Description
本発明は、有機合成反応の試薬として、或いは、医薬、農薬、電子材料及び発光材料の中間体化合物として有用なトリフルオロメチルピリジントリオールボレート塩に関する。
特許文献1には、一定の化学構造を有する有機トリオールボレート塩を含んでなる有機合成反応用試薬及びそれらの製造方法が開示されている。しかしながら、特許文献1には、トリフルオロメチル基を有するピリジントリオールボレート塩は記載されていない。
非特許文献1には、種々の2-ピリジルイミノジアセテート-ボロネートが開示されている。しかしながら、非特許文献1にも、トリフルオロメチル基を有するピリジントリオールボレート塩は記載されていない。

本発明の目的は、トリフルオロメチル基を有するピリジントリオールボレート塩を提供することにある。本化合物は、有機合成反応の試薬として、或いは、医薬、農薬、液晶等の電子材料及び発光材料用の希土類錯体などの中間体化合物として有用であると考えられるが、製造が困難であるなどの理由から、従来は製造されてこなかった。
本発明者らは、トリフルオロメチル基を有するピリジントリオールボレート塩の製造時における中間生成物の安定性に着目して反応条件等を検討した結果、一定の条件においてのみ目的の化合物が製造できるとの知見を得て、本発明を完成した。
すなわち、本発明は、下記(1)~(4)である。

本発明によれば、有機合成反応の試薬として、或いは、医薬、農薬、電子材料及び発光材料の中間体化合物として有用な上記式(I)のトリフルオロメチルピリジントリオールボレート塩(以下、本発明化合物)が提供される。本発明化合物は、脱水三量化しないため、純粋な化合物として得ることができる。また、塩基による活性化を必要とせず、安定かつ高活性な化合物として得ることができるため、有機合成反応(特にクロスカップリング反応)用試薬として特に優れている。
本発明に係るトリフルオロメチルピリジントリオールボレート塩は、下記式(I)で表わされる化合物である。

尚、ハロアルキル基とは上記アルキル基の水素原子がハロゲン原子で置換された基を意味する。このハロアルキル基は、全ての水素原子がハロゲン原子に置換されていてもよく、一部の水素原子のみが置換されていてもよい。

式(I)における、用語「アリール基」は、炭素数6~10のものが好ましい。具体的には、例えばフェニル基、ナフチル基などである。

式(I)における、用語「ヘテロ環」は、単環式複素環の他、縮合複素環が含まれる。単環式複素環としては、例えばオキシラニル基などの3員複素環;フリル基、テトラヒドロフリル基、チエニル基、ピロリル基、ピロリニル基、ピロリジニル基、ジオキソラニル基、オキサゾリル基、イソキサゾリル基、チアゾリル基、イソチアゾリル基、イミダゾリル基、イミダゾリニル基、イミダゾリジニル基、ピラゾリル基、ピラゾリニル基、ピラゾリジニル基、トリアゾリル基、オキサジアゾリル基、チアジアゾリル基、テトラゾリル基などの5員複素環;ピラニル基、ピリジル基、ピペリジニル基、ジオキサニル基、オキサジニル基、モルホリニル基、チアジニル基、ピリダジニル基、ピリミジニル基、ピラジニル基、ピペラジニル基、トリアジニル基などの6員複素環が挙げられる。これら単環式複素環の中では、O、S及びNからなる群より選ばれる少なくとも1種の原子を1~4個含有する5若しくは6員複素環が望ましい。縮合複素環としては、例えばベンゾフラニル基、イソベンゾフラニル基、ジヒドロベンゾフラニル基、ジヒドロイソベンゾフラニル基、ベンゾチエニル基、イソベンゾチエニル基、ジヒドロベンゾチエニル基、ジヒドロイソベンゾチエニル基、テトラヒドロベンゾチエニル基、インドリル基、イソインドリル基、ベンゾオキサゾリル基、ベンゾチアゾリル基、インダゾリル基、ベンズイミダゾリル基、ベンゾジオキソラニル基、ベンゾジオキサニル基、クロメニル基、クロマニル基、イソクロマニル基、クロモニル基、クロマノニル基、キノリル基、イソキノリル基、シンノリニル基、フタラジニル基、キナゾリニル基、キノキサリニル基、インドリジニル基、キノリジニル基、イミダゾピリジル基、ナフチリジニル基、プテリジニル基、ジヒドロベンゾオキサジニル基、ジヒドロベンゾオキサゾリノニル基、ジヒドロベンゾオキサジノニル基、ベンゾチオキサニル基などが挙げられる。これら縮合複素環の中では、O、S及びNからなる群より選ばれる少なくとも1種の原子を1~4個含有する8~10員縮合複素環が望ましい。これらヘテロ環は置換基を有していてもよい。その置換基としては、前記アリール基と同様のものが挙げられる。
式(I)における、用語「ハロゲン原子」は、例えばフッ素原子、塩素原子、臭素原子、ヨウ素原子などである。置換基としてのハロゲン原子の数は1又は2以上であってよく、2以上の場合、各ハロゲン原子は同一でも相異なってもよい。また、ハロゲン原子の置換位置はいずれの位置でもよい。
本発明において、式(I)で表わされるトリフルオロメチルピリジントリオールボレート塩(本発明化合物)は、下記式(I-1)で表わされるものが好ましい。
[式中、R1、R2及びnは、式(I)におけるものと同じ意味を示す。]
本発明において、式(I)および式(I-1)中、R1は、ハロゲン原子、アルキル基、ハロアルキル基、アリール基、ヘテロ環、-NR5R6、-OR5又は-SR5であることが好ましく、R5はアルキル基であり、R6はアルキル基であることが好ましい。
また、本発明において、式(I)および式(I-1)中、nは、0又は1の整数である場合が好ましい。
本発明化合物は、例えば、以下の方法で製造することができる。ここで、式(II)中のXはハロゲン原子を表し、R1及びnは前述の通りである。R1の種類によっては、保護される工程又は脱保護される工程を含んでもよい。
ハロゲン化トリフルオロメチルピリジンを反応溶媒に溶解し、有機リチウム(organolithium)を添加してリチオ化する。さらにトリアルキルボレート(trialkyl boronate)を添加後、トリオール(triol)を添加して反応させる方法が挙げられる。
また、ハロゲン化トリフルオロメチルピリジンとトリアルキルボレートを反応溶媒に溶解し、有機リチウムを添加後、トリオールを添加して反応させる方法が挙げられる。
また、ハロゲン化トリフルオロメチルピリジンとトリアルキルボレートを反応溶媒に溶解し、有機リチウムを添加後、トリオールを添加して反応させる方法が挙げられる。
本発明化合物の製造方法に用いられる反応溶媒としては、反応原料及び反応生成物に対して不活性であれば特に限定されないが、例えばn-ヘキサン、n-ヘプタン、シクロヘキサンなどの炭化水素類;ベンゼン、トルエン、キシレンなどの芳香族炭化水素類;ジエチルエーテル、ジメトキシエタン、ジエトキシエタン、テトラヒドロフラン、シクロペンチルメチルエーテルなどのエーテル類などの有機溶媒が挙げられる。これらは単独で用いても、2種以上を適宜組み合わせて用いてもよい。また、使用する反応溶媒やその組み合わせにより反応の選択性を変えることができる。反応溶媒は、式(II)の化合物に対して通常1~20倍量を用いるが、望ましくは1~15倍量である。
有機リチウムの具体例としては、例えばメチルリチウム、エチルリチウム、プロピルリチウム、プロペニルリチウム、イソプロピルリチウム、ブチルリチウム、ブテニルリチウム、ブチニルリチウム、イソブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウム、ペンチルリチウム、ペンテニルリチウム、イソペンテニルリチウム、イソペンチルリチウム、ネオペンチルリチウム、へキシルリチウム、ヘキセニルリチウム、ヘキシニルリチウム、へプチルリチウム、オクチルリチウム、ノニルリチウム、デシルリチウム、シクロプロピルリチウム、シクロへキシルリチウム、ベンジルリチウム、フェニルリチウム、トリルリチウム、m-アニシルリチウム、キシリルリチウム、メシチルリチウム、ペンタフルオロフェニルリチウム、ビフェニルリチウム、チオフェニルリチウム、ピリジルリチウム、インデニルリチウム、ナフチルリチウム、フリルリチウムなどが挙げられる。有機リチウムは、式(II)の化合物に対して通常1~4倍モル、望ましくは1~2.5倍モル用いることができる。
トリアルキルボレートの具体例としては、例えばトリメチルボレート、トリエチルボレート、トリプロピルボレート、トリイソプロピルボレート、トリブチルボレート、トリイソブチルボレート、トリtert-ブチルボレート、トリペンチルボレート、トリヘキシルボレート、トリシクロヘキシルボレート、トリフェニルボレート、トリベンジルボレート、トリメトキシエチルボレートなどが挙げられる。トリアルキルボレートは、式(II)の化合物に対して通常1~2倍モル、望ましくは1~1.5倍モル用いることができる。
トリオールの具体例としては、例えば1,1,1-トリス(ヒドロキシメチル)エタン、1,1,1-トリス(ヒドロキシメチル)プロパンなどが挙げられる。トリオールは式(II)の化合物に対して通常1~2倍モル、望ましくは1~1.5倍モル用いることができる。
本発明化合物の製造の際の反応温度は、通常-100℃~200℃、望ましくは-80~100℃である。その反応時間は、用いた原料が消費され目的のトリフルオロメチルピリジントリオールボレート塩が生成するまでの時間と同等であれば特に問題ないが、通常1~24時間、望ましくは1~6時間である。
なお、特許文献1に記載の2-ピリジントリオールボレート塩の製造方法(実施例5)と同様の方法では、トリフルオロメチル基を有する本発明化合物を得られなかった。

次に本発明の実施例を記載するが、本発明はこれらに限定されるものではない。まず、本発明化合物の合成例を記載する。
合成例1
2-ブロモ-4-(トリフルオロメチル)ピリジン0.5g(2.2mmol)とトリイソプロピルボレート0.5mL(2.2mmol)とテトラヒドロフラン4mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに2.5M n-ブチルリチウム/n-ヘキサン溶液0.9mL(2.2mmol)をゆっくり加え、-78℃で60分間攪拌した。その後、室温中で85分間攪拌後、1,1,1-トリス(ヒドロキシメチル)エタン 0.26g(2.2mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを10mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.49gの目的物(化合物No.A-1)を得た(収率78.8%)。
2-ブロモ-4-(トリフルオロメチル)ピリジン0.5g(2.2mmol)とトリイソプロピルボレート0.5mL(2.2mmol)とテトラヒドロフラン4mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに2.5M n-ブチルリチウム/n-ヘキサン溶液0.9mL(2.2mmol)をゆっくり加え、-78℃で60分間攪拌した。その後、室温中で85分間攪拌後、1,1,1-トリス(ヒドロキシメチル)エタン 0.26g(2.2mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを10mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.49gの目的物(化合物No.A-1)を得た(収率78.8%)。
合成例2
2-ブロモ-5-(トリフルオロメチル)ピリジン0.5g(2.2mmol)とn-ヘキサン8mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに1.65M n-ブチルリチウム/n-ヘキサン溶液1.3mL(2.2mmol)をゆっくり加え、-78℃で50分間攪拌した。続いてトリイソプロピルボレート0.5mL(2.2mmol)を加え、-78℃で45分間攪拌した後、テトラヒドロフランを8mL加え、引き続き55分間攪拌した。その後、室温中で25分間攪拌した後、1,1,1-トリス(ヒドロキシメチル)エタン0.26g(2.2mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを15mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.37gの目的物(化合物No.B-1)を得た(収率59.9%)。
2-ブロモ-5-(トリフルオロメチル)ピリジン0.5g(2.2mmol)とn-ヘキサン8mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに1.65M n-ブチルリチウム/n-ヘキサン溶液1.3mL(2.2mmol)をゆっくり加え、-78℃で50分間攪拌した。続いてトリイソプロピルボレート0.5mL(2.2mmol)を加え、-78℃で45分間攪拌した後、テトラヒドロフランを8mL加え、引き続き55分間攪拌した。その後、室温中で25分間攪拌した後、1,1,1-トリス(ヒドロキシメチル)エタン0.26g(2.2mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを15mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.37gの目的物(化合物No.B-1)を得た(収率59.9%)。
合成例3
2-ブロモ-6-(トリフルオロメチル)ピリジン0.5g(2.2mmol)とトリイソプロピルボレート0.5mL(2.2mmol)とテトラヒドロフラン4mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに2.5M n-ブチルリチウム/n-ヘキサン溶液0.9mL(2.2mmol)をゆっくり加え、-78℃で60分間攪拌した。その後、室温中で90分間攪拌後、1,1,1-トリス(ヒドロキシメチル)エタン0.26g(2.2mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを10mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.45gの目的物(化合物No.C-1)を得た(収率72.4%)。
2-ブロモ-6-(トリフルオロメチル)ピリジン0.5g(2.2mmol)とトリイソプロピルボレート0.5mL(2.2mmol)とテトラヒドロフラン4mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに2.5M n-ブチルリチウム/n-ヘキサン溶液0.9mL(2.2mmol)をゆっくり加え、-78℃で60分間攪拌した。その後、室温中で90分間攪拌後、1,1,1-トリス(ヒドロキシメチル)エタン0.26g(2.2mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを10mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.45gの目的物(化合物No.C-1)を得た(収率72.4%)。
合成例4
2,6-ジブロモ-4-(トリフルオロメチル)ピリジン0.5g(1.6mmol)とトリイソプロピルボレート0.37mL(1.6mmol)とテトラヒドロフラン4mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに2.5M n-ブチルリチウム/n-ヘキサン溶液0.64mL(1.6mmol)をゆっくり加え、-78℃で64分間攪拌した。その後、室温中で96分間攪拌後、1,1,1-トリス(ヒドロキシメチル)エタン0.19g(1.6mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを10mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.38gの目的物(化合物No.A-2)を得た(収率64.4%)。
2,6-ジブロモ-4-(トリフルオロメチル)ピリジン0.5g(1.6mmol)とトリイソプロピルボレート0.37mL(1.6mmol)とテトラヒドロフラン4mLとを加えた反応容器内を、窒素雰囲気下にした後、-78℃まで冷却した。そこに2.5M n-ブチルリチウム/n-ヘキサン溶液0.64mL(1.6mmol)をゆっくり加え、-78℃で64分間攪拌した。その後、室温中で96分間攪拌後、1,1,1-トリス(ヒドロキシメチル)エタン0.19g(1.6mmol)を加え、還流温度まで加熱後、還流下で30分間攪拌した。室温まで冷却後、n-ヘプタンを10mL加えて結晶を析出させた後、ろ過にて結晶を回収し、減圧下で乾燥する事で0.38gの目的物(化合物No.A-2)を得た(収率64.4%)。
合成例5
2-ブロモ-6-メトキシ-4-(トリフルオロメチル)ピリジンを用いて合成例4と同様の方法により化合物No.A-3を得た(収率51.2%)。

参考例1(本発明化合物No.B-1を利用したクロスカップリング反応)
化合物No.B-1 140mg(0.5mmol)と4-ブロモ-安息香酸メチル36mg(0.17mmol)とジクロロパラジウム2mg(0.011mmol)とジフェニルホスフィノプロパン1.4mg(0.0034mmol)、ヨウ化銅3mg(0.017mmol)とジメチルホルムアミド1.2mLとを反応容器に加え、窒素雰囲気下80℃で24時間攪拌した。室温まで冷却した後、飽和塩化アンモニウム水溶液を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄後、硫酸ナトリウムで乾燥し、濃縮してシリカゲルカラムクロマトグラフィー(溶離液:n-ヘプタン/酢酸エチル=4/1)で精製して、白色結晶の4-(5-(トリフルオロメチル)ピリジン-2-イル)安息香酸メチル14.7mg(収率31.2%、融点98.6~100.2℃)を得た。

Claims (4)
- 下記式(I):
で表されるトリフルオロメチルピリジントリオールボレート塩。 - R1がハロゲン原子、アルキル基、ハロアルキル基、アリール基、ヘテロ環、-NR5R6、-OR5又は-SR5であり、R5がアルキル基であり、R6がアルキル基である、請求項1又は2に記載のトリフルオロメチルピリジントリオールボレート塩。
- nが0又は1の整数である、請求項1又は2に記載のトリフルオロメチルピリジントリオールボレート塩。
