Description
ペプチド等は、診断・検査薬、医薬品として広く利用されており、その原料は非常に高価である。そのため、ペプチド等を含む原料液を濃縮する際には、ペプチド等を変性させず、収率高く回収することが重要である。
また、溶媒を分子レベルで透過させる膜を用いた逆浸透(RO:Reverse Osmosis)法が知られている。RO法は、原料液を、当該原料液の浸透圧より高い所定の圧力に昇圧したうえで、RO膜モジュールに供給し、原料液中の溶媒だけがRO膜を透過するように構成して、原料液中の溶媒(典型的には水)を除去することにより、原料液を濃縮する方法である(特許文献2)。
これらとは更に別の原料液の濃縮方法として、膜蒸留(MD:Membrane distillation)法が知られている。膜蒸留法の1つの方法として、原料液と、この原料液よりも温度の低い冷却水とを膜を介して接触させ、原料液から冷却水へ原料液中の溶媒蒸気が移動することにより、原料液が濃縮されるDCMD法(Direct contact MD)がよく知られている(特許文献4)。
特許文献5には、正浸透法と膜蒸留とを組み合わせた方法が記載されている。特許文献5の技術では、膜蒸留法は正浸透法で用いる誘導溶液の再生に用いられている。
特許文献2に記載のRO法では、原料液の加圧が必要なため、原料液に含まれる溶質のRO膜表面への固着が起こり、回収率が低下する課題があった。また、RO法は、濃縮された原料液中の溶媒(ろ過された溶媒)の浸透圧が、加圧に用いる高圧ポンプの圧力を超えることはないため、RO法による原料液の濃縮率には、ポンプの能力に応じた限界がある。
一方、特許文献4の膜蒸留法では、蒸気圧の低い成分が優先的に膜を通して系外に除去されるため、溶媒として水および有機溶媒の双方を含む原料液を濃縮した場合、濃縮液には水が多く残る課題があった。これに加えて、濃縮物に水が多く残ると、原料液中の有効成分が析出してしまう課題もあった。
特許文献5の技術では、膜蒸留法は正浸透に用いられる誘導溶液の再生に使用されており、原料液の濃縮には適用されていない。
本発明者らは、溶媒として水および有機溶媒を含む原料液を濃縮する際に、原料液中の主として水を除去する濃縮方法と、主として有機溶剤を除く濃縮方法とを組み合わせることで、溶媒中の水および有機溶媒の比率を任意の割合に調整できることを見出し、本発明をなすに至った。

本発明の濃縮装置を用いると、上記のような利点を有する原料液の濃縮システムを構成することができる。
本発明の原料液濃縮システムは、
溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮システムであって、
溶媒が水および有機溶媒を含有し、
濃縮システムが、
正浸透法によって原料液中の主として水を除く第1の濃縮手段と、
原料液中の水および有機溶媒を蒸発させて除く第2の濃縮手段と
を組み合わせて行う、原料液濃縮システムである。
一方、第2の蒸発手段は、溶媒を蒸発させて除く方法によって行われる。第2の濃縮手段では、水および有機溶媒のうちの、蒸気圧が大きい側が優先的に除かれるため、一般的には有機溶媒が除かれる割合の方が高い。
したがって、これらを組み合わせて行う本発明の濃縮システムは、単に濃縮を行うだけでなく、濃縮される原料液中の溶媒における水および有機溶媒の比率を任意に制御しながら濃縮を行うことができる。
本発明の原料液濃縮システムにおける第1の濃縮手段は、正浸透膜を用いた正浸透法である。
図1は、正浸透膜を用いた濃縮方法の作用機構を示す模式図である。
図1において、正浸透膜(120)は、膜の片側では原料液(a)と接し、反対側では原料液(a)よりも浸透圧が高い誘導溶液(d)と接している。図1の正浸透膜(120)は、基材層(121)と、この基材層(121)の片面に形成された活性層(122)とを有する。活性層(122)は、主として水が透過でき、溶質が透過できない、非常に緻密な構造を有しており、基材層(121)のうちの原料液(a)に接する側の面上に形成されている。
正浸透膜の形態は、例えば、中空糸膜状、チューブラー状、および平膜状の、いずれの構造でも構わない。中空糸膜状の正浸透膜は、原料液および誘導溶液が通る流路を、スペーサー等を用いずに形成することができ、均一な濃縮を行えることから、適している。
限外濾過膜の材質としては、例えば、ポリエーテルスルホン、ポリスルホン、ポリケトン、ポリエーテルエーテルケトン、ポリフェニレンエーテル、ポリフッ化ビニリデン、ポリアクリロニトリル、ポリイミン、ポリイミド、ポリベンゾオキサゾール、ポリベンゾイミダゾール、スルフォン化テトラフルオロエチレン、およびポリアミドから成る群から選ばれる少なくとも1種を主成分とする薄膜層を有することが好ましい。
多官能性芳香族酸ハライドとは、一分子中に2個以上の酸ハライド基を有する芳香族酸ハライド化合物である。具体的には、例えば、トリメシン酸ハライド、トリメリット酸ハライド、イソフタル酸ハライド、テレフタル酸ハライド、ピロメリット酸ハライド、ベンゾフェノンテトラカルボン酸ハライド、ビフェニルジカルボン酸ハライド、ナフタレンジカルボン酸ハライド、ピリジンジカルボン酸ハライド、ベンゼンジスルホン酸ハライド等を挙げることができ、これらを単独で、またはこれらの混合物を用いることができる。これらの芳香族酸ハライド化合物におけるハロゲン化物イオンとしては、例えば、塩化物イオン、臭化物イオン、ヨウ化物イオン等を挙げることができる。本発明においては、特にトリメシン酸クロリド単独、またはトリメシン酸クロリドとイソフタル酸クロリドとの混合物、若しくはトリメシン酸クロリドとテレフタル酸クロリドとの混合物が好ましく用いられる。
多官能性酸ハライドおよび多官能性芳香族アミンの界面重合は、定法に従って実施することができる。
誘導溶液の溶媒は、典型的には水である。
アルコールを誘導溶液の溶質として用いるとき、当該誘導溶液の溶媒は好ましくは水であり、この場合の特に好ましい誘導溶液は、イソプロパノールの水溶液である。
アルコールを誘導溶液の溶媒として用いるとき、当該誘導溶液の溶質は、好ましくは、ヒドロキシカルボン酸、常温で固体の有機物等である。
なお、図1では、原料液(a)と、誘導溶液(d)とは対向流であるが、並行流でもよい。
図3の正浸透膜モジュール(100)では、ハウジング(110)内に、複数の中空糸状の正浸透膜(120)が収納され、正浸透膜(120)の両端は、接着樹脂(130)により、ハウジング(110)に接着固定されている。ハウジング(110)の側面には、2本のハウジング側管がある。これらハウジング側管のうちの片方は誘導溶液入口(111)であり、他方は誘導溶液出口(112)である。
ハウジング(110)内は、正浸透膜(120)の外壁および接着樹脂(130)により、原料液(a)が流れる空間と、誘導溶液(d)が流れる空間とに、2分割されており、両空間は、正浸透膜(120)の内壁を介して溶媒が行き来できる他は、流体的に遮断されている。
同様に、ハウジング(110)の側管のうちの誘導溶液入口(111)から誘導溶液(d)を流すと、誘導溶液(d)は、中空糸状の正浸透膜(120)の外側空間を流れて行き、誘導溶液出口(112)から流出する。
これらにより、原料液(a)と誘導溶液(d)とは、正浸透膜(120)を介して接することができる。このとき、原料液(a)から誘導溶液(d)に、両液の浸透圧の違いによって溶媒(主として水)が移動する。正浸透膜法では、このような機構により、濃縮が行われる。
原料液(a)の流量、および/または誘導溶液(d)の流量が大きい場合、正浸透膜(120)の、特に活性層における浸透圧差がより大きくなるため、正浸透膜の膜面積当たりの液体の透過量は大きくなる。
原料液(a)の流れる方向と誘導溶液(d)の流れる方向とは、図3の正浸透膜モジュール(100)では、対向流であるが、並行流でもよい。
図3における接着樹脂(130)としては、機械的強度が良好で、かつ100℃における耐熱性を有することが望まれる。接着樹脂(130)として使用できる樹脂としては、例えば、熱硬化性のエポキシ樹脂、熱硬化性のウレタン樹脂等が挙げられる。耐熱性の観点ではエポキシ樹脂が好ましいが、ハンドリング性の観点ではウレタン樹脂が好ましい。
ハウジング(110)に正浸透膜(120)を接着固定する方法は、中空糸膜モジュール作製に関する既知の接着方法に従えばよい。
本発明の原料液濃縮システムにおける第1の濃縮手段は、原料液中の水および有機溶媒を蒸発させて除く濃縮手段である。
このような濃縮手段としては、例えば、エバポレーター、薄膜蒸留装置、原料液の入った容器内に気体を流して、容器外に溶媒を移動させる方法、膜蒸留法等が挙げられる。これらのうち、膜蒸留法は、原料液から除去される水および有機溶媒の比率を調整し易いため、特に有効な濃縮手段である。
図2は、膜蒸留法を用いた濃縮方法の作用機構を示す模式図である。
図2において、膜蒸留用膜(220)は、膜の片側では原料液(a)と接し、反対側では原料液(a)よりも温度が低い冷却水(CW)と接触している。膜蒸留法では、原料液(a)の方が温度が高く、したがって溶媒(b)の蒸気圧が高い状態にあるため、原料液(a)中の溶媒(b)が蒸発して蒸気となって、膜蒸留用膜(220)を介して、冷却水(CW)側に移動して液化することにより、濃縮が起こる。
膜蒸留法では、原料液(a)の溶媒(b)のうちの、水および有機溶媒の双方が蒸発して除かれるが、これらのうちの、蒸気圧が大きい側が優先的に除かれる。水と有機溶媒とでは、一般的には有機溶媒の蒸気圧の方が高い場合が多く、したがって有機溶媒が除かれる割合の方が高い。
膜蒸留法の別の形態として、原料液(a)と冷却水(CW)とを膜を介して接触させる代わりに、原料液(a)と膜を介した反対側を減圧状態とすることによって蒸気を取り出す、VMD(Vacuum MD)法を適用することもできる。
疎水性を表す方法として、水接触角が用いられる。この方法では、膜の表面に置いた水滴と膜との接触角により、疎水性を評価する。本発明の第2の濃縮手段として用いる膜蒸留法用の膜としては、水接触角は90°以上が好ましく、より好ましくは110°、更に好ましくは120°以上である。水接触角の上限はないが、現実的には上限が150°程度である。
膜蒸留用膜の空隙率は、高い蒸気透過性を得る観点から、好ましくは60%以上、より好ましくは70%以上であり、膜自体の強度が良好に維持され、長期使用の際に破断等の問題が発生し難くする点で、好ましくは90%以下、より好ましくは85%以下である。
中空糸膜状の膜蒸留用膜を用いる場合には、蒸気の透過性と膜の機械的強度との両立の観点から、膜厚が、10μm以上1,000μm以下であることが好ましく、20μm以上500μm以下であることがより好ましい。膜厚が1,000μm以下であれば、高い蒸気透過性を得ることができ、他方、膜厚が10μm以上であれば、膜が変形することなく使用することができる。
中空糸膜状の膜蒸留用膜の外径は、300μm以上5,000μm以下が好ましく、より好ましくは350μm以上4,000μm以下であり、内径は、200μm以上4,000μm以下が好ましく、より好ましくは250μm以上3,000μm以下である。この範囲のサイズの中空糸膜とすると、膜強度と膜の有効面積とのバランスが良好となる。
膜蒸留用膜の疎水性をより向上するために、膜蒸留用膜の少なくとも一部に、疎水性ポリマーが付着したものも使用することができる。疎水性ポリマーは、例えば、疎水性構造を有するポリマーであってよい。疎水性構造としては、非極性基または低極性基、非極性骨格または低極性骨格等が挙げられる。非極性基または低極性基としては、例えば、炭化水素基、含フッ素基等が挙げられ、非極性骨格または低極性骨格としては、例えば、炭化水素主鎖、シロキサン主鎖等が挙げられる。
(ア)シロキサン結合を有するポリマーとして、例えば、ジメチルシリコーンゲル、メチルフェニルシリコーンゲル、有機官能基(アミノ基、フルオロアルキル基等)を有する反応性変性シリコーンゲル、シランカップリング剤と反応して架橋構造を形成するシリコーン系ポリマー等、およびこれらの架橋体であるポリマーゲル
(イ)フッ素原子含有ポリマーとして、側鎖にフッ素原子含有基を持つポリマー、ここで、フッ素原子含有基は、(パー)フルオロアルキル基、(パー)フルオロポリエーテル基、アルキルシリル基、フルオロシリル基等である。
特に、疎水性ポリマーが、炭素数1~12の(パー)フルオロアルキル基および/または(パー)フルオロポリエーテル基を有する、(メタ)アクリレート系モノマーおよび/またはビニル系モノマーの重合体であることが好ましい。
図4の膜蒸留用膜モジュール(500)では、ハウジング(210)内に、複数の中空糸状の膜蒸留用膜(220)が収納され、膜蒸留用膜(220)の両端は、接着樹脂(230)により、ハウジング(210)に接着固定されている。ハウジング(210)の側面には、2本のハウジング側管がある。これらハウジング側管のうちの片方は冷却水入口(211)であり、他方は冷却水出口(212)である。
ハウジング(210)内は、膜蒸留用膜(220)の外壁および接着樹脂(230)により、原料液(a)が流れる空間と、冷却水(CW)が流れる空間とに、2分割されており、両空間は、膜蒸留用膜(220)の外壁を介して溶媒の蒸気が行き来できる他は、流体的に遮断されている。
同様に、ハウジング(210)の側管のうちの冷却水入口(211)から冷却水(CW)を流すと、冷却水(CW)は、中空糸状の膜蒸留用膜の外側空間を流れて行き、冷却水出口(212)から流出する。
これらにより、原料液(a)と冷却水(CW)とは、膜蒸留用膜(220)を介して接することができる。このとき、原料液(a)から冷却水(CW)に、両液の溶媒の蒸気圧の違いによって溶媒の蒸気が移動して、冷却水(CW)中で液化することにより、原料液(a)の濃縮が行われる。
膜蒸留法では、原料液(a)中の溶媒(b)のうちの、水および有機溶媒の双方が蒸発して、蒸気として膜蒸留用膜(220)を通過して冷却水(CW)に移動する。このとき、移動する水と有機溶媒との比率が、両者の蒸気圧の大小によって定まり、通常は有機溶媒が優先的に移動することは上述した。
原量液(a)の流れる方向と冷却水(CW)の流れる方向とは、図4の膜蒸留用膜モジュール(500)では対向流であるが、並行流でもよい。
原料液(a)の温度、および冷却水(CW)の温度の設定に、特に制限はないが、原料液(a)の溶質の組成が変化しない範囲で設定することが好ましい。
図4における接着樹脂(230)としては、機械的強度が良好で、かつ100℃における耐熱性を有することが望まれる。接着樹脂(230)として使用できる樹脂としては、例えば、熱硬化性のエポキシ樹脂、熱硬化性のウレタン樹脂等が挙げられる。耐熱性の観点ではエポキシ樹脂が好ましいが、ハンドリング性の観点ではウレタン樹脂が好ましい。
ハウジング(210)に膜蒸留用膜(220)を接着固定する方法は、中空糸膜モジュール作製に関する既知の接着方法に従えばよい。
図5~図10に、本発明の濃縮システムの例を示した。
図5の濃縮システムは、第2の濃縮手段として、原料液の入った容器内に気体を流して、容器外に溶媒を移動させる方法を適用した例である。
図5のシステムは、正浸透膜モジュール(100)と、原料液タンク(200)と、誘導溶液タンク(300)と、トラップ(400)とを有し、原料液タンク(200)には、送風機(BL)が設置されている。
図5のシステムでは、原料液(a)を満たした原料液タンク(200)の下部より、ポンプ(P)により、正浸透膜モジュール(100)の一方の空間(例えば、中空糸状正浸透膜の側の空間)に、適切な流量で原料液(a)を送る。この際に、流量計等(図示せず)により原料液(a)の流量を、手動または自動で調整する。一方、誘導溶液(d)を満たした誘導溶液タンク(300)から、ポンプ(P)により、正浸透膜モジュール(100)の他方の空間(例えば、中空糸状正浸透膜の外側空間)に、誘導溶液(d)が供給される。そして、原料液(a)と誘導溶液(d)との浸透圧の違いにより、原料液(a)から溶媒(b)の一部(主として水)が誘導溶液(d)側に移動して濃縮が起こり、濃縮された原料液(a)は、正浸透膜モジュール(100)より出て行き、原料液タンク(200)に戻って循環する。
正浸透膜モジュール(100)による濃縮では、分子径の小さい水が主として誘導溶液(d)側に移動するため、濃縮後の原料液(a)における溶媒(b)の組成は、有機溶媒の比率が高くなるのが一般的である。
原料液タンク(200)から排出された溶媒蒸気は、トラップ(400)によって、再度液化して回収することが可能である。
原料液タンク(200)内への送風方法によると、原料液(a)の液面で濃縮が起こるため、必要に応じて原料液タンク(200)に攪拌機能を設けることにより、組成が安定した濃縮を行うことができる。
第2の濃縮手段による濃縮では、水および有機溶媒の双方が除去されるが、蒸気圧の大きい有機溶媒が優先的に除かれるため、濃縮後の原料液(a)における溶媒(b)の組成は、水の比率が高くなるのが一般的である。
図6のシステムは、正浸透膜モジュール(100)と、原料液タンク(200)と、誘導溶液タンク(300)と、膜蒸留用膜モジュール(500)と、冷却水タンク(700)とを有する。
図6の濃縮システムでは、原料液タンク(200)に、正浸透膜モジュール(100)と膜蒸留用膜モジュール(500)とが、並列に接続されている。
正浸透膜モジュール(100)による濃縮は、図5の濃縮システムと同じ方法で行う。
膜蒸留用膜モジュール(500)による濃縮では、原料液タンク(200)の底部から、膜蒸留用膜モジュール(500)の一方の空間(例えば、中空糸状膜蒸留用膜の内側の空間)に、ポンプ(P)を用いて原料液(a)を適切な流量で供給する。原料液(a)は、中空糸膜の内を通過した後に、原料液タンク(200)に戻され循環する。この戻り位置は、原料液タンク(200)の底部であって、膜蒸留用膜モジュール(500)に供給する位置から遠い位置にすることが、原料液(a)が過度の濃縮されることを抑制し、濃縮された原料液(a)中の溶質を、原料液タンク(200)の壁面等で失うことを防ぐ点で好ましい。
一方、膜蒸留用膜モジュール(500)の他方の空間(例えば、中空糸状膜蒸留用膜の外側空間)には、原料液(a)よりも低温の冷却水(CW)を導入する。
冷却水(CW)は、原料液(a)からの溶媒が混入することにより、容量が増えるため、冷却水タンク(700)は、十分に大きくするか、一部をオーバーフロー等でタンク外に排出する等の対応が必要である。図6では、これらの機構は図示していない。
膜蒸留用膜モジュール(500)による濃縮では、蒸気圧の高い有機溶媒が除去され易く、したがって濃縮された原料液(a)における溶媒(b)の組成は、水の比率が高くなるのが一般的である。
図7の濃縮システムでは、原料液(a)を第1の濃縮手段としての正浸透膜モジュール(100)で濃縮し、正浸透膜モジュール(100)によって濃縮された原料液(a)は、製品タンク(600)に移送される。そして、製品タンク(600)から、タンクの底部よりポンプ(P)にて、第1の濃縮手段によって濃縮された後の原料液(a)が、膜蒸留用膜モジュール(500)の一方の空間に適切な流量で供給され、膜蒸留法による濃縮が行われる。膜蒸留用膜モジュール(500)によって濃縮された後の原料液(a)は、製品タンク(600)に戻され循環する。
正浸透膜モジュール(100)から製品タンク(600)に移送される濃縮後の原料液(a)は、溶媒(b)中の有機溶媒比率が高くなった状態である。しかし、膜蒸留用膜モジュール(500)を通過して膜蒸留法によって濃縮されると、溶媒(b)中の水の比率が回復する。
図7のシステムでは、製品タンク(600)に比重計(HM)および液面計(LG)が設置されている。比重計(HM)により、所望の水/有機溶媒比率の比重と比較することができる。液面計(LG)により、目標濃縮倍率まで濃縮が行えたか否かが確認できる。
図8の濃縮システムでは、原料液(a)を、先ず膜蒸留用膜モジュール(500)に送り、膜蒸留法によって濃縮する。濃縮後の原料液(a)は、製品タンク(600)に移送される。そして、濃縮後の原料液(a)は、製品タンク(600)から正浸透膜モジュール(100)に送られ、正浸透法による濃縮後、製品タンク(600)に戻され循環する。
図8の濃縮システムにおける膜蒸留用膜モジュール(500)および正浸透膜モジュール(100)による濃縮は、それぞれ、図7の濃縮システムで示したものとほぼ同様であるが、原料液(a)は、膜蒸留用膜モジュール(500)を1回通過するのみであり、正浸透膜モジュール(100)には濃縮は循環して行われる点が、図7のシステムと異なる。
膜蒸留法による濃縮では、膜蒸留用膜モジュール(500)を1回通過しただけでは十分な濃縮が行えない場合には、膜蒸留用膜モジュール(500)を出た原料液(a)の一部を製品タンク(600))に送り、残りの一部を、原料液タンク(200)に戻して、循環して濃縮を行う態様にしてもよい。
図5~図8のシステムにおいては、比重計(HM)にて原料液(a)の比重を追跡することができ、これにより、原料液(a)の溶媒中の水および有機溶媒の比率をモニターすることができる。そして、必要に応じて、各濃縮方法の操作を停止または減速させて、溶媒(b)中の水および有機溶媒の比率を目標値に近づけながら、濃縮を行うことが可能である。
最終的には、液面計(LG)で原料液(a)の容積を測定し、目標濃縮倍率になった時点で濃縮を完了することができる。
原料液(a)の性状をモニターする項目としては、例示した比重の他に、pH、導電率等を用いることができ、これらと、液面計測定値、液の重量等を同時にモニターすることにより、原料液(a)の溶媒(b)中の有機溶媒および水の比率を調整しながら濃縮を行うことができる。
このフィルターは、例えば、原料液タンク中、正浸透膜モジュールの出口、膜蒸留膜モジュールの出口、配管系中等の、任意の位置に配置することができる。
フィルターの材質は、例えば、紙、樹脂、ガラス、無機物等、特に限定されるものではなく、原料液に対して化学的及び物理的に安定なものから適宜に選択して使用してよい。フィルターの目開きは、析出物の大きさに応じて、適宜設定されてよい。目詰まり防止の観点からは、例えば、目開きが1μm以上のフィルターが好ましい。
図9では、システムの構成要素として、フィルター(201)の設置態様を示すため、フィルター(201)とともに、正浸透膜モジュール(100)、原料液タンク(200)、および誘導溶液タンク(300)、ならびにこれらの近傍の配管系等が示されているのみであるが、これらに、例えば、送風機、膜蒸留用膜モジュール等の適宜の第2の濃縮システムを接続して、本発明の濃縮システムとすることができる。
原料液(a)を貯蔵して濃縮する原料液タンク(200)が、フィルター(201)および原料液ろ過用配管を有し、
フィルター(201)は、原料液(a)の液面よりも上部に設定されており、
原料液ろ過用配管は、原料液タンク(200)の底部に入り口を有し、かつ、フィルター(201)の上部に出口を有し、
原料液ろ過用配管によって、原料液(a)が、原料液タンク(200)の底部からフィルター(201)の上部に運搬されて、フィルター(201)を通過して、原料液タンク(200)内の原料液(a)に合流するように構成されている。
このような構成により、原料液(a)から析出物(a’)が析出した場合でも、この析出物(a’)は、フィルター(201)によって原料液(a)から除かれるから、固液分離を行いながら、原料液(a)の液体部分のみを、第1または第2の濃縮ユニットに供給して濃縮を行うことができる。
原料液(a)を移送する流量は、特に限定はされないが、第1の濃縮方法に用いる正浸透膜処理における原料液の供給流量と、同等か、それ以下とすることが好ましい。
図10では、システムの構成要素として、仕切り板(201)、第1室(202)、および第2室(203)の設置態様を示すため、正浸透膜モジュール(100)、原料液タンク(200)、および誘導溶液タンク(300)、ならびにこれらの近傍の配管系等が示されているのみであるが、これらに、例えば、送風機、膜蒸留用膜モジュール等の適宜の第2の濃縮システムを接続して、本発明の濃縮システムとすることができる。
原料液タンク(200)の内部が、仕切り板(201)によって第1室(202)と第2室(203)とに分割されており、
第1室(202)の底部から原料液(a)を抜き出して、第1の濃縮ユニットである正浸透膜モジュール(100)に供給する手段と、

第2室内の原料液(a)が、複数の入口(205a、205b、205c、205d)のうちの少なくとも1つから採取され、第1室第2室連結配管(204)を通過して、出口(206)から排出されて第1室(202)に戻るように構成されている。
本発明の濃縮方法で濃縮対象となる原料液は、溶質および溶媒を含有する溶液または分散液である限り、特に限定されるものではなく、いかなる溶質および溶液を用いてもよい。しかしながら、正浸透膜法および膜蒸留法によると、ともに例えば50℃以上に加熱することを要さずに濃縮可能であることから、熱をかけずに濃縮することが望ましい原料溶液への適用が有効である。
適用される原料液としては、例えば、食品;医薬品;海水;ガス田、油田等から排出される随伴水等を挙げることができる。しかしながら、加熱を要さずに濃縮可能であるとの本発明の利点を考慮すると、溶質として、加熱により分解が懸念される有価物、特に、医薬品原料、医薬品原体、および医薬品中間体(以下、これらを総称して「医薬品原料等」という。)を含む溶液または分散液への適用が有効である。
必須アミノ酸としては、例えば、トリプトファン、リジン、メチオニン、フェニルアラニン、トレオニン、バリン、ロイシン、イソロイシン等があげられる。非必須アミノ酸としては、例えば、アルギニン、グリシン、アラニン、セリン、チロシン、システイン、アスパラギン、グルタミン、プロリン、アスパラギン酸、グルタミン酸等が挙げられる。
医薬品原料等の溶質として好ましい非天然アミノ酸として、例えば、標識アミノ酸、機能化アミノ酸等が挙げられる。
標識アミノ酸は、アミノ酸骨格と標識化合物とが結合した非天然アミノ酸であり、その具体例としては、例えば、側鎖に芳香環を含むアミノ酸骨格に、標識化合物が結合したアミノ酸等が挙げられる。
機能化アミノ酸としては、例えば、光応答性アミノ酸、光スイッチアミノ酸、蛍光プローブアミノ酸、蛍光標識アミノ酸等が挙げられる。
タンパク質は、一般的には、アミノ酸残基が結合した化合物のうちのペプチドよりも長鎖のものを指す。本明細書におけるタンパク質としては、例えば、タンパク製剤として適用されるものが好ましい。
タンパク製剤としては、例えば、インターフェロンα、インターフェロンβ、インターロイキン1~12、成長ホルモン、エリスロポエチン、インスリン、顆粒状コロニー刺激因子(G-CSF)、組織プラスミノーゲン活性化因子(TPA)、ナトリウム利尿ペプチド、血液凝固第VIII因子、ソマトメジン、グルカゴン、成長ホルモン放出因子、血清アルブミン、カルシトニン等が挙げられる。
単糖類としては、例えば、グルコース、フルクトース、ガラクトース、マンノース、リボース、デオキシリボース等が挙げられる。二糖類としては、例えば、マルトース、スクロース、ラクトース等が挙げられる。
本明細書における糖鎖とは、二糖類を除く概念であり、例えば、グルコース、ガラクトース、マンノース、フコース、キシロース、グルクロン酸、イズロン酸等が挙げられる。糖鎖誘導体としては、例えば、N-アセチルグルコサミン、N-アセチルガラクトサミン、N-アセチルノイラミン酸等の、糖類誘導体等が挙げられる。
核酸としては、例えば、オリゴヌクレオチド、RNA、アプタマー、デコイ等が;
抗生物質としては、例えば、ストレプトマイシン、バンコマイシン等が;
それぞれ挙げられる。
溶質の分子量が過度に小さいと、正浸透膜、および膜蒸留時の半透膜を透過する場合があり、分子量が過度に大きいと、膜表面への溶質の付着が起こる場合があり、好ましくない。
溶質の数平均分子量は、分子量が比較的小さい場合(例えば分子量500以下の場合)には、溶質の化学式からの計算により、求めることができる。一方、溶質の分子量が比較的大きい場合(例えば分子量500超の場合)の数平均分子量は、GPCによって測定されたポリエチレングリコール換算の数平均分子量として、求めることができる。
本発明では、溶媒として、水および有機溶媒の双方を含む原料液を、溶媒組成を維持したまま濃縮できる利点を有する。したがって、本発明の原料液の溶媒は、水および有機溶媒の双方を含有するものである。有機溶媒は、水と相溶するものであることが好ましく、例えば、アルコール、ケトン、非プロトン性極性溶媒等であってよい。
本発明における原料液の溶媒として、好ましくは、水と、アセトニトリル、イソプロパノール、アセトン、メタノール、およびエタノールから選ばれる1種以上と、の混合物であることが好ましい。
原料液に含まれる溶質が、旋光物質であれば、旋光度のオンライン測定によって、濃縮運転中に溶質濃度を知ることができる。
近赤外分光分析をオンラインで行うと、濃縮運転中に、例えば、溶質濃度、溶媒組成等の情報を知ることができる。
溶媒組成を知るために使用されるオンライン測定としては、例えば、比重測定、pH測定、導電率測定、液位測定、近赤外分光分析、重量測定等が挙げられる。
これらのオンライン測定は、それぞれ、公知の測定手段を用いて行うことができる。
オンライン測定の測定値に応じて溶媒組成を調整する方法としては、例えば、目標の溶質濃度および溶媒組成と、オンライン測定値とが、どの程度ずれているかを算出し、そのずれの程度に応じて、第1の濃縮手段および第2の濃縮手段それぞれについて、運転するか、濃縮速度を変更するか、または停止するかを決定して、目標の溶質濃度および溶媒組成に近づくように調整することが挙げられる。濃縮速度の変更方法としては、例えば、濃縮温度の変更、各モジュールへの原料液の供給速度の変更等が挙げられる。
例えば、ICP-MS(誘導結合高周波プラズマ質量分析)、核磁気共鳴分光(NMR)法、ガスクロマトグラフィー質量分析(GC/MS)法、比色法、蛍光法、高速液体クロマトグラフ(HPLC)等の各種の公知の分析方法を用いることができる。
本発明の別の観点によると、原料液の濃縮装置が提供される。
本発明の濃縮装置は、以下のいずれかであってよい。
溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮装置であって、

本発明の原料液濃縮方法は、
溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮方法であって、
前記溶媒が水および有機溶媒を含有し、
前記濃縮方法が、
正浸透法によって前記原料液中の水を除く第1の濃縮方法と、
前記原料液中の水および有機溶媒を蒸発させて除く第2の濃縮方法と
を、組み合わせた方法である。
本発明の原料液の濃縮システムについて説明した事項のうち、濃縮方法に適用可能な事項は、本発明の濃縮方法に制限なく適用することができる。
内径0.7mm、外径1.0mmのポリエーテルスルホン製の中空糸膜限外濾過膜を基材膜として、その130本を、2cm径、10cm長の円筒状プラスチックハウジングに充填し、両端部を接着剤で固定することにより、有効膜内表面積約0.02m2の中空糸支持層モジュールを作製した。
0.5L容器に、m-フェニレンジアミン10gおよびラウリル硫酸ナトリウム0.8gを入れ、更に純水489.2を加えて溶解し、界面重合に用いる第1溶液を0.5kg調製した。
別の0.5L容器に、トリメシン酸クロリド0.8gを入れ、n-ヘキサン399.2gを加えて溶解し、界面重合に用いる第2溶液0.4kgを調製した。
これらの溶液を、中空糸膜内側を通過するように、上記の中空糸支持層モジュールに通液して、中空糸の内側面上で界面重合を行い、中空糸支持層上に活性層を形成した。その後、中空糸内側を純水で洗浄を行うことにより、正浸透膜モジュールを作成した。
内径0.7mm、外径1.3mm、ASTM-F316-86に準拠して求めた平均孔径0.21μm、最大孔径0.29μm、空隙率72%のPVDF製の多孔質中空糸膜を長さ15cmに切出した。
モジュール製作は、接着樹脂として熱硬化性のエポキシ樹脂を使用し、遠心接着により中空糸膜をハウジング内に接着固定した。これにより、中空糸膜を、非接着固定部位の長さが約10cmとなり、中空糸膜の内表面の合計膜面積が約0.02m2となるように調整した。この仕様の膜モジュールを3本作製した。
この膜モジュールにフロロテクノロジー社製のFS-392Bを3倍に濃縮した状態で通液し、中空糸膜の外側に塗布してから乾燥させることにより、膜蒸留用膜モジュールを得た。
上記の方法で得られた膜蒸留用膜モジュールのうち、1モジュールを解体して多孔質中空糸膜の性状を測定した。多孔質中空糸膜の水接触角は23℃の温度および50%の相対湿度の条件下で、2μLの純水を滴下し、液滴と中空糸膜外側表面とが形成する角度を画像解析により算出して接触角を求めた。測定は5回行い、数平均値を算出した。中空糸膜外側表面の接触角は、132°であり、とても強い疎水性を示していた。
実施例1では、図5に示した構成の濃縮装置1で濃縮を行った。
ステンレス製の原料液タンク(200)に原料液(a)1Lを入れた。原料液タンク(200)の底部から、正浸透膜モジュール(100)に、原料液(a)が供給でき、かつ、正浸透膜モジュール(100)によって濃縮された原料液(a)を原料液タンク(200)の底部に戻すように、配管が組んである。この正浸透膜モジュール(100)では原料液(a)中の溶媒(b)のうち、主として水が、誘導溶液(d)に移動して、原料液(a)の濃縮が行われる。
また原料液タンク(200)には、送風機(BL)により空気がタンクの上部空間に導入される空気入口と、この空気によって気化された溶媒(b)(水および有機溶媒)とが排出される出口が設けられている。これにより、原料液(a)中の溶媒(b)のうちの水および有機溶媒の双方が除去され、原料液(a)の濃縮が行われる。
原料液タンク(200)から排出された空気および溶媒(b)は、トラップ(400)に送られて、水および有機溶媒が回収される。
また、原料溶液タンクには液面計(LG)と比重計(HM)が接続されている。
原料液タンク(200)は、密閉状態であるが、原料液(a)が減少してもタンク内部の圧力が常圧に維持される構造となっている。
正浸透膜モジュール(100)の側管には、誘導溶液(d)を連続して供給できるように、入側配管と出側配管が連結されている。誘導溶液(d)は、誘導溶液タンク(300)内に貯留され、ポンプ(P)、流量計(図示せず)、および流量を調整するバルブ(図示せず)を介して正浸透膜モジュール(100)に供給される。なお、誘導溶液(d)としては、25wt%の塩化マグネシウム水溶液を用いた。
実施例2~5、ならびに比較例1および2では、図6に示した構成の濃縮装置2で濃縮を行った。
ステンレス製の原料液タンク(200)に原料液(a)1Lを入れた。原料液タンク(200)の底部から、正浸透膜モジュール(100)および膜蒸留用膜モジュール(500)に、それぞれ、原料液(a)が供給でき、かつ、それぞれのモジュールによって濃縮された原料液(a)を原料液タンク(200)の底部に戻すように、配管が組んである。原料液タンク(200)は、密閉状態であるが、原料液(a)が減少しても、タンク内部の圧力が常圧に維持される構造となっている。また、原料液タンク(200)には、液面計(LG)および比重計(HM)が接続されている。
原料液タンク(200)から正浸透膜モジュール(100)までの配管には、ポンプ(P)、流量計(図示せず)、流量を調整するバルブ(図示せず)があり、流量を適宜コントロールできる。
正浸透膜モジュール(100)の側管には、誘導溶液(d)が連続して供給できるように、入側配管と出側配管が連結されている。誘導溶液(d)は、誘導溶液タンク(300)内に貯留され、ポンプ(P)、流量計(図示せず)、および流量を調整するバルブ(図示せず)を介して正浸透膜モジュール(100)に供給される。なお、誘導溶液(d)としては、25wt%の塩化マグネシウム水溶液を用いた。
膜蒸留用膜モジュール(500)の側管には、冷却水(CW)が連続して供給できるように、入側配管と出側配管が連結されている。冷却水(CW)は、冷却水タンク(700)内に貯留され、ポンプ(P)、流量計(図示せず)、および流量を調整するバルブ(図示せず)を介して、膜蒸留用膜モジュール(500)に供給される。冷却水(CW)は、冷却水(CW)を一定温度に調整するための恒温装置(図示せず)および熱交換器(図示せず)を介して、膜蒸留用膜モジュール(500)に供給される。
冷却水(CW)には、膜蒸留用膜モジュール(500)にて原料液(a)から溶媒(b)(水および有機溶媒)が移動するので、濃縮運転の継続に伴って容積が増える。そのため、冷却水タンク(700)には、オーバーフロー口が設けられ、増えた分の冷却水(CW)の一部を系外に排出する構造になっている。
実施例では、原料液タンク(200)内の原料液(a)の液位が所定の目標値に達するまで濃縮運転を行うように、実験を行った。
濃縮装置1および2で濃縮を行う際には、下記のコントロール方法にしたがった。
正浸透膜モジュール(100)と、濃縮装置1の場合は送風機(BL)または濃縮装置2の場合は膜蒸留用膜モジュール(500)と、の双方を運転する。
(2)目標液位の+20%~0%まで
濃縮された原料液(a)の比重を測定し、以下の基準で運転する。
a)比重が適正な場合:正浸透膜モジュール(100)、および送風機(BL)(濃縮装置1)または膜蒸留用膜モジュール(500)(濃縮装置2)の双方の運転を継続する。
b)比重が高い場合:この場合は、濃縮された原料液(a)の溶媒(b)中の、水の割合が過剰であることを意味する。したがってこの場合には、送風機(BL)(濃縮装置1)または膜蒸留用膜モジュール(500)(濃縮装置2)の運転は停止して、正浸透膜モジュール(100)のみ運転を継続する。
c)比重が低い場合:この場合は、濃縮された原料液(a)の溶媒(b)中の、有機溶媒の割合が過剰であることを意味する。したがってこの場合には、正浸透膜モジュール(100)の運転を停止して、送風機(BL)(濃縮装置1)または膜蒸留用膜モジュール(500)(濃縮装置2)の運転を継続する。
(3)目標液位を下回った場合:
濃縮された原料液(a)の比重を測定し、以下の基準で運転する。
a)比重が適正な場合:直ちに濃縮運転を終了する。
b)比重が高い場合:送風機(BL)(濃縮装置1)または膜蒸留用膜モジュール(500)(濃縮装置2)の運転を停止し、正浸透膜モジュール(100)のみ運転を継続して、適正比重になった時点で濃縮運転を終了する。
c)比重が低い場合:正浸透膜モジュール(100)の運転を停止して、送風機(BL)(濃縮装置1)または膜蒸留用膜モジュール(500)(濃縮装置2)の運転を継続して、適正比重になった時点で濃縮運転を終了する。
実施例1では、濃縮装置1を用いた。
水900mLおよびアセトニトリル100mLの混合溶媒に、溶質としてトリプトファンを0.3wt%添加した溶液を、原料液(a)の模擬液として原料液タンク(200)に充填した。
正浸透膜モジュール(100)に、この原料液(a)と、誘導溶液(d)としての25wt%濃度の塩化マグネシウム水溶液と、を流した。
また、原料液タンク(200)の上部空間に、送風機(BL)にて空気を10L/分の流量で流し続けた。
目標とする濃縮倍率は10倍以上、溶媒(b)中のアセトニトリルの比率は10体積%を目標とした。
8時間の濃縮運転の結果、濃縮倍率が10倍以上であり、アセトニトリルの比率が10体積%の濃縮液が得られた。この濃縮時間の間の原料液の温度は27~30℃であった。
また、以下の分析結果:
濃縮に使った誘導溶液(塩化マグネシウム水溶液)をUV可視分光光度計UV-2400PC(SHINADZU)にて分析したところ、トリプトファンが検出されなかったこと、および
濃縮された原料液中のトリプトファンについての分析により、トリプトファンの変性が起きていないことが確認されたこと
から、原料液中の溶質を損なうことなく、原料液の非加熱濃縮が行われたことが検証された。
実施例2では、濃縮装置2を用いた。
水900mLおよびアセトニトリル100mLの混合溶媒に、溶質としてトリプトファンを0.3wt%添加した溶液を、原料液(a)の模擬液として原料液タンク(200)に充填した。
正浸透膜モジュール(100)に、この原料液(a)と、誘導溶液(d)としての25wt%濃度の塩化マグネシウム水溶液と、を流した。
一方、膜蒸留用膜モジュール(500)には、30℃に温度調節された原料液(a)を。と、10℃に温度調節された冷却水(CW)と、を流して、正浸透膜法および膜蒸留法の双方による濃縮を行った。
目標とする濃縮倍率は10倍以上、溶媒(b)中のアセトニトリルの比率は10体積%を目標とした。
6時間の濃縮運転の結果、濃縮倍率が10倍以上であり、溶媒(b)中のアセトニトリルの比率が10体積%の濃縮液が得られた。この濃縮時間の間の原料液の温度は29~33℃であった。
また、以下の分析結果:
濃縮に使った誘導溶液(塩化マグネシウム水溶液)、および冷却水を前記UV可視分光光度計にて分析したところ、トリプトファンが検出されなかったこと、
濃縮された原料液中のトリプトファンについての分析により、トリプトファンの変性が起きていないことが確認されたこと、および
濃縮後の原料液(a)に含まれるマグネシウム濃度をICP-MS法で分析したところ、誘導溶液から濃縮液へのマグネシウムの移動速度は、1時間当たり、膜面積1m2当たり、0.14g/m2/hrに過ぎなかったこと
から、原料液中の溶質を損なうことなく、原料液の非加熱濃縮が行われ、かつ、濃縮物への誘導溶液の混入は、ごく微量であることが検証された。
実施例3では、濃縮装置2を用いた。
水900mLおよびアセトニトリル100mLの混合溶媒に、溶質としてインスリン、ヒト、組み換え体(富士フィルム和光純薬製)を0.6wt%添加した溶液を、原料液(a)の模擬液として原料液タンク(200)に充填した。
正浸透膜モジュール(100)に、この原料液(a)と、誘導溶液(d)としての25wt%濃度の塩化マグネシウム水溶液と、を流した。
一方、膜蒸留用膜モジュール(500)には、30℃に温度調節された原料液(a)を。と、10℃に温度調節された冷却水(CW)と、を流して、正浸透膜法および膜蒸留法の双方による濃縮を行った。
目標とする濃縮倍率は10倍以上、溶媒(b)中のアセトニトリルの比率は10体積%を目標とした。
6時間の濃縮運転の結果、濃縮倍率が10倍以上であり、溶媒(b)中のアセトニトリルの比率が10体積%の濃縮液が得られた。この濃縮時間の間の原料液の温度は27~32℃であった。濃縮に使った塩化マグネシウム溶液、および冷却水を前記UV可視分光光度計にて分析したところ、インスリン、ヒト、組み換え体は検出されなかったため、濃縮は行われたと判断した。
原料液(a)の模擬液の溶媒として、水500mLおよびアセトニトリル500mLを混合して用い、濃縮目標を以下のように変更した他は、実施例2と同様にして、濃縮運転を行った。
目標とする濃縮倍率は10倍以上、溶媒(b)中のアセトニトリルの比率は50体積%を目標とした。
6時間の濃縮運転の結果、濃縮倍率が10倍以上であり、溶媒(b)中のアセトニトリルの比率が50体積%の濃縮液を得ることができた。この濃縮時間の間の原料液の温度は37~42℃であった。
実施例5では、濃縮装置2を用いた。
水900mLおよびアセトニトリル100mLの混合溶媒に、溶質としてトリプトファンを0.3wt%添加した溶液を、原料液(a)の模擬液として原料液タンク(200)に充填した。
正浸透膜モジュール(100)に、この原料液(a)と、誘導溶液(d)としてのイソプロパノール50wt%濃度の水溶液と、を流した。
一方、膜蒸留用膜モジュール(500)には、30℃に温度調節された原料液(a)と、10℃に温度調節された冷却水(CW)と、を流して、正浸透膜法および膜蒸留法の双方による濃縮を行った。
目標とする濃縮倍率は10倍以上、溶媒(b)中のアセトニトリルの比率は10体積%、溶媒(b)中のイソプロパノールの比率は5体積%未満を目標とした。
4時間の濃縮運転の結果、濃縮倍率が10倍以上であり、溶媒(b)中のアセトニトリルの比率が10体積%であり、溶媒(b)中のイソプロパノールの比率が1体積%未満の濃縮液が得られた。この濃縮時間の間の原料液の温度は29~33℃であった。
また、以下の分析結果:
濃縮に使った誘導溶液(イソプロパノール水溶液)、および冷却水を前記UV可視分光光度計にて分析したところ、トリプトファンが検出されなかったこと、および
濃縮された原料液中のトリプトファンについての分析により、トリプトファンの変性が起きていないことが確認されたこと
から、原料液中の溶質を損なうことなく、原料液の非加熱濃縮が行われたことが検証された。
原料液(a)の模擬液の溶媒として、水900mLおよびイソプロパノール100mLを混合して用い、濃縮目標を以下のように変更した他は、実施例2と同様にして、濃縮運転を行った。
目標とする濃縮倍率は10倍以上、溶媒(b)中のイソプロパノールの比率は10体積%を目標とした。
6時間の濃縮運転の結果、濃縮倍率が10倍以上であり、溶媒(b)中のイソプロパノールの比率が10体積%の濃縮液を得ることができた。この濃縮時間の間の原料液の温度は8~12℃であった。
膜蒸留用膜モジュール(500)を使用せず、正浸透膜モジュール(100)のみを使用して濃縮運転を開始し、この状態で濃縮倍率8倍まで運転を行い、続いて正浸透膜モジュール(100)を使用せず、膜蒸留用膜モジュール(500)のみを使用して濃縮運転を継続し、濃縮倍率10倍まで運転を行った以外は、実施例2と同様にして、濃縮運転を行った。
原料液(a)を、正浸透膜モジュールにて8倍まで濃縮するのに要した時間は、9時間であり、その後、膜蒸留用膜モジュールにて10倍まで濃縮するのに要した時間は、1時間であった(累計10時間)。正浸透モジュールによる濃縮に時間がかかったのは、濃縮中に溶媒中のアセトニトリル濃度が高くなって、正浸透膜モジュールによる正浸透処理の速度が遅くなったためと考えられる。
また、8倍濃縮時点の原料液(a)では、溶媒中のアセトニトリルの比率は21体積%であり、出発組成から大きく変わっていた。しかしながら、10倍濃縮後の原料液(a)では、溶媒(b)中のアセトニトリルの比率が15体積%であり、出発時と同じ溶媒組成に濃縮することができた。
10倍濃縮後の原料液(a)に含まれるマグネシウム濃度をICP-MS法で測定したところ、誘導溶液から濃縮液に移動したマグネシウムの移動速度は、1時間当たり、膜面積1m2当たり、2.2g/m2/hrであった。
正浸透膜モジュール(100)を使用せず、膜蒸留用膜モジュール(500)のみを使用して濃縮運転を開始し、この状態で濃縮倍率8倍まで運転を行い、続いて膜蒸留用膜モジュール(500)を使用せず、正浸透膜モジュール(100)のみを使用して濃縮運転を継続し、濃縮倍率10倍まで運転を行った以外は、実施例2と同様にして、濃縮運転を行った。
原料液(a)が1.2倍に濃縮された時点で、原料液(a)の一部に、白濁が生じた。トリプトファンが析出したものと思われる。
また、このときの原料液(a)は、溶媒(b)中のアセトニトリルの比率が3.6体積%まで低下していた。
原料液(a)を、10倍濃縮するまでに要した累計時間は、7時間であった。
10倍濃縮後の原料液(a)では、溶媒中のアセトニトリル比率は、7.7体積%であり、出発時よりも溶媒中のアセトニトリルの比率が低下した。
10倍濃縮後の原料液(a)に含まれるマグネシウム濃度をICP-MS法で測定したところ、誘導溶液から濃縮液に移動したマグネシウムの移動速度は、1時間当たり、膜面積1m2当たり、0.16g/m2/hrであり、濃縮液への誘導溶液の混入は、ごく微量であることが検証された。
膜蒸留用膜モジュール(500)を使用せず、正浸透膜モジュール(100)のみを使用した他は、実施例と同様にして、濃縮運転を行った。
正浸透膜モジュール(100)のみによって10倍に濃縮された原料液(a)は、溶媒(b)中のアセトニトリルの比率が35体積%に上昇しており、目標組成の濃縮液は得られなかった。この濃縮時間の間の原料液の温度は25~27℃であった。
濃縮液に含まれるマグネシウム濃度をICP-MS法で測定したところ、誘導溶液から濃縮液に移動したマグネシウムの移動速度は、1時間当たり、膜面積1m2当たり、3.5g/hr/m2であり、実施例2に比べると大きい値であった。その理由は、濃縮過程での原料液(a)の溶媒中のアセトニトリルの比率が高くなり、正浸透膜に、例えば膨潤等の構造変化があったためと考えられる。
正浸透膜モジュール(100)を使用せず、膜蒸留用膜モジュール(500)のみを使用した他は、実施例と同様にして、濃縮運転を行った。
膜蒸留用膜モジュール(500)のみによると、濃縮倍率10倍未満の段階で、溶媒(b)中のアセトニトリルの比率が1体積%未満となり、目標倍率までの濃縮ができなかった。この濃縮時間の間の原料液の温度は26~30℃であった。

Claims (28)
- 溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮方法であって、
前記溶媒が水および有機溶媒を含有し、
前記濃縮方法が、
正浸透法によって前記原料液中の水を除く第1の濃縮方法と、
前記原料液中の水および有機溶媒を蒸発させて除く第2の濃縮方法と
を、組み合わせた方法である、
原料液濃縮方法。 - 前記原料液中の水および有機溶媒を蒸発させて除く濃縮方法が、膜蒸留法である、請求項1に記載の原料液濃縮方法。
- 前記第1の濃縮方法の後に、前記第2の濃縮方法を実施する、請求項1または2に記載の原料液濃縮方法。
- 前記第2の濃縮方法の後に、前記第1の濃縮方法を実施する、請求項1または2に記載の原料液濃縮方法。
- 前記原料液の濃縮物中の溶質濃度および溶媒組成のうちの少なくとも1つをオンラインで測定し、前記測定された値に応じて、前記第1の濃縮方法および前記第2の濃縮方法の実施、濃縮速度の変更、または停止を決定する、請求項1~4のいずれか一項に記載の原料液濃縮方法。
- 前記原料液の濃縮物中の溶質濃度および溶媒組成のうちの少なくとも1つの測定を、前記濃縮物の比重測定、pH測定、導電率測定、液位測定、旋光度測定、近赤外分光分析、および重量測定から成る群から選択される1種以上の測定手段によって行う、請求項5に記載の原料液濃縮方法。
- 前記溶媒が、
水と、
アセトニトリル、メタノール、エタノール、およびイソプロパノールからなる群から選ばれる1種以上と
を含む、混合物である、
請求項1~6のいずれか一項に記載の原料液濃縮方法。 - 前記溶質が、アミノ酸、ペプチド、タンパク質、糖、ワクチン、核酸、抗生物質、抗体薬物複合体(ADC)、およびビタミン類からなる群から選ばれる1種以上である、請求項1~7のいずれか一項に記載の原料液濃縮方法。
- 前記溶質の数平均分子量が100~50,000のである、請求項1~8のいずれか一項に記載の原料液濃縮方法。
- 前記原料液の温度が1℃以上50℃以下の範囲に調整されている、請求項1~9のいずれか一項に記載の原料液濃縮方法。
- 前記正浸透法に用いる誘導溶液の溶質として、メタノール、エタノール、イソプロパノール、およびt-ブタノールから選ばれるアルコールを用いる請求項1~10のいずれか一項に記載の原料液濃縮方法。
- 第1の濃縮方法、および第2の濃縮方法のうちの少なくとも一方を実施しているときに、
原料液をろ過する、固液分離工程を行う、
請求項1~11のいずれかに記載の原料液濃縮方法。 - 溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮システムであって、
前記溶媒が水および有機溶媒を含有し、
前記濃縮システムが、
正浸透法によって前記原料液中の水を除く第1の濃縮手段と、
前記原料液中の水および有機溶媒を蒸発させて除く第2の濃縮手段と
を、組み合わせたシステムである、
原料液濃縮システム。 - 前記原料液中の水および有機溶媒を蒸発させて除く濃縮手段が、膜蒸留法である、請求項13に記載の原料液濃縮システム。
- 前記第1の濃縮手段の後段に、前記第2の濃縮手段が配置されている、請求項13または14に記載の原料液濃縮システム。
- 前記第2の濃縮手段の後段に、前記第1の濃縮手段が配置されている、請求項13または14に記載の原料液濃縮システム。
- 前記原料液の濃縮物中の溶質濃度および溶媒組成のうちの少なくとも1つをオンラインで測定する測定手段を有し、前記測定手段で測定された値に応じて、前記第1の濃縮手段および前記第2の濃縮手段の運転、濃縮速度の変更、または停止を決定する、請求項13~16のいずれか一項に記載の原料液濃縮システム。
- 前記原料液の濃縮物中の溶質濃度および溶媒組成のうちの少なくとも1つを測定する測定手段が、前記濃縮物の比重測定、pH測定、導電率測定、液位測定、旋光度測定、近赤外分光分析、および重量測定から成る群から選択される1種以上の測定手段である、請求項17に記載の原料液濃縮システム。
- 前記溶媒が、
水と、
アセトニトリル、メタノール、エタノール、およびイソプロパノールからなる群から選ばれる1種以上と
を含む、混合物である、
請求項13~18のいずれか一項に記載の原料液濃縮システム。 - 前記溶質が、アミノ酸、ペプチド、タンパク質、糖、ワクチン、核酸、抗生物質、およびビタミン類からなる群から選ばれる1種以上である、請求項13~19のいずれか一項に記載の原料液濃縮システム。
- 前記溶質の数平均分子量が100~50,000のである、請求項13~20のいずれか一項に記載の原料液濃縮システム。
- 前記原料液の温度が1℃以上50℃以下の範囲に調整されている、請求項13~21のいずれか一項に記載の原料液濃縮システム。
- 前記正浸透法に用いる誘導溶液の溶質として、メタノール、エタノール、イソプロパノールから選ばれるアルコールを用いる請求項13~22のいずれか一項に記載の原料液濃縮システム。
- 溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮装置であって、
原料液を貯留するための原料液タンクに、
正浸透法によって前記原料液の濃縮を行う、第1の濃縮ユニットと、
前記原料液中の水および有機溶媒を蒸発させて除くことによって前記原料液の濃縮を行う、第2の濃縮ユニットと
が連結された、装置。 - 溶質および溶媒を含有する原料液を濃縮して、原料液の濃縮物を得る、原料液の濃縮装置であって、
原料液を貯留するための原料液タンクに、
正浸透法によって原料液の濃縮を行う、第1の濃縮ユニットと、
膜蒸留法によって原料液の濃縮を行う、第2の濃縮ユニットと、
が連結された、装置。 - 原料液タンク中の原料液の溶質濃度および溶媒の組成のうちの少なくとも1つをオンラインで測定し、前記測定された溶媒組成に応じて、前記第1の濃縮ユニットおよび前記第2の濃縮ユニットの運転、濃縮速度の変更、および停止を自動で決定および実行する、請求項24または25に記載の装置。
- 前記原料液タンクが、フィルターおよび原料液ろ過用配管を有し、
前記フィルターは、前記原料液タンク内の原料液面よりも上部に設置されており、
前記原料液ろ過用配管は、前記原料液の底部に入り口を有し、かつ、前記フィルターの上部に出口を有し、
前記原料液ろ過用配管によって、前記原料液が、前記原料液タンクの底部から前記フィルターの上部に運搬されて、前記フィルターを通過して前記原料液タンク内の原料液に合流するように構成されている、
請求項24~26のいずれか一項に記載の装置。 - 前記原料液タンクの内部が、仕切り板によって第1室と第2室とに分割されており、
前記装置は、
前記第1室の底部から原料液を抜き出して、前記第1の濃縮ユニットおよび前記第2の濃縮ユニットのうちの少なくとも一方に供給する手段と、
前記第1の濃縮ユニットおよび前記第2の濃縮ユニットのうちの少なくとも一方から得られた濃縮された原料液を、前記第2室に戻す手段と、
前記第1室および前記第2室を連結する、第1室第2室連結配管と
を有し、
前記第1室第2室連結配管は、
前記第2室内の異なる高さに連結する複数の弁と、
前記第1室の底部に開口する出口と
を有し、
前記第2室内の原料液が、前記複数の弁のうちの少なくとも1つから採取され、前記第1室第2室連結配管を通過して、前記出口から排出されて前記第1室に戻るように構成されている、
請求項24~26のいずれか一項に記載の装置。
