Description
しかしながら、いずれも、単一のリソソーム酵素の欠損が原因のリソソーム病の治療を目的としており、複数のリソソーム酵素が欠損しているリソソーム病に対して有効ではない。さらに、現在販売されている上記治療剤は非常に高価であり、安価な治療剤の開発が求められる。
この事実に基づき、リソソーム病の酵素補充療法において欠損酵素を効果的にリソソーム中に送達させるために、高リン酸化されたリソソーム酵素を得る研究が進められている。例えば、単離したリソソーム酵素をマンノース6リン酸残基で修飾する方法や(特許文献1参照)、特殊な細胞系を用いて高リン酸化された組み換えリソソーム酵素を作製する方法(特許文献2参照)が報告されている。しかしながら、これらの技術では、多種類のリソソーム酵素を一度に且つ安価に製造することは難しい。
したがって、ML-IIおよびML-IIIの治療には、上記したような単一酵素製剤の投与では効果がなく、理論上、欠損したリソソーム酵素の全てを補充する必要があると考えられる。しかしながら、ML-IIおよびML-IIIにおいて欠損した50種類以上もの酵素の全てを個々に単離・精製することは非常に困難である。また、上記したような組み換えリソソーム酵素製剤は非常に高価なので、このような高価な酵素製剤を数十種類用いることは、経済的にも困難である。
このような情況から、ML-IIおよびML-IIIのようなリソソーム病には、従来の単一組み換えリソソーム酵素を用いた酵素製剤は適用できず、新たな有用な治療剤が求められている。
(1)細胞を培養して得られたリソソーム酵素群を有効成分として含むリソソーム病治療用医薬組成物、
(2)細胞がリソソーム酵素にマンノース6リン酸残基を付加する機能を有する細胞である、上記(1)記載の医薬組成物、
(3)細胞がリソソーム病に罹患していない対象由来の細胞である、上記(1)記載の医薬組成物、
(4)リソソーム酵素群が下記工程:
細胞に、両親媒性アミン、リソソーム指向性アミン、イオノフォア、およびV-ATPase阻害薬からなる群より選択される1または複数の試薬を添加して培養し、
培養上清を回収し、
得られた培養上清を精製および/または濃縮する、
ことを含む方法により得られたものである、上記(1)~(3)のいずれか1つに記載の医薬組成物、
(5)試薬が、塩化アンモニウム、クロロキン、モネンシン、ニゲリシン、およびバフィロマイシンA1からなる群より選択される、上記(4)記載の医薬組成物、
(6)細胞が正常皮膚線維芽細胞、COS-1細胞、NIH3T3細胞、HEK293細胞、HeLa細胞およびCHO細胞からなる群より選択されるものである、上記(1)~(5)のいずれか1つに記載の医薬組成物、
(7)リソソーム酵素が組み換え酵素ではない、上記(1)~(6)のいずれか1つに記載の医薬組成物、
(8)リソソーム酵素群がマンノース6リン酸残基を有するリソソーム酵素を含むものである、上記(1)~(7)のいずれか1つに記載の医薬組成物、および
(9)ムコリピドーシスII型またはIII型の治療用である、上記(1)~(8)のいずれか1つに記載の医薬組成物
を提供する。
また別の態様として、本発明は、リソソーム病の治療のための、細胞を培養して得られたリソソーム酵素群の使用を提供する。
また別の態様として、本発明は、細胞を培養して得られたリソソーム酵素群を患者に投与することを含む、リソソーム病の治療方法を提供する。
また別の態様として、本発明は、リソソーム病治療用医薬組成物の製造のための、細胞を培養して得られたリソソーム酵素群の使用を提供する。
本発明に用いる細胞として、好ましくは、リソソーム酵素にマンノース6リン酸残基を付加する機能を有する細胞またはリソソーム病に罹患していない対象由来の細胞が用いられる。
本発明において「リソソーム病に罹患していない対象由来の細胞」とは、健康な対象由来の正常細胞、およびリソソーム病に罹患していないが他の疾病に罹患している対象由来の細胞を意味する。
該対象は、哺乳動物対象、例えば、ヒト、サル、ウシ、イヌ、ネコ、マウス、ラット、ハムスター、ウサギ、マーモセット、ヒツジ、ヤギ等であり、好ましくは、ヒトである。
本発明に用いる細胞としては、限定するものではないが、例えば、皮膚、腎臓、胚、卵巣、子宮等に由来する細胞が挙げられ、好ましくは、皮膚線維芽細胞、腎線維芽細胞、胚性線維芽細胞等、さらに好ましくは、正常皮膚線維芽細胞、COS-1細胞、NIH3T3細胞、HEK293細胞、HeLa細胞、CHO細胞等、またさらに好ましくは、正常ヒト線維芽細胞が挙げられる。
本発明のリソソーム酵素群は、上記細胞を培養することにより得られる2種類以上のリソソーム酵素からなる酵素混合物であって、マンノース6リン酸残基を有するリソソーム酵素を含む。
本発明のリソソーム酵素群は、例えば、5種類以上、8種類以上、または12種類以上、好ましくは20種類以上、さらに好ましくは30種類以上、またさらに好ましくは40種類以上、最も好ましくは50種類以上のリソソーム酵素からなる。
本発明のリソソーム酵素群は、例えば、リソソーム病に罹患していない対象由来の細胞を培養し、得られた細胞から公知の手段・方法を用いて抽出することにより得ることができる。好ましくは、リソソーム病に罹患していない対象由来の細胞に、適当な酵素抽出用試薬を添加して培養し、培養上清を回収し、得られた培養上清を精製および/または濃縮する工程を含む方法によって得られる。
細胞に上記のような試薬を添加して培養することにより得られたリソソーム群をリソソーム病の治療に用いる試みは、本発明が初めてである。
培養上清は、適宜所望の程度まで精製・濃縮すればよい。
また、該リソソーム酵素群は、リソソーム中にすでに存在する正常なリソソーム酵素を排出させることにより得られるので、マンノース6リン酸残基で修飾された状態のリソソーム酵素を含むことができる。
したがって、本発明によれば、マンノース6リン酸残基を付加する工程を必要とせずに、マンノース6リン酸残基を有する多種類のリソソーム酵素を一度に得ることができ、容易にリソソーム病の治療に用いることができる。
本発明のリソソーム酵素群は、また、研究用のリソソーム酵素試薬として利用することもできる。
10cm培養皿において、10%FBS(ウシ胎児血清)および推奨量の抗生剤/抗菌剤(Gibco(登録商標)Antibiotic-Amtimycotic,Life Technologies)を含有するDMEM培地(以下、「標準培養培地」という)中でコンフルエントな状態まで培養した正常皮膚線維芽細胞に、培地中20mM濃度となるように塩化アンモニウムを添加し、培養した。培養7日後、上清を回収し、0.2μm滅菌フィルターでろ過して大きい細胞破片および生存細胞を除去した。ろ過後の上清中のリソソーム酵素活性を下記の方法によって測定した。
次いで、培養上清を0℃にて、分画分子量5000の分子量フィルター(Vivaspin 15R,Sartorius Stedim Biotech GmbH)のスピンカラムで遠心分離して塩化アンモニウムを除去した。得られたリソソーム酵素群を最終的に、α-マンノシダーゼ活性が600nmol/h/μLとなるように無血清DMEMを用いて希釈し、下記の実施例においてサンプルとして用いた。該サンプル中の使用時の酵素活性は、α-マンノシダーゼ 364.6nmol/h/μL、α-フコシダーゼ 38.4nmol/h/μL、α-ガラクトシダーゼ 5.6nmol/h/μL、α-グルコシダーゼ 10.3nmol/h/μL、β-ガラクトシダーゼ 35.0nmol/h/μL、β-グルコシダーゼ 25.9nmol/h/μL、総β-ヘキソサミニダーゼ 2650.4nmol/h/μL、β-ヘキソサミニダーゼA 522.9nmol/h/μL、β-グルクロニダーゼ 14.3nmol/h/μL、およびガラクトセレブロシダーゼ 0.57nmol/h/μLであった。なお、酵素活性の測定方法は下記にしたがう。
下記の実施例におけるtERTは、該サンプルを培地に添加して細胞を7日間培養して行った。その後、培養液を通常の標準培養培地に置き換えて24時間以内に各アッセイを行った。
得られたリソソーム酵素群中にリソソーム酵素が含まれることを確認するために、上記の塩化アンモニウム添加培養後の培養上清を用いて、リソソーム酵素活性を測定した。対照として、塩化アンモニウムを添加していない正常皮膚線維芽細胞の培養上清、および塩化アンモニウムを添加していないML-II患者由来の皮膚線維芽細胞(以下、「ML-II細胞」という)の培養上清中のリソソーム酵素活性を測定した。
リソソーム酵素の活性は、合成4-メチルウンベリフェリル基質を用いる標準的な方法で測定した。簡単に言えば、試料を酸性リン酸またはクエン酸バッファー中において37℃で1時間、該合成基質と共にインキュベートし、365nmの励起波長および450nmの発光波長にてマイクロプレートリーダーで蛍光を測定した。ガラクトセレブロシダーゼの場合、6-ヘキサデカノイルアミノ-4-メチルウンベリフェリル β-D-ガラクトピラノシド(Slater and Frith Ltd.)を基質として用い、385nmの励起波長および450nmの発光波長にて蛍光を測定した。酵素活性は、nmol/h/μLとして計算した。
結果を図1に示す。図1において、各酵素活性は、正常細胞の培養上清中の酵素活性に対する比率(倍)で表す。
図1から明らかなように、塩化アンモニウムを添加した正常皮膚線維芽細胞の培養上清は、少なくとも10種類のリソソーム酵素(α-マンノシダーゼ、α-フコシダーゼ、α-ガラクトシダーゼ、α-グルコシダーゼ、β-ガラクトシダーゼ、β-グルコシダーゼ、β-ヘキソサミニダーゼA、β-ヘキソサミニダーゼB、β-グルクロニダーゼ、およびガラクトセレブロシダーゼ)を含んでいた。また、該培養上清中の酵素活性パターンは、ML-II細胞の培養上清中の酵素活性パターンと類似していた。
ML-IIでは、リソソーム酵素にマンノース6リン酸残基が付加されていないため、リソソーム酵素がマンノース6リン酸受容体に認識されず、リソソームへ運搬されないので、リソソーム中においてリソソーム酵素が枯渇し、細胞外にマンノース6リン酸残基が付加されていないリソソーム酵素が溢れ出ている。したがって、ML-II細胞の培養上清中には多種類のリソソーム酵素が含まれる。このことは、ML-II細胞の培養上清の酵素活性測定結果と一致する。
実施例1で得られたリソソーム酵素群をML-II細胞に投与してtERTを行い、リソソーム酵素が活性な状態で該細胞内に取り込まれることを確認するために、細胞内リソソーム酵素活性を測定した。
実施例1で得られたリソソーム酵素群サンプルをML-II細胞の培養培地に添加し、所定の期間培養後、細胞を培養培地から単離し、プロテアーゼ阻害剤を含有する水中でソニケーションし、得られた懸濁液のタンパク質濃度を常法により測定した。得られた懸濁液を用いて、実施例1に記載の方法により、リソソーム酵素活性を測定した。酵素活性は、nmol/h/mg proteinとして計算した。対照として、正常皮膚線維芽細胞、およびtERT処理していないML-II細胞を用いた。
結果を図2に示す。
図2から明らかなように、tERTによって、細胞内のリソソーム酵素活性が上昇したことから、リソソーム酵素が細胞内に取り込まれたことが確認できた。
さらに、経時的なリソソーム酵素のML-II細胞内取り込みを図3に示す。
リソソーム酵素は少なくとも7日間、持続的に細胞内に取り込まれた。細胞内酵素活性は7日間上昇し続けたので、本実施例の上記の評価は、tERT処理の7日後に回収した細胞を用いて行った。また、下記の実施例4~9においても、tERT処理の7日後の細胞を用いた。
リソソーム酵素群が細胞表面のマンノース6リン酸受容体を介して細胞内に取り込まれていることを確認するために、マンノース6リン酸を同時投与し、細胞内取り込み阻害について試験した。
実施例1で得られたリソソーム酵素群サンプルをML-II細胞の培養培地に添加し、同時に、マンノース6リン酸を添加しない(0mM)、あるいはマンノース6リン酸を5mMまたは10mMとなるように添加した。細胞を所定時間培養後、実施例1および実施例2と同様の方法により、細胞内酵素活性を測定した。
結果を図4に示す。
リソソーム酵素の細胞内への取り込みは、マンノース6リン酸により、濃度依存的に競合的に阻害された。
細胞内に取り込まれたリソソーム酵素がリソソームまで運搬されているか確認するために、細胞の免疫染色を行った。
実施例1に記載のようにML-II細胞をtERT処理し、下記の方法により蛍光染色した。対照として、正常皮膚線維芽細胞およびtERT処理しないML-II細胞を用いた。
細胞を3.7%ホルムアルデヒドで1時間固定し、次いで、0.1%Triton-X100で15分間透過処理し、室温にて1時間、1%ウシ血清アルブミンでブロッキングした。一次抗体として、モノクローナル抗Lamp-2抗体(H4B4)、ポリクローナル抗カテプシンB抗体(S-12)、ポリクローナル抗カテプシンD抗体(H-75)、およびポリクローナル抗β-グルコシダーゼ抗体(H-300)(全て、Santa Cruz Biotech. Inc.,Santa Cruz,CA,USAから入手)を1:100濃度で用いて、室温で1時間処理した。次いで、二次抗体(Alexa Fluor 488または555、Invitrogen)を1:1000希釈で用いて、室温で1時間処理した。LysoTracker Red DND-99(Molecular Probes Inc.#7528)は0.2μM濃度で用いて、37℃で1時間処理した。全ての蛍光画像は、蛍光顕微鏡(BX51,Olympus)または共焦点レーザー走査顕微鏡システム(Leica TCS SP-2,Leica Microsystems)を用いて獲得した。リソソームマーカーとしてLamp-2およびLysoTrackerを用いた。
結果を図5および図6に示す。
図5および図6に示されるように、ML-II細胞ではカテプシンBおよびカテプシンDが確認できなかった。一方、tERT処理したML-II細胞では、カテプシンBおよびカテプシンDの画像がリソソーム(Lamp-2)の画像と重なったことから、細胞に投与されたリソソーム酵素混合物からリソソーム酵素がリソソームに運搬されたことが確認できた。
リソソーム酵素のなかでも、β-グルコシダーゼは、マンノース6リン酸依存性の運搬システムではなく、LIMP-2というタンパク質でリソソームに運搬されていることが証明されている(Reczek D,et al.,Cell 2007;131(4):770-83)。したがって、ML-II細胞は、β-グルコシダーゼを欠損していない。この以前の知見は、上記実験結果と一致する。
ML-IIでは、リン脂質およびコレステロールが蓄積する。したがって、細胞中のリン脂質およびコレステロール量を測定することにより、ML-IIに対するtERTの効果を評価した。
実施例1に記載のようにML-II細胞をtERT処理し、市販の測定キットを用い、製造者の指示書にしたがって、細胞中のリン脂質およびコレステロール量を測定した(リン脂質:Phospholipids C-test,Wako Pure Chemicals,Co.、およびコレステロール:Amples Red Cholesterol Assay Kit(A12216),Molecular Probes,Invitrogen)。対照として、正常皮膚線維芽細胞およびtERT処理しないML-II細胞を用いた。
結果を図7aおよび図7bに示す。
リン脂質およびコレステロールは、正常細胞に比べてML-II細胞において蓄積しているが、tERTによって蓄積量が減少した。
ML-IIにおけるオートファジー異常およびミトコンドリア異常に対するtERTの効果を評価した。対照として、正常細胞およびtERT処理していないML-II細胞を用いた。
実施例1に記載のようにML-II細胞をtERT処理し、ウェスタンブロッティングによりLC3-IIの量を測定した。一次抗体として、ポリクローナル抗LC3抗体(PM036)およびポリクローナル抗β-アクチンHRP DirecT抗体(PM053-7)(MBL Co.Ltd.,Nagoya,Japan)を用いた。
結果を図8に示す。tERTにより、LC3-IIの量が減少した。
また、蛍光免疫染色を実施例4の記載と同様に行った。ミトコンドリアのマーカーとしてMitoTracker Red CMXRos(Molecular Probes Inc.#7512)を37℃で1時間、200nM濃度で用いた。オートファジーのマーカーとして、ポリクローナル抗LC3抗体(PM036)(MBL Co.Ltd.,Nagoya,Japan)を用いた。
蛍光顕微鏡で観察したところ、tERTにより、LC3の小胞が減少し、ミトコンドリアの形態も明らかに改善を示したことが確認された。
ML-IIは、皮膚線維芽細胞における封入体(inclusion body)の存在によって特徴付けられる。封入体は、リソソーム酵素の異常により分解できない基質がリソソームに蓄積することが原因であり、同時に、リソソームの数も顕著に増加する。tERTのこれらの現象に対する効果を下記の方法によって評価した。対照として、正常細胞およびtERT処理していないML-II細胞を用いた。
実施例1に記載のようにML-II細胞をtERT処理し、トリプシンを用いて細胞を標準的な方法で回収し、小型のチューブに入れた。該細胞のペレットを、LysoTracker Red DND-99(1μM)およびDAPI(1μg/ml)の両方を含有する標準培養培地中に再懸濁し、37℃で1時間インキュベートした。インキュベーション後、遠心分離により該細胞を回収し、PBSで1回リンスした後、該細胞をPBS中に再懸濁した。該細胞懸濁液の蛍光強度をマイクロプレートリーダー(Vorona Fluorescence Microplate Reader MTP810Lab)を用いて、励起波長530nm/発光波長590nm(LysoTrackerの場合)および、励起波長365nm/発光波長450nm(DAPIの場合)で測定した。実施例4の記載と同様にして、蛍光画像を得た。該蛍光染色の結果、tERTによりリソソームの染色が減少したことが分かった。
さらに、LysoTracker/DAPIの蛍光強度比率を算出し、各細胞あたりのリソソーム量(数および大きさ)を評価した。
結果を図9示す。tERTにより、各細胞あたりのリソソーム量が減少した。
さらに、光学顕微鏡写真観察では、tERTにより、封入体の減少が確認された(図10)。また、電子顕微鏡写真観察では、tERTにより、均質な小胞の蓄積が解消されたことが確認された。
蛍光標識したセラミド(BODIPY-Cer)を用いて、正常細胞、ML-II細胞およびtERT処理したML-II細胞におけるエンドサイトーシスによって取り込まれた物質の細胞内輸送を評価した。
ガラス底皿中で皮膚線維芽細胞を調整した。BSAに複合体化したBODIPY FLC5-セラミド(Invitrogen #B22650)を入手し、標準培養培地中2.5nM濃度に希釈した。該培地中で30分間、細胞を培養した。培養後、該細胞をPBSで1回洗浄し、実施例4に記載の共焦点顕微鏡で直接観察した。実施例4の記載と同様にして、蛍光画像を得た。
蛍光画像を解析したところ、正常細胞ではセラミドはゴルジ体に運ばれたが、ML-IIでは細胞質内の小胞に閉じこめられていた。一方、tERT処理したML-II細胞では、セラミドは、細胞質内の小胞に多少残っているものの、ゴルジ体へ運搬されるようになった。
また、氷冷培地中において、原形質膜に対するBODIPY-Cerの接着について試験した結果、正常細胞、ML-II細胞およびtERT処理したML-II細胞間に違いはなかった。
マンノース6リン酸受容体に対する抗体を用いて、細胞表面の該受容体を介して取り込まれた該抗体の細胞内輸送を評価した。
マンノース6リン酸受容体抗体[モノクローナル抗マンノース6リン酸受容体抗体(2G11)(Abcam)]を1:75濃度で含有する標準培養培地中で細胞をインキュベートした。30分間の取り込み試験の場合、該抗体含有培地で細胞を30分間インキュベートした。インキュベーション後、氷冷PBSで洗浄し、すぐに3.7%ホルムアルデヒド中で固定した。細胞を0.1%Triton-X100で15分間透過性処理し、室温で1時間、1%ウシ血清アルブミンでブロッキングした後、細胞を二次抗体で処理した。1時間の取り込みおよび2時間のインキュベーション試験の場合、該抗体含有培地で細胞を1時間インキュベートし、洗浄し、次いで、抗体を含有しない標準培養培地中で培養した。その後の工程は、上記の30分取り込み試験と同様である。二次抗体としてAlexa Flour 488標識抗体(Life Technologies Corp.)を用いた。実施例4の記載と同様にして、蛍光画像を得た。
蛍光画像を解析したところ、正常細胞では、小胞に留まっていたが、ML-II細胞ではゴルジ体に留まっていた。一方、tERT処理したML-II細胞では、正常細胞と同じ小胞のパターンを示した。
10cm培養皿において、標準培養培地中でコンフルエントな状態まで培養した正常皮膚線維芽細胞に、培地中20mM濃度となるように塩化アンモニウム、培地中20μM濃度となるようにクロロキン、または培地中20nM濃度となるようにバフィロマイシンA1を添加し、培養した。培養5日後および7日後に上清を回収し、0.2μm滅菌フィルターでろ過して大きい細胞破片および生存細胞を除去した。ろ過後の上清中のリソソーム酵素活性を実施例1の記載と同じ方法で測定した。対照として、いずれの試薬も添加していない標準培養培地中で培養した正常皮膚線維芽細胞の培養上清中のリソソーム酵素活性を測定した。
結果を表1および2に示す。なお、表1および表2中、β-Galはβ-ガラクトシダーゼ、total β-Hexは総β-ヘキソサミニダーゼ、β-Hex Aはβ-ヘキソサミニダーゼA、β-Glucはβ-グルコシダーゼ、α-Manはα-マンノシダーゼ、α-Fucはα-フコシダーゼ、α-Glucはα-グルコシダーゼ、α-Galはα-ガラクトシダーゼ、β-Glucuroはβ-グルクロニダーゼ、α-Iduroはα-L-イデュロニダーゼ、GALCはガラクトセレブロシダーゼを示す。SCMは、いずれの試薬も添加していない標準培養培地(standard culture medium)を示す。CQはクロロキンを示し、BafiloはバフィロマイシンA1を示す。酵素活性は、nmol/h/μLで示す。
10cm培養皿において、標準培養培地中でコンフルエントな状態まで培養したCOS-1細胞に、培地中20mM濃度となるように塩化アンモニウムを添加し、培養した。培養3日後に上清を回収し、0.2μm滅菌フィルターでろ過して大きい細胞破片および生存細胞を除去した。ろ過後の上清中のリソソーム酵素活性を実施例1の記載と同じ方法で測定した。対照として、塩化アンモニウムを添加していない標準培養培地中で培養したCOS-1細胞の培養上清中のリソソーム酵素活性を測定した。
結果を表3に示す。なお、表3中、β-Galはβ-ガラクトシダーゼ、total β-Hexは総β-ヘキソサミニダーゼ、β-Hex Aはβ-ヘキソサミニダーゼA、β-Glucはβ-グルコシダーゼ、α-Manはα-マンノシダーゼ、α-Fucはα-フコシダーゼ、α-Glucはα-グルコシダーゼ、α-Galはα-ガラクトシダーゼ、β-Glucuroはβ-グルクロニダーゼ、α-Iduroはα-L-イデュロニダーゼ、GALCはガラクトセレブロシダーゼを示す。NH4Cl(-)は、塩化アンモニウムを加えない場合の培養上清を示し、NH4Cl(+)は、塩化アンモニウムを加えた場合の培養上清を示す。酵素活性は、nmol/h/μLで示す。
実施例1で得られたリソソーム酵素群サンプルを、クラッベ病およびテイ・サックス病の患者由来の皮膚線維芽細胞(以下、各々、「クラッベ病細胞」および「テイ・サックス病細胞」という)の培養培地に添加し、7日間培養した。細胞を培養培地から単離し、プロテアーゼ阻害剤を含有する水中でソニケーションし、得られた懸濁液のタンパク質濃度を常法により測定した。得られた懸濁液を用いて、実施例1に記載の方法により、リソソーム酵素活性を測定した。酵素活性は、nmol/h/mg proteinとして計算した。対照として、正常皮膚線維芽細胞、およびtERT処理していないクラッベ病細胞またはtERT処理していないテイ・サックス病細胞を用いた。
結果を図11に示す。
図11に示されるように、本発明のリソソーム酵素群を用いるtERTによって、クラッベ病およびテイ・サックス病でそれぞれ欠損しているリソソーム酵素(クラッベ病についてガラクトセレブロシダーゼ、テイ・サックス病についてβ-ヘキソサミニダーゼA)の細胞内酵素活性が上昇した。
Claims (9)
- 細胞を培養して得られたリソソーム酵素群を有効成分として含むリソソーム病治療用医薬組成物。
- 細胞がリソソーム酵素にマンノース6リン酸残基を付加する機能を有する細胞である、請求項1記載の医薬組成物。
- 細胞がリソソーム病に罹患していない対象由来の細胞である、請求項1記載の医薬組成物。
- リソソーム酵素群が下記工程:
細胞に、両親媒性アミン、リソソーム指向性アミン、イオノフォア、およびV-ATPase阻害薬からなる群より選択される1または複数の試薬を添加して培養し、
培養上清を回収し、
得られた培養上清を精製および/または濃縮する、
ことを含む方法により得られたものである、請求項1~3のいずれか1項記載の医薬組成物。 - 試薬が、塩化アンモニウム、クロロキン、モネンシン、ニゲリシン、およびバフィロマイシンA1からなる群より選択される、請求項4記載の医薬組成物。
- 細胞が正常皮膚線維芽細胞、COS-1細胞、NIH3T3細胞、HEK293細胞、HeLa細胞およびCHO細胞からなる群より選択されるものである、請求項1~5のいずれか1項記載の医薬組成物。
- リソソーム酵素が組み換え酵素ではない、請求項1~6のいずれか1記載の医薬組成物。
- リソソーム酵素群がマンノース6リン酸残基を有するリソソーム酵素を含むものである、請求項1~7のいずれか1項記載の医薬組成物。
- ムコリピドーシスII型またはIII型の治療用である、請求項1~8のいずれか1項記載の医薬組成物。
