Abstract

 セルロース由来糖液を、限外ろ過膜、ナノろ過膜、逆浸透膜のいずれか1以上の分離膜に通じてろ過することによる糖液の製造方法において、ろ過により汚染した分離膜を酸性物質および芳香族化合物を含む洗浄水を用いて洗浄する。汚染した分離膜を効果的に洗浄することができる。

Description

糖液の製造方法
 本発明は、セルロースから糖液を製造する方法に関する。
 近年、石油資源枯渇と地球温暖化への懸念から、石油代替原料としてのバイオマス利用が注目されている。特に、デンプンやセルロースなどの多糖を含むバイオマスを加水分解し、糖を得る方法に関して活発に研究が行われている。その理由は、糖を原料とした微生物発酵により様々な化学品を生産できるからである。
 セルロース由来糖液には、糖のほか、リグニン、シリカ、カルシウム塩、凝集タンパク質および未分解のセルロースなど水不溶性の微粒子、オリゴ糖、多糖、タンニンおよびタンパク質などの水溶性高分子、ならびに低分子の発酵阻害物質、無機塩および有機酸などが不純物として含まれており、それら不純物の除去方法として、微粒子を精密ろ過膜によって非透過側にろ別する方法、水溶性高分子を限外ろ過膜によって非透過側にろ別する方法、発酵阻害物質などの低分子化合物をナノろ過膜や逆浸透膜によって透過側から除去する方法がそれぞれ知られている(特許文献1)。

 本発明者らは、分離膜を利用してセルロース由来糖液から不純物を除去する工程において、長期運転に伴って分離膜に目詰まりが生じることを見出した。そこで本発明は、分離膜を利用してセルロース由来糖液を製造する方法において、セルロース由来糖液の不純物、特に水溶性高分子が要因となる分離膜の目詰まりを洗浄する方法を提供することによってセルロース由来糖液から効率的に不純物を除去する方法を提供することを課題とする。
 発明者らは、上述の問題を解決するため、様々な洗浄方法につき鋭意検討を行った結果、芳香族化合物のみを含む水溶液を用いて膜洗浄を行っても特別な洗浄効果は得らなかったが、驚くべきことに、芳香族化合物と酸性物質とを共に含む水溶液を用いて膜洗浄を行うことにより、著しい洗浄効果が得られることを発見した。本発明はかかる知見に基づいて完成されたものである。
 すなわち、本発明は以下の(1)~(7)の構成を有する。
(1)セルロース由来糖液を、限外ろ過膜、ナノろ過膜および逆浸透膜からなる群から選択される1以上の分離膜に通じてろ過する工程を含む糖液の製造方法において、ろ過後の分離膜を酸性物質および芳香族化合物を含む洗浄水を用いて洗浄することを含むことを特徴とする、糖液の製造方法。
(2)酸性物質が硫酸、塩酸、硝酸、シュウ酸、クエン酸および酢酸からなる群から選択される1種類または2種類以上であることを特徴とする、(1)に記載の糖液の製造方法。
(3)芳香族化合物がヒドロキシメチルフルフラール(HMF)、フルフラール、クマル酸、フェルラ酸、クマルアミド、フェルラアミドおよびバニリンからなる群から選択される1種類または2種類以上であることを特徴とする、(1)または(2)に記載の糖液の製造方法。
(4)洗浄水がセルロース由来糖液をナノろ過膜および/または逆浸透膜に通じて得られたろ液由来であることを特徴とする、(1)~(3)のいずれかに記載の糖液の製造方法。
(5)セルロース由来糖液が精密濾過膜で濾過されたものであることを特徴とする、(1)~(4)のいずれかに記載の糖液の製造方法。
(6)洗浄水を分離膜でクロスフロー濾過することにより分離膜を洗浄することを特徴とする、(1)~(5)のいずれかに記載の糖液の製造方法。
(7)洗浄水の膜面線速度が5~50cm/secであることを特徴とする、(6)に記載の糖液の製造方法。
 本発明によりセルロース由来糖液特有の不純物による分離膜の目詰まりを、膜劣化を抑えながら低コストで除去できるため、目詰まりした分離膜をセルロース由来糖液の製造工程に再利用することができる。
図1は、本発明を実施するための分離膜洗浄装置の、基本構成の略図である。 図2は、糖液の膜処理を行う機能と、分離膜の洗浄を行う機能とを兼用する分離膜洗浄装置の略図である。 図3は、糖液の膜処理を行う機能と、分離膜の洗浄を行う機能とが独立した分離膜洗浄装置の略図である。 図4は、セルロース含有バイオマスの前処理物からセルロース由来糖液を製造するシステム全体の略図である。
 本発明でいうセルロース由来糖液とは、セルロース含有バイオマスを加水分解し、その結果得られた糖液のことを指す。セルロース含有バイオマスの加水分解の方法は特に限定されないが、水熱処理、酸処理、アルカリ処理、酵素糖化などを適宜組み合わせた手法であることが好ましい。セルロース由来糖液には、グルコース、キシロース、マンノースなどの単糖、セロビオース、セロオリゴ糖、キシロオリゴ等などの水溶性多糖を含んでおり、こうした糖類を微生物の発酵原料(炭素源)として使用することができ、微生物によりエタノール、乳酸、アミノ酸へ変換することができる。
 また、セルロース由来糖液には、こうした糖類以外の成分として、加水分解の過程で分解されなかったリグニン、シリカ、カルシウム塩、凝集タンパク質および未分解のセルロースなどの微粒子、オリゴ糖、多糖、タンニンおよびタンパク質などの水溶性高分子、ならびに低分子の発酵阻害物質、無機塩および有機酸など多種多用な不純物を含んでいる。こうした不純物は、水溶性成分と、水不溶性成分の2種に分類することができ、水不溶性成分は、あらかじめセルロース由来糖液の固液分離により固形分として除去することが好ましい。
 セルロース由来糖液の固液分離の方法としては、遠心分離、精密ろ過膜によるろ過などの手法があるが、精密ろ過膜によるろ過は、ミクロンオーダーの水不溶性成分まで除去することができるため、本発明におけるセルロース由来糖液は、後段の分離膜によるろ過に加えて、精密ろ過膜によりあらかじめろ過されることが好ましい。なお、精密ろ過膜については、WO2010/067785号に記載のものを使用することができる。
 本発明では、前述のセルロース由来糖液を限外ろ過膜、ナノろ過膜、逆浸透膜のいずれか1以上の分離膜に通じてろ過することを特徴とする。セルロース由来糖液の分離膜によるろ過については、WO2010/067785号に記載の方法に準じて実施することができる。また、本発明で使用される分離膜に関しても、WO2010/067785号に記載の内容のものを使用することができる。
 セルロース由来糖液を前述の分離膜に通じてろ過する場合、その長期使用に伴って分離膜の目詰まりがおこってしまう。本発明では芳香族化合物および酸性物質を共に含む水溶液(以下、洗浄水ともいう)によって、目詰まりした分離膜を洗浄することを特徴とする。以下、芳香族化合物および酸性物質を共に含む水溶液による分離膜の洗浄工程(以下、膜洗浄工程ともいう。)について説明する。
 本発明でいう芳香族化合物とは、ヒュッケル則に従う環状炭化水素、すなわちπ電子の数が4n+2(nは0を含めた正の整数)を満たす環状炭化水素の構造を有する化合物のことであり、好ましくは、ヒドロキシメチルフルフラール(HMF)、フルフラール、クマル酸、フェルラ酸、クマルアミド、フェルラアミド、バニリンなどが挙げられる。本発明で使用する洗浄水は芳香族化合物を1種類のみ含んでもよいし、2種類以上含んでもよい。
 本発明でいう酸性物質とは、アレニウスの定義による酸、すなわち水溶液中で水素イオンを生じる物質のことであり、例えば硫酸、硝酸、塩酸などの無機酸、酢酸、シュウ酸、クエン酸などの有機酸などが挙げられる。本発明で使用する洗浄水は、前記酸性物質を1種類のみ含んでもよいし、2種類以上含んでもよい。
 本発明の膜洗浄工程では、分離膜を芳香族化合物および酸性物質を共に含む洗浄水で洗浄することを特徴とする。芳香族化合物のみを含む水溶液では特別な膜洗浄効果は得られず、酸性物質を共に含む場合に限り、分離膜表面および内部に付着し、膜目詰まりを誘起している成分を効率的に除去でき、著しく分離膜のろ過速度を回復させることができるためである。なお、酸性物質のみを含む水溶液により、膜の目詰まりはある程度解消されるが、共に芳香族化合物を含む場合に限り、洗浄効果は更に著しくなる。
 本発明の膜洗浄工程において、分離膜目詰まり成分中の、カルシウム塩、タンパク質、オリゴ糖などは酸性物質により除去され、またリグニン由来の発酵阻害物質などは芳香族化合物により除去されることが予想される。上記の通り、芳香族化合物のみを含む水溶液では特別な洗浄効果が得られず、酸性物質を共に含む場合に限り著しく分離膜のろ過速度を回復できた理由は定かではないが、おそらく目詰まり成分中のリグニン由来発酵阻害物質が、カルシウム塩、タンパク質、オリゴ糖などに覆われた構造を有しており、芳香族化合物のみを含む水溶液による洗浄では、洗浄液がリグニン由来発酵阻害物質に到達し得なかったためと考えられる。
 本発明において、洗浄水中の酸性物質の濃度範囲としては、合計100~5000ppmであることがより好ましい。
 洗浄水中の芳香族化合物の濃度範囲としては、合計で500~10000ppmであることが好ましく、5000~10000ppmであることがより好ましい。
 本発明の膜洗浄工程において、洗浄水の温度範囲は特に限定されないが、10~90℃が好ましい。10℃未満では、洗浄水の粘度が著しく高くなり膜洗浄の効果を損なうことがあり、また90℃を超えると熱により分離膜そのものが損傷を引き起こし、逆に膜のろ過性能を低下させてしまうことがあるためである。
 前記芳香族化合物および酸性物質を含む水溶液は、水に該芳香族化合物および酸性物質を添加することにより調製してもよいが、該芳香族化合物を予め含む水溶液に酸性物質を添加することにより調整してもよい。具体例として、セルロース由来糖液をナノろ過膜および/または逆浸透膜に通じて得られたろ液(以下、NFろ液等、という。)が挙げられる。NFろ液等には前記芳香族化合物が含まれていることが知られており、これに酸性物質を0.1~5g/Lの濃度範囲となるよう添加したものが本発明における洗浄水として好ましく使用される。なお、セルロース由来糖液の製造プロセスではNFろ液等を廃液として処分するのが通常であるが、これを分離膜の洗浄水として再利用することにより水が節約できるため、経済的な利点を有する。
 洗浄水による分離膜の洗浄方法としては、洗浄水に分離膜を浸漬する方法、洗浄水を分離膜で全量ろ過する方法、洗浄水を分離膜でクロスフローろ過する方法などがあるが、これらの中でも洗浄水を分離膜でクロスフローろ過する方法は、膜面に対し平行な流れを作ることで洗浄水によって除去された目詰まり成分を洗い流しながら洗浄を行うことができるため好ましい。
 洗浄水を分離膜でクロスフローろ過することによる膜洗浄においては、洗浄水の膜面線速度は5~50cm/secであることが好ましく、より好ましくは10~50cm/secである。その理由として、5cm/sec未満の場合は分離膜表面の洗浄効果が不十分になり、また、50cm/secを超える場合は、これ以上膜面線速度を増しても洗浄への効果はほとんど変わらないためである。なお、洗浄水の膜面線速度は、洗浄水送液ポンプ流量の増減により調節することができる。
 本発明の洗浄水を分離膜でクロスフローろ過する膜洗浄方法では、分離膜の細孔内を洗浄するためには膜間差圧をかけながら洗浄することが好ましいが、一方で膜間差圧を高くしすぎると、膜の目詰まり成分が膜面に対して強く押し付けられることにより、膜表面での洗浄性が低下してしまう。このような観点から、本発明の洗浄水を分離膜でクロスフローろ過する膜洗浄方法では、限外ろ過膜においては5kPa~1MPa、ナノろ過膜または逆浸透膜においては0.5~3MPaの膜間差圧をかけることが好ましい。膜間差圧とは、膜処理において膜をはさんで膜前後に生じる圧力の差、すなわち原液(濃縮液)側とろ液側との差圧のことである。洗浄時の膜間差圧が上記範囲未満の場合、膜の細孔を通る洗浄水が極端に少なくなり、細孔内の洗浄が不十分になることがある。また、洗浄時の膜間差圧が上記範囲を超える場合は、膜の細孔を通る液が過剰になり、洗浄水の消費量が増大するため経済的に不利である。なお、膜の種類にも寄るが、上記膜面線速度および膜間差圧の範囲における洗浄水のろ過流束は、通常0.05~0.5m/day程度である。
 膜洗浄工程による膜洗浄の効果は、目詰まりにより未使用時と比べて低下した膜ろ過流束が洗浄によってどれだけ回復したかをもって評価するものとする。すなわち、洗浄前の目詰まり膜および洗浄後の目詰まり膜のろ過流束を、未使用膜のろ過流束で除した値をろ過率(%)と定義し、洗浄前後のろ過率の差、あるいは洗浄後のろ過率の大きさによって洗浄効果を評価した。なお、ろ過率は通常100%が最大値となると考えられる。本発明では、セルロース由来糖液の膜処理に関し、ろ過率が70%未満に低下した膜は処理速度の低さゆえ実用に不適であるため使用不能とみなし、また、ろ過率が70%以上の膜であれば、処理速度の観点で十分実用的であるため糖液の膜処理に使用可能であるとみなした。すなわち、本発明における膜洗浄工程は、ろ過率が70%未満に低下した分離膜に関し、ろ過率を70%以上まで回復させることにより、セルロース由来糖液のろ過に再使用することを可能にするものである。
 次に、本発明の装置実施形態に関して説明する。本発明を実施するための装置は、洗浄水を保持する洗浄水保持タンクおよび洗浄水を膜に循環するための循環ポンプおよび循環ラインを少なくとも具備する。また、洗浄水の膜面に対する圧力を調節するためのバルブを具備することが好ましい。以下、本発明を実施するための装置について、図面を用いて説明する。なお、本発明の図面において、実線の矢印は液体または固体の流れおよび配管を示している。
 図1は、本発明を実施するための装置の、最も基本的な構成例の略図である。図2は、図1に含まれる必須要件に加え、洗浄水保持タンクに、糖液の膜処理に使用する糖液供給タンク14としての機能を兼備させた応用例の略図である。図3は、図1に含まれる必須要件に加え、糖液の膜処理システムを独立に有しており、バルブの操作によりこれらの膜処理工程、膜洗浄工程を切り替えることが出来る応用例の略図である。図4は、セルロース含有バイオマスの前処理物からセルロース由来糖液を製造するシステム全体の略図である。
 図1の装置に関して詳細に説明する。洗浄水を保持する洗浄水保持タンク1には、水、芳香族化合物および酸性物質が投入される。洗浄水保持タンク1に保持された洗浄水は、送液流量を調節できる循環ポンプ3によってクロスフロー式膜モジュール4に供給され、循環ライン10を経由して再び洗浄水保持タンクへと戻ってくる。また、調節バルブ5の開閉度および循環ポンプの流量操作により、膜間差圧を調節することができる。なお、膜間差圧は、圧力計11~13を用いて算出可能である。すなわち、圧力計11と圧力計13の測定値の平均と圧力計12の測定値との差を膜間差圧とみなせる。
 図2の装置に関して詳細に説明する。図2に示される装置は、糖液の膜処理を行う機能と、膜の洗浄を行う機能とを兼用する。糖液の膜処理の際に、糖液を保持する糖液供給タンク14は、膜の洗浄を行う際には洗浄水保持タンクとしても使用される。洗浄水は、洗浄水調整タンク8に水、芳香族化合物および酸性物質を投入して調整され、洗浄水投入ポンプ9によって糖液供給タンクに投入される。洗浄時、糖液供給タンク14に保持された洗浄水は、送液流量を調節できる循環ポンプ3によってクロスフロー式膜モジュール4に供給され、循環ライン10を経由して再び洗浄水保持タンクへと戻ってくる。また、調節バルブ5の開閉度および循環ポンプの流量操作により、膜間差圧を調節することができる。なお、膜間差圧は、圧力計11~13を用いて算出可能である。すなわち、圧力計11と圧力計13の測定値の平均と圧力計12の測定値との差を膜間差圧とみなせる。
 図3の装置に関して詳細に説明する。図3に示される装置は、糖の膜処理を行う機能と、膜の洗浄を行う機能を別々に備えており、3方バルブ16、17の操作により、その機能を切り替える。糖液の膜処理において、糖液供給タンク14に投入された糖液は、糖液送液ポンプ15によりクロスフロー式膜モジュール4へと供される。膜の洗浄に関しては、洗浄水を保持する洗浄水保持タンク1を備える。洗浄水保持タンク1に保持された洗浄水は、送液流量を調節できる循環ポンプ3によってクロスフロー式膜モジュール4に供給され、循環ライン10を経由して再び洗浄水保持タンクへと戻ってくる。また、調節バルブ5の開閉度および循環ポンプの流量操作により、膜間差圧を調節することができる。なお、膜間差圧は、圧力計11~13を用いて算出可能である。すなわち、圧力計11と圧力計13の測定値の平均と圧力計12の測定値との差を膜間差圧とみなせる。
 図4の装置に関して詳細に説明する。セルロース含有バイオマスは、糖化反応槽21内で糖化酵素と混合され、加水分解される。糖化反応後のスラリーは、スラリー移送手段22により、固液分離装置24へと移送され、固体残渣25と1次糖液とに分離される。1次糖液は、限外ろ過膜供給槽27に保持された後、限外ろ過膜供給ポンプ28によって限外ろ過膜モジュール30に供給され、高分子濃縮液と2次糖液(ろ液)に分離される。高分子濃縮液は限外ろ過膜供給槽27および限外ろ過膜供給ポンプ28によって循環され、更に濃縮される。2次糖液は、ナノろ過膜供給槽31に保持された後、ナノろ過膜供給ポンプ32によってナノろ過膜モジュール34に供給され、濃縮糖液と、NFろ液に分離される。濃縮糖液はナノろ過膜供給槽31およびナノろ過膜供給ポンプ32によって循環され、更に濃縮される。NFろ液は、逆浸透膜供給槽35に保持された後、逆浸透膜供給ポンプ36によって逆浸透膜モジュール39に供給され、RO濃縮液とROろ液に分離される。RO濃縮液は逆浸透膜供給槽35および逆浸透膜供給ポンプ36によって循環され、更に濃縮される。逆浸透膜供給槽内に保持されたRO濃縮液は、芳香族化合物を含む水溶液であり、酸供給タンク44および酸投入ポンプ45によって酸が投入されることにより、逆浸透膜供給槽内で洗浄水が調整される。洗浄水は、逆浸透膜供給槽35から、膜洗浄ポンプ40によって限外ろ過膜モジュール30やナノろ過膜モジュール34に返送され、膜モジュール洗浄に再利用される。ROろ液は、逆浸透膜ろ液槽41に保持され、必要に応じて逆浸透膜ろ液槽ポンプ42によって糖化反応槽21や限外ろ過膜モジュール30、ナノろ過膜モジュール34に返送され、糖化反応の固形分濃度調整や膜モジュール洗浄に再利用される。なお、糖化反応槽21、固液分離装置24、限外ろ過膜供給槽27、ナノろ過膜供給槽31、逆浸透膜供給槽35、逆浸透膜ろ液槽41はいずれも保温装置(それぞれ23、26、29、33、37、43)をそなえており、糖液の温度を保温しながら各工程の処理を行うことができる。
 以下に本発明の実施例を挙げるが、本発明はこれらに限定されるものではない。
 (参考例1)目詰まり膜作製方法
 本発明による膜洗浄の効果を正確に評価するためには、汚染状態が同一の膜を多数準備する必要がある。以下にその作製方法を説明する。
 セルロース含有バイオマスとして、2mmサイズまで粉砕した稲藁を使用した。前記セルロース含有バイオマスを水に浸し、撹拌しながら180℃で5分間オートクレーブ処理(日東高圧株式会社製)した。その際の圧力は10MPaであった。処理後は処理バイオマス成分に遠心分離(3000G)を用いて固液分離した。この溶液成分に“アクセルレース・デュエット”(ジェネンコア協和株式会社製)を添加して50℃で24時間反応させ、溶液成分由来の糖液を得た。その後、フィルタプレス処理(薮田産業株式会社製、MO-4)を行い、未分解セルロースあるいはリグニンを分離除去したバイオマス由来の糖液を得た。更に、本糖液を細孔径0.22μmの精密ろ過膜に供することにより、ミクロンオーダーの不溶性粒子を除去した。このようにして得られた糖液約40Lを、限外ろ過膜、ナノろ過膜または逆浸透膜からなるスパイラル膜モジュールに供した。いずれの膜を使用した場合も操作温度は50℃、膜面線速度は20cm/secとし、操作圧力は限外ろ過膜の場合0.1MPa、ナノろ過膜の場合2MPa、逆浸透膜の場合4MPaでそれぞれろ過を行った。ろ過流束が0.05m/day以下に低下した時点で運転を停止した。このようにしてろ過流束が低下したスパイラル膜モジュールを分解し、膜部分を190mm×140mmのシート状に裁断した。
 以降の実施例では、このようにして得られたシート状の膜に関して、スパイラル膜モジュールのろ過小型試験として使用できる小型の平膜ユニット“SEPACF-II”(GEオスモニクス社製:有効膜面積140cm)を使用して膜の洗浄試験および透過試験を行った。
 (参考例2)ろ過流束測定方法および膜洗浄効果の評価方法
 ろ過流束測定における、限外ろ過膜、逆浸透膜共通の条件として、操作温度25℃、膜面線速度20cm/secとした。操作圧力は、限外ろ過膜の場合0.1MPa、ナノろ過膜の場合2MPa、逆浸透膜の場合4Mpaとした。本条件にて純水のろ過を1分間行い、その間の平均ろ過流束(m/day)を測定した。なお、この際クロスフロー流の循環は行わず、そのまま排水した。また、膜分離装置は参考例1に記載のスパイラルモジュールを想定した小型の平膜ユニットを使用した。
 以降の実施例では、未使用膜、洗浄前の目詰まり膜、洗浄後の目詰まり膜のそれぞれにつき、上記操作にてろ過流束を測定した。そして、洗浄前の目詰まり膜および洗浄後の目詰まり膜のろ過流束測定値を、未使用膜のろ過流束測定値で除した値をろ過率(%)と定義し、膜洗浄前後のろ過率の回復、あるいは洗浄後のろ過率の大きさによって膜洗浄効果を評価した。なお、ろ過率は通常100%が最大値となると考えられる。
 (参考例3)HPLCによる芳香族化合物分析
 水溶液中のHMF、フルフラール、クマル酸、フェルラ酸、クマルアミド、フェルラアミド、バニリンの濃度は、下記に示すHPLC条件で、標品との比較により定量した。なお、クマルアミド、フェルラアミドは、市販の標品が流通していないため、委託合成(委託先:VSN社)により標品を取得した。
機器:高速液体クロマトグラフ“Lachrom elite”(株式会社日立製作所製)
カラム:“Synergi 2.5μm Hydro-RP 100A”(Phenomenex社製)
検出方法:Diode Array 検出器
流速:0.6mL/min
温度:40℃。
 (実施例1)芳香族化合物と酸性物質を含む水溶液による限外ろ過膜の洗浄
 参考例1の方法により得られた、ろ過流束の低下した耐熱性限外ろ過膜(デサル社製、“HWS UF”シリーズ)について、純水を洗浄水とした場合(条件1)、塩酸のみを0.5g/Lの濃度で含む水溶液を洗浄水とした場合(条件2)、フルフラールのみを0.5g/Lの濃度で含む水溶液を洗浄水とした場合(条件3)、塩酸を0.5g/Lの濃度で含み、かつHMF、フルフラール、クマル酸、フェルラ酸、クマルアミド、フェルラアミド、バニリンの群から選ばれる芳香族化合物のうちいずれか1つを0.5g/Lの濃度で含む水溶液を洗浄水とした各場合(条件4~10)、フルフラールを0.5g/Lの濃度で含み、かつ硝酸、硫酸、クエン酸、シュウ酸、酢酸の群から選ばれる酸性物質のうちいずれか1つを0.5g/Lの濃度で含む水溶液を洗浄水とした各場合(条件11~15)の合計15通りの条件を設定し、膜洗浄を実施した。前記洗浄水2Lを用いて、洗浄水温度25℃、操作圧力0.1MPa、膜面線速度30cm/secで20分間膜洗浄を行い、クロスフロー流を循環させた。膜分離装置は参考例1記載のスパイラルモジュールを想定した小型の平膜ユニットを使用した。ろ過流束は、参考例2の方法に従って、膜洗浄前後に測定した。ただし、膜洗浄前のろ過流速は全条件について共通とみなし、1条件について測定した値を、全条件に共通の膜洗浄前ろ過流束とした。参考例2に従い、ろ過流束をろ過率に換算した値を表1に示す。なお、未使用膜のろ過流束測定値は、0.258m/dayであった。表1から明らかなように、純水を使用して膜の洗浄を行った場合、洗浄前と比較すればろ過率は大きく回復したものの、膜の再使用に当たっては不十分であった。また、フルフラールのみを含む水溶液を使用した場合、ろ過率の回復は純水を使用した場合と同等であり、特別な効果は見られなかった。また、塩酸のみを含む水溶液を使用した場合、純水を使用した場合と比較して、ろ過率はより回復したが、依然として膜の再使用に当たっては不十分であった。一方、芳香族化合物と酸性物質を共に含む水溶液を使用した場合、いずれの組み合わせであってもろ過率は格段に回復し、膜の再使用に当たって十分なろ過率が得られた。

 (実施例2)芳香族化合物と酸性物質を含む水溶液によるナノろ過膜の洗浄
 参考例1の方法により得られた、ろ過流束の低下した耐熱性ナノろ過膜(デサル社製“HWS NF”シリーズ)について、操作圧力を2MPaとする以外は実施例1と同様の条件で膜洗浄を実施した。参考例2に従い、ろ過流束をろ過率に換算した値を表2に示す。なお、未使用膜のろ過流束測定値は、0.246m/dayであった。その結果、表2から明らかなように、純水を使用して膜の洗浄を行った場合、洗浄前と比較すればろ過率は大きく回復したものの、膜の再使用に当たっては不十分であった。また、フルフラールのみを含む水溶液を使用した場合、ろ過率の回復は純水を使用した場合と同等であり、特別な効果は見られなかった。また、塩酸のみを含む水溶液を使用した場合、純水を使用した場合と比較して、ろ過率はより回復したが、依然として膜の再使用に当たっては不十分であった。一方、芳香族化合物と酸性物質を共に含む水溶液を使用した場合、いずれの組み合わせであってもろ過率は格段に回復し、膜の再使用に当たって十分なろ過率が得られた。

 (実施例3)芳香族化合物と酸性物質を含む水溶液による逆浸透膜の洗浄
 参考例1の方法により得られた、ろ過流束の低下した耐熱性逆浸透膜(デサル社製“HWS RO”シリーズ)について、操作圧力を4MPaとする以外は実施例1と同様の条件で膜洗浄を実施した。参考例2に従い、ろ過流束をろ過率に換算した値を表3に示す。なお、未使用膜のろ過流束測定値は、0.245m/dayであった。その結果、表3から明らかなように、純水を使用して膜の洗浄を行った場合、洗浄前と比較すればろ過率は大きく回復したものの、膜の再使用に当たっては不十分であった。また、フルフラールのみを含む水溶液を使用した場合、ろ過率の回復は純水を使用した場合と同等であり、特別な効果は見られなかった。また、塩酸のみを含む水溶液を使用した場合、純水を使用した場合と比較して、ろ過率はより回復したが、依然として膜の再使用に当たっては不十分であった。一方、芳香族化合物と酸性物質を共に含む水溶液を使用した場合、いずれの組み合わせであってもろ過率は格段に回復し、膜の再使用に当たって十分なろ過率が得られた。

 (実施例4)膜面線速度の膜洗浄効果に対する影響
 参考例1の方法により得られた、ろ過流束の低下した耐熱性ナノろ過膜(デサル社製“HWSNF”シリーズ)について、膜面線速度条件を、5cm/sec、10cm/sec、30cm/sec、50cm/sec、70cm/sec、90cm/secの合計6通り設定し、その他の条件については実施例2の条件5と同様の条件で膜洗浄を実施した。参考例2に従い、ろ過流束をろ過率に換算した値を表4に示す。なお、未使用膜のろ過流束測定値は、0.246m/dayであった。その結果、表4からも明らかなように、膜面線速度が高いほどろ過率の回復は良好であり、50cm/sec以上で上限に達した。

 (実施例5)芳香族化合物濃度の洗浄効果に対する影響
 実施例2の条件5と同様の方法において、洗浄水中のフルフラール濃度に関し500ppm、1000ppm、3000ppm、5000ppm、7000ppm、10000ppmの6通りを設定して膜洗浄を行った。参考例2に従い、ろ過流速をろ過率に換算した値を表5に示す。なお、未使用膜のろ過流速測定値は、0.246m/dayであった。表5から明らかなように、フルフラール濃度が高くなるにつれて膜洗浄効果が高まっており、5g/L以上の濃度で最大の膜洗浄効果が得られた。

 (実施例5)セルロース由来糖液をナノろ過膜に通じて得られたろ液による膜洗浄
 参考例1の方法に従い、セルロース由来糖液をナノろ過膜に通じたNFろ液を得た。更に、NFろ液の一部を、逆浸透膜(“UTC-80”、東レ株式会社製)を用いて、常温にて操作圧力6MPaでろ過を行い、各成分濃度がおよそ6倍、10倍、20倍となったRO濃縮液を調整した(以下、それぞれ6倍NFろ液、10倍NFろ液、20倍NFろ液、という。)。これらの液中の芳香族化合物濃度を、参考例3の方法により分析した結果を表6に示す。

 洗浄水を、前記NFろ液、6倍NFろ液、10倍NFろ液、20倍NFろ液に、塩酸を0.5g/Lとなるように添加したものとすること以外、実施例2と同様の条件で膜洗浄を実施した。参考例2に従い、ろ過流束をろ過率に換算した値を表7に示す。なお、未使用膜のろ過流束測定値は、0.246m/dayであった。表6、7から明らかなように、芳香族化合物濃度が高いほど膜洗浄効果が高まった。

 本発明はセルロース由来糖液を限外ろ過膜、ナノろ過膜、逆浸透膜のいずれか1以上の分離膜に通じて濾過する工程を含む糖液の製造方法における分離膜の洗浄方法として利用することができる。

Claims (7)

  1.  セルロース由来糖液を、限外ろ過膜、ナノろ過膜および逆浸透膜からなる群から選択される1以上の分離膜に通じてろ過する工程を含む糖液の製造方法において、ろ過後の分離膜を酸性物質および芳香族化合物を含む洗浄水を用いて洗浄することを含むことを特徴とする、糖液の製造方法。
  2.  酸性物質が硫酸、塩酸、硝酸、シュウ酸、クエン酸および酢酸からなる群から選択される1種類または2種類以上であることを特徴とする、請求項1に記載の糖液の製造方法。
  3.  芳香族化合物がヒドロキシメチルフルフラール(HMF)、フルフラール、クマル酸、フェルラ酸、クマルアミド、フェルラアミドおよびバニリンからなる群から選択される1種類または2種類以上であることを特徴とする、請求項1または2に記載の糖液の製造方法。
  4.  洗浄水がセルロース由来糖液をナノろ過膜および/または逆浸透膜に通じて得られたろ液由来であることを特徴とする、請求項1~3のいずれかに記載の糖液の製造方法。
  5.  セルロース由来糖液が精密濾過膜で濾過されたものであることを特徴とする、請求項1~4のいずれかに記載の糖液の製造方法。
  6.  洗浄水を分離膜でクロスフロー濾過することにより分離膜を洗浄することを特徴とする請求項1~5のいずれかに記載の糖液の製造方法。
  7.  洗浄水の膜面線速度が5~50cm/secであることを特徴とする請求項6に記載の糖液の製造方法。
