本発明の一実施の形態に係る不妊の改善剤組成物(以下、「不妊の改善剤組成物」又は単に「組成物」と略称する場合がある。)は、ヒシ科植物の果皮及び果実の一方又は双方に含まれる1又は複数の化合物を有効成分として含んでいる。
実施例1:ヒシ果皮抽出物及びそれを含む経口投与剤の調製
(1)ヒシ科植物の熱水抽出物(以下、「ヒシ果皮抽出物」と略称する。)の調製
収穫後のトウビシ(Trapa bispinosa)の果実を乾燥させ、乾燥果皮を回収した。フードプロセッサーを使用して乾燥果皮を粉末化し、熱水抽出(乾燥果皮の1重量部に対し6重量部の90℃の熱水を使用)し、最終的に得られるヒシ果皮抽出物のポリフェノール含量が25重量%以上となるよう予め定めた所定の濃縮率で抽出液を濃縮した。濃縮液67重量%に対し33重量%のデキストリンを添加し、スプレードライヤーで噴霧乾燥した。このようにして得られた粉末(ポリフェノール含量25重量%以上)を、ヒシ果皮抽出物として、以下の試験に使用した。
上記(1)で調製したヒシ果皮抽出物100mgに、賦型剤としてコーンスターチ187mg及びステアリン酸カルシウム3mgを加え、これをゼラチンハードカプセルに封入した。後述する実施例2~4において、被験者には、上記ハードカプセルを一日一カプセル、食前に水又はぬるま湯で服用していただいた。また、ヒシ果皮抽出物の代わりにデキストリンを用いて調製したものをプラセボ(ヒシ果皮抽出物を含む経口投与剤と外観上区別できない。)として用いた。

ジカルボニル結合の切断活性は、ジカルボニル化合物である1-フェニル-1,2-プロパンジオン(PPD)を分解し得られる安息香酸濃度をHPLC法にて測定することで算出した。
98%PPD溶液4.2μLを50mMリン酸緩衝液(pH7.4)/50%メタノール(メタノール緩衝液)25mLへ溶解し、最終反応液中の濃度が1.0mMとなるようにPPD溶液を調製した。調製したPPD溶液900μLへ任意濃度のヒシ果皮抽出物溶液100μLを添加し、混和後、37℃にて振とうさせながら4時間反応させた。反応液へ2NHClを200μL添加し反応を停止させ、0.45μmフィルターにてろ過を施しHPLC用サンプルとした。
AGE架橋切断活性は、タイプIコラーゲンとAGE-BSA間に形成された架橋をELISA法にて測定し、ヒシ果皮抽出物添加による架橋切断率を算出することにより評価した。
反復ART(高度生殖補助医療)不成功の難治性不妊の女性患者5名を被験者として、3ヶ月間にわたり、ヒシ果皮抽出物を含む経口投与剤を投与した。3ヶ月間の投与後、全員について血中HbA1c濃度が0.1~0.4ポイント低下した。その他、アトピー性皮膚炎の症状が軽快した被験者が1名、アレルギー性鼻炎(花粉症)の症状が軽快した被験者が1名、化粧乗りが改善した被験者が1名いた。被験者のうち、アトピー性皮膚炎及び糖尿病に罹患しており、これまで13回の体外受精が不成功に終わっていた40代女性1名が、試験期間中に体外受精による妊娠に成功した。
2014年7月~2015年3月に、反復ART(高度生殖補助医療)不成功の難治性不妊症32例(実施例6の5例を含む)に対し、説明と同意のもと、ARTの1~2ヶ月前よりヒシ果皮抽出物を連日投与(1日1回、朝食開始直前に100mgを内服)しながら、ARTを施行した。
Claims (5)
- ヒシ科に属する植物の果皮及び果実の一方又は双方に含まれる1又は複数の化合物を有効成分として含むことを特徴とする不妊の改善剤組成物。
- 前記有効成分として、果皮及び果実の一方又は双方の抽出物又は該抽出物より分離される1又は複数の化合物を含むことを特徴とする請求項1記載の不妊の改善剤組成物。
- タンパク質と糖からの終末糖化産物の生成に関連する1又は複数の反応を阻害する活性を有することを特徴とする請求項1又は2記載の不妊の改善剤組成物。
- 終末糖化産物の分解に関連する1又は複数の反応を促進する活性を有することを特徴とする請求項1から3のいずれか1項記載の不妊の改善剤組成物。
- アレルギー症状を軽減する活性を有することを特徴とする請求項1から4のいずれか1項記載の不妊の改善剤組成物。
