Abstract

알러지 진단용 칩 및 그 알러지 진단용 칩 제조 방법이 개시된다. 본 발명에 따르면, 흡인 유닛의 내부에 상압(normal pressure)보다 낮은 압력인 음압(negative pressure)을 형성함으로써 상부 플레이트 내의 항원이 포함된 용액을 하방으로 이동시켜, 멤브레인 상에 항원이 흡착된 라인 형태의 코팅부를 복수로 형성하는 코팅부 형성 단계; 를 포함하는 알러지 진단용 칩 제조 방법이 제공된다.

기술분야
본 발명은 알러지 진단용 칩 및 그 알러지 진단용 칩 제조 방법에 관한 것이다.
시료 내의 성분을 분석하기 위한 진단용 칩은 멤브레인 형태의 고정체 상에 항원 등을 코팅함으로써 제조되는 것이 일반적이다. 이후, 제조된 진단용 칩 상에 코팅된 항원과 분석하고자 하는 시료를 반응시킴으로써 여러가지 생물학적 정보를 얻어낼 수 있다.
이러한 진단용 칩은 크게 도트(dot)형과 라인(line)형으로 구별될 수 있다. 도트형 진단 칩은 고정체 상에 항원 등이 도트 형태로 코팅된 진단 칩을 의미하고, 라인형 진단 칩은 고정체 상에 항원 등이 라인 형태로 코팅된 진단 칩을 의미한다. 이 중 도트형 진단 칩은 고정체 상에 항원 등을 도트 형태로 일일이 코팅해야 하는 번거로움이 있다. 반면, 라인형 진단 칩은 고정체 상에 항원 등을 라인 형태로 코팅한 후 이를 원하는 형태로 잘라서 스트립(strip) 형태로 쓸 수 있기 때문에, 생산성의 측면에서 이점이 있다.
이러한 라인형 진단 칩에서는, 코팅에 의해 형성된 라인들 간의 간격이 작을수록 동일 면적 대비 코팅되는 라인의 수가 증가하게 되므로 한 번의 검출만으로도 보다 다양한 정보를 얻을 수 있다. 따라서, 라인형 진단 칩의 경우 동일한 면적 대비 코팅되는 라인의 수를 보다 극대화시키는 고집적화가 이루어질 필요가 있다.
따라서, 본 발명이 해결하고자 하는 과제는, 종래 기술에 비해 고집적화된 알러지 진단용 칩을 제조하는 것이다.
상기 목적을 달성하기 위한 본 발명의 일 측면에 따르면, 항원이 포함된 용액이 수용될 수 있는 공간이 형성되는 상부 플레이트, 상기 상부 플레이트의 하부에 구비되는 흡인 유닛, 및 상기 상부 플레이트의 하부에 구비되고 상기 흡인 유닛 상에 장착되는 하부 플레이트를 준비하는 단계; 상기 하부 플레이트의 상면에 멤브레인을 구비하는 멤브레인 구비 단계; 상기 흡인 유닛의 내부에 상압(normal pressure)보다 낮은 압력인 음압(negative pressure)을 형성함으로써 상기 상부 플레이트 내의 상기 항원이 포함된 용액을 하방으로 이동시켜, 상기 멤브레인 상에 상기 항원이 흡착된 라인 형태의 코팅부를 복수로 형성하는 코팅부 형성 단계; 를 포함하는 알러지 진단용 칩 제조 방법이 제공된다.
복수의 상기 코팅부의 두께는 0.2mm 내지 0.5mm일 수 있다.
상기 코팅부 형성 단계에서, 복수의 상기 코팅부 간의 간격은 0.5mm 내지 0.9mm일 수 있다.
상기 멤브레인 구비 단계는, 상기 흡인 유닛과 상기 멤브레인 사이에 상기 음압과 상압 간의 차이에 의한 상기 멤브레인의 손상을 방지하기 위한 보호판(plate)를 구비하는 것을 더 포함할 수 있다.
상기 멤브레인 구비 단계와 상기 코팅부 형성 단계 사이에 구비되고, 상기 멤브레인이 하방으로 밀착될 수 있도록 상기 코팅 유닛의 상부 영역 중 일부를 추가로 가압하는 가압 단계; 를 더 포함할 수 있다.
상기 목적을 달성하기 위한 본 발명의 다른 측면에 따르면, 항원이 포함된 용액이 수용될 수 있는 공간이 형성되는 상부 플레이트, 상기 상부 플레이트의 하부에 구비되는 흡인 유닛, 및 상기 상부 플레이트의 하부에 구비되고 상기 흡인 유닛 상에 장착되는 하부 플레이트를 준비하는 단계; 상기 하부 플레이트의 상면에 멤브레인을 구비하는 멤브레인 구비 단계; 상기 흡인 유닛의 내부에 상압(normal pressure)보다 낮은 압력인 음압(negative pressure)을 형성함으로써 상기 상부 플레이트 내의 상기 항원이 포함된 용액을 하방으로 이동시켜, 상기 멤브레인 상에 상기 항원이 흡착된 라인 형태의 코팅부를 복수로 형성하는 코팅부 형성 단계; 를 포함하고, 상기 코팅부 형성 단계에서 상기 음압은 7초 이상 형성되는 알러지 진단용 칩 제조 방법이 제공된다.
상기 목적을 달성하기 위한 본 발명의 또 다른 측면에 따르면, 시트 형태의 멤브레인; 및 상기 멤브레인 상에 항원이 코팅된 영역인 복수의 코팅부; 를 포함하고, 상기 코팅부는 일정한 두께(t)를 가지고, 상기 복수의 코팅부 간에는 일정한 간격(d)으로 서로 이격되어 있고, 상기 코팅부 간의 간격(d)에 대한 상기 코팅부의 두께(t)의 비(t/d)는 0.40 내지 0.55인 알러지 진단용 칩이 제공된다.
상기 코팅부의 두께(t)는 0.2mm 내지 0.5mm일 수 있다.
상기 복수의 코팅부 간의 간격(d)은 0.5mm 내지 0.9mm일 수 있다.
본 발명에 따르면, 종래 기술에 비해 고집적화된 알러지 진단용 칩을 제조할 수 있다.

이하, 도면을 참고하여, 본 발명에 따른 알러지 진단용 칩 제조 방법 및 그 제조 방법에 사용될 수 있는 제조 장치에 대해 설명하도록 한다.
칩 제조 방법 및 칩 제조 장치
도 1은 본 발명에 따른 알러지 진단용 칩 제조 방법에 사용될 수 있는 제조 장치의 구조를 도시한 사시도이고, 도 2는 본 발명에 따른 알러지 진단용 칩 제조 방법에 사용될 수 있는 제조 장치의 구조를 도시한 측단면도이다.
도 1 및 도 2에 도시된 바와 같이 본 발명에 따른 알러지 진단용 칩 제조 장치(10, 이하, '제조 장치'라 부르기로 한다)는 항원이 포함된 용액이 수용될 수 있는 상부 플레이트(100)를 포함할 수 있다. 상부 플레이트(100)는 외부로부터 항원이 포함된 용액을 공급받아 하방으로 배출하는 구성일 수 있다.
상부 플레이트(100)의 하부에는 압력 차이에 의해 상부 플레이트(100) 내부에 수용된 항원이 포함된 용액을 하방으로 이동시킬 수 있는 흡인 유닛(200)이 구비될 수 있다. 본 발명에 따른 흡인 유닛(200)은 하기에서 살펴볼 바와 같이 흡인 유닛 내부의 압력과 외부의 압력 차이에 의해 외부의 공기를 흡인 유닛(200)의 하방으로 이동시킴으로써, 그에 따라 상부 플레이트(100) 내부의 항원이 포함된 용액이 하방으로 배출되도록 하는 구성일 수 있다.
한편, 도 1 내지 도 2를 참고하면, 상부 플레이트(100)의 하부에는 흡인 유닛(200) 상에 장착되는 하부 플레이트(150)가 구비될 수 있다.
전술한 내용 및 도 1 내지 도 2를 참고하면, 본 발명에 따른 알러지 진단용 칩 제조 방법은, 항원이 포함된 용액이 수용될 수 있는 공간이 형성되는 상부 플레이트(100), 상부 플레이트(100)의 하부에 구비되는 흡인 유닛(200), 및 상부 플레이트(100)의 하부에 구비되고 흡인 유닛(200) 상에 장착되는 하부 플레이트(150)를 준비하는 단계를 포함할 수 있다. 또한, 본 발명에 따른 알러지 진단용 칩 제조 방법은, 하부 플레이트(150)의 상면에 멤브레인을 구비하는 멤브레인 구비 단계를 더 포함할 수 있다.
도 3은 본 발명에 사용될 수 있는 제조 장치 중 흡인 유닛의 구조를 도시한 평면도이고, 도 4는 본 발명에 사용될 수 있는 제조 장치 중 흡인 유닛의 구조를 도시한 측단면도이다.
도 3 및 도 4에 도시된 바와 같이 흡인 유닛(200)에는 상부가 개방되어 있는 공간인 공간부(210)가 형성될 수 있고, 하부 플레이트(150)는 공간부(210)의 상부에 탑재될 수 있다.
전술한 바와 같이 상부 플레이트(100)의 내부에 수용된 항원이 포함된 용액은 흡인 유닛에 의해 하방으로 배출될 수 있다. 또한, 항원이 포함된 용액이 하방으로 배출될 때, 하부 플레이트(150)의 상면에는 얇은 두께를 갖는 멤브레인이 구비되는데, 하방으로 배출된 용액은 멤브레인 상에 분사됨으로써 멤브레인에 항원이 코팅될 수 있다.
도 4에 도시된 바와 같이 흡인 유닛(200)에는, 공기가 이송되는 경로를 제공하는 유로(220)가 구비될 수 있다. 유로(220)는 복수로 구비될 수 있다. 도 4에는 유로(220)가 3개가 구비된 경우가 도시되어 있다.
또한, 흡인 유닛(200)은 유로(220)와 연결되며 내부에 음압을 형성할 수 있는 음압 형성부(미도시)가 형성될 수 있다. 본 발명에 따르면 음압 형성부의 내부에 존재하는 공기가 제거되는 경우 음압 형성부의 내부는 상압(normal pressure)보다 낮은 상태인 음압(negative pressure) 상태가 된다. 따라서, 외부의 압력(즉, 상압)과 읍압 형성부 내부의 압력(즉, 음압) 간에는 압력 차이가 존재하게 된다. 이때, 흡인 유닛(200)의 유로(220)를 개방하게 되면 상기 음압 형성부 내부의 압력과 외부의 압력 간의 차이에 의해 외부의 공기가 음압 형성부 내부에 공급됨과 동시에 상부 플레이트(100) 내부에 수용되어 있던 항원이 멤브레인에 분사되어 코팅된다.
따라서, 본 발명에 따른 알러지 진단용 칩 제조 방법은, 흡인 유닛(200)의 내부에 상압(normal pressure)보다 낮은 압력인 음압(negative pressure)을 형성함으로써 상부 플레이트(100) 내 항원이 포함된 용액을 하방으로 이동시켜, 멤브레인 상에 항원이 흡착된 라인 형태의 코팅부를 복수로 형성하는, 코팅부 형성 단계를 포함할 수 있다.
한편, 음압 형성부 내에서 음압은, 음압 형성부 내의 공기가 외부로 배출됨으로써 형성될 수 있다. 이후, 코팅부 형성 단계에서 음압 형성부 내부로 공기가 유입되고, 그에 따라 음압 형성부 내의 압력은 점차 증가하게 된다. 그러다가 음압 형성부 내의 압력이 상압과 동일하게 되면 코팅부 형성 단계가 종료될 수 있다.
본 발명에 따른 알러지 진단용 칩 제조 방법의 코팅부 형성 단계에서, 음압은 7초 내지 120초 동안 형성될 수 있다. 코팅부 형성 단계에서 음압이 7초 미만으로 형성되는 경우에는, 멤브레인에 항원을 코팅하는 과정에서 항원이 포함된 용액이 멤브레인에 라인 형태로 제대로 코팅되지 못하고 번지는 문제가 발생할 수 있다. 반대로, 코팅부 형성 단계에서 음압이 120초를 초과하여 형성되는 경우에는, 멤브레인에 항원을 코팅하는 공정에 지나치게 많은 시간이 소모되어 생산성이 떨어질 수 있다. 보다 바람직하게, 코팅부 형성 단계에서 음압은 7초 내지 90초 동안 형성될 수 있다.
또한, 코팅부 형성 단계에서, 음압과 상압 간의 차이는 1.8bar 내지 2.2bar 일 수 있다. 코팅부 형성 단계에서 음압과 상압 간의 차이가 1.8bar 미만인 경우에는 멤브레인의 항원 코팅 과정에서 상부 플레이트(100) 내의 용액이 분사되는 속도가 충분하지 않게 되어 항원 코팅의 품질이 저하될 수 있다. 반면, 코팅부 형성 단계에서 음압과 상압 간의 차이가 2.2bar를 초과하는 경우에는 멤브레인의 항원 코팅 과정에서 용액이 분사되는 속도 및 읍압 형성부에 유입되는 공기의 속도가 지나치게 커지게 되어 멤브레인이 손상될 수 있다.
다시 도 1 및 도 2를 참고하면, 제조 장치(10)는 상부 플레이트(100)를 하방으로 가압하는 가압 유닛(300)을 더 포함할 수 있다. 본 발명에 따른 가압 유닛(300)은 멤브레인에 항원을 코팅하는 과정에서 상부 플레이트(100)를 하강시켜 하부 플레이트(150)에 밀착 및 가압함으로써 하부 플레이트(150)의 상면에 구비된 멤브레인에 항원의 코팅이 원활하게 이루어지도록 하는 구성일 수 있다.
도 2에 도시된 바와 같이 가압 유닛(300)은 상부 플레이트(100)의 상부의 양 측부를 하방으로 가압하는 측면 가압부(310) 및 상부 플레이트(100)의 중앙부를 하방으로 가압하는 중앙 가압부(320)를 포함할 수 있다. 측면 가압부(310)와 중앙 가압부(320) 간에는 서로 독립적으로 구동될 수 있다.
따라서, 본 발명에 따른 알러지 진단용 칩 제조 방법은 멤브레인이 하방으로 밀착될 수 있도록 코팅 유닛의 상부 영역 중 일부를 추가로 가압하는 가압 단계를 더 포함할 수 있다. 본 발명에 따른 가압 단계는 전술한 멤브레인 구비 단계와 코팅부 형성 단계 사이에 이루어질 수 있다.
계속해서 도 1 및 도 2를 참고하면, 측면 가압부(310)의 내측에는 계단 형상의 단차(310a)가 형성될 수 있다. 이때, 단차(310a)는 상부 플레이트(100)의 양 측부에 대응되는 형상을 가질 수 있다. 도 2에는 상부 플레이트(100)의 양 측부가 수직하게 형성되고, 상부 플레이트(100)의 양 측부와 마주보는 단차(310a) 역시 수직하게 형성된 경우가 도시되어 있다. 단차(310a)가 상부 플레이트(100)의 양 측부에 대응되는 형상을 가지는 경우, 측면 가압부(310)는 상부 플레이트(100)를 하방으로 가압하는 역할을 수행할 뿐만 아니라, 상부 플레이트(100)가 좌우로 흔들리지 않도록 상부 플레이트(100)를 파지(grip)하는 역할도 수행할 수 있다.
도 5는 본 발명에 사용될 수 있는 제조 장치 중 상부 플레이트의 일 예의 구조를 도시한 사시도이고, 도 6은 본 발명에 사용될 수 있는 제조 장치 중 상부 플레이트의 일 예의 구조를 도시한 평면도이고, 도 7은 도 6의 A-A 선을 따라 절단된 상부 플레이트의 구조를 도시한 단면도이다.
도 5 내지 도 7에 도시된 바와 같이 일 예에 따르면 상부 플레이트(100)는 항원이 포함된 용액이 주입되는 주입구(112a)가 형성되는 상면부(112) 및 상면부(112)의 하부에 구비되고 항원이 포함된 용액이 하부 플레이트(150, 도 2 참조)를 향해 배출되는 하면부(114)를 포함할 수 있다. 도 5 내지 도 7에 도시된 바와 같이 본 발명에 따른 상부 플레이트의 일 예에 따르면 상부 플레이트(100)의 주입구(112a)는 도트(dot) 형상을 갖는 복수의 공급홀일 수 있다. 이하, 본 명세서에서 도면부호 112a를 '공급홀'이라 부르기로 한다.
한편, 본 발명의 상부 플레이트(100)의 일 예에서 하면부(114)에는 라인 형상을 갖는 슬릿(114a)이 복수로 형성될 수 있다. 본 발명에 따르면 상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)을 통해 항원이 포함된 용액이 라인 형태로 배출됨으로써 멤브레인에 항원이 라인 형태로 코팅될 수 있다. 즉, 본 명세서에서 슬릿(114a)은 물리적인 구성을 의미하는 것이 아니라, 항원이 포함된 용액이 배출되는 공간을 의미한다.
상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)의 폭은 0.2mm 내지 0.5mm일 수 있다. 본 발명에 따르면 상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)의 폭이 0.2mm 내지 0.5mm이므로, 멤브레인에 항원이 포함된 용액을 라인 형태로 코팅할 때 멤브레인에 코팅된 라인의 두께 역시 0.2mm 내지 0.5mm가 되도록 할 수 있다. 따라서, 종래 기술에 비해 멤브레인에 코팅된 라인의 두께를 줄임으로써, 진단용 칩의 고집적화를 도모할 수 있다. 보다 바람직하게 상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)의 폭은 0.3mm 내지 0.5mm일 수 있다.
한편, 전술한 바와 같이 코팅부 형성 단계에서, 상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)을 통해 항원이 포함된 용액이 배출되므로 코팅부의 두께 슬릿(114a)의 폭에 대응될 수 있다. 즉, 코팅부 형성 단계에서 형성된 복수의 코팅부의 두께는 0.2mm 내지 0.5mm일 수 있다. 보다 바람직하게는, 복수의 코팅부의 두께는 0.3mm 내지 0.5mm일 수 있다.
계속해서 도 5 내지 도 7을 참고하면, 상부 플레이트(100)는 상면부(112)와 하면부(114)에 형성되고 상면부(112)와 하면부(114)를 연결하는 연결부(116)를 포함할 수 있다. 상부 플레이트(100)의 연결부(116)는 상부 플레이트(100)의 몸체를 형성하는 것으로 이해될 수도 있다.
이때, 도 7에 도시된 바와 같이 상부 플레이트(100)의 연결부의 내부에는 복수의 박판(116a)들이 서로 이격되게 구비되어 있을 수 있다. 복수의 박판(116a)들 사이에는 서로 밀폐된 복수의 내부 공간이 형성될 수 있는데, 항원이 포함된 용액은 복수의 박판(116a)들 사이에의 내부 공간에 수용될 수 있다.
한편, 상부 플레이트(100)에 구비된 복수의 박판(116a)들에 의해 형성된 복수의 내부 공간들은 각각 상부 플레이트(100)의 하면부에 형성된 복수의 슬릿(114a)들 중 하나와 연통될 수 있다. 또한, 상부 플레이트(100)의 상면부(112)에 형성된 복수의 공급홀(112a)들 각각은 상부 플레이트(100)의 연결부(116)에 형성된 복수의 내부 공간들 중 하나와 연통될 수 있다.
따라서, 상부 플레이트(100)의 공급홀(112a)들 중 하나를 통해 공급된 항원이 포함된 용액은 복수의 박판(116a)들에 의해 형성되고 상기 공급홀(112a)과 연통된 복수의 내부 공간에 유입된 후 상기 복수의 내부 공간과 연통된 슬릿(114a)을 통해 하방으로 배출될 수 있다.
상부 플레이트(100)의 연결부(116)에 형성된 복수의 박판(116a)의 두께는 0.5mm 내지 0.9mm일 수 있다.
한편, 도 7에 도시된 바와 같이 상부 플레이트(100)의 하면부(114)에 형성된 슬릿(114a)들은 연결부(116)에 구비된 복수의 박판들(116a)에 의해 형성되는 복수의 내부 공간들의 끝부와 연결될 수 있다. 또한, 상부 플레이트(100)의 하면부(114)에 형성된 슬릿(114a)들 간의 간격은 연결부(116)에 구비된 복수의 박판(116a)들의 두께과 동일할 수 있다. 따라서, 박판(116a)의 두께는 코팅부 형성 단계에서 형성된 코팅부들 간의 간격과 동일할 수 있다. 즉, 본 발명에 따른 알러지 진단용 칩의 제조 방법의 코팅부 형성 단계에서 복수의 코팅부 간의 간격은 0.5mm 내지 0.9mm일 수 있다. 복수의 코팅부 간의 간격이 0.5mm 미만인 경우에는 멤브레인에 항원을 코팅하는 과정에서 흡인 유닛(200)의 가동에 의한 급격한 유체의 흐름에 의해 박판(116a)이 손상될 수 있다. 반면, 복수의 코팅부 간의 간격이 0.9mm를 초과하는 경우에는 멤브레인에 코팅되는 코팅부들 간의 폭이 지나치게 커지게 되어 진단용 칩의 고집적화가 불가능하게 된다.
상부 플레이트(100)에 형성된 상면부의 공급홀(112a), 복수의 박판(116a)들에 의해 형성된 내부 공간, 및 하면부의 슬릿(114a)은 서로 일대일로 연통될 수 있다. 따라서, 상부 플레이트(100)의 상면부(112)에 형성된 복수의 공급홀(112a)의 개수, 상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)의 개수, 및 상부 플레이트(100)의 연결부(116)에 형성된 복수의 내부 공간의 개수는 서로 동일할 수 있다. 도 7에는 상부 플레이트(100)의 연결부(116)에 형성된 복수의 내부 공간들 중 일부가 상면부(112)의 공급홀(112a)과 연통되지 않는 것처럼 도시되어 있지만, 도 6에 도시된 바와 같이 공급홀(112a)은 상부 플레이트의 상면부(112)의 전 영역에 걸쳐 형성될 수 있기 때문에, 도 7에 도시된 내용이 본 문단의 상기 내용과 부합하지 않는 것으로 이해되어서는 안될 것이다.
그러나, 이와 달리 상부 플레이트(100)에 형성된 상면부의 공급홀(112a)은 복수의 박판(116a)들에 의해 형성된 내부 공간과 n(n은 2 이상의 정수) 대 1로 대응될 수도 있다. 예를 들어, 상부 플레이트(100)에 형성된 상면부의 공급홀(112a)은 복수의 박판(116a)들에 의해 형성된 내부 공간과 2대 1로 대응될 수 있다.
도 8은 본 발명에 사용될 수 있는 제조 장치 중 상부 플레이트의 다른 예의 구조를 도시한 사시도이고, 도 9는 본 발명에 사용될 수 있는 제조 장치 중 상부 플레이트의 다른 예의 구조를 도시한 평면도이고, 도 10은 도 9의 B-B 선을 따라 절단된 상부 플레이트의 구조를 도시한 단면도이다. 하기에서, 상부 플레이트의 다른 예에 대한 설명 중 상부 플레이트의 일 예에 대한 설명과 중첩되는 부분은 생략하고, 상부 플레이트의 일 예와 다른 내용 위주로 설명하도록 한다.
도 8 내지 도 9에 도시된 바와 같이 다른 예에 따르면 상부 플레이트(100)의 상면부(112)에는 상부 플레이트(112)로 항원이 포함된 용액이 공급될 수 있도록 라인 형상을 갖는 슬릿(112b)이 주입구로서 복수로 형성될 수 있고, 상부 플레이트(100)의 상면부(112)에 형성된 복수의 슬릿(112b)들 각각은 상부 플레이트(100)의 연결부(116)에 형성된 복수의 내부 공간들 중 하나와 연통될 수 있다.
따라서, 상부 플레이트(100)의 상면부(112)의 슬릿(112b)들 중 하나를 통해 공급된 항원이 포함된 용액은 복수의 박판(116a)들에 의해 형성되고 상기 슬릿(112b)과 연통된 복수의 내부 공간에 유입된 후 상기 복수의 내부 공간과 연통된 하면부(114)의 슬릿(114a)을 통해 하방으로 배출될 수 있다.
또한, 상부 플레이트(100)에 형성된 상면부의 슬릿(112b), 복수의 박판(116a)들에 의해 형성된 내부 공간, 및 하면부의 슬릿(114a)은 서로 일대일로 연통될 수 있다. 따라서, 상부 플레이트(100)의 상면부(112)에 형성된 복수의 슬릿(112b)의 개수, 상부 플레이트(100)의 하면부(114)에 형성된 복수의 슬릿(114a)의 개수, 및 상부 플레이트(100)의 연결부(116)에 형성된 복수의 내부 공간의 개수는 서로 동일할 수 있다. 도 10에는 상부 플레이트(100)의 연결부(116)에 형성된 복수의 내부 공간들 중 일부는 상면부(112)의 슬릿(112b)과 연통되지 않는 것처럼 도시되어 있지만, 도 9에 도시된 바와 같이 슬릿(112b)은 상부 플레이트의 상면부(112)의 전 영역에 걸쳐 형성되어 있기 때문에, 도 9에 도시된 내용이 상기 본 문단의 상기 내용과 부합하지 않는 것으로 이해되어서는 안될 것이다.
한편, 상부 플레이트(100)는 전술한 공급홀(112a)과 슬릿(112b)이 혼합되어 형성된 형태를 가질 수도 있다. 즉, 본 발명에 따른 상부 플레이트(100)는 공급홀(112a)과 슬릿(112b)이 모두 형성된 구조를 가질 수도 있다.
도 11은 본 발명에 사용될 수 있는 제조 장치 중 하부 플레이트의 구조를 도시한 사시도이고, 도 12는 도 11의 C-C 선을 따라 절단된 하부 플레이트의 구조를 도시한 측단면도이다.
도 11 및 도 12에 도시된 바와 같이 하부 플레이트(150)는 복수의 슬릿(162a)이 형성되는 상면부(162)와 복수의 슬릿(164a)이 형성되는 하면부(164)를 포함할 수 있다. 상부 플레이트의 경우와 동일하게 하부 플레이트(150)에 형성된 슬릿들(162a, 164a) 역시 물리적인 공간을 의미한다.
하부 플레이트(150)의 상면부(162) 및 하면부(164)에 각각 형성된 슬릿들(162a, 164a)은 항원이 포함된 용액이 멤브레인에 코팅되는 과정에서, 외부의 공기들이 흡인 유닛으로 유입되는 경로를 제공할 수 있다. 즉, 전술한 바와 같이 멤브레인의 코팅 과정에서 흡인 유닛(200)의 내부에서 음압이 형성되면 외부의 공기들은 하부 플레이트(150)의 상면부(162)에 형성된 슬릿(162a)과 하면부(164)에 형성된 슬릿(164a)을 순차적으로 통과한 후 흡인 유닛으로 공급될 수 있다. 한편, 상부 플레이트의 경우와 유사하게 하부 플레이트(150)에도 상면부(162)와 하면부(164)를 연결하는 연결부가 형성될 수 있고 연결부의 내부에는 서로 이격되어 있는 복수의 박판들이 형성될 수 있다. 복수의 박판들 사이의 내부 공간 각각은 상면부(162)의 슬릿(162a)들 중 하나 및 하면부(164)의 슬릿(164a)들 중 하나와 연통될 수 있다.
한편, 본 발명에 따른 상부 플레이트에서 박판의 두께에 대한 슬릿의 폭의 비는 0.40 내지 0.55일 수 있다.
도 13은 본 발명에 따라 제조된 알러지 진단용 칩의 구조를 도시한 평면도이다.
도 13에 도시된 바와 같이 본 발명에 따른 알러지 진단용 칩(400)은 전술한 본 발명에 따른 알러지 진단용 칩 제조 방법에 의해 제조된 구성일 수 있다.
알러지 진단용 칩(400)은 시트 형태의 멤브레인(400a) 및 멤브레인(400a) 상에 항원이 코팅된 영역인 코팅부(410)를 포함할 수 있다. 코팅부(410)는 복수개 형성될 수 있고, 일정한 두께를 갖는 띠 형상을 가질 수 있다.
알러지 진단용 칩(400)은 일정한 폭(W) 및 길이(L)를 갖는 직사각형 형상을 가질 수 있고, 일정한 두께(t)를 갖는 복수의 코팅부(410)는 알러지 진단용 칩(400)의 폭(W) 방향을 따라 서로 일정한 간격(d)만큼 이격되어 구비될 수 있다.
본 발명에 따른 알러지 진단용 칩(400)은 종래 기술에 비해 칩의 동일 면적 대비 많은 수의 코팅부를 형성할 수 있으므로, 알러지 진단용 칩(400)의 고집적화가 가능할 수 있다. 이를 위해, 본 발명에 따른 알러지 진단용 칩(400)에서 코팅부(410) 간의 간격(d)과 코팅부(410)의 두께(t) 간에는 일정한 비율을 가질 수 있다.
즉, 본 발명에 따른 알러지 진단용 칩(400)에서 코팅부(410) 간의 간격(d)에 대한 코팅부(410)의 두께(t)의 비(즉, t/d)는 0.40 내지 0.55일 수 있다.
또한, 본 발명에 따르면 코팅부(410)의 두께(t)는 0.2mm 내지 0.5mm일 수 있다. 또한, 코팅부(410) 간의 간격(d)은 0.5mm 내지 0.9mm일 수 있다.
한편, 본 발명이 코팅부 형성 단계에서 상압과 음압 간의 차이를 일정 수치 범위 내에서 조절을 함에도 불구하고 멤브레인의 손상이 일어날 수 있다. 예를 들어, 코팅부 형성 단계에서 음압과 상압 간의 압력 차이에 의해 외부의 공기가 하부 플레이트를 거쳐 흡인 장치로 유입되는데, 하부 플레이트(150)의 상면에 구비된 멤브레인이 하부 플레이트(150)의 상면부(162)와 직접 접촉하는 경우 상면부(162)의 슬릿(162a)에 빨려 들어가는 등의 문제가 발생할 수 있다.
따라서, 상기 문제점을 방지하기 위해 본 발명에 따른 알러지 진단용 칩 제조 방법의 멤브레인 구비 단계는, 흡인 유닛(200)과 멤브레인 사이에 음압과 상압 간의 차이에 의한 멤브레인의 손상을 망지하기 위한 보호판(plate)를 구비하는 것을 더 포함할 수 있다. 보호판은 면(cotton) 재질을 포함할 수 있다. 보호판이 면 재질을 포함하거나 면 재질로 이루어진 경우 코팅부 형성 단계에서 보호판 자체의 재질로 인해 멤브레인이 손상되는 것을 방지할 수 있다.
실시예 1
체결을 통해 흡인 유닛의 공간부의 상부에 하부 플레이트를 고정시킨 후 하부 플레이트의 상면에 멤브레인을 구비하였다. 또한, 상부 플레이트의 양 측부를 측면 가압부로 파지한 후 측면 가압부와 중앙 가압부를 하방으로 이동시켜 상부 플레이트를 하부 플레이트 및 멤브레인에 밀착시켰다. 또한, 흡인 유닛의 음압 형성부의 내부에 있는 공기를 배출하여 상압보다 2 bar만큼 낮은 음압 상태를 만들었다.
이후, 흡인 유닛의 유로를 개방함으로써 외부의 공기가 음압 형성부에 유입되면서 멤브레인에 항원이 포함된 용액을 코팅하여 진단용 칩을 제조하였다. 흡인 유닛의 유로는 7초동안 개방되었다. 음압 형성부 내에 공기가 유입되면서 음압 형성부 내의 압력은 점차 상승하였고 코팅이 완료된 후 음압 형성부 내의 압력은 상압에 도달하였다.
실시예 2
흡인 유닛의 유로가 30초동안 개방된 것을 제외하면, 실시예 1과 동일하게 멤브레인에 항원이 포함된 용액이 코팅되었다.
실시예 3
흡인 유닛의 유로가 60초동안 개방된 것을 제외하면, 실시예 1과 동일하게 멤브레인에 항원이 포함된 용액이 코팅되었다.
실시예 4
흡인 유닛의 유로가 120초동안 개방된 것을 제외하면, 실시예 1과 동일하게 멤브레인에 항원이 포함된 용액이 코팅되었다.
비교예
흡인 유닛의 유로가 5초동안 개방된 것을 제외하면, 실시예 1와 동일하게 멤브레인에 항원이 포함된 용액이 코팅되었다.
실험예 1
실시예와 비교예에 따라 제조된 진단용 칩의 항원 코팅 상태를 관찰하였다. 관찰한 결과는 도 14에 도시되어 있다. 도 14의 (a)는 비교예에 따른 진단용 칩의 코팅 상태를 도시한 것이고, 도 14의 (b) 내지 (e)는 각각 실시예 1 내지 실시예 4에 따른 진단용 칩의 코팅 상태를 도시한 것이다.
도 14의 (a)에 도시된 바와 같이, 비교예의 경우 진단용 칩에 코팅된 라인 주변에 항원이 포함된 용액의 일부가 번져 있는 것을 확인할 수 있었다. 즉, 비교예의 경우 음압이 형성되는 시간이 충분히 확보되지 못해 멤브레인에 코팅이 제대로 이루어지지 못하였음을 확인할 수 있다.
도 14의 (b) 내지 (e)에 도시된 바와 같이, 실시예 1 내지 실시예 4의 경우 진단용 칩에 코팅된 라인에만 코팅이 정상적으로 이루어졌음을 확인할 수 있었다.
실험예 2
실시예 1 내지 4 및 비교예에 따라 제조된 진단용 칩에서 항원 포함된 용액이 코팅된 코팅층의 두께(t)와 코팅층 간의 간격(d)을 측정하였다.
실시예 1 내지 4에 따라 제조된 진단용 칩에서 코팅층의 두께(t)는 각각 0.4mm로 측정되었고, 코팅층 간의 간격(d)은 0.75mm로 측정되었다. 따라서, 코팅부 간의 간격(d)에 대한 상기 코팅부의 두께(t)의 비(t/d)는 0.533으로 측정되었다.
반면, 비교예의 경우 용액이 균일하게 코팅되지 못하고 코팅된 라인 주변으로 용액의 일부가 번졌으므로 코팅층의 두께 및 코팅층 간의 간격을 측정할 수 없었다.
이상에서 본 발명은 비록 한정된 실시예와 도면에 의해 설명되었으나, 본 발명은 이것에 의해 한정되지 않으며, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자에 의해 본 발명의 기술사상과 아래에 기재될 특허청구범위의 균등범위 내에서 다양한 실시가 가능함은 물론이다.

Claims (9)

  1. 항원이 포함된 용액이 수용될 수 있는 공간이 형성되는 상부 플레이트, 상기 상부 플레이트의 하부에 구비되는 흡인 유닛, 및 상기 상부 플레이트의 하부에 구비되고 상기 흡인 유닛 상에 장착되는 하부 플레이트를 준비하는 단계;
    상기 하부 플레이트의 상면에 멤브레인을 구비하는 멤브레인 구비 단계;
    상기 흡인 유닛의 내부에 상압(normal pressure)보다 낮은 압력인 음압(negative pressure)을 형성함으로써 상기 상부 플레이트 내의 상기 항원이 포함된 용액을 하방으로 이동시켜, 상기 멤브레인 상에 상기 항원이 흡착된 라인 형태의 코팅부를 복수로 형성하는 코팅부 형성 단계; 를 포함하는 알러지 진단용 칩 제조 방법.
  2. 청구항 1에서,
    복수의 상기 코팅부의 두께는 0.2mm 내지 0.5mm인 알러지 진단용 칩 제조 방법.
  3. 청구항 1에서,
    상기 코팅부 형성 단계에서,
    복수의 상기 코팅부 간의 간격은 0.5mm 내지 0.9mm 인 알러지 진단용 칩 제조 방법.
  4. 청구항 1에서,
    상기 멤브레인 구비 단계는,
    상기 흡인 유닛과 상기 멤브레인 사이에 상기 음압과 상압 간의 차이에 의한 상기 멤브레인의 손상을 방지하기 위한 보호판(plate)를 구비하는 것을 더 포함하는 알러지 진단용 칩 제조 방법.
  5. 청구항 1에서,
    상기 멤브레인 구비 단계와 상기 코팅부 형성 단계 사이에 구비되고, 상기 멤브레인이 하방으로 밀착될 수 있도록 상기 코팅 유닛의 상부 영역 중 일부를 추가로 가압하는 가압 단계; 를 더 포함하는 알러지 진단용 칩 제조 방법.
  6. 항원이 포함된 용액이 수용될 수 있는 공간이 형성되는 상부 플레이트, 상기 상부 플레이트의 하부에 구비되는 흡인 유닛, 및 상기 상부 플레이트의 하부에 구비되고 상기 흡인 유닛 상에 장착되는 하부 플레이트를 준비하는 단계;
    상기 하부 플레이트의 상면에 멤브레인을 구비하는 멤브레인 구비 단계;
    상기 흡인 유닛의 내부에 상압(normal pressure)보다 낮은 압력인 음압(negative pressure)을 형성함으로써 상기 상부 플레이트 내의 상기 항원이 포함된 용액을 하방으로 이동시켜, 상기 멤브레인 상에 상기 항원이 흡착된 라인 형태의 코팅부를 복수로 형성하는 코팅부 형성 단계; 를 포함하고,
    상기 코팅부 형성 단계에서 상기 음압은 7초 이상 형성되는 알러지 진단용 칩 제조 방법.
  7. 시트 형태의 멤브레인; 및
    상기 멤브레인 상에 항원이 코팅된 영역인 복수의 코팅부; 를 포함하고,
    상기 코팅부는 일정한 두께(t)를 가지고, 상기 복수의 코팅부 간에는 일정한 간격(d)으로 서로 이격되어 있고,
    상기 코팅부 간의 간격(d)에 대한 상기 코팅부의 두께(t)의 비(t/d)는 0.40 내지 0.55인 알러지 진단용 칩.
  8. 청구항 7에서,
    상기 코팅부의 두께(t)는 0.2mm 내지 0.5mm인 알러지 진단용 칩.
  9. 청구항 7에서,
    상기 복수의 코팅부 간의 간격(d)은 0.5mm 내지 0.9mm인 알러지 진단용 칩.
