본 발명은 화학발광법을 이용하여 수중에 존재하는 크롬의 농도를 연속적으로 모니터링하기 위한 미세유체칩 및 이를 포함하는 크름 검출 장치에 관한 것이다.
【배경기술】
크롬은 합금, 안료, 피혁, 직물공업, 촉매, 목재 방부제 등에 주로 사용되는 물질로 화학적으로 안정한 성질 때문에 금속의 부식을 막기 위한 도금 재료로도 많이 사용된다. 상기한 공업활동이 증가함에 따라 크롬의 방출로 인해 지표수 및 지하수가 오염된다.
자연상태의 크름 이온은 다양한 산화가로 존재하지만 수중에는 가장 안정한 크름 [m]와 크롬 [IV]만이 존재한다. 크롬 [m]는 포도당, 콜레스테를, 지방 대사에 관여하는 필수영양소로 부족할 경우 체중이 감소하고, 혈액에서 포도당을 제거하는 기능에 손상을 준다. 그리고 장시간 피부에 노출되었올 경우 알레르기 또는 암을 유발할 수 있다. 그러나 물에 대한 용해도와 생체막에 대한 투과성이 낮아 독성은높지 않은 것으로 알려져 있다. 이와는 달리 크롬 [IV]는 물에 대한 용해도와 이동성이 높고, 산화력과 생체막에 대한 투과성이 높아 신장, 간, 폐 등의 여러 장기에 좋지 않은 영향을 주며, 피부 또는 점막에 염증을 일으키는 것으로 알려져 있다.
상기한 바와 같이 크름 [m]와 크름 [IV]의 서로 다른 특성과 독성 때문에 이들 각각의 농도를 정확히 분석하는 것이 매우 중요하다. 특히 크름 [m]와 크롬 [IV]는 환경에 따라 산화 -환원 반웅을 통해 서로 쉽게 바뀌므로 수질의 지속적인 모니터링이 필요하다. 우리나라의 경우 수질오염물질 배출허용기준에 따르면 청정지역 내에 허용되는 총크롬의 농도를 0.5 ppm, 그 외의 지역은 2ppm 이하로 규제하고 있으며, 크롬 [IV]의 농도는 청정지역에서 0.1 ppm, 그 외의 지역은 0.5 ppm 이하로 제한하고 있다. 그리고 미국환경보호국 (Environmental Protection Agency, EPA)에서는 크름 [IV]를 발암물질로 규정하고 있으며 , 음용수
내에 허용되는 총크름의 농도를 ippm이하로 제한하고 있다.
수중에 크름을 분석하기 위한 종래기술로서 화학발광반웅을 이용한 크롬분석은 아래의 [식 1]과 같이 염기성 조건에서 크름 [m] 촉매 하에
과산화수소에 의해 산화된 루미놀 (luminol,
5-amino-2,3-dihydro-l,4-phthalazinedione)에서 방출되는 빛 (425 run)의 세기를 측정하는 것이다. 이때 방출되는 빛의 세기는 크롬 [m]의 농도에 비례하기 때문에 빛의 세기를 측정함으로써 크름 [m]를 정량분석할 수 있다.

화학발광법으로 크롬 [m]와 크롬 [IV]를 분석하는 방법은 다음과 같다.
먼저시료를 환원제와 반응하여 시료 내에 크롬 [IV]를 크름 [m]로 환원시켜 총크톰의 농도를 측정한다. 그리고 환원제를 사용하지 않고 크롬 [in]의 농도를 측정한 다음, 총크롬과 크름 [m]의 농도 차를 계산하여 크롬 [IV]의 농도를 알 수 있다. 종래기술은 상기한 방법을 적용하여 수중에 크름 분석을 위한 미세유체칩을 개발하였으나, 다음과 같은 문제점들을 가지고 있다ᅳ
첫째, 염기성 조건에서 과산화수소에 의한 루미놀 산화반웅은 반웅속도가 매우 빠르므로 최대화학발광을 효율적으로 측정하기 위해서는 루미놀과
과산화수소가 흔입되는 화학발광시약 흔합채널의 길이를 최소화해야 한다. 그리고 크롬 [IV]의 농도를 정확하게 분석하기 위해서는 시료 내에 크롬 [IV]를 크롬 [m]로 100% 환원시켜야 함에 따라 층분한 반웅시간이 요구된다. 따라서 환원채널의 길이를 화학발광시약 흔합채널의 길이보다 상대적으로 길게 디자인해야 한다. 그러나 종래기술은 화학발광시약 흔합채널의 길이와 크롬 [IV]의 크름 [m]로의 환원을 위한 환원채널의 길이를 동일하게 하여 크룸 [m]와 크름 [IV]의 농도를 정확하게 검출할 수 없고, 검출감도가 떨어진다.
둘째, 루미놀올 이용한 화학발광반응은 크롬 [m]뿐만 아니라 다른 금속이온들 (철 [π], 코발트 [π], 구리 [π], 니켈 [π] 등)에 의해서도 일어난다.
종래기술은 이러한 방해물질들에 대한 효과를 고려하지 않아, 시료 내에 크름의 농도를 선택적으로 분석할 수 없다.
셋째, 다른 금속이온들이 존재하는 현장시료 내에 크롬 [m] 분석 시 주입한 증류수와 흔합하는 과정에서 시료의 pH 변화에 의해 크롬 [m]와 금속이은의 흔합결정이 생성되어 크름 [m]도 함께 침전되는 공침현상이 발생할 수 있다. 공침현상이 발생하면 미세유체칩으로 주입된 시료 내에 크롬 [m]의 농도가 감소하여 분석오차를 유발하며, 상기 침전으로 인해 미세유체칩이 막히는 결과를 초래한다.
넷째, 크름 [m]와 크름 [IV]를 분석하기 위해 2개의 검량곡선 (총크름과 크름 [m]의 검량곡선)이 필요하다.
다섯째, 미세유체칩을 투명한 유리기판으로 제작하여 주변의 빛이 검출기로 유입됨에 따라 검출감도가 떨어지고 장치의 운용에 어려움을 야기한다. 【발명의 상세한 설명】
【기술적 과제】
본 발명은 화학발광법으로 수중에 크롬을 연속적으로 모니터링하기 위해 연속흐름상에서 분리과정 없이 크름 [m]와크롬 [IV]를 고효율, 고감도로 검출하고, 방해물질에 의한 영향 없이 크롬을 선택적으로 검출할 수 있는 미세유체칩 및 이를 갖는 크롬 검출 장치를 제공하고자 한다.
【기술적 해결방법】
본 발명의 일 측면에 따른 미세유체칩은 시료와 시약을 흔합하는 환원채널과 시약들을 흔합하는 시약 흔합채널, 및 일측 단부에 상기 환원채널과 상기 시약 흔합채널이 병렬로 연결되고 타측 단부에 배출구가 연결된 검출채널이 형성된 제 1 기판, 및 상기 검출채널과 마주하며 빛이 투과하도록 형성된 검출부를 구비하며 상기 제 1기판에 결합되는 게 2기판을 포함한다.
상기 환원채널은 상기 시약 흔합채널보다 더 길게 형성될 수 있으며, 상기 거 U 기판과 제 2 기판은 빛을 흡수할 수 있는 색을 갖는 물질로 이루어지거나, 빛을 흡수할 수 있는 색으로 착색될 수 있다.

본 발명의 일 측면에 따른 크름 검출장치는 시료와 시약을 흔합하는 환원채널과 시약들을 흔합하는 시약 흔합채널, 및 일측 단부에 상기 환원채널과 상기 시약 흔합채널이 병렬로 연결되고 타측 단부에 배출구가 연결된 검출채널이 형성된 제 1 기판, 및 상기 검출채널과 마주하며 빛이 투과하도록 형성된 검출부를 구비하며 상기 제 1 기판에 결합되는 제 2 기판을 포함하는 미세유체칩과, 상기 검출부와 마주하도록 배치된 검출기, 및 상기 미세유체칩에 시료 및 시약을 공급하는 공급부를 포함한다.
상기 환원채널은 상기 시약 흔합채널보다 더 길게 형성될 수 있으며, 상기 제 1 기판과 제 2 기판은 빛을 흡수할 수 있는 색을 갖는 물질로 이루어지거나, 빛을 흡수할 수 있는 색으로 착색될 수 있다.
상기 제 1 기판에는 시료의 주입을 위한 시료 주입구와 환원제의 주입을 위한 환원제 주입구, 제 1 시약의 주입을 위한 제 1 시약 주입구, 및 제 2 시약의 주입을 위한 제 2 시약 주입구가 형성될 수 있다.
상기 공급부는 상기 시료 주입구로 시료를 공급하고, 상기 환원제 주입구로 아황산칼륨을 공급하며, 상기 제 1 시약 주입구로 염기성 조건의 완층용액에 용해된 루미놀을 공급하고, 상기 제 2 시약 주입구로 염기성 조건의 완충용액에 용해된 과산화수소를 공급할 수 있다.
상기 공급부는 상기 제 1 시약 및 상기 제 2 시약과 함께 브롬이온을 상기 미세유체칩으로 공급할 수 있으며, 상기 공급부는 상기 제 1 시약 및 상기 제 2 시약과 함께 에틸렌디아민테트라아세트산 (EDTA; ethylenediaminetetraacetic acid)을
상기 미세유체칩으로 공급할 수 있다.
상기 검출채널은 중첩 배치되며 일측 단부와 타축 단부가 교호적으로 연결되고, 중앙에서 양쪽 측단으로 갈수록 감소하도록 형성될 수 있다.
【유리한 효과】
본 발명에서는 미세유체칩 내에서 시료 내의 크름 [IV]가 크롬 [m]로 모두 환원될 수 있도록 환원채널을 제작하여 크롬 [m]와 크롬 [IV]의 정확한 분석이 가능하다.
또한, 화학발광시약이 주입되는 시약 흔합채널과 시료가 주입되는 환원채널의 길이를 다르게 하여 비대칭적으로 디자인함으로써 시약 흔합채널에서 일어나는 화학발광의 손실을 최소화하여 낮은 검출한계와 높은 감도로 크롬을 검출하는 것이가능하다.
또한, 검출채널에서 화학발광시약에 포함된 킬레이트제와 시료가 만나 시료 내에 존재하는 방해효과를 줄 수 있는 금속이온들을 제거함으로써 연속흐름상에서 크름의 선택적인 검출이 가능하다.
또한, 크롬 [in]분석 시 환원제 주입구로 시료를 주입하여 크롬 [in]의 침전 또는 방해금속과의 공침현상이 발생하지 않아 정확한 크름 농도의 분석이 가능하다.
또한, 크롬 [m]의 검량곡선의 기울기가총크롬의 검량곡선의 기울기의 2배임에 따라 총크롬의 검량곡선만으로 크롬 [m]와 크롬 [IV]의 농도를 분석하는 것이 가능하여 분석시간을 줄일 수 있다.
본 실시예의 미세유체칩은 주변의 빛이 검출기로 들어가는 것올 방지하기 위해 빛을 흡수하는 재료로 제작하여 높은 감도의 안정한 화학발광신호를 얻을 수 있고, 현장에 적용이 용이하다.

【발명의 실시를 위한 형태】
이하, 첨부한 도면을 참고로 하여 본 발명의 실시예에 대하여 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 상세히 설명한다. 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
도 1은 본 발명의 일 실시예에 따른 미세유체칩의 상판을 도시한 평면도이고, 도 2는 본 발명의 일 실시예에 따른 미세유체칩의 하판을 도시한 평면도이다.
본 실시예에 따른 미세유체칩 (30)은 상판 (제 1 기판 )(10)과 하판 (제 2 기판) (20)을 포함한다. 상판 (10)은 사각판 형상으로 이루어지며, 상판 (10)에는 시료 주입구 (11), 환원제 주입구 (12)와 제 1 시약 주입구 (13)과 게 2 시약 주입구 (14)가 형성되어 있다. 또한, 시료 주입구 (11)에는 시료 주입관 (52)과의 결합을 위한 체결부 (31)가 형성되며, 환원제 주입구 (12)에는 환원제 주입관 (53)과의 결합을 위한 체결부 (32)가 형성된다.
제 1 시약 주입구 (13)에는 시약 주입관 (54)과의 결합을 위한 체결부 (34)가 형성되어 있으며, 제 2 시약 주입구 (14)에는 시약 주입관 (55)과의 결합을 위한 체결부 (35)가 형성되어 있다.
또한, 상판 (10)에는 시료채널 (lla), 환원제채널 (12a), 제 1시약채널 (13a), 제 2시약채널 (14a) 환원채널 (15), 시약 흔합채널 (16) 및 검출채널 (17)이 형성된다. 시료 주입구 (Π)에는 시료채널 (lla)이 연결 형성되며, 환원제 주입구 (12)에는 환원제채널 (12a)이 연결 형성된다. 시료 주입구 (11)를 통해서 크롬을 포함하는 시료가 주입되며, 환원제 주입구 (12)를 통해서 크롬 [IV]를 환원시키기 위한 아황산칼륨 등의 환원제가주입된다.
시료채널 (lla)과 환원제채널 (12a)에는 환원채널 (15)이 연결 형성되며, 환원채널 (15)에서 시료와 환원제가 흔합되고 시료 내에 포함된 크롬 [IV]는 환원제에 의해 크롬 [ΙΠ]로 환원된다. 환원채널 (15)은 중첩 배치되며 일측 단부와 타측 단부가 교호적으로 연口결된다. 환원채널 (15)의 일측 단부는 시료채널 (lla) 및 환원제채널 (12a)과 연결되고, 타측 단부는 검출채널 (17)과 연결된다.
거 11 시약 주입구 (13)에는 제 1 시약채널 (13a)이 연결 형성되며, 제 2 시약 주입구 (14)에는 제 2 시약채널 (14a)이 연결 형성된다. 제 1 시약 주입구 (13)에는 염기성 조건의 완층용액에 용해된 투미놀, 브롬 이온 그리고 EDTA가 주입될 수 있으며, 거] 2 시약 주입구 (14)에 상기한 루미놀의 용해에 사용된 염기성 조건의 완층용액에 용해된 과산화수소, 브롬 이온 그리고 EDTA가 주입될 수 있다.
제 1 시약채널 (13a) 및 제 2 시약채널 (14a)에는 시약 흔합채널 (16)이 연결 형성되며, 시약 흔합채널 (16)에서 루미놀과 과산화수소가 흔합된다. 시약 혼합채널 (16)은 중첩 배치되며 일측 단부와 타측 단부가 교호적으로 연결된다. 시약 흔합채널 (16)의 일측 단부는 제 1 시약채널 (13a) 및 제 2 시약채널 (14a)과 연결되고, 타측 단부는 검출채널 (17)과 연결된다.
검출채널 (17)의 일측 단부에는 환원채널 (15)과 시약 흔합채널 (16)이 연결 형성되고, 타측 단부에는 반웅에 사용된 시약과 시료를 배출할 수 있도록 배출구 (18)가 형성되어 있다ᅳ 배출구 (18)에는 배출관이 결합될 수 있도록 체결부 (33)가 형성되어 있다.
검출채널 (17)은 중첩 배치되며 일측 단부와타측 단부가 교호적으로 연결된다. 중첩된 검출채널 (17)의 길이는 중앙에서 양쪽 측단으로 갈수톡 감소하며, 검출채널 (17)의 중심은 원형상으로 이루어진 검출부 (21)의 중심과 맞도록 배치된다.
검출채널 (17)의 일측 단부에 환원채널 (15)과 시약 흔합채널 (16)이 함께 연결되므로 검출채널에서 만난 시료와 시약이 흔합되어 시료 내의 방해효과가 제거됨과 동시에 화학발광반웅이 일어난다.
환원채널 (15)의 길이는 시약 흔합채널 (16) 길이의 2배 내지 6배가 된다. 이에 따라 환원채널 (15)을 통과하면서 환원제에 의해 크름 [IV]가크름 [m] 로 모두 환원될 수 있다. 또한, 시약 흔합채널 (16)의 길이가 짧게 형성되므로 시약 혼합채널 (16)에서 일어나는 화학발광의 손실을 최소화하여 낮은 검출한계와 높은 감도로 크름을 검출할 수 있다.
체결부들 (31, 32, 33, 34, 35)은 폴리머 또는 튜빙 등으로 이루어지며, 관 또는 튜브가 용이하게 착탈 결합될 수 있도록 홀을 갖는다.
상판 (10)은 광 흡수성을 갖는 폴리머로 이루어지며, 특히 검정색의 폴리디메틸실록산 (PDMS,polydimethylsiloxane)으로 이루어질 수 있다. 다만 상판 (10)은 광 흡수성을 갖는 다양한 색을 갖는 물질로 이루어지거나, 광 흡수성을 갖는 색으로 착색될 수 있다.
한편, 하판 (20)에는 상판 (10)의 검출채널 (17)과 대웅되는 하부에
검출부 (21)가 형성된다. 하판 (20)은 검정색의 PDMS로 이루어지며, 검출부 (21)는 화학발광반웅에 의해 발생한 빛이 통과할 수 있도록 투명한 PDMS로 채워진다. 검출부 (21)는 검정색 PDMS에 홀을 형성한 후, 투명 PDMS를 주입하여 형성할 수 있다. 다만, 하판 (20)이 착색된 경우에는 검출부만 착색되지 아니하고 투명하게 형성된다.
상기한 바와 같이 상판 (10)과 하판 (20)이 광 흡수성을 갖도톡 형성되면, 외부에서 입사된 빛을 흡수하여 안정한 화학발광신호를 얻을 수 있고, 검출한계와
감도를 향상시킬 수 있다.
이하에서는 상판 (10)과 하판 (20)의 제조 방법에 대하여 설명한다. 준비된 실리콘 웨이퍼 상에 포토레지스트를 스핀 코팅한 후, 포토 마스크를 설치한 상태에서 자외선을 조사한 다음 경화 및 현상을 거쳐 양각 패턴을 형성한다. 양각 패턴이 형성된 실리콘 웨이퍼 상에 검정색 PDMS를 웨이퍼 위에 붓고 경화 후에 상판 (10)을 웨이퍼에서 분리시키면 음각이 형성된 검정색
PDMS의 상판을 얻을 수 있다. 하판 (20)도 상판 (10)과 동일한 방법으로 제조한다. 상판 (10)과 하판 (20)은 결합되는 부분을 아크 방전으로 표면처리 한 후, 상판 (10)의 검출채널 (17)의 중심에 하판 (20)의 검출부 (21)의 증심이 일치하도록 결합한다.
시료와 환원제는 환원채널 (15)을 거쳐 검출채널 (17)로 이동하며, 시약은 시약 흔합채널 (16)을 거쳐 검출채널 (17)로 이동하여 화학발광반웅을 일으킨다. 검출채널 (17)에서 화학발광반웅이 일어난 후의 시료와 시약은 배출구 (18)로 이동한다.
도 3은 본 발명의 일 실시예에.따른 크롬 검출장치를 도시한 구성도이다. 도 3을 참조하여 설명하면, 본 실시예에 따른 크롬 검출장치는 외형을 이루는 하우징 (41) 내에 삽입 설치되며 검출부 (21)의 아래에 배치된 검출기 (42)와 미세유체칩 (30), 및 미세유체칩 (30)에 시료 및 시약을 공급하는 공급부 (51)를 포함한다. 하우징 (41)은 대략 직육면체의 상자 형태로 이루어진다. 하우징 (41)은 빛을 흡수할 수 있도록 검정색으로 착색된 금속으로 이루어지며,
미세유체칩 (30)은 하우징 (41)의 상면에 고정 설치된다. 이에 따라 외부의 빛이 검출기 (42)로 들어가는 것을 차단하여 화학발광반웅에 의해 생성된 빛만 검출부 (21)를 통해 검출기 (42)로 입사된다.
한편, 시료 주입구 (11)에 설치된 체결부 (31)에는 시료주입관 (52)이 연결 설치되고, 환원제 주입구 (12)에 설치된 체결부 (32)에는 환원제 주입관 (53)이 연결 설치된다. 또한, 제 1 시약 주입구 (13)에 설치된 체결부 (34)에는 계 1 시약
주입관 (54)이 연결 설치되고, 제 1 시약 주입구 (14)에 설치된 체결부 (35)에는 제 2 시약 주입관 (55)이 연결 설치된다.
공급부 (51)에는 연동 펌프가 설치되며 공급부 (51)는 시료주입관 (52)으로 시료를 공급하고, 환원제 주입관 (53)으로 아황산칼륨을 공급한다. 또한,
공급부 (51)는제 1 시약 주입관 (54)으로 루미놀을 공급하고, 제 2 시약 주입관 (55)으로 과산화수소를 공급한다.
공급부 (51)는 시약과 시료를 연속적으로 공급하며, 공급된 시료와 시약은 반응한 후, 배출구 (18)을 통해서 배출된다. 이에 따라 본 실시예에 따른 크롬 검출 장치는 크롬을 연속적으로 모니터링할 수 있다.
공급부 (51)는 시약과 함께 브름이온을 미세유체칩 (30)으로 공급하며, 이에 따라 화학발광세기가 증가하여 크롬 검출장치의 감도가 향상되고, 크름
검출장치는더 낮은 검출한계를 갖게 된다. 또한, 공급부 (51)는 시약과 함께
EDTA를 공급한다 .EDTA는 염기성 조건에서 금속이은과 반응하여 착물을 형성하는 바, 이에 따라 시료에 포함된 루미놀 화학발광반웅에 참여하는 다른 금속이온들을 제거하여 크름의 농도가 부정확하게 측정되는 것을 방지할 수 있다. 검출기 (42)에는 전원 (61)이 연결 설치된다. 또한, 검출기 (42)에는
검출기 (42)에서 발생된 신호를 가공 처리하는 디지털 멀티미터 (62)가 연결 설치되고, 디지털 멀티미터 (62)는 가공된 신호를 연결된 컴퓨터 (63)로 전달한다.
[실험예 1]
실험예 1에서는 본 실시예에 따른 미세유체칩과 이를 포함하는 크롬 검출장치를 이용하여 크롬의 농도에 따른 화학발광세기를 측정하였다.
화학발광시약은 시료에서 크롬 [m]를 검출하기 위한 것으로 염기성 조건 하에서 크롬 [ΙΠ]와 반웅하여 화학발광을 나타낸다. 화학발광시약은 루미놀과
과산화수소로 이투어지며, 화학발광의 세기는 루미놀과 과산화수소의 농도에 따라 달라진다.
화학발광법을 위한 완충용액은 30.09 g의 붕산 (H3B03)과 10.2 g의 브롬화 칼륨 (KBr), 2.92 g의 EDTA((HOOCCH2)2HNCH2CH2NH(CH2COOH)2)를 중류수에 녹인 다음 5M의 수산화칼륨 (KOH) 용액을 이용하여 pH를 10.9로 조절하고 최종부피를 1L로 하여 제조한다. 화학발광시약은 0.02 g의 루미놀을 완충용액에 녹여 pH가 10.9가 되도록 조절한 뒤 최종부피가 100 mL가 되도록 제조한다. 산화제로 0.868 mL의 과산화수소를 상기 완층용액에 녹여 pH가 10.9가 되도록 조절한 뒤 최종부피가 100 mL가 되도록 제조한다. 크롬 [IV]를 크톰 [ΙΠ]로 환원하기 위한 환원제는 아황산칼륨 (K2SO3)0.1104g을 증류수에 녹여 pH가 2.5가 되도록 조절한 뒤 최종부피가 100 mL가 되도록 제조한다. 크름 [ΙΠ]와 크름 [IV]의 표준시료는 각각
질산크롬 9수화물 (Cr(N03)3'9H20)과 크롬산 칼륨 (K2Cr04)을 증류수에 녹여 1000 ppm의 용액을 제조한 다음 회석하여 5 ppb, 50 ppb, 125 ppb, 250 ppb, 500 ppb, 1000 ppb의 농도로 각각 제조한다.
화학발광시약인 루미놀은 제 1 시약주입구 (13)를 통해서 주입하고, 과산화수소는 제 2 시약주입구 (14)를 통해 주입한다. 시료는 시료 주입구 (11)로 주입하고, 환원제는 환원제 주입구 (12)를 통해 각각 주입한다. 각 용액의 주입 시 사용된 연동펌프의 유속은 7.5μΙ7ηήη이다.
도 4a는 화학발광시약에 0.1 M의 브롬이온의 유무와 크롬 [IV]의 농도에 따른 화학발광신호를 나타낸 그래프이고, 도 4b는 화학발광시약에 0.1 M의 브롬이온의 유무에 따른 크롬 [IV]의 검량곡선을 나타낸 그래프이다.
본 실험예에 사용된 표준시료는 5 ppb, 50 ppb, 125 ppb, 250 ppb, 500 ppb, 1000 ppb크롬 [IV]이며, 환원제를 주입하고 ΐΜ의 브롬이온을 첨가한 화학발광시약과 브롬이온을 첨가하지 않은 화학발광시약을 사용하여 각 농도에 따른
화학발광신호를 연속적으로 측정하였다. 브롬이온은 염기성 조건 하에 금속이온, 과산화수소 그리고 루미놀이 반웅하여 생성된 복합체를 공격한다. 이로 인해 화학발광을 일으키는 들뜬 상태의 3-아미노프탈레이트 (3-aminophthalate)가 더 많이 생성되어 화학발광세기가 증가한다. 도 4a에서 나타낸 바와 같이 0.1 M의
브롬이온을 첨가한 화학발광시약을 사용하였을 때 브름이온을 첨가하지 않은 화학발광시약을 사용하였을 때보다 화학발광세기가 크게 증가한 것을 알 수 있다. 도 4b에서 0.1 M의 브름이은을 첨가한 화학발광시약의 검량곡선은 y =
0.00100x + 0.00289 (R2=0.9999)이며, 브롬이은을 첨가하지 않은 화학발광시약의 검량곡선은 y = 0.000182x + 0.000212 (R2=0.9999)이다. 여기서 y는 화학발광세기를 의미하며 ,χ는 농도, R2은 선형계수를 의미한다. 이러한 검량곡선은 분석하고자 하는 물질로 제조한 표준시료를 사용해 낮은 농도부터 농도 순으로
화학발광세기를 측정하여 분석하고자 하는 물질의 농도에 따른 화학발광세기의 관계를 선으로 나타낸 것이다.
도 4b에 도시된 바와 같이 0.1 M의 브름이온을 첨가한 화학발광시약올 사용하였을 때 브름이온을 첨가하지 않은 화학발광시약올 사용하였을 때보다 검량곡선의 기울기가 약 5.5배 증가한 것을 알 수 있다.
그리고 0.1 M의 브롬이온을 첨가하였을 때 검출한계는 0.32 ppb로,
브롬이온을 첨가하지 않았을 때의 검출한계 1.3ppb에 비해 약 4배 낮은 검출한계를 가진다. 따라서 화학발광시약에 0.1 M 브롬이온을 첨가함으로써 더 높은 감도와 낮은 검출한계로 크름을 검출하는 것이 가능하다.
[실험예 2]
본 실험예 2에서는 미세유체칩의 상판 (10)과 하판 (20) 모두를 검정색
PDMS로 제작한 것과 투명 PDMS로 제작한 것을 크롬 검출장치에 적용하여 총크롬의 화학발광세기를 측정하였다. 본 실험예는 암실에서 수행되었다.
도 5는 검정색 미세유체칩과 투명한 미세유체칩으로부터 얻은 크름 [IV]의 농도에 따론 화학발광신호를 나타낸 그래프이다.
도 5에 도시된 바와 같이 투명 PDMS로 제작한 미세유체칩은 외부의 빛이 검출기로 들어가 바탕신호 (background signal)의 세기와 신호의 잡음이 크게 증가하였고, 크롬 [IV] 표준시료 (5 ppb, 50 ppb, 125 ppb, 250 ppb, 500 ppb, 1000 ppb)로부터 얻은 화학발광신호 또한 주변의 빛과 화학발광반웅에서 발생한 빛의 산란에 의해 신호가 불안정하고 잡음이 큰 것을 알 수 있다.
그리고 바탕신호의 세기가 크게 증가함에 따라 50 ppb 이하의 크롬 [ΙΠ]의 화학발광신호는 바탕신호와 구별할 수 없다. 이와 같이 투명 PDMS로 제작한 미세유체칩으로부터 얻은 화학발광신호는 바탕신호의 세기와 잡음의 증가에 따라 검출한계가 높아지고, 검출할 수 있는 크롬의 농도범위가 축소된다.
반면, 검정색 PDMS로 제작한 미세유체칩은 외부의 빛이 검출기로 들어가는 것을 차단하여 바탕신호의 세기와 잡음이 감소하였으며, 화학발광신호가 매우 안정적이다. 크롬 검출장치는 검정색 PDMS로 제작한 미세유체칩을 사용함으로써 외부의 빛을 효과적으로 차단하여 더 높은 감도와 낮은 검출한계로 크롬을 검출하는 것이 가능하고, 검출할 수 있는 크롬의 농도범위를 확장할 수 있다.
[실험예 3]
본 실험예 3에서는 500 ppb 크름 [m]를 시료 주입구에 주입하고, 냉각 CCD(CooledCCD)카메라로 화학발광을 관찰하였다. 도 6은 검출채널에서 나타나는 500 ppb 크롬 [III]의 화학발광을 넁각 CCD(CooledCCD)카메라로 관찰한사진이다. 화학발광은 검출채널 (17)의 가운데 부분에서 가장 강하게 나타내며, 시작 부분과 끝 부분에서의 화학발광세기는 가운데 부분과 비교하여 상대적으로 약한 것을 알
수 있다. 이는 화학발광의 생성부터 소멸까지 화학발광반응으로부터 발생한 대부분의 빛을 측정할 수 있도록 미세유체칩의 디자인과 시약의 조건이
최적화되어 고감도의 크름 검출이 가능함을 보여주는 결과이다.
[실험예 4]
본 실험예 4에서는 환원채널의 길이를 100cm로 일정하게 고정하고, 시약 흔합채널 (16)의 길이를 각각 Ocm, 20 cm, 60 cm, 100 cm로 변화시키면서
화학발광세기를 측정하였다.
도 7a는 시약 흔합채널의 길이 변화에 따른 크롬 [IV]의 화학발광신호를 나타낸 그래프이고, 도 7b는 시약 흔합채널의 길이 변화에 따른 크롬 [IV]의 검량곡선을 나타낸 그래프이다.
본 실험예에 사용된 표준시료는 5ppb, 50 ppb, 125 ppb, 250 ppb, 500 ppb, 1000 ppb 크름 [IV]이며, 환원제를 주입하여 각 농도에 따른 화학발광신호를 연속적으로 측정하였다. 염기성 조건에서 과산화수소에 의한 루미놀 산화반웅의 반웅속도가 매우 빠르므로, 화학발광시약인 루미놀과 과산화수소는 촉매 역할을 하는 크롬 [ΠΙ]가 없는 경우에도 시약흔합채널 (16)에서 반웅하여 화학발광이 일어난다. 따라서 도 7a에 나타낸 바와 같이 시약 흔합채널 (16)의 길이가 길어질수톡 검출채널 (17)에서 나타나는 화학발광의 세기가 감소하며, 도 7b는 도 7a로부터 얻은 검량곡선으로 시약 흔합채널 (16)의 길이가 길어짐에 따라 검량곡선의 기울기가 감소하는 것을 알 수 있다. 그러나 시약 흔합채널 (16) 없이 검출채널에 시약을 바로 주입할 경우 (시약 흔합채널 Ocm)에는 환원채널 (15)과의 길이의 차이에서 발생하는 큰 압력차로 인해 화학발광신호가 매우 불안정하여 도 7b의 검량곡선에서 500 ppb 크름 [IV]의 화학발광세기가 선형성에서 벗어나는 것을 알 수 있다.
검출장치의 감도는 검량곡선의 기울기와 측정의 재현성에 의해 결정된다. 도 7b에서 시약 흔합채널 (16)의 길이가 Ocm일 때를 제외한 나머지의 경우에 대해서 측정의 재현성은 같다고 가정할 수 있다. 따라서 시약 흔합채널 (16)의 길이에 따른 크름 검출 감도는 검량곡선의 기을기에 의해 결정되므로, 시약 흔합채널 (16)의 길이가 20 cm일 때 가장 높다ᅳ
[실험예 4]
본 실험예 4에서는 크롬 [IV]가 크롬 [ΠΙ]로 환원되는 환원율을 확인하기
위해 총크름 농도를 looo ppb로 동일하게 하고, 크름 [m]와 크름 [IVᅵ의 비율을 각각 3:7, 5:5, 7:3으로 다르게 하여 준비한 표준시료를 미세유체칩 A와
미세유체칩 B에 각각 순차적으로 주입하여 화학발광신호를 측정하였다.
도 7은 환원채널이 시약 흔합채널에 비하여 더 길게 형성된
미세유체칩 A (환원채널 (15): 100cm, 시약 흔합채널 (16):20 cm)와 환원채널과 시약 흔합채널의 길이가 동일한 미세유체칩 B (환원채널 (15): 20 cm, 시약 흔합채널 (16): 20 cm)에서 1000 ppb총크틈 내에 크름 [ΠΙ]와 크름 [IV]의 비율이 다른 표준시료들로 측정한 화학발광신호를 나타낸 그래프이다.
미세유체칩 A는 시료에 포함된 크롬 [IV]가 크름 [m]로 모두 환원되어 시료 내의 크롬 [m]와 크롬 [IV]의 조성비가 다름에도 동일한 화학발광세기를 보인다. 미세유체칩 B는 시료 중에 포함된 크름 [IV]의 일부분만이 크름 [m]로 환원되어 서로 다른 조성의 시료가 각기 다론 화학발광세기를 보인다.
상기한 바와 같이 본 발명에 따르면 환원채널 (15)의 길이가 시약
흔합채널 (16)의 길이보다 더 길게 형성됨으로써 크롬 [IV]의 농도를 정확하게 측정할 수 있다.
[실험예 5]
본 실험예 5는 넓은 농도 범위의 크롬 [IV]가 크롬 [m]로 100% 환원되는지 확인하기 위하여 5 ppb, 50 ppb, 125 ppb, 250 ppb, 500 ppb, 1000 ppb의 크롬 [ΙΠ] 그리고 크롬 [IV] 표준시료 각각올 시료 주입구 (11)에 순차적으로 주입하고, 환원제 주입구 (12)에 환원제를 주입하여 화학발광신호를 측정하였다.
도 8a는 환원제를 주입하여 얻은 크롬 [ΙΠ]와 크롬 [IV]의 농도에 따른 화학발광신호를 나타낸 그래프이고, 도 8b는 환원제를 주입하여 얻은 크롬 [m]와 크롬 [IV]의 검량곡선을 나타낸 그래프이다.
도 8a는 각 농도의 크롬 [ΠΙ]와 크롬 [IV]는 거의 동일한 화학발광세기를 나타낸다.
도 8b에서 크롬 [ΙΠ]의 검량곡선은 y = 0.00 ΙΟΟχ- 0.00115 (R2=0.9999)이며, 크롬 [IV]의 검량곡선은 y = 0.00100x + 0.00127 (R2=0.9999)이다. 여기서 y는
화학발광세기를 의미하며 ,χ는 농도, R2은 선형계수를 의미한다.
크롬 [ΠΙ]와 크롬 [IV]의 검량곡선은 기울기가 동일하고 y절편이 아주 미세하게 상이하므로 각각의 검량곡선을 하나의 그래프에 도시한 결과 도 8b와
같이 2개의 검량곡선이 겹치는 것을 알 수 있다ᅳ
도 8의 결과로부터 미세유체칩과 이를 포함하는 화학발광 검출장치는 넓은 농도 범위 (5~1000ppb)의 크름 [IV]를 크름 [ΙΠ]로 100%환원할 수 있어, 분석오차 없이 크롬 [mᅵ와 크롬 [IV]의 농도를 정확하게 검출할 수 있음을 알 수 있다.
[실험예 6]
본 실험예 6은 5 ppb, 50 ppb, 125 ppb, 250 ppb, 500 ppb, 1000 ppb의 총크롬과 크롬 [m] 표준시료 각각으로부터 얻은 농도에 따른 화학발광신호를 측정하였다. 도 9a는 총크름과 크름 [m]의 농도에 따른 화학발광신호를 나타낸 그래프이고, 도 %는 총크름과 크름 [m]의 검량곡선을 나타낸 그래프이다.
동일한 농도의 시료에 대해 크롬 [m]의 화학발광세기가 총크롬
화학발광세기의 2배인 것을 알 수 있다. 도 8a에 나타낸 바와 같이 환원제를 주입할 경우, 동일한 농도의 크롬 [m]와 크롬 [IV]는 거의 유사한 화학발광세기를 갖는다. 그러나 본 실험예 6은 크름 [m] 분석 시, 시료 주입구 (Π)와 환원제 주입구 (12)로 시료를 주입하고, 총크롬 분석 시, 시료 주입구 (11)로만 시료를 주입하고 환원제 주입구 (12)로 환원제를 주입함에 따라 시료의 농도가 1/2로 희석되어 동일한 농도의 크롬 [m]의 화학발광세기는 총크롬의 화학발광세기의
2배이다.
도 9b는 도 9a에 도시한 농도에 따른 화학발광신호를 총크름과 크롬 [ΠΙ] 각 z|에 대하여 세 번 반복하여 얻은 검량곡선으로 재현성이 매우 높아
상대표준편차가 매우 작은 것을 알 수 있다. 도 9b에서 크룸 [ΠΙ]의 검량곡선은 y = 0.00200x + ().00112(R2=0.9999)이며, 총크롬의 검량곡선은 y - 0.00100x + 0.00289 (R2=0.9999)이다. 여기서 y는 화학발광세기를 의미하며, x는 농도 , R2은 선형계수를 의미한다.
본 실험예 6에 따르면 크롬 [ΙΠ] 검량곡선의 기울기는 총크롬 검량곡선의 기울기의 2배임을 알 수 있다. 이러한 관계를 이용하여 총크름 검량곡선으로 현장시료 내에 존재하는 총크롬과 크롬 [ΠΙ], 크름 [W]의 농도를 분석한 결과 2개의 검량곡선 (총크롬과 크롬 [m] 검량곡선)으로 계산한 결과와 유사함을 알 수 있다
(표 2). 이와 같이 본 발명에 따른 크롬 검출장치는 총크름 검량곡선만으로 총크롬과 크롬 [ΠΙ],크름 [IV]의 정확한 분석이 가능하며 분석시간을 크게 줄일 수
있다.
본 실험예 6으로부터 얻은 총크롬과 크롬 [ΙΠ] 각각의 검출한계는 0.32 ppb, 0.13ppb이다. 그리고 선형구간은 5ppb~1000ppb로 법적규제농도를 포함하고 있어 시료의 희석이나 농축 등의 전처리 없이 크름 검출이 가능하다.
[실험예 7]
루미놀 화학발광반웅은 크름 [m] 이외에 다른 금속이온들 (코발트 [π], 철 [π], 구리 [π], 니켈 [π] 등)도 참여하며, 크롬을 포함한 금속이온들의 물의 용해도와 이동도는 pH에 크게 의존한다. 예를 들어 산성비 등의 원인으로
발생하는 물의 산성화로 인해 물의 pH가 4.0 4.5까지 내려가 금속이은의 농도가 증가함으로 인해 방해금속이은들에 의한 영향이 더욱 심각해진다. 이에 본 실험예
7은 물의 산성화로 인한 pH 변화에도 다론 금속이온들에 의한 방해효과 없이 크롬을 선택적으로 검출 가능한지 확인하였다.
방해금속이은들에 의한 효과는 EDTA를 사용하여 제거할 수 있다 .EDTA는 염기성 조건에서 금속이온과 반응하여 착물을 형성하며, 착물이 형성된
금속이온은 화학발광에 참여하지 않는다. 반면, 크롬 [ΠΙ]는 EDTA와 반웅 시 다른 금속이은에 비해 상대적으로 느린 속도로 착물을 형성하므로, 시료 내에 크롬을 선택적으로 분석하는데 적용이 가능하다.
도 10은 크름 [ΙΠ]와 철 [Π]가 흔합된 시료에 EDTA의 유무와 환원제의 유무에 따른 크름 [ΠΙ]의 화학발광신호를 나타낸 그래프이다. 도 10에서 (a)는 500 ppb 크름 [m]만 용해되어 있는 시료로부터 측정된 화학발광신호로 다른 시료들의 화학발광세기에 기준이 되는 것으로 (a)의 화학발광세기를 100%로 정하였다.
도 10에서 (b)는 500 ppb 크름 [ΙΠ]와 5ppm 철 [Π]가 용해된 시료를 EDTA를 넣지 않은 루미놀, 과산화수소로 분석한 것으로 117%의 화학발광세기를 나타낸다.
17%의 화학발광세기의 증가는 방해물질인 5ppm 철 [Π]로부터 발생한 것이다. 도 10에서 (c)는 (b)와 동일한 시료를 주입하고, 루미놀, 과산화수소가 용해된 완충용액에 각각 10 mM EDTA를 용해한 시약을 주입하여 측정한
화학발광신호로 (a)와 동일한 100%의 화학발광세기를 보였다. 상기한 결과로부터
10 mM EDTA를 사용하여 5 ppm 철 [Π]에 의한 방해효과 없이 선택적으로
크름 [m]를 검출할 수 있음을 확인하였다.
도 10에서 (d)는 (c)와 동일한 실험조건에서 두 개의 시료 주입구 중에
하나의 주입구로 환원제를 주입하여 얻은 화학발광신호로 50%에 해당하는 화학발광세기를 보였다 .50%의 화학발광세기는 환원제와 주입으로 시료의 농도가 1/2로 회석되었기 때문이다ᅳ 상기 결과로부터 10mM의 EDTA를 사용하였을 때 pH가 2.5인 환원제를 주입하여도 화학발광신호에 영향을 끼치지 않는다는 것을 알 수 있다.
도 10의 결과로부터 산성화로 인해 방해금속이온의 농도가 높아졌을 때에도, 분리과정 없이 EDTA를 사용하여 크롬 [m]와 크름 [IV]를 선택적으로 검출할 수 있음을 알 수 있다.
[실험예 8]
본 실험예 8은 크름 [m]분석 시 증류수의 주입으로 인한 침전물의 발생 유무를 확인하기 위해 500 ppb 크롬 [ΙΠ]와 5ppm 철 [Π]를 용해한 용액의 pH를 4.5로 조절하여 준비한 시료를 시료 주입구 (11)와 환원제 주입구 (12)로 주입하였을 때와, 시료 주입구 (11)로 500ppb 크름 [m]와 철 [Π]를 용해한 용액을 주입하고, 환원제 주입구 (12)로 증류수를 주입하였을 때의 화학발광신호를 확인하였다.
종래기술은 시료 내에 크름 [m]검출 시 두 개의 주입구 중에 하나의 주입구로 증류수 (pH 5.8 6.2)를 주입하였다. 상기의 경우 환원채널 (20)에서 시료와 증류수가 섞이는 과정에서 시료의 pH가 변하여 크롬 [ΙΠ]의 침전을 유발할 수 있다.
도 11은 크름 [m]와 철 [π]가 흔합된 시료에 증류수를 주입하여 얻은 크롬 [ΠΙ]의 화학발광신호를 나타낸 그래프이다. 도 11에서 (a)는 500 ppb
크롬 [m]를 시료 주입구 (11)와 환원제 주입구 (12)로 주입하여 얻은 화학발광신호로 이것의 화학발광세기를 100%로 정하였다. 도 11에서 (b)는 pH4.5의 500 ppb 크롬 [ΙΠ]와 5ppm 철 [Π]를 용해한 시료를 시료 주입구 (11)와 환원제 주입구 (12)로 각각 주입하여 얻은 화학발광신호로 도 11에서 (a)보다 17% 높은 화학발광세기를 보였다. 도 11에서 (d)만 시약에 EDTA를 첨가하여 화학발광신호를 얻었다.
따라서 도 11의 (b)에서 17%의 화학발광세기의 증가는 5ppm 철 [Π]에 의한 방해효과로부터 발생한 것이다.
도 11에서 (c)는 시료 주입구로는 500 ppb 크롬 [ΠΙ]와 5ppm 철 [Π]를 용해한 시료를 주입하고, 환원제 주입구로는 증류수를 주입하여 얻은
화학발광신호이다. 만약 증류수의 주입으로 시료가 어떠한 영향도 받지 않았다면,
시료의 농도가 1/2로 회석되어 58.5%의 화학발광세기를 보여야 한다. 그러나 도 11에서 (c)는 17.3%의 화학발광세기를 보였다. 이는 환원채널에서 ρΗ4·5의 시료가 증류수와 섞인 후 ρΗ가 높아져 크톰 [ΠΙ]의 침전 또는 철과 크롬의 공침현상이 발생하여 화학발광세기가 41.2% 줄어든 것이다. 도 11에서 (d)는 (c)와 동일한 방법으로 시료와 증류수를 주입하고, EDTA가 용해된 시약을 주입하여 얻은 화학발광신호로 시료 내에 용존된 상태로 남아있는 철 [Π]가 EDTA에 의해 제거되어 5.4%의 화학발광세기가 감소한 것을 의미한다. 상기 결과로부터 화학발광법으로 크름 [m]를 검출할 때, 증류수를 주입하면 분석오차가 발생하고, 이러한 경우 EDTA를 주입한다고 하더라도 다른 금속이은들에 의한 방해효과를 제거할 수 없음을 알 수 있다.
[실험예 9]
본 실험예 9에서는 EDTA를 첨가하지 않은 시약과 10mM의 EDTA를 첨가한 시약을 사용하여 루미놀 화학발광반웅에 참여하는 철 [Π], 코발트 [Π], 구리 [Π], 니켈 [Π]와 크름이 흔합된 pH4.5인 시료의 화학발광을 검출하였다.
500ppb의 크롬 [ΠΙ]와 상기 방해금속이온들 각각이 500 ppb 또는 5 ppm 용해된 시료에 pH를 질산을 이용하여 4.5로 조절하였다. 이때 ,ρΗ 조절에 사용되는 질산용액의 부피는 시료 부피의 1/1000 이하로 시료 내에 크름과 방해금속의 농도 변화는 무시할 만 하다. 그리고 500 ppb의 총크름 (250 ppb의 크롬 [ΙΠ]와 250 ppb의 크름 [IV]를 포함)과 방해금속이온들 각각이 500 ppb또는 5 ppm 용해된 시료에 질산을 이용하여 pH를 4.5로 조절하였다.
화학발광시약은 lOmM의 EDTA를 첨가한 완층용액과 EDTA를 첨가하지 않은 완충용액에 루미놀과 과산화수소를 각각 넣어 제조하였다.

[표 1]은 500 ppb의 크롬 [ΙΠ]와 방해금속이온이 용해된 시료의 경우 500 ppb 크롬 [ΠΙ] 시료의 화학발광세기를 100%로 정하고 , 500 ppb 총크톰과 방해금속이온이 용해된 시료의 경우, 500 ppb의 총크름 시료의 화학발광세기를 100%로 하여 EDTA를 첨가하지 않은 화학발광시 약과 10 mM의 EDTA를 첨 가한 화학발광시 약을 주입하였을 때 각 시료에 대한 화학발광세기를 나타낸 것 이다. 화학발광세기 옆의 괄호에 표기 한 값은 각 시료에 대한 화학발광세기를 3번 측정하여 얻은
상대표준편차이다.
EDTA를 첨가하지 않은 시 약을 주입하였올 때, 시료에 존재하는
방해금속이은에 의해 측정 된 화학발광세기는 100%를 초과하였다. 그러나 10 mM의 EDTA를 첨가한 화학발광시 약을 사용한 경우에는 검 출채널에서
방해금속이온이 EDTA에 의해 제거되 어 500 ppb의 크롬 [ΠΙ], 500 ppb의 총크롬 표준시료의 화학발광세기 와 같은 것을 알 수 있으며 , 방해금속이온의 농도가 크름 농도의 10배 (5 ppm)인 시료에 대해서도 방해효과 없이 500 ppb의 크름 [ΙΠ]와 총크름을 선택적으로 분석할 수 있다.
[실험 예 10]
본 실험 예 10은 본 실시 예에 따른 검출장치의 성능을 확인하기 위 해 공장에서 채수한 시료 내에 총크름과 크름 [m], 크롬 [IV]의 농도를 분석한 것이다.
0.45 μπι pore를 갖는 필터를 거 친 시료를 미세유체칩 에 주입하여 크름 농도를 측정 하였다.
표 2는 시료 내에 총크롬과 크롬 [m], 크롬 [IV]의 농도를 분석 한 결과와
원자흡광광도법과 흡광법으로 분석한 결과를 비교한 것으로 원자흡광광도법은 총크름 농도만을 분석할 수 있으며, 검출한계는 20ppb이다. 그리고 흡광법은 크롬 [IV]와 1,5-디페닐카바자이드 (1,5-diphenylcarbazide)의 반응으로 생성되는 붉은색 착물의 540nm의 파장에서의 흡수도를 자외선-가시광선 분광광도계로 측정하여 분석하였으며, 다른 물질에 의한 방해효과가 없어 크롬 [IV] 분석에 있어 선택성이 높은 분석법이다.
본 실험예 10에 따른 시료 내의 크름을 본 실시예에 따른 미세유체칩을 사용하여 화학발광법으로 분석한 결과 시료 1, 시료 2, 시료 3에 포함된 총크름의 농도는 각각 137 ppb, 112 ppb, 128 ppb로 원자흡광광도법으로 분석한 결과와 유사하고, 각각의 시료에 대해 세 번 분석한 결과 원자흡광광도법 보다 더 낮은 상대표준편차를 얻었다.

상기에서는 본 발명의 바람직한 실시예에 대하여 설명하였지만, 본 발명은 이에 한정되는 것이 아니고 특허청구범위와 발명의 상세한 설명 및 첨부한
도면의 범위 안에서 여러 가지로 변형하여 실시하는 것이 가능하고 이 또한 본 발명의 범위에 속하는 것은 당연하다.

시료와 시약을 흔합하는 환원채널과 시약들을 흔합하는 시약 흔합채널, 및 일측 단부에 상기 환원채널과 상기 시약 흔합채널이 병렬로 연결되고 타측 단부에 배출구가 연결된 검출채널이 형성된 제 1 기판; 및
상기 검출채널과 마주하며 빛이 투과하도록 형성된 검출부를 구비하며 상기 제 1 기판에 결합되는 제 2 기판;을 포함하는 미세유체칩.

제 4항에있어서,
상기 제 1 기판에는 게 1 시약의 주입을 위한 제 1 시약 주입구와 제 2 시약의 주입을 위한 제 2 시약 주입구가 형성되고, 상기 제 1 시약 주입구는 제 1 시약채널을 통해서 시약 흔합채널과 연결되며, 상기 제 2 시약 주입구는 제 2 시약채널을 통해서 시약 흔합채널과 연결된 미세유체칩.

상기 검출채널은 중첩 배치되며 일측 단부와 타측 단부가 교호적으로 연결되고, 중앙에서 양쪽 측단으로 갈수록 감소하도록 형성된 미세유체칩.

상기 환원채널과 상기 시약 흔합채널은 중첩 배치되며 일측 단부와 타측 단부가 교호적으로 연결된 미세유체칩.

시료와 시약을 흔합하는 환원채널과 시약들을 흔합하는 시약 흔합채널, 및 일측 단부에 상기 환원채널과 상기 시약 흔합채널이 병렬로 연결되고 타측 단부에 배출구가 연결된 검출채널이 형성된 제 1 기판, 및 상기 검출채널과 마주하며 빛이 투과하도톡 형성된 검출부를 구비하며 상기 제 1 기판에 결합되는 제 2 기판을 포함하는 미세유체칩;
상기 검출부와 마주하도록 배치된 검출기; 및
상기 미세유체칩에 시료 및 시약을 공급하는 공급부;
를 포함하는 크롬 검출장치.

제 8항에있어서,
상기 환원채널은 상기 시약 흔합채널보다 더 길게 형성된 크름 검출장치.

상기 공급부는 상기 시료 주입구로 시료를 공급하고, 상기 환원제 주입구로 아황산칼륨을 공급하며, 상기 제 1 시약 주입구로 염기성 조건의 완층용액에 용해된 루미놀을 공급하고, 상기 제 2 시약 주입구로 염기성 조건의
완층용액에 용해된 과산화수소를 공급하는 크름 검출장치.
