Description
本発明は、目的物質を細胞質に送達するためのペプチド、細胞質送達剤及び物質導入剤に関するものである。
タンパク質、核酸の生細胞内への導入は細胞機能の計測・解析や細胞機能の調節に非常に有用なアプローチである。例えば、現在、蛍光タンパク質との融合により、種々の細胞内タンパク質の局在や挙動の観察が行われている。しかし、蛍光タンパク質の融合による細胞内局在や挙動への影響やタンパク質活性への影響は捨てきれず、また、融合タンパク質の細胞内発現量の調節も難しい。従って、これらの厳密な評価においては、複数の方法での検証が望ましい。融合する蛍光タンパク質の蛍光特性にも制約があり、適切な蛍光団で化学標識したタンパク質を細胞内に導入できればこの問題の解決に資する。また、近年、天然タンパク質の認識能を活用した種々のバイオセンサーが開発されてきており(非特許文献1)、細胞内計測への応用が期待されている。しかし、このような化学修飾されたタンパク質を細胞外から細胞内へと導入しようとする際に、細胞外から加えたタンパク質の少なからぬ量がエンドソームに保持され、細胞質へと放出・拡散しないために、これらのタンパク質の細胞内可視化や計測の所望の機能を発揮できない場合が多い。
一方、近年、種々のバイオ高分子医薬品の開発が急速に進んでいる。中でも抗体は非常に高い標的特異性を持つため、低分子医薬品に代わる分子標的薬として世界中で開発が進められている。しかし、一般に抗体は細胞質内に移行しないため、その標的は細胞膜上の受容体もしくは細胞外の疾患関連因子に限られているのが現状である。細胞質中には細胞骨格関連タンパク質や、キナーゼ関連因子など種々の疾病治療に際して標的となりうるものが多い。そのため、高分子の効率的細胞内導入法が確立すれば、抗体医薬の適用範囲は大幅に拡大する。さらにsiRNAなどの核酸(DNA、RNA)、医薬も細胞内導入の目的物質である。
以上により、抗体を始めとする生理活性タンパク質、核酸、医薬を生細胞の細胞質へ効果的に導入する方法の確立が求められている。
エンドソームに内包されたタンパク質や薬物を細胞質に放出する代表的なエンドソーム不安定化ペプチドとして、GALA(非特許文献2)、インフルエンザ・ヘマグルチニンHA2タンパク質由来ペプチド(非特許文献3)などが挙げられる。これらは、pH依存的膜融合ペプチドであり、エンドソーム内のpHが5程度になると膜融合性を発揮し、エンドソーム膜を傷害し、内包物を細胞質に放出するとされる。また、膜透過ペプチドとして知られるHIV-1 TatペプチドとHA2ペプチドの連結体(非特許文献4)なども報告されており、CreとTatの融合タンパク質等の生理活性タンパク質の細胞内導入に用いられた例が報告されている。
また、高い膜傷害性を有するハチ毒メリチンにおいて、塩基性アミノ酸のリジンをpH感受性のマレイン酸誘導体で保護し、エンドソーム内でのpH低下により保護基が脱離することにより、エンドソーム膜を選択的に損傷させ、エンドソーム内包物を細胞質に放出される試みも報告されている(非特許文献5)。
さらに、ポリエチレンイミンなどのポリカチオン性ポリマーのプロトン化の程度がpHの低下に伴い上昇することにより、エンドソームの膨潤を誘起し、内容物(核酸など)を細胞質に放出することが報告されている(プロトンスポンジ効果:非特許文献6)。また種々のpH感受性ポリマーが遺伝子導入に用いられている(特許文献1)。
特許文献2は、抗体をはじめとするタンパク質を生細胞に導入することができるペプチドを開示しているが、その導入効率に改善の余地があった。

効果的な細胞機能の計測・解析のためには、細胞外から導入する生理活性タンパク質、核酸、医薬、あるいは蛍光標識タンパク質やセンサー分子の大部分が機能・活性発現の場である細胞質に導入できることが望ましい。しかし、従来の方法では、細胞の生理的取込機構であるエンドサイトーシスにより取り込まれたこれらの分子を、満足できる効率でエンドソームから細胞質へ放出することができないことが問題点であった。バイオ医薬品の細胞内への送達という観点からも、エンドサイトーシスで取り込まれた抗体などのタンパク質、核酸、医薬を効果的に細胞質に送達することが期待されるが、抗体などのタンパク質、核酸、医薬を満足できる効率で細胞質に放出できる方法は報告されていない。
本発明は、エンドサイトーシスで取り込まれたタンパク質、核酸、医薬などの目的物質を高効率で細胞質に送達することを目的とする。
本発明者は、塩基性両親媒性ペプチドLycotoxinのアミノ酸の一部を酸性アミノ酸と置換することで、抗体やセンサー分子、核酸、医薬などの目的物質のエンドソームから細胞質への放出を亢進させ得ることを見出した。
本発明は以下のペプチド、タンパク質、核酸、医薬などの目的物質の細胞質送達剤及び物質導入剤を提供するものである。

本発明のペプチドは、細胞表面と効果的に相互作用することにより、抗体などのタンパク質、核酸、医薬などの目的物質(細胞内導入分子)とともに、エンドソーム内に取り込まれる効果を奏する。
本発明のペプチドは、エンドソーム内におけるpHの減少ならびにエンドソーム成熟化に伴う膜構成成分の変化に伴いエンドソーム膜の不安定化を誘起し、ペプチドとともにエンドソーム内に移行した抗体やセンサー分子、核酸、医薬などの目的物質の細胞質への放出を促進する効果を奏する。
本発明のペプチドは、蛍光標識をはじめとした化学修飾を施したタンパク質やセンサー分子などの目的物質の細胞質内への送達を通して、当該目的物質の細胞内局在や挙動、また、細胞内環境の計測・制御を可能にする効果を奏する。
本発明のペプチドは、抗体、核酸のようなサイズの大きな生理活性物質を細胞質内に送達する効果を奏する。
本発明の第1の好ましい実施形態は、12位と16位のヒスチジン残基のアラニン置換である。
特許文献2のL17Eは二つのヒスチジン(H)を含んでいる。ヒスチジンは細胞外のpHでは荷電せず、エンドソーム内のpHでは正に荷電すると考えられる。L17Eのエンドソーム膜選択的な傷害性は、ヒスチジンの正電荷により向上すると考えられたが、意外にもヒスチジン残基により低下することが分かった。このため、12位と16位のヒスチジン残基をアラニンで置換することでエンドソーム膜選択的な傷害性の向上を図った。
2つのヒスチジン残基をアラニンで置換したHA/E1はL17Eに比べ高いヘリックス性を示し(図1)、高分子薬物モデルとしてのAlexa488-dextranのエンドソームからの放出活性は大きく向上した(図2)。
さらに、21位をGlu(E)で置換したH12A/H16A/L17E/Q21E置換体(=HA/E2)は、酸性脂質膜存在下で高いヘリックス性を示し(図1)、HA/E1と同等のエンドソーム内包物放出活性を有していた(図2)。また、L17Eに比べて有意に高いエンドソーム内包物放出活性を有していた。
12位、16位のヒスチジンをアラニン以外の親水性アミノ酸(グルタミン、セリン)に置換したH12Q/H16Q/L17E/Q21E、H12S/H16S/L17E/Q21Eの活性はL17Eに比べて劣るものであり、ヒスチジンの置換は、アラニンが望ましいと結論された。
本発明の第2の好ましい実施形態は、17位(E)、21位(Q)の2-アミノアジピン酸(Aad)、2-アミノピメリン酸又は2-アミノスベリン酸への置換である。
さらなるペプチドのヘリックス構造の安定化と膜親和性の向上を期待して、細胞膜に対してのエンドソーム膜の選択的不安定化に重要な役割を担うL17E中の17位のグルタミン酸(E)を、側鎖のメチレン鎖が一つ多い2-アミノアジピン酸(Aad)に置換した誘導体(L17Aad)を合成した。ヒスチジン残基が配列中に存在しているために、L17Aadのヘリックス含量自体はあまり向上しなかったが(図1)、図3に示すようにL17Eに比べて有意に高いエンドソーム内包物放出活性を示すことが明らかとなった。
本発明の第3の好ましい実施形態は、12位と16位のヒスチジン残基をアラニン置換し、さらに17位のグルタミン酸残基の2-アミノアジピン酸(Aad) 2-アミノピメリン酸又は2-アミノスベリン酸への置換である。
第1及び第2の好ましい実施形態で得られた結果を基に、L17Eのヒスチジンをアラニンに置換すると共に17位のグルタミン酸をアミノアジピン酸に置換したペプチドHAadを合成した。HAadは図1に示すように酸性脂質膜存在下に高いヘリックス性を示すことが明らかとなり、図4ならびに実施例に示すようにL17Eに比べてエンドソームに取り込まれた薬物を格段に効果的に細胞質に放出する活性を有することが明らかとなった。
本発明の第4の好ましい実施形態は、5個のLys(K)残基のArg(R)、ホモアルギニン、オルニチン、ホモリジン又は2-amino-7-guanidinoheptanoic acidへの置換である。
本発明のペプチドは、特許文献2に記載の以下のL17Eの誘導体である。

細胞内に導入される目的物質は特に以上のものに限定される必要はなく、膜透過性を有するペプチド、各種トランスフェクション試薬など他の細胞導入剤と併用し、これらの細胞質内移行効率の更なる向上のために本発明を用いることも可能である。
R1とR2がともに目的物質ではない本発明のペプチドは、目的物質と組み合わせて組成物、好ましくは医薬組成物として脊椎動物、好ましくはヒトを含む哺乳動物に投与してもよい。目的物質が本発明のペプチドに結合していない場合、目的物質はタンパク質、ペプチド、核酸、医薬、糖あるいはこれらの標識物質、合成高分子、リポソーム、有機/無機/半導体微粒子などから構成され、スペーサーを含まない。
本発明のペプチドと目的物質を含む組成物または医薬組成物において、目的物質100質量部に対し、本発明のペプチドを1~10000質量部、好ましくは5~200質量部、より好ましくは10~200質量部含む。
本発明のペプチドは、目的物質に直接あるいは適当なスペーサーを介して共有結合的に結合してもよく、直接あるいは目的物質と相互作用する他分子を介して非共有結合的に結合して複合体を形成してもよい。さらに、エンドソームに内包されるベクター(細胞導入剤)に含ませてもよい。例えば、本発明のペプチドは、ベクターの内部に含まれていてもよく、ベクターの構成成分に直接又はスペーサーを介して結合されていてもよい。あるいは、本発明のペプチドがベクターの構成成分と相互作用する他分子を介して非共有結合的に複合体を形成することで、ベクターの表面に含ませてもよい。さらにベクターの内部に目的物質とともに本発明のペプチドを内包させて、目的物質がエンドソームに取り込まれた後に、エンドソームから細胞質への移行を促進することができる。本発明のペプチドと目的物質が非共有結合的に結合した複合体は、本発明の組成物または医薬組成物に包含される。

本明細書において、スペーサーとしては、エステル結合(-CO-O-、-O-CO-)、エーテル結合(-O-)、アミド結合(NHCO, CONH)、糖鎖リンカー、ポリエチレングリコールリンカー、ペプチドリンカーなどが挙げられる。ペプチドリンカーとしては、タンパク質を構成する天然の20種のアミノ酸の少なくとも1種を含むリンカーが挙げられ、ペプチドリンカーのアミノ酸の数としては、1~20個、1~15個、1~12個、1~10個、1~8個、1~6個あるいは1~4個が挙げられるがこれらに限定されない。ペプチドリンカーとして、アルギニンダイマー、アルギニントリマー、アルギニンテトラマー、リジンダイマー、リジントリマー、リジンテトラマー、グリシンダイマー、グリシントリマー、グリシンテトラマー、グリシンペンタマー、グリシンヘキサマー、アラニン-アラニン-チロシン(AAY)、イソロイシン-ロイシン-アラニン(ILA)、アルギニン-バリン-リジン-アルギニン(RVKR)等が例示される。スペーサーは2価であっても多価であってもよい。

本発明のペプチドが多量体の場合、デンドリマーのような分岐型の多価リンカー、あるいは金属錯体等で連結されたものであってもよい。デンドリマーは、例えば以下の式(IB)で表わされる。

本発明のペプチドを目的物質と共存させると、目的物質のエンドソームから細胞質への移行を促進することができるが、特定の細胞へ導入するための特異性はない。したがって、本発明のペプチドと目的物質をDDSにより標的細胞に供給するか、細胞特異的なベクターと本発明のペプチドを組み合わせることにより、特定細胞の細胞質に目的物質を導入することができる。細胞特異的なベクターとして、細胞特異的な抗体、リガンドなどが表面に導入されたベクターが例示される。ベクターとしてはリポソーム(カチオニックリポソーム、アニオニックリポソーム)、リピッドマイクロスフェア、リポフェクタミン、親水性セグメントと内核形成セグメント(疎水性セグメント、カチオン性セグメント、金属錯体形成セグメント等)を含むブロックコポリマーから構成される高分子ミセル、高分子中空キャリア(特許第5142313号に記載のポリサルコシン誘導体を含む薬物担体、WO2004/105799に記載の非荷電性セグメントと荷電性セグメントとを有するブロック共重合体からなる静電結合型高分子ミセルなど)、ナノゲル、高密度リポタンパク質(HDL)、合成ポリマーやナノ粒子などが挙げられる。自己集合型核酸由来ベクター、ウイルス外殻タンパク質由来ベクターであってもよい。例えばベクターがリポソームの場合には、R1として炭素数10以上のアシル基を使用するか、N末端にコレステリルオキシカルボニル基やC末端にコレステリル基などの脂溶性の大きい基、あるいはホスファチジルエタノールアミンなどのリン脂質を必要に応じて適当なスペーサーを介してエステル結合又はアミド結合で本発明のペプチドと結合させることで、本発明のペプチドを含むリポソームを得ることができる。スペーサーとしては、PEG(ポリエチレングリコール)、アミド基(-CONH-、-NHCO-)、エステル基(-COO-,-O-CO-)、エーテル基(-O-)、アミノ基(-NH-)、アルキレン(メチレン、エチレン、プロピレン、ブチレン、ペンチレン、ヘキシレンなど)、アミノ酸などが挙げられる。アミノ酸は側鎖のCOOHまたはNH2基を介して結合してもよい。ウイルス(外殻タンパク質)由来ベクターとしては、レトロウイルスベクター、アデノウイルスベクター、センダイウイルスベクター等が挙げられる。自己集合型核酸由来ベクターとしては、例えばWO 2012144560 A1、WO2016/006628に記載のものが挙げられる。「本発明のペプチドの標的細胞へ親和性を高める分子」と本発明のペプチドとのコンジュゲ-ションにより、当該細胞のエンドソーム内に本発明のペプチドをより効率的/効果的に移行・蓄積させることで、エンドソーム膜の不安定化効果と目的物質の細胞質への送達効果を高めることも可能である。「本発明のペプチドの標的細胞への親和性を高める分子」としては、細胞の表面に提示される受容体(葉酸受容体、トランスフェリン受容体、グルコース等の糖類の受容体、上皮細胞増殖因子(EGF)や血管内皮増殖因子(VEGF)などの増殖因子受容体等)に結合するリガンド、あるいは脂肪酸、疎水性ペプチド、ポリアルギニンなどの塩基性ペプチド、糖鎖などがある。
さらに、本発明のペプチドと目的物質は、直接、あるいは目的物質と相互作用する他分子を介して非共有結合的な複合体を形成することができる。このような非共有結合的な複合体としては、

目的物質を細胞に送達すべき生物種は、脊椎動物、好ましくは哺乳動物である。哺乳動物としては、例えば、ヒト、サル、ウシ、ヒツジ、ヤギ、ウマ、ブタ、ウサギ、イヌ、ネコ、ラット、マウス、モルモット等が挙げられる。

HAad (40 μM)存在下にHeLa細胞を高分子薬物モデルとしてのAlexa488-dextran (10 kDa)(200 μg/mL)とα-MEM(-)培地中で1 hインキュベーションすることにより、約75%の細胞内(サイトゾル)へのAlexa488-dextranの放出が見られた。一方、L17Eを用いた際には約50%であった。
HAad (40 μM)存在下にHeLa細胞を高分子薬物モデルとしてのAlexa488-dextran (10 kDa)(200 μg/mL)とα-MEM(-)培地中で1 hインキュベーションすることにより、約75%の細胞内(サイトゾル)へのAlexa488-dextranの放出が見られた。一方、L17Eを用いた際には約50%であった。
細胞内に放出されたAlexa488-dextranのシグナルを50%の細胞に対して認めるために必要なペプチド濃度は、HAadが20 μMであるのに対し、L17Eは40 μMであった(EC50が1/2)。
実施例7.デキストラン(10 kDa)の細胞内放出効率比較 (2)
HeLa細胞に較べて細胞内へのデキストランの放出が難しいNIH3T3細胞において、実施例6と同様に、HAad、HA/E2、L17E (各40 μM)存在下に細胞とデキストラン(10 kDa)をDMEM(-)培地中で1 hインキュベーションしたところ、全体のそれぞれ約35%、25%、17%の細胞で細胞内へのデキストランの放出が認められた(HAad処理による細胞内放出はL17Eによるものの2倍)。
HeLa細胞に較べて細胞内へのデキストランの放出が難しいNIH3T3細胞において、実施例6と同様に、HAad、HA/E2、L17E (各40 μM)存在下に細胞とデキストラン(10 kDa)をDMEM(-)培地中で1 hインキュベーションしたところ、全体のそれぞれ約35%、25%、17%の細胞で細胞内へのデキストランの放出が認められた(HAad処理による細胞内放出はL17Eによるものの2倍)。

実施例6と同様に、HAad、HA/E2、L17E (各40 μM)存在下に細胞とAlexa488で蛍光標識したヒト免疫グロブリン(IgG、~160 kDa)(500 μg/mL)をα-MEM(-)培地中で1 hインキュベーションしたところ、全体のそれぞれ約70%、45%、35%の細胞で細胞内へのヒト免疫グロブリンの放出が認められた(図7、HAad処理による細胞内放出はL17Eによるものの2倍)。
実施例6と同様に、HAad、HA/E2、L17E (各40 μM)存在下に細胞とAlexa488で蛍光標識したヒト免疫グロブリン(IgG、~160 kDa)(500 μg/mL)をα-MEM(-)培地中で1 hインキュベーションしたところ、全体のそれぞれ約70%、45%、35%の細胞で細胞内へのヒト免疫グロブリンの放出が認められた(図7、HAad処理による細胞内放出はL17Eによるものの2倍)。

HAad、L17E (各40 μM)存在下に細胞とAlexa488で蛍光標識したヒト免疫グロブリン(IgG、~160 kDa)(50 μg/mL:実施例8の1/10量)をα-MEM(-)培地中で1 hインキュベーションしたところ、HAadで処理した細胞では、全体の約50%の細胞で細胞内へのヒト免疫グロブリンの放出が認められたのに対し、L17Eで処理した場合は約10%の細胞でしかこれが認められなかった(図8,HAad処理による細胞内放出はL17Eによるものの5倍。また、50%の細胞内でIgGの放出が認められるのに必要なIgG量は1/10)。
実施例10.ヒト免疫グロブリン(IgG、~160 kDa)の細胞内放出効率比較 (3)
実施例8及び9で、HAadがより低いIgG投与量でも高いエンドソーム内包IgGの細胞内への放出能を有することが確認された。これを基に、投与するペプチド量が低減可能かに関して検討した。

HAad、L17E (各20 μM:実施例8及び9の1/2量)存在下に細胞とAlexa488で蛍光標識したヒト免疫グロブリン(IgG、~160 kDa)(500 μg/mL:実施例8と同量)をα-MEM(-)培地中で1 hインキュベーションしたところ、HAad 20 μMで処理した細胞では、全体の約37%の細胞で細胞内へのヒト免疫グロブリンの放出が認められたのに対し、L17Eで処理した場合は約15%の細胞でしかこれが認められなかった(図9,低濃度におけるHAad処理による細胞内放出はL17Eによるものの2.5倍。また、細胞内に放出されたヒト免疫グロブリンのシグナルを35%の細胞に対して認めるために必要なペプチド濃度は、L17Eが40 μMであるのに対し、HAadはその半分の20 μMである)。
実施例11.毒素タンパク質saporinの細胞内送達による殺細胞アッセイ
細胞内への送達によりがん細胞を死滅される細胞毒としてsaporinが知られている。HAad、HA/E2、L17E (各40 μM)存在下にRAW264.7細胞とsaporin (10 μg/mL) をα-MEM(-)培地中で1 hインキュベーションし、その後、細胞を洗浄し、培地をDMEM(+)に交換し更に6 hインキュベーションした。WST-8 assayで細胞生存率を検定した(図10)。この結果、HAadおよびHA/E2存在下でsaporinを処理した細胞の生存率はそれぞれ4、6%とL17E存在下での生存率 (16%)と比較して有意に減少し、またHAad存在下の生存率は、L17E存在下の1/4であった。
細胞内への送達によりがん細胞を死滅される細胞毒としてsaporinが知られている。HAad、HA/E2、L17E (各40 μM)存在下にRAW264.7細胞とsaporin (10 μg/mL) をα-MEM(-)培地中で1 hインキュベーションし、その後、細胞を洗浄し、培地をDMEM(+)に交換し更に6 hインキュベーションした。WST-8 assayで細胞生存率を検定した(図10)。この結果、HAadおよびHA/E2存在下でsaporinを処理した細胞の生存率はそれぞれ4、6%とL17E存在下での生存率 (16%)と比較して有意に減少し、またHAad存在下の生存率は、L17E存在下の1/4であった。
実施例12. Creタンパク質の細胞内送達による遺伝子組換えアッセイ
特許文献2の実施例と同様にして作製したloxP-DsRed-loxP-EGFPをコードするプラスミドをトランスフェクションしたHeLa細胞に、His-tag融合Cre (Cre-His6) (10 μM)をHAad、HA/E2、L17E (各40 μM)存在下、α-MEM(-)中1 hインキュベーションした。この系は、Creタンパク質が細胞内に放出されることで遺伝子組換えが誘起されるとEGFPが発現するように設計してある。細胞を洗浄し、α-MEM(+)で24 h培養後、共焦点顕微鏡観察した(図11)。ペプチド処理細胞群では、コントロール細胞群 (Cre-His6のみ投与)に比べ有意にEGFP発現細胞の割合が増加した。HAad、HA/E2、L17E処理時のGFP発現細胞の割合はそれぞれ41%、36%、30%であった。放出ペプチド非投与時のEGFP発現効率は9%であったことを考慮すると、ペプチド投与時には遺伝子組換えが、非投与時に較べ32、27、22%上昇したと考えることが可能であり、HAad投与時にはL17E投与時にくらべ有意に遺伝子組換えが促進された。
特許文献2の実施例と同様にして作製したloxP-DsRed-loxP-EGFPをコードするプラスミドをトランスフェクションしたHeLa細胞に、His-tag融合Cre (Cre-His6) (10 μM)をHAad、HA/E2、L17E (各40 μM)存在下、α-MEM(-)中1 hインキュベーションした。この系は、Creタンパク質が細胞内に放出されることで遺伝子組換えが誘起されるとEGFPが発現するように設計してある。細胞を洗浄し、α-MEM(+)で24 h培養後、共焦点顕微鏡観察した(図11)。ペプチド処理細胞群では、コントロール細胞群 (Cre-His6のみ投与)に比べ有意にEGFP発現細胞の割合が増加した。HAad、HA/E2、L17E処理時のGFP発現細胞の割合はそれぞれ41%、36%、30%であった。放出ペプチド非投与時のEGFP発現効率は9%であったことを考慮すると、ペプチド投与時には遺伝子組換えが、非投与時に較べ32、27、22%上昇したと考えることが可能であり、HAad投与時にはL17E投与時にくらべ有意に遺伝子組換えが促進された。
実施例13.ペプチド処理におけるミトコンドリアに対する傷害性
ミトコンドリア膜は細胞膜に比べより酸性脂質に富んでいると言われていることから、上記のペプチドのミトコンドリア膜への影響をJC-1アッセイにより調べた。JC-1試薬はミトコンドリア膜が健常に保たれ、膜電位が保たれている場合は赤色(J-aggregate)の、ミトコンドリア膜が傷害され、膜電位が正常に保たれていない場合は緑色(J-monomer)の蛍光を発する。HeLa細胞をα-MEM(-)中、HAad、HA/E2、L17E (40 μM)と1 hインキュベーションした後、JC-1アッセイを行い、フローサイトメトリーにより赤色蛍光を発する細胞の割合を検定した。ATP合成を阻害する脱共役剤(carbonyl cyanide-p-trifluoromethoxyphenylhydrazone, FCCP) [α-MEM(-)中、40μM、1 hインキュベーション]、カチオン性の遺伝子のトランスフェクション試薬であるリポフェクタミン LTXとpCI vector (空ベクター)との複合体(標準プロトコルに従いα-MEM(+)中、一夜インキュベーション)の影響に関しても同様に検討した(図12)。
ミトコンドリア膜は細胞膜に比べより酸性脂質に富んでいると言われていることから、上記のペプチドのミトコンドリア膜への影響をJC-1アッセイにより調べた。JC-1試薬はミトコンドリア膜が健常に保たれ、膜電位が保たれている場合は赤色(J-aggregate)の、ミトコンドリア膜が傷害され、膜電位が正常に保たれていない場合は緑色(J-monomer)の蛍光を発する。HeLa細胞をα-MEM(-)中、HAad、HA/E2、L17E (40 μM)と1 hインキュベーションした後、JC-1アッセイを行い、フローサイトメトリーにより赤色蛍光を発する細胞の割合を検定した。ATP合成を阻害する脱共役剤(carbonyl cyanide-p-trifluoromethoxyphenylhydrazone, FCCP) [α-MEM(-)中、40μM、1 hインキュベーション]、カチオン性の遺伝子のトランスフェクション試薬であるリポフェクタミン LTXとpCI vector (空ベクター)との複合体(標準プロトコルに従いα-MEM(+)中、一夜インキュベーション)の影響に関しても同様に検討した(図12)。
結果として、FCCP処理の細胞ではほとんどの細胞が緑色の蛍光を発するように変化した。一方、L17E処理細胞では赤色を示す細胞の割合がペプチド未処理の場合と比べて僅かに低下したのに対して、HA/E2、HAad処理した場合には未処理の場合と有意な差が見られなかった。したがって、これらのペプチドのミトコンドリア膜に対しての影響はほとんど無く、HA/E2、HAadの傷害性はL17Eよりも一層低いことが示唆された。汎用されているリポフェクタミンには若干のミトコンドリア傷害活性を有するが、本発明のペプチドHA/E2、HAadにはミトコンドリア傷害性がみられないことが示された。
実施例14. 核局在化シグナル融合緑色蛍光タンパク質 (NLS-EGFP) の核集積
実施例6と同様に、HAad, L17E (各40 μM) 存在下に細胞とNLS-EGFP (10 μM) をα-MEM(-)培地中で1 hインキュベーションした。NLS-EGFPが活性を保持したまま細胞内に放出されていれば、NLSにより緑色の蛍光が核に局在する様子が観察される。結果として、全体のそれぞれ約84%、56%の細胞で核へのNLS-EGFPの局在が認められた (図13)。HAadの高いエンドソーム内包物放出活性をこの系でも確認することができた。
実施例6と同様に、HAad, L17E (各40 μM) 存在下に細胞とNLS-EGFP (10 μM) をα-MEM(-)培地中で1 hインキュベーションした。NLS-EGFPが活性を保持したまま細胞内に放出されていれば、NLSにより緑色の蛍光が核に局在する様子が観察される。結果として、全体のそれぞれ約84%、56%の細胞で核へのNLS-EGFPの局在が認められた (図13)。HAadの高いエンドソーム内包物放出活性をこの系でも確認することができた。
実施例15. 血清存在下ペプチド処理24時間後の細胞生存率の評価
HAad, HA/E2, L17E (各10, 20, 40 μM) で細胞をα-MEM(+)培地中で24 hインキュベーションした後の細胞生存率をWST-8 assayにより評価した (図14)。どのペプチドのどの濃度においても24時間後の生存率はほぼ100%で、L17Eと同様に、HA/E2およびHAadに顕著な毒性は見られなかった。

実施例16
1)H-IWLTALZFLGZAAAZAXAZQXLSZL-amide [HAad-homoR] (Z=homoarginine、X= L-2-aminoadipic acid)の調製
Francesco L. Brancia., et al., Anal. Chem., 2004, 76 (10), 2748-2755に従って標記化合物を合成した。ヘミ硫酸O-メチルイソ尿素(1.23 g)の水溶液にアンモニア水を添加しpH 11.0とする。HAad[H-IWLTALKFLGKAAAKAXAKQXLSKL-amide]の水溶液(1.3 mM, 38 μL)をこれに加え、ヘミ硫酸O-メチルイソ尿素、1cの最終濃度がそれぞれ0.5M、5 μM、総液量10 mLとする。室温一夜反応。1% トリフルオロ酢酸水溶液を同体積加えて反応停止後、凍結乾燥、逆相HPLCで精製し、質量分析(MALDI TOFMS)で目的物が得られたことを確認した[実測値2965.8; 理論値 2965.8 (M+H)+]。
2)H-IWLTALZFLGZAAAZAXAZQXLSZL-amideのAlexa488-dextran (10 kDa)の細胞質への放出活性
HeLa細胞とAlexa488-dextran (Molecular Probes、10 kDa) (200 μg/mL)とHAad、あるいはHAad-homoR (20 μM)をα-MEM(-)中1 h処理、細胞を洗浄し、共焦点顕微鏡で観察した。HAad処理細胞では約40%の細胞で細胞質へのAlexa488-dextranの放出が見られたのに対し、HAad-homoR処理細胞では75%の細胞でこれが観察された。

HeLa細胞とAlexa488-dextran (Molecular Probes、10 kDa) (200 μg/mL)とHAad、あるいはHAad-homoR (20 μM)をα-MEM(-)中1 h処理、細胞を洗浄し、共焦点顕微鏡で観察した。HAad処理細胞では約40%の細胞で細胞質へのAlexa488-dextranの放出が見られたのに対し、HAad-homoR処理細胞では75%の細胞でこれが観察された。
5個のLys(K)のアミノ基をグアニジノ基に変換してホモアルギニンにすることにより、細胞内に導入されたタンパク質の細胞質への放出活性が顕著に増大することが明らかになった。
