Abstract
Description
事前に閉鎖系培養容器を含む閉鎖系流路を自動培養装置に設置する。閉鎖系流路は閉鎖系培養容器101、細胞懸濁液の入った細胞ボトル106、培地の入った培地ボトル112、培養上清を回収する培養上清バッグ113等と、それらをつなぐ流路回路の流路チューブから成る。閉鎖系流路を設置後、設置正常性を確認する。
自動培養装置の実施する自動培養スケジュールを決定する。細胞播種、培地交換、培養上清回収、気体交換、顕微鏡観察、検査用組織回収、移植用組織回収等の操作を行う日時、液量等の条件を制御用端末110の入力部より入力する。
適切な電磁弁を開閉後、ポンプを作動させ細胞ボトル106より細胞懸濁液を吸引する。細胞懸濁液を閉鎖系培養容器101まで送液する。全ての閉鎖系培養容器に対し播種終了後、閉鎖系培養容器を設置する培養容器ベースに取り付けたアクチュエータを作動させ培養容器ベースへ傾きを与え揺動し細胞分布を一様化する。
細胞播種直後、各培養容器の内部へ所定量の気体を送気する気体交換を行う。気体交換は培養期間中、1日に数回程度の頻度でも実施する。供給する気体は例としてCO2濃度5%を含む空気を用いる。気体は各閉鎖系培養容器へガスボンベから気体フロメータにより流量を制御し加湿ボトルを通し、水分子を飽和させた状態で供給する。閉鎖系培養容器101へ送気後の不要な気体はフィルタを介し流路外へ排出する。フィルタは必要に応じ流路内の圧力を調整する。フィルタは、例えば0.22μm以上の粒子を通さない品質のものを使用する。
自動培養装置内に設置した顕微鏡を用い細胞画像を取得する。自動培養装置内に設置した光源を適宜発光させ、細胞に焦点を合わせ撮像する。取得した細胞画像は制御部102内の記憶部のデータベースに保存し、自動培養装置の制御用端末110のディスプレイ上で閲覧し細胞の状態を作業者が適宜確認する。また自動細胞撮影時以外は作業者が必要に応じ顕微鏡を手作業で操作し、細胞の観察、撮影を行う。
培地交換は培養期間中、数日に一度の頻度で実施する。冷蔵庫内の培地ボトル112で4℃にて保管されている培地を予熱ボトルまで送液し予熱する。最初に閉鎖系培養容器101から古い培地を排出する。この時、アクチュエータにより閉鎖系培養容器を傾け排出効率を向上させる。排出後、速やかに新しい培地を閉鎖系培養容器内へ供給する。古い培地は最終的に培養上清バッグ113へ排出する。必要に応じ培養上清バッグ内の培養上清を回収し、培地成分分析により細胞の生育状態を評価する。尚、培地交換は古い培地が入った状態で新しい培地を押し出す方式により実施しても構わない。
S50で移植予定日の前日であることが確認された場合、複数個の閉鎖系培養容器を同時培養している場合は、閉鎖系培養容器の一部を検査用に回収する。自動培養装置の扉を開け、検査用の閉鎖系培養容器101の流路チューブを熱溶着等の手段により無菌切断し取り出す。取り外した閉鎖系培養容器は安全キャビネット又はCPC外へ搬送し、速やかに検査を実施する。例えば生体試料の細胞数、生存率、特定タンパク質の発現等を評価する。
ステップS4と同じ操作による培養を行う。そしてステップS9を実施する直前に、ステップS6と同じ操作による培地交換を行う。必要に応じステップS5と同じ操作による顕微鏡観察も実施する。
ステップS7による評価の結果、移植に適した状態と判定した場合、生体試料を回収し再生医療治療に用いる。S7同様、閉鎖系培養容器を閉鎖系流路から無菌的に分離させインキュベータ103から取り出す。必要に応じ安全キャビネット内へ運び処理を行う。
手術室へ到着後、再生組織を閉鎖系培養容器から取り出す。開封時、閉鎖系培養容器の外側は菌等の生物や粒子が付着している可能性があるため、閉鎖系培養容器内が清浄性を維持するよう無菌的に開封する。
培養に用いた閉鎖系流路を取り外す。続いて、装置の内部へ適切な操作により、滅菌ガスによる滅菌或いはエタノールによる消毒を施し、清浄な状態にする。自動培養装置の各種ソフトを終了させ、自動培養装置の作動を終了させる。

- 培養容器と、前記培養容器を保持する保持部材を備え、
前記培養容器と前記保持部材は、培養に必要な液体或いは気体の送液或いは送気を行うコネクタ部を除いて閉鎖された培養空間を形成し、
前記保持部材は前記培養空間とは独立した空間を有する閉鎖系培養容器を用いて細胞培養を行い、
前記独立した空間により前記閉鎖系培養容器に対する衝撃を緩衝する、
ことを特徴とする細胞培養装置。 - 請求項1に記載の細胞培養装置であって、
前記独立した空間は半開放の空洞であり、前記閉鎖系培養容器の外部との間で気体の移動が自由である、
