Description
本発明は、金属イオン内包フラーレンを用いた高感度分子検出装置に関する。
癌は早期発見が望まれている。例えば、癌患者の呼気には、癌患者ではない者の呼気には含まれない物質が含まれていることが知られている。
例えば、食道がん患者には、2―ブタノン、酢酸、アセトン、アセトニトリルという4種類の分子が多く含まれていることがわかっている。他のがんについては、他の分子を含んでいる。また、今後、臨床の現場において数々の分子が見いだされることが予想される。
なお、図3に示す分子は肺がん患者の呼気に多く含まれるガス基質である。
そして、そのような物質の検出を利用して癌の早期発見を行うことが試みられている。現在、検出技術としては、例えば、以下の技術が試みられている。(1)MEMにより形成したSiダイヤフラムに塗布した官能膜にガスを吸着させ、次に吸着によって生じた官能膜の力学的歪を電流として検知する技術(カンチレバー方式)。(2)ガス分子の吸着による抵抗変化を検出する技術(半導体センサー方式)。(3)遺伝子操作したマウスの臭覚受容体に匂いを分子を通して蛍光を検出する技術(バイオチップ方式)。(4)変動電場中に希薄気体を通すと、特定の質量断面積比の分子だけが検知器に到達することを利用した技術(対称場イオン移動分析方式)。
一方、内包フラーレンを用いた技術として、本発明者は、特許文献1に記載の内包フラーレンを用いた光電変換装置を提供している。その装置は、アルカリイオン内包フラーレンとアニオン性色素とからなる超分子複合体を含む内包フラーレンを用いた光電変換装置である。
例えば、食道がん患者には、2―ブタノン、酢酸、アセトン、アセトニトリルという4種類の分子が多く含まれていることがわかっている。他のがんについては、他の分子を含んでいる。また、今後、臨床の現場において数々の分子が見いだされることが予想される。
なお、図3に示す分子は肺がん患者の呼気に多く含まれるガス基質である。

一方、内包フラーレンを用いた技術として、本発明者は、特許文献1に記載の内包フラーレンを用いた光電変換装置を提供している。その装置は、アルカリイオン内包フラーレンとアニオン性色素とからなる超分子複合体を含む内包フラーレンを用いた光電変換装置である。
前述した(1)~(4)の検出技術によれば、いずれも検出濃度はよくてもppbレベルである。また、(4)の技術では」大がかりな装置を必要とする。
癌初期においては、呼気に含まれる物質濃度も微量であることを考えるとより高感度の検出技術が望まれる。
本発明は、pptレベルの濃度でも検出可能な金属イオン内包フラーレンを用いた高感度分子検出装置及を提供することを目的とする。

極めて簡単な構成にもかかわらず、例えば、癌患者の呼気に特有的に含まれる分子をpptレベルの濃度でも検出可能となる。
検出対象は、癌患者の呼気に特有的に含まれる分子に限らない、気体、固体、液体のいずれでも対象となる。

電子供与体と金属イオン内包フラーレンとの複合錯体は、ドナー・アクセプターの構成をなしており、光照射により電化分離状態を生成し、酸化力を保有する。特許文献1においては、この酸化力を利用して光電変換装置を構成した。
本発明者は、かかる特性を利用して他の装置への利用を検討したところ、エネルギーレベルを設計すれば分子の検出を行うことが可能となるとの知見を得た。
すなわち、図1に示すように、本体1内部に電子供与体2と金属イオン内包フラーレン3との超分子錯体を配置し、それを挟んで一対の電極5,6を配置する構成を基本構成とし(この構成は光電変換装置とほぼおなじである。)つつも、検出分子のエネルギーレベルに合わせて電子供与体と金属イオン内包フラーレンとの複合錯体のエネルギーレベルを、例えば、図2に示す状態に設定しておき、所定波長の光を照射すれば、分子から離脱した電子をほぼ一対一で捕獲することができることを知見した。
図1に示す例では、電極は透明電極であり、SnO2からなる。一方の電極は本例ではPtを使用している。
図2ではHOMO(最高被占起動:電子に占有されている最もエネルギーの高い分子軌道)とLUMO(最低空起動:電子に占有されていない最もエネルギーの低い分子軌道)は次のように設定されている。

すなわち、図1に示すように、本体1内部に電子供与体2と金属イオン内包フラーレン3との超分子錯体を配置し、それを挟んで一対の電極5,6を配置する構成を基本構成とし(この構成は光電変換装置とほぼおなじである。)つつも、検出分子のエネルギーレベルに合わせて電子供与体と金属イオン内包フラーレンとの複合錯体のエネルギーレベルを、例えば、図2に示す状態に設定しておき、所定波長の光を照射すれば、分子から離脱した電子をほぼ一対一で捕獲することができることを知見した。
図1に示す例では、電極は透明電極であり、SnO2からなる。一方の電極は本例ではPtを使用している。
図2ではHOMO(最高被占起動:電子に占有されている最もエネルギーの高い分子軌道)とLUMO(最低空起動:電子に占有されていない最もエネルギーの低い分子軌道)は次のように設定されている。

かかる設定においては、基底状態(図2上段)では、検出知将分子を導入しても電子の移動は生じない。
一方、励起状態、すなわち、図2下段に示すように、光を照射した状態では、色素においては、HOMOの電子は、励起されてLUMOの準位に移動し、HOMOに空席が生じる。すなわち、電荷分離状態が生成され、酸HOMO化力を保有する。
光励起状態において、検出対象分子がイオン化すれば、分離した電子は、色素の空席に移動する。一方、色素において励起された電子は金属イオン内包フラーレンのLUMOに移動し、電流が流れる。
仮に、呼気500cc中に1pptだけ検出対象分子が含まれているとする。500ccの呼気に含まれる分子の数は、
5L/25.36L×6.02×1023≒1.19×1022個である。
1pptだけ検出対象分子が含まれているとすると、その個数は、
1.19×1022×10-12≒1.19×1010個である。
1電子酸化で放出する電荷量は、
6×10-19C×1.19×1010≒1.9×10-9C
この電荷量を1秒間で流せば、1.9nA=10pAである。
他の検出技術では、間接的に(例えば歪から)電気信号に変換しているのに対し、本発明においては、検出対象分子から電子を奪い、機械的量に変化することなく直接電気信号に変化するため、極めて感度の高い検出装置が達成される。
なお、色素としては、π電子系化合物、例えば、ポルフィリン類、金属キレート化合物、ポリアニリン類、芳香族多環化合物、ポリアセン系骨格構造を有する化合物などが例示できる。
一方、励起状態、すなわち、図2下段に示すように、光を照射した状態では、色素においては、HOMOの電子は、励起されてLUMOの準位に移動し、HOMOに空席が生じる。すなわち、電荷分離状態が生成され、酸HOMO化力を保有する。
光励起状態において、検出対象分子がイオン化すれば、分離した電子は、色素の空席に移動する。一方、色素において励起された電子は金属イオン内包フラーレンのLUMOに移動し、電流が流れる。
仮に、呼気500cc中に1pptだけ検出対象分子が含まれているとする。500ccの呼気に含まれる分子の数は、
5L/25.36L×6.02×1023≒1.19×1022個である。
1pptだけ検出対象分子が含まれているとすると、その個数は、
1.19×1022×10-12≒1.19×1010個である。
1電子酸化で放出する電荷量は、
6×10-19C×1.19×1010≒1.9×10-9C
この電荷量を1秒間で流せば、1.9nA=10pAである。
他の検出技術では、間接的に(例えば歪から)電気信号に変換しているのに対し、本発明においては、検出対象分子から電子を奪い、機械的量に変化することなく直接電気信号に変化するため、極めて感度の高い検出装置が達成される。
なお、色素としては、π電子系化合物、例えば、ポルフィリン類、金属キレート化合物、ポリアニリン類、芳香族多環化合物、ポリアセン系骨格構造を有する化合物などが例示できる。
これらのエネルギーレベルは、分子軌道法により容易に求めることができる。なお、図3に示す分子のエネルギーレベルも容易に知ることができる。
分子軌道法は、各原子の原子軌道を組み合わせて1電子分子軌道を作り、それを最適化して近似性が最も高い1電子分子軌道を求める。ついで、エネルギーの低い1電子分子軌道から順に2個ずつ(スピンを逆にして)その分子が持つすべての電子を収納する。そのうえで、電子の存在確立の空間分布を計算し、電子が分子の周りにどのように広がっているかを明らかにする。このようにして分子の電子状態を知ることができる。ただ、分子の励起エネルギーとHOMO-LUMOギャップは必ずしも一致しない。HOMO、LUMOについては、分子軌道法により定性的に求めておき、より正確には、既知の検出対象分子を用いて、図1に示す装置を構成し、電流が流れるかどうかを確認することにより、図2に示すエネルギーレベルの関係が達成されているかどうかの確認をとることでき、逆に、該色素のHOMO、LUMO状態を知ることができる。
なお、色素と金属イオン内包フラーレンとの錯体は適宜の溶媒とともに容器本体1内に保持すればよい。また、電極5側に、金属イオン内包フラーレンがくるようにすることが好ましい。
分子軌道法は、各原子の原子軌道を組み合わせて1電子分子軌道を作り、それを最適化して近似性が最も高い1電子分子軌道を求める。ついで、エネルギーの低い1電子分子軌道から順に2個ずつ(スピンを逆にして)その分子が持つすべての電子を収納する。そのうえで、電子の存在確立の空間分布を計算し、電子が分子の周りにどのように広がっているかを明らかにする。このようにして分子の電子状態を知ることができる。ただ、分子の励起エネルギーとHOMO-LUMOギャップは必ずしも一致しない。HOMO、LUMOについては、分子軌道法により定性的に求めておき、より正確には、既知の検出対象分子を用いて、図1に示す装置を構成し、電流が流れるかどうかを確認することにより、図2に示すエネルギーレベルの関係が達成されているかどうかの確認をとることでき、逆に、該色素のHOMO、LUMO状態を知ることができる。
なお、色素と金属イオン内包フラーレンとの錯体は適宜の溶媒とともに容器本体1内に保持すればよい。また、電極5側に、金属イオン内包フラーレンがくるようにすることが好ましい。
本発明は、医療用分野に最適に適用されるが、分野のみならず、物質の検出が必要なあらゆる分野において適用可能である。
Claims (6)
- 容器本体内部へ検出分子を導入するための導入口を有する容器と、
前記容器の内部に包含された、色素と金属イオン内包フラーレンとの錯体と、
一対の電極と、
前記容器の内部に光を照射するための光照射手段と、
前記電極間を流れる電流を測定するための電流計と、
からなり、
光照射のない基底状態においては電子の移動はなく、
光照射による励起状態においては、
励起により生じた空席に、前記検出分子から分離した電子が移動するように電子軌道のエネルギーレベルが設定されていることを特徴とする金属イオン内包フラーレンを用いた高感度分子検出装置。 - 前記金属はアルカリ金属である請求項1記載の金属イオン内包フラーレンを用いた高感度分子検出装置。
- 前記アルカリ金属はLiである請求項1記載の金属イオン内包フラーレンを用いた高感度分子検出装置。
- 前記フラーレンはC60である請求項1ないし3のいずれか1項記載の金属イオン内包フラーレンを用いた高感度分子検出装置。
- アルカリ金属はリチウムである請求項2記載のイオン内包フラーレンはLi+@C60である請求項1又は2記載の金属イオン内包フラーレンを用いた高感度分子検出装置。
- 前記色素は、ポリ-3-ヘキシルチオフェン(P3HT)などのポリチオフェン、ポリp-フェニレン、ポリp-フェニレンビニレン、ポリアニリン、ポリピロール、PEDOT、P3OT、POPT、MDMO-PPV、MEH-PPVなどの高分子重合体、その誘導体のいずれかである請求項1ないし5のいずれか1項記載の金属イオン内包フラーレンを用いた高感度分子検出装置である。
