하지만, 위 방법에 의해 제조된 젖산은 여전히 발효공정에 기인한 금속, 다당류 및 단백질과 같은 불순물을 다량 함유하고 있기 때문에 PLA 중합급 락타이드를 제조하는데 부적합하다. 따라서, 고순도 락타이드 제조를 위해서 추가 전처리 과정이 필요하며, 이를 위해 통상적으로 상기 산성화된 젖산으로부터 알콜과의 에스테르화 반웅을 통해 알킬 락테이트를 제조하고 증류를 통해 알킬 락테이트를 분리한 후, 산촉매 존재하에서 가수분해하여, 알콜 및 수분을 분리함으로써 순수한 ¾산을 제조하게 된다. 하지만, 상기 락타이드 제조 공정은 알킬 락테이트 제조 후 가수분해 및 증류 공정을 추가로 필요로 하기 때문에 너무 복잡하다는 단점이 있다. 한편, 락타이드를 제조하는 또 다른 방법으로 미국 특허 제 6ᅳ 569, 989호 및 일본 특허 제 209, 370호 등에 99% 이상의 순도의 알킬 락테이트를 유기 주석 (Organic Sn)계 촉매를 이용하여 예비중합 및 해중합 공정을 통하여 락타이드로 전환하는 기술이 소개되어 있다. 하지만, 실제 젖산 발효액을 이용하여 알킬 락테이트를 제조하는 경우 물과 알킬 락테이트의 공비 (azeotrope) 특성에 의해 99% 이상의 고순도 알킬 락테이트를 단순 증류공정에 의해 얻는 것이 용이하지 않다. 만일, 젖산 발효액으로부터 알킬 락테이트 제조 후 별도의 수분제거 공정을 거치지 않고 수분이 다량 함유된 상태의 알킬 락테이트를 기 공지된 유기주석계 촉매를 이용하여 예비중합시키는 경우, 유기주석계 촉매가 가수분해로 인해 비활성화되는 문제점이 있다. 따라서, 젖산 발효액으로부터 제조한 알킬 락테이트로부터 유기주석계 촉매를 이용하여 락타이드를 제조하기 위해서는 예비중합 단계 전에 제을라이트와 같은 흡착제를 이용한 탈수 처리, 공비 증류 또는 투과증발과 같은 물을 제거해주는 별도의 공정을 필요로 하게 된다. 이러한 추가 처리 공정은 젖산 발효액으로부터 락타이드까지의 제조 생산성을 떨어뜨려 락타이드 제조 원가의 상승을 초래하게 된다. 이에 본 발명자들은 젖산 발효액으로부터 얻은, 수분이 다량 함유된 알킬 락테이트로부터 수분 제거 공정 없이 락타이드를 제조하기 위한 방법을 연구하던 중, 알킬 락테이트의 증합에 납 (Pb)계 촉매를 사용함으로써 상기 문제를 해결할 수 있음을 확인하고 본 발명을 완성하였다. 발명의 요약
따라서, 본 발명의 목적은 젖산 발효액과 알콜 간의 에스테르화 반응에 의해 제조되는, 수분이 다량 함유된 알킬 락테이트를 직접 사용하여 간단하면서도 경제적으로 고순도의 락타이드를 제조할 수 있는 방법을 제공하는 것이다.
상기 목적에 따라, 본 발명은 1) 알킬 락테이트를 납 (F )계 촉매 하에서 중합하여 젖산 을리고머를 제조하는 단계; 및 2) 상기 젖산 올리고머를 해중합하여 락타이드를 수득하는 단계를 포함하는, 알킬 락테이트로부터 락타이드를 제조하는 방법을 제공한다.
본 발명에 따른 락타이드 제조 방법은 젖산 발효액에서 얻어지는 수분이 다량 함유된 알킬 락테이트로부터 별도의 수분 제거 공정, 즉, 제올라이트를 이용한 수분흡착 제거 및 투과증발 등을 거치지 않고도 직접 예비중합 및 해중합 과정을 통해 락타이드를 제조할 수 있기 때문에 공정이 간편하며, 또한, 락타아드 제조를 위해 알킬 락테이트 제조 후 젖산 제조를 위한 추가적인 가수분해과정이 필요하지 않으며, 나아가 가수분해에 의해 젖산을 수득한 후 락타이드를 제조하는 기존 방법보다 저온에서 락타이드의 제조가 가능한 장점이 있다.

도 2는 젖산 발효액으로부터 락타이드를 제조하는 공정에 본 발명의 방법을 적용한 예의 모식도이다.
발명의 상세한설명 이하, 본 발명을 상세히 설명한다. 본 발명은 1) 알킬 락테이트를 납 (Pb)계 촉매 하에서 중합하여 젖산 을리고머를 제조하는 단계; 및 2) 상기 젖산 올리고머를 해중합하여 락타이드를 수득하는 단계를 포함하는, 알킬 락테이트로부터 락타이드를 제조하는 방법을 제공한다. 본 발명에 따른 단계 1은 알킬 락테이트를 납 (Pb)계 촉매 하에서 중합하여 젖산 올리고머를 제조하는 단계이다.
상기 중합 단계에 사용되는 알킬 락테이트는 C广 C14 알킬 락테이트 중 어느 하나 이상일 수 있으며, 바람직하게는, 중합 속도를 고려할 때 메틸 락테이트, 에틸 락테이트, 프로필 락테이트 및 부틸 락테이트로 이루어지는 군으로부터 선택되는 어느 하나 이상일 수 있고, 보다 바람직하게는, 젖산 발효액으로부터 알킬 락테이트 생성 속도 향상 및 후속 단계에서 증류에 의한 분리의 용이성을 고려할 때 메틸 락테이트 또는 에틸 락테이트일 수 있다.
상기 본 발명에 사용된 알킬 락테이트는 시판중인 제품일 수 있다. 또한, 상기 본 발명에 사용된 알킬 락테이트는 당업계에 알려진 공정을 이용하여 젖산 발효액으로부터 얻어진 것, 예를 들어, 젖산 발효액을 알콜과 에스테르화 반웅시켜 얻어진 것일 수 있으며, 10% 이하의 수분 함량을 갖는 것일 수 있다. 이하 설명할 촉매의 작용으로 인해, 상기 수분을 함유한 알킬 락테이트도 추가의 수분 제거 공정 없이 직접 사용될 수 있다. 한편, 본 발명은 상기 중합 단계에서 납 (Pb)계 촉매를 사용하는 것을 특징으로 한다. 본 단계에서 사용되는 납 (Pb)계 촉매는 다양한 형태의 화합물이 사용될 수 있으며, 바람직하게는 PbO, Pb304, Pb02, Pb(N03)2, PbCl2, Pb(0Ph)2( Pb(0Ac)2 또는 PbO (쉘) /실리카 (코어)를 사용하는 것이 유리하다.
상기 납 (Pb)계 촉매는 알킬 락테이트 대비 0.05 내지 10 중량 %의 양으로 사용되는 것이 바람직하며, 0.5 내지 3 중량 %의 양이 보다 바람직하다. 촉매의 양이 0.5% 이내인 경우 중합 속도가 느린 단점이 있으며, 10% 이상인 경우 중합 속도 조절이 힘들고 해중합 단계에서 락타이드의 광학 순도가 저하되는 문제점이 있다. 순수한 알킬 락테이트의 경우 모노부틸주석 (monobutyltin) 또는 주석 옥틸레이트 (tin octylate)와 같은 유기주석계 촉매를 이용하여 중합하는 기술은 기존에 공지되어 있다 (미국 특허 제 6, 569, 989호 참조). 하지만 유기주석계 촉매의 사용시 수분과의 반응에 의해 촉매가 비활성화되는 문제점이 있다. 한편, SnO와 같은 무기주석계 촉매의 경우에도 수분에 대한 안정성이 있으나 젖산과 달리 알킬 락테이트에 대해서는 촉매 활성이 거의 없기 때문에 사용하기 어려운 단점이 있다. 이에 본 발명에서는 납 (Pb)계 촉매를 사용하는 것을 특징으로 하며, 본 발명자들은 납 (Pb)계 촉매를 적용한 결과 수분이 존재하는 상황에서도 알킬 락테이트를 중합할 수 있는 탁월한 능력이 있음을 확인하였다. 본 발명에 따른 단계 1의 중합 과정은 120 내지 180°C 범위의 온도 및
100 내지 500 토르 (13 내지 67 kPa)의 압력하에서 수행하는 것이 바람직하다. 상기 온도 및 압력은 사용하는 알킬 락테이트의 종류에 따라 달라질 수 있다. 본 발명에 따른 단계 2는 상기 젖산 올리고머를 해중합 (depolymerization)하여 락타이드를 수득하는 단계이다. 상기 해중합 과정은 당해 기술분야의 숙련자에게 널리 알려져 있다. 상기 해중합 과정은 160 내지 23CTC 범위의 해중합 온도에서 5 내지 100 토르 (0.7 내지 13.5 kPa)에서 수행하는 것이 바람직하며, 16CTC 미만인 경우 해중합 속도가 느린 문제점이 있으며 230°C를 초과하는 경우 생성되는 락타이드의 광학 순도가 저하되는 문제점이 있다. 본 발명의 방법을 이용한 결과, 90% 이상의 순도를 갖는 락타이드를 얻을 수 있으며, 단계 1에서 주입한 납 (Pb)계 촉매는 최종 락타이드에서 전혀 검출되지 않는다. 이하, 본 발명을 하기 실시예에 의해 더욱 구체적으로 설명한다.
단, 하기의 실시예는 본 발명을 예시하기 위한 것일뿐, 본 발명의 내용이 하기 실시예에 의해 한정되는 것은 아니다. 실시예 1: 메틸 락테이트를 이용한 락타이드의 제조 수분을 5¾ 함유하고 있는 메틸 락테이트 50 g(TCI사)을 증류장치가 연결된 반웅기에 주입한 후 Pb00.5g을 추가로 주입하였다. 이 때, 반응기 내부 온도는 140 °C, 압력은 200 토르가 되도록 유지하였다. 약 10시간 중합 반웅 후 제조된 젖산 올리고머의 분자량은 2,200 g/nrol로 확인되었다.
상기 단계에서 제조된 젖산 을리고머가 포함된 반웅기 내부 온도를 180°C, 압력은 10 토르로 유지하였다. 이후 약 2시간 동안 증기 상태로 얻어지는 락타이드를 응축시켜 회수하였다. 이때, 회수된 락타이드의 순도를 기체 크로마토그래피로 분석한 결과 95%로 확인되었다. 실시예 2: 에틸 락테이트를 이용한 락타이드의 제조 -(1) 수분을 5¾ 함유하고 있는 에틸 락테이트 50 g(Alfa사)을 증류장치가 연결된 반웅기에 주입한 후 Pb00.5g을 추가로 주입하였다. 이 때, 반웅기 내부 온도는 160°C, 압력은 400 토르가 되도록 유지하였다. 약 10시간 중합 반응 후 제조된 젖산 을리고머의 분자량은 2,500 g/ηωΐ로 확인되었다.
상기 단계에서 제조된 젖산 을리고머가 포함된 반웅기 내부 온도를 180°C, 압력은 10 토르로 유지하였다. 이후 약 2시간 동안 증기 상태로 얻어지는 락타이드를 응축시켜 회수하였다. 이때, 회수된 락타이드의 순도는 97%로 확인되었다. 실시예 3: 에틸 락테이트를 이용한 락타이드 제조 -(2) 수분올 5% 함유하고 있는 에틸 락테이트 50 g(Alfa사)을 증류장치가 연결된 반응기에 주입 후 PbO 0.05 g을 추가로 주입하였다. 이 때, 반응기 내부 온도는 160°C, 압력은 400 토르가 되도톡 유지하였다. 약 12시간 반웅 후 제조한 젖산 올리고머의 분자량은 1,500 g/mol로 확인되었다.
상기 단계에서 제조된 올리고머가 포함된 반웅기 내부 온도를 180°C, 압력은 10 토르로 유지하였다. 이후 약 2시간 동안 증기 상태로 얻어지는 락타이드를 웅축 후 회수하였다. 이때, 회수된 락타이드의 순도는 92%로 확인되었다. 실시예 4: 에틸 락테이트를 이용한 락타이드 제조 -(3) 수분을 5% 함유하고 있는 에틸 락테이트 50 g(Alfa사)을 증류장치가 연결된 반웅기에 주입 후 PbO 2.5 g을 추가로 주압하였다. 이 때, 반응기 내부 온도는 160°C, 압력은 400 토르가 되도록 유지하였다. 약 8시간 반웅 후 제조한 젖산 을리고머의 분자량은 2,500 g/m 로 확인되었다.
상기 단계에서 제조된 올리고머가 포함된 반응기 내부 온도를 180°C, 압력은 10 토르로 유지하였다. 이후 약 2시간 동안 증기 상태로 얻어지는 락타이드를 웅축 후 회수하였다. 이때, 회수된 락타이드의 순도는 88%로 확인되었다. 실시예 5: 에틸 락테이트를 이용한 락타이드 제조 -(4) 실시예 2에서 PbO 대신에 Pb02를 사용하는 것을 제외하고, 실시예 2의 과정을 반복하여 락타이드를 회수하였다. 상기 과정 중 제조된 젖산 올리고머의 분자량은 2,700 g/mol로 확인되었고 회수된 락타이드의 순도는 96¾>로 확인되었다. 실시예 6: 에틸 락테이트를 이용한 락타이드 제조 -(5) 실시예 2에서 PbO 대신에 Pb304를 사용하는 것을 제외하고, 실시예 2의 과정을 반복하여 락타이드를 회수하였다. 상기 과정 중 제조된 젖산 올리고머의 분자량은 2,600 g/mol로 확인되었고, 회수된 락타이드의 순도는 95%로 확인되었다. 실시예 7: 에틸 락테이트를 이용한 락타이드 제조 -(6) 실시예 2에서 PbO 대신에 Pb304를 사용하는 것을 제외하고, 실시예 2의 과정을 반복하여 락타이드를 회수하였다. 상기 과정 중 제조된 젖산 을리고머의 분자량은 2,350 g/nrol로 확인되었고, 회수된 락타이드의 순도는 96%로 확인되었다. 실시예 8: 부틸 락테이트를 이용한 락타이드 제조 수분을 5% 함유하고 있는 부틸 락테이트 50 g(Aldrich사)을 증류장치가 연결된 반응기에 주입 후 PbO 0.5g을 추가로 주입하였다. 이 때, 반웅기 내부 온도는 180°C, 압력은 400 토르가 되도록 유지하였다. 약 10시간 반웅 후 제조한 젖산 올리고머의 분자량은 1,850 g/m이로 확인되었다.
상기 단계에서 제조한 올리고머가 포함된 반웅기 내부 온도를 200°C, 압력은 10 토르로 유지하였다. 이후 약 2시간 동안 증기 상태로 얻어지는 락타이드를 웅축 후 회수하였다. 이때, 회수된 락타이드의 순도는 90%로 확인되었다. 비교예 1: Sn(0ct)2를 이용한 에틸 락테이트 중합 반웅 상기 실시예 2에서 PbO 대신 Sn(0ct)2 촉매를 사용하는 것을 제외하고 동일한 방법으로 진행한 결과, 에틸 락테이트의 중합 반웅에 의한 젖산 올리고머의 생성이 일어나지 않았다. 비교예 2: SnO를 이용한 에틸 락테이트 증합 반웅 상기 실시예 2에서 PbO 대신 SnO 촉매를 사용하는 것을 제외하고 동일한 방법으로 진행한 결과, 에틸 락테이트의 중합 반응에 의한 젖산 을리고머의 생성이 일어나지 않았다. 비교예 3: SnC204를 이용한 에틸 락테이트 중합 반웅 상기 실시예 2에서 PbO 대신 SnC204 촉매를 사용하는 것을 제외하고 동일한 방법으로 진행한 결과, 에틸 락테이트의 중합 반웅에 의한 젖산 을리고머의 생성이 일어나지 않았다.

Claims

특허청구범위 :
1. 1) 알킬 락테 이트를 납 (Pb)계 촉매 하에서 중합하여 젖산 을리고머를 제조하는' 단계 ; 및
2) 상기 젖산 올리고머를 해중합하여 락타이드를 수득하는 단계
를 포함하는 , 알킬 락테 이트로부터 락타이드를 제조하는 방법 .
2. 게 1항에 있어서 , 상기 알킬 락테이트가 d Cu 알킬 락테이트 중 어느 하나 이상인 것을 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
3. 제 2항에 있어서, 상기 알킬 락테이트가 메틸 락테이트, 에 틸 락테이트, 프로필 락테이트 및 부틸 락테이트로 이루어지는 군으로부터 선택되는 어느 하나 이상인 것을 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
4. 제 1항에 있어서, 상기 알킬 락테 이트가 젖산 발효액으로부터 얻어진 것을 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
5. 제 4항에 있어서, 상기 알킬 락테아트가 젖산 발효액과 알콜 간의 에스테르화 반웅을 통해 얻어진 것을 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
6. 제 1항에 있어서, 상기 납계 촉매가 PbO, Pb304> Pb02, Pb(N03)2, PbCl2, Pb(OPh)2, Pb(OAc)2 및 PbO (쉘) /실리카 (코어 )로 이루어진 군으로부터 선택되는 어느 하나 이상인 것을 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
7. 게 1항에 있어서, 상기 납계 촉매가 알킬 락테이트 대비 0.05 내지 10 중량 %의 양으로 사용되는 것올 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
8. 제 1항에 있어서, 상기 단계 1이 120 내지 180°C 범위의 온도 및 100 내지 500 토르의 압력 조건 하에서 수행되는 것을 특징으로 하는, 알킬 락테이트로부터 락타이드를 제조하는 방법 .
9. 게 1항에 있어서 , 상기 단계 2가 160 내지 230°C 범위의 온도 및 5 내지 100 토르의 압력 조건 하에서 수행되는 것을 특징으로 하는, 알킬 락테 이트로부터 락타이드를 제조하는 방법 .
