Abstract
測定条件下における還元型ピロロキノリンキノンの安定性を維持した状態で定量分析を可能とする高速液体クロマトグラフィー法を提供することを目的とする。前記目的は次の方法により達成することができる。試料から還元型ピロロキノリンキノン又はその塩を含む高速液体クロマトグラフィー用サンプルを調製するサンプル調製工程と、固定相として逆相カラムを用い、移動相として、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む溶離液を用いる高速液体クロマトグラフィー法により、前記高速液体クロマトグラフィー用サンプルに含まれる前記還元型ピロロキノリンキノン又はその塩を前記固定相及び前記移動相との相互作用を利用して前記試料から分離する分離工程と、を有する、高速液体クロマトグラフィー分析方法。
Description
本発明は、酸化型および還元型ピロロキノリンキノン又はその塩の定量分析方法に関する。
ピロロキノリンキノン(Pyrroloquinoline quinone以下、単に「PQQ」ということがある)は、ピロール環とキノリン環が縮合したものがo-キノン構造をとる物質である。この物質は酸化型PQQと言われることがある。このキノン構造は容易に還元され、還元型ピロロキノリンキノンになる。また、還元型PQQは空気による酸化で酸化型PQQに変化する。このように酸化型ピロロキノリンキノンと還元型ピロロキノリンキノンの間で酸化還元が容易におこなわれることが知られている。この酸化型PQQと還元型PQQは異なるUVスペクトルであることが知られている(非特許文献1)。
またPQQは、細胞の増殖促進作用、抗白内障作用、肝臓疾患予防・治療作用、創傷治癒促進作用、抗アレルギー作用、逆転写酵素阻害作用、グリオキシラーゼI阻害作用および制癌作用などの多くの重要な生理活性を有するとされ、PQQ利用の産業上の重要性が高まっている。実際、食品としてピロロキノリンキノンジナトリウムが使用されている(特許文献1)。
機能性を利用する機能性表示食品制度では食品中のPQQの正確な測定データを算出する必要が求められている。多くの食品では単独での製品はなく、色々な物質と混合して提供される。一方でPQQは多くの食品成分と反応しやすく、分析時に妨害を受けやすい。また、分析スケールはカプセルや錠剤に含まれるmgオーダーの含有量の分析である。
これまでにPQQの分析方法は酸化型PQQを対象とする分析方法が使われてきた。高速液体クロマトグラフィー(HPLC)は不揮発性の物質を分析する方法であり、高感度分析も開発されている。しかし、酸化型PQQと還元型PQQの定量に適した分析方法は報告されてこなかった。また、酸化型PQQと還元型PQQの分離について詳しい報告はされていない。
ピロロキノリンキノンは還元性物質と反応することで還元型ピロロキノリンキノンに容易に変化する。この分析時の妨害を抑制する方法については非特許文献1に報告されている。この方法はアスコルビン酸による妨害を抑制するために、測定前に空気酸化によるアスコルビン酸を除去する工程を経た後にHPLC分析をする方法である。この分析条件ではピロキノリンキノンの酸化型と還元型の分離はできていないために還元剤であるアスコルビン酸を除去し、すべてを酸化型として検出する方法である。
一方、アスコルビン酸に代表される還元剤は色々な食品に配合されている。特に飲料では酸味料としての役割も合わさって、大量に配合されている。
このような食品にPQQが同時に配合された場合、還元剤の量が多く、空気酸化で除去する方法では長時間の処理が必要か、分析できない可能性が高い。そのため、還元型PQQを直接分析可能にして、正確な配合量を算出する分析方法が望まれてきた。
また、酸化型PQQと還元型PQQの分離を行うことで混合物内での状態を知る方法が望まれてきた。さらには定量を行うためにすべてを還元型PQQに変換させる前処理方法も望まれている。

従来法では、還元型ピロロキノリンキノンは空気(液体に含まれる溶存酸素を含む)により酸化されるなどその測定条件下における安定性に問題がある。そのため、還元型ピロロキノリンキノンの正確な定量方法は確立されていなかった。
仮に、還元型ピロロキノリンキノンを測定条件下において安定させることができ、その量を定量することが可能となれば、HPLCを用いた方法において還元型PQQと酸化型PQQを分離でき、酸化型と還元型の正確な定量可能な方法を確立することができる。また、これに加えて、試料に含まれる酸化型と還元型のピロロキノリンキノンの総量を知りたい場合には、すべてを還元型にして測定する新たな方法を確立することも可能となる。
本発明は上記問題点に鑑みてなされたものであり、測定条件下における還元型ピロロキノリンキノンの安定性を維持した状態で定量分析を可能とする高速液体クロマトグラフィー法を提供することを目的とする。
また、これにより、飲料のようなmgオーダーのPQQとその他の多数の共存成分を含む測定対象物を測定対象とした際に、共存成分による影響を受けることなく、当該測定対象物に含まれるPQQを迅速かつ簡便に定量することができる。
本発明者らは、上記課題を解決するために鋭意検討した結果、固定相として逆相カラムを用い、移動相として所定の溶離液を用いることにより、上記課題を解決し得ることを見出して、本発明を完成するに至った。

本発明によれば、測定条件下における還元型ピロロキノリンキノンの安定性を維持した状態で定量分析を可能とする高速液体クロマトグラフィー法を提供することができる。
またこれにより、飲料のようなPQQとその他の多数の共存成分を含む測定対象物を測定対象とした際に、共存成分による影響を受けることなく、当該測定対象物に含まれるPQQを迅速かつ簡便に定量できる分析方法を提供することができる。特に還元性物質を含む場合、還元型PQQとして検出することで食品をはじめとして様々な製品におけるPQQの定量が広い濃度範囲で確立され、PQQ類に関連した健康食品や医薬品等の開発に資することができる。
以下、本発明の実施の形態(以下、「本実施形態」という。)について詳細に説明するが、本発明はこれに限定されるものではなく、その要旨を逸脱しない範囲で様々な変形が可能である。
〔分析方法〕
本実施形態の高速液体クロマトグラフィー分析方法は、試料から還元型ピロロキノリンキノン又はその塩を含むHPLC用サンプルを調製するサンプル調製工程と、固定相として逆相カラムを用い、移動相として、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む溶離液を用いる高速液体クロマトグラフィ法により、前記HPLC用サンプルに含まれる前記還元型ピロロキノリンキノン又はその塩を前記固定相及び前記移動相との相互作用を利用して前記試料から分離する分離工程と、を有する。
本実施形態の高速液体クロマトグラフィー分析方法は、試料から還元型ピロロキノリンキノン又はその塩を含むHPLC用サンプルを調製するサンプル調製工程と、固定相として逆相カラムを用い、移動相として、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む溶離液を用いる高速液体クロマトグラフィ法により、前記HPLC用サンプルに含まれる前記還元型ピロロキノリンキノン又はその塩を前記固定相及び前記移動相との相互作用を利用して前記試料から分離する分離工程と、を有する。
一般に、還元型ピロロキノリンキノンは空気酸化しやすい。分析中で還元型ピロロキノリンキノンの酸化が進行すると、正確な分析結果を得ることができない。これに対して、本実施形態の分析方法によれば、還元型ピロロキノリンキノンが測定中に酸化型に変換されることを抑制することができ、還元型ピロロキノリンキノンの安定性を保った状態で分析をすることが可能となる。
そのため、還元型ピロロキノリンキノンと酸化型ピロロキノリンキノンを含む試料の測定においては、すべてを還元型ピロロキノリンキノンに変換して測定することができる。これにより、一回の測定により試料に含まれるピロロキノリンキノンの総量を定量することが可能となる。
また、本実施形態においては、上記所定の移動相を用いることにより、還元型ピロロキノリンキノンと酸化型ピロロキノリンキノンの分離能が向上し、それぞれを分離して定量することも可能となる。
〔サンプル調製工程〕
サンプル調製工程は、試料から還元型ピロロキノリンキノン又はその塩を含むHPLC用サンプルを調製する工程である。なお、サンプル調製工程前には、必要に応じて、後述する前処理工程、還元処理工程、分液工程、及び遠心分離工程を行ってもよい。

(分析対象)
本実施形態の分析方法における分析対象となるピロロキノリンキノンを以下に示す。PQQには、下記式(1)で表される酸化型PQQと、下記式(2)で表される還元型PQQがある。通常、ピロロキノリンキノンと表示された場合は酸化型PQQを示す。食品としてはピロロキノリンキノンジナトリウムn水和物が販売されており、これも分析対象に含まれる。環境に応じて酸化型PQQが相対的に多い状態から還元型PQQが相対的に多い状態まで様々取りうる。例えば、溶液中では、還元が進みやすく還元型PQQが相対的に多く存在し、酸化環境下では酸化型PQQが相対的に多く存在する。この点、本実施形態の分析方法によれば、酸化型PQQ及び還元型PQQの総量を定量することができる。

また、分析対象となる上記ピロロキノリンキノンの塩としては、特に制限されないが、例えば、アルカリ金属及びアルカリ土類金属などの金属との塩;アンモニウムカチオン等の非金属との塩が挙げられる。特に、アルカリ金属塩の一種であるジナトリウム塩は、食品として多用されており、分析対象としては重要である。
(試料)
ピロロキノリンキノンを含む試料(測定対象物)としては、特に制限されないが、例えば、食品(飲料を含む)、医薬品、医薬部外品等が挙げられる。食品としては、例えば、一般食品、保険機能食品(例えば、特定保健用食品、機能性表示食品、及び栄養機能食品)等が挙げられる。より具体的な試料としては、例えば、カプセル剤、錠剤、散剤、顆粒剤などの経口投与用の薬剤及びサプリメント;飲料;ゼリー;グミ;レトルト食品などその他食品が挙げられる。試料については、上記の他、飲食品ではない化粧料、洗浄料、その他外用剤なども対象とすることができ、PQQを含む製品全般を対象とすることができる。
ピロロキノリンキノンを含む試料(測定対象物)としては、特に制限されないが、例えば、食品(飲料を含む)、医薬品、医薬部外品等が挙げられる。食品としては、例えば、一般食品、保険機能食品(例えば、特定保健用食品、機能性表示食品、及び栄養機能食品)等が挙げられる。より具体的な試料としては、例えば、カプセル剤、錠剤、散剤、顆粒剤などの経口投与用の薬剤及びサプリメント;飲料;ゼリー;グミ;レトルト食品などその他食品が挙げられる。試料については、上記の他、飲食品ではない化粧料、洗浄料、その他外用剤なども対象とすることができ、PQQを含む製品全般を対象とすることができる。
(前処理工程)
サンプル調製工程において、測定対象物の形態に応じた前処理工程を行ってもよい。例えば、測定対象物がカプセル剤であればカプセルを切断したり、錠剤であれば錠剤を粉砕したりするなど、サンプルが混合されやすいよう適切に処理することができる。

(還元処理工程)
サンプル調製工程の前には、試料に対して還元剤を添加する還元処理工程が行われてもよい。この際、試料が酸化型ピロロキノリンキノンを含む場合、試料に含まれるすべての酸化型ピロロキノリンキノンが還元型ピロロキノリンキノンに変換することが好ましい。これにより、試料に含まれる還元型ピロロキノリンキノンと酸化型ピロロキノリンキノンの総量を、一回の分析により定量すること可能となる。
サンプル調製工程の前には、試料に対して還元剤を添加する還元処理工程が行われてもよい。この際、試料が酸化型ピロロキノリンキノンを含む場合、試料に含まれるすべての酸化型ピロロキノリンキノンが還元型ピロロキノリンキノンに変換することが好ましい。これにより、試料に含まれる還元型ピロロキノリンキノンと酸化型ピロロキノリンキノンの総量を、一回の分析により定量すること可能となる。
また別の観点として、試料には、サンプル調製工程の前後又はその工程中において酸化型PQQと反応し、不特定の類縁体を形成する成分が含まれることがある。このような成分は、分析の妨害を起こし得る。特に、アミノ基含有物質はこのような妨害を起こしやすい傾向にある。より具体的には、必須アミノ酸、テアニン、オルニチン、タンパク含有物質が挙げられる。
このように、試料が酸化型PQQと反応し得る化合物を含む場合には、PQQを還元型にする操作を行うことができる。例えば、試料に対して還元剤を添加することにより試料中の酸化型PQQを還元型PQQにする還元処理工程が行われてもよい。本実施形態の分析方法における主な測定対象となる還元型PQQは酸化型PQQと異なり、アミノ酸等の任意成分と反応をしないという点で、安定性に優れる。これにより、HPLC用サンプル中に共存する任意成分の妨害を受けずに定量分析が可能となる。また、還元型PQQの液体クロマトグラフィーの溶出時間は、酸化型PQQと異なり、クロマトグラムのピークの分離も容易になる。
還元剤としては、特に制限されないが、例えば、アスコルビン酸などの従来公知の物質が挙げられる。還元剤を用いることにより、還元型PQQの生成が効率的及び優先的に進行し、上記妨害物質による影響を排することができる。還元剤として添加するアスコルビン酸の態様は、特に制限されず、水溶液であっても、粉末状であってもよい。水溶液である場合には、溶媒として水を用いても緩衝液を用いてもよい。
試料に対する還元剤の添加方法は、特に制限されないが、水又は緩衝液などの水溶液中で、ピロロキノリンキノン又はその塩とアスコルビン酸とを共存させる方法が挙げられる。当該方法により還元型PQQを生成する反応の温度と時間は、PQQ濃度や妨害物質濃度によって調整することができ、特に制限されない。
このなかでも、反応温度は、好ましくは0~120℃であり、より好ましくは10~90℃であり、さらに好ましくは20~80℃である。また、反応時間は、好ましくは5分~2日であり、より好ましくは10分~24時間であり、さらに好ましくは10分~10時間である。さらに、水溶液のpHは、好ましくは1~3であり、より好ましくは1.5~2.5である。
還元剤の使用量は、ピロロキノリンキノンに対して大過剰で使用することが好ましい。より具体的には、還元剤の使用量は、ピロロキノリンキノン1質量部に対して、好ましくは100~1000000質量部であり、より好ましくは500~500000質量部であり、さらに好ましくは1000~100000質量部である。
(分液工程)
また、試料には、サンプル調製工程においてPQQの抽出を阻害することにより分析の妨害を起こす可能性のある化合物も含まれ得る。このような化合物としては、特に制限されないが、例えば、脂溶性成分が挙げられる。脂溶性成分としては、より具体的には、食用油脂、非食用油脂、動物性油脂、及び植物性油脂等の油脂類;リポ酸;ドコサヘキサエン酸(DHA)、エイコサペンタエン酸(EPA),レシチン等の脂質;ビタミンE等の脂溶性ビタミン類及びその誘導体;グリセリルホスホリルコリン、ステアリン酸マグネシウムが挙げられる。このなかでも、油脂は妨害効果が高く、定量化の妨げになりやすい。
また、試料には、サンプル調製工程においてPQQの抽出を阻害することにより分析の妨害を起こす可能性のある化合物も含まれ得る。このような化合物としては、特に制限されないが、例えば、脂溶性成分が挙げられる。脂溶性成分としては、より具体的には、食用油脂、非食用油脂、動物性油脂、及び植物性油脂等の油脂類;リポ酸;ドコサヘキサエン酸(DHA)、エイコサペンタエン酸(EPA),レシチン等の脂質;ビタミンE等の脂溶性ビタミン類及びその誘導体;グリセリルホスホリルコリン、ステアリン酸マグネシウムが挙げられる。このなかでも、油脂は妨害効果が高く、定量化の妨げになりやすい。
このように試料が脂溶性成分を含む場合には、これを除去する操作を行うことができる。例えば、水と相溶しない有機溶剤をさらに混合し、測定対象物中の脂溶性成分を有機溶剤に溶解させた後、有機溶剤を取り除くことにより、測定対象物中の脂溶性成分が除去されたHPLC用サンプルを調製する分液工程を行ってもよい。特に、測定対象物が脂溶性成分を含む場合には、分液操作を行うことにより、有機層に脂溶性成分を抽出し、水層にPQQ(酸化型または還元型)を抽出することで、PQQ(酸化型または還元型)の定量精度がより向上する傾向にある。
分液処理において用い得る有機溶媒としては、特に制限されないが、例えば、酢酸エチル、クロロホルム、塩化メチレン、シクロヘキサン、トルエン、ヘキサン、ジメチルホルムアミド、ジメチルスルホキシド、アセトンが挙げられる。このなかでも、水溶性が低いという観点から、酢酸エチル、クロロホルム、塩化メチレンが好ましく、より好ましくは生分解性の高い酢酸エチルである。なお、分液操作は脂溶性成分を含まないと考えられる場合には、省略することも可能である。分液操作を行うことにより、PQQの定量精度がより向上する傾向にある。
(遠心分離工程)
また、サンプル調製工程においてPQQの抽出を阻害するその他の物質として、粉末成分が挙げられる。粉末成分としては、特に制限されないが、例えば、シリカ等の無機粉体;コメ粉等の有機粉体などが挙げられる。

このように試料が粉末成分を含む場合には、これを除去する操作を行うことができる。例えば、測定対象物が、難溶性成分等の粉末成分を含む場合には、遠心分離操作を行ってもよい(遠心分離工程)。また、分液操作の過程で水層と有機層を分離させるために遠心分離操作を利用してもよい。
(希釈工程)
HPLC用サンプルには、PQQあるいはその他の成分の濃度をHPLC測定に適した濃度に調整するために、希釈剤を加えてもよい。希釈剤としては、特に制限されないが、例えば、メタノールやアセトニトリルなど水と混和する有機溶媒や、水が挙げられる。また、希釈剤としてはこれらの混合溶媒を用いてもよい。
HPLC用サンプルには、PQQあるいはその他の成分の濃度をHPLC測定に適した濃度に調整するために、希釈剤を加えてもよい。希釈剤としては、特に制限されないが、例えば、メタノールやアセトニトリルなど水と混和する有機溶媒や、水が挙げられる。また、希釈剤としてはこれらの混合溶媒を用いてもよい。
希釈剤が水を含む場合、酸性が好ましい。また、希釈剤の添加時にアスコルビン酸等の還元剤をさらに添加してもよい。これにより、還元型PQQをより安定に保つことができる。
さらに、希釈剤としては、HPLC溶離液を用いてもよい。これにより、HPLCに導入するサンプル組成とHPLC溶離液の組成との相違が少なくなる。そのため、ショックピーク、ピーク形状の乱れ、溶出時間の変動を抑止することができる。
(HPLC用サンプル)
上記のようにして得られるHPLC用サンプルは、酸化型ピロロキノリンキノン、還元型ピロロキノリンキノン、又はその塩とアスコルビン酸とが反応して得られた還元型PQQ又はその塩と、未反応のアスコルビン酸と、測定対象物に含まれるその他の各成分が含まれ得る。
上記のようにして得られるHPLC用サンプルは、酸化型ピロロキノリンキノン、還元型ピロロキノリンキノン、又はその塩とアスコルビン酸とが反応して得られた還元型PQQ又はその塩と、未反応のアスコルビン酸と、測定対象物に含まれるその他の各成分が含まれ得る。
HPLC用サンプルに含まれるアスコルビン酸の含有量は、好ましくは1~40質量%であり、より好ましくは3~20質量%であり、さらに好ましくは3~10質量%である。
〔分離工程〕
分離工程は、固定相として逆相カラムを用い、移動相として、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む溶離液を用いる高速液体クロマトグラフィ法により、HPLC用サンプルに含まれる還元型ピロロキノリンキノン又はその塩を固定相及び移動相との相互作用を利用して試料から分離する工程である。
分離工程は、固定相として逆相カラムを用い、移動相として、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む溶離液を用いる高速液体クロマトグラフィ法により、HPLC用サンプルに含まれる還元型ピロロキノリンキノン又はその塩を固定相及び移動相との相互作用を利用して試料から分離する工程である。
これにより、HPLC用サンプルに含まれる還元型PQQおよび酸化型PQQを、それぞれ定量することができる。また、還元工程を経る場合には、試料に含まれる還元型PQQおよび酸化型PQQの総量を定量することができる。
なお、定量方法としては、特に制限されないが、例えば、標準添加法、内部標準法、絶対検量線法が挙げられる。また、定量法は求める精度と補正を考慮して採用すればよい。
本実施形態において用い得る標準添加法は、常法と同様であり、例えば、酸化型PQQもしくは還元型PQQ及びその塩を検出可能な検出器を用いて、HPLC用サンプルのクロマトグラムを得て、当該ピークの面積に基づき予め定めた検量線から、HPLC用サンプルに含まれるイミダゾピロロキノリン及びその塩を定量する方法が挙げられる。検量線の作成にあっては、初めに、濃度が既知の標準溶液を予め調製し、当該標準溶液をHPLC用サンプルに添加して、検量線作成用のサンプル群を調製する。
本実施形態に高速液体クロマトグラフィー(HPLC)等の液体クロマトグラフィー(LC)が挙げられる。HPLC装置は、分離カラム、および分離溶液を分離カラムに送り込むポンプを備える。HPLC装置は、それ以外の要素、例えば、オートサンプラー、ヒーター、分離された成分を検出する検出器等備えていてもよい。検出器としては、例えば、UV検出器や蛍光検出器、質量分析装置が挙げられる。
(固定相)
固定相となる分離カラムとしては、逆相カラムを用いる。逆相カラムとしては、例えば、オクタデシルシリル化シリカゲル充填剤を充填したカラム(オクタデシルシリル(ODS)カラム)、オクチル基を導入したシリカゲルを充填したカラム(C8カラム)、エチル基を導入したシリカゲルを充填したカラム(C2カラム)、これらにイオン交換樹脂を配合したカラムが挙げられるが、特にODSカラムが好ましい。
固定相となる分離カラムとしては、逆相カラムを用いる。逆相カラムとしては、例えば、オクタデシルシリル化シリカゲル充填剤を充填したカラム(オクタデシルシリル(ODS)カラム)、オクチル基を導入したシリカゲルを充填したカラム(C8カラム)、エチル基を導入したシリカゲルを充填したカラム(C2カラム)、これらにイオン交換樹脂を配合したカラムが挙げられるが、特にODSカラムが好ましい。
充填剤の粒径は、好ましくは5.0μm以下であり、より好ましくは1.7~5.0μmである。充填剤の粒径が上記範囲内であることにより、分離能がより向上する傾向にある。
(移動相)
移動相となる溶離液は、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む。このような溶離液を用いることにより、還元型PQQを安定な状態で測定することができ、また、酸化型PQQと還元型PQQを分離することも可能となる。
移動相となる溶離液は、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む。このような溶離液を用いることにより、還元型PQQを安定な状態で測定することができ、また、酸化型PQQと還元型PQQを分離することも可能となる。
溶離液は、リン酸又は塩酸の一方、あるいは、リン酸及び塩酸の両方を含む。「0.050~1.5質量%のリン酸及び/又は塩酸」とは、溶離液がリン酸又は塩酸の一方を含む場合にはその濃度をいい、溶離液がリン酸及び塩酸の両方を含む場合には、その総量をいう。これを踏まえて、リン酸及び/又は塩酸の濃度は、0.050~1.5質量%であり、好ましくは0.085~1.5質量%であり、より好ましくは0.1~1.5質量%であり、さらに好ましくは0.2~1質量%である。リン酸及び/又は塩酸の濃度が上記範囲内であることにより、分離能がより向上し、還元型PQQを安定な状態で測定することができ、また、酸化型PQQと還元型PQQを分離することも可能となる。なお、リン酸及び塩酸に加えて、その他の酸性分を添加してもよい。
溶離液は、メタノール又はアセトニトリルの一方、あるいは、メタノール及びアセトニトリルの両方を含む。「20~50体積%のメタノール及び/又はアセトニトリル」とは、溶離液がメタノール又はアセトニトリルの一方を含む場合にはその濃度をいい、溶離液がメタノール及びアセトニトリルの両方を含む場合には、その総量をいう。これを踏まえて、メタノール及び/又はアセトニトリルの含有量は、20~50体積%であり、好ましくは25~45体積%であり、より好ましくは27.5~42.5体積%である。また、メタノール及び/又はアセトニトリルの含有量が、30~50体積%、30~45体積%、又は30~42.5体積%であることも好ましい。メタノール及び/又はアセトニトリルの含有量が上記範囲内であることにより、分離能がより向上し、還元型PQQを安定な状態で測定することができ、また、酸化型PQQと還元型PQQを分離することも可能となる。なお、メタノール及びアセトニトリルに加えて、その他の有機溶剤を添加してもよい。
溶離液のpHは、好ましくは2.8以下であり、より好ましくは2.3以下であり、さらに好ましくは2.1以下である。溶離液のpHが上記範囲内にあることにより、分離能がより向上し、還元型PQQを安定な状態で測定することができ、また、酸化型PQQと還元型PQQを分離することも可能となる。
溶出法としては、特に制限されないが、例えば、送液中において移動相(溶離液)の組成を変化させないアイソクラティック溶出法、及び、送液中において移動相(溶離液)の組成を変化させるグラジエント溶出法が挙げられる。溶出法は、分離能に応じて適宜選択することができる。
本実施形態の分析方法は、PQQ定量分析において以下の利点、特徴を有する。第一に、アスコルビン酸のような還元性物質が入った場合にも酸化型PQQと還元型PQQを検出することができる。第二に、すべてを還元型PQQとして検出することで定量計算が容易になる。また、酸化型PQQを還元型PQQに変換するアスコルビン酸は安価で安全である。
以下、本発明を実施例及び比較例を用いてより具体的に説明する。本発明は、以下の実施例によって何ら限定されるものではない。
以下に分析に用いた各試料の調製方法を記載する。なお、実施例において用いた試薬は、特に記載がない限り和光特級の試薬を用いた。ピロロキノリンキノンジナトリウムは三菱瓦斯化学株式会社製BioPQQ(登録商標)を使用した。
〔実施例1:酸化型PQQと還元型PQQ分離条件〕
(溶離液1)
実施例1で使用した溶離液は、水とメタノールを体積比73:27で混合し、その混合液に対して、リン酸を0.34質量%となるように添加したものである。

ピロロキノリンキノンジナトリウム水溶液(40mg/L)とアスコルビン酸水溶液(2g/L)を混合して分析サンプル1を調製した。分析サンプル1の調製から120分以内に、上記したHPLC分析条件で分析操作を行った。そのクロマトグラムを図1に示す。

また、別途、アスコルビン酸水溶液単独、十分に空気に曝して酸化型としたピロロキノリンキノンジナトリウム水溶液単独をサンプルとして用いて、上記したHPLC分析条件で分析操作を行って、それぞれクロマトグラムを得た。アスコルビン酸水溶液単独をサンプルとした場合のクロマトグラムから、アスコルビン酸の溶出時間が1.1分であることが分かった。また、十分に空気に曝したピロロキノリンキノンジナトリウム水溶液単独をサンプルとした場合のクロマトグラムから、酸化型ピロロキノリンキノンジナトリウムの溶出時間が10.2分であることが分かった。
分析サンプル1を用いて得られたクロマトグラムのなかから、アスコルビン酸と酸化型ピロロキノリンキノンジナトリウムの溶出時間に対応するピークを特定するとともに、アスコルビン酸と酸化型ピロロキノリンキノンジナトリウムの溶出時間以外のピークが還元型ピロロキノリンキノンジナトリウムに由来するものであると同定した。ここで、還元型ピロロキノリンキノンジナトリウムの溶出時間は、17.4分であった。
さらに、上記のように同定した各成分の溶出時間が、それぞれアスコルビン酸と、酸化型ピロロキノリンキノンジナトリウムと、還元型ピロロキノリンキノンジナトリウムとを示すものであることを別の観点から確認するために、上記検出器UVに加えて、フォトダイオードアレイ分析により、各溶出時間におけるUV-VISの吸収を測定した。
具体的には、HPLC分析装置で分離しながら、送液を止めることなく逆相カラムから流出する溶離液のスペクトルを測定した。溶出時間10.2分の時のUV-VISの吸収スペクトルを図2に示し、溶出時間17.4分の時のUV-VISの吸収スペクトルを図3に示す。
図2では、270nmに最大吸収を示すスペクトルが得られ、図3では、320nmに最大吸収を示すスペクトルが得られた。このスペクトルの特徴は、これまで報告されてきた酸化型ピロロキノリンキノンジナトリウムと還元型ピロロキノリンキノンジナトリウムのスペクトルの特徴と一致した(文献(Itohら非特許文献1))。これにより、溶出時間10.2分におけるピークが酸化型ピロロキノリンキノンジナトリウムに由来するものであり、溶出時間17.4分におけるピークが還元型ピロロキノリンキノンジナトリウムに由来するものであることが改めて確認できた。
〔比較例1:既存の分析方法〕
(溶離液2:HPLC溶離液(100mM CH3COOH/100mM CH3COONH4=30/70(pH5.1))
6.0gのCH3COOHを蒸留水で溶解し、総量が1Lとなるようにメスアップして100mM CH3COOH(1液)を調製し、これとは別に、7.71gのCH3COONH4を蒸留水で溶解し、総量が1Lとなるようにメスアップして100mM CH3COONH4(2液)を調製した。その後、300mLの1液と700mLの2液を混合して、HPLC溶離液を得た。なお、得られた緩衝液のpHが5.1±0.2であることを確認した。
(溶離液2:HPLC溶離液(100mM CH3COOH/100mM CH3COONH4=30/70(pH5.1))
6.0gのCH3COOHを蒸留水で溶解し、総量が1Lとなるようにメスアップして100mM CH3COOH(1液)を調製し、これとは別に、7.71gのCH3COONH4を蒸留水で溶解し、総量が1Lとなるようにメスアップして100mM CH3COONH4(2液)を調製した。その後、300mLの1液と700mLの2液を混合して、HPLC溶離液を得た。なお、得られた緩衝液のpHが5.1±0.2であることを確認した。

上記と同様にして分析サンプル1を調製した。分析サンプル1の調製から120分以内に、上記したHPLC分析条件で分析操作を行い、クロマトグラムを得た。また、別途、アスコルビン酸水溶液単独、十分に空気に曝したピロロキノリンキノンジナトリウム水溶液単独をサンプルとして用いて、上記したHPLC分析条件で分析操作を行って、それぞれクロマトグラムを得た。
アスコルビン酸水溶液単独をサンプルとした場合のクロマトグラムから、アスコルビン酸の溶出時間が1.1分であることが分かった。また、十分に空気に曝したピロロキノリンキノンジナトリウム水溶液単独をサンプルとした場合のクロマトグラムから、酸化型ピロロキノリンキノンジナトリウムの溶出時間が2.1分であることが分かった。
分析サンプル1を用いて得られたクロマトグラムでは、溶出時間1.1分と2.1分の二か所においてピークが見られ、それ以上のピークは確認できなかった。この結果から、比較例1の分析条件では、酸化型ピロロキノリンキノンジナトリウムと、還元型ピロロキノリンキノンジナトリウムとは分離できなかったことが確認できた。
〔実施例2~7、比較例2〕
下記表1に示すように使用したカラムと溶離液を変更したこと以外は、実施例1の分析条件と同様にして分析を行い、分析サンプル1中の酸化型ピロロキノリンキノンジナトリウムと還元型ピロロキノリンキノンジナトリウムが分離できるか否かを確認した。

表1に示すように、溶離液のリン酸をギ酸に変更した比較例2では、酸化型ピロロキノリンキノンジナトリウム、及び還元型ピロロキノリンキノンジナトリウムが分離できないことが分かった。
〔実施例8~16:前処理工程を含む分析〕
酸化型ピロロキノリンキノンジナトリウム(100mg/L)を含む水溶液と、表2に記載の前処理液とを混合して、所定の条件で放置し(前処理工程)、各分析サンプルを調製した。得られた各分析サンプルを用いて、下記HPLC分析条件で分析を行った。

〔実施例17~27:還元型PQQの安定性〕
酸化型ピロロキノリンキノンジナトリウム(100mg/L又は500mg/L)を含む水溶液と、表2に記載の前処理液とを混合して、50℃で1時間放置し(前処理工程)、各分析サンプルを調製した。得られた各分析サンプルを用いて、下記HPLC分析条件で分析を行った。24時間後以上、還元型PQQの析出を確認したあと、分析した。
酸化型ピロロキノリンキノンジナトリウム(100mg/L又は500mg/L)を含む水溶液と、表2に記載の前処理液とを混合して、50℃で1時間放置し(前処理工程)、各分析サンプルを調製した。得られた各分析サンプルを用いて、下記HPLC分析条件で分析を行った。24時間後以上、還元型PQQの析出を確認したあと、分析した。

以上のとおり、実施例25~27によれば、還元型ピロロキノリンキノンは析出しやすく、アスコルビン酸単独では長時間保存すると分析サンプルから析出してしまう危険性があった。これに対して、実施例17~24に示される通り、シクロデキストリンまたは有機溶媒を使用することにより、還元型ピロロキノリンキノンを溶液中で安定化できることが分かった。
〔定量性実験1〕
ピロロキノリンキノンジナトリウムを20mg/L含む水溶液、ピロロキノリンキノンジナトリウムを40mg/L含む水溶液、及びピロロキノリンキノンジナトリウムを80mg/L含む水溶液を準備した。これら水溶液に、それぞれ、前処理液として10質量%アスコルビン酸水溶液を混合し、常温(23℃)で1時間放置することで、3つの分析サンプルを調製した。その後、下記HPLC分析条件で分析を行った。
ピロロキノリンキノンジナトリウムを20mg/L含む水溶液、ピロロキノリンキノンジナトリウムを40mg/L含む水溶液、及びピロロキノリンキノンジナトリウムを80mg/L含む水溶液を準備した。これら水溶液に、それぞれ、前処理液として10質量%アスコルビン酸水溶液を混合し、常温(23℃)で1時間放置することで、3つの分析サンプルを調製した。その後、下記HPLC分析条件で分析を行った。

3つの分析サンプルそれぞれについて得られたクロマトグラムに基づいて、還元型ピロロキノリンキノンジナトリウムの溶出時間4.0に現れるピーク面積を算出した。この際、還元型ピロロキノリンキノンジナトリウムの溶出時間4.0に現れるピーク面積は、検出器UVの259nmの波長の吸収により得られたクロマトグラムと、320nmの波長の吸収により得られたクロマトグラムの両方で算出した。
259nmの波長の吸収により得られたクロマトグラムに基づくピーク面積を縦軸とし、分析サンプルの調製において用いたピロロキノリンキノンジナトリウムの濃度を横軸として、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度との関係を図4に示す。また、同様に、320nmの波長の吸収により得られたクロマトグラムにおいても、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度との関係を図4に示す。
直線Aは259nmの波長の吸収により得られたクロマトグラムに基づく、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度との関係を示す直線であり、直線Bは320nmの波長の吸収により得られたクロマトグラムに基づく、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度との関係を示す直線である。
〔定量性実験2〕
さらに、異なる分析条件においても定量性が確認できるか否かを確認した。まず、ピロロキノリンキノンジナトリウムを20mg/L含む水溶液、ピロロキノリンキノンジナトリウムを40mg/L含む水溶液、ピロロキノリンキノンジナトリウムを60mg/L含む水溶液、及びピロロキノリンキノンジナトリウムを80mg/L含む水溶液を準備した。これら水溶液に、それぞれ、前処理液としてアスコルビン酸2質量%及びγ-シクロデキストリン2質量%を含む水溶液を混合し、50℃で1時間放置することで、4つの分析サンプルを調製した。その後、下記HPLC分析条件で分析を行った。
さらに、異なる分析条件においても定量性が確認できるか否かを確認した。まず、ピロロキノリンキノンジナトリウムを20mg/L含む水溶液、ピロロキノリンキノンジナトリウムを40mg/L含む水溶液、ピロロキノリンキノンジナトリウムを60mg/L含む水溶液、及びピロロキノリンキノンジナトリウムを80mg/L含む水溶液を準備した。これら水溶液に、それぞれ、前処理液としてアスコルビン酸2質量%及びγ-シクロデキストリン2質量%を含む水溶液を混合し、50℃で1時間放置することで、4つの分析サンプルを調製した。その後、下記HPLC分析条件で分析を行った。

4つの分析サンプルそれぞれについて得られたクロマトグラムに基づいて、還元型ピロロキノリンキノンジナトリウムの溶出時間9.7に現れるピーク面積を算出した。この際、還元型ピロロキノリンキノンジナトリウムの溶出時間9.7に現れるピーク面積は、320nmの波長の吸収により得られたクロマトグラムで算出した。
320nmの波長の吸収により得られたクロマトグラムに基づくピーク面積を縦軸とし、分析サンプルの調製において用いたピロロキノリンキノンジナトリウムの濃度を横軸として、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度との関係を図5に示す。
直線は320nmの波長の吸収により得られたクロマトグラムに基づく、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度との関係を示す直線である。
図4~5に示されるように、ピーク面積と分析サンプル中のピロロキノリンキノンジナトリウムの濃度は直線性を有しており、これらはピーク面積からピロロキノリンキノンジナトリウムの濃度を求めるための良い検量線となりうることが分かった。なお、この検量線は、最小二乗法によるR2は0.999以上であり、濃度20から80mg/Lで高い相関性を維持することが確認できた。
〔実施例28〕
表4に示す組成を有する飲料500mLを調製した。得られた飲料1gに対して、前処理液としてアスコルビン酸2質量%及びγ-シクロデキストリン2質量%を含む水溶液1gを混合し、50℃で1時間放置することで、分析サンプルを調製した。

その結果、得られたクロマトグラムを図6に示す。本分析において、分析サンプルに含まれるピロロキノリンキノンの濃度は21.8mg/Lと算出された。また、定量性試験2において求めた検量線を用いて、ピーク面積から分析サンプルに含まれるピロロキノリンキノンの濃度を算出したところ21.6mg/Lと推定された。分析サンプルに実際に含まれるピロロキノリンキノンの濃度に対する、検量線により算出したピロロキノリンキノンの濃度の割合(回収率)は、99%であった。この分析結果により、定量性に優れた結果が得られることが分かった。
〔比較例7〕
比較例1の条件で上記の飲料を分析した。回収率は103%と算出された。また、酸化型PQQと還元型PQQの分離が完全に行われておらず、分析方法として適切でなかった。その結果を図7に示す。

〔実施例29〕

実施例29で使用した溶離液は、水とメタノールを体積比70:30で混合し、その混合液に対して、塩酸(36%塩酸=濃塩酸)を0.4質量%となるように添加したものである。

ピロロキノリンキノンジナトリウム水溶液(40mg/L)と、前処理としてアスコルビン酸2質量%及びγ-シクロデキストリン2質量%を含む水溶液を1:1で混合して分析サンプルを調製した。分析サンプルの調製から60分以内に、上記したHPLC分析条件で分析操作を行った。
ピロロキノリンキノンジナトリウム水溶液(40mg/L)と、前処理としてアスコルビン酸2質量%及びγ-シクロデキストリン2質量%を含む水溶液を1:1で混合して分析サンプルを調製した。分析サンプルの調製から60分以内に、上記したHPLC分析条件で分析操作を行った。
図8に前処理前のPQQ水溶液(40mg/L)のクロマトグラムを示す。PQQのピークを6.4分に検出した。また、図9に前記の前処理したPQQ水溶液の結果を示す。前処理液のアスコルビン酸は1.2分にピークを示し、還元型PQQ(20mg/L)のピークを18.9分に検出した。
またHPLC分析装置で分離しながら、送液を止めることなく逆相カラムから流出する溶離液のスペクトルを測定した。図8における溶出時間6.4分の時のUV-VISの吸収スペクトルを図10に示し、図9における溶出時間18.9分の時のUV-VISの吸収スペクトルを図11に示す。これにより、溶離液として塩酸を使用した場合にも酸化型と還元型PQQを分離できたことが示された。
Claims (6)
- 試料から還元型ピロロキノリンキノン又はその塩を含む高速液体クロマトグラフィー用サンプルを調製するサンプル調製工程と、
固定相として逆相カラムを用い、移動相として、0.050~1.5質量%のリン酸及び/又は塩酸と、20~50体積%のメタノール及び/又はアセトニトリルと、を含む溶離液を用いる高速液体クロマトグラフィー法により、前記高速液体クロマトグラフィー用サンプルに含まれる前記還元型ピロロキノリンキノン又はその塩を前記固定相及び前記移動相との相互作用を利用して前記試料から分離する分離工程と、を有する、
