 이루어진 군에서 선택된 질환 예방 또는 개선용 건강식품을 제공할 수 있다.
상기 건강식품은 상기 펩타이드 또는 이들의 혼합물 이외에 다른 식품 또는 식품 첨가물과 함께 사용되고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분의 혼합양은 그의 사용 목적 예를 들어 예방, 건강 또는 치료적 처치에 따라 적합하게 결정될 수 있다.
상기 건강식품에 함유된 화합물의 유효용량은 상기 치료제의 유효용량에 준해서 사용할 수 있으나, 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 범위 이하일 수 있으며, 유효성분은 안전성 면에서 아무런 문제가 없기 때문에 상기 범위 이상의 양으로도 사용될 수 있음은 확실하다.
상기 건강식품의 종류에는 특별한 제한이 없고, 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제등을 들 수 있다.
이하, 본 발명의 이해를 돕기 위하여 실시예를 들어 상세하게 설명하기로 한다. 다만 하기의 실시예는 본 발명의 내용을 예시하는 것일 뿐 본 발명의 범위가 하기 실시예에 한정되는 것은 아니다. 본 발명의 실시예는 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해 제공되는 것이다.
<실시예 1> 펩타이드 준비
뱀의 독, 특히 단백질로 알려진 물질으로부터 염증의 치료를 위한 저분자 물질을 개발하기 위하여 예의 노력한 결과 약효를 가지는 펩타이드를 발견하기에 이르렀다. 하기의 11 내지 23개 아미노산 서열을 가지는 펩타이드를 준비하였다.

<실시예 2> 건선 마우스 모델에서 펩타이드의 효과 확인
C56BL/6J 마우스에 이미퀴모드 (Imiquimod, IMQ) 20 μg/day/mouse를 6 일간 매일 1회 피부 외용제로 투여하여 건선 증상을 유발시켰다.
실시예 1에서 분리된 펩타이드 P1, P2 및 P4를 각각 1 μg이 되도록 주사용 생리식염수에 혼합한 펩타이드 혼합물 (P124)과 대조군 (control) 크림을 피부외용제로 5 일간 매일 투여한 후 면역세포 특징을 확인하였다.
펩타이드 투여 후 말초혈액세포를 FACS (flow cytometry)로 분석한 결과, 도 1과 같이 수지상세포 활성화 마커인 CD40 positive (+) 세포와 CD83+ 세포의 빈도가 감소한 것을 확인할 수 있었으며, 도 2와 같이 RORγt를 발현하는 세포의 빈도 감소가 확인되었다. RORγt는 Th17 세포의 마커로 Th17 세포의 증가는 건선의 악화와 연관성이 있다.
또한, 도 3과 같이 펩타이드 투여군에서 염증반응을 억제하는 기능이 있는 조절 T 세포 (regulatory T cell)의 빈도가 증가하는 것이 확인되었다.
펩타이드 투여 후 복강 대식세포를 FACS (flow cytometry)로 분석한 결과, 도 4와 같이 Ly6G를 발현하는 호중구 세포 (Neutrophil)의 빈도가 감소하는 것을 확인할 수 있었다. 과도한 호중구의 발현은 싸이토카인을 과도하게 분비하여 과도한 염증 반응을 일으키는 것으로 알려져 있다.
펩타이드 투여 후 림프절에서 분리한 림프구(lymphocytes)를 FACS (flow cytometry)로 분석한 결과, 도 5와 같이 CD8+NK1.1+ 세포의 빈도 감소가 확인되었다. CD8+NK1.1+ 세포는 IFNγ를 분비하고 미생물 병원체를 제어하는 innate immune 반응 역할을 하지만, 과도한 발현은 염증의 유발과 관련성이 있는 것으로 알려져 있다.
한편, 건선 증상이 유발된 마우스 동물 모델에 대조군 (control), 대조군 크림 (Control cream) 및 펩타이드 혼합물 (P124)를 피부외용제로 5 일간 매일 투여 후 증상의 변화를 비교한 결과, 도 6과 같이 대조군과 대조군 크림 그룹에서는 6일 경과 후에 피부의 염증 증상이 확인되었으나 펩타이드 처리군에서는 건선 증상이 거의 남아 있지 않은 것을 확인할 수 있다.
<실시예 3> 염증성 장질환 동물모델에서 펩타이드의 효과 확인
Dextran sulfate sodium (DSS)로 장염을 유발시킨 마우스 동물모델에 실시예 1에서 분리된 펩타이드 P1, P2 및 P4를 각각 1 μg이 되도록 주사용 생리식염수에 혼합한 펩타이드 혼합물 (P124)을 1일 1회, 4일간 연속으로 경구 투여하였다.
펩타이드 혼합물 (P124)을 경구 투여한 마우스의 비장을 적출하여 확인한 결과, 도 7과 같이 비장의 크기가 감소한 것이 확인되었다.
또한, 펩타이드 혼합물이 투여된 장염 마우스 모델의 면역세포를 확인한 결과, 도 8과 같이 말초혈액세포에서 CD40+ 수지상세포와 CD86+ 수지상세포의 빈도가 감소하였으며, 복강 내 대식세포에서도 펩타이드 투여로 CD83+ 세포 및 CD86+ 세포의 빈도가 감소한 것을 확인할 수 있었다.
수지상세포 활성화마커의 과발현은 만성염증 지속과 관련이 있는 것으로 알려져 있으며, 상기 결과에서 펩타이드 투여는 수지상세포 활성화마커를 감소시키는 것으로 확인됨에 따라, 상기 펩타이드 투여에 의해 염증성 장 질환의 증상이 호전되는 것이 확인되었다.
또한, 도 9와 같이 장염 모델 마우스에 펩타이드 투여 후 복강 내 대식세포에서 대식세포 마커인 CD11b+ 세포의 빈도 감소가 확인되었으며, 비장세포에서도 CD11b+ 세포의 빈도 감소가 나타났다. 비장세포에서는 자연살해세포 (natural killer cell) 마커인 NK1.1+ 세포의 빈도는 증가하였고, 호중구 마커인 Ly6G+ 세포를 발현하는 세포의 빈도는 감소하였다. Th17+ 세포 분화에 관여하는 유전자인 ROR gammaT 발현세포의 빈도는 감소하였다.
상기 결과들로부터 본 발명의 펩타이드는 염증 유발 세포의 빈도를 감소시켜 대장염증상을 호전시킬 수 있음이 확인되었다.
한편, 펩타이드 1, 2 및 4를 각각 다른 형광을 결합(conjugation)한 후 정상 마우스에 경구로 투여하고, 48 및 72 시간 경과 후 복강 내 대식세포에서 형광 표지 세포의 빈도를 측정하였다.
그 결과, 도 10과 같이 P2 및 P4 펩타이드는 48 시간까지 유지되었으며, P1은 72 시간까지 유지되었다.
상기 결과로부터 경구로 투여된 펩타이드가 복강 내에 도달하여 면역반응에 관여할 수 있으며, 대장염 증상의 완화를 유도한 것으로 확인되었다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
