Description
本发明属于保健品技术领域,具体涉及一种全面抗氧化保健食品的配方及制备方法。
自由基(Free radical)是人体生命活动中各种生化反应的中间代谢产物,具有高度的化学活性,是机体有效的防御系统,若不能维持一定水平则会影响机体的生命活动。生物体内自动氧化、酶促氧化、呼吸带入、药物代谢以及受到外界电磁辐射、吸食烟草、空气污染等都会导致机体内自由基的过量产生。但自由基产生过多而不能及时地清除,它就会攻击机体内的生命大分子物质及各种细胞器,造成机体在分子水平、细胞水平及组织器官水平的各种损伤,加速机体的衰老进程并诱发各种疾病。
体内自由基过量会削弱细胞的抵抗力,使机体易受细菌和病菌感染;产生破坏细胞的化学物质,形成致癌物质;阻碍细胞的生长修复;破坏DNA、细胞膜、细胞器等;破坏蛋白质和酶类,导致炎症和衰老;破坏脂肪结构,使脂质过氧化,引起动脉粥样硬化等一系列心脑血管疾病;破坏碳水化合物,使透明质酸降解,导致关节炎等。
人体内的活性氧(ROS)主要包括过氧自由基(ROO·)、羟基自由基(·OH)、超氧阴离子(O

目前国内的抗氧化功能保健食品,并未具体对其能够清除哪种氧自由基进行确切研究及说明,抗氧化概念笼统且没有明确的评价指标。
发明内容
为了解决现有技术的不足,本发明提供了一种全面抗氧化保健食品的配方及制备方法。
本发明的目的通过以下技术方案来实现:
一种全面抗氧化保健食品,所述食品主要由以下成分按质量百分比组成:葡萄籽提取物10%~60%,虎杖提取物0.05%~30%,姜黄提取物0.05%~30%,绿茶提取物0.05%~30%,芦荟凝胶冻干粉200:10.05%~30%,柑橘生物类黄酮0.05%~30%,其中所述葡萄籽提取物中多酚含量≥85%,所述姜黄提取物中姜黄素含量≥90%,所述虎杖提取物中白藜芦醇含量≥45%,所述绿茶提取物中茶多酚含量≥90%。

S1、原料准备,按质量百分比称取原料:葡萄籽提取物10%~60%,虎杖提取物0.05%~30%,姜黄提取物0.05%~30%,绿茶提取物0.05%~30%,芦荟凝胶冻干粉200:1 0.05%~30%,柑橘生物类黄酮0.05%~30%,其中所述葡萄籽提取物中多酚含量≥85%,所述姜黄提取物中姜黄素含量≥90%,所述虎杖提取物中白藜芦醇含量≥45%,所述绿茶提取物中茶多酚含量≥90%;
S2、筛选,将准备好的原料并经过70-90目筛进行筛选,选出粒度均匀的原料待用;
S3、混合,将过筛后的原料按照等量递增的方法混合均匀;
S4、辅料添加,在S3中加入辅料,进行再次混合,制粒,干燥,制备成所需的胶囊剂型或片剂所述辅料为微晶纤维素10%~50%,硬脂酸镁5%~40%,二氧化硅2%~35%。
本发明的有益效果体现在:本发明中的保健配方通过多种抗氧化物的组合,发挥了协同抗氧化作用,并通过可靠的科学方法得到了清除不同自由基能力的佐证,可以全面缓解氧化应激反应对机体的损伤,保护机体的细胞、组织、器官,从而达到延缓衰老,预防疾病的功效。
以下结合实施例具体阐述本发明的技术方案,本发明揭示了一种 具有全面抗氧化功能的保健食品,所述抗氧化能力包括清除过氧自由基(ROO·)、羟自由基(·OH)、超氧阴离子(O2-·)、单线态氧(1O2)、过氧亚硝酸基(ONOO-)的能力。
以下实施例中葡萄籽提取物中多酚含量≥85%,姜黄提取物中姜黄素含量≥90%,虎杖提取物中白藜芦醇含量≥45%,绿茶提取物中茶多酚含量≥90%。

葡萄籽提取物97mg,虎杖提取物1.25mg,姜黄提取物1mg,绿茶提取物0.25mg,芦荟凝胶冻干粉200:1 0.25mg,柑橘生物类黄酮0.25mg,微晶纤维素120mg,硬脂酸镁45mg,二氧化硅35mg。将所述原料过80目筛,将过筛后的原料按照等量递增混合后,加入辅料,制粒,干燥,按照常规工艺制备成胶囊剂型或片剂。

一种具有全面抗氧化功能的保健食品,按质量百分比用下述原料制成:

葡萄籽提取物30mg,虎杖提取物0.15mg,姜黄提取物0.15mg, 绿茶提取物60mg,芦荟凝胶冻干粉200:1 47.7mg,柑橘生物类黄酮87mg,微晶纤维素30mg,硬脂酸镁30mg,二氧化硅15mg。将所述原料过80目筛,将过筛后的原料按照等量递增混合后,加入辅料,制粒,干燥,按照常规工艺制备成胶囊剂型或片剂。
本发明中的辅料与现有技术中制备胶囊或片剂类似,可以根据具体需求采用其他用于胶囊、片剂所需的稀释剂、润滑剂、助流剂等,在此不做具体细化。
本发明还揭示了一种以ORAC 5.0分析技术及CAA细胞抗氧化活性评价方法为依托,对所发明全面抗氧化保健食品实例1~6进行了评价和测试。验证了本发明一种全面抗氧化功能的保健食品的作用,能够清除过氧自由基(ROO·)、羟自由基(·OH)、超氧阴离子(O
2-·)、单线态氧(
1O
2)、过氧亚硝酸基(ONOO
-)。
ORAC 5.0分析技术法原理为:将检测剂(荧光探针)和活性氧/活性氮(ROS/RNS)产生剂(底物)加入样品(抗氧化剂)中,ROS/RNS攻击荧光探针,产生荧光变化(荧光强度增强或减弱),一旦受到抗氧化剂作用,体系中的抗氧化剂竞争性抑制检测剂与底物反应,从而抑制荧光变化。该方法以Trolox作为阳性对照,结合荧光强度的变化评价样品(抗氧化剂)的ROS/RNS清除能力。
细胞抗氧化活性(cellular antioxidant activity,CAA)评价方法,模拟生物体内部环境,检测待测样品氧自由基吸收能力的方法。其原理为:荧光探针DCFH-DA可以自由穿过细胞膜,进入细胞内,并被细胞内的酯酶水解生成DCFH,后者不能自由穿过细胞膜,因此就被装载入细胞内。在细胞内,活性氧自由基可以把没有荧光的DCFH氧化成有荧光的DCF。该方法以Quercetin作为阳性对照,通过检测DCF的荧光强度变化评价细胞内活性氧自由基的水平。
通过该两项技术对所发明保健食品实例1~6的自由基清除能力进行了评价和测试,ORAC值(过氧自由基清除能力)在5000~6900μmolTE/g,HORAC值(羟基自由基清除能力)在9000~16500μmolTE/g,SORAC值(超氧阴离子清除能力)在3600~5200μmolTE/g,SOAC值(单线态氧清除能力)在1200~6520μmolTE/g,NORAC值(过氧亚硝基清除能力)在260~440μmolTE/g,细胞抗氧化活性CAA值在155~240μmol QE/g,成功表明具有很明显全面的抗氧化能力。
当然本发明尚有多种具体的实施方式,在此就不一一列举。凡采用等同替换或者等效变换而形成的所有技术方案,均落在本发明要求保护的范围之内。

- 如权利要求1所述的一种全面抗氧化保健食品的制备方法,其特征在于:包括如下步骤,S1、原料准备,按质量百分比称取原料:葡萄籽提取物10%~60%,虎杖提取物0.05%~30%,姜黄提取物0.05%~30%,绿茶提取物0.05%~30%,芦荟凝胶冻干粉200:1 0.05%~30%,柑橘生物类黄酮0.05%~30%,其中所述葡萄籽提取物中多酚含量≥85%,所述姜黄提取物中姜黄素含量≥90%,所述虎杖提取物中白藜芦醇含量≥45%,所述绿茶提取物中茶多酚含量≥90%;S2、筛选,将准备好的原料并经过70-90目筛进行筛选,选出粒度均匀的原料待用;S3、混合,将过筛后的原料按照等量递增的方法混合均匀;S4、辅料添加,在S3中加入辅料,进行再次混合,制粒,干燥,制备成所需的胶囊剂型或片剂所述辅料为微晶纤维素10%~50%,硬脂酸镁5%~40%,二氧化硅2%~35%。
