本発明者らは、予備試験により、1.5%オラネジン(オラネキシジングルコン酸塩として1.508(W/V)%を含む液、株式会社大塚製薬工場製)に、サンセットイエローFCF(黄色5号)を添加すると、沈殿を生じることを確認した。そこで、サンセットイエローFCF(黄色5号)の添加により生じる沈殿を溶解させるために、界面活性剤として終濃度が2(W/V)%となるようにラウロマクロゴールを添加した着色製剤(オラネジン消毒液着色製剤)を調製し、殺菌効力を試験した。
使用量に応じて作製量を変更した。その場合、試薬、培地及び溶媒量は同一比率で増減した。
10.0gのポリソルベート80を量り採り、これに約80mLの蒸留水、大豆レシチン1.17gを加え、加熱、撹拌した。リン酸水素二ナトリウム1.01g、リン酸二水素カリウム0.04g、チオ硫酸ナトリウム水和物0.5g、Tamol(登録商標)NN 8906 1.0gを加え、溶解するまで加熱、撹拌した。溶解後、室温まで冷却し、水酸化ナトリウム水溶液若しくは塩酸でpH7.8~7.9に調整した。全量が100mLとなるよう蒸留水を加えた後、高圧蒸気滅菌(121℃、20分)を行った。
サブロー・ブドウ糖LP寒天培地(SABLP培地、組成:ブドウ糖40.0g/L、ペプトン10.0g/L、カンテン15.0g/L、レシチン1.0g/L、ポリソルベート80 7.0g/L)73.0gをガラス製三角フラスコに量り採り、純水1000mLを加え、スターラーで撹拌した。これを高圧蒸気滅菌(121℃、20分)した。使用するまで47℃に設定した温浴中に保存した。
1.5(W/V)%のオラネキシジングルコン酸塩に対して、微量のサンセットイエローFCF(黄色5号)及び終濃度が2(W/V)%となるようラウロマクロゴールを添加して、試験物質を調製した。
SABLP培地上に増殖した8種の試験菌[スタフィロコッカス・アウレウス(Staphylococcus aureus)ATCC29213、エンテロコッカス・フェカリス(Enterococcus faecalis)ATCC15606、スタフィロコッカス・エピデルミディス(Staphylococcus epidermidis)ATCC12228、セラチア・マルセッセンス(Serratia marcescens)ATCC14756、アシネトバクター・バウマニ(Acinetobacter baumannii)ATCC19606、エシェリキア・コリ(Escherichia coli)ATCC25922、シュードモナス・イルージノウザ(Pseudomonas aeruginosa)ATCC27853、カンジダ・アルビカンス(Candida albicans)ATCC90028]のコロニーを白金耳で採取し、蒸留水でMcFarland5の菌液を調製した。これを試験菌液とした。
蒸留水3mLに試験菌液150μLを添加し、よく混合した。直ちに、この菌混合液0.5mLを中和剤4.5mLに加え、よく混合した。これを101倍希釈液とした。101倍希釈液0.5mLを中和剤4.5mLに加えて、更に10倍希釈した。同様の操作により希釈を繰り返して10倍希釈系列(101~105倍希釈液までの計5段階)を作製した。103~105倍希釈液を1mLずつシャーレに分注し、温浴中に保存していたSABLP培地を約15mL加えて混釈平板を作製した。混釈平板が固化した後、倒置し、35±2℃で2日間好気培養した。混釈平板で増殖したコロニーを目視で計数した。この操作を3回行った。
それぞれの試験物質3mLに試験菌液150μLを添加し、よく混合した。これを反応液とした。反応は室温(23.0±3.0℃)で行った。30秒及び60秒後に反応液0.5mLを中和剤4.5mLに加え、よく混合した。これを101倍希釈液とした。101倍希釈液0.5mLを中和剤4.5mLに加えて10倍希釈した。これを102倍希釈液とした。101及び102倍希釈液を1mLずつシャーレに分注し、温浴中に保存していたSABLP培地を約15mL加えて混釈平板を作製した。混釈平板が固化した後、倒置し、35±2℃で2日間好気培養した。混釈平板で増殖したコロニーを目視で計数した。この操作を3回行った。
初期生菌数(CFU/mL)の常用対数値とその平均値を求めた。この平均値と各作用時間での試験物質作用後の生菌数(CFU/mL)の対数値の平均値からLog Reductionを次式で求めた。

結果を図1及び図2に示す。図1において、殺菌効力があった組成物(「1-2-3 試験物質作用後の生菌数の測定」において混釈平板で増殖したコロニー数が、オラネジン消毒液無色製剤と同数であった組成物)を○で表した。また、図2に、オラネジン消毒液着色製剤の殺菌効力をLog Reductionを示す。カンジダ・アルビカンス(Candida albicans)ATCC90028以外の菌種に対しては、従来のオラネジン消毒液無色製剤と、オラネジン消毒液着色製剤とは同等の殺菌効力を示したが、カンジダ・アルビカンス(Candida albicans)ATCC90028に対しては、オラネジン消毒液着色製剤は30秒で殺菌効力が低下した。
実施例1により、界面活性剤としてラウロマクロゴールを用いたオラネジン消毒液着色製剤は、カンジダ・アルビカンス(Candida albicans)ATCC90028に対する殺菌効力が低下することがわかった。そこで、本試験では、種々の界面活性剤を用いてオラネジン消毒液着色製剤を調製し、カンジダ・アルビカンス(Candida albicans)ATCC90028に対する殺菌効力を試験した。
試験物質として、1.5(W/V)%のオラネキシジングルコン酸塩に対して、微量のサンセットイエローFCF(黄色5号)及び図3、4に記載の界面活性剤を添加した水溶液としたものを用いたこと、並びに、試験菌としてカンジダ・アルビカンス(Candida albicans)ATCC90028を用いたことを除いて、実施例1と同様に行った。
結果を図3及び図4に示す。
実施例2により、界面活性剤としてアルキルジメチルアミンオキシドであるラウリルジメチルアミンオキシドを用いたときに、現行のオラネジン消毒液無色製剤と同等の殺菌効力を有する着色製剤を調製できることがわかった。そこで、本試験では、ラウリルジメチルアミンオキシド以外のアルキルジメチルアミンオキシドを用いて調製したオラネジン消毒液着色製剤の殺菌効力を試験した。
試験物質として、1.5(W/V)%のオラネキシジングルコン酸塩に対して、微量のサンセットイエローFCF(黄色5号)及び図5、6に記載の界面活性剤を添加した水溶液としたものを用いたこと、並びに、試験菌としてカンジダ・アルビカンス(Candida albicans)ATCC90028を用いたことを除いて、実施例1と同様に行った。
結果を図5及び図6に示す。
実施例2により、界面活性剤としてラウリルジメチルアミンオキシドを用いたときに、現行のオラネジン消毒液無色製剤と同等の殺菌効力を有する着色製剤を調製できることがわかった。そこで、本試験では、ラウリルジメチルアミンオキシドと、他の界面活性剤とを併用して調製したオラネジン消毒液着色製剤の殺菌効力を試験した。
試験物質として、1.5(W/V)%のオラネキシジングルコン酸塩に対して、微量のサンセットイエローFCF(黄色5号)及び図7~図12に記載の界面活性剤を添加した水溶液としたものを用いたこと、並びに、試験菌としてカンジダ・アルビカンス(Candida albicans)ATCC90028を用いたことを除いて、実施例1と同様に行った。なお、図中、LDAOは、カチオン性界面活性剤であるラウリルジメチルアミンオキシドを表し、POE(20)POP(4)セチルエーテルは、ポリオキシエチレン(20)ポリオキシプロピレン(4)セチルエーテルを表す。
結果を図7~図12に示す。図7、9、11中、殺菌効力があった組成物(「1-2-3 試験物質作用後の生菌数の測定」において混釈平板で増殖したコロニー数が、オラネジン消毒液無色製剤と同数であった組成物)を○で表した。
結果を図7及び図8の試験番号1~3に示す。LDAOの添加量に依らず、組成物は30秒及び60秒で殺菌効力が低下しなかった。また、試験番号1において、1.5(W/V)%のオラネキシジングルコン酸塩に着色料を添加したときの沈殿が、2.0(W/V)%のラウリルジメチルアミンオキシドによって溶解したことから、0.01(W/V)%のオラネキシジングルコン酸塩に着色料を添加したときの沈殿は、0.013(W/V)%のラウリルジメチルアミンオキシドによって溶解することが示唆された。
結果を図7及び図8の試験番号4~18に示す。ラウロマクロゴールを単独で添加した組成物は、30秒で殺菌効力が低下した(図7、試験番号18)。一方で、ラウロマクロゴール及びLDAOを添加した組成物では、ラウロマクロゴール及びLDAOの合計濃度に対するLDAO濃度の比率が0.18以上である組成物は30秒及び60秒で殺菌効力が低下しなかった。
結果を図9及び図10に示す。POE(20)POP(4)セチルエーテルを単独で添加した組成物は、30秒及び60秒で殺菌効力が低下した。一方で、POE(20)POP(4)セチルエーテル及びLDAOを添加した組成物では、POE(20)POP(4)セチルエーテル及びLDAOの合計濃度に対するLDAO濃度の比率が0.12以上である組成物は30秒及び60秒で殺菌効力が低下しなかった。
結果を図11及び図12に示す。LDAO、ラウロマクロゴール及びPOE(20)POP(4)セチルエーテルの合計濃度に対するLDAO濃度の比率が0.06以上である組成物は60秒で殺菌効力が低下せず、0.28以上である組成物は30秒及び60秒で殺菌効力が低下しなかった。
Claims (15)
- オラネキシジングルコン酸塩と、着色料と、アルキルジメチルアミンオキシドとを含むことを特徴とする、皮膚消毒用組成物。
- オラネキシジングルコン酸塩の濃度が、0.01~20(W/V)%であることを特徴とする、請求項1に記載の皮膚消毒用組成物。
- 着色料が、法定タール色素であることを特徴とする、請求項1又は2に記載の皮膚消毒用組成物。
- 法定タール色素が、サンセットイエローFCFであることを特徴とする、請求項3に記載の皮膚消毒用組成物。
- アルキルジメチルアミンオキシドの濃度が、0.01(W/V)%以上であることを特徴とする、請求項1~4のいずれかに記載の皮膚消毒用組成物。
- ポリオキシエチレンアルキルエーテルをさらに含むことを特徴とする、請求項1~5のいずれかに記載の皮膚消毒用組成物。
- アルキルジメチルアミンオキシド及びポリオキシエチレンアルキルエーテルの合計濃度に対するアルキルジメチルアミンオキシド濃度の比率が、0.18以上であることを特徴とする、請求項6に記載の皮膚消毒用組成物。
- ポリオキシエチレンポリオキシプロピレンアルキルエーテルをさらに含むことを特徴とする、請求項1~5のいずれかに記載の皮膚消毒用組成物。
- アルキルジメチルアミンオキシド及びポリオキシエチレンポリオキシプロピレンアルキルエーテルの合計濃度に対するアルキルジメチルアミンオキシド濃度の比率が、0.12以上であることを特徴とする、請求項8に記載の皮膚消毒用組成物。
- ポリオキシエチレンアルキルエーテル及びポリオキシエチレンポリオキシプロピレンアルキルエーテルをさらに含むことを特徴とする、請求項1~5のいずれかに記載の皮膚消毒用組成物。
- アルキルジメチルアミンオキシド、ポリオキシエチレンアルキルエーテル及びポリオキシエチレンポリオキシプロピレンアルキルエーテルの合計濃度に対するアルキルジメチルアミンオキシド濃度の比率が、0.28以上であることを特徴とする、請求項10に記載の皮膚消毒用組成物。
- アルキルジメチルアミンオキシドが、炭素数10~16のアルキル基を有するアルキルジメチルアミンオキシドであることを特徴とする、請求項1~11のいずれかに記載の皮膚消毒用組成物。
- アルキルジメチルアミンオキシドが、ラウリルジメチルアミンオキシド、デシルジメチルアミンオキシド、ミリスチルジメチルアミンオキシド及びヤシ油アルキルジメチルアミンオキシドから選択されることを特徴とする、請求項12に記載の皮膚消毒用組成物。
- ポリオキシエチレンアルキルエーテルが、ラウロマクロゴールであることを特徴とする、請求項6~7及び10~13のいずれかに記載の皮膚消毒用組成物。
- ポリオキシエチレンポリオキシプロピレンアルキルエーテルが、ポリオキシエチレン(20)ポリオキシプロピレン(4)セチルエーテルであることを特徴とする、請求項8~14のいずれかに記載の皮膚消毒用組成物。
