Abstract

본 발명은 고마리(Polygonum thunbergii) 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물에 관한 것으로, 본 발명의 조성물은 피부에 자극을 일으키지 않으면서 엘라스타아제(elastase) 및 콜라게나아제(collagenase) 저해활성이 우수하여 피부 주름 개선을 위한 기능성 화장품의 소재로 사용할 수 있다.

Description

고마리 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물
본 발명은 고마리(Polygonum thunbergii) 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물에 관한 것으로, 더욱 상세하게는 엘라스타아제(elastase) 및 콜라게나아제(collagenase) 저해 활성을 가지는 고마리 메탄올 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물에 관한 것이다.
피부는 외부 자극으로부터 우리 몸을 보호해주는 인체 최전방의 방어 장기이며, 미적인 관점에서도 그 관심이 점점 높아지고 있다. 이에 따라 다양한 기능성 화장품에 대한 연구가 활발한데, 그 중에서도 주름완화 및 미백 기능에 대한 다양한 기능성 화장품들이 출시되고 있는 실정이다.
주름의 생성 원인으로는 여러 가지가 보고되고 있는데, 우선 피부가 과다한 자외선에 노출되면 피부 구성성분인 콜라겐(collagen) 분해가 촉진되어 피부가 탄력을 잃고 주름이 생성될 수 있다. 또한, 피부가 과도한 온도변화 및 습도저하나 바람 등에 의해 지나치게 건조해지면 외부에 대한 방어벽으로서의 피부 기능이 저하되어 주름이 생기기도 한다. 그리고, 피부가 활성 산소종이나 자유 라디칼에 노출되면 산화 작용에 의해 과산화 지질이 생성되고 그로 인해 콜라겐 등의 피부 구성 단백질이 변형되어 주름이 생성될 수 있다. 이러한 외부적 요인과 내부적 요인에 의한 주름 발생을 억제하고자 다양한 화장품 조성물이 연구되고 있는 실정이다.
특히 화학성분이 함유된 기능성 화장품의 경우 피부 자극을 유발하여 민감성 피부에 적합하지 않은 문제점이 있는바, 피부에 자극이 적고 친환경적인 천연 식물 유래의 기능성 화장품에 대한 관심이 집중되고 있다.
고마리(Polygonum thunbergii)는 고만이, 꼬마리라고도 하며, 양지바른 들이나 냇가에서 자란다. 높이는 약 1 m이고, 줄기의 능선을 따라 가시가 나며 털이 없다. 잎은 어긋나고 잎자루가 있으나, 윗부분 잎에는 잎자루가 없고 잎 모양은 서양 방패처럼 생겼으며 길이 4~7 cm, 너비 3~7 cm이다. 꽃의 형태와 피는 시기, 잎의 생김새 등에 변이가 많으며 메밀과 비슷하다. 고마리는 한방에서는 줄기와 잎을 지혈제로 쓰고, 고마리꽃은 자연정화기능이 뛰어나 눈을 밝게 하고 시력을 증진시키며 이질에도 효험이 있으며, 류머티즘에 좋다고 알려져 있다.
고마리 추출물을 유효성분으로 함유하는 화장료 조성물에 관한 종래 기술로는 한국공개특허 제2015-0000719호에 고마리 추출물을 유효성분으로 미백 화장료 조성물이 개시되어 있으나, 본 발명과 같이 고마리 추출물을 유효성분으로 함유하는 주름 개선용 화장료 조성물에 관한 기술은 전무한 실정이다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명자들은 고마리(Polygonum thunbergii)의 메탄올 추출물을 제조하여 그 기능을 시험해 본 결과, 고마리 메탄올 추출물이 엘라스타아제 및 콜라게나아제 활성을 억제하는 것을 확인함으로써, 본 발명을 완성하였다.
상기 과제를 해결하기 위해, 본 발명은 고마리(Polygonum thunbergii) 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물을 제공한다.
본 발명의 고마리 추출물은 천연 유래 추출물이기 때문에 피부에 부작용을 일으키지 않는 안정한 특징이 있고, 엘라스타아제 및 콜라게나아제 활성을 저해함으로써 피부 주름생성 및 노화를 방지할 수 있으므로, 이를 유효성분으로 포함하는 본 발명의 조성물은 피부에 자극을 일으키지 않는 피부 주름 개선을 위한 기능성 화장품의 소재로 사용될 수 있을 것이다.
본 발명의 목적을 달성하기 위하여, 본 발명은 고마리(Polygonum thunbergii) 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물을 제공한다.
본 발명의 피부 주름 개선용 화장료 조성물에서, 상기 고마리 추출물은 고마리 잎, 줄기, 꽃 및 뿌리로 이루어진 군으로부터 선택되는 하나 이상의 추출물일 수 있으며, 바람직하게는 전초(식물의 전체) 추출물일 수 있으나, 이에 제한되지 않는다.
또한, 본 발명의 피부 주름 개선용 화장료 조성물에서, 상기 고마리 추출물은 건조된 고마리의 물, 메탄올, 에탄올, 프로판올, 부탄올 또는 이의 혼합 용매 추출물일 수 있고, 바람직하게는 건조된 고마리의 메탄올 추출물일 수 있고, 구체적으로는 작두를 사용하여 2~3 cm로 자른 고마리를 30~50℃의 온도에서 2~4일간 건조시킨 후, 400~600 g의 건중량에 75~85% 메탄올을 넣고 초음파 기기를 이용하여 추출하고 여과하여 제조된 것일 수 있다.
또한, 본 발명의 피부 주름 개선용 화장료 조성물에서, 상기 고마리 추출물은 조성물 중량에 대하여 0.001~97% 함유할 수 있으며, 바람직하게는 0.01~20% 함유할 수 있으나, 이에 제한되지 않는다.
본 발명의 고마리 추출물은 주름 개선 효과를 갖는 것을 특징으로 하는데, 상기 주름 개선은 엘라스타아제 및 콜라게나아제 활성을 억제함으로써 피부 탄력을 유지하는 단백질인 엘라스틴 및 콜라겐의 분해를 방지하여 피부 탄력이 저하되지 않도록 하여 주름이 생기지 않도록 하는 것을 특징으로 한다.
본 발명의 화장료 조성물은 스킨, 스킨 소프트너, 스킨토너, 아스트린젠트, 로션, 밀크로션, 모이스쳐 로션, 영양로션, 마사지크림, 영양크림, 아이 크림, 모이스쳐 크림, 핸드크림, 에센스, 영양에센스, 팩, 클렌징 폼, 클렌징 워터, 클렌징 로션, 클렌징 크림, 바디로션, 바디클렌져, 비누 및 파우더로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가질 수 있으나, 이에 제한되지 않는다.
또한, 상기 화장료는 본 발명의 고마리 추출물에 추가로 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온봉쇄제 및 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제, 지질 소낭 또는 화장품에 통상적으로 사용되는 임의의 다른 성분과 같은 화장품학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다.
이하, 본 발명을 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
제조예 1: 고마리 추출물 제조
고마리(Polygonum thunbergii)의 추출물은 고마리 전초를 이용하였다. 고마리의 전초를 작두를 이용하여 2.54 cm 정도로 고르게 자른 후, 건조 오븐에서 40℃로 3일간 건조시켰다. 약 500 g의 식물 건중량을 이용하여, 80% 메탄올에서 초음파 기기를 이용하여 추출하였으며, 이들 액체를 멤브레인 필터(0.2 ㎛)에 통과시켜 여과한 후, 진공상태에서 용매를 제거하여 메탄올 추출물을 남겼다. 이러한 과정을 통하여, 고마리의 구성성분 중 섬유소 등은 정제 과정을 통해 걸러지고, 고마리 추출물은 고마리의 미량성분을 주로 포함하는 메탄올 추출물이 된다. 사용한 모든 용매들은 분석용 등급으로써, Sigma-Aldrich(미국)나 Merck(미국)에서 구입하였다.
실험예 1: 주름개선 효과 검증
(1) 엘라스타아제(Elastase) 저해 활성 측정
돼지의 췌장 엘라스타아제(Porcine pancreas elastase) 저해 활성 측정은 기질로서 N-숙시닐-(L-Ala)3-p-나이트로아닐라이드를 사용하여 37℃에서 20분간 p-나이트로아닐라이드의 생성량을 405 nm에서 측정하였다. 즉, 각 시험용액을 일정 농도가 되도록 조제하여 0.5 ㎖씩 시험관에 취하고 50 mM Tris-HCl 완충액(pH 8.6)에 녹인 엘라스타아제(2.5 U/㎖) 용액 0.5 ㎖를 가한 후 기질로 50 mM Tris-HCl 완충액(pH 8.6)에 녹인 N-숙시닐-(L-Ala)3-p-나이트로아닐라이드(0.5 mg/㎖)를 첨가하여 20분간 반응시켜 측정하였다.
(2) 콜라게나아제(Collagenase) 저해활성 측정
반응구는 0.1M Tris-HCl 완충액(pH 7.5)에 4mM 염화칼슘(CaCl2)을 첨가하여, 0.3 mg/㎖의 4-페닐아조 벤질옥시 카보닐-Pro-Leu-Gly-Pro-D-Arg(Pz-peptide)를 녹인 기질액 0.25 ㎖ 및 고마리 추출물 0.1 ㎖의 혼합액에 0.2 mg/㎖의 콜라게나아제 0.15 ㎖를 첨가하여 실온에서 20분간 방치한 후 6% 시트르산(citric acid) 0.5 ㎖를 넣어 반응을 정지시킨 후, 에틸 아세테이트 1.5 ㎖을 첨가하여 320 nm에서 흡광도를 측정하였다.
실시예 1: 고마리 추출물의 엘라스타아제 저해 활성
엘라스타아제는 진피 내 피부 탄력을 유지하는 기질 단백질인 엘라스틴의 분해에 관여하며, 엘라스틴을 분해할 수 있는 비특이적 가수분해효소이다. 또한, 체내의 엘라스틴을 분해하는 백혈구 과립 효소 중의 하나로 이상조직에서는 활성이 높아져 조직파괴의 직접적인 원인이 되며, 피부의 주름 및 탄력성 소실 등을 유발한다. 따라서 엘라스타아제 저해제는 피부의 주름을 개선하는 효과가 있다.
고마리 메탄올 추출물의 엘라스타아제 저해 활성 검증을 위해, 50, 100 및 500 ㎍/㎖ 농도의 엘라스타아제 저해 활성을 측정한 결과, 고마리 추출물의 농도가 증가할수록 엘라스타아제 저해 활성이 증가함을 확인하였고, 500 ㎍/㎖ 농도에서는 양성대조구인 우르솔산과 큰 차이를 나타내지 않았다(표 1).

실시예 2: 고마리 추출물의 콜라게나아제 저해 활성
피부 주름은 여러 요소들에 의해 유발되는 것으로 알려져 있으나, 피부 진피는 콜라겐으로 주로 이루어져 콜라겐이 피부 주름 형성에 가장 큰 영향을 미치는 것으로 알려져 있다. 콜라게나아제는 피부 탄력을 유지하는 단백질인 콜라겐을 분해하는 효소로, 피부의 주름을 형성시키고 피부 탄력의 저하를 유발한다. 따라서, 이러한 콜라게나아제의 활성을 저해시킬 경우 피부 주름의 생성을 지연시킬 수 있다.
고마리 메탄올 추출물의 콜라게나아제 저해 활성 검증을 위해, 50, 100 및 500 ㎍/㎖ 농도의 콜라게나아제 저해 활성을 측정한 결과, 고마리 추출물의 농도가 증가할수록 콜라게나아제 저해 활성이 증가함을 확인하였고, 500 ㎍/㎖ 농도에서는 90% 이상의 저해 활성을 나타내었다(표 2).

실시예 1 및 2의 결과들을 통해, 고마리 추출물이 피부 주름 개선을 위한 기능성 화장품의 새로운 소재로서의 가능성이 있음을 알 수 있었다.

Claims (6)

  1. 고마리(Polygonum thunbergii) 추출물을 유효성분으로 함유하는 피부 주름 개선용 화장료 조성물.
  2. 제1항에 있어서, 상기 고마리 추출물은 고마리 잎, 줄기, 꽃 및 뿌리로 이루어진 군으로부터 선택되는 하나 이상의 추출물인 것을 특징으로 하는 피부 주름 개선용 화장료 조성물.
  3. 제1항에 있어서, 상기 고마리 추출물은 건조된 고마리의 물, 메탄올, 에탄올, 프로판올, 부탄올 또는 이의 혼합 용매 추출물인 것을 특징으로 하는 피부 주름 개선용 화장료 조성물.
  4. 제1항에 있어서, 상기 고마리 추출물은 엘라스타아제 및 콜라게나아제 활성을 억제하는 것을 특징으로 하는 피부 주름 개선용 화장료 조성물.
  5. 제1항에 있어서, 상기 화장료 조성물은 스킨, 스킨 소프트너, 스킨토너, 아스트린젠트, 로션, 밀크로션, 모이스쳐로션, 영양로션, 마사지크림, 영양크림, 아이 크림, 모이스쳐 크림, 핸드크림, 에센스, 영양에센스, 팩, 클렌징폼, 클렌징 워터, 클렌징 로션, 클렌징 크림, 바디로션, 바디클렌져, 비누 및 파우더로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가지는 것을 특징으로 하는 피부 주름 개선용 화장료 조성물.
  6. 제1항에 있어서, 상기 화장료 조성물은 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온 봉쇄제 및 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제, 지질 소낭 중에서 선택된 하나 이상의 보조제를 추가로 포함하는 것을 특징으로 하는 피부 주름 개선용 화장료 조성물.
