Abstract
標的腫瘍細胞等、所望の細胞で効果的に増殖しうるアデノウイルス(Ad)を提供することを課題とする。また、所望の細胞でAdを効果的に増殖させる、Ad増殖促進方法を提供する。さらに、Adを効果的に増殖しうるAd産生用パッケージング細胞を提供する。標的腫瘍細胞等、所望の細胞で効果的に増殖しうるAdは、AdゲノムにおいてAd由来mivaRNAの機能を抑制すること、又はAdゲノムにおいてmiR-27a及び/又はmiR-27bを阻害することにより作製される。Ad由来mivaRNAの機能抑制、あるいはmiR-27a及び/又はmiR-27bの阻害は、少なくともDicer又はAgo2のいずれか一方を阻害することにより達成される。
Description
本発明は、所望の細胞で効果的に増殖しうるアデノウイルス(以下、「Ad」という。)に関する。さらにAdの増殖促進方法に関する。
本出願は、参照によりここに援用されるところの日本出願特願2013-172810号優先権を請求する。
近年、様々な疾患に対する遺伝子治療法の開発が試みられている。遺伝子治療法開発の鍵は、安全かつ効率良く治療遺伝子を目的の組織や細胞へと送達する技術開発であるが、克服すべき問題点も多く残されているのが現状である。
Adベクターは、その優れた遺伝子導入特性から、遺伝子導入用ベクターとして細胞の形質転換による研究や産業向けの遺伝子組み換え細胞の製造に用いられており、また、遺伝子治療用ベクターとして各種疾患への適用が期待されている。一方、制限増殖型Adの特徴は、正常な細胞では増殖せず、腫瘍細胞や特殊な細胞のみで増殖する点である。Ad(2 型又は5型)の感染は、細胞表面のcoxsackie-adenovirus receptor(CAR)にウイルスのタンパク質の殻(カプシド)から突出するファイバーの先端のノブが吸着することにより始まり、細胞表面のインテグリンとファイバーの根本のペントンベースが結合して細胞内に侵入する。その後ライソゾームを酸性化してライソゾームから脱出し、ウイルスゲノムは核内に至り、ゲノムの複製が始まる。Adは他のDNA ウイルスと比較してウイルスゲノムの核内への移行効率が高く、その結果ベクターとしても遺伝子導入効率が高いと考えられている。
Ad複製は、まずE1Aタンパク質が発現し、他の全ての初期タンパク質であるE1B、E2、E3 及びE4の発現をトランスアクティベートすることにより始まる。一般的に応用されているE1置換型ベクターはこのE1A やE1Bが欠失しているため、E1タンパク質が恒常的に発現しているHEK293細胞では野生型とほぼ同程度の複製が可能である。HEK293細胞以外の細胞ではベクターゲノムを先の侵入方法に則って効率よく核内に移行するものの、それ以降の全てのウイルス由来のタンパク質の発現が起きないためウイルスとして複製することがない。このようなベクターは非増殖型に分類される。
一方、Adを腫瘍細胞でのみ複製させて腫瘍細胞を死滅させるという試みも行われている。正常細胞では増殖せず、腫瘍細胞特異的に感染・増殖し腫瘍細胞を死滅させる制限増殖型Adは、次世代の腫瘍治療薬として期待を集めている。例えば、p53 欠損細胞でのみ増殖が可能なE1Bの変異ウイルスやE1Aを腫瘍細胞特異的プロモーターから発現させ、腫瘍細胞でのみ複製させる等の制限増殖型Adが開発されている。更に、制限増殖型AdにGFP等のレポーター遺伝子を搭載し、腫瘍細胞でのみ複製させることによりGFPを産生させて蛍光発光させ、腫瘍細胞を検出する技術が開発されている。しかしながら、従来の制限増殖型Adは、腫瘍細胞における増殖速度が遅いために、十分な治療効果が得られないことが問題点として挙げられる。
一方、Adを腫瘍細胞でのみ複製させて腫瘍細胞を死滅させるという試みも行われている。正常細胞では増殖せず、腫瘍細胞特異的に感染・増殖し腫瘍細胞を死滅させる制限増殖型Adは、次世代の腫瘍治療薬として期待を集めている。例えば、p53 欠損細胞でのみ増殖が可能なE1Bの変異ウイルスやE1Aを腫瘍細胞特異的プロモーターから発現させ、腫瘍細胞でのみ複製させる等の制限増殖型Adが開発されている。更に、制限増殖型AdにGFP等のレポーター遺伝子を搭載し、腫瘍細胞でのみ複製させることによりGFPを産生させて蛍光発光させ、腫瘍細胞を検出する技術が開発されている。しかしながら、従来の制限増殖型Adは、腫瘍細胞における増殖速度が遅いために、十分な治療効果が得られないことが問題点として挙げられる。
また、遺伝子導入用ベクター、遺伝子治療用ベクター、腫瘍治療薬、腫瘍検出薬、その他の用途に用いられる制限増殖型Ad、非増殖型Ad又はその他のAdを産生するためのAd産生用パッケージング細胞において、Adの増殖速度が遅いために、十分な量のAdを効果的に得られないことが問題点として挙げられる。
Adの増殖を促進するウイルス側因子として小分子RNAであるVA-RNAが挙げられる。VA-RNAにはVA-RNA I とVA-RNA IIが存在するが、ともに約160塩基長の非翻訳RNAポリメラーゼIII転写産物(一本鎖RNA)であり、特に転写量が多いVA-RNA Iは、Ad複製時には細胞あたり~108分子にも達する。VA-RNA Iは、Ad複製時に宿主細胞の抗ウイルス反応としてのプロテインキナーゼR(protein kinase R:PKR)の活性化を阻害する中心的な働きをしており、VA-RNA I非存在下ではPKRが活性化し、翻訳開始因子のeIF-2αのリン酸化が誘導され、結果としてウイルスmRNAの翻訳が阻害されることが報告された(非特許文献1、2)。すなわち、VA-RNAはPKRを阻害することにより、ウイルス遺伝子の翻訳を促進、ひいてはウイルスの増殖を促進していると考察されている。また、VA-RNAはADAR(RNA-specific adenosine deaminase)を阻害することも報告された(非特許文献3)。
VA-RNAが核内で転写されたのち、核外輸送タンパク質であるExportin-5(XPO5)によって核外、すなわち細胞質に輸送され、Dicerによる切断を受けた後mivaRNA(viral miRNA)を形成し、Ago(Argonaute)タンパク質とともにRISC(RNA-induced silencing complex)を形成することが報告された(非特許文献4)。RISCに取り込まれたmivaRNAは、miRNA(microRNA)と同様の機構により感染細胞の遺伝子発現を制御するとともに、ウイルス増殖を促進することが報告された(非特許文献5)。恐らくこれはmivaRNAがmiRNAと同様の機構で感染細胞の遺伝子発現を制御することで、感染細胞内をウイルス増殖に適した環境にしているものと考えられた(非特許文献6)。実際に、mivaRNAのアンチセンスオリゴヌクレオチドを細胞に導入することでAdの増殖が抑制されている。
従来の制限増殖型Adに比べて、標的腫瘍細胞でのウイルス増殖を促進して抗腫瘍効果を高める高い増殖能を有する制限増殖型Adの開発が求められている。

本発明は、標的腫瘍細胞等、所望の細胞で効果的に増殖しうるAdを提供することを課題とする。また、本発明は所望の細胞でAdを効果的に増殖させる、Ad増殖促進方法を提供することを課題とする。さらに、本発明はAdを効果的に増殖しうるAd産生用パッケージング細胞を提供することを課題とする。
本発明者らは上記課題を解決するために、Adゲノムにおいて、Ad由来mivaRNAとウイルス増殖との関係に着目し、鋭意検討を重ねた結果、従来はAd由来mivaRNAが感染細胞の遺伝子発現を抑制しウイルス増殖を促進すると考えられていた(非特許文献5、6)にも関わらず、驚くべきことに、Ad由来mivaRNAの機能を抑制することでAdが細胞内で効果的に増殖し、上記課題を解決しうることを見出し、本発明を完成した。また、Ad由来VA-RNAの切断を抑制することにより、VA-RNAのウイルス増殖促進効果を得ることができることを見出した。
すなわち本発明は、以下よりなる。
1.アデノウイルスの増殖を促進する方法であって、アデノウイルス由来mivaRNAの機能を抑制することを含む、アデノウイルスの増殖促進方法。
2.アデノウイルスの増殖を促進する方法であって、miR-27a及び/又はmiR-27bを阻害することによるアデノウイルスの増殖促進方法。
3.mivaRNAの機能を抑制、あるいはmiR-27a及び/又はmiR-27bを阻害することが、少なくともDicer又はAgo2のいずれか一方を阻害することによる、前項1又は2に記載のアデノウイルスの増殖促進方法。
4.Dicer又はAgo2の阻害が、RNA干渉によるDicer又はAgo2の阻害である、前項3に記載のアデノウイルスの増殖促進方法。
5.RNA干渉によるDicer又はAgo2の阻害が、Dicer又はAgo2に対するsiRNAによる阻害であって、Dicer又はAgo2に対するsiRNAが、当該アデノウイルスに搭載されたDicer又はAgo2に対するsiRNA、アデノウイルス産生用パッケージング細胞に導入されたDicer又はAgo2に対するsiRNA、あるいはDicer又はAgo2に対するsiRNAである、前項4に記載のアデノウイルスの増殖促進方法。
6.Dicer又はAgo2の阻害が、アデノウイルス産生用パッケージング細胞におけるDicer遺伝子又はAgo2遺伝子のノックアウトによる阻害である、前項3に記載のアデノウイルスの増殖促進方法。
7.Dicer又はAgo2の阻害が、Dicer又はAgo2の抗体による阻害、あるいはDicer又はAgo2の阻害剤による阻害である、前項3のアデノウイルスの増殖促進方法。

本発明のAdは、従来のAdに比べて効果的にAdを増殖促進させることができる。本発明のAdを有効成分として含む抗腫瘍剤によれば、効果的に標的となる腫瘍細胞内で増殖が促進され、抗腫瘍効果が発揮される。
本発明のAdの増殖促進方法によれば、従来の方法に比べて、効果的にAdを増殖促進させることができる。標的腫瘍細胞やAd増殖用細胞等、所望の細胞で効果的にAdを増殖させることができれば、効果的に腫瘍溶解等の抗悪性腫瘍効果、腫瘍検出効果やAdの効率的な産生効果を発揮しうる。
本明細書において、「Ad」とはアデノウイルスを意味し、「Adゲノム」は、Adが有する遺伝情報、即ちAdに含まれるDNA配列をいう。
本発明は、Ad由来mivaRNAの機能を抑制することを含む、Adの増殖促進方法に関する。本明細書において、「mivaRNAの機能を抑制する」とは、「mivaRNA産生の阻害」又は「mivaRNAの活性阻害」を意味する。具体的には、「VA-RNAの切断を阻害することによるmivaRNA産生の阻害」又は「mivaRNAとAgo2のRISC形成を阻害することによるmivaRNA活性の阻害」を意味する。このような「mivaRNAの機能を抑制する」ことは、少なくともDicer又はAgo2のいずれか一方を阻害することにより達成される。また、Dicerを阻害することにより「VA-RNAの切断を阻害すること」によって、VA-RNAのウイルス増殖促進効果を得ることもできる。
本発明は、miR-27a及び/又はmiR-27bを阻害することによるAdの増殖促進方法にも関する。本明細書において、miR-27a及び/又はmiR-27bの阻害は、少なくともDicer又はAgo2のいずれか一方を阻害することにより達成される。他の方法としては、例えば後述するmiR-27a又はmiR-27bに対するデコイ核酸による阻害、miR-27a又はmiR-27bに対する阻害剤による阻害、miR-27a又はmiR-27bのノックアウトによる阻害等が挙げられる。
Dicer又はAgo2の阻害の方法としては、特に制限されないが、例えばDicer又はAgo2に対するRNA干渉による阻害、Dicer遺伝子又はAgo2遺伝子のノックアウトによる阻害、あるいはDicer又はAgo2に対する抗体や阻害剤の使用による阻害等が挙げられる。好適には、RNA干渉による阻害や遺伝子のノックアウトによる阻害であり、最も好適にはRNA干渉による阻害である。Dicer又はAgo2に対するRNA干渉による阻害としては、Adゲノム又はAd産生細胞にDicer又はAgo2に対するsiRNA作用させることで達成することができる。Dicer又はAgo2に対するsiRNA作用させるために、Dicer又はAgo2に対するsiRNAの発現カセットをAdゲノムに搭載し、あるいはAd産生細胞に導入することができる。また、Dicer又はAgo2に対するsiRNAをAd産生細胞の培養系に添加してもよい。
Dicer又はAgo2に対するsiRNAとしてはDicer又はAgo2に対してRNA干渉機能を有するsiRNAであればよく、特に制限されず、自体公知又は今後開発されるあらゆるsiRNAであってもよい。自体公知のDicerに対するRNA干渉機能を有するsiRNAやshRNAとしては、以下の各論文に示すshRNAがあげられる。Dicer又はAgo2に対するRNA干渉による阻害や、Dicer遺伝子又はAgo2遺伝子のノックアウトによる阻害の手法は、自体公知の方法、例えばNucleic Acids Research, (2006) Vol. 34, No. 17 4801-4815(doi:10.1093/nar/gkl646)、Cell 141, 1195-1207, June 25 (2010)、Molecular Therapy vol. 17 no. 4, 725-732 apr.(2009)、Nucleic Acids Research, Vol. 38, No. 21, 7736-7748 (2010)、Nucleic Acids Research, Vol. 38, No. 15 5141-5151 (2010)、PNAS, vol.107, no.17, 7904-7909, April 27, (2010)、JOURNAL OF VIROLOGY, Vol. 85, No. 5, p.2342-2350, Mar. 「Adの増殖を促進する方法」において、RNA干渉を用いてDicer又はAgo2を阻害する場合は、Adに搭載されたDicer又はAgo2に対するsiRNAの発現による阻害で達成できる。また同様に、Ad産生用パッケージング細胞に導入されたDicer又はAgo2に対するsiRNAの発現で達成できる。Dicer又はAgo2に対するsiRNAの具体例は、上述のとおりである。ここにおいて、「Dicer又はAgo2に対するsiRNAの発現」とは、Dicer又はAgo2に対するsiRNAを含むsiRNAの発現カセットであってもよい。「Dicer又はAgo2に対するsiRNA」を含むsiRNAの発現カセットは、例えば「Dicer又はAgo2に対するsiRNA」を含むsiRNAの発現カセットであってもよいし、「Dicer又はAgo2に対するshRNA」を含むsiRNAの発現カセットであってもよい。shRNAはsiRNAの前駆体であるため、shRNAが発現されたのちにsiRNAが産生されることも、本明細書において「siRNAの発現」に含まれる。
「Adの増殖を促進する方法」において、Dicer又はAgo2の抗体、あるいはDicer又はAgo2の阻害剤を用いてDicer又はAgo2を阻害する場合は、自体公知の抗体や阻害剤(デコイ核酸やアプタマー核酸を含む)を使用することができる。前記抗体や前記阻害剤によってAd増殖用細胞のDicer又はAgo2を阻害することで、当該Ad増殖用細胞におけるAdの増殖を促進することができる。
「Adの増殖を促進する方法」において、miR-27a又はmiR-27bを阻害する場合は、miR-27a又はmiR-27bに対するデコイ核酸による阻害、miR-27a又はmiR-27bに対する阻害剤による阻害が挙げられる。例えばmiR-27a又はmiR-27bに対するデコイ核酸をAdに作用させ、miR-27a又はmiR-27bを阻害することができる。具体的にはmiR-27a又はmiR-27bに対するデコイ核酸の発現カセットを、Adゲノムに搭載し、あるいはAd産生細胞に導入することができる。また、miR-27a又はmiR-27bに対する阻害剤としては、miR-27a又はmiR-27bに対するアンチセンスオリゴヌクレオチドや、miR-27a又はmiR-27bの阻害剤として市販されている試薬、例えばmiRVana miRNA inhibitor(Ambion社)を用いてもよい。
miR-27aに対するデコイ核酸は、配列番号10に示す塩基配列を少なくとも1又は2以上含む配列であればよい。miR-27aに対するデコイ核酸の具体例として、以下の配列番号12に示す塩基配列からなるオリゴヌクレオチドが挙げられる。配列番号12の下線に示す部位が、miR-27aに対するアンチセンス配列であり、配列番号10に示す塩基配列に該当する。また、miR-27bに対するデコイ核酸は、配列番号11に示す塩基配列を少なくとも1又は2以上含む配列であればよい。同様に、miR-27bに対するデコイ核酸の具体例として、以下の配列番号13に示す塩基配列からなるオリゴヌクレオチドが挙げられる。配列番号13の下線に示す部位が、miR-27bに対するアンチセンス配列であり、配列番号11に示す塩基配列に該当する。遺伝子組換えの手法により、Adや細胞にmiR-27a又はmiR-27bに対するデコイ核酸を発現させるために、適宜必要なオリゴヌクレオチドを含んでいてもよい。

本発明の「Adの増殖を促進する方法」において増殖が促進されるAdは、特定の細胞種でのみ複製できるようにしたAd、即ち制限増殖型Adや非増殖型Adベクターが挙げられるが、特に好適には制限増殖型Adである。本明細書の背景技術の欄でも示した通り、正常細胞では増殖せず、腫瘍細胞特異的に感染・増殖し腫瘍細胞を死滅させる又は腫瘍細胞を検出する制限増殖型Adは、次世代の腫瘍治療薬、腫瘍検出薬として期待を集めているが、従来の制限増殖型Adは腫瘍細胞における増殖速度が遅いために、十分な治療効果が得られないのが問題点であった。本発明の「Adの増殖促進方法」により、Adを腫瘍細胞特異的に感染・増殖させることができれば、Adを腫瘍細胞でのみ複製させて腫瘍細胞を死滅させる抗腫瘍剤又は腫瘍細胞を検出する検出剤として有効に利用することができる。制限増殖型Adは、E1タンパク質の発現が例えば腫瘍細胞でのみ起こるAdであり、NATURE MEDICINE, 7, 781-787 (2001)に報告されているものを例示することができる。例えば、腫瘍細胞特異的に遺伝子発現を制御するプロモーターを搭載するAdが挙げられる。このような機能を発揮しうるプロモーターであれば、自体公知のプロモーター又は今後開発されるプロモーターのいずれであってもよい。例えばhTERTプロモーター、IAI.3Bプロモーター、ミッドカインプロモーター、β-HCGプロモーター、SCCA1プロモーター、cox-2プロモーター、PSAプロモーター、又はその他の腫瘍特異的プロモーターが挙げられる。目的とする遺伝子の発現に用いるウイルスに対応しうる、適切なプロモーターとしては、腫瘍特異的プロモーターの他、例えばCMVプロモーター、SV40後期プロモーター、MMTV LTRプロモーター、RSV LTRプロモーター、SRαプロモーター等を用いることができる。
本発明の増殖が促進される「Ad」は、Adゲノム において、Ad由来mivaRNAの機能を抑制するか、あるいはmiR-27a及び/又はmiR-27bを阻害するように工夫されていることが必要である。具体的には、Dicer又はAgo2に対するデコイ核酸やアプタマー核酸などの発現カセットや、Dicer又はAgo2の遺伝子に対するアンチセンスオリゴヌクレオチドなどの発現カセットが搭載されていてもよい。また、miR-27a又はmiR-27bの抑制のみに着目した場合は、miR-27a又はmiR-27bに対するデコイ核酸やアプタマー核酸などの発現カセットや、miR-27a又はmiR-27bに対するアンチセンスオリゴヌクレオチドなどの発現カセットが搭載されていてもよい。Dicer及びAgo2各々に対するsiRNA 又はshRNA、あるいはmiR-27a又はmiR-27bに対するデコイ核酸は上述したごとくである。
本明細書においてAdは、代表的にはヒトを宿主とする2型、5型、11型、35型の各Ad、ヒト以外を宿主とするサルAd、マウスAd、イヌAd、ヒツジAd及びトリAdなどが挙げられる。
本明細書において、E1領域欠損型Adとは、AdゲノムのうちE1タンパク質を発現させないようにE1遺伝子領域を機能的に欠損するAdをいう。E1遺伝子領域とは、例えばヒト5型Adゲノム(GenBank Accession番号:M73260, M29978)において、具体的には342~3523番目に相当する。E1遺伝子領域を機能的に欠損するとは、例えば、宿主細胞内で機能するE1タンパク質を発現させないようにすることであり、E1タンパク質をコードするゲノム領域のすべてを欠損している必要はない。本発明のAdに使用可能なAdは、E1領域の他に、例えばE3遺伝子領域を機能的に欠損する5型Adであっても良い。5型Adゲノム(GenBank Accession番号:M73260, M29978)のE3領域とは、28133~30818番目又は27865~30995番目に相当する部位をいう。
また、本発明の「Adの増殖を促進する方法」を、少なくとも「Ad」と「Ad増殖用細胞」のいずれかに適用することで、Adの産生を促進することができる。この場合のAdとしては、制限増殖型Adに限定されず、非増殖型Adであってもよい。制限増殖型Ad及び非増殖型Adのいずれであっても、in vitroでウイルス増殖を促進することができれば、効果的にAdを活用することができる。Adを効果的に増殖促進させて作製するための「Ad増殖用細胞」としては、「Ad産生用パッケージング細胞」が挙げられる。
本明細書において、「Ad産生用パッケージング細胞」とは、Adを産生しうる細胞であればよく、特に限定されない。Ad産生用パッケージング細胞に使用可能な細胞は、各種腫瘍細胞の他、HEK293細胞が挙げられる。Adを効果的に増殖促進させて作製するためのAd産生用パッケージング細胞は、Ad由来mivaRNAの機能を抑制するか、あるいはmiR-27a及び/又はmiR-27bを阻害するように工夫されている細胞であることが必要である。Dicer又はAgo2に対するデコイ核酸やアプタマー核酸などの発現カセットや、Dicer又はAgo2の遺伝子に対するアンチセンスオリゴヌクレオチドなどの発現カセットが導入されていてもよい。また、miR-27a又はmiR-27bの抑制のみに着目した場合は、miR-27a又はmiR-27bに対するデコイ核酸やアプタマー核酸などの発現カセットや、miR-27a又はmiR-27bに対するアンチセンスオリゴヌクレオチドなどの発現カセットが導入されていてもよい。Dicer及びAgo2各々に対するsiRNA 又はshRNA、あるいはmiR-27a又はmiR-27bに対するデコイ核酸は上述したごとくである。
本発明は、本発明のAdを有効成分として含む抗腫瘍剤又は腫瘍検出剤にも及ぶ。本発明の抗腫瘍剤は、錠剤、カプセル剤、散剤、顆粒剤、丸剤、液剤、シロップ剤等の経口投与剤、注射剤、外用剤、坐剤、点眼剤等の非経口投与剤などの形態に応じて、経口投与又は非経口投与することができる。好ましくは、腫瘍、筋肉、腹腔等への局部注射、静脈への注射等が例示される。本発明のAdを有効量含む抗腫瘍剤又は腫瘍検出剤は、常法に従って製造され、希釈剤として一般に生理食塩水、細胞培養液等を用いることができる。さらに必要に応じて、殺菌剤、防腐剤、安定剤、等張化剤、無痛化剤などを加えてもよい。
本発明の抗腫瘍剤は、ほぼ全ての悪性腫瘍に適用することができ、治療対象となる癌の種類は、卵巣癌、扁平上皮癌(子宮頚部癌、皮膚癌、頭頚部癌、食道癌、肺癌等)、消化器癌(大腸癌、膵癌、肝癌、胃癌等)、神経芽細胞腫、脳腫瘍、乳癌、精巣癌、前立腺癌などが例示される。
本発明は、悪性腫瘍の治療方法にも及ぶ。具体的には、上述のいずれかの悪性腫瘍の治療及び/又は予防のために、本発明の抗腫瘍剤を有効量投与することによる。本発明の抗腫瘍剤は、そのまま患部に適用することもできるし、あらゆる公知の方法、例えば、静脈、筋肉、腹腔内又は皮下等の注射、あるいは鼻腔、口腔又は肺からの吸入、経口投与、カテーテルなどを用いた血管内投与等により生体(対象となる細胞や臓器)に導入することもできる。また、例えば凍結などの方法により扱いやすくした後、そのまま若しくは賦形剤、増量剤、結合剤、滑沢剤等公知の薬学的に許容される担体、公知の添加剤(緩衝剤、等張化剤、キレート剤、着色剤、保存剤、香料、風味剤、甘味剤等が含まれる。)などと混合することができる。
治療のための投与量は、腫瘍の大きさ・種類、症状の程度、投与経路、投与対象、患者の年齢、体重などに応じて適宜決定することができる。本発明の抗腫瘍剤は、数回にわたり患者に投与してもよいし、複数回のクールに分け、一クール当たりの投与回数、投与間隔などを任意に設定してもよい。一日投与量として、通常有効成分である本発明ウイルスの量を106~1011PFU(plaque forming units)程度、好ましくは109~1011PFU程度とすることができる。また、本発明の抗腫瘍剤を使用する際には、公知の免疫抑制剤等を用いることにより、生体の免疫を抑制し、有効成分として含まれるAdを感染し易くすることもできる。更に、本発明の抗腫瘍剤は、公知の抗癌剤及び放射線からなる群から選ばれる少なくとも1種の抗癌剤を併用することもできる。抗癌剤には、以下のものがあるが、これらに限定されない。
(a)アルキル化活性剤:この製剤は、癌細胞の核酸タンパク質にアルキル基を導入して細胞障害を起こさせる作用を有するものであり、例えばカルボコン、プスルファン(マスタード薬)、ニムスチン(ニトロソウレア類)などがあげられる。
(b)代謝拮抗活性剤:この製剤は、代謝過程で酵素に拮抗して、細胞合成を阻害する作用を有するものであり、例えばメトトレキサート(葉酸系)、メルカプトプリン(プリン系)、シタラビン(ピリミジン系)、フルオロウラシル、テガフール、カルモフールなどがあげられる。
(c)抗生物質:抗癌作用を有する、アクチノマイシンD、プレオマイシン、アドリアマイシン、マイトマイシンCなどがあげられる。
(d)微小管阻害活性剤:この製剤は、微小管に作用して抗腫瘍効果を示すものであり、例えばドセタキセル、パクリタキセル(タキサン)、ビノレルビン、ピンクリスチン、ビンブラスチン(アルカロイド系)などがあげられる。
(e)白金製剤:この製剤は、DNA鎖内又は鎖間結合あるいはDNAタンパク質結合を構成して、DNA合成を阻害する作用を有するものであり、例えば、シスプラチン、アルボプラチン、ネダプラチンなどがあげられる。
(f)トポイソメラーゼ阻害活性剤:トポイソメラーゼを阻害する、イリノテカン(トポイソメラーゼI阻害薬)、ポドフィロトキシン誘導体(トポイソメラーゼII阻害薬)などがあげられる。なお、トポイソメラーゼとは、DNAに一時的に切れ目を入れてDNA鎖のリンキング数を変える反応を触媒する酵素である。
(a)アルキル化活性剤:この製剤は、癌細胞の核酸タンパク質にアルキル基を導入して細胞障害を起こさせる作用を有するものであり、例えばカルボコン、プスルファン(マスタード薬)、ニムスチン(ニトロソウレア類)などがあげられる。
(b)代謝拮抗活性剤:この製剤は、代謝過程で酵素に拮抗して、細胞合成を阻害する作用を有するものであり、例えばメトトレキサート(葉酸系)、メルカプトプリン(プリン系)、シタラビン(ピリミジン系)、フルオロウラシル、テガフール、カルモフールなどがあげられる。
(c)抗生物質:抗癌作用を有する、アクチノマイシンD、プレオマイシン、アドリアマイシン、マイトマイシンCなどがあげられる。
(d)微小管阻害活性剤:この製剤は、微小管に作用して抗腫瘍効果を示すものであり、例えばドセタキセル、パクリタキセル(タキサン)、ビノレルビン、ピンクリスチン、ビンブラスチン(アルカロイド系)などがあげられる。
(e)白金製剤:この製剤は、DNA鎖内又は鎖間結合あるいはDNAタンパク質結合を構成して、DNA合成を阻害する作用を有するものであり、例えば、シスプラチン、アルボプラチン、ネダプラチンなどがあげられる。
(f)トポイソメラーゼ阻害活性剤:トポイソメラーゼを阻害する、イリノテカン(トポイソメラーゼI阻害薬)、ポドフィロトキシン誘導体(トポイソメラーゼII阻害薬)などがあげられる。なお、トポイソメラーゼとは、DNAに一時的に切れ目を入れてDNA鎖のリンキング数を変える反応を触媒する酵素である。
本発明は、悪性腫瘍の高感度な検出方法にも及ぶ。具体的には、上述のいずれかの悪性腫瘍の検出のために、GFP等のレポーター遺伝子を搭載した本発明の腫瘍検出剤を対象となる細胞、培養細胞、培養組織、採取血液または生検組織に添加すること、あるいは生体に投与することによる。本発明の腫瘍検出剤は、そのまま生体の患部に適用することもできるし、あらゆる公知の方法、例えば、静脈、筋肉、腹腔内又は皮下等の注射、あるいは鼻腔、口腔又は肺からの吸入、経口投与、カテーテルなどを用いた血管内投与等により生体(対象となる細胞や臓器)に導入することもできる。また、例えば凍結などの方法により扱いやすくした後、そのまま若しくは賦形剤、増量剤、結合剤、滑沢剤等公知の薬学的に許容される担体、公知の添加剤(緩衝剤、等張化剤、キレート剤、着色剤、保存剤、香料、風味剤、甘味剤等が含まれる。)などと混合することができる。
悪性腫瘍検出のための使用量は、対象の採取条件、培養条件、腫瘍の大きさ・種類、症状の程度、投与経路、投与対象、動物種、患者の年齢、体重などに応じて適宜決定することができる。本発明の腫瘍検出剤は、数回にわたり生体に投与してもよいし、複数回のクールに分け、一クール当たりの投与回数、投与間隔などを任意に設定してもよい。また、生体の血液や生検材料を採取して検査することもできる。例えば、J. Clin. Invest., 119:3172-3181 (2009)やVNATURE MEDICINE, 12:1214-12193181 (2006)を参照しつつ、本発明の腫瘍検出剤の使用量を適宜決定することができる。
さらに、本発明の増殖促進方法によれば、従来の方法に比べて、効果的に有効成分としてのAdを増殖促進させることができる。本発明のAdの増殖が促進される細胞は、卵巣癌、扁平上皮癌(子宮頚部癌、皮膚癌、頭頚部癌、食道癌、肺癌等)、消化器癌(大腸癌、膵癌、肝癌、胃癌等)、神経芽細胞腫、脳腫瘍、乳癌、精巣癌、前立腺癌などの標的腫瘍細胞が挙げられる。標的腫瘍細胞等、所望の細胞で効果的にAdを増殖することができれば、効果的に腫瘍溶解等の抗悪性腫瘍効果や腫瘍検出効果を発揮しうる。そして、本発明の増殖促進方法を、細胞を用いたin vitroでのAdの産生系、例えばHEK293細胞等を用いたウイルスの産生系に適用することで、より効果的にAdを産生することができる。
以下に実施例及び実験例並びに参考例を示して本発明をさらに具体的に説明するが、本発明はこれらにより限定されるものではない。
(実施例1)Dicerに対するshRNAを搭載したAdの作製
本実施例では、Dicerに対するshRNA(以下、実施例、実験例において「shDicer」ということとする。)を搭載したカセットであって、Dicerに対するsiRNAを発現しうるカセットを搭載したAdの作製について説明する。以降、本実施例のAdを「Ad-shDicer」ということとする。作製したAd-shDicerのゲノム構造は図1に示すとおりである。
本実施例では、Dicerに対するshRNA(以下、実施例、実験例において「shDicer」ということとする。)を搭載したカセットであって、Dicerに対するsiRNAを発現しうるカセットを搭載したAdの作製について説明する。以降、本実施例のAdを「Ad-shDicer」ということとする。作製したAd-shDicerのゲノム構造は図1に示すとおりである。
1)Ad-shDicerゲノムの作製
本実施例では、ヒト5型Adゲノム(GenBank Accession番号:M73260, M29978)において、当該ゲノムのうちE1領域(342~3523)並びにE3領域(28133~30818)を欠損しており、E1欠損領域に外来遺伝子を挿入できるベクタープラスミドを元に構築した。当該ベクタープラスミドpAdHM19のE1欠損領域に、腫瘍特異的プロモーターであるhTERTプロモーターと、E1A、IRES、E1B、ポリA(pA) シグナル配列をライゲーションして搭載し、腫瘍溶解性(Oncolytic)Adゲノムを得た。さらに、E3欠損領域に、shDicerの発現カセットを搭載した。本実施例において使用したshDicerの発現カセットにおけるshDicerに対応する配列は、配列番号5に示す塩基配列からなる。RNA干渉機能の発揮に重要であるのは、配列番号5において、下線に示す塩基配列、具体的には配列番号6又は配列番号7に示す塩基配列部分であると考えられる。

2)Ad-shDicerの作製
上記1)で作製したAd-shDicerゲノムを、ウイルスゲノム末端に存在する制限酵素PacIで切断して洗浄し、トランスフェクション試薬Lipofectamine2000TM(Invitrogen社)を用いてHEK293細胞へトランスフェクションし、Ad-shDicerを作製した。

(実験例1-1)Ad-shDicerによる癌細胞内でのAdゲノム増殖能の確認
実施例1で作製したAd-shDicerを各種癌細胞に作用させたときのウイルスゲノム量を確認した。癌細胞は、HeLa細胞(ヒト子宮頸癌細胞)、SK HEP-1細胞(ヒト肝細胞癌細胞)及びH1299細胞(ヒト非小細胞肺癌細胞)について確認した。従来型Oncolytic Adコントロールとして、実施例1で作製したAd-shDicerと同様のゲノム構造であって、shRNAを何ら発現しないAd-E3(-)、及びshDicerの代わりにホタルルシフェラーゼに対するshRNA(shLuc)を発現するAd-shLucを用いた。
実施例1で作製したAd-shDicerを各種癌細胞に作用させたときのウイルスゲノム量を確認した。癌細胞は、HeLa細胞(ヒト子宮頸癌細胞)、SK HEP-1細胞(ヒト肝細胞癌細胞)及びH1299細胞(ヒト非小細胞肺癌細胞)について確認した。従来型Oncolytic Adコントロールとして、実施例1で作製したAd-shDicerと同様のゲノム構造であって、shRNAを何ら発現しないAd-E3(-)、及びshDicerの代わりにホタルルシフェラーゼに対するshRNA(shLuc)を発現するAd-shLucを用いた。
各種癌細胞に対して、細胞が70%コンフルエントの状況になった時に、培養液を用いて調製した各種ウイルス懸濁液をMOI 2となるように作用させ、37℃で24時間感染させた。その後ウイルス液を除去し、培養液を加えて37℃で培養した。培養開始後50時間目のAdゲノムコピー数を定量的PCR法により確認した。その結果、上記3種類のいずれの癌細胞においても、Ad-shDicerの場合は、その他コントロールのAdと比較して、顕著なAdゲノム増殖能が認められた(図2参照)。
(実験例1-2)Ad-shDicerを作用させたときの細胞生存率の確認
実施例1で作製したAd-shDicerを各種癌細胞及び正常細胞に作用させたときの細胞生存率を確認した。実験例1-1に示すコントロールについても同様に確認した。さらに、Adを加えないmockの系もコントロールとした。癌細胞は、HeLa細胞(ヒト子宮頸癌細胞)、H1299細胞(ヒト非小細胞肺癌細胞)及びHepG2細胞(ヒト肝細胞癌細胞)について、正常細胞は、HUVEC(ヒト臍帯静脈血管内皮細胞)、NHLF(ヒト肺線維芽細胞)、PrSC(前立腺ストローマ細胞)について確認した。
実施例1で作製したAd-shDicerを各種癌細胞及び正常細胞に作用させたときの細胞生存率を確認した。実験例1-1に示すコントロールについても同様に確認した。さらに、Adを加えないmockの系もコントロールとした。癌細胞は、HeLa細胞(ヒト子宮頸癌細胞)、H1299細胞(ヒト非小細胞肺癌細胞)及びHepG2細胞(ヒト肝細胞癌細胞)について、正常細胞は、HUVEC(ヒト臍帯静脈血管内皮細胞)、NHLF(ヒト肺線維芽細胞)、PrSC(前立腺ストローマ細胞)について確認した。
各種細胞に対して、細胞が70%コンフルエントの状況になった時に、培養液を用いて調製した各種ウイルス懸濁液をMOI 2となるように作用させ、37℃で24時間感染させた。その後ウイルス液を除去し、培養液を加えて37℃で培養した。測定時点においてAd非作用群の細胞生存率を100%としたとき、培養3日目(day3)及び5日目(day5)の各細胞の生存率を確認した。細胞生存率はアラマーブルー(R)アッセイ法(Invitrogen社)により確認した。
その結果、Ad-shDicerを作用させた場合は、コントロール群と比べて癌細胞で顕著な殺細胞効果が認められ(図3参照)、正常細胞では生存率にほとんど差を認めなかった(図4参照)。
(実験例1-3)Ad-shDicerを作用させたときのDicer mRNA量の確認
実施例1で作製したAd-shDicerを各種癌細胞に作用させたときのDicer mRNA量を測定した。癌細胞は、HeLa細胞(ヒト子宮頸癌細胞)、H1299細胞(ヒト非小細胞肺癌細胞)及びSK HEP-1細胞(ヒト肝細胞癌細胞)について確認した。
実施例1で作製したAd-shDicerを各種癌細胞に作用させたときのDicer mRNA量を測定した。癌細胞は、HeLa細胞(ヒト子宮頸癌細胞)、H1299細胞(ヒト非小細胞肺癌細胞)及びSK HEP-1細胞(ヒト肝細胞癌細胞)について確認した。
各種細胞に対して、細胞が70%コンフルエントの状況になった時に、培養液を用いて調製した各種ウイルス懸濁液をMOI 2となるように作用させ、37℃で48時間感染させた。感染後、感染細胞からRNAを回収し、Dicer mRNA量を定量的RT-PCR法にて測定した。
その結果、上記3種類のいずれの癌細胞においても、Ad-shDicerの場合は、その他コントロールAdと比較して、顕著なDicer mRNA量の低下が認められた(図5参照)。

(実験例1-4)Ago2、Dicer又はXPO5をノックダウンしたときの野生型Adの増殖
各種癌細胞に、Ago2、Dicer又はXPO5に対するsiRNAを作製し、各siRNAを各細胞に導入してRNA干渉させた。前記各細胞に野生型Adを加えて培養したときの、Adゲノム量を確認した。背景技術の欄でも示したように、VA-RNAが細胞核内で転写されたのち、XPO5によって核外に輸送され、細胞質内でDicerによる切断を受けた後、Ago2タンパク質とともにRISCを形成する(図6参照)。そこで、各種癌細胞に対して、Ago2、Dicer又はXPO5に対するsiRNAを導入することで、Adの増殖に対してどのように影響するかを確認した。
各種癌細胞に、Ago2、Dicer又はXPO5に対するsiRNAを作製し、各siRNAを各細胞に導入してRNA干渉させた。前記各細胞に野生型Adを加えて培養したときの、Adゲノム量を確認した。背景技術の欄でも示したように、VA-RNAが細胞核内で転写されたのち、XPO5によって核外に輸送され、細胞質内でDicerによる切断を受けた後、Ago2タンパク質とともにRISCを形成する(図6参照)。そこで、各種癌細胞に対して、Ago2、Dicer又はXPO5に対するsiRNAを導入することで、Adの増殖に対してどのように影響するかを確認した。
本実施例で使用した各siRNAの塩基配列は、配列番号1~4、8及び9に示した。

HeLa細胞(ヒト子宮頸癌細胞)又はH1299細胞(ヒト非小細胞肺癌細胞)に、Ago2、Dicer又はXPO5に対するsiRNAを導入した。コントロールは、control siRNA(Allstars negative control siRNA; Qiagen社)を導入した細胞とした。前記各種癌細胞が70%コンフルエントの状況になった時に、野生型Ad溶液をMOI 5となるように、37℃で24時間感染させた。野生型AdはshDicer発現カセットを搭載しておらず、ウイルスの自己増殖に必須のE1遺伝子がウイルス自身のプロモーターで転写される。
Adを感染した各細胞について、Adゲノムコピー数を定量的PCR法により確認した。その結果、上記2種類のいずれの癌細胞においても、細胞にAgo2又はDicerに対するsiRNAを導入した場合は、コントロールに比べて有意にAdゲノムのコピー数が増加した(図7参照)。一方、XPO5に対するsiRNAを作用させた場合には、Adゲノムのコピー数はコントロールと同等又は低い値を示した(図7参照)。
次に、Ago2又はDicerに対するsiRNAを導入した場合のmivaRNA I又はmivaRNA IIの産生に及ぼす影響を電気泳動で確認した。その結果、何れの細胞においても、mivaRNA I及びmivaRNA IIの産生が抑制されていることが確認された(図8参照)。
HeLa細胞又はH1299細胞の各細胞に、Dicer又はAgo2に対するsiRNAを各々導入した。コントロールはControl siRNA(Allstars negative control siRNA)を用いた。siRNA導入48時間後、VA-RNA発現プラスミドを導入した。プラスミド導入24時間後、VA-RNAのコピー数を定量的RT-PCR法により測定した。すなわち、DicerやAgo2をノックダウンすることで、全長のVA-RNAのコピー数が増加すると考えられる(図9参照)。
以上の結果より、VA-RNAが細胞核内で転写されたのち、XP05によって核外に輸送される段階ではなく、Dicer又はAgo2に対するsiRNAを導入すること等によりAd由来mivaRNAの産生を抑制するか、又はmivaRNAの活性を抑制することで、Adの増殖促進効果が得られることが確認された。
(実験例1-5)各種細胞においてDicerをノックダウンしたときの野生型Adの増殖
本実施例では、各癌細胞にshDicerの発現カセットを導入してRNA干渉させた前記各細胞に野生型Adを加えて培養したときのウイルスゲノム量を確認した。shDicer発現カセットにおけるshDicerに対応する配列は、配列番号5に示す塩基配列からなる。

H1299細胞(ヒト非小細胞肺癌細胞)、SK HEP-1細胞(ヒト肝細胞癌細胞)又はHepG2細胞(ヒト肝細胞癌細胞)に、shDicer発現カセットを導入した。コントロールは、shRNAを導入しない細胞とした。前記各種癌細胞が70%コンフルエントの状況になった時に、野生型Ad溶液をMOI 5となるように、37℃で24時間感染させた。
Adを感染した各細胞について、Adゲノムコピー数を定量的PCR法により確認した。その結果、各癌細胞においても、細胞にshDicer発現カセットを導入した場合は、コントロールに比べて有意にAdゲノムのコピー数が増加した(図10参照)。このことから、Ad由来mivaRNAの産生をshDicerで抑制することで、Adの増殖促進効果が得られることが確認された。
(比較例1)Ago2又はDicerを過剰発現させた場合の野生型Adの増殖
癌細胞に、Ago2又はDicerを過剰発現させたときのAdの増殖を確認した。

HeLa細胞(ヒト子宮頸癌細胞)又はH1299細胞(ヒト非小細胞肺癌細胞)に、Ago2又はDicer発現プラスミドを導入した。コントロールは、何も発現しないコントロールプラスミド(p3xFLAG-CMV10)を導入した細胞とした。前記各種癌細胞が70%コンフルエントの状況になった時に、野生型Ad溶液をMOI 5となるように、37℃で24時間感染させた。
Adを感染した各細胞について、Adゲノムコピー数を定量的PCR法により確認した。その結果、上記2種類のいずれの癌細胞においても、細胞にAgo2又はDicerが過剰発現している場合は、コントロールに比べて有意にAdゲノムのコピー数が減少した(図11参照)。
(実施例2)Ad-shDicerの各癌細胞に対する殺細胞効果
実施例1と同手法により作製したAd-shDicerをHeLa細胞(ヒト子宮頸細胞)又はH1299細胞(ヒト非小細胞肺癌細胞)の各癌細胞に作用させたときの細胞生存率を確認した。実験例1-1に示すコントロールについても同様に確認した。各細胞に対して、細胞が70%コンフルエントの状況になった時に、培養液を用いて調製した各種ウイルス懸濁液をMOI 0.01、0.1、1、10、100となるように作用させ、37℃で24時間感染させた。その後ウイルス液を除去し、培養液を加えて37℃で培養した。測定時点においてAd非作用群の細胞生存率を100%としたとき、培養5日目の各細胞の生存率を確認した。細胞生存率はアラマーブルー(R)アッセイ法(Invitrogen社)により確認した。その結果、本発明のAd-shDicerは、コントロールのAd-E3(-)及びAd-shLucに比べて高い殺細胞効果を示した(図12参照)。

(実施例3)Ad-shDicerの各癌細胞に対する抗癌作用(抗腫瘍効果)
実施例1と同手法により作製したAd-shDicerのマウス皮下腫瘍に対する抗腫瘍効果を検討した。実験例1-1に示すコントロールについても同様に確認した。5週齢のヌードマウス(BALB/c nu/nu、雌)皮下にH1299細胞(ヒト非小細胞肺癌細胞)又はSK-OV-3細胞(ヒト卵巣癌細胞)を各マウスについて2×106個移植し、直径5-6mmの固形癌を形成させた。その後、各々n=6のグループについて、形成した固形癌内にAdを107 ifu(Infectious Units)局所投与した。また初回投与後の3日後、再度Adを局所投与した。その後、経日的に腫瘍径を測定した。その結果、本発明のAd-shDicerはコントロールのAd-E3(-)及びAd-shLucに比べて高い抗腫瘍活性を有していることが示された(図13参照)。
実施例1と同手法により作製したAd-shDicerのマウス皮下腫瘍に対する抗腫瘍効果を検討した。実験例1-1に示すコントロールについても同様に確認した。5週齢のヌードマウス(BALB/c nu/nu、雌)皮下にH1299細胞(ヒト非小細胞肺癌細胞)又はSK-OV-3細胞(ヒト卵巣癌細胞)を各マウスについて2×106個移植し、直径5-6mmの固形癌を形成させた。その後、各々n=6のグループについて、形成した固形癌内にAdを107 ifu(Infectious Units)局所投与した。また初回投与後の3日後、再度Adを局所投与した。その後、経日的に腫瘍径を測定した。その結果、本発明のAd-shDicerはコントロールのAd-E3(-)及びAd-shLucに比べて高い抗腫瘍活性を有していることが示された(図13参照)。
(参考例1)Dicerノックダウン細胞におけるmiR-27a,bの発現量
実験例1-4等の結果より、Dicerのノックダウンにより、Adの増殖が促進されることが確認された。このことより、Dicerをノックダウンした場合、いずれかのmiRNAの産生が抑制されることが予想される。そこで、Dicerノックダウンによりある種のmiRNA量が減少し、それによってウイルス増殖を促進する遺伝子の発現が回復して、Adの増殖が促進している可能性が考えられた。
実験例1-4等の結果より、Dicerのノックダウンにより、Adの増殖が促進されることが確認された。このことより、Dicerをノックダウンした場合、いずれかのmiRNAの産生が抑制されることが予想される。そこで、Dicerノックダウンによりある種のmiRNA量が減少し、それによってウイルス増殖を促進する遺伝子の発現が回復して、Adの増殖が促進している可能性が考えられた。
上記知見に基づき、miR-27a及びmiR-27bに着目し、Dicerノックダウン細胞におけるmiR-27a及びmiR-27bの発現量を測定した。ドキシサイクリン(Dox:テトラサイクリン誘導体)依存的にshDicerを発現するカセットをHeLa細胞に挿入した。HeLa細胞をDoxを100ng/mlを含む培地、又は含まない培地で48時間培養した。miR-27a又はmiR-27b量は定量的RT-PCRにより測定した。その結果、Dox存在下でHeLa細胞を培養したときに、miR-27a及びmiR-27bがDicerノックダウンにより発現低下していることが明らかとなった(図14参照)。
(参考例2)VA-RNA、Dicer及びmiR-27a,bの関係について
従来、VA-RNAはDicerの発現を抑制することが報告されている。そこでVA-RNAを過剰発現させることでmiR-27a及びmiR-27bの発現が抑制されるか否かを検討した。VA-RNAI及びII発現プラスミド(pAdvantage)-DNaeI、pAdVantage、VA-RNAI発現プラスミド(pVAI)又はVA-RNAII発現プラスミド(pVAII)をトランスフェクトし、VA-RNA(I及びII)、VA-RNAI又はVA-RNAIIが過剰発現したHeLa細胞を48時間培養し、miR-27a及びmiR-27bの発現量を定量的RT-PCRにより測定した。その結果、VA-RNAの過剰発現によりmiR-27a及びmiR-27bの発現量が各々減少することが示された(図15A参照)。HeLa細胞をVA-RNA発現プラスミド及びRLuc遺伝子の3'-UTRにmiR-27a又はmiR-27bの相補配列を2コピー含むレポータープラスミド (psiCHECK-2-mR-27aT or -miR-27bT)をトランスフェクトし、48時間培養した。その後ルシフェラーゼ活性を測定した。その結果、miR-27a及びmiR-27bの発現量がVA-RNAにより減少することが示された(図15B参照)。
従来、VA-RNAはDicerの発現を抑制することが報告されている。そこでVA-RNAを過剰発現させることでmiR-27a及びmiR-27bの発現が抑制されるか否かを検討した。VA-RNAI及びII発現プラスミド(pAdvantage)-DNaeI、pAdVantage、VA-RNAI発現プラスミド(pVAI)又はVA-RNAII発現プラスミド(pVAII)をトランスフェクトし、VA-RNA(I及びII)、VA-RNAI又はVA-RNAIIが過剰発現したHeLa細胞を48時間培養し、miR-27a及びmiR-27bの発現量を定量的RT-PCRにより測定した。その結果、VA-RNAの過剰発現によりmiR-27a及びmiR-27bの発現量が各々減少することが示された(図15A参照)。HeLa細胞をVA-RNA発現プラスミド及びRLuc遺伝子の3'-UTRにmiR-27a又はmiR-27bの相補配列を2コピー含むレポータープラスミド (psiCHECK-2-mR-27aT or -miR-27bT)をトランスフェクトし、48時間培養した。その後ルシフェラーゼ活性を測定した。その結果、miR-27a及びmiR-27bの発現量がVA-RNAにより減少することが示された(図15B参照)。
(参考例3)Adの感染及びmiR-27a,bとの関係について
参考例2の結果、VA-RNAによりmiR-27a及びmiR-27bの発現量が減少することが確認されたので、Adの感染及びmiR-27a及びmiR-27bの発現量との関係を確認した。野生型Ad(WT-Ad)をHeLa細胞に100VP(vector particles)/cell加え、各時間感染させたところ、感染後24時間目でmiR-27a及びmiR-27bの発現量が有意に上昇することが確認された(図16参照)。
参考例2の結果、VA-RNAによりmiR-27a及びmiR-27bの発現量が減少することが確認されたので、Adの感染及びmiR-27a及びmiR-27bの発現量との関係を確認した。野生型Ad(WT-Ad)をHeLa細胞に100VP(vector particles)/cell加え、各時間感染させたところ、感染後24時間目でmiR-27a及びmiR-27bの発現量が有意に上昇することが確認された(図16参照)。
(参考例4)miR-27a,bとAdの感染との関係について
次にmiR-27a及びmiR-27bがAdの増殖に及ぼす影響について検討した。HeLa細胞にmimic miR-27a又はmimic miR-27b(合成されたmiR-27a又はmiR-27b)、あるいはmiR-27a又はmiR-27bの阻害剤(miR-27a又はmiR-27bに対するアンチセンスオリゴヌクレオチド:miRVana miRNA inhibitor:Ambion社)を1, 3, 10又は30 nMトランスフェクトし、次に野生型Ad(WT-Ad)を100VP/cell加え、24時間感染させた。Adゲノム量を定量的RT-PCRにより測定した。その結果、mimic miR-27a又はmimic miR-27bを含む系でAdの増殖が抑制され、阻害剤を含む系ではAdの増殖促進が認められた(図17A,B)。H1299細胞についてもmimic miR-27a又はmiR-27bを30 nM、あるいはmiR-27a又はmiR-27bの阻害剤(inhibitor)を30 nM加えたところ、mimic miR-27a又はmimic miR-27bを加えた系ではAdの増殖が抑制されたが、阻害剤(inhibitor)を含む系ではAdの増殖促進が認められた(図17C)。
次にmiR-27a及びmiR-27bがAdの増殖に及ぼす影響について検討した。HeLa細胞にmimic miR-27a又はmimic miR-27b(合成されたmiR-27a又はmiR-27b)、あるいはmiR-27a又はmiR-27bの阻害剤(miR-27a又はmiR-27bに対するアンチセンスオリゴヌクレオチド:miRVana miRNA inhibitor:Ambion社)を1, 3, 10又は30 nMトランスフェクトし、次に野生型Ad(WT-Ad)を100VP/cell加え、24時間感染させた。Adゲノム量を定量的RT-PCRにより測定した。その結果、mimic miR-27a又はmimic miR-27bを含む系でAdの増殖が抑制され、阻害剤を含む系ではAdの増殖促進が認められた(図17A,B)。H1299細胞についてもmimic miR-27a又はmiR-27bを30 nM、あるいはmiR-27a又はmiR-27bの阻害剤(inhibitor)を30 nM加えたところ、mimic miR-27a又はmimic miR-27bを加えた系ではAdの増殖が抑制されたが、阻害剤(inhibitor)を含む系ではAdの増殖促進が認められた(図17C)。
(実施例4)miR-27a,b を阻害するタフデコイRNA 発現カセットを搭載したAdの作製
本実施例では、miR-27a又はmiR-27bに対するタフデコイRNA(以下、実施例、実験例において「TuD-27a又はTuD-27b」ということとする。)を搭載したカセットであって、miR-27a又はmiR-27bに対するsiRNAを発現しうるカセットを搭載した腫瘍溶解性(Oncolytic)Adの作製について説明する。以降、本実施例のAdを「Oncolytic Ad-TuD-27a」又は「Oncolytic Ad-TuD-27b」ということとする。作製したOncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bのゲノム構造は図18に示すとおりである。
本実施例では、miR-27a又はmiR-27bに対するタフデコイRNA(以下、実施例、実験例において「TuD-27a又はTuD-27b」ということとする。)を搭載したカセットであって、miR-27a又はmiR-27bに対するsiRNAを発現しうるカセットを搭載した腫瘍溶解性(Oncolytic)Adの作製について説明する。以降、本実施例のAdを「Oncolytic Ad-TuD-27a」又は「Oncolytic Ad-TuD-27b」ということとする。作製したOncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bのゲノム構造は図18に示すとおりである。
1)Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bゲノムの作製
本実施例では、実施例1と同様にヒト5型Adゲノム(GenBank Accession番号:M73260, M29978)において、当該ゲノムのうちE1領域(342~3523)並びにE3領域(28133~30818)を欠損しており、E1欠損領域に外来遺伝子を挿入できるベクタープラスミドを元に構築した。当該ベクタープラスミドpAdHM19のE1欠損領域に、腫瘍特異的プロモーターであるhTERTプロモーターと、E1A、IRES、E1B、ポリA(pA)シグナル配列をライゲーションして搭載し、Oncolytic Adゲノムを得た。
本実施例では、実施例1と同様にヒト5型Adゲノム(GenBank Accession番号:M73260, M29978)において、当該ゲノムのうちE1領域(342~3523)並びにE3領域(28133~30818)を欠損しており、E1欠損領域に外来遺伝子を挿入できるベクタープラスミドを元に構築した。当該ベクタープラスミドpAdHM19のE1欠損領域に、腫瘍特異的プロモーターであるhTERTプロモーターと、E1A、IRES、E1B、ポリA(pA)シグナル配列をライゲーションして搭載し、Oncolytic Adゲノムを得た。
さらに、E3欠損領域に、TuD-27a又はTuD-27bの発現カセットを搭載した。本実施例において使用したTuD-27a又はTuD-27bに対応する配列は、配列番号12又は13に示す塩基配列からなる。下線部は、miR-27a又はmiR-27bに対するアンチセンス配列である。

2)Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bの作製
上記1)で作製したTuD-27a又はTuD-27bゲノムを、ウイルスゲノム末端に存在する制限酵素PacIで切断して洗浄し、トランスフェクション試薬Lipofectamine2000TM(Invitrogen社)を用いてHEK293細胞へトランスフェクションし、Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bを作製した。
上記1)で作製したTuD-27a又はTuD-27bゲノムを、ウイルスゲノム末端に存在する制限酵素PacIで切断して洗浄し、トランスフェクション試薬Lipofectamine2000TM(Invitrogen社)を用いてHEK293細胞へトランスフェクションし、Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bを作製した。
(実験例4-1)Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bを作用させたときの細胞生存率の確認
実施例4で作製したOncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bをHeLa細胞又はH1299細胞に作用させたときの細胞生存率を確認した。タフデコイRNAを何ら発現しないAd-E3(-)に示すコントロール(従来型Oncolytic Ad)についても同様に確認した。さらに、Adを加えないmockの系もコントロールとした。
実施例4で作製したOncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bをHeLa細胞又はH1299細胞に作用させたときの細胞生存率を確認した。タフデコイRNAを何ら発現しないAd-E3(-)に示すコントロール(従来型Oncolytic Ad)についても同様に確認した。さらに、Adを加えないmockの系もコントロールとした。
各細胞に対して、細胞が70%コンフルエントの状況になった時に、培養液を用いて調製した各種ウイルス懸濁液をMOI 2となるように作用させ、37℃で24時間感染させた。その後ウイルス液を除去し、培養液を加えて37℃で培養した。測定時点においてAd非作用群の細胞生存率を100%としたとき、培養3日目(day3)及び5日目(day5)の各細胞の生存率を確認した。細胞生存率はアラマーブルー(R)アッセイ法(Invitrogen社)により確認した。その結果、Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bを作用させた場合、コントロール群よりも顕著な殺細胞効果が認められた(図19参照)。
(実験例4-2)Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bによる癌細胞内でのAdゲノム増殖能の確認
実施例4で作製したOncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bをHeLa細胞又はH1299細胞に作用させたときのウイルスゲノム量を確認した。実験例4-1と同様にコントロール(従来型Oncolytic Ad)についても同様に確認した。

各細胞に対して、細胞が70%コンフルエントの状況になった時に、培養液を用いて調製した各種ウイルス懸濁液をMOI 2となるように作用させ、37℃で24時間感染させた。その後ウイルス液を除去し、培養液を加えて37℃で培養した。培養開始後48時間目のAdゲノムコピー数を定量的PCR法により確認した。その結果、Oncolytic Ad-TuD-27a又はOncolytic Ad-TuD-27bを作用させた場合、コントロール群よりも癌細胞内での高い増殖能が認められた(図20参照)。
以上詳述したように、本発明のAdは、従来のAdに比べて効果的にAdを増殖促進させることができる。本発明のAdを有効成分として含む抗腫瘍剤によれば、効果的に標的となる腫瘍細胞内で増殖が促進され、抗腫瘍効果が発揮される。これにより、より効果的な抗腫瘍薬を提供することができ、有用である。
本発明の抗腫瘍剤において有効成分として含まれるAdは、所望の標的細胞において増殖促進能を有するので、通常の制限増殖Adよりも低い濃度で使用することができ、経済的にも有用である。
さらに、本発明の増殖促進方法によれば、従来の方法に比べて、効果的にAdを増殖促進させることができる。標的腫瘍細胞等、所望の細胞で効果的にAdを増殖することができれば、効果的に腫瘍溶解等の抗悪性効果を発揮しうる。そして、本発明の増殖促進方法を、細胞におけるAdの産生系に適用することで、より効果的にAdを産生することができ、有用である。
Claims (15)
- アデノウイルスの増殖を促進する方法であって、アデノウイルス由来mivaRNAの機能を抑制することを含む、アデノウイルスの増殖促進方法。
- アデノウイルスの増殖を促進する方法であって、miR-27a及び/又はmiR-27bを阻害することによるアデノウイルスの増殖促進方法。
- mivaRNAの機能を抑制、あるいはmiR-27a及び/又はmiR-27bを阻害することが、少なくともDicer又はAgo2のいずれか一方を阻害することによる、請求項1又は2に記載のアデノウイルスの増殖促進方法。
- Dicer又はAgo2の阻害が、RNA干渉によるDicer又はAgo2の阻害である、請求項3に記載のアデノウイルスの増殖促進方法。
- RNA干渉によるDicer又はAgo2の阻害が、Dicer又はAgo2に対するsiRNAによる阻害であって、Dicer又はAgo2に対するsiRNAが、当該アデノウイルスに搭載されたDicer又はAgo2に対するsiRNA、アデノウイルス産生用パッケージング細胞に導入されたDicer又はAgo2に対するsiRNA、あるいはDicer又はAgo2に対するsiRNAである、請求項4に記載のアデノウイルスの増殖促進方法。
- Dicer又はAgo2の阻害が、アデノウイルス産生用パッケージング細胞におけるDicer遺伝子又はAgo2遺伝子のノックアウトによる阻害である、請求項3に記載のアデノウイルスの増殖促進方法。
- Dicer又はAgo2の阻害が、Dicer又はAgo2の抗体による阻害、あるいはDicer又はAgo2の阻害剤による阻害である、請求項3のアデノウイルスの増殖促進方法。
- miR-27a又はmiR-27bの阻害が、miR-27a又はmiR-27bに対するデコイ核酸、あるいはmiR-27a又はmiR-27bに対する阻害剤による阻害である、請求項2に記載のアデノウイルスの増殖促進方法。
- アデノウイルスゲノム において、Dicer又はAgo2に対するsiRNAの発現カセット、あるいはmiR-27a又はmiR-27bに対するデコイ核酸の発現カセットが搭載されていることを特徴とするアデノウイルス。
- Dicerに対するsiRNAが、以下の1)又は2)に示すオリゴヌクレオチドを含むRNAであり、Ago2に対するsiRNAが、以下の3)又は4)に示すオリゴヌクレオチドを含むRNAであり、miR-27aに対するデコイ核酸が、以下の5)に示すオリゴヌクレオチドを含むRNAであり、miR-27bに対するデコイ核酸が、以下の6)に示すオリゴヌクレオチドを含むRNAである、請求項9に記載のアデノウイルス;

- アデノウイルス産生用パッケージング細胞におけるゲノムにおいて、少なくともDicer又はAgo2に対するsiRNAの発現カセット、あるいはmiR-27a又はmiR-27bのデコイ核酸の発現カセットが導入されていることを特徴とする、アデノウイルスを作製するためのパッケージング細胞。
- Dicerに対するsiRNAが、以下の1)又は2)に示すオリゴヌクレオチドを含むRNAであり、Ago2に対するsiRNAが、以下の3)又は4)に示すオリゴヌクレオチドを含むRNAであり、miR-27aに対するデコイ核酸が、以下の5)に示すオリゴヌクレオチドを含むRNAであり、miR-27bに対するデコイ核酸が、以下の6)に示すオリゴヌクレオチドを含むRNAである、請求項13に記載のパッケージング細胞:
