Abstract
物質の細胞層透過を促進させるための技術を提供する。 下記一般式(1)で表わされる化合物からなる細胞層透過促進剤を提供するものである。また、上記細胞層透過促進剤を含み、生体への薬剤の吸収を補助するためのものである、薬剤吸収補助用組成物を提供するものである。また、上記細胞層透過促進剤を含み、生体に吸収させるべき薬剤を更に含む、医薬組成物を提供するものである。
Description
本発明は、細胞層透過促進剤、薬剤吸収補助用組成物、及び医薬組成物に関する。
上皮組織等を形成する細胞層では、隣り合う細胞同士の間隙がタイトジャンクション(TJ: Tight Junction)等の細胞接着構造によりシールされ、異物に対するバリア機能を果たしている。このため、細胞と細胞の間をすり抜ける細胞間隙経路での物質透過は、生体の恒常性維持のためには、きわめて制限的といえる。この細胞接着構造を一次的に緩和して、所望の物質を細胞間隙経路で透過させることにより、薬剤等の生体内への吸収を高めようとするドラッグデリバリーシステム(DDS: Drug Delivery System)の考え方がある(非特許文献1~5)。ペプチド、タンパク質、抗体、ワクチン、核酸など、近年開発の盛んなバイオ医薬品やその候補物質の中には低バイオアベイラビリティーのものもあり、ドラッグデリバリーシステム(DDS: Drug Delivery System)の技術は、そのような薬剤の適用可能性の範囲を広げ、また、注射投与ではなく、非侵襲的な、経口、経皮、経鼻、経肺、経粘膜などの投与方法により、患者のQuality of Lifeの向上にも繋がるので、その技術の開発が望まれている。
一方、下記非特許文献6には、シュードモナス菌由来の抗菌性物質として、環状リポペプチドの報告がある。

しかしながら、上記非特許文献6は、物質の細胞層透過を促進させる技術について、着眼するものではなかった。
よって、本発明の目的は、物質の細胞層透過を促進させるための技術を提供することにある。
上記目的を達成するため、本発明者らが鋭意研究した結果、特定の環状リポペプチドには、物質の細胞層透過を促進する作用効果があることを見出し、本発明を完成するに至った。
すなわち、本発明は、第1に、下記一般式(1)で表わされる化合物からなる細胞層透過促進剤を提供するものである。

Xは、ロイシン又はその保存的置換、より好ましくは、ロイシン、イソロイシン、ノルロイシン、tert-ブチルアラニン、tert-ロイシン、バリン、シクロへキシルグリシン、又はシクロヘキシルアラニン、又はアラニンであり、

本発明によれば、特定の環状リポペプチドには、物質の細胞層透過を促進する作用効果があるので、これを利用して、優れた細胞層透過促進剤、薬剤吸収補助用組成物、及び医薬組成物を提供することができる。

上記化合物の一般式中、Rは置換基を有してもよい炭素数1~9のアルキル基として表わされるが、典型的には、そのアルキル基は任意に水酸基を有していてもよく、より典型的には置換基として1~4個の水酸基のみを有するアルキル基であってもよく、更により典型的には置換基として1~2個の水酸基のみを有するアルキル基であってもよい。また、アルキル基長としては炭素数1~6であってもよく、炭素数1~3であってもよい。更に、後述の実施例を考慮すると、Rとしては下記式(4)又は(5)で表わされる基であることが、より典型的である。また、下記式(4)又は(5)で表わされる基のアルキル基長は炭素数9であるが、アルキル基長としては炭素数6であってもよく、炭素数3であってもよい。
本発明に用いる上記化合物としては、上記非特許文献6に示されるように、少なくともその1種は、ある種のシュードモナス菌に含まれていることも明らかとなっており、そのような菌株、変異株、遺伝子導入株等から抽出して調製してもよく、その他の微生物等の他の生物資源から抽出して調製してもよいが、工業的に生産する観点からは、人工的に合成されたものであることがより好ましい。合成法としては、液相法、アジフェーズ法、固相法等が挙げられる。なかでも、中間体の精製を省略可能であることから固相法によって合成することがより好ましい。
以下には、固相法の一例として、Fmoc固相合成法の具体例を述べる。ただし、以下に示す好ましい合成法の記載は、本発明の範囲が、それらの方法で得られたものに限る趣旨のものではない。
(Fmoc固相合成法)
Fmoc固相合成法の手順としてはまず、樹脂上の保護基であるFmoc(9-フルオレニルメトシキカルボニル)基を20% (v/v) ピペリジン/DMF溶液中、室温で20分間反応させ除去する。樹脂を洗浄後、種々の縮合剤を用い、アミノ酸を縮合する。このサイクルを繰り返すことで目的ペプチドを獲得する。縮合剤としてはカルボジイミド試薬、ジフェニルリン酸アジド、BOP試薬、HATU試薬などが用いられる。
Fmoc固相合成法の手順としてはまず、樹脂上の保護基であるFmoc(9-フルオレニルメトシキカルボニル)基を20% (v/v) ピペリジン/DMF溶液中、室温で20分間反応させ除去する。樹脂を洗浄後、種々の縮合剤を用い、アミノ酸を縮合する。このサイクルを繰り返すことで目的ペプチドを獲得する。縮合剤としてはカルボジイミド試薬、ジフェニルリン酸アジド、BOP試薬、HATU試薬などが用いられる。
本発明においては、固相合成は、例えば、N末端から13番目のD-Gln残基を起点に合成を行うことができる。すなわち、樹脂上のFmoc基を20% (v/v) ピペリジン/DMF溶液にて脱保護後、側鎖無保護のFmoc-D-Glu-Oallylを縮合する。脱保護と縮合を繰り返し、8番目のD-Val残基まで伸長する。伸長後、脱保護を行い、N(((9H-フルオレン-9-イル)メトキシ)カルボニル)-O-((2R)-2-(((アリルオキシ)カルボニル)アミノ)-3-メチルペンタノイル)-D-セリンを樹脂に導入する。樹脂上で環を形成するために、パラジウム触媒を用い、アリル基、Alloc(アリルオキシカルボニル)基を除去する。除去後、樹脂上で環を形成する。以降、Fmoc-D-Glu-Oallylの場合と同様にして順次C末端側から伸長を行うことで、目的ペプチドを得ることができる。
なお、この手法では、縮合させるアミノ酸を変更することにより様々な環状ペプチドが樹脂上で合成できる。例えば、N末端から13番目のD-Gln残基を他のアミノ酸に変更したい場合、樹脂との結合位置を13番目から10番目のD-Glnへ変更することで置換可能である。
本発明に係る細胞層透過促進剤は、上記化合物からなる、もしくは上記化合物を用いる。すなわち、後述する実施例で示されるように、上記化合物を細胞層に作用させるとその細胞接着構造に何らかの影響を与えて、細胞間隙経路の物質の透過性が亢進する。この場合、適用対象としては、具体的には、皮膚、肺、鼻、消化管、腸管、その他粘膜組織等が挙げられ、組織の種類に特に制限はない。また、細胞同士が単層に形成した形態であろうと、多層に形成した形態であろうと、その層形状の種類にも特に制限はない。また、ヒトを対象にするばかりではなく、ペット、家畜等の動物に由来する細胞層が対象であってもよい。更に、場合によっては、必要に応じて生体外に形成させた細胞層に適用してもよい。
一方、本発明に係る薬剤吸収補助用組成物は、上記化合物からなる細胞層透過促進剤を含み、生体への薬剤の吸収を補助するための組成物である。すなわち、この組成物で、適用の対象とする細胞層として、例えば、皮膚、肺、鼻、消化管、腸管、その他粘膜組織等を、生体に吸収させるべき所望の薬剤とともに処置することで、上記化合物が有効成分となって細胞間隙経路での物質等が亢進するので、その経路による所望の薬剤の生体への吸収率を向上させることができる。この場合、組成物の形態は、上記化合物を含んでいればよく、特に制限はないが、適用の対象とする細胞層の種類に応じて、適宜製剤的基材とともに当業者に知られた製剤手法により、例えば、軟膏、クリーム、ローション、乳液等の塗布剤、パッチ剤、貼付剤、エアロゾル剤、吸引剤、坐剤、トローチ剤、舌下錠剤、口腔内崩壊剤、ゼリー状剤、液剤、ソフトカプセル、ハードカプセル剤、粉末剤、顆粒剤、錠剤等の形態を採用し得る。組成物中の上記化合物の含有量としては、例えば0.0001~100質量%、より典型的には0.001~75質量%、更により典型的には0.01~50質量%、特に典型的には0.1~30質量%、特により典型的には1~20質量%等である。
本発明を適用する薬剤としては、親水性、親油性、もしくは両親媒性であってもよい低分子化合物、ペプチド、タンパク質、抗体、ワクチン、核酸など、特に制限されるものではないが、例えば、少なくとも分子量500以上、より好ましくは1000以上、更により好ましくは2000以上の物質に適用して、その生体への吸収率を向上して、その物質の薬剤としての適用可能性の範囲を広げ、また、注射投与ではなく、非侵襲的な、経口、経皮、経鼻、経肺、経粘膜などの投与方法の開発により、患者のQuality of Lifeの向上にも繋がるので、好ましい。
本発明による他に形態としては、上記薬剤吸収補助用組成物の形態中に、上記化合物と、生体に吸収させるべき所望の薬剤とを、その同一の形態中に含有せしめて、医薬組成物の形態と成してもよい。
本発明に係る上記化合物の投与量としては、適用の対象とする細胞層の種類や、生体に吸収させるべき薬剤の種類によっても異なり、一概ではないが、例えば、典型的には、細胞層の表面積1cm2当りへの1回適用量として0.01μg~10mg程度であってよく、より典型的には0.1~1mg程度であってよく、更により典型的には1μg~0.1mg程度であってよい。また、後述する実施例で示されるように、上記化合物による細胞間隙の開口作用は、従来知られたアクチン重合阻害剤のような不可逆的なものではなく、可逆的であり、処置後には、比較的速やかに、おそらくは生体恒常性維持作用によりタイトジャンクション等の細胞接着構造の正常性が回復するので、異物に対するバリア機能を損ねることなく安全に使用することが可能である。
以下実施例を挙げて本発明を具体的に説明するが、これらの実施例は本発明の範囲を限定するものではない。
[調製例1](cu538-1及びcu538-2の固相合成)
Fmoc固相合成法により、下記に示すcu538-1及びcu538-2を合成した。なお、これらの化合物は下記に化学構造式とともに矢印で示したOH基の位置に異性体が存在しラセミ体として得られ、これをHPLCにより分離精製することにより調製した。

具体的には、以下のとおり合成した。
PP 製濾過カラムにFmoc-NH-SAL Resin (0.37 mmol/g, 渡辺化学工業株式会社) 300 mg (0.111 mmol) を量りとり、ジメチルホルムアミド (DMF) 溶液中室温で40分間樹脂を膨潤させた。膨潤後、20% (v/v) ピペリジン/DMF溶液中、室温で20分間反応させることで樹脂上の保護基Fmoc(9-フルオレニルメトシキカルボニル) 基を除去した。DMFで10回洗浄し、1-ヒドロキシ-7-アザベンゾトリアゾール (HOAt, 0.333 mmol, 3 eq)、O-(7-アザ-1H-ベンゾトリアゾール-1-イル)-N, N, N', N' -テトラメチルウロニウムヘキサフルオロホルファート(HATU, 0.333 mmol, 3 eq)、N, N-ジイソプロピルアミン (DIPEA, 0.333 mmol, 3 eq)存在下で、Fmoc-D-Glu-Oallyl(0.333 mmol, 3 eq)を室温で30分間DMF中にて反応させ、樹脂上にアミノ酸を導入した。次のアミノ酸を縮合させるために、DMFで10回洗浄し、20% (v/v) ピペリジン/DMF溶液中、室温で20分間反応させることで樹脂上の保護基Fmoc 基を除去した。以下、Fmoc-D-Glu-Oallylの場合と同様にして順次C末端からFmoc-Leu-OH(0.333 mmol, 3 eq)、Fmoc-Leu-OH(0.333 mmol, 3 eq)、Fmoc-D-Gln(Trt)-OH(0.333 mmol, 3 eq)、Fmoc-D-Leu-OH(0.333 mmol, 3 eq)、Fmoc-D-Val-OH(0.333 mmol, 3 eq)を導入してペプチド鎖を伸長させた。20% (v/v) ピペリジン/DMF溶液中、室温で20分間反応させることで樹脂上のFmoc基を除去した。DMFで10回洗浄し、N, N'-ジイソプロピルカルボジイミド(DIPCI, 0.333 mmol, 3 eq)、1-ヒドロキシ-1H-ベンゾトリアゾールハイドレート(HOBt・H2O, 0.333 mmol, 3 eq)存在下で、N(((9H-フルオレン-9-イル)メトキシ)カルボニル)-O-((2R)-2-(((アリルオキシ)カルボニル)アミノ)-3-メチルペンタノイル)-D-セリン(0.333 mmol, 3 eq)を室温で90分間DCM(ジクロロメタン)中にて反応させ、樹脂上にアミノ酸を導入した。導入後、DMFで5回洗浄、メタノールで5回洗浄、ジエチルエーテルで5回洗浄後、樹脂を乾燥させた。乾燥樹脂に対して、テトラキス(トリフェニルホスフィン)パラジウム(0) (0.111 mmol, 1 eq)、フェニルシラン(1.11 mmol, 10 eq)存在下、室温で180分間DCM中にて反応させ、樹脂上のアリル基、Alloc(アリルオキシカルボニル)基を除去した。樹脂上で環を形成するため、DCMで5回洗浄、0.5% (w/v) ソディウムN,N-ジエチルジチオカルバメートトリハイドレート/DMF溶液で5回洗浄、DMFで5回洗浄し、N, N'-ジイソプロピルカルボジイミド(DIPCI, 0.555 mmol, 5 eq)、1-ヒドロキシ-1H-ベンゾトリアゾールハイドレート(HOBt・H2O, 0.555 mmol, 5 eq)存在下、室温で90分間DMF中にて環を形成した。樹脂上のFmoc基を除去するため、DMFで10回洗浄し、20% (v/v) ピペリジン/DMF溶液中、室温で5分間反応させることで樹脂上のFmoc 基を除去した。DMFで10回洗浄し、N, N'-ジイソプロピルカルボジイミド(DIPCI, 0.333 mmol, 3 eq)、1-ヒドロキシ-1H-ベンゾトリアゾールハイドレート(HOBt・H2O, 0.333 mmol, 3 eq)存在下で、Fmoc-Gln(Trt)-OH(0.333 mmol, 3 eq)を室温中90分間DMF中にて反応させ、樹脂上にアミノ酸を導入した。次のアミノ酸を縮合させるために、DMFで10回洗浄し、20% (v/v) ピペリジン/DMF溶液中、室温で20分間反応させることで樹脂上のFmoc 基を除去した。以下、Fmoc-D-Glu-Oallylの場合と同様にして順次C末端からFmoc-D-Leu-OH(0.333 mmol, 3 eq)、Fmoc-D-Val-OH(0.333 mmol, 3 eq)、Fmoc-D-Gln(Trt)-OH(0.333 mmol, 3 eq)、 Fmoc-D-Glu(OtBu)-OH(0.333 mmol, 3 eq)、 Fmoc-Leu-OH(0.333 mmol, 3 eq)、(±)-3-(tert-ブチルジメチルシロキシ)デカン酸(0.333 mmol, 3 eq)を導入し、DMFで5回洗浄、メタノールで5回洗浄、ジエチルエーテルで5回洗浄後、樹脂を乾燥させた。各種側鎖保護基の除去および脱樹脂のため、水(0.6 ml)存在下でトリフルオロ酢酸 (TFA) 11.4ml中にて3時間反応させた。反応液を濾過し、濾液を集め、窒素気流下でTFAを揮発させることにより留去し、ジエチルエーテルで洗浄し、デカンテーションにより溶液を除去した。得られた残渣を減圧下で乾燥し、70%(v/v)アセトニトリル水溶液に溶解し、高速液体クロマトグラフィーを用いて精製することにより、白色固体を得た(cu538-1 : 8.18 mg, 収率8%、cu538-2:7.75 mg, 収率 8%)。なお、得られた化合物のMS(質量)、及びcu538-2のNMRを分析したところ、下記のデータを得た。

PP 製濾過カラムにFmoc-NH-SAL Resin (0.37 mmol/g, 渡辺化学工業株式会社) 100 mg (0.037 mmol) を量りとり、ジメチルホルムアミド (DMF) 溶液中室温で40分間樹脂を膨潤させた。合成法はcu538-1,2と同様の手法を用い、N末端1残基目のロイシンまで伸長を行う。Fmoc基を除去後、N, N'-ジイソプロピルカルボジイミド(DIPCI, 0.111 mmol, 3 eq)、1-ヒドロキシ-1H-ベンゾトリアゾールハイドレート(HOBt・H2O, 0.111 mmol, 3 eq)存在下で、デカン酸(0.111 mmol, 3 eq)を導入し、DMFで5回洗浄後、メタノールで5回洗浄、ジエチルエーテルで5回洗浄後、樹脂を乾燥させた。各種側鎖保護基の除去および脱樹脂のため、水(0.2 ml)存在下でトリフルオロ酢酸 (TFA) 3.8 ml中にて3時間反応させた。反応液を濾過し、濾液を集め、窒素気流下でTFAを揮発させることにより留去し、ジエチルエーテルで洗浄し、デカンテーションにより溶液を除去した。得られた残渣を減圧下で乾燥し、70%(v/v)アセトニトリル水溶液に溶解し、高速液体クロマトグラフィーを用いて精製することにより、白色固体を得た(2.6 mg, 収率 4%)。なお、得られた化合物のMS(質量)を分析したところ、下記のデータを得た。

[調製例3](cu592の固相合成)
調製例1と同様にして、Fmoc固相合成法により、下記に示すcu592を合成した。

具体的には、以下のとおり合成した。

イヌ腎臓上皮細胞由来のMDCK II細胞を使用して、トランスウェルを用いた上皮細胞層のモデルを作成し、物質透過性評価試験を行った。具体的には、対数増殖期のMDCK II細胞を、3.4~4.0×104 cellsの細胞数でトランスウェル(6.5mm径、コラーゲンコート、孔サイズ0.4μm)(商品名「corning transwell 3495」、コーニング社製)に播種し、基底部側ウェルとして24穴ウェルプレートを使用して、頂端側のウェルと基底部側のウェルの各ウェルに細胞生育培地600μLを入れた。毎日培地交換して層形成させ、播種から3日後に培地を測定用の Hank's balanced salt solution(HBSS)に交換して、37℃で1時間インキュベート後、層形成を、抵抗値測定システム(商品名「Millicell-ERS」、ミリポア社製)を使用して経上皮電気抵抗(TEER)を測定することにより確認した。その層形成の確認後、頂端側の培地を、分子量4000のマーカー物質(FD4:蛍光標識デキストラン)(シグマ社製)を1.0w/v%の濃度となるよう添加したHBSSの100μLに置換し、各試験物質を下記に示す最終濃度となるよう添加した。マーカー物質の添加後5時間まで30分間30分間おきにトランスウェルを新しいHBSSが入った基底部側ウェルに移し、残った基底部側ウェルのHBSS(以下「レシーバー溶液」という。)は、その50μLずつを96穴ウェルプレートの1ウェルに回収した。その回収したHBSSについて、蛍光プレートリーダー(商品名「Wallac 1420 ARVO MX」、パーキンエルマー社製)を使用して、励起波長485nm、蛍光波長528nmで蛍光強度を測定した。

結果を図1に示す。
図1に示されるように、陰性コントロールでは、分子量4000のマーカー物質であるFD4のレシーバー溶液への透過は限定的であった。一方、陽性コントロールとして、アクチン重合阻害剤であるLatrunclin A(LatA)を添加すると、試験開始5時間にわたり、継続的なマーカー物質のレシーバー溶液への透過量の増加・蓄積がみられた。アクチン重合阻害により、細胞接着構造を不可逆的に緩和することが知られている。
これに対し、「cu538-1」や「NY-9」の添加により、試験開始2時間の時点で、陰性コントロールに比べてほぼ4倍、「cu538-2」の添加では、陰性コントロールに比べてほぼ8倍、レシーバー溶液への透過量が増加した。ただし、試験開始2時間以降5時間までに、レシーバー溶液への透過量の増加・蓄積がほとんどみられなかった。
これは、「cu538-1」や「cu538-2」や「NY-38」の添加により、細胞間隙経路の物質透過の亢進が起こるものの、その効果は可逆的、すなわちタイトジャンクション等の細胞接着構造の正常性の回復が速やかで、それ以上の細胞間隙経路の物質透過量の増加・蓄積がみられなかったものと考えられた。

図2に示されるように、「NY-38」にも細胞層透過促進の効果が認められた。その効果は、試験例1において細胞層透過促進の効果が明らかにされた「cu538-2」よりも顕著であった。

Claims (4)
- 下記一般式(1)で表わされる化合物からなる細胞層透過促進剤。
Xは、ロイシン又はその保存的置換、より好ましくは、ロイシン、イソロイシン、ノルロイシン、tert-ブチルアラニン、tert-ロイシン、バリン、シクロへキシルグリシン、又はシクロヘキシルアラニン、又はアラニンであり、
XXは、バリン又はその保存的置換、より好ましくは、バリン、イソロイシン、ノルロイシン、tert-ブチルアラニン、tert-ロイシン、ロイシン、シクロへキシルグリシン、又はシクロヘキシルアラニン、又はアラニンであり、
Yは、グルタミン酸又はその保存的置換、より好ましくは、グルタミン酸又はアスパラギン酸、又はアラニンであり、
Zは、グルタミン又はその保存的置換、より好ましくは、グルタミン又はアスパラギン、又はアラニンであり、
ZZは、任意のα-アミノ酸、より好ましくは、グルタミン又はアラニンであり、
YZは、セリン、スレオニン、ホモセリン、ジアミノプロパノイックアシッド、又はジアミノブチリックアシッドであり、
R’は、sec-ブチル又はその保存的置換、より好ましくは、sec-ブチル、イソプロピル、イソペンチル、n-ブチル、tert-ブチル、シクロへキシル、又はシクロへキシルメチルである。) - 請求項1又は2に記載の細胞層透過促進剤を含み、生体への薬剤の吸収を補助するためのものである、薬剤吸収補助用組成物。
- 請求項1又は2に記載の細胞層透過促進剤を含み、生体に吸収させるべき薬剤を更に含む、医薬組成物。
