Abstract
Description
動脈硬化は血管内膜における炎症反応の亢進や、コレステロールの蓄積により、血管壁の弾力性が低下した状態である。それにより円滑な血流の維持が困難になり、血栓が形成され易くなる。この要因の1つとして、血管内皮細胞のNO産生能の低下が多くの研究により指摘されている。このようにNOは、血管機能にとって非常に重要な調節因子となることから、血管内皮細胞のNO産生能を増強する試みにより、動脈硬化をはじめとした血管内皮機能の低下を予防または改善する研究が進められている(非特許文献1、2)。さらに近年、日常活動におけるNOの有用性が着目され、例えば、運動時に体内でのNO濃度を高めることで、運動パフォーマンスの向上や酸素消費効率の増加、および筋代謝を向上させトレーニング効率を上昇させる研究も報告されている(非特許文献3)。すなわち、血管内皮細胞のNO産生能を増強することにより、動脈硬化をはじめとした血管内皮機能の低下に起因する虚血性血管疾患を予防または改善することに加え、例えば日常の活動で、運動効率等を上昇することが期待できる。
(1) グルタチオンまたはその塩を有効成分として含有する一酸化窒素(NO)濃度上昇剤。
(2) NO濃度を上昇することにより、血管拡張または血流促進する(1)記載のNO濃度上昇剤。
(3) NO濃度を上昇することにより、血管内皮機能低下に関連する症状を予防または改善する(1)記載のNO濃度上昇剤。
(4) 血管拡張または血流促進することにより、運動パフォーマンス向上、筋代謝向上、脂肪蓄積抑制、肌質向上、記憶学習力向上、注意集中力向上、肩凝り改善、冷え症改善またはむくみ抑制する(2)記載のNO濃度上昇剤。
(5) 血管内皮機能低下に関連する症状が、脳梗塞、心筋梗塞、狭心症、末梢動脈閉塞、肺高血圧症、腎機能障害、網膜症および性機能障害から選ばれる虚血性疾患の少なくとも一つの症状である(3)記載のNO濃度上昇剤。
(6) グルタチオンまたはその塩が、還元型グルタチオン若しくはその塩、または/および酸化型グルタチオン若しくはその塩である(1)~(5)のいずれかに記載のNO濃度上昇剤。
(7) NO濃度を上昇させるための方法であって、NO濃度を上昇させる必要のある対象者に、対象者のNO濃度を上昇させるのに十分な量のグルタチオンまたはその塩を摂取させる、または投与する工程を含む、方法。
(8) NO濃度を上昇させることにより、血管拡張または血流促進をする(7)記載のNO濃度を上昇させるための方法。
(9) NO濃度を上昇させることにより、血管内皮機能低下に関連する症状を予防または改善する(7)記載のNO濃度を上昇させるための方法。
(10) 血管拡張または血流促進により、運動パフォーマンス向上、筋代謝向上、脂肪蓄積抑制、肌質向上、記憶学習力向上、注意集中力向上、肩凝り改善、冷え症改善またはむくみを抑制する(8)記載のNO濃度を上昇させるための方法。
(11) 血管内皮機能低下に関連する症状が、脳梗塞、心筋梗塞、狭心症、末梢動脈閉塞、肺高血圧症、腎機能障害、網膜症および性機能障害から選ばれる虚血性疾患の少なくとも一つの症状である(9)記載のNO濃度を上昇させるための方法。
(12) グルタチオンまたはその塩が、還元型グルタチオン若しくはその塩、または/および酸化型グルタチオン若しくはその塩である(7)~(11)のいずれかに記載のNO濃度を上昇させるための方法。
(13) 摂取または投与対象者の体内NO濃度の上昇に使用するためのグルタチオンまたはその塩。
(14) 摂取または投与対象者の体内NO濃度の上昇による血管拡張または血流促進に使用するための(13)記載のグルタチオンまたはその塩。
(15) 摂取または投与対象者の体内NO濃度の上昇による血管内皮機能低下に関連する症状の予防または改善に使用するための(13)記載のグルタチオンまたはその塩。
(16) 血管の拡張または血流の促進により、運動パフォーマンス向上、筋代謝向上、脂肪蓄積抑制、肌質向上、記憶学習力向上、注意集中力向上、肩凝り改善、冷え症改善またはむくみの抑制に使用するための(14)記載のグルタチオンまたはその塩。
(17) 血管内皮機能低下に関連する症状が、脳梗塞、心筋梗塞、狭心症、末梢動脈閉塞、肺高血圧症、腎機能障害、網膜症および性機能障害から選ばれる虚血性疾患の少なくとも一つの症状である(15)記載のグルタチオンまたはその塩。
(18) グルタチオンまたはその塩が、還元型グルタチオン若しくはその塩、または/および酸化型グルタチオン若しくはその塩である(13)~(17)のいずれかに記載のグルタチオンまたはその塩。
(19) NO濃度上昇剤の製造のためのグルタチオンまたはその塩の使用。
(20) NO濃度を上昇することにより血管拡張または血流促進するNO濃度上昇剤の製造のための(19)記載のグルタチオンまたはその塩の使用。
(21) NO濃度を上昇することにより血管内皮機能低下に関連する症状を予防または改善するNO濃度上昇剤の製造のための(19)記載のグルタチオンまたはその塩の使用。
(22) 血管を拡張または血流を促進することにより運動パフォーマンス向上、筋代謝向上、脂肪蓄積抑制、肌質向上、記憶学習力向上、注意集中力向上、肩凝り改善、冷え症改善またはむくみを抑制するNO濃度上昇剤の製造のための(20)記載のグルタチオンまたはその塩の使用。
(23) 血管内皮機能低下に関連する症状が、脳梗塞、心筋梗塞、狭心症、末梢動脈閉塞、肺高血圧症、腎機能障害、網膜症および性機能障害から選ばれる虚血性疾患の少なくとも一つの症状である(21)記載のグルタチオンまたはその塩の使用。
(24) グルタチオンまたはその塩が、還元型グルタチオン若しくはその塩、または/および酸化型グルタチオン若しくはその塩である(19)~(23)のいずれかに記載のグルタチオンまたはその塩の使用。
還元型グルタチオンとはγ-L-Glu-L-Cys-Glyの構造を有するトリペプチドを表し、酸化型グルタチオンとは還元型グルタチオン2分子がSS結合により結合したグルタチオンジペプチドを表す。本発明で用いられる還元型グルタチオンおよび酸化型グルタチオンは、どのような製造法によって得られたものであってもよい。
金属塩としては、ナトリウム塩、カリウム塩等のアルカリ金属塩、マグネシウム塩、カルシウム塩等のアルカリ土類金属塩、アルミニウム塩、亜鉛塩等があげられる。
有機アミン付加塩としては、モルホリン、ピペリジン等の塩があげられる。
アミノ酸付加塩としては、グリシン、フェニルアラニン、リジン、アスパラギン酸、グルタミン酸等の塩があげられる。
本発明のNO濃度上昇剤としては、グルタチオンまたはその塩をそのまま摂取または投与することも可能であるが、通常各種の製品形態または製剤として提供するのが望ましい。
該製品または製剤の摂取または投与形態は、体内でのNO濃度上昇に際し最も効果的なものを使用するのが望ましく、経口摂取もしくは投与または、例えば静脈内、腹膜内もしくは皮下投与等の非経口投与をあげることができるが、経口摂取または投与が好ましい。
摂取または経口投与に適当な、例えばドリンク剤のような液体調製物は、水、蔗糖、ソルビトール、果糖等の糖類、ポリエチレングリコール、プロピレングリコール等のグリコール類、ごま油、オリーブ油、大豆油等の油類、p-ヒドロキシ安息香酸エステル類等の防腐剤、パラオキシ安息香酸メチル等のパラオキシ安息香酸誘導体、安息香酸ナトリウム等の保存剤、ストロベリーフレーバー、ペパーミント等のフレーバー類などを添加して製剤化することができる。
また、経口摂取または投与に適当な製品形態または製剤は、例えば摂取期間、摂取回数、摂取量など摂取当たりの単位包装形態の錠剤、散剤、顆粒剤、丸剤、懸濁剤、乳剤、浸剤・煎剤、カプセル剤、ドリンク剤、液剤、エリキシル剤、エキス剤、チンキ剤、流エキス剤に加工製造することができる。
例えば「1回摂取当たりの単位包装形態」としては、製品形態または製剤がドリンク剤であるような場合10 mg以上のグルタチオンまたはその塩が懸濁あるいは溶解したドリンクが1回摂取あたり飲み切りの形態でビン等に入れられている形態が挙げられる。
非経口投与に適当な、例えば注射剤は、好ましくは受容者の血液と等張であるグルタチオンまたはその塩を含む滅菌水性剤からなる。例えば、注射剤の場合は、塩溶液、ブドウ糖溶液または塩溶液とブドウ糖溶液の混合物からなる担体等を用いて注射用の溶液を調製する。
本発明のNO濃度上昇剤中のグルタチオンまたはその塩の濃度は、製品形態または製剤の種類、該製品または製剤の摂取または投与により期待する効果等に応じて適宜選択されるが、グルタチオンまたはその塩として、通常は0.1~100重量%、好ましくは0.5~80重量%、特に好ましくは1~70重量%である。
本発明のNO濃度上昇剤は、生体内においてより高いNO濃度の上昇作用を介して血管内皮機能低下に関連する症状の予防または改善、または血管拡張および血流促進効果を発揮するために使用することがきる。本発明のNO濃度上昇剤は、血管内皮機能低下に関連する症状の予防または改善、または、効果的な血管拡張および血流促進効果が期待できることから、虚血性疾患の予防または改善、あるいは運動パフォーマンス向上、筋代謝向上、脂肪蓄積抑制、肌質向上、記憶学習力向上、注意集中力向上、肩凝り改善、冷え症改善、むくみ抑制に適用できる。
さらに本発明においては、NO濃度上昇剤を製造するために、グルタチオンまたはその塩を使用され得る。
さらにまた本発明は、血管内皮機能低下の予防または改善方法を包含する。本発明の方法は、血管内皮機能低下の予防または改善させる必要のある対象者に、該対象者の血管内皮機能低下の予防または改善に十分な量のグルタチオンまたはその塩を摂取させる、または投与する工程を含む。
以下に、実施例を挙げて本発明をさらに詳細に説明するが、本発明はこれらにより限定されるものではない。下記に、グルタチオンのNO濃度上昇効果を調べた試験例を示す。
試験例
8週齢雄性SD系ラットは日本SLCより購入した。イソフルラン麻酔下で、頸動脈にカテーテルを留置し、覚醒後、1週間の順化期間をおいた。ラットには固形餌(日本クレア製CE-2)と水道水を自由摂取させ、温度20 ℃~24 ℃、湿度50~60%、12時間明暗サイクルで飼育した。また、還元型グルタチオンは協和発酵バイオ株式会社製を用いた。順化後、頸動脈に留置したカテーテルよりベースラインの血液サンプルを採取した。続いて、ラットに精製水(コントロール群)、還元型グルタチオン25 mg/kg/day(グルタチオン-25 mg/kg群)、還元型グルタチオン50 mg/kg/day(グルタチオン-50 mg/kg群)を1日1回、3日間強制経口投与し、最終投与直前および最終投与後から4時間後まで経時的に、頸動脈に留置したカテーテルより採血を行った。なお、最終投与前16時間は絶食した。血液サンプルより血漿を得た後、血液中における安定NO代謝産物であるNitrite (NO2 -)およびNitrate (NO3 -) 濃度をHPLC法 (ENO-20, EICOM) により定量した。すなわち、血中のNO濃度は、これらの総和であるNOxとして表わし、各採血時点におけるベースラインからのNOx増加量を算出した。
還元型グルタチオン120 kg、環状オリゴ糖19 kg、セルロース57 kgおよびプルラン1 kgを流動層造粒乾燥機で造粒した。得られた造粒物とステアリン酸カルシウム3 kgとをコニカルブレンダーで混合した後、ロータリー圧縮成形機で圧縮成形して錠剤を製造する。
実施例1で製造する錠剤の表面をシェラック溶液でコーティングして腸溶錠剤を製造する。
還元型グルタチオン120 kg、環状オリゴ糖19 kg、セルロース57 kg、ステアリン酸カルシウム3 kgおよびプルラン1 kgを、コニカルブレンダーで混合する。得られる混合物20 kgと0.2 kgの二酸化ケイ素とを混合攪拌して得られた混合物をカプセル充填機に投入、ハードカプセルに充填してハードカプセルを得る。得られるハードカプセルの表面をツェイン溶液でコーティングして腸溶カプセルを製造する。
酸化型グルタチオン1.28 kg、エリスリトール3 kg、クエン酸0.05 kg、人工甘味料3g、香料0.06kgを液温70 ℃で水50 Lに攪拌溶解し、クエン酸でpHを3.3に調整後、プレート殺菌を用いて滅菌して瓶に充填後、パストライザー殺菌し、飲料を製造する。
酸化型グルタチオン20 mg、アルギニン20 mgと適当量の果糖ぶどう糖液糖、食塩、クエン酸、香料、クエン酸Na、乳酸Ca、ピロリン酸鉄、グルコン酸Ca、塩化K、塩化Mg、甘味料とを配合し555mLの飲料を製造する。
酸化型グルタチオン100 mg、アルギニン100 mg、アラニン2.5 mg、グリシン2.5 mg、ロイシン 2.5 mg、イソロイシン1.3 mg、バリン 1.3 mgと適当量の香料、甘味料を配合し300 mLの飲料を製造する。
エタノール10.0重量%、還元型グルタチオン2.0重量%、1,3-ブチレングリコール5.0重量%、精製水83.0重量%を配合し化粧水を製造する。
ポリエチレングリコール(PEG55)、モノステアレート2.00重量%、自己乳化型モノステアリン酸グリセリン5.00重量%、セチルアルコール4.00重量%、スクワラン6.00重量%、2-エチルヘキサン酸トリグリセリル6.00重量%、1,3-ブチレングリコール7.00重量%、L-ヒスチジン3.00重量%、還元型グルタチオン1.00重量%、精製水66.00重量%を配合しクリームを製造する。
酸化型グルタチオン3.00重量%、L-セリン1.00重量%、水溶性コラーゲン(1%水溶液)1.00重量%、クエン酸ナトリウム0.10重量%、クエン酸0.05重量%、甘草エキス0.20重量%、1,3-ブチレングリコール3.00重量%、精製水91.65重量%を配合しローションを製造する。
ポリビニールアルコール13.00重量%、L-アスパラギン酸1.00重量%、還元型グルタチオン5.00重量%、ラウロイルヒドロキシプロリン1.00重量%、水溶性コラーゲン (1%水溶液) 2.00重量%、1,3-ブチレングリコール3.00重量%、エタノール5.00重量%、精製水70.00重量%を配合しパックを製造する。
ヒドロキシエチルセルロース(2%水溶液)12.0重量%、キサンタンガム(2%水溶液) 2.0重量%、還元型グルタチオン2.0重量%、1,3-ブチレングリコール6.0重量%、濃グリセリン4.0重量%、ヒアルロン酸ナトリウム(1%水溶液)5.0重量%、精製水69.0重量%を配合し美溶液を製造する。
Claims (24)
- グルタチオンまたはその塩を有効成分として含有する一酸化窒素 (NO) 濃度上昇剤。
- NO濃度を上昇することにより、血管拡張または血流促進する請求項1記載のNO濃度上昇剤。
- NO濃度を上昇することにより、血管内皮機能低下に関連する症状を予防または改善する請求項1記載のNO濃度上昇剤。
- 血管拡張または血流促進することにより、運動パフォーマンス向上、筋代謝向上、脂肪蓄積抑制、肌質向上、記憶学習力向上、注意集中力向上、肩凝り改善、冷え症改善またはむくみを抑制する請求項2記載のNO濃度上昇剤。
- 血管内皮機能低下に関連する症状が、脳梗塞、心筋梗塞、狭心症、末梢動脈閉塞、肺高血圧症、腎機能障害、網膜症および性機能障害から選ばれる虚血性疾患の少なくとも一つの症状である請求項3記載のNO濃度上昇剤。
- グルタチオンまたはその塩が、還元型グルタチオン若しくはその塩、または/および酸化型グルタチオン若しくはその塩である請求項1~5のいずれか一項に記載のNO濃度上昇剤。
- NO濃度を上昇させるための方法であって、NO濃度を上昇させる必要のある対象者に、対象者のNO濃度を上昇させるのに十分な量のグルタチオンまたはその塩を摂取させる、または投与する工程を含む、方法。
- NO濃度を上昇させることにより、血管拡張または血流促進をする請求項7記載のNO濃度を上昇させるための方法。

- 摂取または投与対象者の体内NO濃度の上昇に使用するためのグルタチオンまたはその塩。

- 血管内皮機能低下に関連する症状が、脳梗塞、心筋梗塞、狭心症、末梢動脈閉塞、肺高血圧症、腎機能障害、網膜症および性機能障害から選ばれる虚血性疾患の少なくとも一つの症状である請求項21記載のグルタチオンまたはその塩の使用。
