Abstract
Es wird ein Verfahren zur Herstellung eines Mikrosiebs angegeben, bei dem in einem ersten Schritt ein Träger bereitgestellt wird, in einem zweiten Schritt eine Fotolackschicht mit einer festlegbaren Dicke auf einen Träger aufgebracht wird, in einem Belichtungsschritt die Fotolackschicht mittels Einwirkung von Strahlung unter Verwendung einer Maske, die die Struktur des Mikrosiebs festlegt, belichtet wird, in einem Entwicklungsschritt eine Entwicklung des Fotolacks vorgenommen wird, dadurch gekennzeichnet, dass die Dicke der Fotolackschicht so gewählt wird, dass in einen Teilbereich der Fotolackschicht die zur Belichtung verwendete Strahlung nur noch so geringfügig eindringt, dass praktisch keine Vernetzung des Fotolacks mehr stattfindet.

Beschreibung
Verfahren zur Herstellung eines Mikrosiebs Die Erfindung betrifft ein Verfahren zur Herstellung eines Mikrosiebs .
Für anspruchsvolle Trennaufgaben beispielsweise in der Medizintechnik oder Biotechnologie werden zunehmend Mikrosiebe verwendet. So kann die Anreicherung oder Extraktion bestimmter Zellen aus menschlichem Blut mittels Filtration des Bluts durch ein Mikrosieb erfolgen (Mikrofiltration) . Mikrosiebe besitzen im Unterschied zu den herkömmlichen Mikrofiltern aus schwammartigen Polymer- oder Keramikmembranen eine definierte Porengeometrie und sind daher deutlich effizienter und besser klassifizierend .
Zur Optimierung eines Filtrationsverfahrens ist eine frei wählbare Porengeometrie sowie Porendichte und -Verteilung des Mikrosiebs vorteilhaft. Im Gegensatz zu schwammartigen Filtermembranen dringen die zurückgehaltenen Partikel bei Mikro- sieben kaum oder gar nicht in die Oberfläche ein. Somit sind sie zum einen leichter mikroskopisch identifizierbar und können zum anderen leichter vom Filter abgelöst werden, wenn weitere Analysemethoden dies erfordern.
Eine Art bekannter Mikrosiebe sind die sog. Track-Etched- Membranen. Bei diesen wird eine Polymerfolie mit Schwerionen beschossen und die von den Schwerionen in der Folie hinter- lassene Spur anschließend mit einem Ätzprozess zu einer Pore erweitert. Diese Membranen weisen bedingt durch ihr Herstellungsverfahren eine räumlich unregelmäßige Porenverteilung auf. Je nach Porengröße ist die maximale Anzahl von Poren pro Flächeneinheit erheblich begrenzt. So ist es mit Track- Etched-Membranen bei einem Porendurchmesser von 8 Mikrometern beispielsweise nur möglich, einen Porenanteil an der Gesamtfläche der Membran von maximal 5% zu erreichen. Außerdem durchläuft eine Vielzahl von Poren das Grundmaterial der
Membran nicht senkrecht, sondern schräg. Ferner treten Doppelporen auf, welche eine gemeinsame Pore mit einem größeren als dem nominellen Durchmesser ergeben. Aus der WO 2011/139445 AI ist ein Verfahren zur Herstellung von Mikrosieben bekannt, bei dem ein photostrukturierbares Trockenresist in Form einer Epoxidharz-Folie mittels eines photolithographischen Prozesses zu einem Mikrosieb strukturiert wird. Nachteilig an dem bekannten Verfahren ist, dass stets ein Ablösungsschritt zur Lösung des Mikrosiebs vom bei der Herstellung verwendeten Träger erforderlich ist oder alternativ ein Schritt, bei dem das Mikrosieb in Teilbereichen hinterspült wird. In der WO 2011/139445 AI wird hierzu insbesondere ein Ätzschritt vorgeschlagen, mit dem ein Kupferträ- ger vom Mikrosieb entfernt wird.
Es ist Aufgabe der vorliegenden Erfindung, ein vereinfachtes Verfahren zur Herstellung eines Mikrosiebs anzugeben. Diese Aufgabe wird durch ein Verfahren mit den Merkmalen von An- spruch 1 gelöst. Die Unteransprüche betreffen vorteilhafte Ausgestaltungen des Verfahrens.
Bei dem erfindungsgemäßen Verfahren zur Herstellung eines Mikrosiebs wird eine Fotolackschicht mit einer festlegbaren Dicke auf einen Träger aufgebracht. Weiterhin wird eine Belichtung des Fotolacks unter Verwendung einer Maske vorgenommen, wobei die Maske die Struktur des Mikrosiebs festlegt. Schließlich wird eine Entwicklung des Fotolacks vorgenommen. Dabei wird die Dicke der Fotolackschicht so gewählt, dass in einen Teilbereich der Fotolackschicht die zur Belichtung verwendete Strahlung nur noch so geringfügig eindringt, dass praktisch keine Vernetzung des Fotolacks stattfindet. Für die Erfindung wurde erkannt, dass bei einigen negativ arbeitenden Fotolacken die Durchbelichtung dicker Fotolackschichten aufgrund von der Absorption der verwendeten Strahlung Grenzen gesetzt sind. Ein Beispiel hierfür ist der Lack
der AZ nLOF Familie: Bei diesem Fotolack liegt die Lichtintensität bei einer Wellenlänge von 365 nm nach 10 μπι Lackschichtdicke bereits unter 1 % der einfallenden Leistung. Diese Intensität reicht auch bei langen Belichtungszeiten nicht für eine ausreichende Quervernetzung aus.
Bei der Herstellung des Mikrosiebs wird dieser Effekt ausgenutzt: Vorteilhaft dient der nicht belichtete Teil in der Fotolackschicht als Opferschicht. Bei der Entwicklung der Mik- rosieb-Strukturen wird der nicht belichtete Teilbereich der Fotolackschicht mit aufgelöst. Dadurch löst sich das Mikro- sieb automatisch vom verwendeten Träger. Ein zusätzlicher Verfahrensschritt, beispielsweise ein Ätzschritt, ist unnötig und kann entfallen. Auch das Erzeugen oder Bereitstellen ei- ner Opferschicht, die eine Trennung des Mikrosiebs vom Träger ermöglicht, ist unnötig. Daneben sind fotolithographische Prozesse vorteilhaft weit verbreitet und erlauben einen hohen Substratdurchsatz und damit eine kostengünstige Fertigung. Die Maske ist insbesondere so gestaltet, dass das Mikrosieb eine Löcherstruktur erhält, wobei die Löcher einen, insbesondere gleichförmigen, Durchmesser zwischen ca. 1 Mikrometern und ca. 50 Mikrometern aufweisen. Es ist eine besonders bevorzugte Ausgestaltung, dass die Löcher einen gleichförmigen Durchmesser zwischen 5 Mikrometern und ca. 25 Mikrometern, insbesondere zwischen ca. 7 Mikrometern und ca. 15 Mikrometern, aufweisen.
Bevorzugt wird als Dicke der Fotolackschicht wenigstens die Eindringtiefe der zur Belichtung verwendeten Strahlung gewählt, wobei die Eindringtiefe hier diejenige Tiefe im
Fotolack bezeichnet, bei der die Strahlungsintensität halbiert ist. Insbesondere kann die Dicke der Fotolackschicht wenigstens doppelt so groß gewählt werden wie die Eindring- tiefe.
Vorteilhaft ist es, wenn ein Fotolack mit einer geringen Eindringtiefe verwendet wird, beispielsweise mit einer Eindringtiefe von höchstens 5 μπι, insbesondere höchstens 1 μπι. Als absolute Dicke der Fotolackschicht wird bevorzugt eine Dicke von zwischen 1 μπι und 20 μπι gewählt, in einer besonderen Ausgestaltung eine Dicke von zwischen 1 μπι und 5 μπι.
In einer weiteren vorteilhaften Ausgestaltung des Verfahrens wird die Maske so gestaltet, dass eine Verankerungsstruktur nach der Entwicklung des Fotolacks erzeugt wird, wobei die Verankerungsstruktur so gestaltet wird, dass das Mikrosieb nach der Entwicklung am Träger haftet. Dazu wird zweckmäßig neben der Gestaltung der Maske auch die Entwicklungszeit so kurz gewählt, dass die Verankerungsstrukturen nicht vom Träger gelöst werden.
In diesem Fall führt die Entfernung des nicht belichteten Teils der Fotolackschicht nicht zur vollständigen Ablösung des geformten Mikrosiebs von dem Träger. Stattdessen wird der überwiegende Teil des Mikrosiebs so geformt, dass ein Abstand zwischen dem Mikrosieb und dem Träger gewährleistet ist.
Die hergestellten Mikrosiebe sind beispielsweise für die Ana- lyse von sogenannten Circulating Tumor Cells (CTCs) verwendbar. Dabei spielen auch bei der Zellanalyse sowohl die Dicke des Mikrosiebs als auch die Durchmesser und die Periodizität der Poren eine entscheidende Rolle. Mit den beschriebenen Verfahren lassen sich Mikrosiebe für die Zellanalyse in ge- eigneter Dicke und mit einem geeigneten Porendurchmesser kostengünstig und einfach herstellen.
Das so erhaltene Mikrosieb kann insbesondere zum Trennen fester Stoffe und/oder oder zum Zurückhalten fester Stoffe aus einem Flüssigkeits- und/oder Gasstrom verwendet werden. Unter einem Mikrosieb kann also allgemein auch ein Mikrofilterele- ment verstanden werden. Das Mikrosieb kann insbesondere eine (Trenn- ) Membran sein.
Das Mikrosieb kann insbesondere eingesetzt werden zur Anreicherung oder Extraktion bestimmter Zellen aus zellhaltigen Körperflüssigkeiten, z.B. aus Blut, Urin,
Biopsieflüssigkeiten, Speichel usw., einschließlich aus Humanblut oder aus natürlichen oder künstlich erzeugten Zell- suspensionen oder Verdünnungen davon.
Ein bevorzugtes, jedoch keinesfalls einschränkendes Ausfüh- rungsbeispiel für die Erfindung wird nunmehr anhand der Figur der Zeichnung näher erläutert. Dabei sind die Merkmale schematisiert dargestellt.
Die einzige Figur zeigt ein Herstellungsverfahren zur Her- Stellung eines Mikrosiebs 120 mit einer definierten Porenverteilung und definierten Porengeometrien mit Verfahrensschritten der Fotolithographie.
In einem ersten Schritt 110 wird ein Träger 130 für das zu erstellende Mikrosieb 120 bereitgestellt. Der Träger 130 ist beispielsweise ein Silizium-Wafer oder eine Glasplatte. Auch andere im Wesentlichen glatte und gerade waferartige Träger 130 können verwendet werden. Voraussetzung ist lediglich, dass der 130 Träger für die typischen Schritte der Fotolitho- graphie geeignet, insbesondere chemisch stabil, ist.
In einem zweiten Schritt 111 wird eine Fotolackschicht 140 auf dem Träger erzeugt. Die Fotolackschicht 140 kann dabei in bekannten Herstellungsvarianten aufgebracht werden, bei- spielsweise mittels Aufschleudern (spin coating) oder Aufsprühen .
Hierbei wird besonders das Verhältnis der Dicke 141 der Fotolackschicht 140 und der optischen Dichte des verwendeten Fo- tolacks beachtet. Die Dicke 141 wird so groß gewählt, dass ein Teilbereich 142 der Fotolackschicht 140 beim nachfolgenden Belichtungsschritt 112 unbelichtet bleibt oder so geringfügig belichtet wird, dass keine ausreichende Quervernetzung
im Fotolack erfolgt. Dadurch wird erreicht, dass bei der Entwicklung der Teilbereich 142 der Fotolackschicht 140 zusammen mit dem Fotolack in den Poren 121 des Mikrosiebs 120 entfernt wird. Damit wird der gesamte Fotolack, der die Verbindung mit dem Träger 130 ausmacht und bewirkt, entfernt, sodass sich das Mikrosieb 120 vollständig vom Träger 130 löst. Die Dicke der Fotolackschicht 140 ist in der Figur stark übertrieben zur Dicke des Trägers 130 dargestellt. Beispielsweise kann als Fotolack ein Material der AZ nLOF Serie verwendet werden. Dieses Material hat eine derartige optische Dichte, dass das für die Belichtung zu verwendende Licht eine Fotolackschicht 140 mit einer Dicke 141 von 10 μπι nur noch zu etwa 1% durchdringt. Hierdurch ergibt sich ein mehrere μπι dicker Teilbereich 142, der nicht ausreichend belichtet ist. Dabei ist die Dicke des Teilbereichs 142 abhängig von der Dicke 141 der Fotolackschicht 140 und der Belichtungszeit. Ergibt sich aus der gewünschten Porengeometrie oder der gewünschten Dicke des Mikrosiebs 120 eine ver- gleichsweise große Belichtungszeit, kann die Dicke 141 der
Fotolackschicht 140 vergrößert werden, beispielsweise auf 20 μιτι, um wieder einen ausreichenden Teilbereich 142 zu erhalten . Bekanntermaßen kann vor der weiteren Bearbeitung der Fotolackschicht 140 ein Ausheizschritt, beispielsweise bei 110 °C, erfolgen.
In einem Belichtungsschritt 112 findet sodann die Belichtung des Fotolacks statt. Dazu wird der die Fotolackschicht 140 mit geeigneter Strahlung, beispielsweise mit Strahlung von 365 nm Wellenlänge, beaufschlagt. Eine geeignet gestaltete Maske 145 verhindert dabei an denjenigen Stellen, die später zu Poren 121 des Mikrosiebs 120 werden sollen, die Belich- tung . Dabei ist es möglich, ein einzelnes Mikrosieb 120 zu erstellen. Alternativ kann auch gleichzeitig eine Zusammensetzung aus einer Mehrzahl von Mikrosieben 120 hergestellt werden. Dazu ist die Maske 145 geeignet gestaltet, indem bei-
spielsweise Trennbereiche 146 zwischen den einzelnen zu erzeugenden Mikrosieben 120 ebenso wie die zu erzeugenden Poren 121 von der Maske 145 abgeschattet werden. Die im Belichtungsschritt 112 einsetzende Vernetzung des Fotolacks führt zur Bildung der eigentlichen Mikrosiebstruktur. Um eine möglichst große Genauigkeit der so erzeugten Mikrostrukturen zu gewährleisten, wird in anderen Anwendungen der Fotolithographie die Dicke der Fotolackschicht nicht allzu groß gegenüber der Eindringtiefe der Strahlung in den
Fotolack gewählt. Dadurch wird eine Vernetzung des Fotolacks in der gesamten Fotolackschicht erzwungen.
Ganz im Gegensatz dazu wird vorliegend jedoch wie bereits be- schrieben die Fotolackschicht 140 dick gegenüber der Eindringtiefe gewählt, so dass in jedem Fall ein Teilbereich 142 verbleibt, der von der Belichtung nicht mehr ausreichend betroffen ist. Hierdurch bildet der Teilbereich 142 automatisch eine Opferschicht.
Im Entwicklungsschritt 113 findet in für sich genommen bekannter Weise eine Entwicklung des Fotolacks, beispielsweise mit TMAH-Lösung, statt. Der Teilbereich 142 löst sich dabei zusammen mit den weiteren unbelichteten Teilen der Fotolack- Schicht 140 auf. Dadurch findet automatisch eine Ablösung des Mikrosiebs 120 vom Träger 130 statt. Der Vorteil dieses Prozessablaufs liegt also darin, dass keine zusätzliche Opferschicht notwendig ist und der Fotolack nicht nur zur Strukturierung des Mikrosiebes 120 dient, sondern auch gleich das Material für das eigentliche Mikrosieb 120 darstellt.
Die so erhaltene Mikrosiebstruktur kann nachfolgend beispielsweise zur Stabilisierung einer weiteren Temperaturbehandlung ausgesetzt werden. Weiterhin kann eine Auftrennung der Mikrosiebstruktur in kleinere Mikrosiebe 120 erfolgen.
In einem zweiten Ausführungsbeispiel für die Erfindung wird ein Mikrosieb 120 hergestellt, bei dem im Endergebnis die er-
zeugte Fotolackstruktur sich nicht völlig vom Träger 130 ablöst, sondern Verankerungsstrukturen verbleiben, mittels derer das Mikrosieb 120 auf dem Träger 130 verankert bleibt. Dabei wird ausgenutzt, dass bei der Entwicklung der Fotolackschicht 140 die Entwicklerflüssigkeit nur mit einer gewissen Geschwindigkeit die Strukturen des Mikrosiebs 120
hinterspülen kann und somit nicht überall gleichschnell für eine Ablösung der Fotolackschicht 140 von dem Träger 130 sor- gen kann.
Claims
1. Verfahren zur Herstellung eines Mikrosiebs (120), bei dem
- in einem ersten Schritt (110) ein Träger (130) bereitge- stellt wird,
- in einem zweiten Schritt (111) eine Fotolackschicht (140) mit einer festlegbaren Dicke (141) auf einen Träger (130) aufgebracht wird,
- in einem Belichtungsschritt (112) die Fotolackschicht (140) mittels Einwirkung von Strahlung unter Verwendung einer Maske
(145) , die die Struktur des Mikrosiebs (120) festlegt, belichtet wird,
- in einem Entwicklungsschritt (113) eine Entwicklung des Fotolacks vorgenommen wird,
dadurch gekennzeichnet, dass die Dicke (141) der Fotolackschicht (140) so gewählt wird, dass in einen Teilbereich (142) der Fotolackschicht (140) die zur Belichtung verwendete Strahlung nur noch so geringfügig eindringt, dass praktisch keine Vernetzung des Fotolacks mehr stattfindet.
2. Verfahren gemäß Anspruch 1, bei dem die Dicke (141) der Fotolackschicht (140) wenigstens so groß wie die Eindringtiefe der zur Belichtung verwendeten Strahlung gewählt wird, wobei die Eindringtiefe die Tiefe im Fotolack bezeichnet, bei der die Strahlungsintensität halbiert ist.
3. Verfahren gemäß Anspruch 2, bei dem die Dicke (141) der Fotolackschicht (140) wenigstens doppelt so groß wie die Eindringtiefe der zur Belichtung verwendeten Strahlung gewählt wird.
4. Verfahren gemäß einem der vorangehenden Ansprüche, bei dem ein Fotolack mit einer Eindringtiefe von höchstens 1 μπι verwendet wird.
5. Verfahren gemäß einem der vorangehenden Ansprüche, bei dem die Dicke (141) der Fotolackschicht (140) zwischen 1 μπι und 20 μπι, insbesondere zwischen 1 μπι und 5 μπι beträgt.
6. Verfahren gemäß einem der vorangehenden Ansprüche, bei dem im Entwicklungsschritt (113) wenigstens eine Verankerungsstruktur erzeugt wird, wobei die Verankerungsstruktur so ge- staltet wird, dass das Mikrosieb (120) nach der Entwicklung am Träger (130) haftet.
