본 발명자들은 임상에서 사용가능하도록 유전자를 도입하지 않으며 비신경세포 특이적 유전자가 발현되지 않는 보다 안전하고 안정적인 유도 신경줄기세포를 제조하는 방법을 개발하고자 예의 연구노력한 결과, 분화가 완료된 비신경 세포를 특정 성분이 포함된 배지에서 배양할 경우, 상기 비신경 세포를 유도 신경줄기세포로 전환시킬 수 있음을 확인하고, 본 발명을 완성하였다.
본 발명의 하나의 목적은 비신경 세포의 유도 신경줄기세포로의 전환유도용 조성물을 제공하는 것이다.

도 8은 유도 신경줄기세포를 마우스의 뇌에 주입하고 4주 후에 in vivo 생존과 분화에 대한 분석을 형광면역염색을 통해서 분석한 결과를 나타내는 도이다. 구체적으로, 3가지 대표적 신경세포인 뉴런 (도 8a: Tuj1; green), 별아교세포 (도 8b: astrocyte (GFAP; green)), 희소돌기아교세포 (도 8c: oligodendrocyte (MBP; green))로 분화한 것을 면역형광염색을 확인한 결과이다.
본 발명자들은 보다 안전하고 안정적인 유도 신경줄기세포를 제조하는 방법을 개발하고자 다양한 연구를 수행하던 중, 5-AZA(5-Azacytidine)를 포함하는 배지에 분화가 완료된 비신경 세포를 접종하고 배양하면, 상기 비신경 세포가 리프로그래밍되어 유도 신경줄기세포로 전환됨을 확인하였다. 아울러, 상기 5-AZA의 전환 효율을 향상시킬 수 있는 보조성분을 검색한 결과, CHIR99021, Forskolin 및 SB431542를 상기 배지에 모두 추가할 경우, 유도 신경줄기세포가 안정적으로 제조됨을 확인하였다. 그러나, 상기 3가지 추가성분을 부분적으로 배지에 추가할 경우에는 비신경 세포의 일부가 유도 신경줄기세포로 전환되지 않음을 확인하였다.
따라서, 비신경 세포를 리프로그래밍하여 유도 신경줄기세포를 제조할 경우, 5-AZA가 필수적인 역할을 수행하며, 이를 보조하는 성분으로는 CHIR99021, Forskolin 및 SB431542를 사용할 수 있음을 알 수 있었다. 상기 5-AZA가 포함된 배지를 사용하여 비신경 세포를 유도 신경줄기세포로 전환하는 기술은 지금까지 전혀 알려지지 않았고, 본 발명자들에 의하여 최초로 개발되었다.
상술한 목적을 달성하기 위한 하나의 양태로서, 본 발명은 5-AZA(5-Azacytidine)를 포함하는 비신경 세포의 유도 신경줄기세포로의 전환유도용 조성물을 제공한다.
본 발명의 용어 "5-AZA(5-Azacytidine)"란, 하기 화학식 1의 구조식을 갖는 시티딘의 유도체 화합물을 의미한다. 상기 5-AZA는 골수이형성 증후군(Myelodysplastic syndrome)의 치료제로서 사용된다.
본 발명에 있어서, 상기 5-AZA는 비신경 세포를 유도 신경줄기세포로 전환시키는 유효성분으로 사용될 수 있는데, 일 예로서, 비신경 세포 배양용 배지에 5-AZA를 첨가하여 사용할 수 있고, 이때, 배지에 첨가되는 양은 특별히 제한되지 않는다.

본 발명의 용어 "비신경 세포"란, 신경세포가 아닌 모든 분화 또는 미분화된 세포를 의미하며, 본 발명의 조성물 또는 방법이 적용될 수 있는 표적세포 역할을 한다. 본 발명의 비신경 세포는 인간, 원숭이, 돼지, 말, 소, 양, 개, 고양이, 생쥐 및 토끼 등의 다양한 동물로부터 유래한 세포일 수 있으며, 바람직하게는 인간으로부터 유래한 세포일 수 있으나, 이에 의하여 유도 신경줄기세포로 리프로그래밍시킬 수 있는 세포가 제한되는 것은 아니다.
본 발명에 있어서, 상기 비신경세포는 체세포 등이 될 수 있다.
본 발명에서 "체세포"는 분화능 및 자가재생능이 제한된 성체를 구성하는 세포를 의미하는데, 구체적으로는 상기 체세포는 인간의 피부, 모발, 지방, 혈액 등을 구성하는 체세포일 수 있고, 바람직하게는 인간 섬유아세포 또는 인간 제대혈 세포일 수 있으나, 이에 제한되지는 않는다.
본 발명의 용어 "유도 신경줄기세포"란, 분화된 세포에 리프로그래밍 기술을 이용하여 신경줄기세포와 유사 또는 동일한 다분화능(pluripotency)을 가진 미분화 상태의 줄기세포를 확립하는 방식으로 만들어진 세포를 의미한다. 상기 유도 신경줄기세포는 신경줄기세포와 동일 또는 유사한 특성을 가지고 있는데, 구체적으로는 비슷한 세포형태를 보여주고, 유전자 및 단백질 발현 패턴이 유사하며, 생체 내외에서 다분화능을 가질 수 있다.
본 발명에 있어서, 상기 유도 신경줄기세포는 신경세포(뉴런), 별아교세포, 희소돌기아교세포, GABA성 신경 세포 또는 도파민성 신경 세포 등으로 분화가능한 미분화 세포일 수 있다.
본 발명에서 제공하는 전환유도용 조성물은 섬유아세포 등과 같은 다양한 비신경 세포에 작용하여 상기 비신경 세포 전체를 유도 신경줄기세포로 전환시키는데 사용될 수 있는데, 상기 조성물은 유효성분인 5-AZA 이외에도 CHIR99021, Forskolin 및 SB431542를 추가로 포함할 수 있다. 상기 5-AZA와 CHIR99021, Forskolin 및 SB431542를 모두 포함하는 배지를 사용할 경우에는, 비신경 세포를 모두 유도 신경줄기세포로 전환시킬 수 있으나, 상기 상기 5-AZA와, CHIR99021, Forskolin 및 SB431542 중의 한가지 또는 두가지 성분을 포함하는 배지를 사용할 경우에는, 비신경 세포의 일부가 유도 신경줄기세포로 전환되지 않기 때문에, 가급적 상기 5-AZA와 CHIR99021, Forskolin 및 SB431542를 모두 사용함이 바람직함을 알 수 있었다.
본 발명의 용어 "CHIR99021"이란, C22H18Cl2N8의 화학식으로 표시되고, 하기 화학식 2의 구조식을 갖는 화합물을 의미한다. 상기 CHIR99021은 GSK3 저해제 및 Wnt 신호전달 경로의 활성화제로서 사용된다.
본 발명에 있어서, 상기 CHIR99021는 비신경 세포를 유도 신경줄기세포로 전환시키는 보조성분으로 사용될 수 있는데, 일 예로서, 비신경 세포 배양용 배지에 CHIR99021을 첨가하여 사용할 수 있고, 이때, 배지에 첨가되는 양은 특별히 제한되지 않는다.

본 발명의 용어 "포스콜린(Forskolin)"이란, "coleonol"이라고도 호칭되고, C22H34O7의 화학식으로 표시되며, 하기 화학식 3의 구조식을 갖는 랍데인 디테르펜(labdane diterpene)류 화합물을 의미한다. 상기 포스콜린은 cAMP의 수준을 증가시키는 제제로서 사용되고 있다.
본 발명에 있어서, 상기 Forskolin은 비신경 세포를 유도 신경줄기세포로 전환시키는 보조성분으로 사용될 수 있는데, 일 예로서, 비신경 세포 배양용 배지에 포스콜린을 첨가하여 사용할 수 있고, 이때, 배지에 첨가되는 양은 특별히 제한되지 않는다.

본 발명의 용어 "SB431542"란, C22H16N4O3의 화학식으로 표시되고, 하기 화학식 4의 구조식을 갖는 화합물을 의미한다. 상기 SB431542는 TGF-β, ALK5, ALK4, ALK7 등의 다양한 성분의 억제제로서 사용된다.
본 발명에 있어서, 상기 SB431542는 비신경 세포를 유도 신경줄기세포로 전환시키는 보조성분으로 사용될 수 있는데, 일 예로서, 비신경 세포 배양용 배지에 SB431542를 첨가하여 사용할 수 있고, 이때, 배지에 첨가되는 양은 특별히 제한되지 않는다.

다른 하나의 양태로서, 본 발명은 상기 전환유도용 조성물을 포함하는 비신경 세포의 유도 신경줄기세포로의 전환유도용 키트를 제공한다.
본 발명의 키트는 비신경 세포를 5-AZA를 포함하는 배지에서 배양하여, 유도 신경줄기세포로 전환시키는 방법에 사용될 수 있는데, 특별히 이에 제한되지 않으나, 상기 비신경 세포를 배양하기 위한 배지, 상기 비신경 세포를 배양하기 위한 배양용기, 비신경 세포의 배양에 적합한 한 종류 또는 그 이상의 다른 구성 성분 조성물, 용액 또는 장치가 포함될 수도 있고, 그 종류가 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용되는 형태의 키트를 사용할 수 있다.
구체적인 일 예로서, 본 발명의 키트는 (a) 5-AZA를 포함하는 조성물; 및 (b) 비신경 세포 배양용 배지 조성물이 각각 개별 용기에 담긴 형태, 또는 하나 이상의 구획으로 나누어진 한 개의 용기 내에 담긴 형태로 포장된 형태가 될 수 있다.
다른 일 예로서, 본 발명의 키트는 (a) 상기 5-AZA를 포함하는 제1 조성물; (b) CHIR99021, 포스콜린 및 SB431542를 포함하는 제2 조성물; 및 (c) 비신경 세포 배양용 배지 조성물이 각각 개별 용기에 담긴 형태, 또는 하나 이상의 구획으로 나누어진 한 개의 용기 내에 담긴 형태로 포장된 형태가 될 수 있다.
또 다른 하나의 양태로서, 본 발명은 비신경 세포로부터 전환된 유도 신경줄기세포를 제조하는 방법 및 상기 방법으로 제조된 유도 신경줄기세포를 제공한다.
구체적으로, 본 발명의 비신경 세포로부터 전환된 유도 신경줄기세포를 제조하는 방법은 (a) 비신경 세포를 5-AZA를 포함하는 배지에서 7 내지 14일 동안 배양하는 단계; 및 (b) 상기 (a) 단계에서 배양된 비신경 세포를 5-AZA를 포함하지 않는 배지에서 구상체(Sphere)가 형성될 때까지 배양하는 단계를 포함한다.
이때, 상기 (a) 단계 및 (b) 단계의 배양과정은 당업계에 알려진 적당한 배지와 배양조건에 따라 이루어질 수 있다. 이러한 배양 과정은 당업자라면 선택되는 세포에 따라 용이하게 조정하여 사용할 수 있다. 
일 예로서, 상기 비신경 세포의 종류는 상술한 바와 동일하고, 상기 5-AZA를 포함하는 배지에서 배양하기 전에 일반적인 배지에서 배양하여 배양에 최적화된 상태를 갖추도록 하는 단계를 추가로 포함할 수 있다.
다른 예로서, 상기 (a) 단계에서 사용되는 배지에는 CHIR99021, 포스콜린 및 SB431542를 추가로 포함할 수 있으며, 상기 배지에 5-AZA를 2일 간격으로 첨가할 수 있다.
또 다른 예로서, 상기 (b) 단계에서 사용되는 배지는 성장인자를 포함하는 줄기세포 배양용 배지를 사용할 수 있고, 상기 성장인자는 특별히 이에 제한되지 않으나, bFGF(basic fibroblast growth factor), EGF(epidermal growth factor) 등을 단독으로 또는 혼합하여 사용할 수 있다.
또 다른 예로서, 상기 (a) 및 (b) 단계에서 사용되는 배양용기는 PLO(Poly-L-Ornithin)와 FN(Fibronectin)이 코팅된 배양용기를 사용할 수 있는데, 상기 (b) 단계에서 배양하여 구상체(Sphere)가 형성된 후에는 상기 코팅된 배양용기와 아무것도 코팅되지 않은 배양용기를 번갈아 사용하여 계대배양을 수행할 수 있다.
또 하나의 양태로서, 본 발명은 상기 제조방법에 의해 제조된 유도 신경줄기세포 또는 이로부터 분화된 신경세포 또는 신경교세포를 제공한다.
상기 유도 신경줄기세포에 대하여는 상기에서 설명한 바와 같다.
상기 유도 신경줄기세포가 신경세포, 별아교세포 및 희소돌기아교세포로 분화할 수 있음을 실시예에서 확인하였는 바, 상기 유도 신경줄기세포로 분화된 신경세포 또한 수득할 수 있다.
또 하나의 양태로서, 본 발명은 상기 제조방법에 의해 제조된 유도 신경줄기세포 또는 이로부터 분화된 신경세포 또는 신경교세포를 유효성분으로 포함하는, 신경세포 재생용 세포치료제를 제공한다.
본 발명에 따른 유도 신경줄기세포는 신경관련세포로 분화할 수 있는 세포에 해당하므로, 신경세포 재생에 유용한 바 이를 세포치료제로 사용할 수 있다.
본 발명의 세포치료제는 1㎖ 당 1.0×105개 내지 1.0×109개, 바람직하게는 1.0×106개 내지 1.0×108개, 보다 바람직하게는 1.0×107개의 세포를 포함할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 세포치료제는 동결되지 않은 채 사용되거나 차후 사용을 위해 동결될 수 있다. 동결되어야 할 경우, 표준 냉동보존제 (예를 들어 DMSO, 글리세롤, 에피라이프 (Epilife) 세포 동결 배지 (Cascade Biologics))가 동결 전 세포 집단에 첨가될 수 있다.
또한, 상기 세포치료제는 약학적 분야에서 통상의 방법에 따라 환자의 신체 내 투여에 적합한 단위투여형의 제제로 제형화시켜 투여할 수 있으며, 상기 제제는 1회 또는 수회 투여에 의해 효과적인 투여량을 포함한다. 이러한 목적에 적합한 제형으로는 비경구투여 제제로서 주사용 앰플과 같은 주사제, 주입 백과 같은 주입제, 및 에어로졸 제제와 같은 분무제 등이 바람직하다. 상기 주사용 앰플은 사용 직전에 주사액과 혼합 조제할 수 있으며, 주사액으로는 생리 식염수, 포도당, 만니톨, 링거액 등을 사용할 수 있다. 또한, 주입 백은 염화폴리비닐 또는 폴리에틸렌 재질의 것을 사용할 수 있으며, 박스터 (Baxter), 벡톤 디킨슨 (Becton Dickinson), 메드셉 (Medcep), 내셔날 호스피탈 프로덕츠 (National Hospital Products) 또는 테루모 (Terumo) 사의 주입 백을 예시할 수 있다.
상기 세포치료제에는 하나 또는 그 이상의 약학적으로 허용가능한 통상의 불활성 담체, 예를 들어, 주사제의 경우에는 보존제, 무통화제, 가용화제 또는 안정화제 등을, 국소 투여용 제제의 경우에는 기제 (base), 부형제, 윤활제 또는 보존제 등을 추가로 포함할 수 있다.
이렇게 제조된 본 발명의 세포치료제는 당업계에서 통상적으로 사용하는 투여방법을 이용하여 이식 및 기타 용도에 사용되는 다른 줄기세포와 함께 또는 그러한 줄기세포와의 혼합물의 형태로 투여될 수 있으며, 바람직하게는 치료가 필요한 환자의 질환 부위에 직접 생착 또는 이식하거나 복강에 직접 이식 또는 주입하는 것이 가능하나 이에 한정되지는 않는다. 또한, 상기 투여는 카테터를 이용한 비외과적 투여 및 질환부위 절개 후 주입 또는 이식 등 외과적 투여방법 모두 가능하나 카테터를 이용한 비외과적 투여방법이 보다 바람직하다. 또한 통상의 방법에 따라 비경구적으로, 예를 들면 직접 병변에 투여하는 것 외에 조혈계 줄기세포 이식의 일반적 방법인 혈관 내 주입에 의한 이식도 가능하다.
상기 세포의 1일 투여량은 1.0×104 내지 1.0×1010 세포/kg 체중, 바람직하게는 1.0×105 내지 1.0×109 세포/kg 체중을 1회 또는 수회로 나누어 투여할 수 있다. 그러나, 유효성분의 실제 투여량은 치료하고자 하는 질환, 질환의 중증도, 투여경로, 환자의 체중, 연령 및 성별 등의 여러 관련 인자에 비추어 결정되어야 하는 것으로 이해되어야 하며, 따라서, 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
또 하나의 양태로서, 본 발명은 상기 유도 신경줄기세포 또는 이로부터 분화된 신경세포 또는 신경교세포를 피험체에 투여하는 단계를 포함하는 피험체의 신경세포 손상의 억제 또는 회복방법을 제공한다. 구체적으로 본 발명의 비신경 세포를 5-AZA(5-Azacytidine), CHIR99021, Forskolin 및 SB431542를 포함하는 배지에서 배양하는 단계; 상기 단계에서 배양된 비신경 세포를 5-AZA(5-Azacytidine), CHIR99021, Forskolin 및 SB431542를 포함하지 않는 배지에서 구상체(Sphere)가 형성될 때까지 배양하는 단계; 및 상기 세포를 배양하는 단계에 의해 리프로그래밍된 유도 신경줄기세포 또는 이로부터 분화된 신경세포를 투여함으로써 피험체의 신경세포 손상을 억제하거나 또는 회복시킬 수 있다.
본 발명에서 "피험체"란 소, 개, 돼지, 닭, 양, 말, 인간을 포함한 포유동물을 포함하나 이에 제한되는 것은 아니다. 바람직하게는 리프로그래밍된 유도 신경줄기세포 또는 그 배양물의 투여는 복강 또는 혈관 내 투여, 병변으로의 직접 투여 또는 관절의 활강(Synovial cavity) 내 투여 등일 수 있다.
상기 신경세포 손상의 억제 또는 회복은 신경세포 손상질환의 예방 또는 치료하는 것을 포함할 수 있다.
또 하나의 양태로서, 본 발명은 본 발명의 제조방법에 의해 제조된 유도 신경줄기세포; 또는 상기 유도 신경줄기세포로부터 분화된 신경세포 또는 신경교세포를 유효성분으로 포함하는, 신경세포손상 질환 예방 또는 치료용 약학적 조성물을 제공한다.
상기 설명한 바와 같이, 본 발명의 제조방법에 의해 제조된 유도 신경줄기세포; 또는 상기 유도 신경줄기세포로부터 분화된 신경세포 또는 신경교세포를 유효성분으로 포함하는, 신경세포손상 질환 예방 또는 치료용 약학적 조성물을 신경세포가 손상된 부분에 도입시켜 신경세포손상 질환 예방 또는 치료용도로 사용할 수 있다.
본 발명에서 "신경세포 손상질환"은 신경세포의 변형, 손실 등이 원인이 되어 발생하는 질환으로, 파킨슨씨병, 알츠하이머, 피크병(Pick's disease), 헌팅톤병(Huntington's disease), 근위축성 측면 경화증(amyotriophic lateral sclerosis), 허혈성 뇌질환(stroke), 탈수초질환(demyelinating disease), 다발성 경화증, 간질, 퇴행성 신경질환, 척추 손상(spinal cord injury) 등을 포함할 수 있으나, 상기 예에 제한되지 않는다.
본 발명에서 "예방"은 상기 조성물의 투여로 신경세포 손상질환의 발병을 억제 또는 지연시키는 모든 행위를 포함하며, "치료"는 상기 조성물의 투여로 신경세포 손상 질환의 증세가 호전되거나 이롭게 되는 모든 행위를 포함한다.
또 하나의 양태로서, 본 발명은 본 발명의 유도 신경줄기세포 또는 유도 신경줄기세포로부터 분화된 신경세포 또는 신경교세포를 개체에 이식하는 단계를 포함하는, 신경세포손상 질환 예방 또는 치료하는 방법을 제공한다. 상기 유도 신경줄기세포, 신경세포, 신경세포손상 질환, 예방, 치료는 상기 설명한 바와 같다.
이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: 유도 신경줄기세포의 제조
우선, 10% FBS를 포함하는 DMEM 배양배지가 담겨진 Poly-L-Ornithin(PLO)와 Fibronectin(FN)이 코팅된 세포배양 디쉬(cell culture dish)에 비신경 세포인 인간의 섬유아세포를 1 x 104/㎠의 농도로 접종하고, 하루동안 배양하였다. 이어, 상기 배지를, N2 supplement가 포함된 DMEM/F12 배양배지와 B27 supplement가 포함된 Neurobasal 배양배지가 1:1(v/v)로 혼합된 배지로 교체하고, 상기 교체된 배지에 DMSO에 용해시킨 5-AZA, CHIR99021, 포스콜린 및 SB431542를 첨가하고 배양하면서, 2일 간격으로 상기 첨가물을 첨가하여 총 7일 동안 1차 배양하였다.
상기 1차 배양이 종료된 후, 배양된 세포에 Accutase를 처리하여 배양된 세포를 회수하고, 상기 회수된 세포를 ReNcell NSC maintenance 배양배지와 StemPro NSC SFM 배양배지가 1:1(v/v)로 혼합되고, bFGF(basic fibroblast growth factor)와 EGF(epidermal growth factor)가 첨가된 배지가 담겨진 PLO/FN 코팅 배양용기에 접종하여, 1주일 동안 2차 배양하였다. 상기 2차 배양이 종료된 후, 배양된 세포를 회수하고, 회수된 세포를 상기 2차 배양에 사용된 배지가 담겨진 코팅되지 않은 배양용기에 접종하여 3차 배양하였다. 3차 배양을 수행하면서, 구상체(Sphere)가 형성되면, 안정적인 신경줄기세포 주 확립을 위해 PLO/FN 코팅된 배양용기와 코팅되지 않은 배양용기에 반복적으로 계대배양하였다(도 2)
도 2는 비신경 세포를 5-AZA, CHIR99021, 포스콜린 및 SB431542가 첨가된 배지와 이들이 첨가되지 않은 배지에서 배양하는 과정에서 시간의 경과에 따라 발생하는 세포형태의 변화를 나타내는 현미경 사진이다. 도 2에서 보듯이, 최초 배양시에는 섬유세포의 형태를 나타내었으나, 28일이 경과된 후에는 줄기세포의 특징인 구상체를 형성함을 확인하였다.
상기 계대배양이 종료된 후, 형성된 유도 신경줄기세포에서 신경줄기세포의 특이적 형태(morphology)가 PLO/FN 코팅된 배양용기에서의 부착(attached)된 형태와 PLO/FN 코팅되지 않은 배양용기에서의 부유(suspension)된 형태로 모두 나타나는 것을 확인하였다(도 3).
도 3은 신경줄기세포의 특이적 형태(morphology)를 나타내는 현미경 사진이다. 구체적으로 부착(attached)배양(왼쪽)과 부유(suspension)배양(오른쪽) 시에 신경 줄기세포의 특징적 형태를 나타낸 것이다.
실시예 2: 신경줄기세포 특이적 마커분석
상기 실시예 1에서 제조된 유도 신경줄기세포가 종래에 알려진 신경줄기세포와 동일한 것인지를 확인하기 위하여, 신경줄기세포 마커분석을 수행하였다.
실시예 2-1: 마커 유전자 분석
상기 실시예 1에서 제조된 유도 신경줄기세포(ciNSC)와 기존에 잘 알려진 H9 배아줄기세포로부터 분화된신경줄기세포(H9-NSC)에서 신경줄기세포 특이적 마커 유전자인 SOX2, NESTIN 및 PAX6의 발현수준을 Conventional RT-PCR 기법을 이용하여 비교하였다(도 4a). 이때, 대조군으로는 hDF를 사용하고, 내부대조군으로는 GAPDH를 사용하였다.
도 4a는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포(ciNSC)와 종래의 H9 배아줄기세포로부터 분화된신경줄기세포(H9-NSC)에서 발현된 SOX2, NESTIN 및 PAX6의 발현수준을 비교한 결과를 나타내는 사진이다. 도 4a에서 보듯이, 신경줄기세포의 대표적 특이적 유전자인 SOX2, NESTIN 및 PAX6가 본 발명의 방법으로 제조된 유도 신경줄기세포(ciNSC)에서는 H9-NSC와 유사하게 발현됨을 확인하였다.
또한, 부착 배양하여 제조된 유도 신경줄기세포(ciNSC attach), 부유 배양으로 제조된 유도 신경줄기세포(ciNSpC), 및 기존에 잘 알려진 H9 배아줄기세포로부터 분화된 신경줄기세포(H9-NSC)에서 신경줄기세포 특이적 마커 유전자인 SOX2, PAX6, CXCR4, BRN2, SLC1A3 및 OLIG2와 섬유아세포 특이적 마커 유전자인 COL1A2, S100A4 및 ACTA2의 발현수준을 Quantitative RT-PCR 기법을 이용하여 비교하였다(도 4b). 이때, 대조군으로는 hDF를 사용하였다.
그 결과, Quantitative RT-PCR을 통해서 섬유아세포 특이적인 마커의 발현은 거의 되지 않음을 확인하였는바, 본 발명의 방법으로 제조된 유도 신경줄기세포가 종래에 알려진 신경줄기세포의 특징을 갖음을 확인하였다.
실시예 2-2: 면역형광염색을 이용한 마커 단백질 분석
상기 실시예 1에서 제조된 유도 신경줄기세포를 대상으로, 핵에서 발현되는 마커 단백질인 SOX2, PAX6 및 HMGA2; 세포질에서 발현되는 마커 단백질인 NESTIN; 및 신경줄기세포의 자가재생 능력을 나타내는 마커 단백질인 Ki67의 면역염색을 수행하였다(도 5a).
도 5a는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포에서 발현되는 핵내 마커 단백질(SOX2, PAX6 및 HMGA2), 세포질내 마커 단백질(NESTIN) 및 신경줄기세포의 자가재생 능력을 나타내는 마커 단백질(Ki67)의 발현수준을 확인한 면역형광염색 결과를 나타내는 현미경 사진이다. 또한, 도 5b는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포에서 발현되는 핵내 마커 단백질(SOX2, PAX6) 및 신경줄기세포의 자가재생 능력을 나타내는 마커 단백질(Ki67)의 발현수준을 image-J를 이용하여 %로 계측한 결과이다.
도 5에서 보듯이, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 공지된 신경줄기세포의 핵내 및 세포질내 마커를 발현하였고, 자가재생 능력을 나타냄을 확인하였다.
상기 도 4 및 5의 결과로부터, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 정상적인 신경줄기세포와 동일한 마커를 발현함을 알 수 있었다.
실시예 3: 유도 신경줄기세포의 분화능 분석
상기 실시예 1에서 제조된 유도 신경줄기세포가 다른 신경줄기세포와 마찬가지로 다양한 신경세포로 분화될 수 있는지의 여부를 확인하고자, 상기 유도 신경줄기세포를 공지된 방법으로, 신경세포(Neuron), 별아교세포(Astocyte) 또는 희소돌기아교세포(Oligodendrocyte)로 각각 분화시키고, 신경세포의 마커인 NF(Neurofilament)와 alpha-internexin, 별아교세포의 마커인 GFAP(glial fibrillary acidic protein) 및 희소돌기아교세포의 마커인 O4를 면역형광염색을 통해 확인하였다(도 6a, 6c 및 6d).
도 6a는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포로부터 분화된 신경세포를 대상으로 NF와 alpha-internexin을 면역형광염색한 결과를 나타내는 현미경 사진이다. 도 6b는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포로부터 분화된 신경세포를 대상으로 NF와 alpha-internexin을 DAPI염색 후, 각각의 발현되는 마커를 image-J를 이용하여 %로 계측한 결과이다.
도 6a, 6b에서 보듯이, 상기 분화된 신경세포에서 NF와 alpha-internexin이 발현됨을 확인하였으므로, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 신경세포로 분화될 수 있는 능력을 나타냄을 확인하였다.
도 6c는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포로부터 분화된 별아교세포를 대상으로 GFAP를 면역형광염색한 결과를 나타내는 현미경 사진이다. 도 6c, 6e에서 보듯이, 상기 분화된 별아교세포세포에서 GFAP가 발현됨을 확인하였으므로, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 별아교세포로 분화될 수 있는 능력을 나타냄을 확인하였다.
도 6d는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포로부터 분화된 희소돌기아교세포를 대상으로 O4를 면역형광염색한 결과를 나타내는 현미경 사진이다.
도 6e는 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포로부터 분화된 별아교세포 및 희소돌기아교세포를 대상으로 DAPI염색 후, 각각의 발현되는 마커를 image-J를 이용하여 %로 계측한 결과이다.
도 6d, 6e에서 보듯이, 상기 분화된 희소돌기아교세포에서 O4가 발현됨을 확인하였으므로, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 희소돌기아교세포로 분화될 수 있는 능력을 나타냄을 확인하였다.
상기 도 6a 내지 6e의 결과로부터, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 정상적인 신경줄기세포와 동일한 분화능을 나타냄을 알 수 있었다.
실시예 4: 전환인자의 역할 분석
상기 실시예 1에 개시된 유도 신경줄기세포의 제조방법에서 보듯이, 비신경 세포를 4가지 화합물이 포함된 배지에서 배양하여, 유도 신경줄기세포로 전환시킬 수 있는데, 이때, 사용되는 4가지 화합물 중에서 가장 중요한 역할을 수행하는 화합물이 무엇인지를 확인하고자 하였다.
구체적으로, 5-AZA(5), CHIR99021(C), 포스콜린(F) 및 SB431542(S) 대신에, 5-AZA 단독(5), CHIR99021/포스콜린(CF), 5-AZA/CHIR99021(C5), 포스콜린/SB431542(FS), 5-AZA/포스콜린(F5), 5-AZA/SB431542(S5), CHIR99021/포스콜린/SB431542(CFS), 5-AZA/CHIR99021/포스콜린(CF5), 5-AZA/CHIR99021/SB431542(CS5), 5-AZA/포스콜린/SB431542(FS5), 5-AZA/CHIR99021/포스콜린/SB431542(4Fs)를 사용하는 것을 제외하고는, 실시예 1과 동일한 방법으로 각각의 2차 배양된 세포를 수득하였다. 상기 수득한 각각의 2차 배양된 세포를 대상으로 섬유아세포의 마커인 aSMA의 발현수준과 신경줄기세포의 마커인 SOX2, PAX6, OLIG2 및 NESTIN 의 발현수준을 측정하고 비교하였다(도 7a 및 7b). 이때, 음성 대조군으로는 DMSO만을 처리하여 배양한 세포를 사용하였고, 양성 대조군으로는 4가지 화합물을 모두 사용하여 실시예 1의 방법으로 최종 제조된 유도 신경줄기세포(ciNSC)를 사용하였다.
도 7a는 5-AZA, CHIR99021, 포스콜린 및 SB431542를 다양한 조합으로 처리하여 배양된 세포에서 측정된 섬유아세포의 마커인 aSMA의 발현수준을 비교한 결과를 나타내는 그래프이다. 도 7a에서 보듯이, 3가지 조합 즉 5-AZA 단독(5), 5-AZA/포스콜린(F5) 및 5-AZA/CHIR99021/포스콜린/SB431542(4Fs)를 사용한 경우에는 aSMA가 발현되지 않았으나, 다른 조합을 사용한 경우에는 aSMA가 발현됨을 확인하였다. 상기 aSMA는 섬유아세포의 마커이므로, 상기 aSMA가 발현되었다는 것은 모든 섬유아세포가 유도 신경줄기세포로 전환되지 않고, 일부만이 전환되었다는 것을 의미하는 것으로 해석될 수 있다.
따라서, 5-AZA 단독(5), 5-AZA/포스콜린(F5) 또는 5-AZA/CHIR99021/포스콜린/SB431542(4Fs)를 사용한 경우에는, 2차 배양만을 수행한 수준에서도 최종적으로 제조된 유도 신경줄기세포(ciNSC)와 유사한 수준으로 모든 섬유아세포가 유도 신경줄기세포로 전환됨을 알 수 있었다.
도 7b는 5-AZA(5), CHIR99021(C), 포스콜린(F) 및 SB431542(S)를 다양한 조합으로 처리하여 배양된 세포에서 측정된 유도 신경줄기세포의 마커인 SOX2, PAX6, OLIG2 및 NESTIN 의 발현수준을 비교한 결과를 나타내는 그래프이다. 도 7b에서 보듯이, 모든 조합을 사용한 경우에 SOX2, PAX6, OLIG2 및 NESTIN 이 발현됨을 확인하였다. 상기 SOX2, PAX6, OLIG2 및 NESTIN 은 유도 신경줄기세포의 마커이므로, 상기 SOX2, PAX6, OLIG2 및 NESTIN 가 발현되었다는 것은 섬유아세포가 유도 신경줄기세포로 전환되었다는 것을 의미하는 것으로 해석될 수 있다.
상기 도 7a와 7b의 결과에서 보듯이, 5-AZA(5), CHIR99021(C), 포스콜린(F) 및 SB431542(S)를 다양한 조합으로 사용할 경우, 5-AZA/포스콜린(F5)을 제외한 대부분의 조합이 섬유아세포를 유도 신경줄기세포로 전환시킬 수 있음을 알 수 있었다. 그러나, 대부분의 조합에서는 섬유아세포와 유도 신경줄기세포가 함께 공존하는 형태이고, 5-AZA 단독(5) 및 5-AZA/CHIR99021/포스콜린/SB431542(4Fs)를 사용한 경우에만, 섬유아세포가 모두 유도 신경줄기세포로 전환되어, 섬유아세포가 존재하지 않음을 알 수 있었다.
특히, 5-AZA를 단독으로 사용할 경우(5) 보다는 4가지 화합물을 모두 사용할 경우에 보다 유도 신경줄기세포로의 전환효율이 우수한 것으로 확인되었다.
따라서, 비신경 세포를 유도 신경줄기세포로 전환시킴에 있어서, 가장 핵심적인 요소는 5-AZA이며, 나머지 CHIR99021, 포스콜린 및 SB431542는 상기 5-AZA의 효과를 보조하는 역할을 수행하는 것으로 분석되었다.
실시예 5: in vivo 에서의 생존과 분화 확인
유도 신경줄기세포(ciNSC)는 accutase를 사용하여 분리하고 PBS에 현탁시켰다. 유도 신경줄기세포(ciNSC)는 주입 후 추적을 위해 CM-DiI (Molecular Probes)로 표지하였다. 유도 신경줄기세포(ciNSC)는 37℃의 수조에서 15분 동안 CM-DiI로 배양한 다음, 4℃에서 10분 동안 인큐베이션 하였다. CM-DiI로 표지된 유도 신경줄기세포(ciNSC)를 1x105 세포/μl의 밀도로 PBS에 현탁시켰다. 마우스 (4 주령)를 5% 이소플루란으로 마취시키고 페이셜 마스크(facial mask)를 통해 2% 이소플루란을 투여하여 유지시켰다. 유도 신경줄기세포(ciNSC)는 정위 방법(stereotaxic) 장치와 울트라 마이크로펌프(micropump) (World Precision Instruments, Sarasota, FL, USA)를 사용하여 해마에 이식시켰다. 주입 좌표는 후방 1.4mm, 브레그마(bregma)에서 2 mm, 깊이 1.5 mm이다. 각 마우스는 분당 250nl의 속도로 2μl PBS에서 약 2x105 세포를 수용하였다. 이식 후, 두피를 봉합하고 동물을 마취로부터 회복시켰다.
이식 된 유도 신경줄기세포(ciNSC)를 추적하기 위해 마우스를 0.1M PBS (pH 7.4)로 관류시킨 다음 0.1M PBS에서 4 % PFA로 관류시켰다. 그 후, 뇌를 조심스럽게 단리하고 4% PFA에 밤새 담가 고정시킨 후, 30% 수크로스로 옮겨 48시간 동안 담갔다. 뇌는 침투 혼합물 (OCT compound; Sakura Finetek, Tokyo, Japan)로 몰딩하고, -70℃에서 밤새 보관 한 다음 저온 유지 장치 (CM 3050, Leica, Wetzlar, Germany)를 사용하여 저온 절개를 하였다. 이는 ICC와 유사한 절차로 수행하였다. 구체적으로, 조직을 0.05% Triton X-100으로 20분 동안 인큐베이션시킨 후, 비특이적인 항체의 결합을 차단하기 위해 5% NGS로 배양하였다. 그 후, 조직을 4℃에서 하룻밤 동안 5% NGS에서 권장 희석 배율에 따라 1차 항체와 함께 배양하였다. 조직을 실온에서 1 시간 동안 2차 (Alexa 488- 또는 Alexa 546-) 표지 항체 (1:1000)와 함께 배양하였다. 핵은 DAPI로 10분 동안 염색하였고, 이미지는 공초점 현미경 (Nikon)을 사용하여 캡처하였다.
도 8은 유도 신경줄기세포를 마우스의 뇌에 주입하고 4주 후에 in vivo 생존과 분화에 대한 분석을 형광면역염색을 통해서 분석한 결과를 나타내는 도이다. 도 8a 내지 8c에서 보듯이, 주입된 유도 신경줄기세포(CMDi; red)는 4주 후에도 주입된 부분에서 생존하고 있음을 확인하였다. 또한, 3가지 대표적 신경세포인 뉴런 (도 8a: Tuj1; green), 별아교세포 (도 8b: astrocyte (GFAP; green)), 희소돌기아교세포 (도 8c: oligodendrocyte (MBP; green))로 분화한 것을 확인할 수 있었다.
이를 통해, 본 발명에서 제공하는 방법에 의하여 제조된 유도 신경줄기세포는 in vivo에서도 정상적인 신경줄기세포와 동일한 분화능을 나타냄을 알 수 있었다.
이상의 설명으로부터, 본원이 속하는 기술분야의 당업자는 본원이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로 이해해야만 한다. 본원의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본원의 범위에 포함되는 것으로 해석되어야 한다.

Claims (19)

  1. 5-AZA(5-Azacytidine)를 포함하는 비신경 세포의 유도 신경줄기세포로의 전환유도용 조성물.
  2. 제1항에 있어서,
    상기 비신경 세포는 동물 유래 체세포인 것인, 조성물.
  3. 제1항에 있어서,
    상기 조성물은 CHIR99021, Forskolin 및 SB431542를 추가로 포함하는 것인 조성물.
  4. 제1항에 있어서,
    상기 유도 신경줄기세포는 SOX2, NESTIN 및 PAX6 유전자를 발현시키고, HMGA2, NESTIN 및 Ki67 단백질을 발현시키며, 신경세포, 별아교세포세포 또는 희소돌기아교세포로 분화될 수 있는 것인, 조성물.
  5. 제1항 내지 제4항 중 어느 한 항의 조성물을 포함하는, 비신경 세포의 유도 신경줄기세포로의 전환유도용 키트.
  6. 제5항에 있어서,
    상기 키트는 (a) 5-AZA를 포함하는 조성물; 및 (b) 비신경 세포 배양용 배지 조성물이 각각 개별 용기에 담긴 형태, 또는 하나 이상의 구획으로 나누어진 한 개의 용기 내에 담긴 형태로 포장된 형태인 것인, 키트.
  7. 제5항에 있어서,
    상기 키트는 (a) 상기 5-AZA를 포함하는 제1 조성물; (b) CHIR99021, 포스콜린 및 SB431542를 포함하는 제2 조성물; 및 (c) 비신경 세포 배양용 배지 조성물이 각각 개별 용기에 담긴 형태, 또는 하나 이상의 구획으로 나누어진 한 개의 용기 내에 담긴 형태로 포장된 형태인 것인, 키트.
  8. (a) 비신경 세포를 5-AZA를 포함하는 배지에서 7 내지 14일 동안 배양하는 단계; 및
    (b) 상기 (a) 단계에서 배양된 비신경 세포를 5-AZA를 포함하지 않는 배지에서 구상체(Sphere)가 형성될 때까지 배양하는 단계를 포함하는, 비신경 세포로부터 전환된 유도 신경줄기세포를 제조하는 방법.
  9. 제8항에 있어서,
    상기 (a) 단계에서 사용되는 배지에 CHIR99021, 포스콜린 및 SB431542를 추가로 포함하는 것인, 방법.
  10. 제8항에 있어서,
    상기 (b) 단계에서 사용되는 배지는 성장인자를 포함하는 줄기세포 배양용 배지인 것인, 방법.
  11. 제10항에 있어서,
    상기 성장인자는 bFGF(basic fibroblast growth factor), EGF(epidermal growth factor) 또는 이들의 조합인 것인, 방법.
  12. 제8항에 있어서,
    상기 (a) 및 (b) 단계는 PLO(Poly-L-Ornithin)와 FN(Fibronectin)이 코팅된 배양용기에서 수행되는 것인, 방법.
  13. 제8항에 있어서,
    상기 (b) 단계에서 배양하여 구상체(Sphere)가 형성된 후에는, 상기 코팅된 배양용기와 아무것도 코팅되지 않은 배양용기를 번갈아 사용하여 계대배양을 수행하는 단계를 추가로 포함하는 것인, 방법.
  14. 제8항 내지 제13항 중 어느 한 항의 방법으로 제조된, 유도 신경줄기세포.
  15. 제14항에 있어서,
    상기 유도 신경줄기세포는 SOX2, NESTIN 및 PAX6 유전자를 발현시키고, HMGA2, NESTIN 및 Ki67 단백질을 발현시키며, 신경세포, 별아교세포 또는 희소돌기아교세포로 분화될 수 있는 것인, 세포.
  16. 제14항의 유도 신경줄기세포; 또는 상기 유도 신경줄기세포로부터 분화된 신경세포 또는 신경교세포를 유효성분으로 포함하는, 신경세포 재생용 세포 치료제.
  17. 제14항의 유도 신경줄기세포; 또는 상기 유도 신경줄기세포로부터 분화된 신경세포 또는 신경교세포를 유효성분으로 포함하는, 신경세포손상 질환 예방 또는 치료용 약학적 조성물.
  18. 제17항에 있어서, 상기 신경세포손상 질환은 파킨슨씨병, 알츠하이머, 피크병(Pick's disease), 헌팅톤병(Huntington's disease), 근위축성 측면 경화증(amyotriophic lateral sclerosis), 허혈성 뇌질환(stroke), 탈수초질환(demyelinating disease), 다발성 경화증, 간질, 퇴행성 신경질환 및 척추 손상(spinal cord injury)으로 이루어진 군에서 선택되는 신경세포손상 질환 예방 또는 치료용 약학적 조성물.
  19. 제14항의 유도 신경줄기세포; 또는 상기 유도 신경줄기세포로부터 분화된 신경세포 또는 신경교세포를 개체에 이식하는 단계를 포함하는, 신경세포손상 질환 예방 또는 치료하는 방법.
