Abstract
표적분자(핵산, 지질, 당, 단백질, 펩타이드 등의 생체분자나 호르몬, 저분자 화학물질, 독성물질, 이온 등을 모두 포함)를 인식하는 DNA/RNA 압타머를 선별하기 위해서는 Systematic Evolution of Ligand Exponential Enrichment (SELEX) 과정을 수반한다. 일반적으로 SELEX를 수행하기 위해서는 표적분자를 기판이나 비드 표면에 고정시키는 과정이 필수적으로 요구된다. 또한 SELEX과정의 각 round에서 실제 압타머 라이브로리가 표적물질과 잘 결합하고 있는지에 대해 관찰할 수 있는 positive/negative 모니터링이 불가능하기 때문에 수회에 걸친 round이후 선별된 압타머를 분석함으로써 SELEX과정이 올바르게 진행되고 있는지를 간접적으로 확인하고 있다. 이러한 종래의 문제점들을 획기적으로 해결함과 동시에 보다 손쉽고 간편한 SELEX 기법을 구성하기 위해, 본 발명에서는 금나노입자를 이용한 새로운 SELEX 기법을 제시하고자 한다.
Description
본 발명은 압타머의 선별 방법에 관한 것이다. 압타머는 목적 물질에 대하여 특이적인 결합 능력을 갖는 폴리뉴클레이티드 분자로서, 단백질 센서, 나노센서 등에 활용될 수 있다. 기존 압타머의 선별 방법은 스크리닝 과정에서 많은 시간이 소요되어 어느 효능 이상의 압타머를 수득하기 위하여 오랜 시간이 걸리는 문제가 있다.
Systemic Evolution of Ligand Exponential Enrichment (SELEX) 기술은 목적 물질에 특이적으로 결합하는 압타머를 선별하기 위한 기술로서, 무작위 서열의 폴리뉴클레오티드 라이브러리를 생성하고 증폭시킨 후 목적 물질과 결합된 압타머를 분리하는 방법이다.
양적 SELEX (positive SELEX)는 SELEX 과정을 여러번 반복하여 목적 물질에 대한 높은 특이성을 갖는 압타머의 순도를 높이는 과정이다.
음적 SELEX (negative SELEX)는 SELEX 공정에 쓰이는 시료 또는 목적 물질과 상동성을 갖는 물질에 대하여 결합 능력을 갖는 압타머를 함으로써 목적 물질에 대하여 선택적인 특이성을 갖는 압타머를 선별하는 과정이다.
압타머는 타겟에 특이적으로 결합할 수 있다는 점에서 항체와 쓰임이 비슷하다. 그러나 in vitro에서 실험자가 원하는 표적물질에 대해 인공적으로 합성된 핵산 라이브러리로부터 선정할 수 있다는 점에서 압타머는 항체가 개발될 수 없는 다양한 범위의 타겟에 대해 용도에 맞게 개발될 수 있다. 압타머를 선정하기 위해서는 일반적으로 SELEX (Nature, 1990, 346, 818-822) 라는 스크리닝 방식이 사용되는데 이는 10-20 round 정도가 반복되며, 타겟과 라이브러리의 결합, 결합하지 않은 서열의 세척, 타겟과 결합한 서열의 해리, 해리된 서열의 증폭 과정으로 이루어져 있다. 일부 타겟의 경우 세척과정을 위해 타겟을 modification 하여 표면에 고정시키는 과정이 필요하며, SELEX round가 완료된 이후에는 선정된 압타머의 성능을 개선시키기 위한 post-SELEX과정이 필요하다. 또한, SELEX round 진행 중 3-5라운드 완료마다 monitoring이 수반되어야 한다. 그러므로 압타머를 선정하기 위해서는 최소 수개월의 시간이 필요하다.
압타머 선정과정을 간소화하고 더 나은 압타머를 선정하기 위해 SELEX를 변형시키는 다양한 방법들이 보고되어 있다. Graphene oxide를 사용하여 타겟을 표면에 고정하지 않고 압타머를 선정할 수 있는 GO-SELEX(Chem. Commum, 2012, 48, 2071-2073), click chemistry를 이용하여 post-SELEX 과정을 간소화할 수 있는 Click-SELEX(Nature Protocols, 2018, 13, 1153-1180) 등이 대표적이다. 그러나 모니터링은 여전히 SELEX에서 중요 과제이며, 이는 실험자가 SELEX round가 진행되는 동안은 압타머가 원하는 방향으로 선정되고 있는지 확인할 수 없기 때문이다. SELEX 과정 안에는 라이브러리를 증폭하는 PCR 에서의 오류, target의 낮은 순도, 과한 selective pressure 등 SELEX가 실패할 수 있는 변수들이 다수 포함되어 있으므로 모니터링은 SELEX의 진행 경과를 판단하기 위해 필수적이다.

DNA검출, 단백질 검출, 중금속 이온, 면역측정법 등)은 널리 수행되어왔으나, 나노입자의 색상 변화를 통한 압타머의 선정 과정을 모니터링 하는 방법은 보고된 바가 없다. 본 기술에서 ssDNA와 타겟물질 간의 결합력이 높을 경우, 금 나노입자는 염을 만났을 때 나노입자 표면이 변성이 되어 응집이 발생하고, 응집과정에서 금 나노입자는 색깔 변화를 유도하게 된다. 이는 나노입자 크기에 따른 표면 플라즈몬 및 빛의 산란 정도가 변화하기 때문에 발생하는 현상이다. 즉, 일반적인 금 나노입자의 경우 적갈색을 띄고 있으나, 응집 시에는 입자 크기가 커지고 보다 큰 파장을 산란하게 되어 금 나노입자가 녹아 있는 용액의 색깔이 그 크기에 따라 보라색, 푸른색, 잿빛 등으로 변하게 된다. 반면에, ssDNA와 타겟의 결합력이 약할 시에는 위와 같은 금 나노입자의 응집이 억제가 되어 색깔 변화가 덜 발생하게 해당 SELEX라운드에서의 ssDNA pool과 타겟과의 결합력을 색깔의 변화를 통해 신속하고 빠르게 분석 할 수 있다. 용액 내 금 나노입자 색깔의 변화는 간단한 흡광분석만으로 손쉽게 정량이 가능하며, 매 라운의 마지막에 염을 추가하여 쉽게 모니터링이 가능하기 때문에 기존 방법에 비해 보다 효과적인 SELEX 과정 평가가 가능하다.
진술한 바와 같이, 금 나노입자의 단일가닥 DNA에 대한 친화력과 염으로 인한 응집현상을 활용하면 보다 신속하고 간편하게 SELEX과정을 분석하고 모니터링 할 수 있다. 특히 별도의 monitoring 과정이 필요하지 않고, 기존의 SELEX와 비교하였을 때 금나노입자의 색 변화만으로 진행 수준을 판단하여 압타머 선정 과정을 유연하고 실험자 편의에 맞게 진행 할 수 있다. 또한 SELEX를 하기 위해 타겟을 화학적으로 modification 할 필요가 없는 labeling-free 방식으로서, 원하는 타겟의 원형 그대로 압타머 선정을 진행할 수 있다. 이를 이용하면 관심 물질에 대한 압타머 선정이 간편해질 뿐만 아니라 선정된 압타머를 이용한 타겟 이미징, 의료 진단, 환경 또는 식품분야에서의 독성 센싱, 타겟 특이적 약물 전달 치료 등에 광범위하게 활용하여 발전을 기대할 수 있다.

실험예 1. 테스트를 위한 금 나노입자의 합성
본 발명에 의한 단백질 분해효소 활성분석 검증을 위해 카르복실기로 변형된 금 나노입자가 합성되었다.
금 나노입자는 흔히 알려진 방법대로, 시트르산(citrate)으로 환원 및 안정화 하는 방법으로 합성되었다. 요약하자면, 1 mM 농도의 사염화금(HAuCl4ㆍ3H2O, Sigma-Aldrich) stock solution 20 mL를 50 mL 의 증류수에 넣고 지속적으로 교반시켜 최종 농도 300 nM의 용액을 만들었다. 이 용액에, 30 mM 시트르산 소듐(C6H5Na3O7ㆍ2H2O, Sigma-Aldrich) 2 mL이 더해져 최종 농도 600 nM이 되도록 하였고, 교반시켰다. 환원시키고 금 콜로이드를 형성하기 위해, 용액을 끓이면서 교반시켰다.
실험예 2. 금 나노입자에 기초한 SELEX(Gold-SELEX)를 수행하기 위한 ssDNA library 준비
Random ssDNA library 는 A,G,C,T염기의 비율이 1:1:1;1이 되도록 준비하였으며, 3'과 5' 양 말단에 DNA 증폭을 위한 프라이머 자리를 위치시켰다. Citrate로 안정화된 금나노입자는 대부분의 ssDNA를 흡착할 수 있으나, 서열의 염기 구성에 따라 일부 ssDNA와는 친화력이 떨어질 수 있다. 금나노입자와의 결합력이 낮은 서열이 타겟물질과의 친화도와 상관 없이 금나노입자로부터 탈락하여 positive SELEX과정에 선정되는 것을 방지하기 위하여 Gold-SELEX 의 시작 단계에서 다음의 두가지 방법으로 ssDNA library를 준비했다.
첫번째 방법은 초반 1-4라운드 가량에 추가적인 원심분리 과정을 넣는 방법이다. 하기 실시예 중 Brassinlide Gold-SELEX 에 해당된다. 금나노입자 75ul와 ssDNA 150nM, 1X phosphate buffered saline(1X, PBS의 조성은 다음과 같다. 0.137M Sodium chloride, 2.7mM Potassium chloride, 4.3mM, Sodium phosphate (dibasic, anhydrous), 1.4mM Potassium phosphate (monobasic, anhydrous)) 20ul, 증류수를 섞어 최종 부피가 190ul가 되도록 하고 10분동안 나노입자와 ssDNA가 붙을수 있도록 한다. 금나노입자와 친화력이 없는 ssDNA 서열을 제거하기 위해 6500g에서 10분동안 원심분리해준 뒤에 상층액을 버리고, 남은 금나노입자+ssDNA를 0.1X PBS 190ul로 희석하여 이후 SELEX과정에 사용했다.
두번째 방법은 타겟물질과 첫 positive SELEX과정을 진행하기 전에 수행할 수 있으며, 하기 예시 중 Bisphenol A Gold-SELEX에 해당된다. 금나노입자 75ul와 ssDNA 150nM, 1X PBS 20ul, 증류수를 섞어 총 190ul의 샘플을 준비하여 상온에서 10분동안 ssDNA와 금나노입자를 결합시킨다. 6500g에서 10분동안 원심분리하여 상층액의 나노입자에 결합하지 않은 서열을 제거하고 남아있는 금나노입자+ssDNA를 40ul 증류수에 희석하였다. 95℃에서 10분 끓여 ssDNA를 나노입자로부터 해리시킨 뒤에 13000rpm으로 1분 원심분리하여 상층액의 ssDNA만 취한다. PCR하여 해리된 ssDNA를 증폭하고 해당 과정을 1-3 cycle반복하여 금나노입자와 결합하지 않는 서열을 ssDNA library로 부터 제거할 수 있으며 준비된 library는 Gold-SELEX에 사용되었다.
실험예 3. 금 나노입자의 비색법에 기초한 Brassinolide(이하 BL)와 Bisphenol A(이하 BPA)에 대한 Gold-SELEX 의 monitoring과 압타머 선정
Gold-SELEX의 활용성을 확인하기 위하여, BL과 BPA 두가지 표적물질에 대한 압타머 선정을 진행하였다. Positive SELEX는 타겟과 ssDNA의 결합력을 높히는 SELEX과정이다. ssDNA library 30pmol, 75ul 금나노입자, 1X PBS, 증류수를 섞어 총 190ul의 반응물을 준비하여 금나노입자와 ssDNA가 붙을 수 있도록 10분동안 상온에서 반응시킨 뒤 첫번째 사진촬영과 흡광도 측정을 진행하였다. 사진촬영은 모두 동일한 장소와 조건에서 한대의 휴대전화 내장 카메라로 진행되었으며, 흡광도는 마이크로 플레이트 리더를 사용하여 투명 96 웰 플레이트에서 500-800nm의 파장 범위를 측정하였다. 측정 뒤에 10ul의 타겟을 원하는 농도만큼 첨가하고 10-30분 동안 상온에서 반응시켰으며, 이 과정의 농도, 시간, 온도는 타겟의 종류와 SELEX의 진행상황에따라 달라질 수 있다. 첫번째와 동일한 방법으로 사진촬영과 흡광도 측정을 완료한 뒤에 샘플에 1M NaCl을 10첨가하고 15분 동안 색 변화를 기다렸다. 이후 색 변화가 안정된 뒤에 마지막 사진촬영과 흡광드 측정을 진행하였다. 촬영이 끝난 샘플은 1.5ul 마이크로 튜브로 옮겨 6500g에서 10분동안 원심분리하여 ssDNA+target이 포함되어 있는 상층액만 취했다.
상층액에서 얻은 ssDNA는 Ethanol precipitation과정을 통해 타겟으로부터 분리되었다. 3M Sodium acetate 20ul, 차가운 (-20℃ 보관) 100% 에탄올 660ul를 샘플에 첨가하여 -20℃에서 30분 동안 반응하였다. 14000rpm에서 20분 동안 원심분리하여 상층액을 버리고, 차가운 70%에탄올 1mL을 첨가해 DNA펠렛을 세척한 뒤 15분동안 추가로 원심분리하였다. 상층액을 모두 제거하고 10분동안 상온에서 펠렛을 말린 뒤에 40ul 증류수를 넣어 DNA를 녹여내었다. 이런 과정으로 얻은 ssDNA는 polymerase chain reaction(PCR)을 통해 증폭되어 다음 라운드에 사용되었다.
한편 ssDNA와 counter target의 결합력을 낮춰 선정된 압타머의 특이성을 높히기 위해 Negative SELEX도 진행되었다. 첫번째 촬영 및 측정까지의 과정은 positive SELEX와 동일하며 target 대신 배제하고자 하는 counter taraget을 원하는 농도만큼 10ul를 샘플에 섞어주었다. 이후 3번째 촬영 및 측정 과정까지 positive SELEX와 동일하다. 측정이 끝난 샘플을 1.5ul 마이크로 튜브로 옮겨 6500g에서 10분동안 원심분리한 뒤에 상층액을 버리고 금나노입자+ssDNA 펠렛에 40ul의 증류수를 첨가하여 95℃에서 10분 끓여 나노입자로부터 counter target과 결합하지 않은 DNA를 해리시켰다. 해리된 DNA는 PCR 과정을 통해 증폭하여 다음 라운드에 사용하였다.
도 9 및 10은 식물유래 스테로이드 호르몬의 일종인 brassionlide(BL)에 결합하는 DNA압타머를 선정한 Gold-SELEX 결과이다. 압타머 또는 항체가 존재하지 않는 물질에 대하여 Gold-SELEX를 총 9라운드 완료하였으며, BL에 높은 결합력을 보이는 서열을 선정하기 위한 positive SELEX 2단계, beta-sitosterol에 대한 결합력이 낮은 서열을 선정하기 위한 negative SELEX 1단계로 이루어져 있다. 타겟 물질 (브라시놀리드)과 논타겟 물질 (B-시토스테롤)의 구조는 각각 구조식 1 및 2와 같다.

도 9와 10은 SELEX전과정을 금나노입자의 색상 변화에 의해 모니터링한 결과이다. 색상 변화는 620nm에서의 흡광도와 520nm에서의 흡광도의 비율로 측정되었으며, 타겟 또는 counter target과의 DNA결합력이 높을수록 염을 추가한 뒤의 금나노입자가 적색에서 보라색을 거쳐 푸른색으로 변화하게 되고, 그 결과 520nm에서의 흡광도가 낮아지고 620nm의 흡광도가 올라가기 때문에 E620/E520 의 값이 올라가게 된다.
도 9와 10에서, 1-4라운드까지 BL에 대한 positive SELEX가 진행될 수록 타겟에 대한 ssDNA pool의 결합력이 높아져 색상이 붉은색에서 보래색으로 바뀌었다. 5-7라운드 동안은 라운드가 진행될 수록 b-sitosterol에 대한 DNA pool의 결합력이 감소하여 금나노입자의 색상이 붉은색으로 바뀌었다. 7라운드의 경우, 모니터링 결과에 근거하여 6라운드 산물이 충분한 타겟 특이성을 획득하였다고 판단하였으며 같은 6라운드 산물을 사용하여 positive SELEX인 7라운드를 추가로 진행했고 이를 7-P라운드라 했다. 그 결과 4라운드와 비교하여 7-P의 타겟과의 친화도가 감소하였음을 확인하였고, 7-N은 폐기하고 7-P 부터 9라운드까지 positive SELEX를 추가로 진행하였다. 그리고 BL에 대한 결합력이 4라운드만큼 다시 회복되었을 때 SELEX를 완료하였다.
도 11 및 12는 내분비계교란물질(환경호르몬)의 일종인 비스베놀 A(BPA) (구조식 3)에 결합하는 DNA 압타머를 선정한 Gold-SELEX결과이다. 기존에 다른 SELEX방법으로 압타머가 개발되어있는 알려진 타겟에 대해 본 발명으로 새로운 압타머 선정을 완료함으로써 기존의 SELEX방법과 비교하여 Gold-SELEX의 압타머 선정 효율을 증명하고자 하였다.
BPA에 대한 SELEX 는 총 11라운드가 진행되었으며, 한번의 positive SELEX와 Negative SELEX로 구성되어 있다. 타겟 물질과 논타겟 물질은 각각 비스페놀 A (구조식 3) 및 비스페놀 S (구조식 4)였다.

도 11, 도 12는 620nm와 520nm에서의 흡광도 비율을 이용한 SELEX전체 과정을 모니터링한 결과이다. 모든 측정은 BL SELEX와 동일하게 진행되었다.
도 11, 도 12에서, 1-8라운드까지 BPA에 대한 positive SELEX가 진행될 수록 타겟에 대한 ssDNA pool의 결합력이 높아져 색상이 붉은색에서 보라색으로 바뀌는것을 확인하였다. 8라운드의 경우 DNA pool이 BPA에 대한 충분한 결합력을 얻었다고 생각되어, 7라운드 산물의 타겟 특이성을 확인하기 위해 Bisphenol S(BPS)를 타겟과 동일한 농도로 사용하여 counter SELEX를 추가로 진행하였다. 그 결과 7라운드 산물은 BPS에 대해서도 BPA와 비슷한 결합력을 나타냄을 확인했다. 타겟 특이적 결합력이 높은 BPA 압타머를 선정하기 위해 8-P는 폐기하였고, 8-N부터 11라운드까지 Negative-SELEX를 진행하였다. BPS에 대한 결합력이 2라운드 수준으로 감소한 뒤에 SELEX를 완료하였다.
실험예 4. Gold-SELEX 의 시퀀싱 결과 확인
SELEX가 완료된 후, 서열확인을 위해 BL SELEX 9라운드 산물, BPA SELEX의 11라운드 산물을 PCR로 증폭하여 t-vector cloning하여 Dh5a competent cell에 transformation 시켰다. 50-100개의 colony를 취하여 플라스미드를 prep한 뒤에 T7 promoter로 시퀀싱하였다. 그 결과 유사도가 높고 출현 빈도가 높은 서열을 위주로 분석하였으며 결과는 아래 표와 같다.
실험예 5. 금 나노입자의 비색법에 기초한 BL과 BPA에 대한 압타머 결합력 및 특이성 확인
각각의 최종 SELEX 라운드의 산물로부터 가장 타겟에 대한 친화도가 높은 서열 한가지씩을 선정하였으며, 각각의 후보들에 대해 타겟 결합력과 특이성을 확인하였다. 모든 실험은 Gold-SELEX와 같은 조건으로 실험하였다.
도 13 내지 15는 BL 에 대해, 도 16 내지 18는 BPA에 대해 선정된 압타머의 target과 counter target들에 대하여 압타머의 결합력과 특이성을 확인한 결과이다. 도 13이 최종적으로 선정된 BL 압타머인 BLA9-20의 2차 구조이고, 도 16은 최종적으로 선정된 BPA압타머인 nBPA40의 2차 구조이다. 두 구조 모두 mfold web server(M. Zuker. Mfold web server for nucleic acid folding and hybridization prediction. Nucleic Acids Res. 31 (13), 3406-3415, 2003.)로부터 열역학적으로 안정성이 가장 높은 서열로 예측, 선정되었다. 그리고 도 14, 15, 17, 및 18로부터 선정된 압타머들은 각각의 타겟에 대한 대한 결합력이 가장 높으며, 비슷한 구조의 다른 물질들에 대해서는 낮은 상대적으로 낮은 결합력을 가지고 있음을 확인할 수 있다. 도 14 및 17에서 측정된 흡광도 비율 E620/E520은 정확한 비교를 위해 ((분석물이 들어갔을 때의 Ext. Ratio)-(분석물이 없을 때의 Ext. Ratio ))/(분석물이 없을 때의 Ext. Ratio)의 식으로 normalize 되었다. 농도별 표준 시약은 실험 전에 농도별로 희석하여 준비되었으며, 100% 에탄올에 희석되었다. 도 14 및 15의 실험에 사용된 물질들의 구조가 도 15 하단에 나타나 있으며, 모두 식물체내에서 BL이 생합성 되는 과정의 중간 산물이다. 도 17 및 18에 사용된 물질들의 종류와 구조가 도 18 하단에 나타나 있다.
실험예 6. 알려진 BPA 압타머와 Gold-SELEX로 선정된 BPA압타머의 비교
다른 SELEX방법으로 선정되어 이미 알려져있는 BPA DNA 압타머와 Gold-SELEX로 선정된 새로운 압타머 서열 두가지의 타겟 감지 능력을 금나노입자 비색법으로 비교하였고, multi align기법으로 두 압타머의 염기서열을 확인하였다.
실험조건은 Gold-SELEX 압타머 선정 과정과 동일하게 진행되었으며, 도 19와 도 20의 결과에서 확인할 수 있듯, Gold-SELEX로 선정된 압타머인 nBPA40이 기존에 발표되어있는 압타머보다 금나노입자 비색 센서로 활용하기에 적합한 것으로 보인다. 그리고 서로 다른 방법으로 선정되었음에도 불구하고, 도 21의 결과로 보아 nBPA40과 기존의 압타머 간 유사성이 높은 부분들이 발견되었다. 붉은색의 경우 90%이상의 유사성, 푸른색의 경우 50-90% 유사성, 검정색이 경우 50% 미만의 유사성을 보임을 의미한다.
실험예 7. 브라시놀리드 결합성 압타머의 개량화
도 22 내지 도 25는 BLA9-20 압타머를 개량화하여 브라시놀리드와의 결합력을 비교한 결과이다. 도 22에서 나타낸 바와 같이, BLA9-20 (총 60 bp)에서 표시된 부분만을 절단하여 구성한 truncated 압타머 (tBLA9-20v1, 총 29 bp, 서열번호 17) 형태와, single base pair를 추가하여 구성한 truncated 압타머 (tBLA9-20v2, 총 31 bp, 서열번호 18)로 제작하였다. 이 2가지의 truncated 압타머에 대해 브라시놀리드의 결합력을 금 나노입자의 비색법으로 측정한 결과 tBLA9-20v2가 tBLA9-20v1보다는 상대적으로 높이 측정되었으며 tBLA9-20v2 결합친화도는 BLA9-20에 비해 다소 높게 측정 (K
d=54.2 nM)되었다 (도 23). tBLA9-20v1과 tBLA9-20v2에 브라이놀리드를 처리하기 전과 처리 후에 2차구조를 circular dichroism (CD)을 사용하여 비교한 결과 tBLA9-20v1의 경우는 브라이놀리드의 결합이후 2차구조가 바뀌는 반면, tBLA9-20v2의 경우는 2차구조에 변화가 없다는 것을 알 수 있다 (도 24 및 도 25).
실험예 8. 브라시놀리드 결합성 압타머를 이용한 브라시놀리드의 검출 방법
도 26은 압타머를 이용한 브리시놀리드 검출방법으로서 항체를 이용한 면역침강법 (immunoprecipitation)과 유사한 압타머 침강법 (aptamer precipitation)으로 활용하여 검출할 수 있음을 나타낸다. 즉, 아비딘으로 코팅된 마이크로비드 표면을 바이오틴-압타머 (biotin- tBLA9-20v2)를 결합시키고 각각 다른 농도의 브라시놀리드가 포함된 애기장대 추출물과 반응 후 세척할 수 있다. 애기장대 추출물에 포함된 브라시놀리드는 비드 표면에 강하게 결합되어 있게 되는데 최종 에탄올을 처리하여 압타머에 결합된 브라시놀리드를 효과적으로 추출 후 금나노입자 비색법으로 정량화 할 수 있다. 도 27은 실제 야생형 애기장대 추출물 (WT, wild-type Arabidopsis)과 브라시놀리드 생합성이 저해되는 배지에서 자란 돌연변이 애기장대 추출물 (Sdet2, BL-deficient mutant Arabidopsis), 그리고 야생형 애기장대에 브라시놀리드가 풍부하게 존재하는 배지에서 자란 추출물 (WT+BL, wild-type Arabidopsis with BL-rich media)을 분석하여 표준 정량 곡선과 비교한 결과 야생형 애기장대에 비해 Sdel2는 상대적으로 적고, WT-BL는 매우 높은 발색범위를 나타내어 정량적으로 분석이 가능함을 확인하였다.
