Abstract

 生体分子の機能を維持しながら標識化反応の進行を早め、標識化剤の減衰を考慮した混合時間を選定できる反応容器、当該反応容器を使用した反応装置を提供する。 生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応容器において、反応容器本体と、当該反応容器本体の表面側に対向して設けた蓋部材とを有し、前記反応容器本体の表面には流路が形成されており、前記流路の中間部に、前記第2の化合物の溶液の溶媒を除去して固化する標識化剤固化部が形成されており、前記標識化剤固化部の上流側に前記第1の化合物の供給部及び前記第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に前記混合物の排出部が形成されている。反応装置は、前記反応容器に前記第1の化合物と前記第2の化合物をそれぞれ供給するとともに、前記固化部で固化した第2の化合物の上部に、前記第1の化合物の溶液を往復して送液する送液ユニットを備える。

Description

生体分子標識化反応容器、それを使用する反応装置及び反応方法
 本発明は、生体分子の標識化に使用する反応容器、当該反応容器を使用する反応装置及び反応方法に関する。
 核医学診断(Diagnostic Nuclear Medicine)では、放射性薬剤(放射性物質を含む薬剤)を生体内に投与し、生体機能を反映した画像をPET(Positron Emission Tomography)、ガンマカメラなどを通じて取得する。
 放射性薬剤として、疾患関連物質と相互作用する生体分子を放射性物質で標識化した薬剤は、疾患を特異的に検出する薬剤として期待され、新規薬剤の研究開発が活発に行われている(例えば非特許文献1、非特許文献2、を参照)。
 生体分子の疾患関連物質と相互作用する機能を維持するためには、放射性物質での標識化反応は穏やかな条件で行われることが望ましい。例えば、代表的なPET薬剤である18F標識化物を合成する際には、通常100℃前後の高温で18F標識化工程が行われる(例えば非特許文献3を参照)。しかし、生体分子では高温での機能の失活が懸念されるため、この工程での直接の標識化は行わず、18Fで標識化した18F標識化剤を用いて室温前後で反応させる方法が主流となっている(例えば非特許文献1、非特許文献2、を参照)。
 また、18F標識化剤の多くは非水溶性の化合物であり、反応溶液は生体分子の機能の失活が懸念される、アセトニトリル、エーテル等の有機溶剤溶液となる。そこで、18F標識化剤を反応容器に導入し、溶媒を蒸発乾固させてから、生体分子水溶液を添加し、これもまた、生体分子の機能の失活が懸念される撹拌は行わずに一定時間静置するという製造方法が採用されている(例えば非特許文献1、非特許文献2、を参照)。

 前述したように、非特許文献1、非特許文献2の製造方法では、室温で、有機溶剤を使わず、反応溶液の撹拌も行わないため、生体分子の機能を保持できる可能性は高いが、18F標識化剤は非水溶性の化合物であるため、水に溶解しにくく、反応の進行が遅いという課題がある。
 放射性薬剤の放射能は半減期を有する。特にPET用薬剤の核種の半減期は11C:20分、18F:110分と短い。上記のように反応の進行が遅い場合は、通常の化学反応では反応時間を延長することで、目的物の収率を向上させることができるが、放射性薬剤の反応では反応時間を延長させると放射能が減衰し、放射化学的な収率が低下するという課題がある。
 そこで、本発明は、生体分子の機能を維持する穏やかな反応条件でありながら、反応の進行を早め、また、放射能の減衰を考慮した反応時間を選定できる反応容器、当該容器を使用する反応装置及び反応方法の提供を目的とする。
 前述した課題を解決するために、複数の課題を解決する手段を含んでいるが、本発明の生体分子標識化反応容器の1例を挙げるならば、生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応容器において、反応容器本体と、当該反応容器本体の表面側に対向して設けた蓋部材とを有し、前記反応容器本体の裏面側に前記第1の化合物及び前記第2の化合物の導入部並びに混合物の回収部を有し、前記反応容器本体の表面には流路が形成されており、前記流路の中間部に、前記第2の化合物の溶液の溶媒を除去して固化する標識化剤固化部が形成されており、前記流路において、前記標識化剤固化部の上流側に前記第1の化合物の供給部及び前記第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に前記混合物の排出部が形成されているものである。
 本発明の生体分子標識化反応容器において、前記標識化剤は、放射性標識化剤であり、放射線センサを備えているのが好ましい。
 また、本発明の反応装置の1例を挙げるならば、反応容器内において生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応装置であって、反応容器本体の表面には流路が形成されており、前記流路の中間部に、前記第2の化合物の溶液の溶媒を除去して固化する標識化剤固化部が形成されており、前記標識化剤固化部の上流側に前記第1の化合物の供給部及び前記第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に前記混合物の排出部が形成されている反応容器を含む反応容器ユニットと、前記反応容器に前記第1の化合物と前記第2の化合物をそれぞれ供給するとともに、前記標識化剤固化部で固化した前記第2の化合物の上部に、前記第1の化合物の溶液を往復送液する送液ユニットと、前記反応容器で混合された混合溶液を回収する回収ユニットと、前記反応容器ユニット、前記送液ユニット及び前記回収ユニットを制御する制御ユニットと、を有するものである。
 また、本発明の反応方法の1例を挙げるならば、反応容器本体の表面に流路が形成されており、前記流路の中間部に標識化剤固化部が形成されており、前記標識化剤固化部の上流側に第1の化合物の供給部及び第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に混合物の排出部が形成されている反応容器を使用して、生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応方法において、前記標識化剤固化部に、前記第2の化合物の供給部から前記標識化剤である第2の化合物の溶液を導入し、溶媒を除去し、固化する処理と、前記第1の化合物の供給部から生体分子である第1の化合物の溶液を導入する処理と、前記固化した第2の化合物の上部に、前記第1の化合物の溶液を往復して通過させ、前記第1の化合物と前記第2の化合物を混合する処理と、を有するものである。
 本発明によれば、生体分子の機能を維持する穏やかな反応条件でありながら、反応の進行を早めることができる。また、放射能の減衰を考慮した反応時間を選定することができる。

  本発明に係る反応容器を生体分子放射性標識化反応に応用する場合について説明する。図1は、生体分子放射性標識化反応容器を用いた生体分子放射性標識化反応装置の実施例であり、当該反応装置により放射性生体分子薬剤が製造される。もっとも、本発明の用途は、放射性生体分子薬剤の製造に限らない。
 図1に示す生体分子放射性標識化反応装置1は、送液ユニット101、反応容器ユニット201、回収ユニット301、排気ユニット401、温度調節ユニット501、放射線検出ユニット601、制御ユニット701から構成されている。
 送液ユニット101は、生体分子溶液121、放射性標識化剤溶液122及び混合物溶液304の送液に用いられるユニットである。
 反応容器ユニット201は、生体分子放射性標識化反応容器10を搭載するユニットであり、生体分子溶液121及び放射性標識化剤溶液122を反応させて放射性生体分子薬剤を生成するために用いられる。ここでの生体分子放射性標識化反応容器10が、本発明における反応容器に相当する。
 回収ユニット301は、生体分子放射性標識化反応容器10で混合された溶液(混合物溶液)304の回収に用いられるユニットである。排気ユニット401は、生体分子放射性標識化反応容器10内に導入した放射性標識化剤溶液122の溶媒を気化して排気するユニットである。温度調節ユニット501は、生体分子放射性標識化反応容器10の温度を管理するユニットである。放射線検出ユニット601は、反応容器ユニット201の放射線量を測定するユニットである。制御ユニット701は、送液ユニット101、反応容器ユニット201、回収ユニット301、排気ユニット401、温度調節ユニット501、放射線検出ユニット601を制御するユニットである。制御ユニット701は例えばコンピュータで構成される。
 [各ユニットの詳細構造]
  送液ユニット101は、(1)その内部に溶液の吸引・送液・廃液・閉鎖操作を切り替えるための送液切り替えバルブ102、103、(2)溶液吸引ライン104、(3)溶液廃液ライン105、(4)シリンジ106、(5)シリンジポンプ107、(6)生体分子溶液導入部108、(7)放射性標識化剤溶液導入部109を有している。この他、送液ユニット101には、不図示のデバイス、例えば(1)系内の圧力を監視する圧力センサ、(2)シリンジを固定するためのホルダ、(3)電源スイッチ、(4)異常動作を起こした場合の非常停止スイッチ、(5)通信用コネクタ、(6)小量の原料を導入するサンプルループ、(7)各ラインやサンプルループを切り替えバルブ102、103に接続するフィッティング等を含んでいる。
 反応容器ユニット201は、(1)生体分子放射性標識化反応容器10、(2)放射線センサ202、(3)滞留部203を含んでいる。なお、生体分子放射性標識化反応容器10と生体分子溶液導入部108の間、生体分子放射性標識化反応容器10と放射性標識化剤溶液導入部109の間、生体分子放射性標識化反応容器10と滞留部203の間は、不図示のフィッティングにより接続されている。
 回収ユニット301は、(1)送液・閉鎖操作を切り替えるための回収切り替えバルブ302、(2)混合物溶液回収ライン303を含んでいる。なお、回収切り替えバルブ302と滞留部203の間、回収切り替えバルブ302と混合物溶液回収ライン303の間は、不図示のフィッティングにより接続されている。
 排気ユニット401は、(1)排気・送気・閉鎖操作を切り替えるための排気切り替えバルブ402、(2)排気ライン403を含んでいる。なお、排気切り替えバルブ402と排気ライン403の間は、不図示のフィッティングにより接続されている。
 温度調節ユニット501は、反応容器ユニット201との間において、温度制御信号131Aとフィードバック信号131Bを送受する。これらの信号の送受により、生体分子放射性標識化反応容器10の温度の制御が可能となる。生体分子放射性標識化反応容器10の温度を調節する方法には、例えば循環恒温槽を用いて熱媒体を循環させる方法、ぺルチェ素子、リボンヒーター、プレート型ヒーターを用いる方法などがある。また、温度の制御対象には、例えば生体分子放射性標識化反応容器10の周辺を循環する熱媒体、生体分子放射性標識化反応容器10の外側や内部などが挙げられる。例えば生体分子放射性標識化反応容器10の内部を流れる溶液又は、内部に導入された溶液、内部を流れている溶液に近い場所を温度の制御対象とすることにより、より精密な温度制御が可能となる。
 放射線検出ユニット601は、反応容器ユニット201から放射線検出信号132Aを受信し、送液ユニット101および回収ユニット301へフィードバック信号132B、132Cを送信する。放射線検出ユニット601は、放射線センサ202から放射線検出信号132Aとして与えられる放射線量の検出値により、放射性標識化反応の進行を判定し、その判定結果に基づいて、送液切り替えバルブ102の吸引、送液を切り替えるフィードバック信号132B、回収切り替えバルブ302の閉鎖、回収を切り替えるフィードバック信号132Cを出力する。この切り替えにより反応の進行を把握した目的物の回収が実現可能となる。反応容器ユニット201内における放射線センサ202は、目的とする放射線を検出するのに可能な限り生体分子放射性標識化反応容器10内の流路近くに形成される。また目的とする放射線の検出が可能な範囲で、目的としない放射線が遮蔽される。この構造により、より精密な放射線検出が可能となる。
 制御ユニット701は、前述した6つのユニットの動作を監視・制御する。例えば制御ユニット701は、送液ユニット101との間で、制御信号141A及びフィードバック信号141Bを送受し、送液ユニット101内の動作の監視と制御を実行する。なお、制御ユニット701と他の5つのユニットとの間の通信は、送液ユニット101が中継する。このため、送液ユニット101と反応容器ユニット201との間においてはデータ通信信号142が送受され、送液ユニット101と回収ユニット301との間においてはデータ通信信号143が送受され、送液ユニット101と排気ユニット401との間においてはデータ通信信号144が送受され、送液ユニット101と温度調節ユニット501との間においてはデータ通信信号145が送受され、送液ユニット101と放射線検出ユニット601との間においてはデータ通信信号146が送受される。制御ユニット701は、これらのデータ通信信号を通じ、反応容器ユニット201、回収ユニット301、排気ユニット401、温度調節ユニット501及び放射線検出ユニット601を監視・制御する。
 具体的には、制御ユニット701は、送液ユニット101内の切り替えバルブ102及び103の切り替え制御、シリンジポンプ107の駆動制御によるシリンジ106内への溶液の吸引と送液の制御、シリンジ106内に充填された溶液の不図示の廃液タンク等への廃棄の制御を実行する。また、制御ユニット701は、送液制御や吸引制御の途中停止やその再開も制御する。
 制御ユニット701を用いれば、各シリンジ106のサイズ、溶液吸引ライン104からの溶液の吸引量と吸引速度、反応容器ユニット201への溶液の送液量と送液速度、溶液廃液ライン105への溶液の送液量と送液速度、生体分子放射性標識化反応容器10内での溶液の往復送液及び、生体分子放射性標識化反応容器10の温度設定、つまり溶媒除去温度や、反応温度の設定等を行うことができる。また、送液の「時間遅れ」を設定し、シリンジ106毎に、その送液時間を変更することもできる。
 更に、吸引・送液過程に伴うシリンジ106の動作やバルブの動作に関連する、2つ以上の連続させたい動作を指示する入力ファイルを事前に作成して制御ユニット701に読み込んでおけば、一連の動作の自動制御を実行させることもできる。自動制御機能の搭載により、放射線の被曝を回避及び遠隔自動操作が可能となる。また、前述した入力ファイルを制御ユニット701内の記憶領域に保存し、必要に応じて読み込んで動作させるようにすれば、当該入力ファイルの適宜書き換えも可能である。
 また、制御ユニット701は、送液ユニット101に設置された不図示の圧力センサから得られる系内の圧力データ、温度調節ユニット501から得られる温度情報、放射線検出ユニット601から得られる放射線の検出値や時間データ等を、その内部の記憶領域に記録することができる。また、制御ユニット701は、圧力センサや切り替えバルブなどの耐圧情報に基づいて系内の圧力に対する閾値を予め決定しておけば、系内の圧力が当該閾値を超えた場合に、装置全体を非常停止することもできる。
 ここで、前述した溶液吸引ライン104、溶液廃液ライン105、生体分子溶液導入部108、放射性標識化剤溶液導入部109、滞留部203、混合物溶液回収ライン303、生体分子放射性標識化反応容器10などの材質は、実行する反応に悪影響を与えないもの、送液溶液中の物質が吸着しないものであればよく、また、その内部を流れる溶液の温度や濃度、物性に応じ、適宜、変更することも可能である。かかる材料としては、例えばタングステン、ステンレス、シリコン、ガラス、ハステロイ、シリコン樹脂、フッ素系樹脂などを挙げることができる。また、かかる材料には、グラスライニング、ステンレス、シリコンなどの表面にニッケルや金などのコーティングをしたもの、シリコンの表面を酸化させたものなど、所謂、耐食性を向上させたもの、吸着性を低減させたものも用いることができる。
 続いて、生体分子放射性標識化反応容器10の構造を詳細に説明する。図2に、実施例1の生体分子放射性標識化反応容器10の展開斜視図を示す。
 図2に示すように、生体分子放射性標識化反応容器10は、数mm厚のPEEK板材から形成した反応容器本体20と、この反応容器本体20の上面側に配置するPEEK板材から形成した蓋部材30と、この蓋部材30の上面側に配置するSUS316ステンレス鋼板材から形成した蓋部材40と、反応容器本体20の下面側に配置するPEEK板材から形成したアダプタ部材50と、アダプタ部材50の下面側に配置するSUS316ステンレス鋼板材から形成したアダプタ部材60とを有している。
 生体分子放射性標識化反応容器10は、これらアダプタ部材60、アダプタ部材50、反応容器本体20、蓋部材30、蓋部材40を積層し、それらの周縁部を不図示のネジで締結することにより構成される。
 ここで、蓋部材30は、反応容器本体20に表面側(図中上面)に形成される上方に開いた流路の天井部分を構成する。蓋部材30のうち反応容器本体20との対向面(図中下面)には、反応容器本体20から溶媒を気化して排気するための出口ポート部31が形成されており、それぞれ、蓋部材30の上側面へ連通している。
 蓋部材40のうち蓋部材30との対向面(図中下面)には、反応容器本体20から溶媒を気化して排気するための出口部41が形成されている。出口部41は、蓋部材40の表面(図中上面)に形成された気体排出口42に連通している。
 アダプタ部材50のうち反応容器本体20との対向面(図中上面)には、反応容器本体20に生体分子溶液や放射性標識化剤溶液を導入するための入口ポート部51及び混合物溶液を排出する出口ポート部52が形成されており、それぞれ、アダプタ部材50の下側面へ連通している。
 アダプタ部材60のうちアダプタ部材50との対向面(図中上面)には、反応容器本体20に生体分子溶液や放射性標識化剤溶液を導入するための入口部61及び混合物溶液を排出する出口部62が形成されている。入口部61は、アダプタ部材60の裏面(図中下面)に形成された生体分子溶液導入口63、放射性標識化剤溶液導入口64に連通している。出口部62は、アダプタ部材60の裏面(図中下面)に形成された混合物溶液排出口65に連通している。
 なお、反応容器本体20に形成された流路等の外周部と、蓋部材30に形成された出口ポート部31の外周部と、アダプタ部材50に形成された入口ポート部51及び出口ポート部52の外周部には、フッ素ゴム製のOリング等からなる不図示のシール部材が設置されている。
 本実施例では、反応容器本体20から溶媒を気化して排気するために、蓋部材30に出口ポート部31、蓋部材40に出口部41が形成されているが、これらを形成せず、アダプタ部材50に形成された出口ポート部52、アダプタ部材60に形成された出口部62、混合物溶液排出口65を用いてもよい。
 本実施例の場合、蓋部材30と蓋部材40、アダプタ部材50とアダプタ部材60を、それぞれ別材質で2つずつ設置しているが、これは、反応容器本体20に形成した流路を安定に維持するためであり、SUS316ステンレス鋼板材と、反応容器本体20を形成するPEEK板材の硬度の違いを考慮したからである。
 このため、SUS316ステンレス鋼板材の蓋部材40と反応容器本体20の間に反応容器本体20と同じPEEK材質の蓋部材30を設置し、アダプタ部材60と反応容器本体20の間に、反応容器本体20と同じPEEK材質のアダプタ部材50が設置されている。
 反応容器本体20がSUS316ステンレス鋼板材と硬度が近い材質から形成されている場合には、蓋部材30、40とアダプタ部材50、60は、SUS316ステンレス鋼板材から形成される1部材ずつで構成してもよい。
 本実施例では、蓋部材40とアダプタ部材60を、SUS316ステンレス鋼板材で形成しているが、反応容器本体20内に導入する放射性標識化剤の放射線を遮蔽できる材質で形成してもよい。かかる材質としては、タングステン、鉛、遮蔽用加工物等があげられる。このような放射線を遮蔽する材質を用いる場合は、放射線検出用の空間を蓋部材40とアダプタ部材60のいずれか、あるいはどちらか片方に形成する必要がある。
 本実施例では、反応容器本体20をPEEK板材で形成しているが、反応容器本体20で行われる反応に悪影響を与えないもの、反応容器本体20に導入する溶液中の物質が吸着しないものであればよく、反応の種類に応じて、適宜、変更することもできる。かかる材質としては、例えばステンレス、シリコン、金、ガラス、ハステロイ、シリコン樹脂、フッ素系樹脂などを挙げることができる。また、グラスライニング、金属の表面にニッケルや金などのコーティングをしたもの、シリコンの表面を酸化させたものなど、いわゆる、耐食性を向上させたもの、吸着性を低減させたものを用いてもよい。
 上述したシール部材(ただし図示せず)の材質も、反応に悪影響を与えないものであればよく、実行する反応の種類に応じ、適宜、変更することもできる。例えばシリコン樹脂、フッ素系樹脂などを用いることができる。
 本実施例では、シール部材にフッ素ゴムを用いて分解可能な組立式ユニットとしている。分解可能な反応容器では、その内部に閉塞等が生じた場合に分解して洗浄できるため、メンテナンス性が高い。図2においては、アダプタ部材60、アダプタ部材50、反応容器本体20、蓋部材30、蓋部材40の外周部に、10個の穴又はねじ孔を形成し、ねじ締結を容易にしている。なお、レーザー接合や接着剤など他の方法を用い、反応容器本体20の表裏に蓋部材30やアダプタ部材50を直接固定して分解不可能な反応容器としてもよい。分解不可能な反応容器では、Oリングの破損等によるシールの不具合による、導入溶液等の漏れを回避できる。また、臨床に適用する薬剤を製造する反応に用いる場合には、ディスポーザブルとすることにより、薬剤の品質を確保できる。
 図3に、生体分子放射性標識化反応容器10を構成する反応容器本体20の正面図及び裏面図を示す。図4に、反応容器本体20に形成された送液構造の部分拡大図を示す。
 図3に示すように、反応容器本体20の周縁部には縦方向に4個、横方向に3個の締結用から穴が形成されている。この穴よりも中心側には、角部が丸く形成された長方形の溝29が設けられている。溝29には不図示のシール部材が装着される。
 図3の(a)に示す反応容器本体20の表面側には、溝29よりも中心側に溶媒を除去し、固化させた放射性標識化剤と生体分子溶液とを混合するための流路21が形成されている。図4に、当該流路の部分拡大図を示す。
 本実施例における流路21は、上流側から順番に、生体分子溶液供給部22と、生体分子溶液誘導流路部23と、放射性標識化剤溶液供給部24と、漏斗状の放射性標識化剤固化部25と、排出側流路部26と、混合溶液排出部27とで構成されている。
 生体分子溶液誘導流路部23は、それぞれが一定幅を有し、かつ、幅方向に一定幅ずつ離れて形成された31本の流路(第1の流路)の集合体として構成される。生体分子溶液誘導流路部23を構成する各流路は、それぞれが平行に形成された2つの壁で挟まれた空間として構成される。31本の流路の両端は開口端として形成されている。31本の流路の最上流部には、生体分子溶液121を反応容器本体20の裏面側から導入する生体分子溶液供給部22が形成されている。生体分子溶液供給部22から導入された生体分子溶液121は、図中の下端側に位置する流出口まで帯状の流れとして誘導される。生体分子溶液誘導流路部23の流出口は、放射性標識化剤固化部25の入口側開口に接続されている。
 ここで、生体分子溶液供給部22は、反応容器本体20の表面と裏面を連結する31個の開口(生体分子溶液供給ノズル22A)により形成されている。図4に示すように、生体分子溶液供給ノズル22Aは、反応容器本体20の幅方向に、ノズルの表面直径とほぼ同じ長さ間隔で1列に形成される。この生体分子溶液供給ノズル22Aを通じ、生体分子溶液121が生体分子溶液誘導流路部23に導入される。
 なお、反応容器本体20の裏面には、図3の(b)に示すように、供給液である生体分子溶液121を一時的に溜める生体分子溶液溜部28aが形成されており、生体分子溶液溜部28aの領域内に生体分子溶液供給ノズル22Aの裏面側の開口が位置している。因みに、生体分子溶液溜部28aは、反応容器本体20の裏面に対して厚み方向に窪んだ凹部として形成される。この生体分子溶液溜部28aは、アダプタ部材50の入口ポート部51と連結されている。
 放射性標識化剤溶液供給部24は、生体分子溶液誘導流路部23を構成する31本の流路で挟まれた空間内に配置され、放射性標識化剤溶液122の放射性標識化剤固化部25への導入に用いられる。図4に示すように、生体分子溶液誘導流路部23を平行な31本の流路として形成する場合、各流路の間には、生体分子溶液誘導流路部23と同じ幅を有し、かつ、幅方向に一定幅ずつ離れた30本の流路(第2の流路)が形成される。本実施例では、これら30本の流路の最下流位置に放射性標識化剤溶液供給部24を配置する。
 放射性標識化剤溶液供給部24が配置される第2の流路の流出口は、放射性標識化剤固化部25の入口側開口に接続されている。ここで、放射性標識化剤溶液供給部24は、反応容器本体20の表面と裏面を連結する30個の開口(放射性標識化剤溶液供給ノズル24A)により形成されている。図4に示すように、放射性標識化剤溶液供給ノズル24Aは、反応容器本体20の幅方向に、ノズルの表面直径とほぼ同じ長さ間隔で1列に形成される。この放射性標識化剤溶液供給ノズル24Aを通じ、放射性標識化剤溶液122が放射性標識化剤固化部25に導入される。
 なお、反応容器本体20の裏面には、図3の(b)に示すように、供給液である放射性標識化剤溶液122を一時的に溜める放射性標識化剤溶液溜部28bが形成されており、放射性標識化剤溶液溜部28bの領域内に放射性標識化剤溶液供給ノズル24Aの裏面側の開口が位置している。因みに、放射性標識化剤溶液溜部28bは、反応容器本体20の裏面に対して厚み方向に窪んだ凹部として形成される。この放射性標識化剤溶液溜部28bは、アダプタ部材50の入口ポート部51と連結されている。
 放射性標識化剤固化部25は、反応容器本体20の表面に対して厚み方向に窪んだ凹部として形成される。その底面は、生体分子溶液誘導流路部23よりも低く形成される。これにより、導入した放射性標識化剤溶液122をその溶媒を除去して固化する工程中溜めおくことが可能となる。なお、図2に示すように、蓋部材30の反応容器本体20との対向面には、放射性標識化剤溶液122の溶媒を気化して除去する際の、排気出口ポート部31が、放射性標識化剤固化部25の流入側及び流出側より放射性標識化剤溶液が流出しない程度離れた位置に形成される。また、出口ポート部31は、蓋部材40の出口部41に連結されている。これにより、反応容器本体20の放射性標識化剤固化部25に導入した放射性標識化剤溶液122の溶媒は、蓋部材40の表面(図中上面)側に形成された排気口42から排気される。前述の放射性標識化剤固化部25の底面は蓋部材30の排気出口ポート部31から放射性標識化剤溶液122が流出しない程度の深さを有する。排気出口ポート部31の反応容器本体20側には液体は通さず、気体のみを通過させるフィルターを設置してもよい。このフィルターにより、放射性標識化剤溶液122が排気出口ポート部31に達しても溶液の流出を防ぐ事が可能となる。
 また、放射性標識化剤固化部25は、流入側の流路幅が最も広く、流出側ほどその流路幅が狭くなるように漏斗形状に製造されている。放射性標識化剤固化部25の流出口は、その口幅と同じ流路幅を有する所定長の排出側流路部26に接続される。このため、図4の場合には、放射性標識化剤固化部25に導入された31本の帯状の流れで形成される多層流を幅方向に収縮した状態で排出側流路部26に導入することができる。すなわち、排出側流路部26の流路幅がある程度太くても、その内部に形成される層流の幅を狭くすることが可能となる。このように、固化した放射性標識化剤上を通過した生体分子溶液121を多数の層流として排出側流路部26に導入することにより、送液量を確保しながらも安定した混合が可能となる。
 なお、流路21において反応が進行した混合物溶液は混合物溶液排出部27へと導かれる。混合溶液排出部27は、図3の(a)及び(b)に示すように、反応容器本体20の表面から裏面まで連結する孔として形成されている。従って、混合物溶液は、反応容器本体20の表面から裏面へと導出される。なお、図2に示すように、この反応容器本体20の裏面側の混合溶液排出部27は、アダプタ部材50の出口ポート部52と連結されている。また、出口ポート部52は、アダプタ部材60の出口部62に連結されている。これにより、反応容器本体20の流路において生成された混合物溶液は、アダプタ部材60の裏面(図中下面)側に形成された混合物溶液排出口65から排出される。
 本実施例では、実施例1の反応容器本体20に形成された第2の送液構造を説明する。図5に、生体分子放射性標識化反応容器10を構成する反応容器本体20の正面図及び反応容器本体20に形成された送液構造の部分拡大図を示す。
 本実施例における反応容器本体20の構成は、特に明示しない限り実施例1と同様である。
 本実施例では生体分子溶液供給部22中の生体分子溶液供給ノズル22Aと放射性標識化剤溶液供給部24中の放射性標識化剤溶液供給ノズル24Aは同一流路中に形成されている。生体分子溶液誘導流路部23の流路数は61本となり、生体分子溶液121は放射性標識化剤固化部25に、実施例1の約2倍の層流として供給される。これにより、生体分子溶液121と固化した放射性標識化剤との相互作用が増加し、速やかな反応の進行を実現する。
 なお、放射性標識化剤溶液供給ノズル24Aの表面直径は、各ノズルが形成される流路上流から流れてくる生体分子溶液121の送液を阻害しない大きさで形成される。
 本実施例では、実施例1の反応容器本体20に形成された第3の送液構造を説明する。図6に、生体分子放射性標識化反応容器10を構成する反応容器本体20の正面図及び裏面図、反応容器本体20に形成された送液構造の部分拡大図を示す。
 本実施例における反応容器本体20の構成は、特に明示しない限り実施例1と同様である。
 本実施例における流路21は、一定幅を有し、平行に形成された2つの壁で挟まれ、中間部に放射性標識化剤固化部25が形成された連続した空間として構成される。流路の両端は開口端として形成されている。流路21の最上流部には、生体分子溶液121を反応容器本体20の裏面側から導入する生体分子溶液供給部22が形成されている。生体分子溶液供給部22から導入された生体分子溶液121は、放射性標識化剤固化部25を経由して図中の下端側に位置する混合溶液排出部27まで誘導される。
 ここで、生体分子溶液供給部22は、反応容器本体20の表面と裏面を連結する開口(生体分子溶液供給ノズル22A)により形成されている。この生体分子溶液供給ノズル22Aを通じ、生体分子溶液121が生体分子溶液誘導流路部23に導入される。生体分子溶液供給ノズル22Aの反応容器本体20裏面開口はアダプタ部材50の入口ポート部51と連結されている。
 放射性標識化剤溶液供給部24は、放射性標識化剤固化部25の上流側に配置され、放射性標識化剤溶液122の放射性標識化剤固化部25への導入に用いられる。放射性標識化剤溶液供給部24は、反応容器本体20の表面と裏面を連結する開口(放射性標識化剤溶液供給ノズル24A)により形成されている。図6に示すように、放射性標識化剤溶液供給ノズル24Aの表面直径は、生体分子溶液誘導流路部23、放射性標識化剤固化部25、排出側流路部26と続く流路21の流路幅よりも小さく形成されている。その大きさは生体分子溶液誘導流路部23から導入される生体分子溶液121の送液に支障を与えない大きさで形成される。この放射性標識化剤溶液供給ノズル24Aを通じ、放射性標識化剤溶液122が放射性標識化剤固化部25に導入される。
 放射性標識化剤溶液供給ノズル24Aは、反応容器本体20の裏面に連結されており、裏面側開口部は、アダプタ部材50の入口ポート部51と連結されている。
 放射性標識化剤固化部25は、流路21の中間部に形成した流路である。その底面は、生体分子溶液誘導流路部23、排出側流路部26よりも低く形成されてもよい。また、本実施例では、放射性標識化剤固化部25は直線で形成されているが、放射性標識化剤固化部25に導入される放射性標識化剤溶液122の容量に応じて、蛇行したり、渦巻き状に形成されてもよい。これらにより、導入した放射性標識化剤溶液122をその溶媒を除去して固化する工程中溜めおく容量を増やす事できる。
 本実施例における反応容器本体20表面に形成されている、生体分子溶液誘導流路部23から放射性標識化剤固化部25、排出側流路部26まで続く流路21の流路幅と深さは、放射性標識化剤固化部25で固化した放射性標識化剤で、生体分子溶液121の送液が阻害されない程度の大きさであればよい。小さければ小さいほど同一容量での表面積が大きくなり、生体分子溶液121と固化した放射性標識化剤との相互作用が増加し、速やかな反応の進行を実現する。また流路幅と深さが1mm以下となれば、反応時間低減、反応温度の制御能力向上等の所謂マイクロリアクタの効果が得られる。
 本実施例では、実施例1の反応容器本体20に形成された第4の送液構造を説明する。図7に、生体分子放射性標識化反応容器10を構成する反応容器本体20の正面図及び裏面図、反応容器本体20に形成された送液構造の部分拡大図を示す。
 本実施例における反応容器本体20の構成は、特に明示しない限り実施例1と同様である。
 本実施例では放射性標識化剤溶液供給部24が、実施例2における生体分子溶液誘導流路部23の途中に形成される。放射性標識化剤溶液供給部24以降の生体分子溶液誘導流路部23を本実施例における放射性標識化剤固化部25’、実施例2における漏斗状の放射性標識化剤固化部25と排出側流路部26を併せて本実施例における排出側流路部26’とする。
 本実施例における放射性標識化剤固化部25’の底面は、生体分子溶液誘導流路部23、排出側流路部26’よりも低く形成されてもよい。これにより、導入した放射性標識化剤溶液122をその溶媒を除去して固化する工程中溜めおく容量を増やす事ができる。
 本実施例における反応容器本体20表面に形成されている、生体分子溶液誘導流路部23から放射性標識化剤固化部25’まで続く流路幅と深さは、放射性標識化剤固化部25’で固化した放射性標識化剤で、生体分子溶液121の送液が阻害されない程度の大きさであれば構わない。小さければ小さいほど同一容量での表面積が大きくなり、生体分子溶液121と固化した放射性標識化剤との相互作用が増加し、速やかな反応の進行を実現する。また流路幅と深さが1mm以下となれば、反応時間低減、反応温度の制御能力向上等の所謂マイクロリアクタの効果が得られる。
 また、本実施例では固化した放射性標識化剤と生体分子溶液121が相互作用する放射性標識化剤固化部25’が並列の複数流路となるため、実施例3の性能を維持したまま、一定時間での処理量の増加が実現できる。
 続いて、本実施例で生体分子放射性標識化容器10に形成された放射線センサ202の構造を説明する。放射線センサ202は生体分子放射性標識化反応装置1にて生体分子放射性標識化反応工程に関する放射線を検出するために用いられる。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図8に示す。
 本実施例では放射線センサ202は、放射線検出器204、放射線遮蔽部205から構成される。
 放射線センサ202は反応容器本体20表面に形成された流路中、放射性標識化剤固化部25の垂直上方、蓋部材40上に形成されている。
 検出器204および遮蔽部205の蓋部材40側入り口の大きさ、検出器204の蓋部材40からの距離は、生体分子放射性標識化反応装置1中の放射性標識化剤固化部25以外の部分に位置する放射性物質から発する放射線の影響を受けずに、放射性標識化剤固化部25の放射線を検出可能であればよい。
 因みに検出器204から放射性標識化剤固化部25までの距離206が近いほど、検出感度は高くなる。また、検出器204から放射性標識化剤固化部25までの距離206に相当する距離分、放射性標識化剤固化部25の外側に位置する放射性物質の放射線を検出する。
 遮蔽部205は放射性薬剤固化部25の外側に位置する放射性物質から発する放射線の影響を防ぐために、検出器204の外周に形成される。遮蔽部205の材質は、使用する放射性物質から発する放射線を遮蔽できればよい。かかる材質としてはタングステン、鉛、遮蔽用加工物等が挙げられる。遮蔽物の厚さは遮蔽したい放射線量に対応する厚さ以上であればよい。
 本実施例の場合、放射性物質が位置しない検出器204上部は、生体分子放射性標識化反応装置1を使用する環境から発生する放射線を遮蔽できる厚さで形成される。放射性物質が位置する検出器204左右側部は、図8に示す検出器204から放射性標識化剤固化部25までの距離206から放射性標識化剤固化部25内の距離を差し引いた長さ207に位置する放射性物質から発する放射線を遮蔽できる厚さで形成される。
 遮蔽部205にて検出器の入口の幅を狭くするほど、放射性標識化剤固化部25以外の放射線の検出を防ぐ事が可能である。放射性標識化剤固化部25の放射線を検出可能な幅であれば構わない。
 本実施例は反応容器本体20上の流路や、生体分子放射性標識化反応容器10の構成に応じて、放射性標識化剤固化部25の上方、蓋部材40上に形成する事が可能であり、汎用性に優れる。
 本実施例では実施例5の変形となる生体分子放射性標識化容器10に形成された第2の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図9に示す。特に明示しない点は実施例5と同様とする。
 本実施例では実施例6で蓋部材40に形成された放射線センサ202が、アダプター部材60にもう1つ対向で形成される。
 本実施例では放射線検出器204を対向で2つ使用することにより、高い検出感度を実現できる。
 放射性標識化剤がポジトロン核種の場合には同時計数を行うことにより、遮蔽部205が不要となる。
 本実施例では実施例5の変形となる生体分子放射性標識化容器10に形成された第3の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図10に示す。特に明示しない点は実施例5と同様とする。
 本実施例では実施例6で蓋部材40の放射性標識化剤固化部25上部に形成された放射線センサ202が、蓋部材40の混合物溶液排出部27上部にもう1つ形成される。
 本実施例では異なる位置に放射線検出器204を2つ使用することにより、反応の進行を2つの方法でモニタすることが可能となる。すなわち、放射性標識化剤固化部25上部に形成された放射線センサ202を用いて、生体分子溶液121に溶解して減少する放射性標識化剤の量のモニタが、混合物溶液排出部27上部に設置された放射線センサ202を用いて、混合溶液304中に増加する放射性物質の量のモニタが実現される。
 本実施例では実施例5の変形となる生体分子放射性標識化容器10に形成された第4の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図11に示す。特に明示しない点は実施例5と同様とする。
 本実施例では蓋部材40は遮蔽物タングステンで形成される。かかる材料としては、使用する放射性物質から発する放射線を遮蔽できればよい。タングステン以外に、鉛、コンクリート、遮蔽用加工物等が挙げられる。蓋部材40の厚さは遮蔽したい放射線量に対応する厚さ以上であり、放射性標識化反応容器10の積層で反応容器本体20上に形成される流路が維持される厚さ以上であれば構わない。
 本実施例では蓋部材40に放射性標識化剤固化部25から発する放射線を検出するための空間部208が形成される。空間部208は蓋部材40を貫通し、放射性標識化剤固化部25の放射線を検出でき、図11に示す204から放射性標識化剤固化部25までの距離206から放射性標識化剤固化部25の距離を差し引いた長さ207分の放射性物質の放射線を遮蔽できる幅で形成される。
 検出器204の上部、両横部に形成される遮蔽部205は生体分子放射性標識化反応装置1を使用する環境から発生する放射線を遮蔽できる厚さで形成される。
 本実施例は反応容器本体20上の流路中、放射性標識化剤固化部25の位置が同じものであれば、異なる流路形式の反応容器本体20にも対応可能であり、遮蔽部205の単純な構造を実現できる。
 本実施例では実施例8の変形となる生体分子放射性標識化容器10に形成された第5の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図12に示す。特に明示しない点は実施例8と同様とする。
 本実施例では実施例8で蓋部材40に形成された放射線センサ202が、アダプター部材60にもう1つ対向で形成される。
 これに対応して、本実施例ではアダプター部材60がタングステンで形成される。またアダプター部材60に放射性標識化剤固化部25から発する放射線を検出するための空間部208が形成される。
 本実施例では放射線検出器204を対向で2つ使用することにより、高い検出感度を実現できる。
 本実施例では実施例8の変形となる生体分子放射性標識化容器10に形成された第6の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図13に示す。特に明示しない点は実施例8と同様とする。
 本実施例では実施例8で蓋部材40の放射性標識化剤固化部25上部に形成された放射線センサ202が、蓋部材40の混合物溶液排出部27上部にもう一つ形成される。これに対応して、蓋部材40に混合物溶液排出部27上部にも混合物溶液排出部27の放射線を検出するための空間部208が形成される。
 本実施例では異なる位置に放射線検出器204を2つ使用することにより、反応の進行を2つの方法でモニタすることが可能となる。すなわち、放射性標識化剤固化部25上部に形成された放射線センサ202を用いて、生体分子溶液121に溶解して減少する放射性標識化剤の量のモニタが、混合物溶液排出部27上部に形成された放射線センサ202を用いて、混合溶液304中に増加する放射性物質の量のモニタが実現される。
 本実施例では実施例5の変形となる生体分子放射性標識化容器10に形成された第7の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図14に示す。特に明示しない点は実施例5と同様とする。
 本実施例では蓋部材40内に放射線センサ202を形成する。
本実施例では遮蔽部205の放射性標識化剤固化部25側の開口幅を適切に設定すれば、遮蔽部205は生体分子放射性標識化反応装置1を使用する環境から発生する放射線を遮蔽できる厚さで形成可能である。
 また、検出器204と放射性標識化剤固化部25との距離206が近いため、高い検出感度が実現される。
 本実施例では実施例11の変形となる生体分子放射性標識化容器10に形成された第8の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図15に示す。特に明示しない点は実施例11と同様とする。
 本実施例では実施例11で蓋部材40内に形成された放射線センサ202が、アダプター部材60にもう一つ対向で形成される。
 本実施例では放射線検出器204を対向で2つ使用することにより、更に高い検出感度を実現できる。
 放射性標識化剤がポジトロン核種の場合には同時計数を行うことにより、遮蔽部205が不要となる。
 本実施例では実施例11の変形となる生体分子放射性標識化容器10に形成された第9の放射線センサ202の構造を説明する。
 図2に示した生体分子放射性標識化反応容器10を積層し、反応容器本体20を通る面で切断した断面図を図16に示す。特に明示しない点は実施例11と同様とする。
 本実施例では実施例11で蓋部材40内の放射性標識化剤固化部25上部に形成された放射線センサ202が、蓋部材40内の混合物溶液排出部27上部にもう1つ形成される。
 本実施例では異なる位置に放射線検出器204を2つ使用することにより、反応の進行を2つの方法でモニタすることが可能となる。すなわち、放射性標識化剤固化部25上部に設置された放射線センサ202を用いて、生体分子溶液121に溶解して減少する放射性標識化剤の量のモニタが、混合物溶液排出部27上部に設置された放射線センサ202を用いて、混合溶液304中に増加する放射性物質の量のモニタが実現される。
 続いて、図1~図4、図17~図19を用い、反応容器本体20上で実現される生体分子の放射性標識化反応の工程を説明する。図17に放射性標識化剤溶液を導入する工程、図18に放射性標識化剤溶液の溶媒を除去する工程、図19に生体分子溶液を導入する工程から生体分子放射性標識化反応を行うまでの工程を示す。
 [放射性標識化剤溶液導入と溶媒除去工程]
  まず、図1~図4、図17、図18を用い、放射性標識化剤溶液122の生体分子放射性標識化反応容器10への導入と、放射性標識化剤溶液122の溶媒の除去工程を説明する。
 まず、送液切り替えバルブ102及び排気切り替えバルブ402を閉鎖、回収切り替えバルブ302を回収に切り替える。
 次に、送液切り替えバルブ103の設定を、溶液吸引ライン104から吸引された放射性標識化剤溶液122がシリンジ106に送液されるように切り替える。この後、シリンジポンプ107を図中下方に引き、放射性標識化剤溶液122を溶液吸引ライン104及び送液切り替えバルブ103を経てシリンジ106内に導入する。
 放射性標識化剤溶液122のシリンジ106への導入後、送液切り替えバルブ103を放射性標識化剤溶液122が、シリンジ106から放射性標識化剤溶液導入部109の方向に送液されるように切り替える。
 この切り替え後、放射性標識化剤溶液側のシリンジポンプ107を図中上方に押し、シリンジ106内の放射性標識化剤溶液122を送液切り替えバルブ103及び放射性標識化剤溶液導入部109を経由して生体分子放射性標識化反応容器10に送液する。
 放射性標識化剤溶液導入部109から送液された放射性標識化剤溶液122は、アダプタ部材60の下側面に形成された放射性標識化剤溶液導入口64から生体分子放射性標識化反応容器10内に導かれる。放射性標識化剤溶液導入口64は、不図示のソケットを取り付けるために大径に形成されており、アダプタ部材60、アダプタ部材50の小穴、アダプタ部材50の上側面に設置された入口ポート部51を経て、放射性標識化剤溶液122を放射性標識化剤溶液溜部28bに導く。
 放射性標識化剤溶液溜部28bは、放射性標識化剤溶液122を、全ての放射性標識化剤溶液供給ノズル24Aに対して等しい圧力で供給できる量を溜める最低限度の容量を備えている。
 放射性標識化剤溶液溜部28bを満たした放射性標識化剤溶液122は、放射性標識化剤溶液供給ノズル24Aに等しい圧力にて供給される。その結果、全ての放射性標識化剤溶液供給ノズル24Aからほぼ均一に放射性標識化剤溶液122が吐出され、各放射性標識化剤溶液供給ノズル24Aから放射性標識化剤固化部25に導かれる。
 放射性標識化剤溶液供給ノズル24Aは、図4に示す通り、生体分子溶液誘導流路部23の終端部(最下部)の放射性標識化剤溶液供給部24に形成されている。放射性標識化剤溶液供給ノズル24Aは、生体分子溶液供給ノズル22Aとノズル1個分ずつその位置が反応容器本体20の幅方向にずれている。放射性標識化剤溶液供給ノズル24Aの表面直径は、生体分子溶液誘導流路部23の各流路の流路幅および流路深さとほぼ同じ大きさとなっている。
 放射線センサ202を用い、放射線量の上昇にて放射性標識化剤溶液122の放射性標識化剤固化部25への導入を確認後、排気切り替えバルブ402を排気方向に、送液切り替えバルブ103、回収切り替えバルブ302を閉鎖に切り替える。
 温度調節ユニット501にて生体分子放射性標識化反応容器10を加熱し、放射性標識化剤溶液122の溶媒を除去する。
 放射性標識化剤溶液122の生体分子放射性標識化反応容器10への導入量は、放射性標識化剤固化部25を満たすだけの容量に設定する。放射性標識化剤溶液122の必要量が本容量以上である場合は、放射性標識化剤溶液122の放射性標識化剤固化部25への導入と放射性標識化剤溶液122の溶液除去の工程を、放射性標識化剤固化部25への放射性標識化剤溶液122の導入が必要量に達するまで繰り返す。
 放射性標識化剤固化部25への放射性標識化剤溶液122の導入が必要量に達した後は、生体分子放射性標識化反応容器10以前の放射性標識化剤溶液導入部109、シリンジ106内に残留する放射性標識化剤溶液122を排出し、溶媒或いは空気で満たしてもよい。この工程を行うことによって、反応中の放射能をモニタする際、放射性標識化剤溶液導入部109、シリンジ106内に残留する放射性標識化剤溶液122の影響を防ぐことができる。また溶媒を満たしておくことで、生体分子溶液121を往復送液する際、放射性標識化剤溶液導入部109への逆流をふせぐことができる。
 放射性標識化剤溶液122の生体分子放射性標識化反応容器10への導入工程にて、図示しない別途形成した放射線センサ202(例えば実施例7、10、13)を用い、放射性標識化剤固化部25以外への放射性標識化剤溶液122の流出の有無を確認すると本工程は更に確実となる。
 放射性標識化剤固化部25以外への放射性標識化剤溶液122の流出が確認された場合には、送液切り替えバルブ103を放射性標識化剤溶液122が、生体分子放射性標識化反応容器10からシリンジ106の方向に送液されるように切り替える。この切り替え後、放射性標識化剤溶液側のシリンジポンプ107を図中下方に押し、生体分子放射性標識化反応容器10内の放射性標識化剤溶液122を放射性標識化剤溶液導入部109、送液切り替えバルブ103を経てシリンジ106内に回収する。生体分子放射性標識化反応容器10内での放射性標識化剤溶液122に不純物による汚染の可能性がある場合は、送液切り替えバルブ103を送液、回収切り替えバルブ302を回収のままとし、放射性標識化剤溶液側のシリンジポンプ107を図中上方に押し、生体分子放射性標識化反応容器10内の放射性標識化剤溶液122を、滞留部203、混合物溶液回収ライン303を経由して回収する。
 放射性標識化剤溶液122の溶媒の除去工程において、加熱温度は放射性標識化剤溶液122の溶媒の沸点前後とする。また加熱時間は溶媒が除去されるのに充分な時間とする。予備検討にて、溶媒が除去されるのに必要な時間を確認しておけば、本工程を必要最小時間に設定できる。また生体分子溶液導入部108から気体を導入し、放射線センサ202を用いて放射性標識化剤固化部24の放射線検出量が変動しないことを確認すると、放射性標識化剤溶液122の溶媒除去が完了したことが確認できる。
 放射性標識化剤溶液122の溶媒の除去工程は、前述の生体分子放射性標識化反応容器10の加熱以外に、気体の導入、減圧等の方法でも構わない。
 放射性標識化剤溶液122の溶媒の除去工程が気体を導入する方法の場合は、排気ライン403の1本を送気ラインとし、気体供給元に接続する。かかる気体供給元としてはガスライン、ガスボンベ、気体を貯蔵した風船等が挙げられる。気体は、生体分子放射性標識化反応容器10内で行う反応に影響を起こさない気体であればよい。かかる気体として大気、窒素、アルゴン等が挙げられる。送気ラインと接続した側の排気バルブ402を、送気ライン403を経由して生体分子放射性標識化反応容器10に気体を送る方向に切り替え、気体供給元より送気ライン403、排気バルブ402を経由して生体分子放射性標識化反応容器10内に気体を導入する。導入量は放射性標識化剤溶液122の溶媒が出口ポート部31から排出しない程度の導入量とする。
 放射性標識化剤溶液122の溶媒の除去工程が減圧方法の場合は排気ライン403を真空装置と接続する。かかる真空装置としては真空ポンプ、アスピレータ等が挙げられる。減圧の強度は放射性標識化剤溶液122の溶媒が出口ポート部31から排出しない程度とする。
 以上により、放射性標識化剤溶液122の生体分子放射性標識化反応容器10への導入と、放射性標識化剤溶液122の溶媒の除去が実現される。
 [生体分子溶液導入と反応工程]
  次に、図1~図4、図19を用い、生体分子溶液121の生体分子放射性標識化反応容器10への導入と、生体分子と放射性標識化剤の反応の工程を説明する。
 まず、回収切り替えバルブ302を回収、排気切り替えバルブ402を閉鎖に切り替える。
 次に、送液切り替えバルブ102の設定を、溶液吸引ライン104から吸引された生体分子溶液121がシリンジ106に送液されるように切り替える。この後、シリンジポンプ107を図中下方に引き、生体分子溶液121を溶液吸引ライン104及び送液切り替えバルブ102を経てシリンジ106内に導入する。
 生体分子溶液121のシリンジ106への導入後、送液切り替えバルブ102を生体分子溶液121が、シリンジ106から生体分子溶液導入部108の方向に送液されるように切り替える。
 この切り替え後、生体分子溶液側のシリンジポンプ107を図中上方に押し、シリンジ106内の生体分子溶液121を送液切り替えバルブ102及び生体分子溶液導入部108を経由して生体分子放射性標識化反応容器10に導入する。
 生体分子溶液導入部108から送液された生体分子溶液121は、アダプタ部材60の下側面に形成された生体分子溶液導入口63から生体分子放射性標識化反応容器10内に導かれる。生体分子溶液導入口63は、不図示のソケットを取り付けるために大径に形成されており、アダプタ部材60、アダプタ部材50の小穴、アダプタ部材50の上側面に設置された入口ポート部51を経て、生体分子溶液121を生体分子溶液溜部28aに導く。
 生体分子溶液溜部28aは、生体分子溶液121を、全ての生体分子溶液供給ノズル22Aに対して等しい圧力で供給できる量を溜める最低限度の容量を備えている。
 生体分子溶液溜部28aを満たした生体分子溶液121は、全ての生体分子溶液供給ノズル22Aに等しい圧力にて供給される。その結果、全ての生体分子溶液供給ノズル22Aからほぼ均一に生体分子溶液121が吐出され、各生体分子溶液供給ノズル22Aから生体分子溶液誘導流路部23に導かれる。
 生体分子溶液誘導流路部23は、生体分子溶液供給ノズル22Aの表面直径とほぼ同じ大きさの流路幅であり、流路深さもほぼ同じ大きさである。この流路幅の十数倍程度の流路長さを経た後に、生体分子溶液121は、流路幅が漏斗状に形成された放射性標識化剤固化部25へ流出する。
 生体分子溶液供給部22から導入された生体分子溶液121が誘導される生体分子溶液誘導流路部23においては、生体分子溶液供給ノズル22Aの位置から反応容器本体20の長手方向(溶液の流れ方向)に沿って、生体分子溶液121の多数の帯状の流れとして互いに独立するように、生体分子溶液誘導流路部23を構成する第1の流路の間には、各流路と同じ幅の空間が形成されている。この空間を、本実施例では、第2の流路と呼んでいる。このように、生体分子溶液供給ノズル22Aと放射性標識化剤溶液供給ノズル24Aは、幅方向に交互にずれて配置されるため、多数の生体分子溶液供給ノズル22Aから吐出された生体分子溶液121は、放射性標識化剤固化部25において、帯状に流れる多層流れを形成される。
 生体分子溶液121を放射性標識化剤固化部25の固化した放射性標識化剤上を多層流れで通過させ、排出側流路部26に流入させる。
 放射性標識化剤固化部25で形成された多層流れは、反応容器本体20の長手方向(溶液の流れ方向)に流れるのに伴い、細分された各流れの幅方向断面積が収縮され、加速した流れとなって排出側流路部26へと流入する。すなわち、放射性標識化剤固化部25から排出側流路部26へ移動する際に、溶液の流れの幅は、多層流れの接触界面に垂直な方向に徐々に収縮され、多層流れを構成する個々の溶液層の幅方向長さが狭くなる。
 排出側流路部26は、反応容器本体20の長手方向に延びる流路であり、その終端が混合溶液排出部27に達している。排出側流路部26の長さは、生体分子溶液121と放射性標識化剤の混合に必要な時間が確保できる流さであれば良い。従って、排出側流路部26の流路幅と長さは、生体分子溶液121と放射性標識化剤の拡散時間に応じて決めれば良く、その形状も、図3に示すように直線状である必要はない。例えば蛇行状でもよく、渦巻き状でも良い。目的に応じて流路長を変えられるように、アダプタ部材50の出口ポート部52、アダプタ部材60の出口部62には複数個の穴が形成されている。
 混合した生体分子溶液121と放射性標識化剤122は、反応容器本体20の下部中央に形成された液体排出部26から、アダプタ部材50の出口ポート部52、アダプタ部材60の出口部62、混合物溶液排出口65を経て、滞留部203末端の回収切り替えバルブ302手前まで送液したら、シリンジポンプ107を停止し、送液を停止する。
 放射線センサ202を用い、放射性標識化剤固化部25に残存する放射線を検出する。
 続いて、送液切り替えバルブ102の設定を、生体分子溶液121が滞留部203から流路21方面に逆送液されるように切り替える。この後、シリンジポンプ107を図中下方に引き、滞留部203末端に達した生体分子溶液121を排出側流路部26、放射性標識化剤固化部25、生体分子溶液誘導流路部23を経て生体分子溶液導入部108まで送液したら、シリンジポンプ107を停止し、送液を停止する。
 上記の操作を放射性標識化剤固化部25から発する放射線量が予め設定した値以下に達するまで繰り返す。放射性標識化剤固化部25に残存する放射線量が予め設定した値に達したら、放射性標識化剤固化部25と滞留部203を往復した混合溶液量、回収切り替えバルブ302を経て混合物溶液回収ラインより回収し、シリンジポンプ107を停止して送液を停止する。
 上記往復送液工程では往復送液毎に放射線を検出するが、1回目の検出あるいは数回の検出による放射線量の低下度合いから、予め設定した放射線量の値に達するまでの往復送液回数を算出し、算出した回数往復送液を行ったら、混合物溶液を回収してもよい。また回収前に放射線検出を行い、予め設定した放射線量に達しているか確認してから混合物溶液を回収してもよい。
 また、往復送液中、放射線を1定時間毎に検出し、予め設定した放射線量に達したら、混合物溶液を回収してもよい。
 反応に使用する生体分子溶液121の容量が少量の場合は、必要量の生体分子溶液121を気体あるいは溶媒で挟んで送液する。溶媒で挟んで送液する際は、放射性標識化剤固化部25上は生体分子溶液121のみが通過するように往復送液範囲を、送液量で調整する。生体分子溶液121を気体あるいは溶媒で挟んで送液する際は不図示のサンプルループを用いても良い。
 本実施例では、pgオーダーの微量な放射性標識化剤を取り扱う際の、生体分子放射性標識化反応容器10の各部容量の1実施例を図20に示す表を用いて説明する。図20に示す表は、本実施例の流路各部のサイズ及び容量を示す。反応容器本体20上の送液構造は図3、図4に示す実施例1の構造とする。pgオーダーの微量な放射性標識化剤を取り扱うための溶液濃度を保つために、数10μL~数100μLと低容量の溶液を反応容器本体20に導入する。
 本実施例では、生体分子溶液誘導流路部23は幅、深さ共に200μmの流路が幅200μm間隔で31本形成されている。本容量は7μLである。
 放射性標識化剤溶液122を導入し、溶媒を固化する放射性標識化剤固化部25は本実施例では生体分子溶液誘導流路部23の深さ200μmよりも若干深く形成されており、漏斗状の容量は実測で17μLであった。放射性標識化剤固化部25に導入したい放射性標識化剤溶液122の容量が17μL以上の場合は複数回に分けて導入するかあるいは溶液の濃度を濃くして容量を17μL以下にして導入する。
 本実施例では排出側流路部26は幅、深さ共に500μm、長さ13mmの直線状の流路であり、容量は3μLである。
 流路21の容量27μL以上の生体分子溶液121を使用する際は、混合溶液排出部27・滞留部203及び混合物溶液回収ライン303の容量、生体分子溶液溜部28a・生体分子溶液供給部22及び生体分子溶液導入部108の容量にて必要容量を確保する。
 ここでは、生体分子放射性標識化反応装置1(図1)において、生体分子放射性標識化反応容器10に対してサンプルを送液するために使用する流路の実施例を説明する。図21に、本実施例に係る送液流路の概要を示す。
 生体分子溶液121及び放射性標識化剤溶液122を送液するシリンジ106の容量は、送液する溶液量に応じて選択すれば良い。この本実施例では1mLとする。
 また、溶液吸引ライン104の流路幅は、送液する生体分子溶液121及び放射性標識化剤溶液122の容量に応じて選択すれば良い。ただし、流路幅が狭くなると、シリンジ106への吸引時間が長くなる。本実施例では0.5mmとする。
 また、溶液廃液ライン105の流路幅は、送液する生体分子溶液121及び放射性標識化剤溶液122の容量に応じて選択すれば良い。デッドボリュームを低減するには、流路幅が狭い方が望ましいが、廃液速度を早くするには流路幅が広い方が望ましい。本実施例では1mmとする。
 生体分子溶液導入部108及び放射性標識化剤溶液導入部109の流路幅は、送液する生体分子溶液121及び放射性標識化剤溶液122の容量に応じて選択すれば良い。デッドボリュームを低減するには、流路幅が狭い方が望ましいが、送液能力を高めるには流路幅が広い方が望ましい。本実施例では0.5mmとする。
 滞留部203及び生成物回収ライン303の流路幅は、送液する生体分子溶液121及び放射性標識化剤溶液122の容量に応じて選択すれば良い。反応効率を向上するには1mm以下であることが望ましい。本実施例では0.5mmとする。
 溶液吸引ライン104、廃液ライン105、生体分子溶液導入部108、放射性標識化剤溶液導入部109、滞留部203、生成物回収ライン303の流路長は、操作に支障が無い長さであれば良い。本実施例では以下のように設定する。溶液吸引ライン104の流路長を600mm、廃液ライン105の流路長を600mm、生体分子溶液導入部108の流路長を600mm、放射性標識化剤溶液導入部109の流路長を600mm、滞留部203の流路長を120mmと生成物回収ライン303の流路長を120mmとする。
 送液切り替えバルブ103のデッドボリュームは少ない方が望ましい。そこで、本実施例では、送液切り替えバルブ103のデッドボリュームは3.4μLとする。
 本実施例では、本実施例に係る生体分子放射性標識化反応装置1に、実施例15(図20)に示す寸法の反応容器10、実施例16(図21)に示す寸法の送液流路を適用し、生体分子放射性標識化反応を実行した結果を示す。
  なお、本実施例においては、ニューロテンシン0.3mg/mLホウ酸緩衝液(pH8.9)を生体分子溶液121、50MBqの[18F]SFBジエチルエーテル溶液を放射性標識化剤溶液122とする。
 まず、50MBqの[18F]SFBジエチルエーテル溶液50μLを5回に分けて生体分子放射性標識化反応装置1の反応容器10、放射性標識化剤固化部25に、室温にて、0.5mL/分で送液し、反応容器10を50℃に加熱し、ジエチルエーテルを気化して除去した。
  続いて、ニューロテンシン0.3mg/mLホウ酸緩衝液(pH8.9)50μLを生体分子放射性標識化反応装置1の反応容器10に、室温にて、0.5mL/分で往復送液した。送液、送液バルブ102の切り替え、放射線検出、逆送液の1往復送液で約1分を必要とした。
 本実施例での反応を、実施例5(図8)に示す放射線センサでモニタリングした結果801を、図22にグラフで示す。図22には、得られた混合物溶液304をHPLCにて放射線検出器を用いて分析した結果802を併せて示す。縦軸は放射性標識化剤固化部25の初期放射線量を100%とした放射線量割合、横軸は混合時間を示す。
  802に示す通り、目的物である[18F]SFB標識ニューロテンシンは往復送液による混合時間の増加と共に収率が向上したが、混合時間5分から10分と比較すると、混合時間10分から20分ではその収率の向上は遅くなった。この結果は801に示した放射性標識化剤固化部25の放射線量の減少傾向と一致した。
 本結果より放射性標識化剤固化部25に固化した放射性標識化剤が生体分子溶液の往復送液で生体分子溶液に溶解し、生体分子と反応する事が明らかとなった。
  また、放射性標識化剤固化部25の放射線量の減少をモニタすることにより、往復送液による混合の停止時期を予め決められる事が明らかとなった。
 本実施例と同じ反応条件で生体分子ニューロテンシンの[18F]SFBを用いた標識化反応を行う際は、放射性標識化剤固化部25の放射線量が初期値の20%以下に達したら、混合を停止するように設定すると、有効な混合時間の設定が実現できる。
  また、標識化物の核種の半減期の情報と併せて混合時間を判断することもできる。
 18Fの半減期の減衰を、図23に示したグラフ上に804で示した。縦軸は放射性標識化剤固化部25の初期放射線量を100%とした放射線量割合、横軸は混合時間を示す。図22の802に示した[18F]SFB標識ニューロテンシンの収率が、802の延長線上に進行したとした場合の曲線を803に示した。
 802と803を併せて考慮すると[18F]SFB標識ニューロテンシンの収率の向上を目指して反応時間を延長しても、混合時間100分程度経過すると減衰により、得られる[18F]SFB標識ニューロテンシンの放射線量は減少することがわかる。本反応は放射性標識化剤固化部25での放射線量が設定値に達しなくても、混合時間が100分に達したら混合を停止するという設定も実現可能である。
 本実施例では、実施例17と同様の操作を実施例7(図10)に示す放射線センサでモニタリングした結果を図24にグラフで示す。すなわち放射性標識化剤固化部25と混合溶液排出部27の2か所で、放射線のモニタリングを行った。
  グラフ縦軸は放射性標識化剤固化部25の初期放射線量を100%とした放射線量割合、横軸は混合時間を示す。
 実施例17同様、放射性標識化剤固化部25モニタリング結果を801に、得られた混合物溶液304をHPLCにて放射線検出器を用いて分析した結果を802に、混合溶液排出部27の放射線モニタリング結果を805に示す。
  802に示した目的物である[18F]SFB標識ニューロテンシンの収率の向上の傾向は、805に示した混合溶液排出部27の放射線量の増加傾向とも一致した。
 本結果より放射性標識化剤固化部25の放射線量の減少、混合溶液排出部27の放射線量の増加のいずれのモニタによっても、往復送液による混合の停止時期の予測を行えることが明らかとなった。
 なお、各実施例において、標識化剤として、放射性標識化剤を例に説明したが、本発明は放射性標識化剤に限られることはなく、例えば蛍光性標識化剤でもよい。

Claims (15)

  1.  生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応容器において、
     反応容器本体と、当該反応容器本体の表面側に対向して設けた蓋部材とを有し、
     前記反応容器本体の裏面側に前記第1の化合物及び前記第2の化合物の導入部並びに混合物溶液の回収部を有し、
     前記反応容器本体の表面には流路が形成されており、
     前記流路の中間部に、前記第2の化合物の溶液の溶媒を除去して固化する標識化剤固化部が形成されており、
     前記流路において、前記標識化剤固化部の上流側に前記第1の化合物の供給部及び前記第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に前記混合物溶液の排出部が形成されていることを特徴とする生体分子標識化反応容器。
  2.  請求項1に記載の生体分子標識化反応容器において、
     前記標識化剤は、放射性標識化剤であり、
     放射線センサを備えていることを特徴とする生体分子標識化反応容器。
  3.  請求項2に記載の生体分子標識化反応容器において、
     前記放射線センサは、前記標識化剤固化部の放射線を検出する位置に配置されていることを特徴とする生体分子標識化反応容器。
  4.  請求項2または請求項3に記載の生体分子標識化反応容器において、
     前記放射線センサは、前記混合物溶液の排出部付近の放射線を検出する位置に配置されていることを特徴とする生体分子標識化反応容器。
  5.  請求項1~4の何れか1つに記載の生体分子標識化反応容器において、
     前記標識化剤固化部両端付近の前記蓋部材に、気化した溶媒を排気する排気部が形成されていることを特徴とする生体分子標識化反応容器。
  6.  請求項1~5の何れか1つに記載の生体分子標識化反応容器において、
     前記標識化剤固化部は、反応容器本体の表面に対し厚み方向に窪んだ凹部として形成されていることを特徴とする生体分子標識化反応容器。
  7.  請求項1~6の何れか1つに記載の生体分子標識化反応容器において、
     前記反応容器本体の表面に形成された流路の幅及び深さが1mm以下であることを特徴とする生体分子標識化反応容器。
  8.  請求項1~7の何れか1つに記載の生体分子標識化反応容器において、
     前記標識化剤固化部の上流側には、それぞれが平行に形成された複数の流路からなる生体分子溶液誘導流路部を備えることを特徴とする生体分子標識化反応容器。
  9.  請求項1~7の何れか1つに記載の生体分子標識化反応容器において、
     前記流路は、一定幅を有し、平行に形成された2つの壁で挟まれた空間として形成されていることを特徴とする生体分子標識化反応容器。
  10.  請求項9に記載の生体分子標識化反応容器において、
     前記流路は、一定幅を有し、平行に形成された2つの壁で挟まれた複数の空間として形成され、それぞれ前記標識化剤固化部を備えていることを特徴とする生体分子標識化反応容器。
  11.  反応容器内において生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応装置であって、
     反応容器本体の表面には流路が形成されており、前記流路の中間部に、前記第2の化合物の溶液の溶媒を除去して固化する標識化剤固化部が形成されており、前記標識化剤固化部の上流側に前記第1の化合物の供給部及び前記第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に前記混合物溶液の排出部が形成されている反応容器を含む反応容器ユニットと、
     前記反応容器に前記第1の化合物と前記第2の化合物をそれぞれ供給するとともに、前記標識化剤固化部で固化した前記第2の化合物の上部に、前記第1の化合物の溶液を往復送液する送液ユニットと、
     前記反応容器で混合された混合溶液を回収する回収ユニットと、
     前記反応容器ユニット、前記送液ユニット、前記回収ユニットを制御する制御ユニットと、
     を有する反応装置。
  12.  請求項11に記載の反応装置において、
     前記標識化剤は、放射性標識化剤であり、
     前記反応容器は、放射線センサを備えており、
     前記放射線センサの検出信号に基づいて、放射線量を測定する放射線検出ユニットを有することを特徴とする反応装置。
  13.  請求項12に記載の反応装置において、
     前記制御ユニットは、前記放射線ユニットの放射線検出信号に基づいて、前記送液ユニットによる往復送液を停止することを特徴とする反応装置。
  14.  反応容器本体の表面に流路が形成されており、前記流路の中間部に標識化剤固化部が形成されており、前記標識化剤固化部の上流側に第1の化合物の供給部及び第2の化合物の供給部が形成され、前記標識化剤固化部の下流側に混合物溶液の排出部が形成されている反応容器を使用して、生体分子である第1の化合物と標識化剤である第2の化合物を混合する反応方法において、
     前記標識化剤固化部に、前記第2の化合物の供給部から前記標識化剤である第2の化合物の溶液を導入し、溶媒を除去し、固化する処理と、
     前記第1の化合物の供給部から生体分子である第1の化合物の溶液を導入する処理と、
     前記固化した第2の化合物の上部に、前記第1の化合物の溶液を往復して通過させ、前記第1の化合物と前記第2の化合物を混合する処理と、
     を有する反応方法。
  15.  請求項14に記載の反応方法において、更に、
     前記固化した第2の化合物、または前記混合物の放射線を検出する処理と、
     検出した放射線量が予め設定した数値に達すると往復送液を停止して混合物を回収する処理と、
     を有することを特徴とする反応方法。
