Description
この発明は、アナライトの濃度を間欠的又は連続的に定量するセンシング装置及びセンシング方法に関する。
近時、例えば、アナライトと標識化合物との相互作用により蛍光強度が変化する性質を利用して、前記アナライトの濃度を定量するセンシング装置が開発されている。一適用例として、被検体の体内にセンサ部を埋め込み、グルコース濃度の連続定量を可能にする装置が提案されている(特許第4593957号公報及び特許第4558448号公報参照)。この装置を用いて糖尿病患者の血糖値の時系列データを取得・解析することで、血糖値を安定化するための薬剤の投与プロトコール設定や、生活習慣の改善指導を適切に行える。
しかしながら、特許第4593957号公報及び特許第4558448号公報に開示された技術的思想は、装置の構造の改良により消費電力を低減させることを目的としており、例えば、定量の際のサンプリング間隔を適切に決定することで、消費電力をさらに低減できる余地が十分にある。
本発明は、上記した課題を解決するためになされたものであって、センサを用いた間欠的な定量の際の消費電力を低減可能なセンシング装置及びセンシング方法を提供することを目的とする。
本発明に係るセンシング装置は、アナライトの濃度を間欠的又は連続的に定量する装置であって、定量のサンプリング間隔に従って前記アナライトの濃度に相関する計測信号を取得するセンサ部と、前記センサ部により取得された前記計測信号に基づいて前記アナライトの濃度を定量する濃度定量部と、所定の濃度範囲を複数のゾーンに分類するゾーン分類部と、前記濃度定量部により定量された前記アナライトの濃度についての、前記ゾーン分類部により分類された前記複数のゾーンの属否に応じて前記サンプリング間隔を決定する間隔決定部とを備えることを特徴とする。
このように、所定の濃度範囲を複数のゾーンに分類するゾーン分類部と、定量されたアナライトの濃度についての、前記ゾーン分類部により分類された前記複数のゾーンの属否に応じてサンプリング間隔を決定する間隔決定部とを設けたので、ゾーンの属否によってアナライトの濃度変化を大局的に捉えることができ、過不足ない適時の定量が可能になる。
また、前記センサ部は、標識物質又は被検体にエネルギーを与えて発生させた光現象から前記計測信号を取得する光学センサを含むことが好ましい。これにより、光学センサを用いた間欠的又は連続的な定量の際の消費電力を低減できる。また、光学センサを構成する電子部品の劣化を抑制する効果もある。
さらに、前記光学センサは、蛍光の発生を促す励起光を前記標識物質に向けて放射する励起光源を有し、前記間隔決定部により決定された前記サンプリング間隔に従って前記励起光を放射させるように前記励起光源を制御する光源制御部をさらに備えることが好ましい。励起光の放射がトリガとなって蛍光を得る光学センサの場合、定量頻度を減らすことが消費電力の低減に大きく寄与するため、特に効果的である。また、蛍光色素の劣化を抑制する効果もある。
さらに、前記ゾーン分類部は、前記所定の濃度範囲を、最も高い濃度側に存在する最高濃度ゾーン、最も低い濃度側に存在する最低濃度ゾーン、及び中間の濃度側に存在する少なくとも1つの中間濃度ゾーンに分類することが好ましい。
さらに、前記間隔決定部は、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最高濃度ゾーンでの前記サンプリング間隔を決定することが好ましい。
さらに、前記間隔決定部は、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最低濃度ゾーンでの前記サンプリング間隔を決定することが好ましい。
また、前記間隔決定部は、さらに、直近に複数回定量されて得た前記アナライトの濃度のトレンドに応じて前記サンプリング間隔を決定することが好ましい。トレンドを併せて考慮することで異なる観点から濃度変化を大局的に捉えることが可能になり、一層効果的である。
さらに、現時点で定量された前記アナライトの濃度が前記最高濃度ゾーンに属し、且つ、前記トレンドが上昇状態である場合、前記間隔決定部は、前記トレンドが下降状態である場合に応じた前記サンプリング間隔よりも小さくなるように、前記サンプリング間隔を決定することが好ましい。
さらに、現時点で定量された前記アナライトの濃度が前記最低濃度ゾーンに属し、且つ、前記トレンドが下降状態である場合、前記間隔決定部は、前記トレンドが上昇状態である場合に応じた前記サンプリング間隔よりも小さくなるように、前記サンプリング間隔を決定することが好ましい。
さらに、前記間隔決定部での決定に供される、各前記ゾーンに応じた前記サンプリング間隔のデータ組を一括して設定する設定部をさらに備えることが好ましい。
また、前記間隔決定部での決定に供される、各前記ゾーン及び各前記トレンドの組み合わせに応じた前記サンプリング間隔のデータ組を一括して設定する設定部をさらに備えることが好ましい。
本発明に係るセンシング方法は、アナライトの濃度を間欠的又は連続的に定量するセンシング装置を用いた方法であって、所定の濃度範囲を複数のゾーンに分類する分類ステップと、前記センシング装置が備えるセンサ部を用いて、定量のサンプリング間隔に従って前記アナライトの濃度に相関する計測信号を取得する取得ステップと、取得された前記計測信号に基づいて前記アナライトの濃度を定量する定量ステップと、定量された前記アナライトの濃度についての、分類された前記複数のゾーンの属否に応じて前記サンプリング間隔を決定する決定ステップとを備えることを特徴とする。
また、前記センサ部は、標識物質又は被検体にエネルギーを与えて発生させた光現象から前記計測信号を取得する光学センサを含み、前記取得ステップでは、前記光学センサを用いて前記計測信号を取得することが好ましい。
さらに、前記光学センサは、蛍光の発生を促す励起光を前記標識物質に向けて放射する励起光源を有し、決定された前記サンプリング間隔に従って前記励起光を放射させるように前記励起光源を制御する制御ステップをさらに備えることが好ましい。
さらに、前記分類ステップでは、前記所定の濃度範囲を、最も高い濃度側に存在する最高濃度ゾーン、最も低い濃度側に存在する最低濃度ゾーン、及び中間の濃度側に存在する少なくとも1つの中間濃度ゾーンに分類することが好ましい。
さらに、前記決定ステップでは、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最高濃度ゾーンでの前記サンプリング間隔を決定することが好ましい。

また、前記決定ステップでの決定に供される、各前記ゾーン及び各前記トレンドの組み合わせに応じた前記サンプリング間隔のデータ組を一括して設定する設定ステップをさらに備えることが好ましい。
本発明に係るセンシング装置及びセンシング方法によれば、所定の濃度範囲を複数のゾーンに分類し、定量されたアナライトの濃度についての前記複数のゾーンの属否に応じてサンプリング間隔を決定するようにしたので、ゾーンの属否によってアナライトの濃度変化を大局的に捉えることができ、過不足ない適時の定量が可能になる。
以下、本発明に係るセンシング方法についてこれを実施するセンシング装置との関係において好適な実施形態を挙げ、添付の図面を参照しながら詳細に説明する。
先ず、本実施形態に係るセンシング装置10の構成について、図1~図3を参照しながら説明する。
図1に示すように、センシング装置10は、センサ部12(蛍光センサ14及び温度センサ15を含む。)と、センサ制御回路16と、演算部18と、電源回路20と、ROM22と、RAM24と、クロック発生器26と、入力部27と、表示器28とを基本的に備える。
蛍光センサ14(光学センサともいう)は、アナライトAと標識化合物との相互作用による蛍光Fの強度に応じた計測信号(以下、蛍光信号という。)を取得する。蛍光Fは、アナライトAと標識化合物との結合又は解離に起因する光でもよいし、アナライトAと異なる第3成分と標識化合物との結合又は解離に起因する光であってもよい。いずれの蛍光Fであっても、蛍光信号に基づいてアナライトAの濃度を定量可能である。温度センサ15は、蛍光センサ14近傍での環境温度に応じた信号(以下、温度信号という。)を取得する。
なお、センサ部12は上記した形態に限定されるものではなく、標識物質又は被検体にエネルギーを与えて発生させた現象(例えば、光現象)から、アナライトAの濃度に相関する計測信号を取得可能であればよい。例えば、蛍光センサ14に代替して、吸光度、ラマン散乱光強度等を計測する光学センサや、温度、電気抵抗等を計測する化学センサを用いてもよい。
センサ制御回路16は、蛍光センサ14及び温度センサ15を駆動し、蛍光信号及び温度信号を取得可能に制御する。演算部18は、CPU、MPU等で構成されており、ROM22に記録されたプログラムを読み出し、後述する各種信号処理を実行する。
電源回路20は、演算部18を含むセンシング装置10内の各構成要素に電力を供給する。RAM24は、蛍光センサ14を介して入力された蛍光信号、温度センサ15を介して入力された温度信号の他、本発明に係るセンシング方法を実施するために必要な各種データを読出し又は書込み可能である。クロック発生器26は、所定周期でクロック信号を発生し、演算部18側に供給する。これにより、演算部18は、蛍光信号及び温度信号の取得タイミングの制御が可能である。
入力部27は、演算部18での演算に供される各種情報(例えば、計測モードの設定パラメータ)を入力可能に設けられている。例えば、押圧式ボタンであってもよいし、表示器28に組み込まれたタッチパネルであってもよい。表示器28は、演算部18により定量されたアナライトAの濃度に関する各種情報を可視化して表示する。表示器28は、モノクロ又はカラー表示可能な表示モジュールであり、液晶パネル、有機EL(Electro-Luminescence)、無機ELパネル等で構成されてもよい。
次いで、センサ部12の構造について、図2及び図3を参照しながら詳細に説明する。
図2に示すように、センサ部12は、概略矩形状の筐体30を備える。筐体30内部は中空であり、蛍光センサ14と、母材32と、6本の金属線34、35、36とを収容可能である。筐体30及び母材32は、例えばポリイミド、パリレン(ポリパラキシリレン)、又は環状ポリオレフィン等の樹脂でそれぞれ形成されている。また、外部光を遮光するため、材料中にカーボンブラック等の光遮断性材料を含有させてもよい。また、筐体30の一面(進入面38)は、ハイドロゲルとカーボンブラック等とからなり、アナライトAを通過させるとともに、外部光を遮断する特性を有する。
蛍光センサ14は、下方から上方の順に、シリコン等からなる基体40と、フォトダイオード(Photo Diode:以下「PD」ともいう。)素子42と、図示しない第1保護膜と、フィルタ44と、発光ダイオード(Light Emitting Diode:以下「LED」ともいう。)素子46(励起光源)と、エポキシ樹脂等からなる第2保護膜48と、インジケータ層50とから構成されている。
基体40の表面には、PD素子42が形成されている。PD素子42は、蛍光Fを電気信号に変換する光電変換素子である。PD素子42に代替して、フォトコンダクタ(光導電体)、又はフォトトランジスタ(Photo Transistor、PT)等の各種光電変換素子を用いてもよい。なお、PD素子42と金属線34とは、ボンディングワイヤ52、又は貫通配線等の手段により、電気的に結線されている。
フィルタ44は、LED素子46が発光する励起光Eの波長帯域は遮断し、且つ、前記波長帯域よりも長波長側の蛍光Fを通過する吸収型光学フィルタである。光学フィルタとして、多結晶シリコン等のシリコン膜、炭化シリコン膜、又はガリウムリン膜等を用いてもよい。
LED素子46は、蛍光Fの発生を促す励起光Eを放射する発光素子である。LED素子46に代替して、有機EL素子、無機EL素子、又はレーザダイオード素子等多様な種類の発光素子を用いてもよい。ここでは、蛍光Fの検出量(PD素子42での受光量)を増加させるため、蛍光Fの光透過率が高い発光素子を選択することが好ましい。
インジケータ層50は、進入面38から進入したアナライトA(例えば、グルコース)の濃度に応じた蛍光Fを発光する。インジケータ層50は、標識化合物としての蛍光色素が含まれたベース材料から構成されている。標識化合物(例えば、蛍光色素として、ルテニウム有機錯体やフェニルボロン酸誘導体、フルオロセイン標識デキストラン等のアナライトAと可逆結合する物質)と第3成分(例えば、ローダミン-標識コンカナバリンA)との解離により蛍光Fを発光させる場合、インジケータ層50のベース材料に標識化合物と併せて第3成分を含めてもよい。あるいは、第3成分を混入するための別異の機構を設けてもよい。
図3に示すように、筐体30の内部には、蛍光センサ14近傍での温度信号を取得可能な温度センサ15(図1参照)も配設されている。温度センサ15として、蛍光温度計等の光学型、サーミスタ型、金属薄膜抵抗型、又はPN接合の順方向電流の温度特性を基礎におく半導体型等を用いてもよい。なお、半導体型センサの場合、PD素子42と同様に、基体40上に形成可能である。
金属線34、35、36は、金、アルミニウム又は銅等の導電体で形成され、筐体30内の電気配線としての役割に加えて、剛性増加の機能を有する。センサ部12は、金属線34~36を介してセンサ制御回路16(図1参照)と電気的に接続されている。図3に示すように、金属線34は、2本の金属線34a、34bで構成されている。金属線35は、2本の金属線35a、35bで構成されている。金属線36は、2本の金属線36a、36bで構成されている。例えば、金属線34(又は金属線35)と金属線36との間に図示しない絶縁層を設けることで、両者を電気的に絶縁してもよい。
これにより、金属線34を介して、センサ制御回路16は、PD素子42からの蛍光信号を取得可能である。また、金属線35を介して、センサ制御回路16は、LED素子46に駆動電力を供給可能である。さらに、金属線36を介して、センサ制御回路16は、温度センサ15からの温度信号を取得可能である。
次に、センサ部12の動作について説明する。例えば、センサ部12が針状である場合、被検体に針先端部を穿刺・保持させることで、前記被検体の体内のアナライトAの濃度を連続して測定可能である。その際、一部のアナライトAは、進入面38から筐体30の内部に進入して、インジケータ層50周辺に滞在している。
先ず、センサ制御回路16は、蛍光センサ14の金属線35を介して、LED素子46に駆動電力信号を供給することで、励起光Eは発光される。そして、LED素子46からの励起光Eは、インジケータ層50に入射される。そして、インジケータ層50は、アナライトAと標識化合物との相互作用により、あるいは第3成分を加えた相互作用により、アナライトAの濃度に応じた強度の蛍光Fを発する。
インジケータ層50からの蛍光Fは、LED素子46、フィルタ44等を透過された後、PD素子42により光電変換され、蛍光信号として金属線34を伝送され、センサ制御回路16側に供給される。あわせて、温度センサ15からの温度信号は、金属線36を伝送され、センサ制御回路16側に供給される。
このようにして、蛍光センサ14は蛍光信号を取得するとともに、温度センサ15は温度信号を取得する。なお、図2及び図3に示すセンサ部12は、酵素センサ、グルコースセンサ、pHセンサ、免疫センサ、又は微生物センサ等、多様な用途に適用可能である。また、センサ部12の構成は、本構成に限られることなく種々の構成を採り得ることはいうまでもない。例えば、物理的に分離されたセンサ制御回路16及び演算部18の間を無線で通信可能に設けることで、センサ部12を被検体の体内に完全に埋め込んだ状態で間欠的又は連続的に定量可能である。
続いて、図1に示す演算部18の構成及び動作について、図4及び図5を参照しながら説明する。なお、本明細書では、アナライトAとしてグルコースを用いた定量結果を中心に説明する。
図4は、図1に示す演算部18の機能ブロック図である。なお、本図では、演算部18以外の他の構成要素に関して、本発明に係るセンシング方法に密接に関連する構成要素を中心に図示している。
センサ制御回路16は、所定の時点及び/又は光量で励起光Eを放射するようにLED素子46を駆動制御する光源制御部60と、PD素子42から取得した蛍光信号を蛍光強度F(t)として取得する蛍光信号取得部62とを備える。
演算部18は、蛍光センサ14を用いた計測を開始する旨をセンサ制御回路16(光源制御部60)に対して指示する計測指示部64と、所定の濃度範囲を複数のゾーンに分類するゾーン分類部66と、計測・定量のサンプリング間隔Tsを決定する間隔決定部68と、蛍光信号取得部62からの蛍光強度F(t)に基づいてアナライトAの濃度[A(t)]を定量する濃度定量部70と、演算部18での演算に供される各種パラメータを設定するパラメータ設定部71(設定部)とを備える。
間隔決定部68は、定量された濃度[A(t)]についての各ゾーン(具体的には、Lゾーン、Mゾーン及びHゾーン)の属否を判別する属否判別部72、直近の複数の定量値から濃度[A(t)]の変動傾向(以下、「濃度のトレンド」、又は単に「トレンド」という。)を推定するトレンド推定部74として機能する。
続いて、図1に示すセンシング装置10の動作について、図4の機能ブロック図及び図5のフローチャートを主に参照しながら詳細に説明する。
ステップS1において、パラメータ設定部71は、演算部18での演算に供される各種パラメータの初期設定を行う。設定に先立ち、ユーザ(医師等)は、入力部27を介して、被検体に適したモードを指定する。このモードは、例えば、食後、就寝時、運動時等、被検体の動態に応じてそれぞれ用意されている。そして、パラメータ設定部71は、指定されたモードに対応付けられた各種パラメータを設定する。なお、ステップS2以降が実行されている場合であっても、入力部27からの操作に応じて割り込み処理が発生することでステップS1に戻り、各種パラメータの変更内容を随時反映させることができる。
ステップS2において、ゾーン分類部66は、パラメータ設定部71により供給された一部のパラメータに基づいて、所定の濃度範囲を複数のゾーンに分類する。
図6は、複数のゾーンの分類例を示す概略説明図である。グラフの横軸は時間(分;単位:min)であり、グラフの縦軸はグルコースの濃度、いわゆる血糖値(単位:mg/dl)である。本実施形態では、ゾーン分類部66は、下限値を0[mg/dl]とし、上限値を計測可能な最大値とする濃度範囲において、2つの濃度閾値Dh、Dl(0<Dl<Dhを満たす。)を基準として3つのゾーンに分類する。この結果、0≦[A(t)]<Dlを満たし濃度が最も低い側に存在するゾーンは「Lゾーン」(最低濃度ゾーン)に、Dl≦[A(t)]<Dhを満たし濃度が中位に存在するゾーンは「Mゾーン」(中間濃度ゾーン)に、[A(t)]≧Dhを満たし濃度が最も高い側に存在するゾーンは「Hゾーン」(最高濃度ゾーン)にそれぞれ分類される。
ここで、分類されるゾーンの数は3つに限定されることなく、2つであっても4つ以上であってもよい。また、グラフの縦軸は濃度(血糖値)に限られず、計測された生データ{蛍光強度F(t)、又は蛍光信号そのもの}であってもよい。
ところで、パラメータ設定部71は、間隔決定部68での決定に供される、各ゾーンに応じたサンプリング間隔Tsのデータ組を一括して設定可能である。間隔決定部68は、パラメータ設定部71から前記データ組を取得することで、サンプリング間隔Tsの採り得る値、より詳細には、各ゾーンに応じたサンプリング間隔Tsを予め決定しておく。
図7に示すように、Hゾーン又はLゾーンに属する場合、サンプリング間隔Tsは、Ts=1[min]にそれぞれ設定される。また、Mゾーンに属する場合、サンプリング間隔Tsは、Ts=5[min]に設定される。
間隔決定部68は、さらに、サンプリング間隔Tsの初期値(例えば、Ts=1[min])を決定する。
ステップS3において、演算部18は、アナライトAの計測・定量指示があったか否かを判別する。具体的には、演算部18は、クロック発生器26から入力されたクロック信号のパルス数をカウントし、カウント上限値(時間に換算すると、サンプリング間隔Tsに相当する。)に到達した場合、定量指示があったと判別する。一方、前記カウント上限値に到達していない場合、到達するまでステップS3に留まる。
ステップS4において、計測指示部64は、計測を開始する旨の指示信号を光源制御部60に向けて出力する。そして、光源制御部60は、所定量の駆動電流を所定の時間幅で供給することでLED素子46を発光させる。その後、励起光Eは、インジケータ層50(すなわち、アナライトA、標識化合物又は第3成分)に向けて放射される。
ステップS5において、センサ制御回路16(蛍光信号取得部62)は、蛍光センサ14を介して、アナライトAと標識化合物との相互作用による蛍光F(図3参照)を検出する。蛍光信号取得部62は、蛍光Fの強度に応じた蛍光信号を取得し、この蛍光信号を蛍光強度F(t)に変換した上で(あるいはそのままの値で)演算部18側に供給する。そして、演算部18は、蛍光強度F(t)又は蛍光信号をRAM24に一時的に記憶させる。なお、センサ制御回路16は、蛍光信号の取得に同期して(あるいは非同期的に)、温度センサ15を介して温度信号を取得してもよい。
ステップS6において、濃度定量部70は、取得した蛍光強度F(t)、RAM24から読み出した定量係数等を用いて、アナライトAの濃度[A(t)]を定量する。なお、濃度[A(t)]の定量方法は、蛍光色素の材質、化学反応の性質等に適した種々の手法を採ることができる。
ステップS7において、属否判別部72は、ステップS6で定量された濃度[A(t)]についてのゾーンの属否を判別する。具体的には、属否判別部72は、0≦[A(t)]<Dlを満たす場合、今回の定量値はLゾーンに属するものと判別する。あるいは、属否判別部72は、Dl≦[A(t)]<Dhを満たす場合、今回の定量値はMゾーンに属するものと判別する。あるいは、属否判別部72は、Dh≧[A(t)]を満たす場合、今回の定量値はHゾーンに属するものと判別する。
図8Aに示すように、第1番目の定量点P1、第2番目の定量点P2、第3番目の定量点P3、第4番目の定量点P4、第5番目の定量点P5、及び第6番目の定量点P6が順次得られたとする。ここで、濃度閾値DlをDl=75[mg/dl]とし、本図中にLゾーン及びMゾーンを区画する境界線100を実線で表記している。
単に、両者の値の大小関係(境界線100に対する位置関係)に基づいて判別する場合、定量点P1、P3はLゾーンに属し、定量点P2、P4、P5、P6はMゾーンに属すると判別される。このように、計測に起因する定量ばらつきによって、各ゾーンの属否に関する判別結果に時系列的ゆらぎが生じる場合がある。この場合、後述するサンプリング間隔Tsの更新制御に影響を与えるおそれがある。
これに対し、境界線100を下限とし、破線で示す境界線102を上限とする不感帯104を設けてもよい。具体的には、前回の定量点P1がLゾーンに属し、且つ、今回の定量点P2が不感帯104(Mゾーン内)に属する場合、前回の判別結果(Lゾーンに属する旨)が維持されるようにする。これにより、定量点P1、P2、P3、P4はLゾーンに属し、定量点P5、P6はMゾーンに属すると判別されるようになる。このように、時系列的な判別結果のゆらぎを抑制することで、サンプリング間隔Tsの更新制御を安定的に実行できる。
なお、図8Aでは、トレンドが上昇状態(LゾーンからMゾーンへの遷移)である場合を例示したが、トレンドが下降状態(MゾーンからLゾーンへの遷移)である場合においても同様である。この場合、境界線100、102の位置関係が逆になる点に留意する。また、上記した揺らぎを抑制するため、種々の演算手法を採用し得ることは言うまでもない。
ステップS8において、トレンド推定部74は、直近の複数の定量値からトレンドを推定する。具体的には、トレンド推定部74は、直近の複数の定量値から回帰直線106を求め、その勾配が所定の正値よりも大きい場合に「上昇状態」、この勾配が所定の負値よりも小さい場合に「下降状態」、それ以外の場合は「平坦状態」であると推定する。
図8Bに示すように、例えば、現時点の定量点がP5であり、直近順に定量点P4、P3、P2、P1が既に得られていたとする。この場合、過去の定量点P1、P2、P3、P4を用いて、必要に応じて現時点の定量点P5も併せ用いて、回帰直線106(破線で図示する。)を求めることができる。なお、勾配を算出する手法は、加重平均、最小二乗法を含む種々の最適化手法を用いることができる。また、トレンドの推定に供される定量値の個数は5つに限られず、演算量、処理時間等を総合的に考慮して適宜決定してもよい。さらに、勾配(時間の1次微分)のみならず、曲率(時間の2次微分)を併せ考慮してトレンドを推定してもよい。
ステップS9において、間隔決定部68は、ステップS7で得たゾーンの属否、又はステップS8で得た濃度[A(t)]のトレンドに基づいて、次回のサンプリング間隔Tsを決定・更新する。ここで、間隔決定部68は、Mゾーン(少なくとも1つの中間濃度ゾーン)に応じたサンプリング間隔Tsのいずれよりも小さくなるように、Hゾーン又はLゾーンに応じたサンプリング間隔Tsをそれぞれ決定する。
図7に示すように、間隔決定部68は、Hゾーン又はLゾーンに属すると判別した場合、サンプリング間隔TsをTs=1[min]に決定する。ユーザにとって、特に被検体が高血糖値状態である場合、血糖値の最高ピーク値を正確に知りたい、という要求がある。また、ユーザにとって、特に被検体が低血糖値状態である場合、血糖値の最低ピーク値及び上昇の兆候を正確に知りたい、という要求がある。そこで、間隔決定部68は、定量のサンプル数を増やして診断性能を高めるため、相対的に小さいサンプリング間隔Tsを決定する。
一方、間隔決定部68は、Mゾーンに属すると判別した場合、サンプリング間隔TsをTs=5[min]に決定する。被検体が中血糖値状態である場合、上記した高血糖値状態(又は低血糖値状態)と比べて、被検体の安全状態が維持されている蓋然性が高い。そこで、間隔決定部68は、定量のサンプル数を減らして消費電力の低減を図るため、相対的に大きいサンプリング間隔Tsを決定する。
このように、間隔決定部68は、定量の都度、次のサンプリング間隔Tsを決定する。そうすると、図8Aに示すように、第1~5番目(定量点P1~P5)までは小さいサンプリング間隔Ts=1[min]で定量される。その後、Mゾーンに属すると新たに判別された直後、すなわち第6番目(定量点P6)以降は大きいサンプリング間隔Ts=5[min]で定量される。
ステップS10において、表示器28は、ステップS6における定量結果を表示する。表示処理に先立ち、演算部18は、得られた定量結果のうち、表示器28に表示させる可視情報(以下、定量可視情報という。)を決定した後、その定量可視情報に応じた制御信号を表示器28側に供給する。なお、定量可視情報として、例えば、定量値のみならず、トレンド、定量の成否、定量時刻、診断結果等が挙げられる。
図9A及び図9Bに示すように、表示器28が備える表示画面110上には、血糖値を表す数字112と、トレンドを表すマーク114とが表示されている。なお、図9Aの表示形態は、血糖値が100[mg/dl]であり、トレンドは「上昇状態」であることを示唆する。また、図9Bの表示形態は、血糖値が250[mg/dl]であり、トレンドは「緩やかな下降状態」であることを示唆する。
ところで、入力部27の操作に応じて、数字112及び/又はマーク114の表示の更新頻度を変更自在に設けてもよい。例えば、「定量の都度に更新」を選択することで、ユーザは現在の定量値及びトレンドを即時に把握できる。また、数字112の更新頻度をマーク114の更新頻度と異ならせてもよい。例えば、トレンドが「平坦状態」である場合に定量値(数字112)を更新し、トレンドが「上昇状態」又は「下降状態」である場合に定量値(数字112)を非更新にしてもよい。これにより、過渡状態下であっても表示器28に表示される数字112が目まぐるしく変動しなくなり、ユーザの混乱を防止できる。
ステップS11において、演算部18は、この一連の定量動作の終了指示があったか否かを判別する。終了指示がなかったと判別された場合、ステップS3に戻り、以下ステップS3~S11を繰り返す。一方、終了指示があった場合、センシング装置10は、アナライトAの定量動作を終了する。このように、演算部18は、サンプリング間隔Tsを適宜変更しながら、定量時点t毎の時系列データとしてのアナライトAの濃度[A(t)]を得る。
ところで、濃度変化の追従性を高くするためには、サンプリング間隔Tsを極力小さくして計測・定量の頻度を増やすことが最も望ましい。しかし、計測・定量の頻度が増加するにつれて、消費電力が増大するため好ましくない。そこで、本発明に係るセンシング方法を用いることで、濃度[A(t)]の経時的変化を適切に把握しつつも頻度を抑制した適時の定量が可能になる。
図10は、図7に示す決定方法に従って血中グルコースの濃度を定量し、得られた血糖値の経時的変化を表すグラフである。より詳細には、被検体の食後における体内の血糖値を間欠的又は連続的に定量している。ここで、2つの濃度閾値Dh、Dlをそれぞれ、Dh=140[mg/dl]、Dl=75[mg/dl]とした。本図から理解されるように、Mゾーンでの定量頻度は、Hゾーン及びLゾーンでの定量頻度と比べて約1/5に抑制されている。これにより、ピーク近傍での濃度変化、具体的には、「上昇状態」から「下降状態」への遷移、又は「平坦状態」から「上昇状態(又は下降状態)」への遷移に対する追従性を維持できる。一方、中血糖値状態(Mゾーン)での定量頻度を減らすことで、定量に要する消費電力を低減できる。
以上のように、所定の濃度範囲を複数のゾーン(Lゾーン、Mゾーン、Hゾーン)に分類するゾーン分類部66と、定量されたアナライトAの濃度[A(t)]についての複数のゾーンの属否に応じてサンプリング間隔Tsを決定する間隔決定部68とを設けたので、ゾーンの属否によってアナライトAの濃度変化を大局的に捉えることができ、過不足ない適時の定量が可能になる。
また、計測に供されるセンサは、標識物質又は被検体にエネルギーを与えて発生させた光現象から計測信号を取得する光学センサ(蛍光センサ14)を含んでもよい。これにより、光学センサを用いた間欠的又は連続的な定量の際の消費電力を低減できる。また、光学センサを構成する電子部品の劣化を抑制する効果もある。
さらに、蛍光センサ14は、蛍光Fの発生を促す励起光Eを標識物質に向けて放射するLED素子46を有し、間隔決定部68により決定されたサンプリング間隔Tsに従って励起光Eを放射させるようにLED素子46を制御する光源制御部60をさらに備えてもよい。励起光Eの放射がトリガとなって蛍光Fを得る光学センサの場合、定量頻度を減らすことが消費電力の低減に大きく寄与するため、特に効果的である。また、蛍光色素の劣化を抑制する効果もある。
続いて、サンプリング間隔Tsの決定に関する別の形態について、図11及び図12を参照しながら説明する。
図11は、サンプリング間隔Tsの決定方法を説明する第2の表である。この決定方法は、複数のゾーンの属否のみならず、推定されたトレンドも併せて考慮する点で、図7に示す方法と異なる。この場合、パラメータ設定部71は、間隔決定部68での決定に供される、各ゾーン及び各トレンドの組み合わせに応じたサンプリング間隔Tsのデータ組を一括して設定可能である。
本図に示すように、間隔決定部68は、定量値がHゾーンに属し、且つ、トレンドが「上昇状態」である場合、サンプリング間隔TsをTs=1[min]に決定する。一方、間隔決定部68は、定量値がHゾーンに属しても、トレンドが「平坦状態」又は「下降状態」である場合、サンプリング間隔TsをTs=3[min]に決定する。すなわち、血糖値の経時的変化が最高ピーク時に到達する前であると推定された状態下では、相対的に小さいサンプリング間隔Tsが設定される。
同様に、間隔決定部68は、定量値がLゾーンに属し、且つ、トレンドが「平坦状態」又は「下降状態」である場合、サンプリング間隔TsをTs=1[min]に決定する。一方、間隔決定部68は、定量値がLゾーンに属しても、トレンドが「上昇状態」である場合、サンプリング間隔TsをTs=3[min]に決定する。すなわち、血糖値の経時的変化が最低ピーク時を脱する前であると推定された状態下では、相対的に小さいサンプリング間隔Tsが設定される。このようにすれば、Hゾーン又はLゾーンに属し、且つ、隣接するMゾーンに向かうトレンドでの定量頻度は、図7例での定量頻度と比べて約1/3に抑制される。すなわち、サンプリング間隔Tsの決定の際、複数のゾーンの属否のみならずトレンドを併せて考慮することで、異なる観点から濃度変化を大局的に捉えることが可能になり、一層効果的である。
さらに、間隔決定部68は、上昇又は下降の勾配の大きさに応じて、サンプリング間隔Tsを2以上のレベルに細分して決定してもよい。例えば、Hゾーンに属し、且つ、0~5[mg/dl/min]の上昇状態であれば、サンプリング間隔TsをTs=2[min]に決定する。一方、Lゾーンに属し、且つ、0~5[mg/dl/min]の下降状態であれば、サンプリング間隔TsをTs=2[min]に決定する。これにより、サンプリング間隔Tsを段階的に切り替える制御が可能になり、設定の自由度が一層高くなる。
なお、この発明は、上述した実施形態に限定されるものではなく、この発明の主旨を逸脱しない範囲で自由に変更できることは勿論である。
Claims (22)
- アナライト(A)の濃度を間欠的又は連続的に定量するセンシング装置(10)であって、
定量のサンプリング間隔に従って前記アナライト(A)の濃度に相関する計測信号を取得するセンサ部(12)と、
前記センサ部(12)により取得された前記計測信号に基づいて前記アナライト(A)の濃度を定量する濃度定量部(70)と、
所定の濃度範囲を複数のゾーンに分類するゾーン分類部(66)と、
前記濃度定量部(70)により定量された前記アナライト(A)の濃度についての、前記ゾーン分類部(66)により分類された前記複数のゾーンの属否に応じて前記サンプリング間隔を決定する間隔決定部(68)と
を備えることを特徴とするセンシング装置(10)。 - 請求項1記載のセンシング装置(10)において、
前記センサ部(12)は、標識物質又は被検体にエネルギーを与えて発生させた光現象から前記計測信号を取得する光学センサ(14)を含むことを特徴とするセンシング装置(10)。 - 請求項2記載のセンシング装置(10)において、
前記光学センサ(14)は、蛍光(F)の発生を促す励起光(E)を前記標識物質に向けて放射する励起光源(46)を有し、
前記間隔決定部(68)により決定された前記サンプリング間隔に従って前記励起光(E)を放射させるように前記励起光源(46)を制御する光源制御部(60)をさらに備える
ことを特徴とするセンシング装置(10)。 - 請求項1~3のいずれか1項に記載のセンシング装置(10)において、
前記ゾーン分類部(66)は、前記所定の濃度範囲を、最も高い濃度側に存在する最高濃度ゾーン、最も低い濃度側に存在する最低濃度ゾーン、及び中間の濃度側に存在する少なくとも1つの中間濃度ゾーンに分類することを特徴とするセンシング装置(10)。 - 請求項4記載のセンシング装置(10)において、
前記間隔決定部(68)は、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最高濃度ゾーンでの前記サンプリング間隔を決定することを特徴とするセンシング装置(10)。 - 請求項4又は5に記載のセンシング装置(10)において、
前記間隔決定部(68)は、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最低濃度ゾーンでの前記サンプリング間隔を決定することを特徴とするセンシング装置(10)。 - 請求項4記載のセンシング装置(10)において、
前記間隔決定部(68)は、さらに、直近に複数回定量されて得た前記アナライト(A)の濃度のトレンドに応じて前記サンプリング間隔を決定することを特徴とするセンシング装置(10)。 - 請求項7記載のセンシング装置(10)において、
現時点で定量された前記アナライト(A)の濃度が前記最高濃度ゾーンに属し、且つ、前記トレンドが上昇状態である場合、
前記間隔決定部(68)は、前記トレンドが下降状態である場合に応じた前記サンプリング間隔よりも小さくなるように、前記サンプリング間隔を決定することを特徴とするセンシング装置(10)。 - 請求項7又は8に記載のセンシング装置(10)において、
現時点で定量された前記アナライト(A)の濃度が前記最低濃度ゾーンに属し、且つ、前記トレンドが下降状態である場合、
前記間隔決定部(68)は、前記トレンドが上昇状態である場合に応じた前記サンプリング間隔よりも小さくなるように、前記サンプリング間隔を決定することを特徴とするセンシング装置(10)。 - 請求項1~9のいずれか1項に記載のセンシング装置(10)において、
前記間隔決定部(68)での決定に供される、各前記ゾーンに応じた前記サンプリング間隔のデータ組を一括して設定する設定部(71)をさらに備えることを特徴とするセンシング装置(10)。 - 請求項7~9のいずれか1項に記載のセンシング装置(10)において、
前記間隔決定部(68)での決定に供される、各前記ゾーン及び各前記トレンドの組み合わせに応じた前記サンプリング間隔のデータ組を一括して設定する設定部(71)をさらに備えることを特徴とするセンシング装置(10)。 - アナライト(A)の濃度を間欠的又は連続的に定量するセンシング装置(10)を用いた方法であって、
所定の濃度範囲を複数のゾーンに分類する分類ステップと、
前記センシング装置(10)が備えるセンサ部(12)を用いて、定量のサンプリング間隔に従って前記アナライト(A)の濃度に相関する計測信号を取得する取得ステップと、
取得された前記計測信号に基づいて前記アナライト(A)の濃度を定量する定量ステップと、
定量された前記アナライト(A)の濃度についての、分類された前記複数のゾーンの属否に応じて前記サンプリング間隔を決定する決定ステップと
を備えることを特徴とするセンシング方法。 - 請求項12記載のセンシング方法において、
前記センサ部(12)は、標識物質又は被検体にエネルギーを与えて発生させた光現象から前記計測信号を取得する光学センサ(14)を含み、
前記取得ステップでは、前記光学センサ(14)を用いて前記計測信号を取得する
ことを特徴とするセンシング方法。 - 請求項13記載のセンシング方法において、
前記光学センサ(14)は、蛍光(F)の発生を促す励起光(E)を前記標識物質に向けて放射する励起光源(46)を有し、
決定された前記サンプリング間隔に従って前記励起光(E)を放射させるように前記励起光源(46)を制御する制御ステップをさらに備える
ことを特徴とするセンシング方法。 - 請求項12~14のいずれか1項に記載のセンシング方法において、
前記分類ステップでは、前記所定の濃度範囲を、最も高い濃度側に存在する最高濃度ゾーン、最も低い濃度側に存在する最低濃度ゾーン、及び中間の濃度側に存在する少なくとも1つの中間濃度ゾーンに分類することを特徴とするセンシング方法。 - 請求項15記載のセンシング方法において、
前記決定ステップでは、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最高濃度ゾーンでの前記サンプリング間隔を決定することを特徴とするセンシング方法。 - 請求項15又は16に記載のセンシング方法において、
前記決定ステップでは、前記少なくとも1つの中間濃度ゾーンに応じた前記サンプリング間隔のいずれよりも小さくなるように、前記最低濃度ゾーンでの前記サンプリング間隔を決定することを特徴とするセンシング方法。 - 請求項15記載のセンシング方法において、
前記決定ステップでは、さらに、直近に複数回定量されて得た前記アナライト(A)の濃度のトレンドに応じて前記サンプリング間隔を決定することを特徴とするセンシング方法。 - 請求項18記載のセンシング方法において、
現時点で定量された前記アナライト(A)の濃度が前記最高濃度ゾーンに属し、且つ、前記トレンドが上昇状態である場合、
前記決定ステップでは、前記トレンドが下降状態である場合に応じた前記サンプリング間隔よりも小さくなるように、前記サンプリング間隔を決定することを特徴とするセンシング方法。 - 請求項18又は19に記載のセンシング方法において、
現時点で定量された前記アナライト(A)の濃度が前記最低濃度ゾーンに属し、且つ、前記トレンドが下降状態である場合、
前記決定ステップでは、前記トレンドが上昇状態である場合に応じた前記サンプリング間隔よりも小さくなるように、前記サンプリング間隔を決定することを特徴とするセンシング方法。 - 請求項12~20のいずれか1項に記載のセンシング方法において、
前記決定ステップでの決定に供される、各前記ゾーンに応じた前記サンプリング間隔のデータ組を一括して設定する設定ステップをさらに備えることを特徴とするセンシング方法。 - 請求項18~20のいずれか1項に記載のセンシング方法において、
前記決定ステップでの決定に供される、各前記ゾーン及び各前記トレンドの組み合わせに応じた前記サンプリング間隔のデータ組を一括して設定する設定ステップをさらに備えることを特徴とするセンシング方法。
