本発明は、下記反応式〔I〕に示す工程(A)を含む高純度芳香族メチルアルコールの製造方法、及び反応式〔II〕に示す工程(A-0)と工程(A)を含む高純度芳香族メチルアルコールの製造方法である。
本発明の製造方法の工程(A)は、分解抑制剤の存在下に、一般式(1)で示される芳香族メチルアルコール含有粗製物から前記芳香族メチルアルコールを蒸留精製する方法により、前記一般式(1)で示される高純度芳香族メチルアルコールを得る工程である。
本発明の工程(A)の原料として使用される芳香族メチルアルコール含有粗製物は、一般式(1)で示される芳香族メチルアルコールを含有するものであれば特に限定されないが、例えば、一般式(1)で示される芳香族メチルアルコールを90質量%未満の量で含むものが好ましい。芳香族メチルアルコール含有粗製物として、特に好ましいのは、後述するような工程(A-0)の反応終了後に得られた芳香族メチルアルコール含有粗製物1a~1dのいずれかである。なお、これらの粗製物には水分が含有していても良く、その水分量は、特に限定されない。これらの水分は、工程(A)の蒸留時に共沸(azeotrope)にて除去される。しかしながら、次工程の蒸留の精製効率を考慮した場合、少なくとも前記粗製物は、水分が分離しない程度の水分含有量が望ましい。
本発明における分解抑制剤は、本発明の工程(A)における蒸留の際、芳香族メチルアルコールの分解を抑制する目的で使用される剤を示す。
なお、上記の分解抑制剤を固体のまま使用しても良いし、また、芳香族メチルアルコール含有粗製物が液体又は溶液の場合、分解抑制剤を溶解又は懸濁させて使用しても良い。
本発明の工程(A)で使用される分解抑制剤は、工程(A)の蒸留釜の内容物のpHが、好ましくは8~14、より好ましくは9~14、特に好ましくは10~13となるような条件下にするための必要量が使用される。例えば、その使用量の目安としては、芳香族メチルアルコール含有粗製物に含まれる芳香族メチルアルコールの純分質量に対して、好ましくは10~500,000ppm、更に好ましくは200~50,000ppm、より好ましくは200~30,000ppm、特に好ましくは500~15,000ppm、特により好ましくは1000~10,000ppmである。
分解抑制剤を上記のように使用量することで、本発明の工程(A)は、芳香族メチルアルコール自体の分解や副反応等を抑えながら、高純度メチルアルコールを取得することが可能となる(本願実施例2及び3参照)。
本発明の工程(A)で使用される蒸留方法おける、蒸留前仕込み物は、上述の通り、場合により水分を含む前記芳香族メチルアルコール含有粗製物と分解抑制剤とを含む混合物である。
本発明の工程(A)における蒸留は、蒸留開始前~蒸留終了時に渡って、蒸留釜の内容物のpHが、好ましくは8~14、より好ましくは9~14、特に好ましくは10~14未満であるpH条件下で行われる。なお、pHの確認は、適宜サンプリングし、例えば、pH試験紙、pHメーター等を用いて行う。また、pHの調整も、同様に確認して、適宜分解抑制剤を追加する。
本発明の工程(A)で使用される蒸留方法は、例えば、単蒸留、精留精製等のいずれの方法であってもよい。また、蒸留方式は、バッチ方式、半連続方式、連続方式のいずれの方式であってもよい。目的物である高純度芳香族メチルアルコール(一般式(1))の主留分における含有量を微調整しながら蒸留する必要があるため、これらの蒸留装置には、精留塔を設けることが好ましい。なお、精留塔としては、例えば、棚段式精留塔や充填式精留塔等の通常の蒸留で用いられるものを使用することができ、精留塔の本数、及び蒸留回数は特に制限されない。
本発明の工程(A)における蒸留精製の還流比は、各精留塔の分離状態を確認して適宜決められる。しかしながら、過剰な還流比は、長時間の加熱が必要になり、一般式(1)で示される芳香族メチルアルコールの分解やその他の副反応が促進されるため好ましくない。従って、本発明の工程(A)の蒸留における還流比(=還流量/留出量)は、好ましくは0~50、更に好ましくは0.1~30、特に好ましくは1~15である。
前記充填式精留塔を使用する場合、充填物の種類は特に限定されない。しかしながら、芳香族メチルアルコールは、蒸留温度が高くなると分解し易くなるため、蒸留釜の液温を高く設定しなくてすむよう、精留塔の塔頂部と塔底部との差圧が小さくなるように規則充填物を使用することが好ましい。
本発明の工程(A)における蒸留精製において、蒸留装置の蒸留釜の液温は、前記一般式(3)で示されるビス(アリールメチル)エーテルの副生成物の生成状態により適宜設定される。しかしながら、芳香族メチルアルコールは、蒸留温度が高くなると分解し易くなるため、好ましくは70~240℃、更に好ましくは90~210℃である。
本発明の工程(A)における蒸留において、蒸留装置の圧力は、蒸留装置の蒸留釜の液温と前記一般式(3)で示されるビス(アリールメチル)エーテル等の副生物等の生成状態により適宜設定される。
具体例として、一般式(1)で示される芳香族メチルアルコールがピペロニルアルコールの場合、蒸留装置の蒸留釜の液温が240℃を超えると、ピペロニルアルコール自体の分解や、前記式(3d)で示されるビス(3,4-メチレンジオキシベンジル)エーテルの副生成やその他の反応が促進され、これらが主留分に混入する恐れがあるので好ましくない。従って、ピペロニルアルコール含有粗製物を蒸留精製する場合、本発明の工程(A)の蒸留圧力は、ピペロニルアルコールの沸点133℃/0.66kPa(133℃/5Torr)を考慮し、好ましくは0.013~26.6kPa(0.1~200Torr)、更に好ましくは0.066~13.3kPa(0.5~100Torr)、特に好ましくは0.50~3.9kPa(1~30Torr)で行う。
上記の本発明の製造方法より、簡便な操作かつ、好適な反応時間にて、例えば、前記一般式(3)のような副生物の生成、混入等を抑えた高純度の芳香族メチルアルコールを収率良く製造することが出来る。
本発明の工程(A)により得られる一般式(1)で示される高純度芳香族メチルアルコールは、蒸留時に前記一般式(3)で示されるビス(アリールメチル)エーテルを不純物として混入することがある。従って、本発明の製造方法で高純度芳香族メチルアルコールが得られるかどうかどうかをあらかじめ確認するために、一般式(1)で示される芳香族メチルアルコール含有粗製物に混入してくる前記一般式(3)で示される副生物の含有率を調べておくことが望ましい。
本発明の一般式(1)で示される高純度芳香族メチルアルコールの純度は、好ましくは90%以上、更に好ましくは93%以上、より好ましくは95%以上、特に好ましくは97%以上、特により好ましくは98.5%以上のものである。なお、前記純度は、上記同様、HPLC分析(絶対検量線法)から算出される値である。
蒸留により得られる高純度芳香族メチルアルコール留分以外の初留分及び釜残分等は、例えば、濾別や水洗等の処理により不純物を除き、再度、芳香族メチルアルコール含有粗製物として、工程(A)の蒸留原料に使用してもよい。
本発明の製造方法の工程(A-0)は、前記一般式(2)で示される芳香族メチルハライドと水とを反応させて、前記一般式(1)で示される芳香族メチルアルコール含有粗製物を得る工程である。この芳香族メチルアルコール含有粗製物は、工程(A)における蒸留原料である芳香族メチルアルコール含有粗製物として好適に使用できる。
mは、置換基R3の個数であり、0から3の整数を示す。mが2以上の場合、それぞれのR3は、互いに同一でも異なっていてもよい。
但し、n+mは0から4の整数となる。
また、更に好ましくは下記一般式(2a-1)、(2b-1)、(2d-1)、(2e-1)が挙げられる。
(水の使用量)
本発明の工程(A-0)の加水分解反応で使用される水の量は、前記芳香族メチルハライド1モルに対して、当モル以上あれば特に制限されないが、反応時の撹拌性や反応後の単離・精製操作の効率性を考慮すると、芳香族メチルハライド1モルに対して、好ましくは1~1000モル、更に好ましくは1.25~500モル、より好ましくは1.5~250モル、特に好ましくは2.0~100モル使用される。
本発明の工程(A-0)は、無溶媒で行うことも、別途有機溶媒の存在下にて行うことも出来る。また、その反応系は均一系或いは不均一系のどちらでもよく、更に、均一系の場合、単相系であっても、例えば、水-有機相からなる二相系のような多相系であってもいずれの場合でもあってもよい。
本発明の工程(A-0)で使用できる有機溶媒としては、反応を阻害しないものならば特に限定されないが、例えば、脂肪族炭化水素類(例えば、n-ペンタン、n-へキサン、n-ヘプタン、シクロヘキサン等);脂肪族ハライド類(例えば、塩化メチレン、1,2-ジクロロエタン等);エーテル類(例えば、ジエチルエーテル、ジイソプロピルエーテル、テトラヒドロフラン、ジオキサン等);芳香族炭化水素類(例えば、ベンゼン、トルエン、キシレン等);芳香族エーテル類(例えば、アニソール、1,2-ジメトキシベンゼン、1,3-ジメトキシベンゼン、1,4-ジメトキシベンゼン、1,2-メチレンジオキシベンゼン、1,2-エチレンジオキシベンゼン、ジフェニルエーテル等);芳香族ハライド類(例えば、クロロベンゼン、1,2-ジクロロベンゼン、1,3-ジクロロベンゼン、1,4-ジクロロベンゼン等);芳香族ニトロ化合物類(例えば、ニトロベンゼン等);スルホキシド類(例えば、ジメチルスルホキシド等);スルホン類(例えば、スルホラン等)などが挙げられるが、好ましくはn-ペンタン、n-へキサン、n-ヘプタン、シクロヘキサン、ジエチルエーテル、ジイソプロピルエーテル、テトラヒドロフラン、ジオキサン、ベンゼン、トルエン、キシレン、アニソール、1,2-ジメトキシベンゼン、1,3-ジメトキシベンゼン、1,4-ジメトキシベンゼン、1,2-メチレンジオキシベンゼン、1,2-エチレンジオキシベンゼン、ジフェニルエーテル、クロロベンゼン、ジクロロベンゼン、ニトロベンゼン、アセトニトリル、プロピオニトリル、ベンゾニトリル、ジメチルスルホキシド、スルホラン、塩化メチレン、1,2-ジクロロエタン、更に好ましくはn-ペンタン、n-へキサン、n-ヘプタン、シクロヘキサン、ジイソプロピルエーテル、ベンゼン、トルエン、キシレン、1,2-ジメトキシベンゼン、1,2-メチレンジオキシベンゼン、クロロベンゼン、1,2-ジクロロエタンが使用される。なお、これらの溶媒は、単独又はそれらのうち二種以上を混合して使用してもよい。
本発明の工程(A-0)で使用される有機溶媒の量は、反応溶液の均一性や攪拌性等により適宜調節するが、前記芳香族メチルハライド1gに対して、好ましくは0.1~1000mL、更に好ましくは0.3~500mL、特に好ましくは0.5~200mL使用される。
本発明の工程(A-0)は、別途相間移動触媒やリン酸緩衝剤などの添加物を加えて反応を行ってもよい。
本発明の工程(A-0)の反応は、反応を促進させるために、例えば、有機アンモニウム塩化合物(テトラエチルアンモニウムクロリド、テトラエチルアンモニウムブロミド、テトラブチルアンモニウムクロリド、テトラブチルアンモニウムブロミド、ベンジルトリメチルアンモニウムクロリド、ベンジルトリメチルアンモニウムブロミド、ラウリルトリメチルアンモニウムクロリド、ステアリルトリメチルアンモニウムクロリド等)、及び有機ホスホニウム塩化合物(テトラメチルホスホニウムクロリド、テトラブチルホスホニウムクロリド、テトラメチルホスホニウムブロミド、テトラブチルホスホニウムブロミド、ベンジルトリメチルホスホニウムクロリド、ベンジルトリメチルホスホニウムブロミド、テトラフェニルホスホニウムクロリド及びテトラフェニルホスホニウムブロミド等)よりなる群から選ばれる少なくとも一種の相間移動触媒の存在下にて反応を行ってもよい。なお、本発明の相間移動触媒は、それぞれ単独又は二種以上を混合して使用しても良い。また、そのまま使用しても、例えば、水、前記有機溶媒或いはこれらの混合溶媒に溶解又は懸濁させて使用しても良い。
本発明の工程(A-0)は、リン酸緩衝剤の存在下にて反応を行ってもよい。本発明の工程(A-0)で使用されるリン酸緩衝剤として、例えば、リン酸カリウム、リン酸ナトリウム、リン酸一水素カリウム、リン酸二水素カリウム、リン酸一水素ナトリウム、リン酸二水素ナトリウムなどのアルカリ金属又はアルカリ土類金属のリン酸塩化合物が挙げられる。なお、前記リン酸緩衝剤は、それぞれ単独で使用しても、二種類以上を混合して使用してもよい。また、そのまま使用しても、例えば、水、有機溶媒或いはこれらの混合溶媒に、溶解又は懸濁させて使用してもよい。
〔反応方法〕
本発明の工程(A-0)は、例えば、大気中または不活性ガス雰囲気下にて、前記芳香族メチルハライドと有機溶媒の混合液と水とを混合し、攪拌しながら反応させる等の方法によって行われ得る。なお、本発明の工程(A-0)の反応は、酸性条件下、中性条件下又はアルカリ性条件下のいずれの条件下でも行うことは出来る。
また、反応温度は、好ましくは10℃~110℃、更に好ましくは0℃~100℃、特に好ましくは20℃~90℃であり、反応圧力は、特に限定されない。
本発明の工程(A-0)で得られる芳香族メチルアルコール含有粗製物は、以下の芳香族メチルアルコール含有粗製物1aから当該粗製物1dの状態のものを含み得る。
なお、これらの芳香族メチルアルコール含有粗製物中には、前記非特許文献1にも、開示されている、一般式(3):
芳香族メチルアルコール含有粗製物1aは、本発明の工程(A-0)の反応終了直後の反応混合物を示す。
前記芳香族メチルアルコール含有粗製物1aが、例えば、目的物である芳香族メチルアルコールや反応溶媒などを含む有機層-水層溶液である場合、芳香族メチルアルコール含有粗製物1bは、芳香族メチルアルコール含有粗製物1aから分液・抽出操作により得られた有機層溶液を示す。なお、分液・抽出操作には、前記反応溶媒と同義の有機溶媒を使用してもよい。さらに、不要物があれば、必要に応じて不要物の取り除いてもよい。
芳香族メチルアルコール含有粗製物1cは、前記有機層溶液(芳香族メチルアルコール含有粗製物1b)をアルカリ性水溶液等で洗浄して得られる有機層溶液を示す。このように芳香族メチルアルコール含有粗製物1cは、得られた有機層溶液から、水、飽和食塩水、或いは酸性又はアルカリ性溶液等で、適宜洗浄することで、含有する水溶性成分、酸性又はアルカリ性成分をなるべく除いた後、これを工程(A)の蒸留に使用することで、蒸留時の芳香族メチルアルコールの分解を抑えることが出来る。なお、使用されるアルカリ性水溶液としては、例えば、前述の分解抑制剤を水に溶かしたものを使用することができる。
また、前記工程(A-0)の加水分解反応を、アルカリ性条件下にて行った場合、反応終了後、水を投入して洗浄を行うことが出来る。
芳香族メチルアルコール含有粗製物1dは、前記水、飽和食塩水、或いは酸性又はアルカリ性溶液等で洗浄して得られた有機層溶液(芳香族メチルアルコール含有粗製物1c)から、更に反応溶媒等の有機溶媒を留去して得られる濃縮物、又は芳香族メチルアルコール含有粗製物1b(但し、純分含量が90%以上)から更に反応溶媒を留去して得られる濃縮物を示す。
例えば、前記芳香族メチルアルコール含有粗製物1cから工程(A-0)で使用した反応溶媒や分液・抽出に使用した有機溶媒等を除き、濃縮物として芳香族メチルアルコール含有粗製物1dを取得し、これをそのまま工程(A)の蒸留精製に使用することで、蒸留精製の操作時間を短くすることが出来る。即ち、工程(A)の蒸留精製時間を短くすればするほど、芳香族メチルアルコールの分解や目的物である蒸留された芳香族メチルアルコール中への分解物の混入をより抑えることが出来る。なお、工程(A-0)の反応溶媒の留去の際、前述の工程(A)における分解抑制剤を予め必要量入れて行うことが好ましい。
次に、本発明の高純度芳香族メチルアルコール組成物及び高純度芳香族メチルアルコール組成物を含有する保存容器の製造方法について説明する。本発明の高純度芳香族メチルアルコール組成物は、高純度芳香族メチルアルコールと分解抑制剤を含む組成物である。この組成物の製造方法としては、下記に示す方法1及び方法2がある。
方法1は、固体状態の高純度芳香族メチルアルコールに、本発明の分解抑制剤を加えて、例えば、震とう、攪拌等により混合させて高純度芳香族メチルアルコール組成物を製造し、これを保存容器に充填する方法である。ここで、分解抑制剤の添加は、高純度芳香族メチルアルコールを保存容器に加える前又は加えた後、いずれの場合であってもよい。
方法2は、例えば、加熱等により融解させた高純度芳香族メチルアルコールを保存容器に移す際に、分解抑制剤を加えて、高純度芳香族メチルアルコール組成物として充填された保存容器を製造し、これを保存する方法である。ここで、分解抑制剤の添加は、前記融解した高純度芳香族メチルアルコールを保存容器に加える前又は加えた後、いずれの場合であってもよいが、好ましくは前記融解した高純度芳香族メチルアルコールを保存容器に加えた後に、これが固化する前に分解抑制剤を加える方法である。
本発明の高純度芳香族メチルアルコール組成物では、高純度芳香族メチルアルコールは、前記工程(A)にて、蒸留精製された一般式(1)で示される高純度芳香族メチルアルコール、又は一般式(1)で示される芳香族メチルアルコールの純度が95%以上の芳香族メチルアルコール含有粗製物1dが使用され得る。
上記の高純度芳香族メチルアルコール組成物並びにその製造方法及び保存方法において使用される分解抑制剤は、前記工程(A)における蒸留の際、芳香族メチルアルコールの分解を抑制する目的で使用される剤と同義のものが使用される。なお、高純度芳香族メチルアルコール組成物並びにその製造方法及び保存方法において、分解抑制剤は、水溶液として使用してもよい。
本発明の高純度芳香族メチルアルコール組成物における分解抑制剤の使用量は、前記保存容器の内容物のpHが、好ましくは8~14、より好ましくは9~14、特に好ましくは10~13となるような条件下にするための必要量が使用される。例えば、その使用量の目安としては、芳香族メチルアルコール含有粗製物に含まれる芳香族メチルアルコールの純分含量に対して、好ましくは10~500,000ppm、更に好ましくは200~50,000ppm、より好ましくは200~30,000ppm、特に好ましくは500~15,000ppm、特により好ましくは1000~10,000ppmである。
なお、前期内容物のpHの確認は、適宜サンプリングし、例えば、pH試験紙、pHメーター等を用いて行う。また、pHの調整も、同様に確認して、適宜分解抑制剤を追加する。なお、分解抑制剤の使用量が、例えば、上記の範囲より少ない場合には、芳香族メチルアルコールの分解抑制効果が十分でなく、また多い場合には、懸濁、スラリー状になりハンドリング性が悪くなったり、芳香族メチルアルコール組成物から芳香族メチルアルコールを取り出して使用する際に中和が必要になったり、更には経済的にも望ましくない。
本発明の高純度芳香族メチルアルコール、特に、ピペロニルアルコール、ベラトリルアルコール、及びアニスアルコールなどのアルコキシ基を有する高純度芳香族メチルアルコールは、例えば、周囲からの光や熱等の影響により分解し、前記一般式(3)などの分解物を生成することがある(例えば、本願比較例2参照)。
従って、本発明の上記の方法(方法1、方法2)で得られた高純度芳香族メチルアルコール組成物は、遮光性のある保存容器での保存が望ましい。その材質は、例えば、ガラス、セラミック、プラスチック、金属、紙、木、竹、麻等が挙げられるが、特に制限されない。また、例えば、使用前に汎用の容器の内面を分解抑制剤又はその溶液で洗浄し、乾燥する等の前処理を行った容器を使用してもよい。更に、蒸留時に使用した主留分の受器をそのまま保存容器として使用してもよい。なお、前記保存容器は、その内部が窒素、アルゴン、ヘリウムなどの不活性ガスで充填されていてもよい。
次に、高純度芳香族メチルアルコールの保存方法について説明する。
本発明で得られる高純度芳香族メチルアルコールを含め、芳香族メチルアルコールが電子供与基を有する芳香族メチルアルコールである場合、特にピペロニルアルコール、ベラトリルアルコール、及びアニスアルコールなど、アルコキシ基を有する芳香族メチルアルコールである場合には、その純度に関係なく、以下に示す本発明の保存方法により、保存中の分解を抑制することは可能である。
従って、本発明の保存方法で使用される芳香族メチルアルコールは、前記工程(A)にて、蒸留精製された一般式(1)で示される高純度芳香族メチルアルコール、又は一般式(1)で示される芳香族メチルアルコールの純度が90%以上の芳香族メチルアルコール含有粗製物1d、及び本発明の方法で製造された前記高純度芳香族メチルアルコール組成物である。しかしながら、芳香族メチルアルコール化合物は、例えば、香料等の原料として産業上の使用されること考慮すれば、高純度品である本発明の高純度芳香族メチルアルコール、又は高純度芳香族メチルアルコール組成物を保存する方法として使用することが好ましい。
本発明の保存方法で使用される保存容器の材質は、特に制限されず、例えば、ガラス、セラミック、プラスチック、金属、紙(例えば、紙袋、ダンボール箱等)、木(例えば、木箱等)、竹(例えば、編みかご、竹筒等)、麻(例えば、麻袋等)等が挙げられる。
本発明の芳香族メチルアルコールの保存方法において使用される保存剤は、前記工程(A)における蒸留の際、芳香族メチルアルコールの分解を抑制する目的で使用される分解抑制剤と同義のものが、芳香族メチルアルコール用保存剤として使用される。
上記の芳香族メチルアルコールの保存方法における保存剤の使用量は、前記高純度芳香族メチルアルコール組成物の製造方法に記載のpH範囲(pH8以上)を達成する量であれば、特に制限はない。但し、分解抑制剤の使用量があまりにも多い場合、二次加工の際に、これを取り除くことが煩雑になるので、例えば、その使用量の目安としては、芳香族メチルアルコール組成物に含まれる芳香族メチルアルコール純分含量に対して、好ましくは10~500,000ppm、更に好ましくは200~50,000ppm、より好ましくは200~30,000ppm、特に好ましくは500~15,000ppm、特により好ましくは1000~10,000ppmである。ここで、例えば、保存剤がこの範囲より少ない場合には、高純度芳香族メチルアルコールの分解抑制効果が不十分であり、また多い場合には、懸濁、スラリー状になりハンドリングが悪くなったり、芳香族メチルアルコール組成物から芳香族メチルアルコールを取り出して使用する際に中和が必要になったり、更には経済的にも望ましくない。
なお、本発明の方法で製造された前記高純度芳香族メチルアルコール組成物には、既に保存剤が、上記の量含有している。
本発明で得られた高純度芳香族メチルアルコール組成物の保存状態は、特に制限されないが、保存容器の内部が、遮光状態で、かつ窒素、アルゴン、ヘリウムなどの不活性ガス気流下、又はガス充填下であることが望ましい。
本発明の高純度芳香族メチルアルコール組成物の保存は、通常の室温環境下(例えば、0~50℃)でも、安定に保存することはできる。
しかしながら、本発明の高純度芳香族メチルアルコール、特にアルコキシ基を有する高純度芳香族メチルアルコールは、たとえ遮光状態で保存したとしても加熱環境下のみでも分解することがある。
本発明は、前記分解抑制剤であるアルカリ金属の炭酸化合物、アルカリ土類金属の炭酸化合物、アルカリ金属の炭酸水素化合物、アルカリ金属の水酸化物、アルカリ土類金属の水酸化物、アルカリ金属のリン酸塩化合物、アルカリ土類金属のリン酸塩化合物及び陰イオン交換樹脂よりなる群から選ばれる少なくとも一種の分解抑制剤を安定剤として、一般式(1):
で示される高純度芳香族メチルアルコール又は前記芳香族メチルアルコール含有粗製物に加えることを含む、高純度芳香族メチルアルコールの安定化方法に関する。より詳しくは、前記安定剤を、pH8~14の条件となるように、前記高純度芳香族メチルアルコール又は前記芳香族メチルアルコール含有粗製物に加えることを含む、芳香族メチルアルコールの安定化方法に関する。
本発明で使用される芳香族メチルアルコールは、前記≪芳香族メチルアルコール組成物の保存方法≫に記載の<使用される芳香族メチルアルコール>及び前記≪芳香族メチルアルコールの製造方法≫に記載の(本発明の芳香族メチルアルコール含有粗製物)と同じである。なお、産業の有用性を考慮すれば、好ましくは前記<使用される芳香族メチルアルコール>、(本発明の芳香族メチルアルコール含有粗製物)に記載の芳香族メチルアルコール含有粗製物1b、1c及び1dである。
前記芳香族メチルアルコールの安定化方法において使用される安定化剤は、前記工程(A)における蒸留の際の分解抑制剤と同義のものが、芳香族メチルアルコール用安定化剤として使用される。
前記安定化剤の使用量は、前記工程(A)における蒸留の際の分解抑制剤の使用量と同義である。また、安定化剤は、前記工程(A)における蒸留の際の分解抑制剤の使用と同様に、pH8~14となるように、前記高純度芳香族メチルアルコールに加える。
<後述の実施例1~3及び比較例1の高速液体クロマトグラフィー(HPLC)の分析条件>

温度計、温度調整装置、滴下装置及び撹拌措置を備えたガラス製反応容器に、1,2-メチレンジオキシベンゼン4.89kg(40mol)、トルエン3.9kg(42mol)、パラホルムアルデヒド1.41kg(43.2mol)及び36%塩酸水溶液9.10kg(90.8mol)とから、非特許文献2に記載の方法に従って、目的物であるピペロニルクロリドをトルエン溶液として9.90kg得た。
実施例1(ピペロニルアルコール粗製物1bの合成)
温度計、温度調整装置、滴下装置及び撹拌措置を備えたガラス製反応容器に、水6.9L、リン酸二水素ナトリウム・二水和物(NaH2PO4・2H2O)32g(0.20mol)を加え、液温を48℃にした。この溶液に、参考例1で合成したピペロニルクロリドのトルエン溶液9.88kg(20.1mol、ピペロニルクロリドの含量:3.45kg)を、37%水酸化ナトリウム水溶液2.50kg(23.1モル)を加え、2時間攪拌した後、引き続き、液温90℃にて3時間撹拌した。反応終了後、得られた反応液を室温まで冷却し、有機相を抽出した。また、水相は、再度トルエンを加えて分液し、有機相を抽出した。得られた有機相は、先の有機層と混合し、目的物であるピペロニルアルコール粗製物のトルエン溶液として、ピペロニルアルコール粗製物1bを10.4kg得た。
ピペロニルアルコールの含量(HPLC分析、絶対検量線法):2.73kg。
ピペロニルアルコールの含有率(HPLC分析、絶対検量線法):26質量%)。
ピペロニルアルコールの収率(ピペロニルクロリドの使用量基準):88.7%。
次に、実施例1と同様の方法にて得られたピペロニルアルコールのトルエン溶液から、その一部を、シリカゲルカラムクロマトグラフィーにて精製し、ピペロニルアルコールを白色固体として得た。本発明ではこれをHPLC分析の分析標品として使用した。
MSスペクトル〔CI-MS〕:152[M+1]
1H-NMRスペクトル〔300MHz,CDCl3,δ(ppm)〕:1.83(1H,brs),4.56(2H,s),5.94(2H,s),6.78-6.85(3H,m)。
融点:48.0~53.5℃
温度計、温度調整装置、滴下装置及び撹拌措置を備えたガラス製反応容器に、実施例1と同様の方法で製造したピペロニルアルコール130.24gを純分として含むトルエン溶液(ピペロニルアルコール粗製物1b)577.32gを加えた。次に、このトルエン溶液に分解抑制剤として炭酸ナトリウム2.89g(ピペロニルアルコール粗製物中のピペロニルアルコールの純分質量に対して22000ppm)を加え、減圧にて溶媒を留去した。次いで、得られた残渣を釜温127℃、真空度0.64kPa(4.8Torr)の条件にて単蒸留を行い、主留分として、純度98.0%のピペロニルアルコールを125.92g(回収率94.8%)得た。なお、蒸留中の内容物のpHは、pH試験紙で約10であった。また、主留分、釜残分、溶媒混入分中のピペロニルアルコールを合計すると130.24g(回収率100%)であった。また、得られたピペロニルアルコール中には、式(3d)に記載のビス(3,4-メチレンジオキシベンジル)エーテルは混入していなかった。
分解抑制剤として、水酸化ナトリウムをピペリニルアルコールの純分質量に対して0.1wt%添加した以外は、実施例2と同様の条件で蒸留を行った。その結果、主留分として純度98.9%のピペロニルアルコールを取得収率94.3%で得た。なお、蒸留中の蒸留釜内容物のpHは、pH試験紙で約10であった。また、主留分、釜残分、溶媒混入分中のピペロニルアルコールを合計すると130.24g(回収率100%)であった。また、得られたピペロニルアルコール中には、式(3d)に記載のビス(3,4-メチレンジオキシベンジル)エーテルは混入していなかった。
分解抑制剤を添加しない以外は、実施例2と同様の条件で蒸留を行った。その結果、主留分として純度96.2%のピペロニルアルコールを取得収率82.8%で得た。また、主留分、釜残分、溶媒混入分中のピペロニルアルコールを合計すると回収率は、わずか83.5%であり、工程(A)の蒸留操作中に16.5%のピペロニルアルコールが分解していることが確認された。また、実施例2と同じ主留分の取得条件下で得られたピペロニルアルコール中には、式(3d)に記載のビス(3,4-メチレンジオキシベンジル)エーテルが1.64%も混入していた。
なお、蒸留釜内容物を一部取り出し、pHメーターを用いて、pHを測定したところ、蒸留開始前の蒸留釜内容物のpHは6.0、蒸留開始後の蒸留釜内容物のpHは2.6であった。
実施例4(分解抑制剤(炭酸ナトリウム)の添加による分解抑制効果:ピペロニルアルコール)
参考例1及び実施例1と同様の方法で得たピペロニルアルコールのトルエン溶液から、トルエンを留去し、ピペロニルアルコールの濃縮物を得た。次に、得られた濃縮物に分解抑制剤として炭酸ナトリウムを、1000ppm加え、安定化測定用のサンプルを作製した。
これを、温度140℃の高温槽につけて、経時的(4時間後、8時間後、12時間後、16時間後、20時間後)に、高速液体クロマトグラフィー分析(HPLC:絶対検量線法)を行い、ピペロニルアルコールの純度(質量%換算)を測定した。なお、その保存率は、[各核時間経過後のピペロニルアルコールのHPLC純度(面積%)]/[測定開始のピペロニルアルコールのHPLC純度(面積%)]×100(%)とした。即ち、測定開始時の保存率を100%とし、一定時間経過後の数値が高いほど安定に保存できることを示す。その結果を以下の表1に示す。
分解抑制剤(炭酸ナトリウム)を使用しなかった以外は、前記実施例4の比較として、同様の実験を行った。その結果を以下の表2に示す。
参考例1及び実施例1と同様の方法で、ピペロニルアルコールの純度96.4%(HPLC:絶対検量線法)のピペロニルアルコールのトルエン溶液を得た。次に、得られた前記トルエン溶液を、20mlのガラスフラスコに小分けし、これに分解抑制剤として炭酸ナトリウムを、188ppm(実施例5)、489ppm(実施例6)、710ppm(実施例7)、950ppm(実施例8)をそれぞれ加えた後、トルエンを留去し、安定化測定用のサンプルを作製した。
次に、この安定化測定用のサンプルを、温度160℃の恒温槽につけ、経時的(7時間後、14時間後、21時間後)に、高速液体クロマトグラフィー分析(HPLC:絶対検量線法)を行い、ピペロニルアルコールの純度(質量%換算)を測定した。その結果を以下の表3に示す。
分解抑制剤(炭酸ナトリウム)を使用しなかった以外は、前記実施例4~7の比較として、同様の実験を行った。その結果を以下の表3に示す。
*2:高速液体クロマトグラフィー分析(HPLC:絶対検量線法)によるビス(3,4-メチレンジオキシベンジル)エーテル(式(3d)の含有量(質量%:wt%)を示す。
*3:分解抑制剤は、炭酸ナトリウムを使用。
p-アニスアルコール(和光純薬製)を用いて、これに分解抑制剤として炭酸ナトリウムを10,000ppm加えて、安定化測定用のサンプルを作成した。
次に、この安定化測定用のサンプルを、温度160℃の恒温槽につけ、経時的(5時間後、10時間後)に、高速液体クロマトグラフィー分析(HPLC:絶対検量線法)を行い、p-アニスアルコールの保存率(%)を測定した。なお、その保存率は、実施例4の算出方法同じである。また、実施例4と同様に、測定開始時の保存率を100%とし、一定時間経過後の数値が高いほど安定に保存できていることを示す。その結果を以下の表4に示す。
分解抑制剤(炭酸ナトリウム)を使用しなかった以外は、前記実施例8の比較として、同様の実験を行った。なお、その保存率は、実施例4の算出方法同じである。その結果を以下の表4に示す。表4より、分解抑制剤を入れなかった比較例4では、わずか10時間でほぼ10%の分解が確認された。
実施例10(高純度ピペロニルアルコールの保存安定性(90℃):方法(2)、分解抑制剤:炭酸ナトリウム)
30mLのガラス製容器に、実施例2と同様の方法で得られた高純度ピペロニルアルコール5gを加熱融解して加え、更にこれに炭酸ナトリウム0.025g(高純度ピペロニルアルコールの使用量に対して、5000ppm)を加えた。得られた高純度ピペロニルアルコール組成物が入った容器は密栓し、放冷にて固化させて、固体として高純度ピペロニルアルコール組成物の保存容器を作成した。次に、これを90℃の恒温装置内で28日間保存する試験を行った(保存開始の内容物のpHは、pH試験紙で約10であった)。前記温度下で保存開始後、7日後、14日後、21日後、28日後に前記組成物の高速液体クロマトグラフィー分析(HPLC:絶対検量線法)を行い、純分含量(質量%)としてピペロニルアルコールの純度を算出した。その結果を以下の表5に示す。
保存温度を45℃に変えた以外は、実施例10と同じ方法により保存し、同様に保存開始後、7日後、14日後、21日後、28日後の高速液体クロマトグラフィー分析(HPLC:絶対検量線法)によるピペロニルアルコールの純度(質量%換算)を算出した。その結果を以下の表5に示す。なお、保存開始の内容物のpHは、pH試験紙で約10であった。
分解抑制剤を加えなかった以外は、実施例10及び11とそれぞれ同じ方法で、保存温度を90℃(比較例5)、45℃(比較例6)下にて保存し、同様に保存開始後、7日後、14日後、21日後、28日後のピペロニルアルコールの純度を高速液体クロマトグラフィー分析(HPLC:絶対検量線法)で測定した。その結果を表5に示す。

実施例2と同様の方法で得られた高純度ピペロニルアルコール(ピペロニルアルコールの純度96.6wt%;HPLC:絶対検量線法、式(3)の純度0wt%)10gを用い、分解抑制剤(炭酸ナトリウム)の使用量を、200ppm(実施例12)、500ppm(実施例13)、750ppm(実施例14)、1000ppm(実施例15)に変えた以外は、実施例10と同じ方法により、保存温度を90℃にして保存開始した。なお、保存開始の内容物のpHは、pH試験紙で約10であった。
30日経過後、それぞれのサンプルを高速液体クロマトグラフィー分析(HPLC:絶対検量線法)により、ピペロニルアルコールの純度(質量%換算)を測定した。その結果を以下の表6に示す。

Claims (21)
- 一般式(1):
R1及びR2は、水素原子、炭素原子数1~12のアルキル基、フェニル基、ベンジル基、アリル基又はプロパルギル基を示し、これらの基は置換基を有していても良い。
nは、置換基OR2の個数であり、0から3の整数を示す。nが2以上の場合、R2は、互いに同一又は異なっていてもよい。芳香族環上の置換基(OR1,OR2)が、芳香環上の隣接する炭素上に存在する場合、R1とR2とが互いに結合して、環状構造を形成しても良い。
R3は、ハロゲン原子(フッ素原子、塩素原子、臭素原子及びヨウ素原子)、ニトロ基、シアノ基、メチル基、エチル基、トリフルオロメチル基又はフェニル基を示す。R3と芳香族環上の置換基OR1又はOR2とが、芳香族環上の隣接する炭素上に存在する場合、R3とR1又はR2とが互いに結合して、環状構造を形成しても良い。
mは、置換基R3の個数であり、0から3の整数を示す。なお、mが2以上の場合、R3は、互いに同一又は異なっていてもよい。但し、n+mは0から4の整数である。)
で示される芳香族メチルアルコール含有粗製物を分解抑制剤の存在下にて蒸留し、一般式(1)で示される高純度芳香族メチルアルコールを得る工程を含む、高純度芳香族メチルアルコールの製造方法。 - 芳香族メチルハライドが、4-メトキシベンジルクロリド、3,4-ジメトキシベンジルクロリド、3,4,5-トリメトキシベンジルクロリド、3,4-エチレンジオキシベンジルクロリド、又は3,4-メチレンジオキシベンジルクロリドである、請求項2又は3に記載された高純度芳香族メチルアルコールの製造方法。
- 分解抑制剤が、アルカリ金属の炭酸化合物、アルカリ土類金属の炭酸化合物、アルカリ金属の炭酸水素化合物、アルカリ金属の水酸化物、アルカリ土類金属の水酸化物、アルカリ金属のリン酸塩化合物、アルカリ土類金属のリン酸塩化合物及び陰イオン交換樹脂よりなる群から選ばれる少なくとも一種である、請求項1~4のいずれか一項に記載された高純度芳香族メチルアルコールの製造方法。
- 蒸留を、蒸留釜の液温70~240℃にて行うことを特徴とする、請求項1~5のいずれか一項に記載された高純度芳香族メチルアルコールの製造方法。
- 蒸留を、蒸留釜の内容物のpHが、pH8~14にて行うことを特徴とする、請求項1~6のいずれか一項に記載された高純度芳香族メチルアルコールの製造方法。
- 請求項1~7のいずれか一項に記載した方法により製造された、請求項8に記載された高純度芳香族メチルアルコール。
- 高純度芳香族メチルアルコールが、請求項9に記載された高純度芳香族メチルアルコールである、請求項10に記載された高純度芳香族メチルアルコール組成物。
- 分解抑制剤の使用量が、高純度芳香族メチルアルコール中の芳香族メチルアルコールの純分質量に対して、200~50,000ppmである、請求項10又は11に記載された高純度芳香族メチルアルコール組成物。
- 請求項10~12のいずれか1項に記載された高純度芳香族メチルアルコール組成物が保存された容器。
- 蒸留を、蒸留釜の内容物のpHが、pH8~14にて行うことを特徴とする、請求項15に記載された芳香族メチルアルコールの蒸留方法。
- 安定化剤を、pH8~14となるように、前記高純度芳香族メチルアルコールに加えることを含む、請求項18に記載された高純度芳香族メチルアルコールの安定化方法。
