Abstract

 励起光源10は、測定対象の試料2に励起光4を照射する。イメージセンサ20は、1次元または2次元に配列された複数の画素22を含み、励起光4に応答して試料2から発せられる測定光6を受ける。偏光選択素子30は、試料2とイメージセンサ20の間に挿入され、1次元または2次元に配列された複数の画素32を含む。各画素32は、測定光6の対応する部分を受け、当該画素32に印加される駆動信号S1に応じた偏光方向を有する測定光を選択し、イメージセンサ20に供給するように構成される。測定制御部40は、偏光選択素子30の各画素32に、第1周期Tを有する周期的な駆動信号S1を与えるとともに、第1周期Tを4つの露光時間T=T/4に区分し、各露光時間においてイメージセンサ20の各画素22から得られるデータI、I、I、Iを取得する。

Description

偏光分析装置
 本発明は、蛍光偏光解消法などに利用可能な偏光分析装置に関する。
 液体の粘度測定やバイオ分析の手法として、蛍光偏光解消法が用いられる。蛍光偏光解消法では、励起光偏光方向と水平な成分I||および垂直な蛍光成分Iをそれぞれ測定し、蛍光偏光度P=(I||-I)/(I||+I)として検出する。蛍光偏光解消法を用いると、通常では複数ステップが必要なイムノアッセイ(免疫検定法)がワンステップ化できる。

 しかし蛍光偏光解消法では、偏光方向が互いに垂直な2つの光I||,Iをそれぞれ測定した上で、蛍光偏光度Pを演算により検出する煩雑さのため、多サンプル測定はほとんどない。多点を同時に測定したい場合には、集光点を用いた一点測定を、集光点をスキャンしながら繰り返し行う必要があり、測定時間の長期化、装置の大型化、高コスト化の要因となっていた。
 本発明は係る課題に鑑みてなされたものであり、そのある態様の例示的な目的のひとつは、多点を同時測定可能な偏光分析装置の提供にある。
 本発明のある態様は偏光分析装置に関する。偏光分析装置は、測定対象の試料に励起光を照射する励起光源と、励起光に応答して試料から発せられる測定光を受ける1次元または2次元に配列された複数の画素を含むイメージセンサと、試料とイメージセンサの間に挿入され、1次元または2次元に配列された複数の画素を含み、各画素は、測定光の対応する部分を受け、当該画素に印加される駆動信号に応じた偏光方向を有する測定光を選択し、イメージセンサに供給するように構成された偏光選択素子と、偏光選択素子の各画素に、第1周期Tを有する周期的な駆動信号を与えるとともに、第1周期Tを複数N個(Nは整数)の露光時間T=T/Nに区分したときに露光時間T2_1~T2_Nにおいてイメージセンサの各画素から得られるデータI~Iを取得する測定制御部と、を備える。
 データI~Iは、露光時間T2_1~T2_Nそれぞれにおけるイメージセンサの入射光を積分、積算、累積した値であってもよい。この態様によると、偏光選択素子を用いることにより、直交する2つの偏光成分を測定することができ、画素ごとに得られるI~Iにもとづいて、試料の状態を1次元的あるいは2次元的な多点情報として取得することができる。
 N=4であり、データI~Iが取得されてもよい。
 この場合、測定光のAC振幅(AC値)を取得することができる。
 測定制御部は、各画素ごとに、

 を演算することにより、偏光度Pを測定してもよい。
 測定制御部は、第1周期Tを複数M個(M≧3、M≠N)の露光時間T=T/Mに区分し、露光時間T3_1~T3_Mそれぞれにおいてイメージセンサの各画素から得られる中間データIm~Imを取得し、中間データIm~ImをデータI~Iに変換してもよい。
 偏光選択素子は、液晶パネルと、偏光板と、を含んでもよい。
 これにより、駆動信号に応じた偏光方向を有する測定光を選択することができる。
 駆動信号は、正弦波であってもよいし、矩形波、のこぎり波、台形波などの周期波形であってもよい。「駆動信号がある波形(たとえば正弦波形)を有する」とは、駆動波形が厳密に正弦波形を有する場合のみでなく、それを近似した波形を有する場合も含む。
 なお、以上の構成要素の任意の組み合わせ、本発明の表現を、方法、装置などの間で変換したものもまた、本発明の態様として有効である。
 本発明のある態様によれば、多点を同時測定できる。

 以下、本発明を好適な実施の形態をもとに図面を参照しながら説明する。各図面に示される同一または同等の構成要素、部材、処理には、同一の符号を付するものとし、適宜重複した説明は省略する。また、実施の形態は、発明を限定するものではなく例示であって、実施の形態に記述されるすべての特徴やその組み合わせは、必ずしも発明の本質的なものであるとは限らない。
 図1は、実施の形態に係る偏光分析装置1の基本構成を示す図である。偏光分析装置1は、主として、励起光源10、イメージセンサ20、偏光選択素子30、測定制御部40、を備える。ここでは説明の簡潔化および理解の容易化のため、イメージの拡大、縮小を行わずに実像を測定するものとする。
 励起光源10は、測定対象の試料2に励起光4を照射する。たとえば励起光源10は、レーザである。本実施の形態において試料2は蛍光物質であり、励起光4により励起され、それに応じて発光する。したがって励起光源10の波長は、試料2に応じて選択される。試料2から出射する測定光6a(蛍光)の偏光方向は、試料2の状態、たとえばその温度や粘度、それに含まれる特定の物質の量、物質の種類などに応じて変化する。
 イメージセンサ20は、1次元または2次元に配列された複数の画素22を含み、励起光4に応答して試料2から発せられる測定光6aを受光するように配置される。たとえばイメージセンサ20としては、CCD(Charge Coupled Device)やCMOSセンサ、フォトダイオードアレイなどが利用可能であり、その種類は特に限定されない。後述するように、イメージセンサ20は、露光時間が制御、設定可能であることが要求される。
 偏光選択素子30は、1次元または2次元に配列された複数の画素32を含み、試料2とイメージセンサ20の間に挿入される。本実施の形態において偏光選択素子30は透過型デバイスであり、各画素32は、その裏面に測定光6aの対応する部分を受け、当該画素32に印加される駆動信号S1に応じた偏光方向を有する測定光6bを選択し、イメージセンサ20に供給するように構成される。
 たとえば各画素32は、駆動信号S1がある値SMINのとき、所定の第1方向の偏光成分Iを選択し、駆動信号S1が別の値SMAXのとき、第1方向と⊥な第2方向の偏光成分I||を選択し、通過させる。
 測定制御部40は、偏光選択素子30の各画素32に、第1周期Tを有する周期的な駆動信号S1を与える。また測定制御部40は駆動信号S1と同期して、第1周期TをN個(Nは整数)の露光時間T=T/Nに区分し、露光時間T2_1~T2_Nそれぞれにおいてイメージセンサ20の各画素から得られるデータI~Iを取得する。本実施の形態では、N=4の場合を説明する。
 以上が偏光分析装置1の基本構成である。続いて偏光選択素子30について説明する。
 図2(a)、(b)は、偏光選択素子30の構成例および動作原理を示す図である。
 たとえば、偏光選択素子30は透過型であり、液晶パネル34と偏光板36の組み合わせにより実現できる。液晶パネル34は、複数の画素35および図示しないゲートドライバやソースドライバを含む。各画素35には、独立した駆動信号S1が印加可能となっている。各画素35は、裏面から入射する光の偏光方向を、駆動信号S1のレベル(大きさ)に応じた角度θ、回転させる偏光素子である。たとえば駆動信号S1がSMINであるとき、回転角θは0度であり、駆動信号S1がSMAXであるとき、回転角θは90度であり、駆動信号S1が中間的な値をとるとき、回転角θは中間的な値をとる。駆動信号S1と回転角θの関係は、いわゆるγカーブと関連を有する。この画素35は、図1の画素32に対応する。たとえば8ビットの液晶パネル34では、駆動信号S1は、2=256階調で表され、したがってSMIN=0、SMAX=255に対応する。
 偏光板36は、液晶パネル34にオーバーラップしており、所定方向(図2においてX方向)の偏光成分を選択的に通過し、それと垂直方向(図2においてY方向)の偏光成分を遮断(吸収もしくは反射)する。
 一般的に市販されるLCD(Liquid Crystal Panel)パネルは、液晶パネル34と、液晶パネル34を挟むように配置された2枚の偏光板を含み、2枚の偏光方向が直交している。実施の形態に係る偏光選択素子30は、市販されるLCDパネルを分解し、1枚の偏光板を除去することにより構成することができる。
 図2(a)に示すように、測定光6aは、X方向の偏光成分I||と、Y方向の偏光成分Iを含む。図2(a)では、S1=SMINであり、液晶パネル34の各画素35は、X方向の成分I||および偏光成分Iそれぞれの偏光方向を回転させずに、そのまま通過させる。液晶パネル34を通過した測定光6cのうち、Y方向の偏光成分を有するI||’は、偏光板36を通過せず、X方向の偏光成分を有するI’は、偏光板36を通過する。つまり図2(a)の状態では、測定光6aのうち、X方向の偏光成分Iが選択される。
 図2(b)では、S1=SMAXであり、液晶パネル34の各画素35は、X方向の成分I||および偏光成分Iそれぞれの偏光方向を90度回転させて通過させる。そして液晶パネル34を通過した測定光6cのうち、Y方向の偏光成分を有するI’は、偏光板36を通過せず、X方向の偏光成分を有するI||’は、偏光板36を通過する。つまり図2(b)の状態では、測定光6aのうち、Y方向の偏光成分I||が選択される。
 測定光6aのX方向の偏光成分Iに着目すると、図2(a)のS1=SMINのときに、その透過率αが最大(≒1)となり、図2(b)のS1=SMAXのときに、その透過率αが最小(≒0)となる。中間的な状態(SMAX<S1<SMAX)では、透過率αは、S1に応じた中間値をとる。同様に測定光6aのY方向の偏光成分I||に着目すると、図2(a)のS1=SMINのときに、その透過率α||が最小(≒0)となり、図2(b)のS1=SMAXのときに、その透過率α||が最大(≒1)となる。中間的な状態(SMAX<S1<SMAX)では、透過率α||は、S1に応じた中間値をとる。
 任意の駆動信号S1の値について、偏光選択素子30は、液晶パネル34によって測定光6aの偏光方向が駆動信号S1に応じて回転角θだけ回転されるときに、回転後の偏光方向がX方向と一致する光を通過させるものと把握できる。
 以上が偏光選択素子30の構成例である。続いて図1の偏光分析装置1の動作を説明する。
 図3(a)~(c)は、偏光分析装置1の動作波形図である。図3(a)~(c)には、偏光選択素子30の1画素と、それに対応するイメージセンサ20の1画素の動作が示される。
 図3(a)に示すように、駆動信号S1としては、周期Tの正弦波が与えられる。その結果、偏光選択素子30の偏光成分I||に対する透過率α||は、駆動信号S1と同相で変化し、偏光選択素子30の偏光成分Iに対する透過率αは、駆動信号S1と逆相で変化する。
 図3(b)では、試料2からの測定光6aとして、I||>Iの蛍光が定常的に得られている場合を示している。駆動信号S1が最大値SMAXであるときのイメージセンサ20への入射光6bは、偏光成分I||に相当し、駆動信号S1が最大値SMINであるときのイメージセンサ20への入射光6bは、偏光成分Iを示している。
 図3(c)に示すように、イメージセンサ20は、偏光選択素子30と言い換えれば駆動信号S1と同期している。測定制御部40は、駆動信号S1の第1周期Tを4分割した第2周期Tをイメージセンサ20の露光時間として、露光時間T2_1~T2_4それぞれにおいて得られる入射光6bの受光量(輝度)を示す輝度データI、I、I、Iを出力する。
 以上が偏光分析装置1の動作である。輝度データI~Iは、たとえば蛍光偏光度Pの算出に利用することができる。
 イメージセンサ20と偏光選択素子30の対応する画素の位相関係を高精度に制御可能な場合、駆動信号S1の最小値SMINに対応する露光時間T2_1に得られる輝度データIをIとして、駆動信号S1の最大値SMAXに対応する露光時間T2_3に得られる輝度データIをI||として取得することができる。この場合には、蛍光偏光度Pを、式(1)から求めることができる。

 しかしながら、イメージセンサ20および偏光選択素子30の全画素について、駆動信号S1の位相と露光時間T2_1~T2_4の位相関係を一定に保つことは困難である。つまり、ある画素においては、露光時間T2_1が駆動信号S1の最小値SMINに対応するかもしれないが、別の画素においては、別の露光時間T2_2、T2_3、T2_4が、最小値SMINに対応するかもしれない。この場合、式(1)を用いても、偏光度Pを得ることはできない。
 このような問題に対処するために、測定制御部40は、以下の信号処理を行ってもよい。
 図3(b)に示すように、I||-Iは、測定光6bのAC振幅(AC値という)に対応し、(I||+I)/2は、測定光6bのDC成分(DC値という)に対応する。

 DC値に関しては、輝度データI~Iの平均値をとればよく、したがって、式(3)から求めることができる。

 これは、IとIから得られる振幅と、IとIから得られる振幅の、RMS(Root Mean Square)に相当する。
 当業者によれば、式(3)から得られるDC値は、4つの露光時間T2_1~T2_4と、駆動信号S1の位相関係に依存しないことが理解される。また詳しい証明は省略するが、駆動信号S1が正弦波である場合には、式(4)から得られるAC値は、4つの露光時間T2_1~T2_4と、駆動信号S1の位相関係に依存しない。
 したがって、測定制御部40は、式(5)を演算することにより偏光度Pを測定することができる。

 このように、実施の形態に係る偏光分析装置1によれば、1次元あるいは2次元にわたるすべての画素について、測定光6aのX方向の偏光成分I、測定光6aのY方向の偏光成分I||を別々に測定することなく、偏光度Pを同時に測定することができる。また偏光分析装置1は、測定光6aのX方向の偏光成分I、測定光6aのY方向の偏光成分I||それぞれについても、すべての画素について同時に測定することができる。
 すなわち、偏光分析装置1によれば、試料2の1次元あるいは2次元の偏光度Pの分布、偏光成分Iの分布、偏光成分I||の分布のいずれか、あるいは任意の組み合わせを同時に測定することができる。
 続いて、偏光分析装置1の具体的な用途について説明する。
 図4は、偏光分析装置1の具体的な構成例を示す図である。
 図4の偏光分析装置1は、液体の粘度を測定する。図1の励起光源10、イメージセンサ20、偏光選択素子30、測定制御部40に加えて、光学系50、カラーフィルタ52、を備える。
 試料2は、蛍光色素を混入した液体でありえる。たとえば試料2は、マイクロ流路を有するガラス管などのキャピラリーチューブ内に収容される。蛍光色素としては、フルオレセインなどが例示される。
 励起光源10は、試料2に溶かされた蛍光色素を励起可能な光源であり、蛍光色素としてフルオレセインを用いた場合、波長488nmの半導体レーザを用いてもよい。励起光源10と試料2の間には、励起光の強度を調節するためのND(Neutral Density)フィルタ54を挿入してもよい。イメージセンサ20は、CCDアレイである。偏光選択素子30は、図2に示した構造を有する。カラーフィルタ52は、励起光4がイメージセンサ20に入射するのを防止する。
 光学系50は、試料2を拡大/縮小し、偏光選択素子30およびイメージセンサ20のサイズに適合させるために設けられる。したがって試料2のサイズが偏光選択素子30やイメージセンサ20に比べて小さい場合、拡大光学系が採用され、試料2のサイズが偏光選択素子30やイメージセンサ20に比べて大きい場合、縮小光学系が採用されうる。当業者によれば、光学系50は、試料2、イメージセンサ20、偏光選択素子30のサイズ、位置関係に応じて適宜設計しうることが理解され、本発明において特に限定されるものではない。図4の例では、光学系50は、対物レンズ50a、接眼レンズ50dおよびその他のレンズ群50b、50cを含む。
 測定制御部40は、電源41、コンピュータ42、コンバータ43、ファンクションジェネレータ44、コンバータ45、コンピュータ46を含む。
 電源41は偏光選択素子30に電源電圧を供給する。コンピュータ42は、偏光選択素子30の各画素に対する駆動信号S1を生成する。コンバータ43は、コンピュータ42の出力信号を受け、偏光選択素子30の各画素を制御するインタフェースである。
 ファンクションジェネレータ44は、イメージセンサ20の露光時間を制御するための制御信号を生成する。コンバータ45は、ファンクションジェネレータ44が生成した制御信号にもとづいてイメージセンサ20の各画素の露光時間を制御するインタフェースである。コンピュータ46は、イメージセンサ20から第2周期Tにて出力される画像データ、すなわち各画素ごとの輝度データI~Iを受け、輝度データI~Iにもとづいて、必要な情報を演算する。
 なお測定制御部40の構成は例示に過ぎず、当業者によれば、別のデバイスを用いて同等の機能が実現しうることが理解される。たとえばコンピュータ42やコンピュータ46は、汎用コンピュータではなく、専用設計されたハードウェアおよびソフトウェアの組み合わせを用いてもよい。またファンクションジェネレータ44についても、専用設計された機能IC(Integrated Circuit)や、ソフトウェア制御されるマイコンを用いてもよい。
 図5(a)~(c)は、図4の偏光分析装置1を用いて、溶媒粘度に応じた蛍光偏光度Pを測定した結果を示す図である。図5(a)は蛍光偏光度Pを画像化したものであり、図5(b)は、蛍光偏光度Pの数値であり、図5(c)は、蛍光偏光度と粘度(CP)の関係を示す図である。
 この測定では、駆動信号S1の周波数は3Hzであり、第1周期Tは333msである。駆動信号S1は、正弦波を近似した信号であり、具体的には、1周期を8個の区間に分割し、区間ごとに駆動信号S1を同じ値とした。
 ファンクションジェネレータ44は、イメージセンサ20からの画像データのフレームレートを12Hzに設定した。このときの第2周期Tは、83.2msである。
 試料2としては、4本のガラスキャピラリー(外径0.5mm、内径0.32mm)を用意し、それぞれに、濃度が異なる水とエチレングリコール(EG)の溶液を収容した。EG比は、0,40,80,100である。EG比が高い方が溶液の粘度は高くなる。
 図5(a)~(c)から確認されるように、実施の形態に係る偏光分析装置1によれば、蛍光偏光度Pの2次元分布を測定することができる。
 図6(a)は、実施の形態に係る偏光分析装置1により測定された蛍光偏光度P(LCD-CCD system)と、従来手法(Conventional PF apparatus)により測定された、EG比と蛍光偏光度Pの関係を示す図である。図6(a)に示すように、偏光分析装置1によれば、従来手法と同等の精度で蛍光偏光度Pを測定することができる。なお、従来手法では、2次元画像は取得できないことに留意されたい。
 また、偏光分析装置1を用いて、クロラムフェニコール(Chloramphenicol、バクテリア由来の抗生物質)をイムノアッセイ(免疫検定法)により測定した結果を図6(b)に示す。横軸は、クロラムフェニコールの濃度を対数表示したものであり、縦軸は正規化された偏光度Pである。図6(b)には、あわせて従来の手法により測定された偏光度Pが示される。このように偏光分析装置1は、イムノアッセイによる抗生物質の測定にも利用できる。
 以上、本発明について、実施の形態をもとに説明した。この実施の形態は例示であり、それらの各構成要素や各処理プロセスの組み合わせにいろいろな変形例が可能なこと、またそうした変形例も本発明の範囲にあることは当業者に理解されるところである。以下、こうした変形例について説明する。
(第1変形例)
 実施の形態では、駆動信号S1が正弦波である場合を説明したが、本発明はそれには限定されない。正弦波に代えて、矩形波、台形波などの周期波形であってもよく、さらにはこれらの波形を近似した波形、多階調に量子化(離散化)した波形であってもよい。
(第2変形例)
 図7は、偏光分析装置1の光学系の変形例を示す図である。偏光選択素子30は、試料2とイメージセンサ20の間の、任意の位置に配置することが可能であるが、位置選択的に偏光解析を行いたい場合には、図7に示すように結像面に配置することが望ましい。また偏光選択素子30が十分に薄いとみなせる場合には、試料2あるいはイメージセンサ20に近接して配置してもよい。
(第3変形例)
 実施の形態では、偏光選択素子30を、液晶パネル34を利用して構成する場合を説明したが、本発明はそれには限定されない。偏光選択素子30は、液晶パネル34と偏光板36の組み合わせに代えて、それと等価な機能を有する公知の、あるいは将来利用可能なデバイスを用いることができる。また、偏光選択素子30は透過型デバイスである必要はなく、反射型のデバイスであってもよい。
(第4変形例)
 実施の形態では、測定光として蛍光を測定する場合を説明したが本発明はそれには限定されない。たとえば蛍光に代えて燐光を測定してもよい。あるいは試料が、励起光に応じて、非線形光学現象により2次高調波や3次高調波を発生する場合、それらを測定してもよい。つまり偏光分析装置1による測定対象は、試料2の状態に応じた偏光を有するさまざまな光を用いることができる。
(第5変形例)
 偏光分析装置1の用途は特に限定されるものではなく、イムノアッセイの他、以下の相互作用測定にも利用することができる。
 ・糖鎖-タンパク質の相互作用
 ・タンパク質間の相互作用
 ・DNA-タンパク質の相互作用
 ・ペプチド-タンパク質の相互作用
 ・リガンド-レセプターの相互作用
(第6変形例)
 実施の形態では、N=4として、4つの露光時間T2_1~T2_Nそれぞれにおけるイメージセンサの入射光を積分した値を示す輝度データI~Iを取得し、輝度データI~Iにもとづき、蛍光偏光度Pを測定したが、本発明はそれには限定されない。N≠4の場合であっても、I~Iにもとづいて、試料の状態を1次元的あるいは2次元的な多点情報として取得することができるという効果が得られる。たとえばAC値やDC値以外、あるいは蛍光偏光度P以外の情報が必要な場合には、N≠4とすることが考えられる。また後述の(DC値とAC値に関する考察)に示すように、正弦波やその他の一部の波形では、N=4とした場合に、AC値やDC値、ひいては蛍光偏光度Pを好適に計算できるが、駆動信号として別の波形を用いた場合には、N≠4である方が、AC値やDC値、ひいては蛍光偏光度Pを好適に計算しうる場合もある。
(第7変形例)
 実施の形態では、AC値やDC値、蛍光偏光度Pなど、最終的に生成すべき量を計算するために必要なN個の輝度データI~Iを、測定により直接取得したが、本発明はそれに限定されない。たとえば、第1周期Tを複数M個(M≧3,M≠N)の露光時間T=T/Mに区分し、露光時間T3_1~T3_Mそれぞれにおいてイメージセンサの各画素から得られる中間データIm~Imを取得し、中間データIm~Imを、最終的に生成すべき量を計算するために必要なN個の輝度データI~Iに変換してもよい。
 たとえば、M=8であれば、中間データIm、Imにもとづいて輝度データIを計算し、中間データIm、Imにもとづいて輝度データIを計算し、中間データIm、Imにもとづいて輝度データIを計算し、中間データIm、Imにもとづいて輝度データIを計算できる。
 そのほか、M=3、5、6、7であっても、AC値やDC値、蛍光偏光度Pなどを計算するために必要な輝度データI~Iを得ることができる。
(i)M=3
 以下に変換式の一例を示す。
 I=3/4×Im
 I=1/4×Im+1/2×Im
 I=1/2×Im+1/4×Im
 I=3/4×Im
 図8(a)は、M=3のときの波形図である。左は、正弦波Iと露光時間T3_1~T3_3の関係、右は、位相φとAC値の計算結果を示す。
 I(ωt)=DC+AC×sin(ωt+φ)
 T3_1=0~2π/3
 T3_2=2π/3~4π/3
 T3_3=4π/3~2π
 AC値は、露光時間T3_1~T3_3と、正弦波の位相差φに応じた値をとる。AC値は理想的には2であるが、上記変換式から得られる輝度データI~Iにもとづいて計算されるAC値は、1.42±0.10となる。これはM=N=4として輝度データI~Iを直接取得する場合に比べて、70%まで感度が低下し、7%の相対誤差が生ずることを意味する。感度低下を許容すれば、φをランダムにとって複数回演算すれば、十分小さい誤差となる。
(ii)M=5
 以下に変換式の一例を示す。
 I=Im+1/4×Im
 I=3/4×Im+1/2×Im
 I=1/2×Im+3/4×Im
 I=1/4×Im+Im
 図8(b)は、M=5のときの波形図である。理想的に2であるAC値は、1.76±0.03となり、85%まで感度が低下し、1%の相対誤差が生ずる。感度低下を許容すれば、φをランダムにとって複数回演算すれば、十分小さい誤差となる。
(iii)M=6
 以下に変換式の一例を示す。
 I=Im+1/2×Im
 I=1/2×Im+Im
 I=Im+1/2×Im
 I=1/2×Im+Im
 図8(c)は、M=6のときの波形図である。理想的に2であるAC値は、1.89±0.10となり、94%まで感度が低下し、5%の相対誤差が生ずる。感度低下を許容すれば、φをランダムにとって複数回演算すれば、十分小さい誤差となる。
(iv)M=7
 以下に変換式の一例を示す。
 I=Im+3/4×Im
 I=1/4×Im+Im+1/2×Im
 I=1/2×Im+Im+1/4×Im
 I=3/4×Im+Im
 図8(d)は、M=7のときの波形図である。理想的に2であるAC値は、1.87±0.10となり、93%まで感度が低下し、1%の相対誤差が生ずる。感度低下を許容すれば、φをランダムにとって複数回演算すれば、十分小さい誤差となる。
 当業者によれば、それ以外のMについても、同様の結果が得られることが理解される。また、M=3,5,6,7に関する変換式は、例示したそれらには限定されず、以下のように一般化することができる。
 I=Σi=1~M(K1i×Im
 I=Σi=1~M(K2i×Im
 I=Σi=1~M(K3i×Im
 I=Σi=1~M(K4i×Im
 Σj=1~4ji=1 (i=1,2,・・・M)
 係数Kjiを最適化することで、上の例よりも正確な測定が可能となる場合もある。
 N≠4の場合に一般化すれば、以下の式を得る。
 I=Σi=1~M(Kji×Im)  (j=1,2,…N)
 Σj=1~Nji=C (i=1,2,…M)
 ただしCは任意の定数であり、上の例ではC=1の場合を説明した。
(DC値とAC値に関する考察)
 式(A)で与えられる波形を考える。
 I(ωt)=DC+AC×sin(ωt+φ)   …(A)
 ωは角周波数、tは時間、φは任意の位相である。
 1周期(0≦ωt≦2π)を4つの区間に分割し、それぞれの区間について積分すると以下の式を得る。
 I=∫0~π/2I(ωt)dt
 I=∫π/2~πI(ωt)dt
 I=∫π~3π/2I(ωt)dt
 I=∫3π/2~2πI(ωt)dt
 数学的に、DC値およびAC値は、以下の式で与えられる。
 DC=(I+I+I+I)/4×2/π   …(B)
 AC=√{(I-I+(I-I}/(2√2)…(C)
 図9(a)~(d)は、異なる波形I(ωt)における、位相φと式(C)で与えられるAC値の関係を示す図である。図9(a)は、DC=10,AC=2の正弦波の場合である。式(C)のAC値はφに依存しない。このことは、イメージセンサ20による露光時間T2_1~T2_4と、駆動信号S1の位相差に関わらず、式(3)、(4)が成り立つことの数学的根拠となる。
 図9(b)は、I(ω)が矩形波の場合である。
 I(ωt-φ)=8 (0<ωt<π)
 I(ωt-φ)=12 (π<ωt<2π)
 矩形波の場合、式(C)で計算されるAC値は、φに応じて2.23~3.11までの値をとり、最大11.7%の系統誤差を生ずる。ここでは1回の測定について系統誤差とよぶ。φをランダムにとって複数回演算し、平均値をとることで、精度を高めることができる。駆動信号S1を矩形波とする場合、正弦波に比べて精度は低下するが、偏光分析装置1の制御を簡素化できるという利点がある。
 図9(c)は、I(ωt)がのこぎり波の近似波形の場合である。
 I(ωt)=DC+AC×[sin(ωt+φ)+1/2・sin2(ωt+φ)
  +1/3sin3(ωt+φ)]
 つまりI(ωt)は、のこぎり波をフーリエ級数展開し、ω、2ω、3ωまでとり、第4項以下を切り捨てた波形である。この場合、AC値はφに応じて1.78~2.22までの値をとり、最大7.8%の系統誤差を生ずる。φをランダムにとって複数回の平均値を演算すれば、偶然誤差として扱うことができる。
 図9(d)は三角波の近似波形の場合である。
 I(ωt)=DC+AC×X
 X=sin(ωt+φ)-1/9・sin3(ωt+φ)+
  +1/25・sin5(ωt+φ)-1/81・sin9(ωt+φ)
 この場合、AC値はφに応じて1.94~2.05までの値をとり、最大2.1%の系統誤差を生ずる。φをランダムにとって複数回の平均値を演算すれば、偶然誤差として扱うことができる。
 このように、駆動信号S1の波形として、正弦波以外の波形が採用しうることが数学的に証明される。
 実施の形態にもとづき、特定の語句を用いて本発明を説明したが、実施の形態は、本発明の原理、応用を示しているにすぎず、実施の形態には、請求の範囲に規定された本発明の思想を逸脱しない範囲において、多くの変形例や配置の変更が可能である。

Claims (6)

  1.  測定対象の試料に励起光を照射する励起光源と、
     前記励起光に応答して前記試料から発せられる測定光を受ける1次元または2次元に配列された複数の画素を含むイメージセンサと、
     前記試料と前記イメージセンサの間に挿入され、1次元または2次元に配列された複数の画素を含み、各画素は、前記測定光の対応する部分を受け、当該画素に印加される駆動信号に応じた偏光方向を有する前記測定光を選択し、前記イメージセンサに供給するように構成された偏光選択素子と、
     前記偏光選択素子の各画素に、第1周期Tを有する周期的な前記駆動信号を与えるとともに、前記第1周期Tを複数N個(Nは整数)の露光時間T=T/Nに区分したときに露光時間T2_1~T2_Nそれぞれにおいて前記イメージセンサの各画素から得られるデータI~Iを取得する測定制御部と、
     を備えることを特徴とする偏光分析装置。
  2.  前記N=4であり、データI~Iが取得されることを特徴とする請求項1に記載の偏光分析装置。
  3.  前記測定制御部は、各画素ごとに、
     √{(I-I+(I-I}/(I+I+I+I
     を演算することにより、偏光度Pを測定することを特徴とする請求項2に記載の偏光分析装置。
  4.  前記測定制御部は、前記第1周期Tを複数M個(M≧3,M≠N)の露光時間T=T/Mに区分し、露光時間T3_1~T3_Mそれぞれにおいて前記イメージセンサの各画素から得られる中間データIm~Imを取得し、前記中間データIm~Imを前記データI~Iに変換することを特徴とすることを特徴とする請求項1から3のいずれかに記載の偏光分析装置。
  5.  前記偏光選択素子は、
     液晶パネルと、
     偏光板と、
     を含むことを特徴とする請求項1から4のいずれかに記載の偏光分析装置。
  6.  前記駆動信号は、正弦波、矩形波、のこぎり波、台形波のいずれかであることを特徴とする請求項1から5のいずれかに記載の偏光分析装置。
