Abstract
Description
人工的にペプチドライブラリを作製する方法としては、従来、化学合成による方法、二次代謝産物の生合成酵素を用いる方法、翻訳合成系などが用いられている。
しかしながら、化学合成による方法では、ライブラリの多様性を大きくするのが困難である。また、スクリーニングや、化合物の構造と活性の相関解析にも時間を要する。
これに対し、二次代謝産物の生合成酵素を用いる方法によれば、迅速かつ簡便に、有機化学的手法では得られないような精巧な骨格の構築や化学変換が可能となる。しかしながら、酵素には基質特異性があるため、合成できる化合物の種類が限られ、大規模な化合物ライブラリの構築に適用するのは難しい。
ライブラリを用いたスクリーニングでは、プロテアーゼ活性を有する標的物質を阻害する化合物を同定することもしばしば必要とされる。しかしながら、ペプチド性化合物のライブラリは、プロテアーゼによって切断されてしまうので、標的物質の活性を阻害する化合物を効率よくスクリーニングすることができない。
修飾の有無や程度が特定できないライブラリは、結局、化学合成系と同様に構造と活性の相関解析が必要となるため、有用性に劣るという問題がある。
この生合成において前駆体となるのは、patE遺伝子産物のPatEペプチドである。patE遺伝子は超可変領域(カセット領域)を有するため、その産物は天然のコンビナトリアルライブラリを構築する。
PatEペプチドは、カセット領域の両側に、翻訳後修飾酵素による認識配列を有する。翻訳後修飾酵素として働くのは、PatA、PatD及びPatGである。PatDは、PatEのカセットにおけるCys、Ser、及びThrにアゾリン骨格を導入し、Cysをチアゾリン骨格に、Ser及びThrをオキサゾリン骨格に変換する。
PatAは、PatEのカセット領域のN末端側の認識配列を切断する。
PatGは、2つのドメインからなり、N末端のオキシダーゼドメインは、PatDにより導入されたアゾリン骨格をアゾール骨格に、即ちチアゾリン骨格をチアゾール骨格に変換する。C末端側のペプチダーゼドメインは、PatEのカセット領域のC末端側の認識配列を切断しながら大環状化する。
従って、これらの共通点は、翻訳後修飾酵素であるPatD及びPatGの基質となるために必要であるようにも考えられた。しかしながら、これらのSer/Thr/Cysのうち、どれが修飾されているのか、されていないのかも不明であり、PatD及びPatGの基質特異性についてはこれまで明らかとなっていなかった。
さらに、PatEのリーダー配列や、認識配列を、天然の配列とは異なる所定の配列にしても、アゾリン骨格導入酵素の基質となり得ること;図1Bに示すように、リーダー配列部分を、カセット領域を含むペプチドとは別個のペプチドとしても、当該ペプチドがアゾリン骨格導入酵素を含む反応系に存在すれば、カセット領域にアゾリン骨格が導入されることを確認した(以上、特許文献1)。
しかしながら、基質ペプチドとは別のペプチドとしてリーダー配列を反応系に加える必要があることから、得られるライブラリが複雑となる。また、リーダー配列を別ペプチドとして加えた場合、アゾリン骨格が常に十分に導入されるとは限らなかった。
そこで、本発明は、基質ペプチドに安定的にアゾリン骨格を導入することができることを課題とする。
そして、この知見に基づいて、リーダー配列をアゾリン骨格導入酵素に結合させたところ、図1Cに示すように、アゾリン骨格導入酵素が常時十分に活性化され、リーダー配列を持たない基質ペプチドにもアゾリン環等のヘテロ環を導入できることを見出した。また、リーダー配列は、アゾリン骨格導入酵素のN末端に結合させた場合に特に高く活性化されること、及び、リーダー配列は、ある程度の長さを有するスペーサを介してアゾリン骨格導入酵素に結合させるとより効果的であることを確認した。
さらに、リーダー配列を結合させたアゾリン骨格導入酵素を用いたときには、基質ペプチドにおいて、カセット配列を挟む2つの認識配列をかなり短くできる一方、カセット配列は多様な配列とすることができること;基質ペプチドのN末端に環状化に必要なアミノ酸又はアミノ酸アナログを配して、ヘテロ環が導入されたペプチドを効率よく環状化できること;及び、リーダー配列を結合させたアゾリン骨格導入酵素を用いて得られるライブラリは扱いやすい簡略な構成であることを確認し、本発明を完成するに至った。

また、リーダー配列を結合させたアゾリン骨格導入酵素を用いてヘテロ環を含む化合物ライブラリを構築する場合、リーダー配列を独立したペプチドとして加えないためライブラリ構築の反応条件を簡略化でき、また、ヘテロ環を含む化合物にリーダー配列がないことから、余分なリーダー配列を除去すること無しに活性種のスクリーニングを行うことが出来る。さらに、ヘテロ環を含む化合物にリーダー配列がないことから、大環状骨格を形成する配列デザインを行いやすい。かかるヘテロ環を含む化合物のライブラリをスクリーニングに用いれば、標的物質がプロテアーゼ活性を有する場合であっても、当該標的物質に結合する化合物を探索することができる。
また、ヘテロ環を含む化合物のライブラリは、mRNAディスプレイ法に応用できるので、標的物質に結合活性を有する化合物を濃縮し、得られた化合物のペプチド部分をコードする核酸配列を容易に同定できる。
本発明は、アゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物の製造方法を提供する。
本明細書において「アゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物」とは、下記式(I)で示されるペプチドにおいて、(Xaa3)nに含まれる、Cys、Ser、Thr、2,3-ジアミノ酸、ホモシステイン、ホモセリン、2,4-ジアミノ酸、及びこれらのアナログの少なくとも1つに、アゾリン骨格導入酵素によってヘテロ環が導入されている化合物をいう。

このような構成とすることにより、Cys、Ser、Thr、2,3-ジアミノ酸、ホモシステイン、ホモセリン、2,4-ジアミノ酸、及びこれらのアナログが、(Xaa3)nの偶数番目にくるので、アゾリン骨格導入酵素の性質上、アゾリン環等のヘテロ環が導入されやすくなる。なお、Xaa5が、Cys、Ser、Thr、2,3-ジアミノ酸、ホモシステイン、ホモセリン、2,4-ジアミノ酸、又はこれらのアナログであってもよい。
Xaa6は、特にアゾリン骨格が導入されやすいCysのみとしてもよい。

ここでいう親水性アミノ酸は、Asp、Glu、Arg、Lys、Asn、Gln又はこれらの親水性誘導体をいうが、これらに限定しない。
この中で、無細胞翻訳系を用いる場合、式(I)のペプチドは、これをコードする核酸を調製し、当該核酸を無細胞翻訳系で翻訳することによって得ることができる。式(I)で表されるペプチドをコードする核酸は、生体の翻訳系で用いられる遺伝暗号、リプログラミングした遺伝暗号、又はこれらの組み合わせを用いて、当業者が適宜設計することができる。なお、核酸は、DNAであってもRNAであってもよい。
無細胞翻訳系を用いる方法によれば、非天然アミノアシルtRNAを使用することで、天然アミノ酸に加え、その誘導体や人工のアミノ酸を使用することが出来る。例えば、本発明者らが開発した人工アミノアシル化RNA触媒フレキシザイム(Flexizyme)を利用できる。
これらを含む系に、透析を用いて連続的にエネルギーを供給することで、数100μgから数mg/mLのタンパク質を生産することができる。DNAからの転写を併せて行うためにRNAポリメラーゼを含む系としてもよい。市販されている無細胞翻訳系として、大腸菌由来の系としてはロシュ・ダイアグノスティックス社のRTS-100(登録商標)、再構成型翻訳系としてはPGI社のPURESYSTEM(登録商標)やNew England BioLabs社のPURExpressR In Vitro Protein Synthesis Kit等、小麦胚芽抽出液を用いた系としてはゾイジーン社やセルフリーサイエンス社のもの等を使用できる。

無細胞翻訳系によれば、発現産物を精製することなく純度の高い形で得ることができる。

なお、任意のtRNAに任意のアミノ酸を連結する方法は、フレキシザイムを用いる方法に限られず、その他の方法も本発明に適用可能である。
大環状化に用いられるアミノ酸は、(Xaa2)m、(Xaa3)n、(Xaa4)oのいずれに含まれていてもよい。ヘテロ環が導入されたアミノ酸が、大環状を構成するアミノ酸の1つとなっていてもよいし、大環状を構成しないアミノ酸の1つとなっていてもよい。
大環状化の方法は特に限定されないが、例えば、式(I)で表されるペプチドに、以下の官能基1を有するアミノ酸と、対応する官能基2を有するアミノ酸を含めることにより、大環状化してもよい。官能基1と官能基2は、どちらがN末端側にきてもよい。
例えば、Xaa2のいずれかのアミノ酸を、下記の官能基1を有するものとすることができ、対応する官能基2を有するアミノ酸を(Xaa4)oに含めることにより、式(I)で表されるペプチドを発現させた後、環状化反応を実施できる。また、Xaa2のいずれかのアミノ酸を、官能基2を有するものとし、対応する官能基1を有するアミノ酸を(Xaa4)oに含めても良い。
式中、X1はCl、Br又はIであり、Arは置換基を有していてもよい芳香環である。

本発明に係る方法に用いられるアゾリン骨格導入酵素には、アゾリン骨格導入酵素の基質のリーダー配列又はその部分配列が結合している。
本明細書において「アゾリン骨格導入酵素」は、PatD及びこれと相同性を有する酵素を含む。PatDと相同性を有する酵素としては、例えば、Leeらの報告(Lee, S. W. et al., PNAS vol.105, No.15, 5879-5884, 2008)に含まれるものを用いることができるが、これらに限定されない。また、アゾリン骨格導入酵素は、アゾリン骨格導入活性を有する限り、変異体であってもよい。なお、本明細書においては、「ヘテロ環状化酵素」もアゾリン骨格導入酵素と同義で用いられる。

リーダー配列として、alpha helicityの高い配列を用いてもよい。
配列番号:1~4のどこに位置する部分であるかは、特に限定されない。例えば、アゾリン骨格導入酵素の活性化能を有する限り、配列番号:1~4のアミノ酸配列のうちC末端の4アミノ酸、5アミノ酸、又は6アミノ酸を含むものとしてもよいし、N末端の4アミノ酸、5アミノ酸、又は6アミノ酸を含むものとしてもよく、N末端もC末端も含まない4アミノ酸、5アミノ酸、又は6アミノ酸を含むものしてもよい。
このようなリーダー配列の部分配列が、アゾリン骨格導入酵素活性化能を有するか否かは、例えば、リーダー配列の存在下で、アゾリン骨格導入酵素と基質ペプチドを結合させるなど、公知の方法で確認することができる。
スペーサペプチドのアミノ酸配列は、アゾリン骨格導入酵素と基質ペプチドとの反応に悪影響を及ぼさない限り、どのような配列であってもよい。
また、本発明に係るアゾリン骨格導入酵素は、図2A~Fに示すアミノ酸配列からなるものに加え、これらの配列のいずれか1つと80%以上、85%以上、90%以上、95%以上、又は98%以上の配列同一性を有し、アゾリン骨格導入活性を有するもの、及び、これらの配列のいずれか1つにおいて、1個、2個、3個、4個、5個~10個程度のアミノ酸が、欠失、付加又は置換した配列からなり、アゾリン骨格導入活性を有するものも含まれる。
反応の確認は、例えば、MALDI-TOF-MSを用いて質量変化を測定することにより行うことができる。
また、本発明は、本発明に係るアゾリン骨格導入酵素をコードする核酸も包含する。
本発明は、アゾール骨格導入酵素によって導入されるヘテロ環を含む化合物の製造方法も包含する。
なお、本明細書においては、アゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物と、アゾール骨格導入酵素によって導入されるヘテロ環を含む化合物とを、総称して「ヘテロ環化合物」と呼ぶ場合もある。
アゾール骨格導入酵素として、その基質のリーダー配列又はその部分配列を結合させたものを用いてもよい。あるいは、反応容器内に、独立したペプチドとして、基質のリーダー配列又はその部分配列を加えて、反応を行ってもよい。アゾール骨格導入酵素の基質のリーダー配列又はその部分配列は、アゾリン骨格導入酵素の基質のリーダー配列又はその部分配列と同一としてもよい。
本発明は、アゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物を2種以上含むライブラリ(以下「アゾリン系化合物ライブラリ」という。)の製造方法も包含する。
かかるライブラリの製造方法は、上述したアゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物の製造方法において、(Xaa3)nが異なるペプチドを2種以上含むペプチドライブラリを調製し、このペプチドライブラリをアゾリン骨格導入酵素で修飾する工程を含む。
すなわち、このmRNAライブラリは、(Xaa3)nが異なる多数のペプチドをコードするmRNAを含むものであり、例えば、(Xaa3)nに当たる部分をコードするものとして(NNN)n、(NNK)n、(NNT)n、(NNG)n、などの配列を含むDNAを合成し、これを転写することによって作製することができる。ここで、NはA/C/G/Tのうち任意の一つ、KはG/Tのうち任意の一つを意味し、NNN及びNNKは20種類のタンパク質性アミノ酸のうちの任意の一つを、NNU及びNNGはそれぞれ15及び13種類のタンパク質性アミノ酸のうちの任意の一つをコードする。
(Xaa3)nが(Xaa5-Xaa6)pである場合、(Xaa5-Xaa6)pをコードする部分のmRNAは、例えば、(NNK-WST)n、(NNK-TGT)nなどの配列を含むDNAを合成し、これを転写することによって作製することができる。ここで、NはA/C/G/Tのうち任意の一つ、KはG/Tのうち任意の一つ、WはA/Tのうち任意の一つ、SはC/Gのうち任意の一つを意味し、NNN及びNNKは20種類のタンパク質性アミノ酸のうちの任意の一つ、WSTはSer/Thr/Cysのうちの任意の一つ、TGTはCysをコードする。
(Xaa3)mをコードする核酸の5'末端に、開始コドンを含む(Xaa2)mをコードする核酸を連結し、3'末端側に(Xaa4)oをコードする核酸を連結した核酸を合成し、これを翻訳すれば式(I)で表されるペプチドライブラリをコードする核酸ライブラリを得ることができる。
ATG-GGN-(NNK)x-NYK-TGC-NYK-(NNK)x-NYK-TGC-NYK-(NNK)x
式中、NはA、C、G又はTを表し、KはG又はTを表し、YはC又はTを表し、WはA又はTを表し、SはC又はGを表す。
この核酸によれば、(Xaa2)mがATG-GGNでコードされ、(Xaa3)nが(NNK)x-NYK-TGC-NYK-(NNK)x-NYK-TGC-NYKでコードされ、(Xaa4)oが(NNK)xでコードされる。
このような構成によれば、TGCでコードされるCysの両側が、非親水性アミノ酸となる。
ATG-(NNK)m-[(NYK)- (WST)]n-(NNK)o
式中、NはA、C、G又はTを表し、KはG又はTを表し、YはC又はTを表し、WはA又はTを表し、SはC又はGを表す。
このような核酸を用いれば、WSTは、Ser、Thr及びCysのいずれかを表し、NYKは、非親水性アミノ酸を表すので、(Xaa3)nの偶数番目にSer、Thr又はCysを配し、CysのN末端側に親水性アミノ酸が隣接するのを防ぐことができるので、アゾリン骨格導入酵素による修飾を受けやすいペプチドとすることができる。
(Xaa4)oをコードする核酸として、(NNK)oに代えて、NYK-(NNK)xを用いれば、CysのC末端側にも親水性アミノ酸が隣接するのを防ぐことができる。
このようなペプチド-mRNA複合体ライブラリを用いて、標的物質に結合するペプチドのスクリーニングを行えば、選択されたペプチド-mRNA複合体に対して逆転写反応を行うことにより、cDNAを含む複合体が得られるので、その塩基配列を常法により決定することができる。
ペプチド-mRNA複合体の作製は、例えば、公知の方法でmRNAライブラリの各mRNAの3'末端にピューロマイシンを結合させ、ピューロマイシン結合mRNAライブラリを作製し、このピューロマイシン結合mRNAライブラリを無細胞翻訳系で発現させることにより行うことができる。
こうして、ペプチド-mRNA複合体ライブラリを得た後、アゾリン骨格導入酵素と反応させ、アゾリン系化合物ライブラリを得ることができる。
本発明は、アゾール骨格導入酵素によって導入されるヘテロ環を含む化合物を2種以上含むライブラリ(以下「アゾール系化合物ライブラリ」といい、アゾリン系化合物ライブラリとアゾール系化合物ライブラリを総称して「ヘテロ環化合物ライブラリ」という。)の製造方法も包含する。
当該方法は、上述したアゾール骨格導入酵素によって導入されるヘテロ環を含む化合物ライブラリの製造方法において、ヘテロ環の導入されたペプチドライブラリとアゾール骨格導入酵素とを反応させ、アゾリン骨格導入酵素によって導入されたヘテロ環の少なくとも1つを、アゾール骨格導入酵素で導入されるヘテロ環に変換する工程を含む。
本発明は、アゾリン骨格導入酵素でヘテロ環が導入されたペプチドを2種以上含む新規なアゾリン化合物系ライブラリも包含する。
リーダー配列を結合させることによって、アゾリン骨格導入酵素を活性化させた場合、ライブラリ部分(カセット領域)に当たる(Xaa3)rを挟む認識配列は、今まで考えられていたよりも短い配列で足り、且つ短い配列である場合に、効率よくアゾリン骨格が導入されることが判明した。
よって、本発明に係るアゾリン系化合物ライブラリは、

本発明は、アゾール骨格導入酵素でヘテロ環が導入されたペプチドを2種以上含む新規なアゾール系化合物ライブラリも包含する。
本発明に係るアゾール系化合物ライブラリは、

アゾール系化合物ライブラリにおいては、アゾール骨格導入酵素で修飾されたペプチドのそれぞれが、そのペプチド部分をコードするmRNAと複合体を形成していることも望ましい。この構成により、mRNAディスプレイに応用可能となる。
本発明は、標的物質に結合する化合物を同定するスクリーニング方法も包含する。
本発明のスクリーニング方法の一態様は、本発明に係る方法で製造されたヘテロ環化合物ライブラリと、標的物質を接触させてインキュベートする工程を含む。
本明細書において、標的物質は特に限定されず、低分子化合物、高分子化合物、核酸、ペプチド、タンパク質、糖、脂質等とすることができる。特に、本発明のライブラリによれば、標的物質がプロテアーゼ活性を有する場合にも用いることができる。
標的物質と、ライブラリは、適宜選択された緩衝液中で接触させ、pH、温度、時間等を調節して相互作用させる。
検出可能な標識としては、ペルオキシダーゼ、アルカリホスファターゼ等の酵素、125I、131I、35S、3H等の放射性物質、フルオレセインイソチオシアネート、ローダミン、ダンシルクロリド、フィコエリトリン、テトラメチルローダミンイソチオシアネート、近赤外蛍光材料等の蛍光物質、ルシフェラーゼ、ルシフェリン、エクオリン等の発光物質、金コロイド、量子ドットなどのナノ粒子が挙げられる。酵素の場合、酵素の基質を加えて発色させ、検出することもできる。また、ペプチドにビオチンを結合させ、酵素等で標識したアビジン又はストレプトアビジンを結合させて検出することもできる。
この場合、例えば、ヘテロ環化合物-mRNA複合体ライブラリに対して逆転写反応を行った後、当該ライブラリと固相単体に固定した標的物質とを接触させる。標的物質に結合する複合体を選択し、このDNAをPCRで増幅する。このDNAを用いて、再度ヘテロ環化合物-mRNA複合体ライブラリを作製し、同様の操作を繰り返す。
これにより、標的物質に高い親和性を有するヘテロ環化合物-mRNA複合体が濃縮されるので、濃縮された複合体のmRNAの配列を解析して、標的物質に結合するヘテロ環化合物を効率よく同定することができる。
本発明は、ヘテロ環化合物のスクリーニング用キットを提供する。
本発明のスクリーニング用キットの一態様は、本発明に係る製造方法で製造されたヘテロ環化合物ライブラリ、又は、本発明に係るヘテロ環化合物ライブラリを含む。
本発明のスクリーニング用キットは、他に、標的物質とヘテロ環化合物との結合を検出するのに必要な試薬及び装置を含む。かかる試薬及び装置としては、例えば、固相担体、緩衝液、標識用試薬、酵素、酵素反応停止液、マイクロプレートリーダーが挙げられるがこれらに限定されない。

N末端側の発現に関して、pET16bのプラスミドにPatDの遺伝子を導入し、N末端に10xHisタグが付与されたコンストラクトのプラスミドを用意した。そしてPatD遺伝子のN末端領域をNdeIもしくは、NdeI及びNheIを用いて切断し、リーダー配列、及び長さの異なるGSリンカー領域をコードしたDNAを導入して、リーダー配列、及びGSリンカーがPatDのN末端に結合したLS融合PatDのプラスミドを作製した。
C末端側に関しては、初めにPatD遺伝子のC末端側のストップコドンを消去した後、XhoI及びBamHIを用いて切断し、長さの異なるGSリンカー領域、リーダー配列、及びストップコドンをコードしたDNAを導入して、GSリンカー及びリーダー配列がPatDのC末端に結合したLS融合PatDのプラスミドを作製した。
続いて、これらのプラスミドを大腸菌BL21(DE3)pLysS株に形質転換し、30℃で培養した。O.D.が0.4に達した所でIPTG 0.1mMを添加し、大量発現を誘導した後、15℃でオーバーナイトで培養した。回収した菌体をLysis Buffer(1M NaCl, 25mM Imidazole, 50mM HEPES-Na(pH7.7))で懸濁した後、超音波で破砕した。フィルターで濾過したサンプルを、His-Trap HPカラムを用いて精製した。カラムはあらかじめ17CVのBuffer A(500mM NaCl, 25mM Imidazole, 50mM HEPES-Na(pH7.7))で平衡化し、サンプル注入後、Buffer B(500mM NaCl, 1M Imidazole, 50mM HEPES-Na(pH7.7))濃度を徐々に上げることによって、サンプル中のタンパク質を分離し、純粋なLS融合PatDフラクションを得た。
得たサンプルを、Amicon Ultra(Millipore) 30kDaを用いて約4倍に濃縮した。その後PD-10(GE lifescience)を用いてStore Buffer(200mM NaCl,25mM HEPES(pH7.7), 10% glycerol)にバッファ交換し、Amicon Ultra(Millipore) 30kDaを用いて更に約4倍に濃縮した後、-80℃で保存した。
[2]で調製したDNAを、Kawakamiらの方法(Kawakami et al., Chemistry & Biology 15, 32-42 (2008))に従って5.0μlスケールの無細胞タンパク質発現系で転写・翻訳した後(37℃、1時間)、45 mM HEPES-K (pH 8.4), 7.5 mM DTT, 0.5 mM ATP(それぞれ終濃度)を加えて溶液条件を整えた後、[1]で調製したLS融合PatDを加えた。
LS融合PatDの終濃度は6μMとし、反応温度25℃、反応時間16時間とした。
c-18 tip(Thermo Scientific)を用いて、Wash Buffer (4% MeCN, 0.5% AcOH, 95.5%H2O) にてペプチドの脱塩を行い、Elute Buffer(80% MeCN, 0.5% AcOH, 19.5%H2O)にて脱塩後のペプチドを抽出した。
抽出したペプチドは、Matrixとしてα-cyano-4-hydroxycinnamic acid もしくはsinapinic acidを用いて、MALDI-TOF-MSによってペプチドの質量を測定し、LS融合PatD添加による質量変化の有無を確認した。質量変化により、導入されたアゾリン環の個数がわかる。
[1]で調製した各種LS融合PatDを用いて、PatEと同じ配列を有する基質ペプチドM-GLEAS-VTACITFC-AYDGVEPSと反応させ、[4]の方法で、アゾリン環の個数を求めた。
結果を図3A及びBに示す。いずれのLS融合PatDも、基質ペプチドにアゾリン骨格を導入したが、PatDのN末端にリーダー配列を結合させた酵素は、より導入効率が高かった。以降の実験では、LS-(GS)15-PatDを用いた。
[3]の方法でLS融合PatDと各種基質ペプチドとを反応させ、[4]の方法でアゾリン環の個数を求めた。
カセット配列(CS)のN末端側の認識配列(uRS、本発明の(Xaa2)mに相当)と、C末端側の認識配列(dRS、本発明の(Xaa4)oに相当)が異なる基質ペプチドに対する、LS融合PatDによる修飾を調べた。
結果を図4Aに示す。C末端側の認識配列は3残基程度にしても、反応性は変化しなかった。N末端側の認識配列はなくて問題がなかった。Glyが5残基以上連続した認識配列では、反応性が低下する傾向が見られた。
CysのN末端側に親水性アミノ酸(Glu)が隣接しているために反応性が低くなったカセット配列を用いて、認識配列による反応性の違いを調べた。
結果を図4B-1に示す。uRSをGly、又はGly-Gly-Gly、dRSをAla-Tyr-Asp、Ala-Tyr-Asp-Gly-Val、又はAla-Tyr-Asp-Gly-Val-Glu-Pro-Serとした場合に反応性が高い傾向が見られた。
dRSをAla-Tyr-Asp、又はAla-Tyr-Asp-Gly-Ser-Glyとし、6種類のカセット配列に対するLS融合PatDの反応性の違いを調べた。
結果を図4B-2に示す。いずれの場合もLS融合PatDによる修飾を受けた。
dRSをAla-Tyr-Asp、又はAla-Tyr-Asp-Gly-Ser-Glyとし、疎水性アミノ酸からなるカセット配列に対するLS融合PatDの反応性の違いを調べた。
結果を図4B-3に示す。いずれの場合もLS融合PatDによる修飾を受けた。
以降の実験は、uRSをGly、dRSをAla-Tyr-Asp又はGly-Gly-Glyとして行った。
カセット配列の長さが異なる基質ペプチドに対する、LS融合PatDによる修飾を調べた。
結果を図4Cに示す。カセット配列の長さを変えても、反応性への影響は少ないことが確認された。
カセット配列において、CysのN末端側に親水性アミノ酸を隣接させ、LS融合PatDによる修飾を調べた。野生型PatDについては、親水性残基が反応性を低下させることが知られている。
結果を図4D-1に示す。親水性残基が隣接していても、Cysの修飾は十分に進行することが示された。AspがCysに隣接している場合には、若干反応性が低下する傾向が見られた。
IleとAsnからなるカセット配列で、2つのAsnの位置を変えて、LS融合PatDによる修飾に対する、カセット配列中の親水性アミノ酸の影響を調べた。
結果を図4D-2に示す。CysのN末端側にAsnが隣接すると修飾効率が低下したが、C末端側にAsnが隣接しても問題なく修飾された。
カセット配列におけるCysのC末端側の隣接アミノ酸を各種親水性アミノ酸に変えて、LS融合PatDによる修飾に対する影響を調べた。
結果を図4D-3に示す。いずれの場合も効率よく修飾された。

結果を図4D-4に示す。Asn、塩基性アミノ酸、及び酸性アミノ酸以外の場合に効率よく修飾がおきた。
カセット配列をさらに様々な変化させてPatEとは大きく異なるものとし、LS融合PatDによる修飾を調べた。具体的には、Cys以外のアミノ酸を、すべて疎水性アミノ酸とした場合、すべて親水性アミノ酸とした場合、すべて芳香族アミノ酸とした場合、また、Cysが奇数番目に配置された場合について調べた。結果を図4Eに示す。疎水性アミノ酸、又は芳香族アミノ酸を多く含む場合は、Cysの位置にかかわらず、ほぼすべてのCysにアゾリン環が導入されていた。一方、親水性アミノ酸を多く含む場合、修飾されたCysはわずかであった。
また、Cys以外のアミノ酸を、疎水性アミノ酸+親水性アミノ酸、親水性アミノ酸+芳香族アミノ酸、疎水性アミノ酸+芳香族アミノ酸、疎水性アミノ酸+芳香族アミノ酸+親水性アミノ酸とした場合について調べた。結果を図4Fに示す。親水性アミノ酸は、反応を低下させる傾向が見られた。一方で、st125とst126、st119とst122、st121とst123の比較から、親水性アミノ酸が含まれていても、修飾されるCysに隣接していない場合には、反応は大きくは阻害されないことが示唆された。
[6-9]と同様に、PatEとは大きく異なる配列を用いて、カセット配列の長さを変えてLS融合PatDによる修飾を調べた。
結果を図4G-1に示す。カセット配列の長さを変えても、反応性への影響は少ないことが確認された。
より多様なバリエーションのカセット配列についてLS融合PatDによる修飾を調べるために、芳香族アミノ酸からなるカセット配列を用いた。
結果を図4G-2に示す。カセット配列が芳香族アミノ酸を含んでいても、効率よく修飾された。
天然型のカセット配列であるVal-Thr-Ala-Cys-Ile-Thr-Phe-Cys、又はその後半部分Ile-Thr-Phe-Cysを基にして、(i)Cys、Thr、又はSerの被修飾残基は1残基のみとする、(ii) Cys、Thr、又はSerを他のアミノ酸に置き換える際、芳香族アミノ酸(Phe又はTrp)にする、とのルールに従ったカセット配列に対するLS融合PatDによる修飾を調べた。
結果を図4Hに示す。いずれのカセット配列も効率よく修飾された。
カセット配列に非タンパク質性アミノ酸である2,3-ジアミノ酸(Dap)を含む基質ペプチドに対する、LS融合PatDによる修飾を調べた。基質ペプチドの配列は、fMGI-Dap-FWAYDとした。
結果を図4Iに示す。Dapはイミダゾリン環に修飾されたことが確認された。
LS融合PatDで修飾した後のペプチドを大環状化した。大環状化するペプチドには、N末端をAMBFとし、dRSにWOHを入れた。AMBFとWOHによる大環状化反応を図5Aに示す。
大環状化する場合も、まずペプチドをコードするDNAを用意し、[3]の方法で転写、翻訳した後、LS融合PatDと反応させた。LS融合PatDの終濃度は6μM、反応温度は25℃、反応時間は16時間とした。その後、Sephadex G-10を用いた脱塩カラムにて、溶液条件を167 mM ホウ酸-K(pH 9.0), 100 mM NaClへと変換した。その後、K3Fe(CN)6を加え、終濃度125 mM ホウ酸-K(pH 9.0), 75 mM NaCl, 1 mM K3Fe(CN)6、反応温度37℃の条件で30分反応させ、大環状化させた。
得られたペプチドを、[4]の方法で解析した。
結果を図5B-1及び図5B-2に示す。各基質において、Cysにアゾリン骨格が導入されたうえで、環状化されたことが確認された。st146とst149の構造を図5Cに示す。
本発明の方法によれば、基質ペプチドにリーダー配列を含める必要がないため、N末端に環状化に必要なアミノ酸を配置することができ、アゾリン骨格導入後、そのまま環状化できることが確認された。
Claims (19)
- アゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物の製造方法であって:

(Xaa2)mは、m個の任意のアミノ酸を示し、mは0から10より選択される整数を示し;
(Xaa3)nは、n個の任意のアミノ酸を示すが、少なくとも1つは、Cys、Ser、Thr、2,3-ジアミノ酸、ホモシステイン、ホモセリン、2,4-ジアミノ酸、及びこれらのアナログからなる群より選択されるアミノ酸であり、nは2から40から選択される整数を示し;
(Xaa4)oは、o個の任意のアミノ酸を示し、oは0から10より選択される整数を示す。〕
で表されるペプチドを調製する工程と、
前記ペプチドと、基質のリーダー配列又はその部分配列が結合したアゾリン骨格導入酵素とを反応させて、(Xaa3)nのCys、Ser、Thr、2,3-ジアミノ酸、ホモシステイン、ホモセリン、2,4-ジアミノ酸、及びこれらのアナログの少なくとも1つにヘテロ環を導入する工程と、を含む方法。 - 前記アゾリン骨格導入酵素には、そのN末端に、基質のリーダー配列又はその部分配列が結合している、請求項1に記載の方法。
- 前記リーダー配列又はその部分配列は、以下の配列又はその部分配列からなる、請求項1又は2に記載の方法。

- 前記ヘテロ環を導入する工程の後、ヘテロ環を含む化合物を環状化する工程をさらに含む、請求項1から10のいずれか1項に記載の方法。
- アゾール骨格導入酵素によって導入されるヘテロ環を含む化合物の製造方法であって、
請求項1から11のいずれか1項に記載の方法において、前記ヘテロ環の導入工程の後、ヘテロ環が導入されたペプチドとアゾール骨格導入酵素とを反応させ、アゾリン骨格導入酵素によって導入されたヘテロ環の少なくとも1つを、アゾール骨格導入酵素によって導入されるヘテロ環に変換する工程を含む、方法。 - 請求項1から12のいずれか1項に記載の方法で製造されたヘテロ環を含む化合物。
- 以下のいずれかのアゾリン骨格導入酵素。
(i) 配列番号:5~15のいずれか1つで表されるアミノ酸配列からなる酵素、
(ii) 配列番号:5~15のいずれか1つと80%以上の同一性を有する配列からなる、アゾリン骨格導入活性を有する酵素、及び
(iii) 配列番号:5~15のいずれか1つにおいて、1又は数個のアミノ酸が、欠失、付加又は置換した配列からなる、アゾリン骨格導入活性を有する酵素。 - アゾリン骨格導入酵素によって導入されるヘテロ環を含む化合物を2種以上含むライブラリの製造方法であって:
