Abstract
Description
ところで、近年、生体分子に蛍光色素分子を結合させて蛍光顕微鏡で観察することで像を得るバイオイメージングが盛んに研究されている。従来、緑色蛍光たんぱく(GPC)を用いたバイオイメージングが知られているが(非特許文献2)、標識となる分子が大きくタンパク質-タンパク質間の相互作用によりターゲットとしている生体分子に与える影響が問題となっている。ジアリールエテン化合物は低分子量であるため、高精度のバイオイメージングを達成できる化合物として期待される。しかしながら、当該化合物を生体試料へ導入するためには当該分子の水溶化が必要不可欠である。

(3)式(b)で表される化合物の保護基を除去する脱保護工程、
を含む、前記製造方法。
本発明のジアリールエテン化合物は式(I)で表される。
当該化合物は糖系残基Sgを有する。糖系残基とは、糖およびこれに類似する化合物に由来する基である。Sgは6員環糖、5員環糖、シクリトールおよびこれらを含むオリゴ糖からなる群より選択される糖系化合物から水酸基を除いた1価の糖系残基である。ただし糖系残基は、一部の水酸基が保護基により保護されていてもよい。以下、便宜上、糖系化合物を単に「糖」と、糖系残基を単に「糖残基」とも称する。
5員環糖は5員環骨格を有する糖であり、その例にはリボフラノース、アラビノフラノース、キシロフラノース、エリトロフラノース、トレオフラノース、およびリキソフラノースが含まれるが、これらに限定されない。
シクリトールとはシクロアルカンポリオール(ポリヒドロキシシクロアルカン)すなわち環状糖アルコールであり、その例にはイノシトール等が含まれる。
入手が容易であること等を考慮すると、本発明における糖としてはグルコピラノースが好ましい。
また前述のとおり、糖残基は水酸基の一部が保護されていてもよい。保護基については後で詳しく説明するが、アセチル基等のアシル基が好ましい。保護基が存在する場合、その数は糖残基1個当たり、1~2個が好ましく、1個がより好ましい。
Arは、下記式(A1)または(A2)で表される芳香族基である。
Rは炭素数1~4のアルキル基である。本発明おいて、アルキル基とは鎖状および分岐状の基を含む。よって、炭素数1~4のアルキル基は、具体的にメチル基、エチル基、n-プロピル基、i-プロピル基、n-ブチル基、およびt-ブチル基である。
UはSgとArをつなぐ連結基であり、-(CH2)n-、-CH2-U’-、または-C(=O)-である。nは1~5の整数である。nが大きいと得られるジアリールエテン化合物の水溶性が低下する場合があるので、nは1~3が好ましく1がより好ましい。
U’はArと結合する炭素数1~10のアルキル基である。炭素数が多いと得られるジアリールエテン化合物の水溶性が低下する場合があるので、炭素数は1または2が好ましく1がより好ましい。前述のとおりアルキル基は直鎖状および分岐状の基を含む。
式(I)における-(CY)m-は、当該化合物が脂環骨格を有することを示す。Yは水素原子またはハロゲン原子である。優れたフォトクロミック特性を得るために、Yはハロゲン原子が好ましく、フッ素原子がより好ましい。mは5~7の整数であるが、同様の理由から5であることが好ましい。
本発明のジアリールエテン化合物は任意の方法で製造されるが、以下、好ましい製造方法を説明する。
(1)6員環糖、5員環糖、シクリトールおよびこれらを含むオリゴ糖からなる群より選択される糖系化合物から誘導され、1つの水酸基がハロゲン原子に置換され、かつ他の総ての水酸基が保護基によって保護されたハロゲン化糖誘導体を準備する工程、
(2)前記ハロゲン化糖誘導体と式(a)で表される化合物とを反応させて、式(b)で表される化合物を製造するエーテル化工程、
(3)式(b)で表される化合物の保護基を除去する脱保護工程、を含む。
当該方法のスキームを以下に示す。
本発明で用いるアシルハロゲン化糖は、6員環糖、5員環糖、シクリトールおよびこれらを含むオリゴ糖からなる群より選択される糖から誘導され、一つの水酸基がハロゲン原子に置換され、かつ他の総ての水酸基が保護基によって保護されたアシルハロゲン化糖である。上記スキームにおいて、アシルハロゲン化糖はPSg-haloと示されている。ハロゲン原子に置換される水酸基はアノマー位の水酸基が好ましい。
保護基とは糖中の水酸基が副反応を起こさないように、水酸基を保護するための基である。本発明においては、水酸基の保護に通常使用される基を用いることができる。このような保護基の例には、アシル基、アセタール基、およびシリルエーテル基が含まれる。中でも、脱保護しやすいため、アシル基が好ましく、アセチル基がより好ましい。
1)式(a)で表されるジオール体
本工程ではアシルハロゲン化糖と式(a)で表されるジオール体とを反応させる。
当該ジオール体は公知の方法で調製できる。例えば、下記スキームに示すように、式(j)のジアリールエテン化合物の芳香族基Arに-U-OH基を導入すればよい。
式(j)のジアリールエテン化合物であって、Arがチオフェン骨格である化合物は、非特許文献5、6に記載の方法で合成できる。また、Arがチオフェン骨格である化合物のSを酸化すると、Arがチオフェンスルホン骨格である化合物が得られる。
式(a)のジオール体とアシルハロゲン化糖を反応させることでグリコシル化反応が生じ、式(b)で表される中間体が生成する。
この反応は、式(a)の化合物の水素原子とアシルハロゲン化糖のアノマー位のハロゲンが脱離することで生じるため、水素原子とハロゲンの脱離を促進する反応促進剤の存在下で実施することが好ましい。当該反応促進剤の例には、Ag2O等の酸化銀、およびHgBr2、Hg(CN)2等の水銀塩が含まれる。本発明においては反応をより促進しやすいという理由からAg2Oを用いることが好ましい。Ag2Oの使用量は、式(a)の化合物1molに対して5~10molが好ましい。
本工程で使用する溶媒は限定されないが、前述のとおり系中から水を除去すると反応がより促進されるので、水の溶解度が低い溶媒が好ましい。好ましい溶媒の例には、塩化メチレン等の塩素系炭化水素およびトルエン等の芳香族炭化水素等が含まれる。中でも塩素系炭化水素がより好ましい。
反応温度は、反応促進と副反応抑制の観点から適宜決定される。本発明においては10~40℃が好ましい。
本工程では式(b)で表される中間体の保護基を除去する。保護基の除去は公知の方法で行なうことができる。例えば、アセチル基等のアシル基で水酸基が保護されている場合は、水酸化リチウム、水酸化ナトリウム、炭酸カリウム等のアルカリと反応させることで容易に保護基を除去できる。本発明においては水酸化リチウムを使用することが好ましく、その使用料は式(b)で表される中間体1molに対して5~10molがより好ましい。
本反応に使用する溶媒も限定されないがメタノール等のアルコールが好ましい。反応温度も限定されないが、10~30℃が好ましい。
(1)6員環糖、5員環糖、シクリトールおよびこれらを含むオリゴ糖からなる群より選択され、1つの水酸基を除く他の総ての水酸基が保護基によって保護された保護化糖化合物を準備する工程、
(2)前記保護化糖化合物と式(p)で表される化合物とを反応させて、式(q)で表される化合物を製造するエステル化工程、
(3)式(p)で表される化合物の保護基を除去する脱保護工程、を含む。
前述の理由から、以下、保護化糖化合物を単に「保護化糖」とも称する。当該方法のスキームを以下に示す。
保護化糖は、スキームにおいてPSg-OHと示してある。保護化糖は、1つの水酸基を除いた他の総ての水酸基が保護基で保護されている糖である。保護基としては前述のものを使用できるが、本製造方法においてはアセタール基が好ましく、メトキシメチル基がより好ましい。
保護化糖は公知の方法で調製できる。例えば、フラノース等の糖に保護基を導入することで製造できる。アノマー位の水酸基(1位炭素原子上の水酸基)以外の総ての水酸基が保護されることが好ましい。
本発明で用いる好ましい保護化糖としては下記式(s2)で表される化合物が挙げられる。式(s2)において、R’はメチル基またはエチル基であるが、脱保護しやすいためメチル基が好ましい。
1)式(p)で表されるジカルボン酸体
本工程では保護化糖と式(p)で表されるジカルボン酸体とを反応させる。式(p)中、Ar、Y、およびmは前記のとおり定義される。式(p)のジカルボン酸体は公知の方法で調製できる。例えば、第一の製造方法で述べたように、式(j)のジアリールエテン化合物の芳香族基Arにホルミル基を導入したジホルミル体を得て、当該ホルミル基を酸化することで、式(p)のジカルボン酸体を製造できる。
式(p)のジカルボン酸体と保護化糖との反応により式(q)で表される中間体が生成する。式(q)中、Ar、Y、m、およびPSgは前記のとおり定義される。この反応は、エステル化反応であるので、公知の反応促進剤の存在下で実施することが好ましい。当該反応促進剤の例には、DCC等の脱水剤が挙げられる。DCCの使用量は、式(p)の化合物1molに対して2~5molが好ましい。
本工程で使用する溶媒は限定されないが、前述のとおり系中から水を除去すると反応がより促進されるので、水の溶解度が低い溶媒が好ましい。好ましい溶媒は既に述べたとおりである。
反応温度は、反応促進と副反応抑制の観点から適宜決定される。本発明においては10~40℃が好ましい。
本工程では式(q)の中間体の保護基を除去する。保護基の除去は公知の方法で行なうことができる。例えば、保護基がアセタール基である場合は、過剰量の塩酸等の酸により脱保護することができる。本反応に使用する溶媒も限定されないがメタノール等のアルコールが好ましい。反応温度も限定されないが、10~30℃が好ましい。
この結果、目的化合物を得ることができる。当該化合物は式(I)で表され、かつ連結基Uが-C(=O)-である化合物である。
本発明のジアリールエテン化合物は水または水系溶媒に可溶である。すなわち、本発明では、水、または水の濃度が70重量%以上、好ましくは80重量%以上、より好ましくは90重量%以上の水/有機溶媒(混合溶媒)に溶解することを水溶性であるという。
また、本発明のジアリールエテン化合物は糖残基を有するので、糖残基をさらに修飾して標識性や生体親和性を高めることも可能である。よって本発明のジアリールエテン化合物は生体試料に容易に導入が可能であり、高精度のバイオイメージングを達成しうる。
反応スキームを以下に示す。
アシルハロゲン化糖としてグルコピラノースの1位の炭素原子上の水酸基をBrに置換し、他の総ての水酸基をアセチル基で保護した化合物(ブロモテトラアセトグルコース)を準備した。具体的に、当該化合物は次のようにして合成した。ペンタアセチルグルコピラノース(TCI社製)を過剰量の臭化水素-酢酸溶液(臭化水素:酢酸=1:1(mol比))に溶解し、密栓して室温にて一昼夜反応させることにより定量的にブロモテトラアセトグルコースを得た。
当該化合物はチオフェン環の5位に連結基U(メチレン基)が結合しているジオール体である。当該ジオール体は以下の反応により調製した。
式(a1)のジオール体(69mg、0.16mmol)とブロモテトラアセトグルコース(197mg、0.48mmol)をCH2Cl2(3mL)に溶かし、さらにモレキュラーシーブス4Å(ナカライテスク株式会社製)0.2gを加えて室温で1時間撹拌した。さらにアルゴン気流下、室温で遮光してAg2O(111mg、0.48mmol)を加え24時間撹拌した。反応終了後、不溶物を吸引ろ過して除き、ろ液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=2:1~1:1)で分離精製し、式(b1)で表される中間体(96mg、収率52%)を得た。
当該中間体を、質量分析装置(型番JMS-T100LC、日本電子社製)を用いて分析したところ[M+Na]+=1111というピークを得た。当該分析はESIポジティブモードで測定したため、目的物に溶媒またはガラス由来のNa(原子量23)付加物として得られる。よって、このピークは目的物の質量1088に起因するので、得られた化合物が式(b1)の化合物であることを確認した。
HR-ESI-MS m/z: 1111.21787 [M + Na]+ (Calcd for C45H50F6NaO20S2, 1111.21387).
三つ口フラスコに、38mg(0.03mmol)の式(b1)で表される化合物、および2mLのメタノールを装入し撹拌して均一な溶液とした。フラスコ内に13mgの炭酸カリウム(0.3mmol)を装入し、室温で15時間反応を行なった。反応混合物をBio-Gel P-2 Gel(Bio-Rad社製)でゲル濾過精製し、式(I-1)の化合物であって、2つの水酸基がアセチル基で保護された化合物を得た。脱保護反応における収率は56%であった。
少量(50μL)のメタノールを混合した水溶液を準備し、当該溶液に2.5mgの式(I-1)の化合物を加えたところ、赤紫色の溶液が得られ、溶解することを確認した。同様にして、メタノールを含まない水に式(I-1)の化合物を溶解したところ、赤紫色の溶液が得られ、溶解することを確認した。さらに、メタノール少量(0.2mL)に化合物(I-1)を溶解し、蒸留水(4mL)を加えて95%水溶液を調製し、図1の吸収スペクトル(株式会社日立製作所製、U-4100を使用)を得た。開環体のスペクトルを破線10で示した。紫外光(313nm)を照射すると溶液は紫色に着色し、実線20で表わされるスペクトルを観察した。
脱保護工程において、炭酸カリウム(0.3mmol)の代わりに水酸化リチウム(ジアリールエテン1モルに対して5モル当量)を使用した以外は、実施例1-1と同様にして式(I-1)の化合物を製造した。その結果、式(I-1)の化合物であって、2つの水酸基がアセチル基で保護された化合物を得た。2つのアセチル基を含む化合物を得た。脱保護反応における収率は56%であった。
反応スキームを以下に示す。
当該化合物はベンゾチオフェン環の6位に連結基U(メチレン基)が結合しているジオール体である。当該ジオール体は以下の反応により調製した。
当該ジホルミル体(350mg、667μmol)のTHF-MeOH(3mL/3mL)溶液に、0℃でNaBH4(51mg、2eq.)を2回に分けて加え、0℃で4時間撹拌した。反応液を酢酸エチルで希釈し、有機層を水(3回)、飽和食塩水(1回)で順次洗浄し、硫酸ナトリウムで乾燥した。乾燥剤をろ別して溶媒を減圧留去し、残渣をカラムクロマトグラフィー(ヘキサン:酢酸エチル=2:1)で精製し、式(a2)のジオール体(220mg、62%収率)を淡赤色アモルファスとして得た。当該化合物の質量分析結果は次のとおりであった。
HR-ESI-MS m/z: 551.05427 [M + Na]+ (Calcd for C25H18F6NaO2S2, 551.05501).
当該化合物は以下の反応により調製した。
HR-ESI-MS m/z: 615.03291 [M + Na]+ (Calcd for C25H18F6NaO6S2, 615.03467).
式(a3)のジオール体(14mg、0.024mmol)とブロモテトラアセトグルコース(97mg、0.24mmol)をCH2Cl2(2mL)に溶かし、さらにモレキュラーシーブス4Å(0.1g)を加えて室温で1時間撹拌した。さらにアルゴン気流下、室温で遮光してAg2O(56mg、0.24mmol)を加え24時間撹拌した。反応終了後、不溶物を吸引ろ過して除き、ろ液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=2:1~1:1)で分離精製し、淡緑色アモルファスとして式(b3)で表される中間体(16mg、64%収率)を得た。
当該化合物について実施例1と同様にして質量分析したところ[M+Na]+=1275というピークを得た。このピークは目的物の質量1252に由来するので、得られた化合物が式(b3)で表される中間体であることを確認した。
HR-ESI-MS m/z: 1275.22742 [M + Na]+ (Calcd for C25H18F6NaO6S2, 1275.22483).
式(b3)で表される中間体(10mg、0.008mmol)をメタノール(2mL)に溶かし、室温で水酸化リチウム(3mg、0.08mmol)を加え24時間撹拌した。反応終了後、反応液を減圧濃縮し、残渣をBio-Gel P-2 (H2O)で分離精製し、反応混合物を得た。反応混合物を質量分析したところ、917というピークを得た。このピークは目的化合物の質量916に由来するので、得られた化合物が式(I-2)の化合物であることを確認した。
ESI-MS m/z: 917 [M + H] +.
実施例1と同様にして式(I-2)の化合物の水溶性を評価し、水溶性であることを確認した。具体的に、メタノール少量(0.2mL)に(I-2)の化合物を溶解し、蒸留水(4mL)を加えて95%水溶液を調製し、図2の吸収スペクトル(株式会社日立製作所製、U-4100を使用)を得た。破線10で示す開環体のスペクトルが得られた。紫外光(313nm)を照射すると溶液は黄色に着色し、実線20で示すスペクトルが得られた。また、当該化合物は紫外光の照射で蛍光を示し、実線30で示すスペクトルが得られた。

式(k1)のジホルミル体より、以下の手順にて式(p1)のジカルボン酸体を得た。

そこに式(s3)の保護化糖(480mg、1.64mmol)を乾燥CH2Cl2(2mL)に溶かして加え、室温にて終夜撹拌した。生じた固体を吸引ろ過し、CH2Cl2で洗浄した。溶媒を減圧除去し、シリカゲルカラムクロマトグラフィー(展開溶媒;ジクロロメタン:酢酸エチル=4:1)で精製し、式(q1)の中間体(405mg、0.403mmol、58%収率)を淡黄色粉末として得た。NMR(日本電子株式会社製、GSX400)および質量分析(株式会社島津製作所製GCMS-QP2010)の結果は以下のとおりであった。分析結果は以下のとおりであった。

式(q1)で表される中間体(290mg、0.289mmol)をメタノール(5mL)に溶解した。そこに6mol/Lの塩酸(25mL)を加え、55℃で6時間還流した。その後、炭酸ナトリウムで塩化水素をトラップしながら溶媒を減圧除去した。逆相シリカゲルカラムクロマトグラフィー(展開溶媒;メタノール:水=4:1)で精製し目的とする式(I-3)の化合物(210mg、0.269mmol、93%収率)を得た。分析結果は以下のとおりであった。

実施例1と同様にして式(I-3)の化合物の水溶性を評価し、水溶性であることを確認した。
このようにして得た水溶液の吸収スペクトル(株式会社日立製作所製、U-4100を使用)を図3に示す。曲線10は式(I-3)の化合物(開環体)のスペクトルであり、極大吸収波長は254nmであった。次いで、式(I-3)の化合物の水溶液に254nmの光を照射し、それ以上閉環反応が進行しない状態の光定常状態(PSS)とした。この状態でのスペクトルを曲線20として示す。閉環体の極大吸収波長は595nmであった。600nm以上の可視光を照射することにより元のスペクトルに戻ったことから、可逆的なフォトクロミズム(下記スキーム)を確認した。

式(p2)の化合物(366mg、0.457mmol),N,N-ジシクロヘキシルカルボジイミド(283mg、1.37mmol)、4-ジメチルアミノピリジン(19mg、0.152mmol)を窒素雰囲気下にし、乾燥ジクロロメタン(DCM)(12mL)に溶かし、室温で30分撹拌した。前述のとおりに準備した式(s3)の化合物(401mg、1.37mmol)を乾燥ジクロロメタン(1mL)に溶かした溶液を調製し、前記式(p2)の化合物を含む混合物に添加し、一晩撹拌した。沈殿を吸引ろ過で取り除き、溶媒を減圧除去した。シリカゲルカラムクロマトグラフィー(ジクロロメタン:酢酸エチル=4:1)で精製し、式(q2)の化合物を得た。収量は65mg(0.048mmol)、収率は11%であった。分析結果は以下のとおりであった。
MS (ESI) m/z = 1371.2971 [M+Na]+
式(q2)の化合物(50mg、0.037mmol)をTHF(1mL)に溶かし、メタノール(1mL)、6MのHCl(5mL)を加えた。60℃で6時間還流し、トルエン(10mL)を加え、溶媒を減圧除去した。逆相カラムクロマトグラフィー(メタノール:水=4:1)で精製し、目的化合物を得た。分析結果は以下のとおりであった。
MS (ESI) m/z = 1147.1922 [M+Na]+
この化合物約0.5mgを、0.5mLの水/メタノール(90/10重量比)、および0.5mLのメタノール単独に溶解し溶液を調製した。便宜上、前者を水溶液、後者をメタノール溶液とも称する。溶液の吸収スペクトルを図4、5に示す。曲線10は式(I-4)の化合物(開環体)のスペクトルである。メタノール溶液では、極大吸収波長は450nmで、モル吸光係数は39000であった。この吸収は可視光照射により消失し、元の無色の溶液に戻った。曲線20は閉環体のスペクトルである。閉環体は黄緑色の蛍光(曲線30)を示し、発光極大は520nm、蛍光量子収率は0.71であった。また、開環体に戻すと蛍光は消光した。以上より式(I-4)の化合物は、メタノール中で可逆的なフォトクロミズムと蛍光のスイッチングを示すことが明らかとなった。
式(I-4)のジアリールエテン約0.5mgを、0.5mLのメタノールに溶解し、それに水を加えて水/メタノール(5/1重量比)混合液を調製した。この溶液をアフリカツメガエルの4細胞期胚に注入した。注入直後の様子を図6(a)に示す。左上の割球の黒い点が注入部位である。注入後、約1時間放置して細胞分裂させた(32細胞期)。当該細胞に470~495nmの波長のUVを照射した。その後、蛍光の有無を調べたところ、注入部位周辺に蛍光が確認された(図6(b))。
実施例5で調製した溶液を、10μmにスライスしたアフリカツメガエルの尾芽胚頭部載せ、室温で30分間放置した。その後、尾芽胚頭部を10%メタノール水溶液で洗浄し、470~495nmの波長を照射した。明視野下を図7(a)に示す。このとき、暗視野下にて、515~550nmの波長を検出したところ図7(b)のようにほとんど発光はみられなかった。続いて360~370nmの波長のUVを30秒間照射した後、再度515~550nmの波長を検出した(図7(c))。図7(c)に示すとおり、蛍光シグナルは特に胚の表皮の外層に観察された。

Claims (9)
- 式(I):
Sgは、6員環糖、5員環糖、シクリトールおよびこれらを含むオリゴ糖からなる群より選択される糖系化合物(ただし一部の水酸基が保護されていてもよい)から水酸基を除いた1価の糖系残基であり;
Uは、-(CH2)n-、-CH2-U’-、または-C(=O)-であり(ただし、nは1~5の整数であり、U’はArと結合する炭素数1~10のアルキル基である);
Arは、下記式(A1)または(A2)で表される基であり
XはS、SO2、NR3(R3は炭素数1~3のアルキル基)、またはOであり、
Rは炭素数1~4のアルキル基であり、
R1およびR2は、独立に炭素数1~3のアルキル基であり、
aは0または1、bは0~3の整数であり、
*はUと結合していることを示す);
Yは水素原子またはハロゲン原子であり;
mは5~7の整数である]
で表される、ジアリールエテン化合物。 - 前記Sgがピラノースの水酸基を除いた1価の糖系残基である、請求項1に記載の化合物。
- 前記Sgがピラノースの1位炭素原子上の水酸基を除いた1価の糖系残基である、請求項1または2に記載の化合物。
- 前記Sgがシクリトールの水酸基を除いた1価の糖系残基である、請求項1または2に記載の化合物。
- 前記XがSまたはSO2である、請求項1~4のいずれかに記載の化合物。
- 請求項1~5のいずれかに記載のジアリールエテン化合物の製造方法であって、
(1)6員環糖、5員環糖、シクリトールおよびこれらを含むオリゴ糖からなる群より選択される糖系化合物から誘導され、1つの水酸基がハロゲン原子に置換され、かつ他の総ての水酸基が保護基によって保護されたハロゲン化糖誘導体を準備する工程、
(2)前記ハロゲン化糖誘導体と式(a)で表される化合物とを反応させて、
式(b)で表される化合物を生成するエーテル化工程、
(3)前記式(b)で表される化合物の保護基を除去する脱保護工程、
を含む、前記製造方法。 - 前記工程(1)を、Ag2O存在下で実施する、請求項6に記載の製造方法。
- 前記ハロゲン化糖誘導体におけるハロゲン原子が臭素原子である、請求項6または7に記載の製造方法。
- 前記ハロゲン化糖誘導体における保護基がアシル基である、請求項6~8のいずれかに記載の製造方法。
