Abstract
Description
(1)不純物として含むアセトアミド、2,3,4,5-テトラヒドロピリジン、2-ピペリドン、α-アミノ-ε-カプロラクタム、リジン、およびオルニチンの総含有量が150重量ppm以下であることを特徴とする1,5-ペンタメチレンジアミン。
(2)前記アセトアミドの含有量が70重量ppm以下であることを特徴とする(1)に記載の1,5-ペンタメチレンジアミン。
(3)バイオマス資源由来のリジンから製造される前記(1)から(2)のいずれか一項に記載の1,5-ペンタメチレンジアミン。
(4)1,5-ペンタメチレンジアミンを、温度40℃以上200℃以下、圧力0.2kPa以上1200kPa以下、理論段数5段以上で精留することを特徴とする前記(1)から(3)のいずれか一項に記載の1,5-ペンタメチレンジアミンの製造方法。
バイオマス資源から製造された1,5-ペンタメチレンジアミンには通常不純物として1,5-ペンタメチレンジアミン以外の含窒素有機化合物が含まれる。中でもアセトアミド、2,3,4,5-テトラヒドロピリジン、2-ピペリドン、α-アミノ-ε-カプロラクタム、リジン、およびオルニチンなどの含窒素有機化合物は、1,5-ペンタメチレンジアミンを用いたポリアミド樹脂を重合する際に重合遅延や、着色の原因となる。
本発明の1,5-ペンタメチレンジアミンは、原料1,5-ペンタメチレンジアミンを精留することにより得ることができる。原料1,5-ペンタメチレンジアミンは、グルコースやリジンなどのバイオマス資源由来の化合物から、酵素反応や、酵母反応、発酵反応などによって合成されるものが好ましい。これらの方法は、有機合成法に比べ、反応温度を100℃未満で実施できるため副反応が抑制され、不純物として含まれる含窒素有機化合物の量が少ない高純度な1,5-ペンタメチレンジアミンを得ることが出来るため好ましい。具体的な原料1,5-ペンタメチレンジアミンとして、日本国特開2004-000114号公報、日本国特開2005-006650号公報、日本国特開2004-222569号公報、国際公開第2007/1113127号などに記載された製造方法により得られた1,5-ペンタメチレンジアミンが挙げられる。
原料1,5-ペンタメチレンジアミン中に不純物として含まれるアセトアミド、2,3,4,5-テトラヒドロピリジン、2-ピペリドン、ε-カプロラクタム、α-アミノ-ε-カプロラクタム、リジン、およびオルニチンの含有量の合計を150重量ppm以下とする方法としては、精留がある。本発明における精留は、棚段塔または充填塔のいずれを用いた手法でもよく、さらには連続式でもバッチ式でもよい。
本発明の1,5-ペンタメチレンジアミンは、不純物として含有するアセトアミド、2,3,4,5-テトラヒドロピリジン、2-ピペリドン、α-アミノ-ε-カプロラクタム、リジン、およびオルニチンの含有量の合計が150重量ppm以下であるので、これをポリアミド樹脂の原料として用いることで、良好な色調のポリアミド樹脂を与えることが可能である。
1,5-ペンタメチレンジアミン中の不純物の分析は下記に示す条件でGC法により実施した。

Oven :50℃→(8℃/min)→280℃
Inj. Temp. :280℃
Inj. Mode :Splitless
Inj. Vol. :1.0μL
Det. Temp :280℃
試料0.25gを濃度98wt%の硫酸100mlに対してサンプル濃度0.01g/mLになるように溶解し、オストワルド型粘度計を用いて25℃での流下時間(T1)を測定した。引き続き、濃度98wt%の硫酸のみの流下時間(T2)を測定した。T2に対するT1の比、すなわちT1/T2を相対粘度(ηr)とした。
試料1gを50mLのフェノール/エタノール混合溶液(体積比;フェノール/エタノール=80/20)に、30℃で振とう溶解させて溶液とし、この溶液を0.02Nの塩酸で中和滴定し、要した0.02N塩酸の量を求めた。また、上記フェノール/エタノール混合溶媒(上記と同量)のみを0.02N塩酸で中和滴定し要した0.02N塩酸の量を求める。そしてその差から試料1gあたりのアミノ基量を求めた。
スガ試験機(株)製のカラーコンピューターを用いてペレットのYI値を測定した。該測定方法はJIS規格K7373(2006)(プラスチックの光学的特性試験方法)に従って測定した。
リジン脱炭酸酵素(CadA)を高発現するプラスミドpcadA220は、特開2008-193898号公報に記載の方法で構築した。pcadA220は、改変型のEnterobacter属の酸性フォスファターゼ遺伝子の構成発現型プロモーター及びリボソーム結合部位の下流に、E.coli由来リジン脱炭酸酵素遺伝子(CadA)を連結し、CadAを構成的に高発現するプラスミドである。このプラスミドで、E.coli JM109株(宝酒造(株)製)を形質転換し、得られた形質転換株をE.coli cadA220と命名した。この菌株をLB培地で前培養した後、26mlの培養液を26Lのシード培地(25g/L Glucose、1g/L MgSO4・7H2O、2g/L KH2PO4、5g/L (NH4)2SO4、大豆塩酸加水分解物 無機窒素換算0.45g/L、20mg/L FeSO4・7H2O、20mg/L MnSO4・5H2O、Thiamine HCl、0.1ml/L消泡剤、pH5.0)に接種し、28℃、250rpm、pH7、通気量26L/分で通気撹拌培養を行った。培養pHの調整はアンモニアで行った。22時間後、シード培養液1.3Lを26Lのメイン培地(培地組成はシード培地と同じ)に接種し、通気量26L/分、30℃、250rpm、pH7、通気量26L/分で12時間通気撹拌培養を行った。培地pHの調整はアンモニアで行い、メイン培養培地中の糖が消費されたことを確認して培養を終了し、E.coli cadA220培養液を得た。
原料のリジンは、LLB50(50%(W/V)リジンベース溶液、AJINOMOTO EUROLYSINE S.A.S.)を用いた。終濃度200g/L リジンベース、26.5mg/L ピリドキサルリン酸(和光純薬工業製)、380U/ml リジン脱炭酸酵素(参考例1で調製)となるように650Lの反応液を調製した。なお、反応液は硫酸でpHを7に調整し、酵素溶液を添加することで反応開始とし、硫酸でpHを7に維持しながら、37℃で10時間反応させ、リジンが完全に消費されたことを確認して反応を終了した。反応終了後、反応液のpHを硫酸で5まで下げ、80℃、30分加熱することで殺菌し、1,5-ペンタメチレンジアミン硫酸塩を含む反応液を調整した。この溶液に活性炭を1,5-ペンタメチレンジアミンに対して10wt%添加し、1時間撹拌後、フィルタープレス分離機により、活性炭と菌体を1,5-ペンタメチレンジアミン溶液から分離した。得られた濾過液に、1,5-ペンタメチレンジアミン硫酸塩と2倍モル以上の水酸化カルシウムを添加することによって、1,5-ペンタメチレンジアミン硫酸塩を1,5-ペンタメチレンジアミンに変換し、沈殿物として生成した硫酸カルシウムを遠心分離した。得られた上清を約45wt%になるまで濃縮し、蒸留することにより、1,5-ペンタメチレンジアミンを得た。得られた1,5-ペンタメチレンジアミン中の不純物の含有量は表1の通りであった。
参考例2にて得られた1,5-ペンタメチレンジアミン1LをヘリパックNo.4を充填した蒸留塔(理論段数27段)の塔底に仕込み、内温105℃(ヒーター温度150℃)、圧力1.8kPa、還流比2にて蒸留を実施した。得られた精製1,5-ペンタメチレンジアミン中の含窒素有機化合物の含有量は表1の通りであった。
参考例2にて得られた1,5-ペンタメチレンジアミン1LをヘリパックNo.4を充填した蒸留塔(理論段数15段)の塔底に仕込み、内温105℃(ヒーター温度150℃)、圧力1.8kPa、還流比2にて蒸留を実施した。得られた精製1,5-ペンタメチレンジアミン中の不純物の含有量は表1の通りであった。
参考例2にて得られた1,5-ペンタメチレンジアミン700gを1L3つ口フラスコに仕込み、80℃、1.0kPaにて単蒸留を実施した。得られた精製1,5-ペンタメチレンジアミン中の不純物の含有量は表1の通りであった。
参考例1にて製造した1,5-ペンタメチレンジアミンを用いた以外は実施例1と同様の方法でポリアミド樹脂組成物のペレットを製造した。得られたポリアミド樹脂組成物は淡黄色に着色しており、相対粘度は2.67、アミノ末端基量は5.62×10-5mol/g、YIは11.6であった。
Claims (4)
- 不純物として含むアセトアミド、2,3,4,5-テトラヒドロピリジン、2-ピペリドン、α-アミノ-ε-カプロラクタム、リジン、およびオルニチンの総含有量が150重量ppm以下であることを特徴とする1,5-ペンタメチレンジアミン。
- 前記アセトアミドの含有量が70重量ppm以下であることを特徴とする請求項1に記載の1,5-ペンタメチレンジアミン。
- バイオマス資源由来のリジンから製造される請求項1から2のいずれか一項に記載の1,5-ペンタメチレンジアミン。
- 1,5-ペンタメチレンジアミンを、温度40℃以上200℃以下、圧力0.2kPa以上1200kPa以下、理論段数5段以上で精留することを特徴とする請求項1から3のいずれか一項に記載の1,5-ペンタメチレンジアミンの製造方法。
