Abstract
본 발명은 신규한 저혈당지수 감미제에 관한 것이다. 본 발명의 감미제는 수크로오스를 적게 포함하여 칼로리가 낮고 혈당 지수가 낮지만 단맛이 뛰어나다는 것을 기술적 특징으로 한다. 특히 본 발명의 감미 복합제는 수크로오스 대신 사카라이드 계열 화합물과 고감미제인 수크랄로스 또는 천연고감미제의 종류와 농도를 최적으로 배합하여 단맛을 좋게하는 동시에 그 외에 다른 미감을 극대화 시켰다는 점을 기술적 특징으로 한다.
Description
본 발명은 신규한 저혈당지수 감미제에 관한 것이다.
수많은 감미제 중에서, '설탕'으로 대표되는 전통적인 당질 감미제는 단맛이 강하고, 식품 중 자연성분으로도 가장 널리 분포하고 있으며, 또한 가공 식품 제조 시에도 가장 널리 사용되고 있는 당질이다. 그러나 최근 이들 당질 감미제의 비만, 당뇨병, 충치 등의 유발과 관련된 부정적인 측면이 부각되면서, 세계적으로 설탕을 대신하여 사용할 수 있는 기능성 대체 감미제가 주목 받고 있다.
다양한 식품 및 음료에서 기능성 대체 감미제가 사용되고 있으나, 많은 소비자들이 낮은 감미질과 좋지 않은 뒷맛 때문에 선호하지 않는다는 문제가 있다. 이에 따라 설탕의 감미 특성 및 미감, 풍미 및 뒷맛과 같은 다른 중요한 감각 특성을 모두 제공할 수 있는 기능성 대체 감미제를 개발하고자 하는 노력이 계속되고 있다.
그러나, 공지의 감미제의 배합이 반드시 더 강한 단맛을 일으킨다고 보장할 수 없으며, 감미제의 배합에 의해 통상의 기술자가 예상치 못했던 쓴맛, 텁텁함 등의 좋지 못한 감미질이 발생하여 문제된다. 종래 기술의 경우 회귀분석을 통하여 일반적으로 예측되는 상대 감미도만을 보고하여 왔을 뿐 실제 감미제의 배합이 예상했던 것과 동일한 상대 감미도를 갖는지 여부에 대한 실험을 수행한 바 없다.
이에 본 발명의 발명자들은 상기 문제점을 해소하고자 감미질이 우수한 저칼로리 감미제를 개발하였으며, 상기 감미제는 수크로오스의 함량을 적게 포함하면서도 단맛이 뛰어나다는 것을 기술적 특징으로 한다. 특히 본 발명의 감미 복합제는 수크로오스 대신 사카라이드 계열 화합물과 고감미제인 수크랄로스 또는 천연고감미제의 종류와 농도를 최적으로 배합하여 단맛을 좋게하는 동시에 그 외에 다른 미감을 극대화 시키는 연구를 성공적으로 수행하였다.
본 발명의 목적은

상기 목적을 달성하기 위한 본 발명의 하나의 양태는

본 발명의 감미제는 단위 질량 당 높은 단맛을 제공하며 영양가는 거의 제공하지 않거나 또는 전혀 제공하지 않는 물질을 의미한다.
상기 감미제는 단맛 기여도(SECs; sucrose equivalent concentrations) 수준을 기준으로 중량%가 측정된 것이며, 본 발명에서는 수크로오스 10% 수용액을 기준으로 뛰어난 단맛과 청량감을 내면서도 칼로리를 최소화 할 수 있는 배합비를 확인하기 위해 사용되었다. 본 발명에 있어서, 상기 단맛 기여도 수준은 수크로오스 10% 수용액과 동일한 단맛을 내기 위해 요구되는 기타 감미제의 상대적 농도를 의미한다.
본 발명에 있어서, 수크로오스는 테이블 슈가(Table sugar)로부터 정제된 설탕 또는 설탕 그 자체를 의미하며, 이당체로서 그 화학식은 C

11이다. 본 발명에서는 수크로오스의 함량을 줄이면서도 단맛은 유지하고 감미질이 뛰어난 감미제를 제조하는 것을 목적으로 한다.
본 발명에 있어서, 알룰로오스는 프시코스로도 불리며, 천연 화합물에 소량 존재하는 저칼로리 단당체 화합물을 의미하며, 그 화학식은 C

6이다. D-프럭토오스의 C-3 에피머에 해당하며, 수크로오스의 70%의 단맛을 내는 것으로 알려져 있다.
본 발명에 있어서, 에리스리톨은 당알코올의 한 종류이며 4탄당으로서 트레이톨과 이성질체 관계에 있다. 단맛을 냄에도 불구하고 칼로리가 1g당 0.2 kcal 정도로 칼로리가 매우 낮다.
본 발명에 있어서, 레바우디오사이드 A 및 D는 스테비올 글리코사이드로서 고감미제에 해당한다. 수크로오스의 200배 가량의 단맛을 낸다.
본 발명에 있어서, 타가토오스는 6탄당 단당체로서 수크로오스의 92% 가량의 단맛을 보유하나 38% 가량의 칼로리만 있는 것으로 알려진 감미제이다.
본 발명에 있어서, 수크랄로스는 체내에서 분해되지 않아 칼로리가 없는 인공 감미료로서 수크로오스 보다 320배 내지 1000배 가량의 단맛을 낸다.

또한 본 발명은 상기 어느 하나의 감미제를 포함하는 식품 조성물을 제공한다. 본 발명의 감미제는 흐름성이 좋고 흡습성이 낮으며 용해성이 뛰어난 분말 형태로 사용될 수 있으며, 높은 온도의 식품에 적용해도 감미도가 감소되지 않는다는 장점이 있다. 또한 본 발명은 설탕 대체 감미제로서 상기의 감미제를 포함하는 식품을 제공한다.
상기 식품은 소프트드링크, 커피, 차 등의 기호성 음료, 빵, 과자, 발효유, 유가공품, 콘 후레이크를 포함한 씨리얼류, 쨈, 아이스크림 및 빙과류, 조미식품 및 밥을 포함한 쌀가공품류로 등으로 이루어진 군 중에서 선택되는 어느 하나 이상인 것인, 식품 조성물일 수 있다. 상기 식품에 함유되는 혼합 조성물의 함량은 식품의 종류, 소망하는 감미 정도 등에 따라서 달라지며, 대체적으로 0.1 내지 30 중량%의 양으로 함유되는 것이 바람직하다.
상기 중량%는 조성물의 전체 중량% 대비 화합물이 차지하는 무게를 대비하여 계산하였다. 발명의 실시예에서는 단맛 기여도를 이용하여 화합물의 함량을 표시하였는데, 이를 실제 무게로 환산하여 조성물에서 차지하는 중량%로 표시한 것이다. 예를 들어 수크로오스 4 : 알룰로오스 2 : 타가토오스 2 : 수크랄로스 2는 수크로오스 10 중량%와 대비하였을 때 각각 수크로오스 4 중량%, 알룰로오스 3.704 중량%, 타가토오스 2.74 중량%, 수크랄로스 0.003 중량%와 대응된다. 따라서, 조성물의 전체 중량대비 중량%로 계산하면 이는 각각 수크로오스 38 중량%, 알룰로오스 35 중량%, 타가토오스 26 중량%, 수크랄로스 0.028 중량%로 계산된다.
본 발명의 감미제는 수크로오스의 함량을 적게 포함하면서도 단맛이 뛰어나다는 것을 기술적 특징으로 한다. 특히 본 발명의 감미제는 수크로오스 대신 사카라이드 계열 화합물과 고감미제인 수크랄로스 또는 천연고감미제의 종류와 농도를 최적으로 배합하여 단맛을 좋게하는 동시에 쓴맛을 최소화하고 청량감을 극대화하여 예상하지 못한 감미질을 달성하였다.

실시예 및 실험예
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예 및 실험예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 적용되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1. 상대 감미도의 도출
상대 감미도 및 상대감미도의 타당성을 검증하는 실험 절차에 대한 개략도를 도 1에 나타내었다.
1) 회귀곡선 도출
6 단계의 수크로오스와 6 단계의 표적 감미제 수용액을 1 테스트 세트에서 평가하는 수크로오스-감미제 결합 방법(Sucrose-sweetener combined method, SSC)을 이용하여 평가하였다. 이는 1세트 안에서 설탕과 표적 감미제를 함께 평가하여 평가시 개입될 수 있는 편향성을 제어하기 위함이다. 상기 SSC 방법과 관련된 개념도를 도 2에 나타내었다.
다양한 농도 (1 내지 9%)의 수크로오스 및 표적 감미제 평가를 회귀 분석을 이용한 자료로 평가하였다. 상기 표적 감미제로서 알룰로오스 파우더, 타가토오스, 에리스리톨, 수크랄로스, 레바우디오사이드 A 및 레바우디오사이드 D가 사용되었다. 하기 표1에 회귀 분석에 사용된 각 표적 감미제의 회귀식을 나타내었다.

수크로오스 및 표적 감미제 각각의 C-R (감미제 농도 - 감미제 강도 반응)곡선을 도출하여 도 3 내지 도 8에 나타내었다.
2) 도출된 회귀식을 이용한 상대 감미도 계산
도출된 회귀식을 이용하여 표적 감미제의 상대 감미도를 계산하였다. 예를 들어 수크로오스의 C-R 곡선과 표적 감미제의 곡선을 이용하여 수크로오스 4.5%와 같은 단맛을 내는 표적 감미제의 농도를 구한 후, 수크로오스에 대한 표적 감미제의 비를 계산하여 상대 감미도를 계산하였다. 알룰로오스 파우더, 타가토오스, 에리스리톨, 수크랄로스, 레바우디오사이드 A 및 레바우디오사이드 D의 상대 감미도 곡선을 도 9 및 도 10에 나타내었다.
3) 상대감미도 타당성 감증
에리스리톨의 예측 상대감미도가 정확한지 확인하기 위하여 5% 농도의 수크로오스 수용액과 그에 대응하는 농도의 에리스리톨 수용액을 비교 분석하였다. 타당성 검증 과정을 통해 앞서 조사된 종래 기술의 상대감미도보다 정확성과 타당성을 확보할 수 있었다. 종래 기술의 경우 일반적으로 예측 상대감미도만 보고할 뿐 실제 그 감미도를 갖는지에 대한 추가 확인에 대한 내용이 존재하지 않는다. 타당성 검증과 관련된 자료를 하기 표 2에 나타내었다.

분석 결과 벌크(bulk) 감미제는 고농도에서 비교적 상대감미도가 높게 평가되며, 고감미제는 저농도에서 상대감미도가 높게 평가되는 경향을 보였다. 특히, 고감미제의 경우 첨가 농도가 증가할수록 급격하게 상대 감미도가 떨어지는 것을 확인할 수 있었다.
실시예 2. 4성분 조합 저열량 복합 감미제의 감각적 특성 분석
1) 단맛 기여도 수준을 고려한 복합 감미제의 배합
타겟 식품에 일반적으로 첨가되는 설탕의 농도(10%)와 동일한 단맛 강도 수준을 갖도록 다양한 감미제 4종을 4 : 2 : 2 : 2 비율로 혼합하여 수용액 형태로 제시하였다. 혼합 시 각 감미제의 절대 양이 아닌 단맛 기여도(SECs; sucrose equivalent concentrations) 수준을 고려하여 조합하였다. 이때 적용되는 감미제의 상대감미도는 앞서 타당성 검증 과정을 통해 검증된 자료를 사용하였다. 본 실험에서 수행한 4성분 감미제의 배합을 하기 표 3에 나타내었다.

2) 묘사분석(generic descriptive analysis)
묘사분석을 실시하여 시료들을 평가하였다. 묘사분석은 제품에 대한 감각적 특성을 분석하는 평가 방법이다. 제품을 평가하기 위하여 제품의 감각 특성을 묘사하는 용어 개발 및 용어에 대한 정확한 정의를 패널 간 합의를 통해 결정하였다. 감각적 특성은 단맛, 쓴맛, 짠맛, 비린맛, 인공적인 단맛, 청량감, 점도, 후미떫음/텁텁함, 후미단맛 및 후미쓴맛에 대해 수행하였다.
i) 훈련 및 예비실험(training session)
훈련 과정에서는 다양한 농도의 단맛, 쓴맛, 떫은맛 수용액 시료를 제공하여 순위법을 실시함으로써 기본 맛에 대한 패널들의 예민도를 측정하고 훈련시켰다. 추가로, 강도 평가 시 동일한 평가 기준이 필요하기 때문에 패널들의 해당 능력을 훈련시키기 위한 방법으로 단맛 기본맛 훈련을 진행하였다. 이 때, 7%, 9%, 11% 13% 농도의 sucrose 수용액에 대한 단맛 강도를 15점 척도를 사용하여 평가하게 하였고, 본실험에서 제공하는 단맛 표준 시료 6%(단맛 강도 4점), 10%(단맛 강도 7점), 14%(단맛 강도 10점) sucrose 수용액을 함께 제공하여 비교 평가하게 하였다. 단맛 기본맛 훈련은 매 실험 전 실시하였다.
또한 본 실험에서 제품을 평가할 때 사용되는 제품의 감각적 특성 묘사 용어 개발 및 용어에 대한 정확한 정의를 패널 간 합의를 통해 결정하였다. 본 실험 직전, 실험 방법에 대한 이해와 적응을 목적으로 10% 수크로오스 수용액 시료를 이용하여 본 실험 방법과 동일한 방식의 예비 실험을 진행하였다.
ii) 본실험(main descriptive analysis)
본 실험에서 패널들은 독립된 관능 부스(sensory booth) 안에서, 외관에서 오는 bias를 줄이기 위해 빨간 불을 사용하여 샘플을 평가하였다.
i)단계에서 개발한 저열량 복합 감미 시료의 감각적 특성에 대한 강도를 측정하기 위하여 15점 척도를 사용하였으며, 0점은 ‘느껴지지 않음’, 7점은 ‘보통’, 14점은 ‘매우 강함’을 의미한다. 윌리암 라틴 스퀘어 디자인(Williams Latin Square Design)에 근거하여 시료 평가 순서를 결정한 다음 패널들에게 동시에 시료를 제시하였고, 시료 평가는 한 개의 시료에 대한 평가를 완전히 마친 후 다음 시료를 평가하는 방식으로 이루어졌다. 둔화효과를 방지하기 위하여 하루 최대 시료 평가 개수를 14개로 제한하였다. 한 시료 당 평가 시간은 5분이었으며, 쉬는 동안 따뜻한 물과 크래커를 제공하여 입을 헹구도록 지시하였다. 묘사분석을 통해 얻은 데이터의 신뢰성을 확보하기 위하여 각 시료 당 4반복 평가를 진행하였다. 또한 세션마다 대조군(10% 수크로오스 수용액)을 함께 제공하여 세션 간 시료 비교가 가능하게 하였다. 상기 훈련단계에서 개발한 저열량 복합 감미 시료의 감각적 특성을 하기에 표 4에 나타내었다.

3) 데이터 분석 : 묘사분석을 통해 얻은 데이터를 시료 간 감각적 특성 강도 간의 유의차를 검증하기 위하여 일반적 선형 모델 (general linear model, GLM)을 이용한 다변량분산분석(multivariate analysis of variance, MANOVA)을 실시하였다. 또한 분산분석 결과 유의적 차이가 있는 특성의 개별 평균들에 대한 추가 분석 과정으로 사후분석(Post-hoc analysis)을 실시하였다. 각 시료의 평균 및 표준편차를 하기 표 5에 나타내었다.

분석 결과 감미질이 수크로오스 10%와 유사하면서도 단맛 상승작용이 있던 4성분 조합 시료는 수크로오스를 기초로 한 i) 수크로오스 4 : 알룰로오스 2 : 타가토오스 2 : 수크랄로스 2, ii) 수크로오스 4 : 알룰로오스 2 : 에리스리톨 2 : 레바우디오사이드 A 2, iii) 수크로오스 4 : 알룰로오스 2 : 에리스리톨 2 : 레바우디오사이드 D 2, iv) 수크로오스 4 : 알룰로오스 2 : 수크랄로스 2 : 레바우디오사이드 D 2 및 v) 수크로오스 4 : 타가토오스 2 : 레바우디오사이드 A 2 : 레바우디오사이드 D 2이었다.
상기 농도는 앞서 설명한 대로 단맛 기여도 수준 %를 기초로 작성되었다. 이때 수크로오스 10중량%에 대비하여 각각, 수크로오스 4%는 4 중량%, 타가토오스 2는 2.74 중량%, 알룰로오스 2%는 3.704 중량%, 레바우디오사이드 A 및 D 2%는 0.008 %, 수크랄로스 2%는 수크랄로스 0.003% 중량%에 대응된다.
따라서, 예를들어 수크로오스 4 : 알룰로오스 2 : 타가토오스 2 : 수크랄로스 2는 수크로오스 10 중량%와 대비하였을 때 각각 수크로오스 4 중량%, 알룰로오스 3.704 중량%, 타가토오스 2.74 중량%, 수크랄로스 0.003 중량%와 대응된다. 따라서, 조성물의 전체 중량대비 중량%로 계산하면 이는 각각 수크로오스 38 중량%, 알룰로오스 35 중량%, 타가토오스 26 중량%, 수크랄로스 0.028 중량%로 계산된다.
이상과 같이 실시예를 통하여 본 발명을 설명하였다. 본 발명이 속하는 기술분야의 통상의 기술자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 그러므로 상술한 실시예들은 모든 면에 예시적인 것이며 한정적인 것이 아닌 것으로서 이해되어야 한다. 본 발명의 범위는 상세한 설명보다는 후술하는 특허청구범위에 의하여 나타내어지며, 특허청구범위의 의미 및 범위 그리고 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.

- 수크로오스 29 내지 39 중량%;알룰로오스 26 내지 36 중량%; 및에리스리톨 29 내지 39 중량%;을 포함하고,레바우디오사이드 A 0.050 내지 0.167 중량% 또는 레바우디오사이드 D 0.062 내지 0.133 중량%를 더 포함하는 것인 감미제.
- 제1항에 있어서,수크로오스 31 내지 37 중량%;알룰로오스 28 내지 34 중량%; 및에리스리톨 31 내지 37 중량%;을 포함하고,레바우디오사이드 A 0.055 내지 0.147 중량% 또는 레바우디오사이드 D 0.063 내지 0.123 중량%를 더 포함하는 것인 감미제.
- 제1항에 있어서,수크로오스 33 내지 35 중량%;알룰로오스 30 내지 32 중량%; 및에리스리톨 33 내지 35 중량%;을 포함하고,레바우디오사이드 A 0.060 내지 0.127 중량% 또는 레바우디오사이드 D 0.064 내지 0.113 중량%를 더 포함하는 것인 감미제.
