Definitions
- This patent application deals with an unprecedented “MULTIFUNCTIONAL FLUORIDE DENTIFRICAL COMPOSITION” that describes a toothpaste formulation, which comprises a synergistic association of a small amount of water, a fluorinated agent and two phosphate compounds, which confer anti-demineralizing / regenerating action to the composition, which, in contact with the minerals of the tooth, prevents the loss of calcium and phosphate, which occurs during the demineralization process, while promoting the repair of already demineralized areas at an early stage, that is, at the enamel level.
- the composition also has dental desensitizing action, due to its CaF 2 precipitation capacity, which obliterates the dentinal tubules.
- the present invention belongs to the section of human needs, to the field of medical science, dentistry and hygiene; more specifically, dentistry preparations, for describing a toothpaste formulation, which comprises a synergistic association of its components, giving the composition anti-demineralizing, regenerating and desensitizing action.
- the current formulations add antimicrobial effects, anti-caries, anti-plaque, antidor, among others.
- dentifrice formulations in general, have the following components: humectants, thickeners, vehicle, sweeteners, preservatives, abrasives, surfactants, flavors, flavors and active ingredients. Being, exactly in the assets, the main differences between toothpastes, which are responsible for combating the main oral problems.
- Caries and dental erosion are the main problems faced, in the context of destructive processes of dental tissue, also called demineralization. To combat these problems, fluoride has become the main active agent incorporated.
- Some of these compounds are used as a source of calcium and phosphate, in order to increase the saturation of the oral environment in relation to hydroxyapatite, which includes: nanohydroxyapatite, bioactive glass containing sodium and calcium phosphosilicate (NovaMin ® ), a double chamber system in which calcium salts are separated by the plastic compartment of phosphate salts and sodium fluoride (Enamelon®), dehydrated dicalcium phosphate (DCPD), caseinophosphopeptides (CPP) linked to amorphous calcium phosphate (ACP) (Recaldent ® ), tricalcium phosphate functionalized and calcium glycerophosphate.
- hydroxyapatite which includes: nanohydroxyapatite, bioactive glass containing sodium and calcium phosphosilicate (NovaMin ® ), a double chamber system in which calcium salts are separated by the plastic compartment of phosphate salts and sodium fluoride (Enamelon®), dehydrated dicalcium phosphate (DCPD), caseino
- the present invention brings a formulation that fully meets the maintenance of oral health, solving the problem present in the current state of the art, with regard to the proposal of a dentifrice composition capable of promoting maintenance of complete oral health in a single product.
- the previous PI0705195-6 entitled “Acidized liquid toothpaste formulation with low fluoride concentration and its use”, describes a “liquid” toothpaste formulation (fluid gel), consisting of fluorine with a content less than or equal to 750 ppm, of 4 to 50% abrasives, 20 to 65% humectants, 0.5 to 7% binders, 1 to 6% surfactants, 0.01 to 0.5% preservatives, 0 to 750 ppm fluoridated agents, from 0.25 to 2% of antiplaque agents and acidulating agents in the proportion that guarantee pH of 4.3 to 6.0.
- the proposal of a low concentration of fluoride and reduced pH aims to increase the product's effectiveness in controlling dental caries and decrease the risk of dental fluorosis.
- the reaction that sustains the most acidic pH in the mouth, and that occurs in the invention occurs due to contact with water (saliva), which promotes the hydrolysis of the acid in the composition, forming hydronium, with a lower pH (- 1.7 pKa) than the acid itself (2.7 pKa).
- the previous BR102013006807-1 entitled “Ultrafunctional dentifrice and use of a combination of fluoride agent and protease inhibitor”, describes a dentifrice formulation containing fluoride and protease inhibitors, having a reduced pH, between 4.3 and 6; in order to increase the effectiveness of the product in the control of dental caries, dental erosion and periodontal disease.
- the pH of the present invention is more acidic and specifically adjusted so that the composition is capable of promoting remineralization and dentin desensitization.
- the present invention proposes to increase the fluoride concentration and a great reduction of water, which promotes the increase of the acid to correct the pH, which keeps it around 4.5.
- the present invention has a high concentration of fluorine (1450 ppm F), together with a high concentration of phosphate, it is a bisphosphate composition, which has orphophosphoric acid and tetrasodium pyrophosphate (in a proportion of about 2%, added), at acidulated pH and with very little water.
- the present invention brings a higher concentration of stable phosphates in acidic environment, due to the little presence of water, it promotes a greater instantaneous precipitation of calcium fluoride, when in contact with the tooth and saliva. This precipitation is responsible for the obliteration of the dentinal tubules (remineralization of the carious process and dentin desensitization).
- the amount of water present in the formulation is less than 5%, to favor the greater amount of orthophosphoric acid in the pH adjustment;
- the acidulating agent, orthophosphoric acid has a dual function, in order to guarantee the final pH of the product between 4.3 and 5.3 and, at the same time, to provide phosphate in the formulation (together with tetrasodium pyrophosphate) for the precipitation of calcium fluoride; having a high concentration of fluoride, so that there is more formation of fluorapatite; possessing tetrasodium pyrophosphate, a phosphate compound that acts as an antitartar, remineralizing and desensitizing agent.
- the present invention aims to provide a dentifrice composition for daily use, with the ability to promote anti-demineralizing, regenerating and desensitizing dentin actions, relieving pain and promoting complete maintenance of oral health.
- the present invention describes a toothpaste formulation that comprises the synergistic association of a small amount of water (less than 5%), a fluorinated agent (sodium fluoride) and two phosphate compounds (tetrasodium pyrophosphate (Na 4 P 2 0 7 ) together with phosphoric acid (H 3 P0 4 )), which give it an anti-demineralizing / regenerating action.
- a fluorinated agent sodium fluoride
- two phosphate compounds tetrasodium pyrophosphate (Na 4 P 2 0 7 ) together with phosphoric acid (H 3 P0 4 )
- H 3 P0 4 phosphoric acid
- the composition also has dental desensitizing action due to the CaF 2 precipitation capacity, which obliterates the dentinal tubules.
- the present invention has as main advantages:
- a dentifrice composition of daily use with a reduced water percentage, less than 5%, which favors the greater amount of orthophosphoric acid used in pH adjustment;
- orthophosphoric acid has two functions, that of guaranteeing an acid pH, between 4.3 and 5.3, while providing phosphate to the composition, together with tetrasodium pyrophosphate, for precipitation of calcium fluoride, which promotes the obliteration of dentinal tubules;
- a toothpaste composition of daily use with a higher concentration of fluoride which allows greater formation of calcium fluoride and fluorapatite
- anti-tartar agent tetrasodium pyrophosphate (phosphate compound)
- Figure 1 Visual pain scale.
- Table 3 Components used to obtain the toothpaste composition.
- Carboxymethylcellulose in a proportion of 0.7 to 1.0% w / w of the composition; Glycerin, in a proportion of 45 to 55% w / w of the composition.
- Carboxymethylcellulose is added to glycerin, under slow agitation, between 45,000 to 200,000 rpm at 25 ° C, so that no lumps are formed, and the agitation remains until complete dissolution of the carboxymethylcellulose, which lasts about 15 minutes, and formation of the glycerin + CMC phase.
- Deionized Water in a proportion of 2 to 4.9% w / w of the composition;
- Sodium fluoride in a proportion of 0.25 to 0.5% w / w of the composition;
- Sodium Saccharin in a proportion of 0.2 to 0.5% w / w of the composition;
- Tetrasodium pyrophosphate in a proportion of 0.3 to 1.0% w / w of the composition.
- Sorbitol in a proportion of 3 to 10% w / w of the composition
- Polyethylene glycol - PEG 600 in a proportion of 0, at 2% w / w of the composition; Glycerin + CMC phase, previously prepared, in its entirety. [032] Then, they are added to the mixture, so that they are homogenized with scraper and propeller, for the period of 01 h30 minutes, at speed and temperature indicated by the equipment manufacturer:
- Thickener silica in a proportion of 9 to 11% w / w of the composition
- Abrasive silica in a proportion of 5 to 15% w / w of the composition.
- Triclosan in a proportion of 0.1 to 0.3% w / w of the composition, as an antiseptic agent
- Flavorings in a proportion of 1 to 3% w / w of the composition
- mice in a proportion of 0.1 to 0.2% w / w of the composition
- the pH is corrected, to a value between 4.5 and 5.0; through the addition and homogenization, at speed and temperature indicated by the equipment manufacturer, of orthophosphoric acid, in a proportion of 0.8 to 2.0% w / w of the composition.
- the composition is filled in tubes, hereinafter called the daily regenerating gel.
- the operator performs weight tests throughout the filling process to verify that the specified weight matches that specified.
- the tubes of the repairing gel are packaged and labeled, according to the guidelines of the regulatory body.
- the daily restorative gel may have calcium supplementation in its composition, since materials containing calcium have the potential to increase the remineralization of dental enamel lesions (Exterkate et al. (1993), A Single-section Model for Enamel De- and Remineralization Studies. I. The Effects of Different Ca / P Ratios in Remineralization Solutions; and Tanaka et al. (2012), Optimization of calcium concentration of saliva with phosphoryl oligosaccharides of calcium (POs-Ca) for enamel remineralization in vitro). In this sense, studies have been conducted to find the best ratio of the quantity to be made available, in relation to the other components of the formulation. Initial results showed that a calcium concentration, about 0.5 higher than that of phosphate, a Ca: P ratio of 1.67 provides the optimal rate of remineralization, which corroborates with data from the literature.
- a Ca supplement would enhance this remineralizing action of the described composition.
- calcium supplementation can be performed by adding calcium salts, in a proportion of 0.5 to 6% w / w of the composition, preferably using: Calcium Glycerophosphate, Calcium Carbonate and Hydroxide Calcium.
- Such supplementation can happen by adding directly to the composition (encapsulated or not), or even, as a serum / booster, to be added at the time of use, and found separately in the packaging, acting as an accelerator / enhancer.
- a convenience sample made up of 53 patients from 22 dental surgeons who perform treatments against tooth sensitivity and tooth whitening, received the daily regenerating gel described, so that tooth brushing could be performed after claiming to have some type and sensitivity.
