最近、トリプトファンの代謝酵素であるインドールアミン2, 3-ジオキシゲナーゼ(IDO)がT細胞およびNK細胞の増殖を抑制し、また、制御性T細胞を活性化することによって、宿主の免疫を低下させることが報告されている。IDOは腫瘍組織で発現が亢進しており、IFN-γ刺激により癌細胞および樹状細胞でその発現が誘導される[例えば、ザ・ジャーナル・オブ・クリニカル・インベスチゲーション(J. Clin. Invest.)、117巻、5号、1147~1154頁(2007年)]。ヒトでは必須アミノ酸であるトリプトファンの90%が、IDOをその経路の開始段階に含むキヌレニン経路で代謝され、キヌレニン、さらに3OH-キヌレニン、キノリン酸等に変換される。IDOの活性化は局所あるいは全身のトリプトファン濃度を低下させ、キヌレニン濃度を上昇させ、キヌレニンを含むトリプトファン代謝物はT細胞死およびNK細胞死を誘導する[例えば、ザ・ジャーナル・オブ・エクスペリメンタル・メディスン(J. Exp. Med.)、196巻、4号、447~457頁(2002年)]。また、トリプトファン代謝はCD4陽性CD25陰性T細胞を制御性T細胞に変換させる[例えば、ブラッド(Blood)、109巻、7号、2871~2877頁(2007年)]。INF-γによってIDOの発現が誘導された樹状細胞の培養上清中ではトリプトファン濃度が低下し、キヌレニン濃度が上昇し、これにT細胞を共培養させると未刺激の樹状細胞共存下に比べてT細胞の増殖が抑制される[例えば、ザ・ジャーナル・オブ・エクスペリメンタル・メディスン(J. Exp. Med.)、196巻、4号、447~457頁(2002年)]。
結腸直腸癌および前立腺癌では腫瘍組織におけるIDOの発現亢進が報告されている[例えば、クリニカル・キャンサー・リサーチ(Clin. Cancer. Res.)、12巻、4号、1144~1151頁 (2006年)、ヨーロピアン・ジャーナル・オブ・キャンサー(Eur. J. Cancer)、44巻、15号、2266~2275頁(2008年)]。急性骨髄性白血病細胞ではIDOが恒常的に発現している[例えば、リューケミア(Leukemia)、21巻、353-355頁(2007年)]。子宮内膜癌、メラノーマ、および卵巣癌ではIDOの発現亢進した患者の予後が不良であることが報告されている[例えば、ブリティッシュ・ジャーナル・オブ・キャンサー(Br. J. Cancer.)、95巻、11号、1555~1561頁(2006年)、ジャーナル・オブ・クリニカル・インベスチゲーション(J. Clin. Invest.)、114巻、2号、280~290頁(2004年)、クリニカル・キャンサー・リサーチ(Clin. Cancer Res.)、11巻、16号、6030~6039頁(2005年)]。成人T細胞白血病リンパ腫、および急性骨髄性白血病では血中のキヌレニン/トリプトファン比が上昇している[例えば、リューケミア・リサーチ(Leuk. Res.)、33巻、1号、39~45頁(2009年)、リューケミア・リサーチ (Leuk. Res.)、33巻、3号、490~494頁(2009年)]。メラノーマでは血中のキヌレニン/トリプトファン比が上昇している患者の予後が悪いことが報告されている[例えば、ダーマトロジー(Dermatology)、214巻、1号、8~14頁(2007年)]。以上のように多くの固形癌、および血液癌でIDOおよび/またはキヌレニンが関与していることが考えられている。
また、キヌレニン産生抑制作用および/またはIDO阻害作用を示す化合物は、免疫賦活作用を示すことも知られている[例えば、ネーチャー・イミュノロジー(Nat. Immunol.)、2巻、64~68頁 (2001年)]。
一方、エンドセリン拮抗作用を有するピラジン誘導体が知られている(特許文献1および非特許文献1参照)。
またキヌレニン産生抑制作用を有する含窒素複素環化合物(特許文献11参照)が知られている。
さらに、GLP-1作動活性を有する2,3-ジ置換キノキサリン誘導体(特許文献12参照)が知られている。

式(I)の各基の定義において、
(i) 低級アルキルならびに低級アルカノイルおよび低級アルコキシにおける低級アルキル部分としては、例えば炭素数1~10の直鎖状または分枝鎖状のアルキルが挙げられ、より具体的には、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、イソブチル、sec-ブチル、tert-ブチル、n-ペンチル、ネオペンチル、n-ヘキシル、n-ヘプチル、n-オクチル、n-ノニル、n-デシル等が挙げられる。
(ii) フッ素置換低級アルキルとしては、例えば、置換数1~置換可能な数のフッ素で置換された低級アルキル等が挙げられ、より具体的には、ジフルオロメチル、トリフルオロメチル、(1-フルオロ-1-メチル)エチル、1,1,2,2,2-ペンタフルオロエチル等が挙げられる。
(iii) シクロアルキルとしては、例えば炭素数3~10のシクロアルキルが挙げられ、より具体的には、シクロプロピル、シクロブチル、シクロペンチル、シクロヘキシル、シクロヘプチル、シクロオクチル、シクロノニル、シクロデシル等が挙げられる。

C6-14アリールならびにC6-14アリールオキシ、C7-15アロイル、C7-15アロイルオキシ、およびC6-14アリールオキシカルボニルのアリール部分(vi)としては、例えば炭素数6から14のアリール、より具体的には、フェニル、ナフチル、アズレニル、アントリル等が挙げられる。
ハロゲンは、フッ素原子、塩素原子、臭素原子、ヨウ素原子の各原子を意味する。
化合物(I)の薬理学的に許容される塩としては、薬理学的に許容される酸付加塩、金属塩、アンモニウム塩、有機アミン付加塩、アミノ酸付加塩等が挙げられる。酸付加塩としては、例えば、塩酸塩、硫酸塩、リン酸塩等の無機酸塩;酢酸塩、トリフルオロ酢酸塩、マレイン酸塩、フマル酸塩、酒石酸塩、クエン酸塩、乳酸塩等の有機酸塩が挙げられ、金属塩としては、例えば、ナトリウム塩、カリウム塩等のアルカリ金属塩;マグネシウム塩、カルシウム塩等のアルカリ土類金属塩;アルミニウム塩、亜鉛塩等が挙げられ、アンモニウム塩としては、例えば、アンモニウム、テトラメチルアンモニウム等の塩が挙げられ、有機アミン付加塩としては、例えば、モルホリン、ピペリジン等の付加塩が挙げられ、アミノ酸付加塩としては、例えば、リジン、グリシン、フェニルアラニン、アスパラギン酸、グルタミン酸等の付加塩が挙げられる。
IDOおよび/またはキヌレニン産生に関わる疾患としては、例えば癌(腫瘍)、免疫性疾患、神経変性疾患、感染症等が挙げられる。
免疫性疾患としては、例えば、後天性免疫不全症候群(AIDS)、気管支喘息、花粉症、アレルギー性鼻炎、アトピー性皮膚炎、関節リウマチ、潰瘍性大腸炎、クローン病、多発性硬化症、筋萎縮性側索硬化症、移植片対宿主病等が挙げられる。
感染症としては、例えばウイルス感染症、細菌感染症、真菌感染症、クラミジア感染症、リケッチア感染症等が挙げられる。
上記化合物(I)またはその薬理学的に許容される塩は、中でも、癌(腫瘍)等の予防または治療剤の有効成分として特に好適である。
また、以下に示す製造法において、定義した基が実施方法の条件下で変化するか、または方法を実施するのに不適切な場合、有機合成化学で常用される保護基の導入および脱離方法[例えば、プロテクティブ・グループス・イン・オーガニック・シンセシス第3版(Protective Groups in Organic Synthesis third edition)、グリーン(T. W. Greene)著、ジョン・ワイリー・アンド・サンズ・インコーポレイテッド(John Wiley & Sons Inc.)(1999年)]等を用いることにより、目的化合物を得ることができる。また、必要に応じて置換基導入等の反応工程の順序を変えることもできる。
製造法1

化合物(Va-1)は、化合物(IIIa-1)を、無溶媒で、または反応に不活性な溶媒中、1~10 当量、好ましくは1当量の化合物(IX)と、1~100当量、好ましくは1~5当量の適当な塩基の存在下、-10℃~200℃ の間の温度で、好ましくは30℃~180℃ の間の温度で、5分間~72時間反応させることにより製造することができる。
化合物(IX)は、例えば市販品として得ることができる。

化合物(I)は、化合物(Va-1)を、無溶媒で、または反応に不活性な溶媒中、1~100当量の、好ましくは1~10当量の適当な塩基の存在下、1~20当量の、好ましくは1~4当量の化合物(VI)と、-20℃~200℃の間の温度で、好ましくは室温~80℃の間の温度で、5分間~72時間反応させることにより製造することができる。
適当な塩基としては、例えば水素化ナトリウム、水素化カリウム等のアルカリ金属水素化物;ナトリウムメトキシド、ナトリウムエトキシド、カリウムtert‐ブトキシド等の金属アルコキシド;n-ブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウム等の有機金属塩基;LDA、LiHMDS等の金属アミド;水酸化ナトリウム、水酸化カリウム、水酸化リチウム、炭酸ナトリウム、炭酸カリウム等のアルカリ金属塩;ピリジン、トリエチルアミン、ジイソプロピルエチルアミン、DBU、DBN、DMAP、N-メチルモルホリン等の有機塩基;アンバーリストA-21(ロームアンドハース社製)、AG1-X8(バイオラッド社製)、ポリビニルピリジン、モルホリノメチルポリスチレン等の固体塩基等が挙げられ、中でも、アルカリ金属水素化物、金属アルコキシド等が好ましく、水素化ナトリウム、カリウムtert-ブトキシド等がより好ましい。

化合物(Va-1)は、化合物(IIIa-2)を、無溶媒で、または反応に不活性な溶媒中、1~10 当量、好ましくは1当量の化合物(VIII)と、1~100当量、好ましくは1~5当量の適当な塩基の存在下、-10℃~200℃の間の温度で、好ましくは30℃~180℃の間の温度で、5分間~72時間反応させることにより製造することができる。
適当な塩基としては、例えば水素化ナトリウム、水素化カリウム等のアルカリ金属水素化物;ナトリウムメトキシド、ナトリウムエトキシド、カリウムtert-ブトキシド等の金属アルコキシド;n-ブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウム等の有機金属塩基;LDA、LiHMDS等の金属アミド;水酸化ナトリウム、水酸化カリウム、水酸化リチウム、炭酸ナトリウム、炭酸カリウム等のアルカリ金属塩;ピリジン、トリエチルアミン、ジイソプロピルエチルアミン、DBU、DBN、DMAP、N-メチルモルホリン等の有機塩基;アンバーリストA-21(ロームアンドハース社製)、AG1-X8(バイオラッド社製)、ポリビニルピリジン、モルホリノメチルポリスチレン等の固体塩基等が挙げられ、中でも、アルカリ金属水素化物が好ましく、水素化ナトリウム等がより好ましい。
化合物(VIII)は、例えば市販品として得ることができる。

化合物(I)は、化合物(XIa-1)を用い、製造法3の工程1と同様の操作を行うことにより製造することができる。
上記各製造法における中間体および目的化合物の単離、精製は、通常の有機合成で用いられる方法、例えばろ過、抽出、洗浄、乾燥、濃縮、結晶化、各種クロマトグラフィー等を適宜組み合わせて行うことができる。また、中間体においては、特に精製することなく次の反応に供することも可能である。
化合物(I)の塩を取得したい場合には、化合物(I)が塩の形で得られるときはそのまま精製すればよく、また遊離の形で得られるときは適当な溶媒に、化合物(I)を溶解または懸濁し、酸または塩基等を加えて塩を形成させて単離、精製すればよい。
以下、第1~第4表に本発明の化合物の具体例を示すが、本発明の範囲はこれらの化合物に限定されることはない。
なお、以下の表において、Meはメチルを表す。
試験例1:キヌレニン産生抑制活性
ジャーナル・オブ・バイオロジカル・ケミストリー(J. Biol. Chem.)、263巻、2041~2048頁(1988年)の方法に変更を加えて行った。ヒト胃がん由来細胞株KATO-IIIの培養には10 vol%のFBS(GIBCO、10091-148、lot.665285)を添加したRPMI 1640 (GIBCO、11875)を用いた。試験薬物のDMSO溶液1 μLを199 μLの培地で希釈し、384ウェルプレートの各ウェルに10 μLずつ加えた。次いで、31.25 ng/mLとなるようIFN-γ(Sigma、I-3265)を添加した培地にて50000個/mLで懸濁したKATO-IIIを40 μL各ウェルに加え(2000個/ウェル)、37℃、5%CO2存在下で96時間培養した。なおDMSOの最終濃度は、このアッセイで測定するキヌレニン量に単独で影響を与えないように0.1 vol%以下とした。この培養の後、各ウェルに10 μL の30 w/v% トリクロロ酢酸水溶液を加え、65℃で30分間インキュベートした。このプレートを2500×gで5分間遠心作業に供した後、上清15 μLを別の384ウェルプレートに回収した。回収した上清に15 μLの2 w/v% p-ジメチルアミノベンズアルデヒド酢酸溶液を加え、65℃で20分間インキュベートした後に、480 nmにおける吸光度を測定した。

式A1に従い各評価化合物の阻害率を求めた結果、例えば化合物1~21、23~28、30、32、34、36および37は、10 μmol/Lの濃度で80%以上の阻害率を示した。
本アッセイではKATO-III細胞をIFN-γで処理してIDOの発現を誘導し、培地中のキヌレニンを定量している。培地中のキヌレニン濃度は細胞内のIDO酵素活性に比例して増加することが知られている[例えば、ジャーナル・オブ・バイオロジカル・ケミストリー (J. Biol. Chem.)、263巻、2041~2048頁 (1988年)]。本発明における化合物は、キヌレニン産生抑制作用を示した。IDO阻害剤によりキヌレニン産生が抑制されることが知られている[例えば、ジャーナル・オブ・クリニカル・インベスチゲーション(J Clin Invest.)、117巻、5号、1147~1154頁(1988年)]。このことから、化合物(I)もIDOを阻害する作用を有していると推測される。
試験例2:HepaRG(登録商標)細胞を用いたCYP3A4誘導実験法
HepaRG細胞(Biopredic international社)を用いてCYP3A4誘導活性を評価した。HepaRG細胞は培地で0.72×106 cells/mLの濃度に懸濁し、96ウェルコラーゲンコートプレート(Biopredic international社)の各ウェルに100 μLずつ分注した。HepaRG細胞播種時の培地にはHepaRG Thawing and Seeding Medium 670(Biopredic international社)を、評価化合物添加時の培地にはHepaRG serum-free induction medium 650(Biopredic international社)を使用した。HepaRG細胞を播種した3日後に評価化合物による処理を開始した。96ウェルコラーゲンプレートの各ウェルに200 μLのHepaRG serum-free induction medium 650を添加後、ジメチルスルホキシド(DMSO)に溶解させた評価化合物を最終濃度が1 μmol/Lとなるように各ウェルに0.5 μL添加した。DMSOのみを添加したウェルをネガティブコントロールとした。評価化合物を含む培地は24時間毎に交換し、評価化合物による処理を72時間継続した。評価化合物処理開始後4日目にCYP3A4の典型的な基質であるミダゾラムを用い、代謝活性の変動によりCYP3A4誘導能を評価した。培地を吸引除去し、37 ℃に加温したPBS(150 μL/well)で各ウェルを2回洗浄後、37 ℃に加温した20 μmol/L ミダゾラム含有HepaRG serum-free induction medium 650を各ウェルに100 μL添加し、CO2インキュベーター内で2時間インキュベーションした。反応終了時点で各ウェルより70 μLの反応培地を回収し、内部標準(プロプラノロール、2 μmol/L)を添加したメタノール溶液70 μLと混合した。遠心分離(5000×g、4 ℃、10分)後の上清を分析サンプルとした。ミダゾラムの代謝活性はCYP3A4による代謝産物である1’-ヒドロキシミダゾラムの生成量を液体クロマトグラフタンデム質量分析 (liquid chromatograph-tandem mass spectrometer, LC-MS/MS)装置を用いて分析することによって評価した。LC装置にはAgilent 1100 series(Agilent technologies社)、MSMS装置にはAPI2000(AB sciex社)、およびオートサンプラにはHTC PAL(CTC analytics社)を用いた。分析用のカラムにはCAPCELL PAK C18 ACR column (3 μm, 3 mm I.D. x 35 mm、資生堂より購入)を使用した。移動相Aには10 mmol/L酢酸アンモニウム水溶液、移動相Bにはメタノールを用い、グラジエント条件は0 分(B=30%)→ 0.2分(B=30%)→ 2.2分(B=95%)→ 4分(B=95%)→ 4.01分(B=30%)→ 5分(B=30%)とした。流速は0.6 mL/min、サンプル注入量は30 μLとした。MSMS装置を用いて大気圧イオン化法によりイオンを検出した。検出は多重反応モニタリング(multiple reaction monitoring)法により行い、1’-ヒドロキシミダゾラムの検出イオンはm/z (前駆イオン>プロダクトイオン)= 342.1 > 168.2とした。
・誘導倍率の算出
ネガティブコントロール群(DMSO処理群)に対する評価化合物処理群における代謝活性の比を誘導倍率とする式A2に従い各評価化合物の誘導倍率を求め、結果を第5表に示す。
化合物(I)またはその薬理学的に許容される塩を投与する対象としては、上述したようなキヌレニン産生に関わる疾患の患者が好ましい。中でも、癌(腫瘍)、神経変性疾患、感染症、免疫性疾患等の患者が好適であり、癌 (腫瘍) 等の患者がより好適である。これらの疾患の患者は、公知の診断方法によって選択することができる。また、該疾患の発症を予防するために、上記化合物を、該疾患を発症する可能性がある哺乳類に投与することもできる。化合物(I)またはその薬理学的に許容される塩、またはこれを含有する組成物は、ヒト、ヒト以外の哺乳動物(例:マウス、ラット、ハムスター、モルモット、ウサギ、ネコ、イヌ、ブタ、ウシ、ウマ、ヒツジ、サル等) に対して、経口的または非経口的に投与することができる。
プロトン核磁気共鳴スペクトル(1H-NMR)においては、化合物および測定条件によっては交換性水素が明瞭には観測されないことがある。尚、シグナルの多重度の表記としては通常用いられるものを用いるが、brとは見かけ上、巾広いシグナルであることを表す。

参考例2-1
N-(3-クロロキノキサリン-2-イル)プロパン-1-スルホンアミド (化合物AA)
2,3-ジクロロキノキサリン(5.00 g, 25.1 mmol)、およびプロパン-1-スルホンアミド(3.09 g, 25.1 mmol)をDMSOに溶解し、炭酸カリウム(3.47 g, 25.1 mmol)を加え、150 ℃で1時間撹拌した。混合物に1%酢酸水溶液を加え、室温で3時間撹拌し、析出した固体をろ過し、得られた固体を水で洗浄し、ろ取した固体にジイソプロピルエーテルを加えてスラリー精製し、化合物AA (6.01 g, 収率84%)を得た。
参考例2-2
N-(3-クロロキノキサリン-2-イル)-N-((2-(トリメチルシリル)エトキシ)メチル)プロパン-1-スルホンアミド (化合物AB)
化合物AA (1.22 g, 4.28 mmol)をジクロロメタン(12.0 mL)に溶解し、ジイソプロピルエチルアミン(1.5 mL, 8.6 mmol)、および2-(クロロメトキシ)エチルトリメチルシラン(1.10 mL, 6.40 mmol)を加え、室温で30分間撹拌した。混合物に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸ナトリウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=5/1)で精製し、化合物AB (1.68 g, 収率94%)を得た。

2-メトキシエタンスルホンアミド(化合物AC1)
1-ブロモ-2-メトキシエタン (2.00 mL, 21.3 mmol) を水 (16 mL) に懸濁し、亜硫酸ナトリウム(2.95 g, 23.4 mmol) を加え、24時間還流した。混合物から減圧下で溶媒を留去し、残渣にクロロホルムを加えてスラリー精製し、白色固体 (6.11 g) を得た。白色固体に塩化チオニル (15.5 mL, 213 mmol)、およびDMF (0.082 mL, 1.06 mmol) を加え、100 ℃で3時間撹拌した。混合物から減圧下で溶媒を留去し、クロロホルムを加え、不溶物をろ過し、ろ液から減圧下で溶媒を留去し、得られた残渣に25%アンモニア水溶液(10 mL)を加え、室温で3時間撹拌した。混合物から減圧下で溶媒を留去し、クロロホルムを加え、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル= 3/7) で精製し、化合物AC1 (1.36 g, 収率46%) を得た。

化合物BA1をジクロロメタン(50.0 mL)に溶解し、4-メチルモルホリンN-オキシド (2.03 g, 17.4 mmol)、およびテトラプロピルアンモニウムペルルテナート (153 mg, 0.434 mmol)を加え、室温で2時間撹拌した。混合物をろ過して、ろ液から減圧下で溶媒を留去し、粗生成物の化合物BA2を得た。

tert-ブチル 4-(2,2,2-トリフルオロ-1-ヒドロキシエチル)ピペリジン-1-カルボキシレート (化合物BA)
化合物BA2をTHF (50.0 mL)に溶解し、(トリフルオロメチル)トリメチルシラン(2.08 mL, 13.0 mmol)、およびフッ化テトラブチルアンモニウム(1.0 mol/L THF溶液, 0.868 mL, 0.868 mmol)を加え、室温で15分間撹拌した後、さらにフッ化テトラブチルアンモニウム(1.0 mol/L THF溶液, 8.68 mL, 8.68 mmol)を加え、室温で45分間撹拌した。混合物に飽和塩化アンモニウム水溶液を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸ナトリウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=1/1)で精製し、化合物BA (1.47 g, 収率3工程60%)を得た。

化合物BC1 (7.60 g, 29.2 mmol)、シアン化亜鉛 (5.15 g, 43.8 mmol)、およびテトラキス(トリフェニルホスフィン)パラジウム (6.75 g, 5.84 mmol)をDMF (25 mL)に溶解し、窒素雰囲気下、80 ℃で15時間撹拌した。混合物をセライトろ過し、得られたろ液に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル=9/1)で精製し、化合物BC2 (5.0 g, 収率83%)を得た。

5-(ジエトキシメチル)-N’-ヒドロキシピコリンイミドアミド (化合物BC3)
化合物BC2 (2.3 g, 11.15 mmol) をエタノール (10 mL) に溶解し、50%ヒドロキシルアミン水溶液 (3.32 mL, 55.8 mmol) を加え、室温で30分間撹拌した。混合物から減圧下で溶媒を留去し、水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、白色固体の化合物BC3 (2.4 g, 収率 90%)を得た。

(E)-N’-(シクロプロパンカルボニルオキシ)-5-(ジエトキシメチル)ピコリンイミドアミド(化合物BC4)
化合物BC3 (400 mg, 1.672 mmol) をジクロロメタン (2 mL) に溶解し、ピリジン (0.20 mL, 2.508 mmol)、および塩化シクロプロパンカルボニル (210 mg, 2.006 mmol) を加え、室温で2時間撹拌した。混合物に飽和重曹水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、粗生成物の化合物BC4を得た。

化合物BC4にアセトニトリル(1 mL) を加え、フッ化テトラブチルアンモニウム(1.0 mol/L THF溶液, 1.67 mL, 1.672 mmol)を加え、室温で30分間撹拌した。混合物に飽和食塩水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、粗生成物の化合物BC5 を得た。

トルエン (3 mL) に、n-ブチルリチウム (2.6 mol/L, n-へキサン溶液, 0.94 mL, 2.43 mmol)、および化合物BC1 (575 mg, 2.21 mmol)のトルエン (2 mL) 溶液を-78 ℃で滴下した後、-78 ℃で30分間撹拌した。得られた懸濁液にTHF (2 mL) を加え、-78 ℃で30分間撹拌した後、DMF (0.51 mL, 6.63 mmol) を加え、ゆっくりと室温まで昇温させながら30分間撹拌した。混合物に飽和塩化アンモニウム水溶液、および水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸ナトリウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル= 9/1) で精製し、化合物BF1 (149 mg, 収率32%) を得た。

5-(5-(ジエトキシメチル)ピリジン-2-イル)オキサゾール (化合物BF2)
化合物BF1 (148 mg, 0.707 mmol) をメタノール(5 mL)に溶解し、トルエン-4-スルホニルメチルイソシアニド (345 mg, 1.77 mmol)、および炭酸カリウム (244 mg, 1.77 mmol) を加え、室温で8時間撹拌した。混合物に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、無水硫酸ナトリウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル= 7/3) で精製し、化合物BF2 (161 mg, 収率92%) を得た。

2-ブロモ-5-(1-(tert-ブチルジメチルシリルオキシ)-2,2,2-トリフルオロエチル)チアゾール(化合物BG2)
化合物BG1 (413 mg, 1.58 mmol) をジクロロメタン (4 mL) に溶解し、氷冷下、トリエチルアミン (1.63 mL, 11.8 mmol)、およびトリフルオロメタンスルホン酸tert-ブチルジメチルシリル (1.09 mL, 4.73 mmol)を加え、室温で12時間撹拌した。混合物に水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル= 5/1) で精製し、化合物BG2 (524 mg, 88%) を得た。

化合物BG2 (515 mg, 1.37 mmol) をTHF (5 mL) に溶解し、n-ブチルリチウム(1.65 mol/L, n-へキサン溶液, 1.24 mL, 2.05 mmol) を-78 ℃で滴下した後、-78 ℃で30分間撹拌した。混合物にN-メトキシ-N-メチルアセトアミド(1.38 mL, 13.7 mmol) のTHF溶液 (1 mL) を加え、-78 ℃で2時間撹拌した。混合物に飽和塩化アンモニウム水溶液、および水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=4/1)で精製し、化合物BG3 (437 mg, 94%) を得た。

1-(5-(2,2,2-トリフルオロ-1-ヒドロキシエチル)チアゾール-2-イル)エタノン(化合物BG)
化合物BG3 (356 mg, 1.05 mmol) をTHF (3 mL) に溶解し、酢酸 (0.072 mL, 1.26 mmol)、およびフッ化テトラブチルアンモニウム (1.0 mol/L, THF溶液, 1.26 mL, 1.26 mmol) を加え、室温で1時間撹拌した。混合物に飽和炭酸ナトリウム水溶液を加え、酢酸エチルで抽出した。有機層を飽和塩化アンモニウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥し、ろ過を行い、ろ液から減圧下で溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=1/1)で精製し、化合物BG (214 mg, 91%) を得た。

化合物CA1 (140 mg, 0.263 mmol)を酢酸エチル(1 mL)に溶解し、4 mol/L 塩化水素/酢酸エチル溶液 (1 mL)を加え、50 ℃で2時間撹拌した。混合物から、溶媒を減圧下で留去し、残渣にジイソプロピルエーテルを加えてスラリー精製し、化合物CA2 (83.5 mg, 収率68%)を得た。

実施例28と同様にして化合物LA2 (940 mg) を分取高速液体クロマトグラフィー[CHIRALPAC(登録商標) IC (ダイセル化学工業)、粒子径 5 μm、2 cm (内径)×25 cm (長さ)、クロロホルム/エタノール/酢酸=98/2/0.05 (v/v)、流速6 mL/min、カラムオーブン温度40 ℃、検出波長 277 nm]で分離することにより、エナンチオマーである化合物30 (480 mg)、および化合物31 (460 mg)を得た。なお、保持時間が15.8分である化合物を化合物30、保持時間が18.3分である化合物を化合物31とした。
化合物30: ESIMS m/z: 510 (M + H)+.
化合物31: ESIMS m/z: 510 (M + H)+.
(R)-N-(3-(1-(2-アセチルチアゾール-5-イル)-2,2,2-トリフルオロエトキシ)キノキサリン-2-イル)プロパン-1-スルホンアミド (化合物33)
実施例28と同様にして化合物MA2 (500 mg)を分取高速液体クロマトグラフィー[CHIRALPAC IC(ダイセル化学工業)、粒子径5 μm、2 cm (内径)×25 cm (長さ)、n-ヘキサン/2-プロパノール/エタノール=75/20/5(v/v)、流速5.7 mL/min、カラムオーブン温度40 ℃、検出波長 285 nm]で分離することにより、エナンチオマーである化合物32 (250 mg)、および化合物33 (250 mg)を得た。なお、保持時間が29.7分である化合物を化合物32、保持時間が34.4分である化合物を化合物33とした。
化合物32: ESIMS m/z: 475 (M + H)+.
化合物33: ESIMS m/z: 475 (M + H)+.
(S)-5-(4-(2,2,2-トリフルオロ-1-(3-(プロピルスルホンアミド)キノキサリン-2-イルオキシ)エチル)ピペリジン-1-イル)ピコリンアミド (化合物35)
実施例28と同様にして化合物5 (430 mg)を分取高速液体クロマトグラフィー[CHIRALCEL OZ-H (ダイセル化学工業)、粒子径 5 μm、5 cm (内径)×25 cm (長さ)、メタノール/酢酸=100/0.1 (v/v)、流速35 mL/min、カラムオーブン温度40 ℃、検出波長 246 nm]で分離することにより、エナンチオマーである化合物34 (210 mg)、および化合物35 (220 mg)を得た。なお、保持時間が33.5分である化合物を化合物34、保持時間が53.4分である化合物を化合物35とした。
化合物34: ESIMS m/z: 553 (M + H)+.
化合物35: ESIMS m/z: 553 (M + H)+.
(R)-N-(3-(1-(6-(5-シクロプロピル-1,2,4-オキサジアゾール-3-イル)ピリジン-3-イル)-2,2,2-トリフルオロエトキシ)キノキサリン-2-イル)-2-メトキシエタンスルホンアミド (化合物37)
実施例28と同様にして化合物23 (440 mg)を分取高速液体クロマトグラフィー[CHIRALCEL OJ-H (ダイセル化学工業)、粒子径 5 μm、2 cm (内径)×25 cm (長さ)、メタノール/アセトニトリル/酢酸=95/5/0.1 (v/v)、流速5.7 mL/min、カラムオーブン温度40 ℃、検出波長 260 nm]で分離することにより、エナンチオマーである化合物36 (220 mg)、および化合物37 (220 mg)を得た。なお、保持時間が20.8分である化合物を化合物36、保持時間が32.5分である化合物を化合物37とした。
化合物36: ESIMS m/z: 551 (M + H)+.
化合物37: ESIMS m/z: 551 (M + H)+.
次に、WO2010/053182記載のキヌレニン産生阻害剤である化合物312、および313について、X線結晶構造解析による各化合物の絶対立体配置の決定を行なった。また、化合物312、および313の絶対立体配置とキヌレニン産生抑制活性の関係から、これらと構造類似である本願化合物(I)の各エナンチオマーの絶対立体配置の推定をしたので実施例33で説明する。
生成した結晶を、最大長が0.3 mm程度になるように切断して、イメージングプレート(IP)式X線回折計R-AXIS RAPID-F(株式会社リガク)に取り付け、Cu Kα線(λ=1.54187 Å, 1 Å=10-10 m)を用いて回折測定を行った。プログラムRAPID AUTO バージョン2.40(株式会社リガク)およびCrystalStructureバージョン3.8.2(株式会社リガク)を用いて、IP画像よりの回折強度の抽出と結晶によるX線吸収の補正を行った。
・結晶構造解析
直接法プログラムSHELXS-97(Sheldrick GM. A short history of SHELX. Acta Crystallogr. Sect. A. 2008; 64(1): 112-22.)を用いて初期構造を決定した後、構造精密化プログラムSHELXL-97(Sheldrick GM. A short histry of SHELX. Acta Crystallogr. Sect. A. 2008; 64(1): 112-22.)を用いて、最小二乗法による構造精密化を行った。結晶の絶対構造の判定は絶対構造パラメータ法(Flack HD. On enantiomorph-polarity estimation. Acta Crystallogr. Sect. A. 1983; 39(6):876-81.)を用いて行った。結晶構造図はMercury バージョン2.4.6(Cambridge Crystallographic Data Centre)を用いて作成した。
化合物313の結晶データと構造解析結果の詳細を表8、原子座標と等価等方性変位因子(非水素原子の場合)または等方性変位因子(水素原子の場合)を表9、および結晶中における分子構造を図2に示す。この結晶では、C1”の絶対配置が(S)-型となる構造を採用した場合にx = 0.01(2)となった。
以上の結果から、下記式(I)
製剤例1
常法により、次の組成からなる錠剤を調製する。化合物21 (40 g)、乳糖286.8 g、および馬鈴薯澱粉60 gを混合し、これにヒドロキシプロピルセルロースの10%水溶液120 gを加える。この混合物を常法により練合し、造粒して乾燥させた後、整粒し打錠用顆粒とする。これにステアリン酸マグネシウム1.2 gを加えて混合し、径8 mmの杵をもった打錠機(菊水社製RT-15型)で打錠を行って、錠剤(1錠あたり活性成分20 mgを含有する)を得る。
常法により、次の組成からなるカプセル剤を調製する。化合物21, 200 g、アビセル995 gおよびステアリン酸マグネシウム5 gを常法により混合する。この混合物をカプセル充填機(Zanasi社製、LZ-64型) により、ハードカプセル4号 (1カプセルあたり120 mg容量)に充填し、カプセル剤(1カプセルあたり活性成分20 mgを含有する)を得る。
常法により、次の組成からなる注射剤を調製する。化合物25 (1 g) およびD-マンニトール (5 g) を注射用蒸留水に添加して混合し、さらに塩酸、および水酸化ナトリウム水溶液を添加してpHを6に調整した後、注射用蒸留水で全量を1000 mLとする。得られた混合液をガラスバイアルに2 mLずつ無菌的に充填して、注射剤(1バイアルあたり活性成分2 mgを含有する)を得る。

- R2が、式(II)である請求項1記載の含窒素複素環化合物またはその薬理学的に許容される塩。
- R2Aが、式(III-B)である請求項5記載の含窒素複素環化合物またはその薬理学的に許容される塩。
- R2Aが、式(IV)である請求項5記載の含窒素複素環化合物またはその薬理学的に許容される塩。
- R3がトリフルオロメチルである請求項1~7のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩。
- 下記化合物群から選択される請求項1記載の含窒素複素環化合物またはその薬理学的に許容される塩:
N,N-ジメチル-2-(4-(2,2,2-トリフルオロ-1-(3-(プロピルスルホンアミド)キノキサリン-2-イルオキシ)エチル)ピペリジン-1-イル)イソニコチンアミド (化合物1)、
N,N-ジメチル-2-(4-(2,2,2-トリフルオロ-1-(3-(プロピルスルホンアミド)キノキサリン-2-イルオキシ)エチル)ピペリジン-1-イル)ニコチンアミド (化合物2)、
N,N-ジメチル-6-(4-(2,2,2-トリフルオロ-1-(3-(プロピルスルホンアミド)キノキサリン-2-イルオキシ)エチル)ピペリジン-1-イル)ピコリンアミド (化合物3)、
N-(3-(1-(1-(6-シアノピリジン-3-イル)ピペリジン-4-イル)-2,2,2-トリフルオロエトキシ)キノキサリン-2-イル)プロパン-1-スルホンアミド (化合物4)、
5-(4-(2,2,2-トリフルオロ-1-(3-(プロピルスルホンアミド)キノキサリン-2-イルオキシ)エチル)ピペリジン-1-イル)ピコリンアミド (化合物5)、

- 請求項1~12のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩の有効量を投与する工程を含むキヌレニン産生抑制方法。
- キヌレニン産生抑制剤の製造のための請求項1~12のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩の使用。
- キヌレニン産生抑制に使用するための請求項1~12のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩。
- 請求項1~12のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩を有効成分として含有するキヌレニン産生に関わる疾患の予防または治療剤。
- 請求項1~12のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩の有効量を投与する工程を含む、キヌレニン産生に関わる疾患の予防または治療方法。
- キヌレニン産生に関わる疾患の予防または治療剤の製造のための請求項1~12のいずれかに記載の含窒素複素環化合物またはその薬理学的に許容される塩の使用。
