Description
METHOD FOR NORMALIZING REPRODUCTIVE FUNCTION The invention relates to medicine and can be used to normalize (improve, restore, stimulate) the reproductive function of mammals and, in particular, humans.
Restoring reproductive health in modern society is an important task, since the last decade has been characterized by various technological and natural disasters, a deteriorating environmental situation, urbanization, and the instability of the political and economic situation, which naturally leads to an increase in stressful situations. Reproductive function is not directly involved in adaptation to stress. At the same time, taking a passive position, it temporarily reduces or suspends its function, thus yielding blood flow and energy to other systems that ensure survival under stress. This evolutionarily formed defense mechanism contributes to the survival of the species. But in modern conditions, when stress is part of everyday life, this adaptive reaction leads to the formation of pathology, so the search for new means and methods of restoring reproductive function is an urgent problem.
The literature describes various methods of normalizing reproductive function in animals and humans through the use of various drugs and physical factors.
М., Литера, 2006, Sperling Н. et al. New treatment options for erectile dysfunction. Pharmacologic and no
pharmacologic options. Herz. For example, methods for restoring reproductive function using drugs are known: RU 2363467, RU 2149607, WO 94/22460, US 5932548, Alyaev Yu.G. et al. Violation of sexual and reproductive function in men. M., Litera, 2006, Sperling, H. et al. New treatment options for erectile dysfunction. Pharmacologic and no pharmacologic options. Herz. 2003 Jun; 28 (4): 314-24 and others. A common drawback of these methods is the possibility of side effects of drugs, as well as the development of allergic reactions. In addition, the treatment is high-quality, while the effect of the therapy is not always observed.
As the closest analogue, a method for improving reproductive function by introducing into the body the mineral complex and vitamins according to patent RU 23521 10. However, the known method is invasive, vitamins obtained synthetically can provoke allergic reactions, while the achieved improvement is not persistent.
Т.о. T.O. The objective of the present invention is to provide a method for normalizing (improving, restoring, stimulating) reproductive health while achieving a technical result, which consists in eliminating invasiveness, as well as providing a multi-faceted effect on the body, expressed in protecting the body from the harmful effects of the environment and while improving the functions of both individual systems and the whole organism.
To achieve the specified technical result, a well-known product for a new purpose was used, disclosed in patent RU 2412604 - milk whey enriched with lactates (SMOL), which contains 7% solids, of which 5% whey and 2% sodium hydroxide, or ammonium carbonate, or calcium hydroxide obtained by hydrolysis of whey pasteurized at a temperature of 53-57 ° C under cultivation conditions of Lactobacillus acidophilum and Lactobacillus bulgaricum at a cultivation temperature of 36 to 40 ° C and neutralizing the resulting milk to Slots sodium hydroxide or ammonium carbonate, or calcium hydroxide to achieve the lactose mass fraction of not more than 0.2%. Also after thickening and drying, the preparation can be obtained in condensed or dry form. Condensed, it contains 44% solids, of which 32% are whey solids and 12% solids are sodium hydroxide, or ammonium carbonate, or calcium hydroxide. In dry form - contains 95% solids, of which 68% are whey solids and 27% are calcium hydroxide solids.
The resulting preparation contains whey protein, oligopeptides and free amino acids, macro- and microelements (K, Na, Ca, Mg, Zn, Mn, Cr, Ni, Se); vitamins (A, K, D, E, C, gr. B, PP), does not contain hormones, antibiotics, genetically modified and artificial additives. All its components are of natural origin. In this case, the resin contains a high number of viable lactic acid bacteria up to 7x10000000 CFU / g, the content of calcium lactate in 100 g of the drug reaches 73.3 g.
Thus, the preparation contains a unique complex of lactobacilli Lactobacillus acidophilus, Lactobacillus bulgaricum. Great attention to lactobacilli is due to the fact that representatives of this genus do not participate in the occurrence of any pathological processes in the human body, but, on the contrary, have a positive effect on human health.
Acidophilic lactobacilli are widely used for the prevention and treatment of patients with various types of acute and chronic diseases of the digestive tract, inflammatory processes of the respiratory tract, and bacterial infections of the genitourinary system. Representatives of L. acidophilus are used in the same way as antioxidants and lipid peroxidase lowering agents stimulate the growth of other lactobacilli and bifidobacteria. These microorganisms have antitumor activity and stimulate various parts of the immune system. The fact of the pronounced effect of L. acidophilus on the body’s immune system through the stimulation of monocyte migration, activation of phagocytic activity is established. The resulting product has light antibiotic properties, but unlike antibiotics, it does not kill the intestinal microflora, does not lose its properties, with prolonged incubation, the pathogenic microflora does not develop resistance to it.
Along with calcium lactate, the drug contains a set of vital minerals - potassium, sodium, phosphorus, magnesium, iron, zinc, selenium and trace elements in the form of organic natural compounds, which ensures their high digestibility.
Based on a set of vitamins, SMOL is a biologically complete product. It does not contain vitamins obtained chemically. The presence of vitamins in the preparation is ensured by the preservation of biologically active substances of whey and the cultivation of lactobacilli during the process.
Thus, the revealed new property of SMOL, i.e. the ability to normalize (improve, stimulate) the reproductive function of the body, in combination with the properties of this drug listed above ensure the achievement of the above technical result, i.e. in addition to restoring reproductive function, it additionally provides a multidimensional effect on the body, i.e. protects the body from the harmful effects of the environment and improves the functions of both individual systems and the whole organism.
A study of the effect of SMOL on reproductive function was carried out in an experiment on rabbits, pigs and minks, and preliminary tests in humans were carried out. Since similar data were obtained on all animal species, we present the experimental data obtained on rabbits. Groups of animals were selected and formed according to the method of groups of analogues (age, live weight, breed). The study was conducted on the background of feeding rabbits with a full-fledged standard compound feed, balanced in terms of nutritional energy, amino acid composition, micro and macro elements. The study was carried out separately on females and males.
Research on females. Each of the studied groups - control and three experimental, consisted of 9 rabbits. The control group received standard compound feed, in experimental group 1, SMOL in liquid form was added to the feed in the amount of 9% of the daily norm of the main coma; in experimental group 2, resin was added to the feed in the form of a condensed form, in the amount of 6% of the daily norm of the main feed; in experimental group 3 - in the form of a dry form, in the amount of 1.5% of the daily norm of the main feed. The study was carried out for 45 days.
The effect of additives on the body was assessed by clinical and physiological parameters of blood, and reproductive function of female rabbits: by the effectiveness of coming to the hunt, the effectiveness of insemination, the safety of offspring, the functional state of the ovaries and uterine endometrium.
Efficiency of coming into sexual hunting among females of the experimental groups was higher, differences with control were 12.0-18.1%. In the experimental groups, insemination efficiency reached 100%, while in the control group this indicator did not exceed 40%. Moreover, the yield of rabbits per female was 20-21% higher compared to the control.
1When conducting histological and cytological studies also revealed significant differences. In the experimental groups, a positive effect of the SMOL preparation on the number of oocyte-cumulus complexes (OCC), as well as the maturation and embryonic development of artificially activated rabbit oocytes, was established (Table 1) Table 1
The effect of SMOL additives on the weight, morphofunctional state, and folliculogenesis of rabbit ovaries.
5 The data obtained indicate a higher potency for further embryonic development of oocytes obtained from females of the experimental groups, which explains the data on the high efficiency of insemination in comparison with the control.
2). When analyzing the clinical and physiological parameters of blood 10, it was found that in the experimental groups there was a slight increase in the total number of leukocytes in the blood, which may indicate an indirect immunostimulating effect of the additives used (Table 2).

Analysis of the dynamics of various forms of leukocytes showed that in all groups there was an increase in the number of lymphocytes, which can also indirectly indicate an increase in humoral immunity. Moreover, the greatest effect was observed in experimental group 3 when using the drug in dry form.
3). The introduction of SMOL additives into the diet of female rabbits contributed to the improvement of a number of indicators of non-specific animal immunity (Table 3).

When analyzing the red blood cell in the experimental groups, an increase in indicators of the values of red blood was observed.
15 Research on male rabbits. The effect of SMOL additives on the body was evaluated by clinical and physiological parameters of blood, and reproductive function indicators by indicators: ejaculate volume, sperm concentration in the ejaculate, sperm motility, percentage of sperm with intact acrosome, sperm survival rate outside the body. Groups were formed in the same way as for females (see above) - control and three experimental groups.
The volume of ejaculate in the experimental groups increased by 10%, while in the control group remained at the same level. A similar trend was observed in the dynamics of changes in the concentration of sperm in the ejaculate, i.e. in the experimental groups, by the end of the observation period, the concentration increased by 16.6%. Moreover, the age-related characteristics of animals in the experimental groups were leveled due to the addition of SMOL. An analysis was made of indicators for assessing the state of acrosome of spermatozoa as an indicator characterizing the usefulness of sperm and their fertilizing ability, i.e. indicator, which is accepted as a reliable indicator of assessing the quality of the seed. Before the introduction of additives into the feed, both in the control and in the experimental groups, the acrosome status indicator was high (91-92%), which indicates the usefulness of the sperm of males used in the experiment.
Other important indicators characterizing the fertilizing ability of sperm and the effectiveness of insemination are sperm motility and their survival outside the body.
4In males of the experimental groups, sperm motility increased by an average of 8.0% by the beginning of the experiment by day 21, and by an average of 14.7% by the end of the experiment. In the control group, this indicator did not change significantly, by 2.8% by day 21), by 4.2% by the end of the experiment (Table 4). Table 4
Sperm motility in male rabbits, depending on the introduction of SMOL into the diet
* Tables 4 and 5 show the average data for the three experimental groups, since the indicators obtained in 1, 2 and 3 experimental groups were similar.
5). It was also found a significant improvement in the indicator of sperm survival outside the body in the experimental groups: this indicator compared to the beginning of the experiment increased by 21 days on average by 1, 4 times, by the end of the experiment by 2 times. In the control group, sperm survival rate increased by 1.04% by the 21st day, and by 12.0% by the end of the experiment (Table 5).

92,5% . Thus, the study revealed a positive effect of the SMOL preparation on sperm production, including age males. Moreover, in males with low sperm production the effect of this drug was more pronounced: the volume of the ejaculate, the concentration of sperm in the ejaculate, the mobility and survivability of sperm increased by 25.0, 27.3, 14.3, 148.0%, respectively. In males with normal sperm production, with a 8.3% decrease in ejaculate volume, an increase in sperm concentration, motility and survivability was 12.6, 3.5 and 92.5%, respectively.
Clinical and physiological parameters of blood and nonspecific immunity were similar to those obtained for groups of female rabbits. The phagocytic activity of blood serum increased in the control groups by an average of 16%, in the control group by 1, 2%. The phagocytic index in the experimental groups increased by 10.5%, in the control - 3.9%. A similar trend was observed in the bactericidal and lysosome activity of blood serum, which are humoral factors in the protection of natural resistance, i.e. these indicators increased, respectively, in males of the experimental groups on average by 9.2 and 48.2%, in males of the control group - by 2.2 and 14.3%. confirmed the positive effect of the resin on the resistance of the animal organism.
6). As a result of histological studies of the testes of rabbits, it was found that in the experimental group the diameter of the seminiferous tubules is 8.7% larger than in the control group (Table 6).

Thus, the study objectively confirms that the SMOL preparation can normalize, increase and stimulate the reproductive qualities of males and females of rabbits, while the specific and nonspecific resistance of the body increases and erythropoiesis increases. When using various forms of the drug SMOL (dry, condensed, liquid forms), no significant differences were revealed.
Taking into account the obtained results, preliminary studies of the SMOL preparation were performed in patients with male menopause.
The study involved 7 men aged 46-68 years. All patients complained of fatigue, decreased performance, weakened memory, irritability, while all patients complained of sexual weakness. Based on the studies, it was found that these conditions are the result of predominantly hormonal changes in the body that are characteristic of this age group, and in three cases it was impossible to exclude the impact of various environmental and professional factors.
During the study, patients received SMOL in the form of a dry powder, i.e. in a more convenient form for practical use. The drug was administered orally at a daily dose of 200 mg / kg for 90 days. Observation was carried out for a year. The use of the drug contributed to an improvement in overall well-being, while all patients noted an improvement in sexual function in the form of a decrease in the manifestations of sexual weakness.
Microscopic examination of the ejaculate revealed an increase in the number of sperm (on average from 32 million / ml to 83 million / ml), their motility (on average 1.5-2.0 times), viability (on average from 74% to 87%), a decrease in the pathological forms of sperm (on average from 78%) to 20%) and an increase in the Farris rate (on average from 51 to 200).
7). A regulatory effect on the content of sex hormones in the blood was also revealed, while a significantly significant increase in testosterone was detected not only by the end of the course of treatment, but also throughout the entire observation period (Table 7).

Thus, the results of the studies indicate a positive effect of the used resin on the reproductive function of the body. At the same time, the specific and nonspecific resistance of the body increases and erythropoiesis increases.
