以上の結果は、雑種起源等により、異質な染色体の対合が困難となった時、未解明の分子細胞機構により、減数分裂前に染色体の倍加が起きることを示唆している。従って、対合するべき染色体を持たない半数体の生殖細胞では、特別な処理をしなくても、自律的に染色体が倍加する可能性が考えられる。もし、この様なことが起これば、葯培養におけるゲノム倍加と同様な現象が魚類半数体生殖細胞においても期待しうる。

ドジョウ卵と紫外線照射したキンギョ精子、又は紫外線照射したドジョウ卵とドジョウ精子、キンギョ卵と紫外線照射した四倍体フナ精子、又はゼブラフィッシュ卵と紫外線照射したキンギョ精子を媒精後、ドナーの雌性発生又は雄性発生半数体群を作出する。また、ホスト二倍体として通常ドジョウ卵とドジョウ精子による受精により作出した通常二倍体受精群、ホスト二倍体として通常キンギョ卵とキンギョ精子による受精により作出した通常二倍体受精群、通常ゼブラフィッシュ雌と通常ゼブラフィッシュ雄の自然交配による通常二倍体受精群も作出する。さらに、ホストとする三倍体受精群は、ドジョウの場合は通常ドジョウ卵と四倍体ドジョウ(自然四倍体雄×二倍体雌の人工受精後、第二極体放出阻害により得られた四倍体:ネオ四倍体)の二倍性精子による受精によって、キンギョの場合は通常キンギョ卵と四倍体フナ(自然界から採集された個体)の二倍性精子による受精によって作出する。ドジョウ半数体PGCは、ドジョウ卵×ドジョウ精子、ドジョウ卵×ネオ四倍体ドジョウ精子の胚へ移植し、キンギョ半数体PGCは、キンギョ卵×キンギョ精子、キンギョ卵×四倍体フナ精子の胚へ移植し、ゼブラフィッシュ半数体PGCはゼブラフィッシュ二倍体胚へ移植した。三倍体は二倍体と異なり生殖腺の内部構造に空洞が多く、大型の生殖細胞や運動能のない精子を形成することが知られている(Fujimoto et al., Reproductive capacity of neo-tetraploid loaches produced using diploid spermatozoa from a natural tetraploid male. Aquaculture 308: S133-139(2010))。受精後未受精卵を取り除き、さらに死卵の計数と除去、及び水換えを行う。孵化期には孵化仔魚の形態を観察し、正常仔魚出現率と半数体症候群などの奇形を呈する仔魚の出現数を調査する。
顕微注入あるいはPGCの移植のために、Yamaha, E. and Yamazaki, F.: Electrically fused-egg induction and its development in the goldfish, Carassius auratus. Int. J. Dev. Zool. Sci. 23:977-989. (2006)に従い受精卵より卵膜を除去するとともに、孵化まで培養する。
精子又は卵の遺伝的な不活化の成否を検討するために、キメラ作出時に用いたドナー胚の倍数性を調査する。また、ホストが三倍体又は二倍体であるか否かを検討するために、卵膜除去をしていないコントロールのホスト仔魚の倍数性を調べる。ドナー胚及びホスト仔魚は、核単離液及び核染色液中にて細胞を混合・遊離する。その後、フローサイトメーターにより相対DNA量を測定する。この際、実験に用いた種の正常二倍体魚の鰭のDNA量を二倍体DNA量(2C)の、正常二倍体ドジョウの半数体精子のDNA量を半数体DNA量(1C)の基準とする。半数体以外の細胞をもつドナーが検出された場合、そのドナーを用いて作出したキメラは実験から排除する。ドジョウでは、さらに2n-3nキメラ作出に用いたドナー胚及びホスト仔魚においても、倍数性を調べる。キンギョ卵×四倍体フナ精子から作製した胚又は個体の鰭のDNA量(3C)を三倍体の基準とする。
ドナー半数体及びホスト二倍体又はホスト三倍体PGCsを区別して可視化するために、マーカー、好ましくは緑色蛍光を発するGFP nos1 3’UTR mRNAと赤色蛍光を発するDsRed nos1 3’UTRmRNAを注入する。ドナーとして、卵膜除去卵に蛍光mRNAを注入する。ホストについても同様にドナーと異なるmRNAを注入する。同時に、ホスト三倍体を不妊化するためdndMOを注入する。ゼブラフィッシュでは、PGCがGFP蛍光を発する遺伝子組換え体を用いても良い。
異なる手法により生殖系列キメラを作出する。例えば、BT(Blastomere transplantation)法に従って、胞胚期ドナー胚の胚盤下部の割球を胞胚期ホスト胚に移植することにより生殖系列キメラを誘起する。別の手法としてSPT(single primordial germ cell transplantation)法、またはPPT(poly primordial germ cell transplantation)法により、体節形成期のドナー胚に顕在化しているPGCsを、胞胚期のホスト胚の胚盤下部に移植することで誘起してもよい。さらに、sandwich法で半数体の胞胚期の胚盤の下部を切り出し、ホスト胞胚胚盤の中部に移植することでも誘起する。作製した生殖系列キメラ個体において、半数体PGCが生殖隆起へ到達した個体数を計数する。異なる蛍光mRNAで標識したドナー胚とホスト胚を用いて作出した1n-3n及び2n-3nにおいて、生殖隆起へ到達した半数体PGCを計数する。検出したPGC数により、キメラを異なる水槽に分けて収容し、飼育する。このとき異なる蛍光により区別されるホスト三倍体由来のPGCが観察された個体は、ホストの不妊化に失敗したものとして実験から排除する。1n-3n及び2n-3nにおいては、PGCを観察し、個体をサンプリングする。
ドジョウでは、孵化後の1n-3n、2n-3nと正常二倍体ドジョウ(以下、2nともいう)の成魚(雌雄)、dndMO処理により不妊化していない三倍体(正常ドジョウ卵×四倍体ドジョウ(自然四倍体雄×二倍体雌の人工受精後、第二極体放出阻害により得られたネオ四倍体)精子)(以下、「3n」ともいう)の成魚(雌雄)を、キンギョでは、孵化後の生殖系列キメラと正常二倍体(2n)の成魚(雌雄)、dndMO処理により不妊化していない三倍体(正常キンギョ卵×四倍体フナ精子)(3n)の成魚(雌雄)を固定した後、エタノール中で保存する。固定したキメラは切片にし、組織標本を作製する。その後、二重染色を施し、封入後、観察に供する。ドジョウでは、発達したPGCの増殖を調査するために、キメラの未分化生殖細胞もしくはA型精原細胞もしくは卵原細胞を計数し、孵化稚魚期に蛍光顕微鏡で観察したPGC数に対して何個増殖しているのかを調査する。
ドジョウでは、1n-3n、2n-3n雄成魚に、キンギョでは、1n-3n生殖系列キメラ魚に、ヒト絨毛性生殖腺刺激ホルモン(以下、「hCG」ともいう。)を腹腔内に注射し、排精排卵を誘起する。注射してから一定時間後に、腹部を圧迫し、得られた精液を採精する。採精した精子を人工精漿で希釈し、精液を観察する。その後淡水と混合し精子の運動能を調査する。
ゼブラフィッシュの生殖系列キメラにおいては、正常雌親魚と1:1で水槽に入れ、産卵行動を観察するとともに、産卵した卵に受精卵が含まれているか否かを確認する。
ドジョウでは、1n-3n及び2n-3n成魚、キンギョ及びゼブラフィッシュでは生殖系列キメラ成魚から精巣及び卵巣の一部を取り出し、それらの倍数性をフローサイトメトリーで調査する。また、ドジョウでは、孵化後の1n-3n、2n-3nから採精した精子の倍数性を調べる。コントロールとして、成魚の2nと成魚のdndMO処理により不妊化してしない3nの精巣及び卵巣の倍数性も調査する。
キメラ成魚の発達した生殖腺内の細胞が生殖細胞か否かの検討するために、生殖細胞特異的なmRNAの発現を調査する。孵化後の1n-3n、2n-3nの成魚から生殖腺の一部を取り出し、RNAを抽出する。抽出したRNAは逆転写をし、cDNAを合成する。なお、内部コントロールとしてβ-actinのRT-PCRも行う。PCR反応により、キメラ生殖腺における生殖細胞特異的なmRNAの発現の有無を電気泳動によって確認する。
1n-3n生殖系列キメラから得られた精子と体細胞である鰭の細胞よりDNAを抽出し、RAPDマーカーにより異なる遺伝子構成であることを明らかにする。
1n-3n生殖系列キメラから得られた精子で受精させたことによって得られた個体の遺伝子型を、マイクロサテライトマーカーで検討し、精子の遺伝子型が全て同じであることを確認する。
A.材料と方法
1.ドジョウMisgurnus anguillicaudatusを材料に、下述の発生工学的手法により、ドナーに半数体、ホストに正常二倍体を用いた半数体-二倍体生殖系列キメラ(1n-2n)、及びドナーに半数体、ホストに三倍体を用いた半数体-三倍体生殖系列キメラ(1n-3n)を作出した。1n-2nは正常二倍体個体内で、半数体PGCsがホストの二倍体PGCsと同じように生殖隆起へ移動するのか否かを調べることを目的として作出した。1n-3nは、デッドエンドアンチセンスモルホリノオリゴヌクレオチド(配列番号No.1; 5’-GATCTGCTCCTTCCATTGCGTTTGC-3’)でノックダウンして不妊化処理をした三倍体ホスト個体の内で、半数体PGCsが生殖隆起へ移動し、生殖細胞として増殖するか否か、そして、それらの生殖腺から機能的な配偶子が形成されるか否かを検討するために作出した。将来的に、機能的な配偶子が形成されるか否かを検討する場合、得られた子孫の色素によりドナー由来か否かを判別するために、ドナーとして色彩変異体(劣性形質)のヒドジョウ又はアルビノドジョウを使用した。さらに、1n-3nの対照として、ドナーに正常二倍体、ホストに三倍体を用いた二倍体-三倍体生殖系列キメラ(2n-3n)を作出した。
本発明では、北海道大学大学院水産科学院育種生物学講座にて飼育中のドジョウMisgurnus anguillicaudatusとキンギョCarassius auratusを材料として用いた。ドジョウは北海道岩見沢市北村地区及び北海道北斗市大野町において採取したもの、あるいはその子孫を用いた。
20IU/gのhCG(あすか製薬株式会社製)をドジョウ親魚の腹腔内に注射し、排卵、排精を誘起した。雌では12-14時間後に排卵を確認した後、サランラップ(ポリ塩化ビニリデンフィルム)(旭化成株式会社製)を張った直径90mmプラスチックシャーレ上で腹部を圧迫して採卵した。雄も同様に腹部を圧迫し、得られた精液をヘマトクリット毛細管(テルモ製)で採精した。得られた精液はKurokura’s solution(NaCl 750mg, CaCl2 20mg, NaHCO320mg, KCl 20mg/100ml)で約25倍に希釈した。キンギョにおいては精子のみを使用した。20IU/gのhCGをキンギョ親魚の腹腔内に注射し、排精を誘起した。12-14時間後に、腹部を圧迫し、得られた精液をヘマトクリット毛細管で採精した。
卵及び精子の遺伝的不活化には紫外線照射を用いた。上部に2灯の紫外線殺菌灯(GL15W, National社製)を設置した箱形紫外線照射装置を用いた。卵の遺伝的不活化は、Fujimoto et al Journal of Experimental Zoology, 307A: 449-462, DOI: 10.1002/jez.398 (2007)を一部改良し行った。すなわち、卵を収容するチャンバーとして、底を抜き、サランラップ(旭化成株式会社製)を張った90mmプラスチックシャーレを用いた。また、チャンバー内にはさらに底を抜いた50mmプラスチックシャーレ(直径50mm, 高さ10mm)をセットし、その中に0.5%のウシ血清アルブミンを含有するHank’s solution(0.137M NaCl、5.4mM KCl、0.25mM Na2HPO4、0.44mMKH2 PO4、1.3mM CaCl2、1.0mM MgSO4、4.2mM NaHCO3)を約3ml入れ、卵を一層になるように収容したのち、振盪機(VS2, サクラ精機株式会社、日本)の上に乗せ、水平に振盪しながら上面より紫外線を照射した。なお、紫外線照射量を150mJ/cm2とした。精子の遺伝的不活化において、よく洗浄し水分を拭き取ったガラスシャーレ上にキンギョ精子をKurokura’s solutionで約100倍に希釈し約0.1mmの厚さになるように伸展した後、紫外線を照射した。なお、紫外線照射量は60mJ/cm2とした。キンギョを使用した理由は、ドジョウの雌性発生において、キンギョの精子を使えば、紫外線照射から逃れた精子からの受精卵は致死雑種となり除去できるためである(非特許文献1)。
ドジョウ卵と紫外線照射したキンギョ精子、又は紫外線照射したドジョウ卵とドジョウ精子を媒精後、20℃の水道水を満たしたプラスチックシャーレ内に散布してシャーレの底に付着させる方法でドナーの雌性発生又は雄性発生半数体群を作出した。また、ホスト二倍体として通常ドジョウ卵とドジョウ精子との受精により作出した通常受精群も作出した。受精後、各実験群は20℃に設定したインキュベーター内で培養した。さらに、ホストとする三倍体受精群は通常ドジョウ卵と四倍体ドジョウ(自然四倍体雄×二倍体雌の人工受精後、第二極体放出阻害により得られた四倍体:ネオ四倍体)の二倍性精子による受精によって作出した。三倍体は二倍体と異なり生殖腺の内部構造に空洞が多く、大型の生殖細胞や運動能のない精子を形成することが知られている(Fujimoto et al., Aquaculture 308: S133-139(2010))。受精後2時間以内に未受精卵を取り除き、さらに1日毎に死卵の計数と除去、及び水換えを行った。孵化期には孵化仔魚の形態を観察し、正常仔魚出現率と半数体症候群などの奇形を呈する仔魚の出現数を調査した。なお、ドジョウの発生段階はFujimoto et al Zoological Science, 23:977-989, DOI: 10.2108/zsj.23.977 (2006)の報告に従った。
各実験群の約半数程度の1~2細胞期の受精卵について卵膜除去液(0.1%トリプシンと0.4%尿素を含む淡水魚用リンゲル液;128mM NaCl, 2.8mM KCl, 1.9mM CaCl2, pH 7.0)で処理することにより卵膜を除去した。卵膜除去卵は、約1%のアガロースで底面を被膜処理し20℃の一次培養液(1.6%ニワトリ卵白と0.01% ペニシリンとストレプトマイシンを含む淡水魚用リンゲル液;128mM NaCl, 2.8mM KCl, 1.9mM CaCl2, pH 7.0)を満たしたガラスシャーレ内で体節形成期まで培養した。その後、卵膜除去卵を二次培養液(0.01% ペニシリンとストレプトマイシンを含む 1.8mM CaCl2, 1.8mM MgCl2液)に移し、孵化まで培養した。
精子又は卵の遺伝的な不活化の成否を検討するために、キメラ作出時に用いたドナー胚の倍数性をフローサイトメーター(Partec Ploidy Analyser, PA型)を用いて調査した。また、ホストが三倍体又は二倍体であるか否かを検討するために、卵膜除去をしていないコントロールのホスト仔魚の倍数性を調べた。ドナー胚及びホスト仔魚は、Cystain DNA 2 step kit(Partec)のA液(核単離溶液)150μlの中にそれぞれ入れ、20分間放置し、ボルテックスにより細胞を混合・遊離した。その後、試料を50μmのメッシュ(Cell Trics 50μm, Partec)で濾過し、細胞の破片を除去した後、A液の5倍量のB液(4’,6-ジアミジノ-2-フェニリンドール(DAPI)を含む染色液)を加え、フローサイトメトリーにより相対DNA量を測定した。この際、正常二倍体ドジョウ鰭のDNA量を二倍体DNA量(2C)の、正常二倍体ドジョウの半数体精子のDNA量を半数体DNA量(1C)の基準とした。半数体細胞以外をもつドナーが検出された場合、そのドナーを用いて作出したキメラは実験から排除した。さらに2n-3nキメラ作出に用いたドナー胚及びホスト仔魚においても、倍数性を調べた。
1.コントロールの受精数、孵化仔魚数、正常仔魚数
キメラ作成に用いたドナー及びホストは表1にまとめた。ドナーの半数体は、雌性発生、雄性発生により作出し、一方、ドナーの二倍体は通常受精で作成した。ホストとしては三倍体又は二倍体を用いた(表1)。キメラを作出する際のコントロールとして、何の処理もしていない「コントロール」と、卵膜を除去した「卵膜除去コントロール」、蛍光mRNA及びdndMOを注入した「蛍光mRNA注入コントロール」の群を作った。コントロールにおいては、使用卵数、受精卵数、孵化仔魚数、正常仔魚数、奇形仔魚数を計数した。卵膜除去コントロール及び蛍光mRNA注入コントロールにおいては、受精卵から正常なものだけを選び、孵化仔魚、正常仔魚数、奇形仔魚数を計数した(表2)。その結果、ホストとして作出した個体のほとんどから正常仔魚が得られた。また、ドナーとして作出した雌性発生群及び雄性発生群は、孵化後、ほとんどがいわゆる半数体症候群と判定される奇形仔魚を呈した。受精卵の卵膜除去及び蛍光mRNA注入を行っても、孵化仔魚の率は無処理コントロールと変わらず、これらの処理の悪影響は見られなかった。
精子又は卵の遺伝的な不活化の成否を検討するために、キメラ作出時に用いたドナー胚の倍数性を調査した(表3)。その結果、キメラ作出に用いた雌性発生、雄性発生を行ったドナー胚は、ほとんどが半数体細胞から構成されていた。しかし、一部の胚は細胞が壊れていたため、倍数性を検出できなかった。このとき、倍数性を検出できなかったドナー胚から作出したキメラは実験より除いた。一方、2n-3n作出に用いた正常受精により得たドナー胚は、ほとんどが二倍体細胞から構成されていた。通常卵とネオ四倍体の二倍性精子の受精により作出したほとんどのホスト孵化仔魚の倍数性は、三倍体細胞から構成されていた。しかし、実験5では10個体中に四倍体が1個体生じた。キメラ作出に用いたドナー及びホストはほとんどの場合、半数体、三倍体であることが確認できた。
1.PGCsの標識、ホスト三倍体の不妊化
ドナー半数体及びホスト二倍体又はホスト三倍体PGCsを区別して可視化するために、緑色蛍光を発するGFP nos1 3’UTR mRNAと赤色蛍光を発するDsRed nos1 3’UTRmRNAを注入した(Koprunner, et al., Genes. Dev. 15: 2877-2885 (2001)。ドナーとして、1~2細胞期の卵膜除去卵に蛍光mRNA 100ng/μl 及び5%ビオチンデキストラン(Sigma社製)を含む0.2M KClを顕微注入した。ホストについても同様にドナーと異なるmRNA 100ng/μl及び10%ビオチンデキストランを含む0.2M KClを顕微注入した。同時に、ホスト三倍体を不妊化するため2000~4000pg/胚のdndMOを顕微注入した。
異なる手法により生殖系列キメラの作出を試みた。まず、BT(Blastomere transplantation)法に従って、胞胚期ドナー胚の胚盤下部の割球を胞胚期ホスト胚に移植することにより生殖系列キメラを誘起した。別の手法としてSPT(single primordial germ cell transplantation)法により、体節形成期のドナー胚に顕在化している1~5個のPGCsを、胞胚期のホスト胚の胚盤下部に移植することで誘起した。異なる蛍光mRNAで標識したドナー胚とホスト胚を用いて作出した1n-3n及び2n-3nにおいて、生殖隆起へ到達した半数体PGCを計数した。検出したPGC数により、キメラを異なる水槽に分けて収容し、飼育した。このとき異なる蛍光により区別されるホスト三倍体由来のPGCが観察された個体は、ホストの不妊化に失敗したものとして実験から排除した。1n-3n及び2n-3nにおいては、孵化後8日まで蛍光顕微鏡下でPGCを観察し、孵化後1、2、4、12、15.5、16ヶ月齢の個体を各5~10個体サンプリングした。
1.半数体PGCsの生殖隆起への移動
PGCsのキメラ胚内での移動を観察した。1n-2nキメラ(表4 実験1)を蛍光顕微鏡で観察した結果、GFP蛍光mRNAを注入したドナー1n由来のPGCsは緑色蛍光を発していた(図1)。DsRed蛍光mRNAを注入したホスト2n由来のPGCsは赤色蛍光を発していた(図1)。ドナー1n及びホスト2n由来のPGCsは、移植後の体節形成期にほぼ同様の場所に位置し(図1 A2,3, B2,3,C2,3)、いずれのPGCsも孵化期に生殖隆起へ移動した(図1 D2, 3, E2,3; 表4 実験1)。その後、孵化後8日までドナー1n、ホスト2nのPGCsはいずれも生殖隆起に局在することが確認できた。一方、2n-3nキメラにおいては、ドナー2n由来のPGCsが、dndMO処理により不妊化した三倍体ホストの内で孵化期に生殖隆起まで移動した(表4 実験7, 9)。1n-3nキメラにおいては、2n-3nキメラと同様に、ドナー1nのPGCsがdndMO処理により不妊化した三倍体ホストの内で孵化期に生殖隆起まで移動した(表4 実験2, 3, 4, 5, 6, 8, 9)。dndMO処理を行った3nをホストにしたキメラでは、ホスト由来のPGCsは認められなかった(表4 実験2, 3, 4, 5, 6, 7, 8, 9)。
全実験で作出したキメラにおいて、多くの孵化仔魚が得られ、また仔魚が正常な形態を呈した個体の割合も高かった(表4)。さらに、蛍光顕微鏡下で正常仔魚のドナーPGCsの位置を調べ、ホストPGCsの存(+)否(-)も同時に調べた。ドナーPGCsが生殖隆起に確認された個体には、dndMO処理により不妊化を行ったホスト由来のPGCsは観察されなかった。そして、表3で示したように、生殖隆起にドナー由来のPGCが確認されたキメラにおいて、ドナー胚が半数体であると証明されたキメラを、正式な生殖系列キメラとした。ところで、2つのキメラ作出法を用いたが、SPT法の方がBT法よりも効率的にドナーPGCsが生殖隆起へ移動した生殖系列キメラを誘起することができた。
1.組織学的観察
孵化後1、2、4、12、15.5、16ヶ月齢の1n-3n、2n-3nと正常二倍体ドジョウ(2n)の成魚(雌雄)、dndMO処理により不妊化していない三倍体(通常ドジョウ卵×四倍体ドジョウ(二倍体雌×自然四倍体雄の人工受精後、第二極体放出阻止により得られたネオ四倍体)精子)(3n)の成魚(雌雄)をブアン氏液(ピクリン酸飽和溶液:ホルマリン:酢酸=15:5:1)で6~12時間固定した後、80%エタノール中で保存した。固定したキメラはエタノール系列又はブタノール系列で脱水し、エタノール系列ではキシレンにて透徹後、パラフィンに包埋した。包埋したパラフィンブロックをミクロトームで厚さ6又は8μmの切片にし、組織標本を作製した。その後、ヘマトキシリンエオシン二重染色を施し、封入後、観察に供した。Fujimoto et al(Sexual dimorphism of gonadal structure and gene expression in germ cell-deficient loach, a teleost fish.PNAS, 107(40): 17211-17216 (2010))により、dndMO処理により不妊化した個体は、雄においては生殖細胞がない未発達のチューブ状精巣構造を示し、雌においても、生殖細胞がない未発達のリボン状卵巣構造を示すことが明らかになっていることから、本発明では、特にdndMO処理3nを不妊化のコントロール群として作出しなかった。発達したPGCの増殖を調査するために、1、2、4、12、15.5、16ヶ月齢のキメラの未分化生殖細胞もしくはA型精原細胞もしくは卵原細胞を計数し、孵化稚魚期に蛍光顕微鏡で観察したPGC数に対して何個増殖しているのかを調査した。
12ヶ月齢の1n-3n、2n-3n雄成魚に、2日に分けて20IU/gのhCGを腹腔内に注射し、排精を誘起した。2日目に注射してから12-14時間後に、腹部を圧迫し、得られた精液をヘマトクリット毛細管で採精した。得られた精液はKurokura’s solutionで約10倍に希釈した。希釈した精液を、光学顕微鏡下で形態を観察した。その後淡水と混合し精子の運動能を調査した。
組織像から1n-3nと2n-3n間で特にA型精原細胞のサイズに違いが観察された。そのため、より詳細に1、2、4、12、15.5、16ヶ月齢の1n-3nと2n-3nについて、未分化生殖細胞(A型精原細胞か卵原細胞なのか判別できない発達初期の細胞)及びA型精原細胞、卵原細胞の核の直径を測定し頻度を算出した(図7, 8, 9)。同時にコントロールとして2nとdndMO処理により不妊化していない3nの成魚のA型精原細胞及び卵原細胞の核の直径の頻度も求めた。
12、15.5、16ヶ月齢の1n-3n及び2n-3n成魚から精巣及び卵巣の一部を取り出し、それらの倍数性をフローサイトメトリーで調査した。また、孵化後12ヶ月齢の1n-3n、2n-3nから採精した精子の倍数性を調べた。コントロールとして、成魚の2nと成魚のdndMO処理により不妊化していない3nの精巣及び卵巣の倍数性も調査した。
1.生殖腺の外部形態
2n-3nキメラの固定した生殖腺の外部形態を観察したところ、孵化後1、2ヶ月齢の個体では、細い1対2本の生殖腺が見られた(図2A)。4ヶ月齢になると、部分的に発達した生殖腺が観察できた(図2B)。雌雄の判別が可能な成魚となった雄の12、15.5、16ヶ月齢ではより大きく発達した生殖腺(精巣)が観察できた(図2C)。1n-3nキメラでは孵化後1、2、4ヶ月齢において、細い1対2本の生殖腺が見られ(図2D, E)、12ヶ月齢では、2n-3nと同様に部分的に大きく発達した生殖腺が観察された(図2F)。2nの雌成魚では、腎臓に沿って一層になっている非常に発達した卵母細胞を多数持つ卵巣が見られた(図3A)。dndMO処理により不妊化していない3nの成魚の卵巣は、腎臓に沿って一層に広がった卵巣が見られたが、その卵巣には2n程発達した卵母細胞がなかった(図3B)。15.5ヶ月齢の2n-3n雌成魚は2nと同様に発達した卵母細胞を少数持つ卵巣が見られた(図3C)。一方、1n-3nでは腎臓に沿って一層に広がった卵巣が見られたが、その卵巣には、2n及び2n-3n程発達した卵母細胞は観察されなかった(図3D)。
1n-3n、2n-3nにおいて、キメラ誘起後に蛍光顕微鏡下で生殖隆起に観察されたドナーPGCsを計数、PGCs数(1、2、3、4、5~)に応じてキメラを分けて飼育した。そして生殖腺の組織像から、1~16ヶ月齢の1n-3n及び2n-3nの各個体の未分化生殖細胞(12ヶ月齢以降の個体に関して、雄についてはA型精原細胞、雌については卵原細胞)を計数し、キメラ誘起時に計数したPGCs数(1、2、3、4、5~)に対して未分化生殖細胞が何個増殖、維持又は減少したかを調べた(表5, 6)。その結果、2n-3nでは1ヶ月齢で未分化生殖細胞の増殖を示した個体は見られなかった。しかし、2ヶ月齢では増殖を示した個体が6個体見られ(表5)。中には、当初1個のPGCが40個以上まで増殖した個体も認められた(表5, 図4B)。4ヶ月齢以降においては、多くのキメラ個体で生殖細胞の活発な増殖が確認された(表5, 図4C)。雌雄の判別ができる12、15.5、16ヶ月齢の雄の2n-3nと成魚2nの精巣には、全ての精子形成のステージ、即ち、精原細胞、精母細胞、多くの精細胞と精子が観察された(図5A,C)。dndMO処理をしていない3n 成魚においては、2nよりも大型の生殖細胞が見られ、精細胞と精子は観察されなかった(図5B)。1n-3nでは、当初のPGC数よりも増殖を示した個体が1ヶ月齢キメラでは4個体見られた。PGCsは1個から4個増殖していた(表6, 図4D)。2ヶ月齢においては、2個から10個以上の増殖が見られた(表6, 図4E)。4ヶ月齢以降においては、2n-3nよりも少数個体であるが、生殖細胞の活発な増殖が確認された(表6, 図4F)。12、15.5、16ヶ月齢の雄成魚では、多くの個体の精巣においてサイズの異なる多数の精原細胞とわずかの精母細胞が観察された(図5E)。しかし、16ヶ月齢の1n-3nの2個体中1個体は、2n-3nと同様に全ての精子形成ステージが認められ、多数の精細胞と精子が見られた(図5D)。雌においては、2n成魚、及びdndMO処理をしていない3nの卵巣では、多数の周辺仁期、及び少数の卵黄胞期の卵母細胞が観察された(図6A, B)。15.5ヶ月齢の2n-3nの1個体の卵巣から多数の周辺仁期、及び少数の卵黄胞期の卵母細胞が観察された(図6C)。同様に、16ヶ月齢の1n-3nの卵巣に多数の周辺仁期及び少数の卵黄胞期の卵母細胞が観察された(図6D)。1~16ヶ月齢の全ての1n-3n及び2n-3nのドナーPGCsが増殖、維持、減少した個体の割合を算出した(表7)。2n-3nは最初に測定したPGCの数よりも増殖していた個体は全体の51%で、維持していた個体は5%、減少していた個体は5%、生殖細胞が全くなかった個体は39%であった。一方、1n-3nは増殖が35%、維持が2%、減少が10%、なしが53%であった。1n-3nは2n-3nと比べるとPGCが増殖していた個体の割合が少なく、生殖細胞の無かった個体の割合が多かった。
1ヶ月齢の2n-3nの未分化生殖細胞の核の直径は8~9μmの頻度が高く、平均値は9.2μmを示した(図7A)。2ヶ月齢においては、10~12μmの頻度が高く、最小値7μm、最大値14μm、平均値は10.2μmであった(図7B)。1ヶ月齢の1n-3nの核の直径は9~10μmの頻度が高く、最小値は5μm、最大値は10μm、平均値は7.7μmを示した(図7C)。2ヶ月齢においては7~9μm の頻度が高く、最小値5μm、最大値9μm、平均値は6.8μm であった(図7D)。1n-3nは1、2ヶ月齢において、2n-3nと比べて、小さいサイズの未分化生殖細胞を多く持っており、2ヶ月齢において、その核の直径は2n-3nよりも有意に小さかった(スチューデントt検定、P<0.01)(図8)。雄成魚の2n及び3nの核の直径は9~10μmの頻度が高く、頻度分布は類似していた。2nの最小値は7μm、最大値13μm、平均値9.3μmであり、3nの最小値は8μm、最大値13μm、平均値9.4μmであり、2nと3nの核の直径に有意な差は認められなかった(図9A,B)。雄成魚(12~16ヶ月齢)2n-3nにおいて、A型精原細胞の核は9~10μmの頻度が高く、最小値7μm、最大値11μm、平均値8.8μmであった(図9C, 10)。雄成魚(12~16ヶ月齢)の精細胞と精子が観察された1n-3nの核は9~10μmの頻度が高く、最小値6μm、最大値14μmを示した(図9D)。雄成魚(12~16ヶ月齢)の精細胞と精子が観察されなかった1n-3nの核は9~10μmの頻度が高く、最小値6μm、最大値15μmを示した(図9E)。雄成魚1n-3n中の組織像から正常な精子形成のステージを持ち精細胞と精子が観察された個体は、平均値8.4μmであり、多くの精原細胞、わずかな精母細胞を有し、精細胞と精子が観察されなかった個体の核の平均値は9.6μmであった(図10)。2n-3n、2n-3n、1n-3nの間に有意な差が認められなかった(クラスカル・ウォリス検定、P<0.05)。以上のことから、雄成魚(12、15.5、16ヶ月齢)1n-3nの精巣中のA型精原細胞の核の直径は、2n、dndMO処理をしていない3n、2n-3nと同様の核の大きさをもつ細胞が多いが、1n-3nでのみ大型の核をもつ細胞があることが明らかになった(図9D, E)。
正常雄成魚の鰭は2nの細胞集団から構成されていた(図13A)。しかし精巣には1n、2n、4n相当の倍数性をもつ細胞集団が見られ、1n細胞が優占していた(図13C)。雄成魚3n(dndMO処理により不妊化していない)の鰭は3nの細胞集団から構成されており(図13B)、精巣は3n、6n細胞から構成された(図13D)。雄成魚(12~16ヶ月齢)2n-3nの鰭はすべて3nであり、2n細胞は見られなかった(表7)。2n-3nの発達した精巣では1nと3n(表7)又は1n、2nと3n(図13E)又は1n、2n、3nと4n(図13F)の細胞集団から構成されていた。これらの個体の精巣では1n細胞集団のピークが高く、他に2nと3nの小さなピークが見られた。一方、雄成魚(12~16ヶ月齢)1n-3nの鰭は全て3nであり(表8)、1n細胞は見られなかった。これに対して、発達した精巣を持つ1n-3n個体の精巣では1n、2nと3n(図13G)又は1n、2n、3nと6n(図13H)又は1n、2n、3nと4n(図13I)の倍数性の異なる細胞集団から構成されていた。これらの個体は1nと2nの細胞集団の大きさが同程度で、4n及びホスト生殖腺内の3n体細胞とそのG2/M期と思われる6nの倍数性をもつ細胞集団が検出された。精巣に3nのピークのみ検出された個体はホスト三倍体の生殖腺の体細胞のみをもつと考えられる。一方、2n-3n及び1n-3nの発達していない精巣は、ホストの体細胞と考えられる3nの細胞集団から構成されていた(表8)。
12ヶ月齢の2n-3nにhCGを注射したところ、6個体中4個体から採精でき、正常な精子の形態を示した(図15A)。1n-3nにおいては、6個体中2個体から採精できた。しかし、1n-3nの精子の濃度は2n-3nよりも極端に低かった。精子は2n-3nの採精した精子と同様の正常な形態を示した(図15B)。光学顕微鏡下での淡水添加による精子の運動観察では、2n-3nでは採精できた全4個体の精子の運動が観察できたが、1n-3nでは観察されなかった。採精された精液及び精巣の倍数性をフローサイトメーターにより検出した。その結果、2n-3nの精液中の細胞の倍数性は1nであった(図16A)。採精できた個体の精巣は1n、2nと3n(図16C)又は1n、2n、3nと4n又は1nと3nの細胞から構成されていた(表8)。精巣では1n細胞のピークが高く、2nと3n細胞のピークは低かった。そして、採精できなかった個体の精巣は3n細胞で構成されていた(表8)。1n-3nの精液の倍数性は1nであり(図16B)、採精できた個体のうち1個体の精巣は1n、2n、3nと4n細胞から構成されていた(図16D)。残りの1個体では倍数性を検出できなかった。また、採精できなかった個体の精巣は3n細胞より構成されていた(表8)。
1.vasa mRNAの発現
キメラ成魚の発達した精巣中の細胞が生殖細胞か否かの検討するために、生殖細胞特異的なvasa mRNAの発現を調査した。孵化後12、15.5、16ヶ月の1n-3n、2n-3nの成魚から精巣の一部を取り出し、RNAlater(Applied Biosystems社製)に入れ4℃で冷蔵庫に一晩放置し、その後-20℃で保存した。Nucleo Spin RNAII(MACHEREY NAGEL社製)のマニュアルに従いRNAを抽出した。抽出したRNAはPrime Script RT-PCR Kit(タカラバイオ株式会社製)を用いて、逆転写をし、cDNAを合成した。なお、内部コントロールとしてβ-actinのRT-PCRも行った。RT-PCRのプライマーは下記に示した。

PCRは以下の条件で行った。
94℃1分、そして94℃30秒、60℃30秒、72℃45秒を1サイクルとしてこれを30サイクル行った後、72℃7分の伸長反応を行った。PCR産物は1.2%アガロースゲルで電気泳動後、エチジウムブロマイドで染色し、可視化した。
1.vasa mRNAの発現
RT-PCRによりvasa mRNAの遺伝子発現を調査した。精巣の外部形態を見て、1n-3n及び2n-3nの発達している精巣はvasa mRNAの発現が認められた(図17A)。しかし、発達していない精巣にはvasaの発現は見られなかった。したがって、2n-3n及び1n-3nで発達している精巣中の細胞は生殖細胞であることが確認できた。
A.材料と方法
1.キンギョCarassius auratusを材料に、下述の発生工学的手法により、ドナーに半数体、ホストに正常二倍体を用いた半数体-二倍体生殖系列キメラ(1n-2n)、及びドナーに半数体、ホストに三倍体を用いた半数体-三倍体生殖系列キメラ(1n-3n)を作出した。1n-2n、1n-3nは、デッドエンドアンチセンスモルホリノオリゴヌクレオチド(配列番号No.6;5’-TCCATGCCGCTGTCCACCTGTGATG-3’)でノックダウンして不妊化処理をした二倍体又は三倍体ホスト個体の内で、半数体PGCsが生殖隆起へ移動し、生殖細胞として増殖するか否か、そして、それらの生殖腺から機能的な配偶子が形成されるか否かを検討するために作出した。将来的に、機能的な配偶子が形成されるか否かを検討する場合、キンギョでは、得られた子孫のDNA鑑定を可能にするために半数体を得る雌親魚とホストを得る雌親魚に遺伝的に異なる個体を使用した。
本発明では、北海道大学北方生物圏フィールド科学センターにて飼育中のキンギョCarassius auratusを材料として用いた。
キンギョ又は四倍体フナでは10~20IU/g体重のhCG(あすか製薬)を親魚の腹腔内に注射し、排卵、排精を誘起した。雌では12-14時間後に排卵を確認した後、サランラップ(ポリ塩化ビニリデンフィルム)(旭化成)を張った直径90mmプラスチックシャーレ上で腹部を圧迫して採卵した。雄も同様に腹部を圧迫し、得られた精液をヘマトクリット毛細管(テルモ製)で採精した。得られた精液はキンギョ用人工精漿液で約50~100倍に希釈した。作成した生殖系列キメラにおける配偶子と体細胞の遺伝子を比較するため、採精と採卵を行った親個体の鰭断片を100%アルコールで固定し、-80℃で保存した。
卵及び精子の遺伝的不活化には紫外線照射を用いた。上部に紫外線殺菌灯(GL15W、National社製)を設置した箱形紫外線照射装置を用いた。精子の遺伝的不活化のために、キンギョ用人工精漿液で約50~100倍に希釈した四倍体フナの精子を、よく洗浄し水分を拭き取ったガラスシャーレ上に約0.1mmの厚さになるように伸展した後、紫外線を照射した。なお、紫外線照射量は60mJ/cm2とした。キンギョの雌性発生では四倍体フナ由来の二倍体精子を用いた。この理由は、キンギョの雌性発生では、紫外線照射から逃れた二倍体精子からの受精卵は三倍体となり正常な配偶子形成は行わず、除去できるためである。
キンギョ卵と紫外線照射した四倍体フナ精子を媒精後、20℃の受精液を満たしたプラスチックシャーレ内に散布して受精させる方法でドナーの雌性発生半数体群を作出した。受精液には、0.2%尿素、0.24%NaClを含む水道水を用いた。また、ホストとして通常キンギョと四倍体フナ精子との受精により作出した三倍体群を作出した。ホストとする胚は、受精後1~4細胞期にdnd MO(配列番号No.6; 5’-TCCATGCCGCTGTCCACCTGTGATG-3)を顕微注入しPGCの分化を阻害して不妊を誘導した。キンギョのドナー半数体とホスト三倍体群では、卵膜除去を行わない対照群を作成し、受精後5時間以内に未受精卵を取り除き、さらに1日毎に死卵の計数と除去、及び水換えを行った。ドナー半数体の孵化期には孵化仔魚の形態を観察し、正常仔魚出現率と半数体症候群などの奇形を呈する仔魚の出現数を調査した。なお、発生段階は梶島(1960)の報告に従った。
各実験群の約半数程度の1~2細胞期の受精卵について、0.1%トリプシンと0.4%尿素を含む淡水魚用リンゲル液(128mM NaCl, 2.8mM KCl, 1.9mM CaCl2, pH 7.0)を含む卵膜除去液で処理することにより卵膜を除去した。卵膜除去卵は、約1%のアガロースで底面を被膜処理し20℃の一次培養液(1.6%ニワトリ卵白と0.01% ペニシリンとストレプトマイシンを含む淡水魚用リンゲル液)を満たしたガラスシャーレ内で体節形成期まで培養した。その後、卵膜除去卵を二次培養液(0.01% ペニシリンとストレプトマイシンを含む1.8mM CaCl2, 1.8mM MgCl2液)に移し、孵化まで培養した。
精子の遺伝的な不活化の成否を検討するために、キメラ作出時に用いたドナー胚の倍数性をフローサイトメーター(Partec Ploidy Analyser, PA型)を用いて調査した。また、ホストが三倍体又は二倍体であるか否かを検討するために、卵膜除去をしていないコントロールのホスト仔魚の倍数性を調べた。ドナー胚及びホスト仔魚は、Cystain DNA 2 step kit(Partec)のA液(核単離溶液)150μlの中にそれぞれ入れ、20分間放置し、ボルテックスにより細胞を混合・遊離した。その後、試料を50μmのメッシュ(Cell Trics 50μm, Partec)で濾過し、細胞の破片を除去した後、A液の5倍量のB液(4’,6-ジアミジノ-2-フェニリンドール(DAPI)を含む染色液)を加え、フローサイトメトリーにより相対DNA量を測定した。この際、正常二倍体キンギョの鰭のDNA量を二倍体DNA量(2C)の、正常二倍体キンギョの半数体精子のDNA量を半数体DNA量(1C)の基準とした。さらに2n-3nキメラ作出に用いたドナー胚及びホスト仔魚においても、倍数性を調べた。
卵膜除去した半数体受精卵が1~4細胞期の時にGFP-nos1 3’UTR mRNAを注入し、PGCの顕在化を行った。この卵が胞胚期に達したとき、胚盤周囲の割球を吸引しホスト胞胚へ移植する「割球移植法:BT法」、胚盤の下部を切り取りホスト胞胚の中央部へ移植する「胚盤移植:sandwich法」、および体節形成期に顕在化されたPGCをホスト胞胚へ一個移植する「SPT法」あるいは複数移植する「PPT法」のいずれかで生殖系列キメラ個体を誘導した。ホストとして、上述したdnd MOを顕微注入してPGCの形成を阻害した2n、あるいは3nの胚を用いた。ドナー細胞を移植した胚を培養するとともに蛍光顕微鏡で観察し、GFP蛍光を発するPGCが生殖隆起に確認された個体を生殖系列キメラとして分別して継続飼育した。
1n-3nキメラにおいて配偶子の分化の有無を検討するために、上述の採卵と採精の方法に準じてキメラ個体へのhCG注射を行った。注射した翌日に腹部を圧迫し、排卵あるいは排精の有無を確認した。排精した個体より、精子をヘマトクリット毛細管へ取り、さらに人工精漿溶液に精液を懸濁した。また、同個体より鰭断片を採取し、淡水魚用リンゲル液の中に保存した。鰭あるいは精子懸濁液は、Cystain DNA 2 step kit(Partec)のA液(核単離溶液)150μlの中にそれぞれ入れ、20分間放置し、ボルテックスにより細胞を混合・遊離した。その後、試料を50μmのメッシュ(Cell Trics 50μm, Partec)で濾過し、細胞の破片を除去した後、A液の5倍量のB液(4’,6-ジアミジノ-2-フェニリンドール(DAPI)を含む染色液)を加え、フローサイトメトリーにより相対DNA量を測定した。この際、正常二倍体キンギョの鰭のDNA量を二倍体DNA量(2C)の、正常二倍体キンギョの半数体精子のDNA量を半数体DNA量(1C)の基準とした。一方、これまでに排卵の確認された例はない。
排精の確認されたキメラ個体では、精子がドナーPGCに由来するかを明らかにするため、掛け合わせによる判定と、精子ゲノムと体細胞のゲノムの比較を行った。掛け合わせによる判定では、野生型より得られたキメラの半数体生殖細胞のドナー雌親、キメラのホストの雌雄親、キメラ個体の鰭、キメラの精子からDNAを抽出し、RAPD解析とマイクロサテライトマーカーを用いた遺伝的な解析を行った。DNAはエタノール固定されたサンプルの一部をProteinase Kを250μg/mLの濃度で含むTNES-Urea緩衝液に浸漬し、37℃で一晩インキュベートすることによってタンパク質消化を行った。その後、フェノール・クロロホルム法によってDNAの抽出を行った。
1.キンギョBTキメラ群とコントロール群の受精率、正常発生率、および生殖系列キメラ誘導率
BT(割球移植)法により作製した1n-3nキメラの生残率、正常発生率、および生殖系列キメラの率は表9にまとめた。ドナーの半数体は、雌性発生により作出した。ホストとしてはキンギョ卵と4倍体フナから得た二倍体精子による受精卵に、dnd MOを注入した胚を用いた。キメラを作出する際のコントロールとして、GFP nos1 3’UTR mRNAを注入していない卵膜除去をした半数体個体、および卵膜除去後にdnd MOを顕微注入したホスト三倍体対照群を用いた。移植群、対照群ともに使用卵数、受精卵数、孵化仔魚数、正常仔魚数、奇形仔魚数を計数した。その結果、移植群とホスト三倍体対照群では約半数が正常仔魚となった。また、ドナーとして作出した雌性発生群は、孵化後、全てがいわゆる半数体症候群と判定される奇形を呈した。正常な形態を示した移植群のうち1/4で、生殖腺領域にドナーのPGCと考えられる細胞が観察された。ドナー細胞を供与した半数体個体は、対照胚が孵化する時期にDNA量を測定した結果、全て半数体と考えられるDNA量であった。
Sandwich法(胚盤移植法)により作製した1n-3nキメラの生残率、正常発生率、および生殖系列キメラの率は表10にまとめた。ドナーの半数体は、雌性発生により作出した。ホストとしてはキンギョ卵と4倍体フナから得た二倍体精子による三倍体の受精卵に、dnd MOを注入した胚を用いた。キメラを作出する際のコントロールとして、GFP nos1 3’UTR mRNAを注入していない卵膜除去をした半数体個体、および卵膜除去後にdnd MOを顕微注入したホスト三倍体対照群を用いた。移植群、対照群ともに使用卵数、受精卵数、孵化仔魚数、正常仔魚数、奇形仔魚数を計数した。その結果、ホスト三倍体対照群では93.3%が正常に発生したのに対し移植群では約59%に留まった。一方、半数体対照群では正常に発生した個体はおらず、全てが半数体症候群と判定される奇形仔魚であった。正常な形態を示した移植群のうち1/3で、生殖腺領域にドナーのPGCと考えられる細胞が観察された。
SPT法またはPPT法により作製した1n-2nの生残率、正常発生率、および生殖系列キメラの率は表11にまとめた。ドナーの半数体は、雌性発生により作出した。ホストとして、キンギョ同士の二倍体受精卵を用いた。キメラを作出する際のコントロールとして、卵膜除去後にdnd MOを顕微注入したホスト二倍体、およびホスト二倍体受精卵を用いた。実験群、対照群とも90%以上が正常な発生をした。生殖隆起へドナーPGCが移動した個体は、一つのPGCを移植したSPT群では正常胚の29.2%しか無かったのに対して、5個のPGCsを移植した群では、82.4%出現した。
PPT法(胚盤移植法)により作製した1n-3nキメラの生残率、正常発生率、および生殖系列キメラの率は表12にまとめた。ドナーの半数体は、雌性発生により作出した。ホストとしてはキンギョ卵と4倍体フナから得た二倍体精子による受精卵に、dnd MOを注入した胚を用いた。キメラを作出する際のコントロールとして、GFP nos1 3’UTR mRNAを注入していない半数体個体の卵膜除去卵と非卵膜除去卵、卵膜除去後にdnd MOを顕微注入したホスト三倍体対照群、および卵膜を除去していない三倍体対照群を用いた。移植群、対照群ともに使用卵数、受精卵数、孵化仔魚数、正常仔魚数、奇形仔魚数を計数した。その結果、dnd MOを注入したホスト三倍体対照群、三倍体対照群では90%以上が正常に発生したのに対し移植群では約88.1%に留まった。一方、卵膜除去した半数体対照群では正常に発生した個体はおらず、全てが半数体症候群と判定される奇形仔魚であった。一方、卵膜未除去の半数体対照群では2個体(3.6%)で正常個体が出現した。PGC移植群では、17個体(28.8%)の生殖隆起でドナーPGCが確認された。
複数の半数体PGCsを移植したキメラ個体で、ドナーPGCの動態を追跡した。ホストとした個体は、黒色色素が出現しないアルビノ透明鱗個体を用いた。移植したPGCsは、概ね生殖隆起周辺に位置したが、脊索周辺などに分布するものも見受けられた。移植後4週間後に、ドナーPGCsの分裂が確認された。5週後には外部からは観察できなくなった。そのため、個体を固定し、生殖腺の組織学的な観察を行った結果、生殖腺内でのPGCの増加が確認された。
BTキメラを作製し、キメラおよびその対照群を1年半後に開腹して生殖腺の有無を確認した。この時点で4尾のキメラ個体、1尾のMO対照群、および6尾の三倍体対照群が生残した。キメラ群の一尾で、精巣が確認され、残りの3尾では未発達の線状の生殖腺が確認された。MO対照群の生殖腺は線状を示し、キメラ個体の線状生殖腺と一致した。三倍体対照群では、5尾で精巣が、1尾で卵巣が確認された。キメラ個体からの精液、確認された精巣と線状の生殖腺、三倍体対照群で確認された精巣の断片をFCMにかけ、構成する細胞の倍数性を確認した。その結果、キメラ個体の発達した精巣を持つ個体から得られた精液は1n、精巣を構成する細胞は1n、2n、3nの倍数性を示した。一方、未発達の生殖腺を持つ個体は、ホルモン注射後に得られた総排泄孔からの体液からは3n、生殖腺は3nのみの細胞から構成されていた。三倍体対照群の生殖腺は1.5n、3n、6nに相当する細胞が観察された。どの群も、鰭体細胞は3nの相対的DNA量を示した。これらの結果は、半数体のPGCを移植したキメラのみ1nの精液を産生することを示している。
半数体PGCを不妊化した三倍体に移植したPPTキメラ個体では8尾が生残した。これらのキメラ個体にhCGを注射し、排精およびその倍数性を確認した。2尾(090307#4, 090307#5)より排精され、FCMの結果、鰭の体細胞は三倍体、精子は半数体であることが確認された(図18)。
一方、野生型半数体PGCを不妊化したアルビノ二倍体に移植したPPTキメラ個体のうち、途中死亡した個体を解剖した結果、左右両方に精巣が確認された(図19)。これらの精巣断片の構成細胞の倍数性をFCMで確認した結果、半数体、二倍体の細胞が確認された。さらに、組織学的に精巣を観察した結果、精細胞を含む各段階の雄性生殖細胞が確認された。精子と断定できる細胞は観察されなかった。生き残った1個体(090622)で排精が確認され、FCMで半数体精子であることが確認された(図20)。排精が確認されたキメラ個体(090622)の精子をアルビノ雌個体より得られた卵に受精させた。その結果、子孫(n=70)は全て野生型の特徴である黒い色素を持っていた(表13)。このことから、この精子はドナーとなる野生型の半数体PGCに由来することが確認された。
1.キンギョにおけるRAPD解析
RAPD(Randomly amplified polymorphic DNA)解析ではプライマーとして多型が観察されたOPA-2、OPA-4、OPA-11(オペロン社)を用いた。PCR反応は0.2mLチューブとrTaq DNA Polymerase(TaKaRa)を用い、酵素に添付のバッファーを使用して20μLの反応系(1xPCR buffer (Mg2+含有), 0.2mM dATP, 0.2mM dTTP, 0.2mM dGTP, 0.2mM dCTP, Primer 0.5μM,rTaq DNA Polymerase(TaKaRa)1U, Template DNA 2.5-5ng)を作製した。94℃3分1サイクル、94℃30秒-36℃1分-72℃1分30サイクル、72℃7分1サイクルの反応条件でPCR反応を行い、反応終了後は4℃で保存した。得られたPCR産物は1.5%アガロースゲルを用いて75分間の電気泳動を行い、エチジウムブロマイドによってDNAを染色した後、UVトランスイルミネーターでバンドを検出した。
1.キンギョSandwichキメラにおける配偶子の分化とそのRAPD解析
1年間飼育した2尾のSandwichキメラ個体にhCGを注射し、排精およびその倍数性を確認した。1尾(080430)より排精され、FCMの結果、鰭の体細胞は三倍体、精子は半数体であることが確認された(図21)。25尾生き残ったMO対照群のうち、大型の5尾にhCGを注射したが排精、排卵は確認されなかった。排精が確認されたキメラ個体の体細胞と精子、ホストの雄親の体細胞よりDNAを抽出し、RAPD解析を行った(図22)。キメラの精子DNAを鋳型にして増幅されたバンドに、キメラ個体、ホストの雄親のものとは異なるものが確認され、キメラの精子にはドナー半数性PGCに由来するものが含まれていることが確認された。なお、この個体を作製したドナーおよびホストの両親の細胞のサンプルは保存されておらず、これ以上の解析は行えなかった。
1.キンギョにおけるマイクロサテライト解析のプライマー:
キンギョを用いたキメラにおけるクローン性の確認には、キンギョで開発されたマイクロサテライト(Zheng et al., Molecular Ecology 4, 791-792, 1995, Jung et al., Molecular Ecology Notes 7, 124-126, 2007)と、近縁種であるコイで開発されたマイクロサテライト(Crooijmans et al., Animal Genetics 28, 129-134,1997, Yue et al., Aquaculture 234, 85-98, 2004)を用いた(表14)。キンギョ由来のマイクロサテライトマーカーは、GF-1, 11, 17, 20, 29(Zheng et al., 1995)と、CAL-0120, 0428, 04100, 0410, 0461, 0174 0192, 0156, 0495(Jung et al., 2007)の合計14マーカー、また、コイ由来のマイクロサテライトマーカーはMFW-2, 7, 17(Crooijmans et al., 1997)Cca-02, 04, 12, 19, 22, 67, 86, 91(Yue et al., 2004)の合計12マーカーを用いてドナーとホスト、ならびにキメラとの交配に用いたメス親魚の多型性の検討を行った。このうち多型性が認められたのはGF17, GF29, MFW7, MFW17, CAL0428, CAL0174, CAL0192, Cca12, Cca86, Cca91 の10座であった。
2.キンギョにおけるマイクロサテライト解析
本解析ではキメラに用いたホスト両親DNAとドナー両親DNA、そしてキメラの鰭、精子に由来するDNAをテンプレートDNAとして用いた。そして、解析に用いるマーカーのフォワードプライマーには前述のプライマー配列の5’末端に5’-agtcacgacgttgta-3’(配列番号No.7)を付加したプライマーを使用した。そしてPCR反応と遺伝子型の解析は、アニーリング温度は50℃から56℃の間で各プライマーセットに最適な温度を用いたが、Morishima et al. Genetica 132, 227-241, (2008)に基づいてFAM, VIC, PET, NEDの各蛍光色素を付加したM13プライマー5’-GCCAGTCACGACGTTGTA-3’(配列番号NO.8)を用いたM13-tailed法によるPCRを行い、ABI3030xlオートシークエンサー(ABI)を用いて電気泳動し、遺伝子型はGeneMapper program Ver 3.7(ABI)によって解析した。
排精の確認されたキメラ個体では、一個体のキメラから得られた精子がすべて遺伝的に均一(クローン)であることを確認するために、クローンドジョウの産生する卵をキメラから得られた精子とで受精した雑種を作成し、雑種の形態を示す個体を分離した。次に、この雑種よりDNAを抽出し、キンギョ由来のマイクロサテライトマーカーにより解析した。キメラの半数体生殖細胞のドナー雌親、キメラのホストの雌雄親、キメラ個体の鰭のサンプルも合わせて採集した。これらのサンプルからDNAを抽出し、RAPD解析とマイクロサテライト解析を行った。
以上のドナー由来であることが確認された半数体のPGCが移植されたキメラ個体で、さらに詳細な精子の遺伝的な解析を行った。この解析には、ドナー半数体を誘起した両親の体細胞、ホストの個体を誘起した両親の体細胞、およびキメラ個体の体細胞と精子を材料とし、これらの細胞から抽出したDNAを解析に用いた。そして、ドナー半数体を誘起した両親の体細胞、ホストの個体を誘起した両親の体細胞、およびキメラ個体の体細胞と精子のDNAを解析した。解析には、キンギョおよびコイで報告されているマイクロサテライトマーカーを用いた(表14)。
1.キンギョ半数体PGCを移植したキメラドナーの雌性発生の再検証
移植したドナーPGCが雌由来であるかどうかを明らかにするため、キメラの精子のマイクロサテライトマーカーが雌由来のみで、雄由来が含まれていないかを明らかにした(図23)。その結果、090613#4の個体を除く三個体では、雌由来の座のみが観察され雌性発生が起こっていることが明らかにされた(表15)。
キメラの精子にホスト由来のゲノムが含まれていないかどうかを明らかにした(図24)。その結果、090307#4、090307#5、090622の三個体ではドナー雌のみのマーカーが含まれていたが、090613#4ではホストのみの座が認められた(表16)。この結果より、090307#4、090307#5、090622の三個体の精子はドナーメスに由来すると考えられた。090613#4から得られた精子はホストに由来すると考えられる。
キメラが半数性PGCsに由来して精子を形成している場合、一腹に由来するそれらの精子は、すべてのマイクロサテライトマーカー座においてドナー由来の一つのアレルのみを持ち、かつ精子の間で遺伝的相違の無いクローン精子である必要がある(図25)。そこで、ドナーの母親がヘテロアレルである遺伝子座をもつマイクロサテライトマーカーを用いて、キメラが産する精子のマーカー型を検討した。その結果、090307#4、090622の二個体に由来する精子では、用いたすべてのマーカー座においてドナー親の一方のアレルしかない(ヘミ型)ことが明らかとなった(表17)。090307#5ではヘテロ型の座が3/5で見つかった。このことから、090307#4、989622の二個体の精子は半数性のPGCから由来し、クローンである可能性が高いことが示された。
1.ゼブラフィッシュDanio rerioを材料に、下述の発生工学的手法により、ドナーに半数体、ホストに正常二倍体を用いた半数体-二倍体生殖系列キメラ(1n-2n)を作出した。1n-2nは、デッドエンドアンチセンスモルホリノオリゴヌクレオチド(ゼブラフィッシュでは、配列番号No.9;5’-GCTGGGCATCCATGTCTCCGACCAT-3’)でノックダウンして不妊化処理をした二倍体ホスト個体の内で、半数体PGCsが生殖隆起へ移動し、生殖細胞として増殖するか否か、そして、それらの生殖腺から機能的な配偶子が形成されるか否かを検討するために作出した。将来的に、機能的な配偶子が形成されるか否かを検討する場合、得られた子孫の色素によりドナー由来か否かを判別するために、ドナーとして色彩変異体(劣性形質)のゴールデン系統又は遺伝子組換え個体を使用した。
本発明では、北海道大学北方生物圏フィールド科学センターにて飼育中のゼブラフィッシュDanio rerioとキンギョCarassius auratus雄を材料として用いた。
ゼブラフィッシュでは、採卵の前日に16L-8Dに設定した恒温室内の産卵水槽に雌雄を分離して収容した。ドナー用の卵を得るため、採卵日の照明が点灯した直後に雌個体を混合し、産卵が確認された雌個体を取り上げ、腹部を圧迫してサランラップ上に採卵した。ホスト用の二倍体胚では、採卵日の照明が点灯した直後に、雌雄の親魚を混合して産卵を誘発させ、受精卵を得た。キンギョ精子は採精の前日にhCGを注射し、排精を誘導した。キンギョ精子はヘマトクリット毛細管に採精した後、キンギョ人工精漿に希釈し、遺伝的不活化に使用した。
卵及び精子の遺伝的不活化には紫外線照射を用いた。上部に2灯の紫外線殺菌灯(GL15W、National社製)を設置した箱形紫外線照射装置を用いた。精子の遺伝的不活化のための紫外線照射量は60mJ/cm2とした。雌性発生では二倍体キンギョ由来の半数体精子を使用した。この理由は、ゼブラフィッシュの雌性発生において、キンギョの精子を使えば、紫外線照射から逃れた精子からの受精卵は致死雑種となり除去できるためである。
ゼブラフィッシュ卵に紫外線照射したキンギョ精子を媒精後、28.5℃の受精液を満たしたプラスチックシャーレ内に散布して受精させる方法でドナーの雌性発生半数体群を作出した。受精液には0.2%尿素、0.24%NaClを含む水道水を用いた。また、ホストとして自然産卵させて採取した二倍体群を作出した。ホストとする胚は、受精後1~4細胞期にdnd MO(配列番号No.9;5’-GCTGGGCATCCATGTCTCCGACCAT-3’)を顕微注入しPGCの分化を阻害して不妊を誘導した。ドナー半数体の仔魚の形態を観察し、正常仔魚出現率と半数体症候群などの奇形を呈する仔魚の出現数を調査した。なお、発生段階はKimmel et al. (1995)の報告に従った。
半数体ドナー胚および二倍体ホスト胚は、0.1%トリプシンを含む淡水魚用リンゲル液(128mM NaCl, 2.8mM KCl, 1.9mM CaCl2, pH 7.0)で処理することにより卵膜を除去した。卵膜除去卵は、約1%のアガロースで底面を被膜処理し20℃の一次培養液(1.6%ニワトリ卵白と0.01% ペニシリンとストレプトマイシンを含む淡水魚用リンゲル液;128mM NaCl, 2.8mM KCl, 1.9mM CaCl2, pH 7.0)を満たしたガラスシャーレ内で体節形成期まで培養した。その後、卵膜除去卵を二次培養液(0.01% ペニシリンとストレプトマイシンを含む1.8mM CaCl2, 1.8mM MgCl2液)に移し、孵化まで培養した。
精子又は卵の遺伝的な不活化の成否を検討するために、キメラ作出時に用いたドナー胚の倍数性をフローサイトメーター(Partec Ploidy Analyser, PA型)を用いて調査した。ドナー胚は、Cystain DNA 2 step kit(Partec)のA液(核単離溶液)150μlの中にそれぞれ入れ、20分間放置し、ボルテックスにより細胞を混合・遊離した。その後、試料を50μmのメッシュ(Cell Trics 50μm, Partec)で濾過し、細胞の破片を除去した後、A液の5倍量のB液(4’,6-ジアミジノ-2-フェニリンドール(DAPI)を含む染色液)を加え、フローサイトメトリーにより相対DNA量を測定した。この際、正常ゼブラフィッシュの鰭のDNA量を二倍体DNA量(2C)の、ゼブラフィッシュの半数体精子のDNA量を半数体DNA量(1C)の基準とした。
生殖系列キメラの成否を遺伝的に判別するため、雌性発生半数体ドナーにはゴールデン系統および遺伝子組換え系統を用い、ホストには野生型あるいはゴールデンの系統を用いた。ここで用いた遺伝子組換え系統は、全ての細胞が緑色蛍光(GFP)で標識され、生殖細胞は赤色蛍光(RFP)で標識されている “GFP/RFP double transgenic strain”である。全てのホスト胚は一細胞期から四細胞期の間に100μMのdnd MOを顕微注入することにより不妊化した。また、ゴールデンをドナーとして用いる場合は、PGCsを可視化するため、一細胞期から四細胞期の間に300ng/μlのGFP-nos1 3’UTR mRNAを顕微注入した。
1.胞胚期ドナー胚の胚盤周縁部の割球を微小ガラス針で吸引し、同じ発生段階のホスト胚の胚盤中へ移植した(BTキメラ)。ドナー1個体から、およそ10個体のホストへ割球を移植することが可能だった。ドナーとして用いた全ての胚は初期体節形成期まで培養し、フローサイトメーターにより半数体であることを確認した(図26)。
475個体のキメラを作出し、そのうち66個体でドナー由来のPGCsが移植されていることを確認した。それらの個体では、移植されたPGCs数は5~14個と個体により幅があったものの、ほぼ全ての胚でドナーPGCsは生殖腺形成域へと到達していた(表18)。遺伝子組換え胚をドナーとして用いた場合では、キメラ中のPGCs蛍光は細胞が存在するかぎり観察できる。その特長を用いてドナーPGCsの動態を経時的に確認したところ観察した13個体中5個体で発生14日前後にドナーPGCsの消失が確認された。これらの胚では、同時に移植されたドナー体細胞のGFP蛍光は継続して観察できたことから、PGCs特異的な消失であることが示唆される。一方、その他の個体ではドナーPGCsの増殖が確認された。これらの個体では、少なくとも受精後30日後の段階でもPGCsのRFP蛍光が観察された。
作出したゴールデン系統(1n)→野生型系統(2n)キメラのうち5個体が成熟し交配実験を行うことができた。これら5個体はゴールデン系統をドナーとし、不妊化した野生型系統へ移植したものである。これらキメラはすべて雄の外部形態を呈した。ゴールデン系統の雌と交配を行った所、このうちの1個体から数百以上の受精卵が得られ、その全ての子孫がゴールデン系統の表現形を示した(図27)。同様に、遺伝子組換え系統(1n)→ゴールデン系統(2n)キメラのうち3個体が成熟した。これらのキメラも全て雄の外部形態を呈した。非遺伝子組換えの雌個体と交配実験を行ったところ、そのうちの1個体から得られた138個体の卵の中に8個の受精卵が見いだされた。これらの受精卵を培養したところ、遺伝子組換え系統に由来するGFP蛍光の発現が認められた。もし、ホスト(野生型)の生殖細胞が残留しており、それによって受精したのならば、その子孫はホスト系統の表現形を呈するはずである。しかし、妊性が確認されたキメラを用いた掛け合わせの実験ではどちらもドナー系統の表現型を持つ子孫のみが得られた。これらの結果から、キメラの配偶子は移植された半数体PGCsの由来すると考えられた。
本願において、統計的有意性はスチューデントt-検定、クラスカル・ウォリス検定、シェッフェ法により評価した。0.05未満のp値を有意であると判定した。
本発明によって得られたキメラ魚類によって、魚類において半数体PGCを利用した新たな育種技術の開発につながりうる。
本発明では、まずドジョウで、2nドナーのPGCsをdndMO処理により不妊化した3nホストに移植した生殖系列キメラにおいて、ドナー由来の配偶子形成のみが生じることを確かめようとした。その結果、2n-3nの産出した精子は正常な形態を示し、運動能を有することから精子としての機能をもつと思われた。正常二倍体の精巣は1nの精子、精細胞の他、2nと4nの細胞集団からなるが、2n-3nの発達した精巣は1nと3n又は1n、2nと3n又は1n、2n、3nと4nの細胞集団からなり、これら細胞集団の中では1n細胞のピークが高く、2nと4n細胞のピークは低かった。一般的に魚類において、2nはA、B型精原細胞、二次精母細胞、4nは一次精母細胞、1nは精細胞、精子であると考えられる(小林, 足立ら (2002) 生殖. 「魚類生理学の基礎」.恒星社厚生閣. 東京. pp155-182)。正常二倍体及び2n-3nにおいては、精子形成の全ステージ(精原細胞、精母細胞、多くの精細胞、精子)が組織学的に観察されたことから、フローサイトメーターにより検出された精巣中の2n細胞はA、B 型精原細胞及び二次精母細胞、4n細胞は一次精母細胞、ピークが高く検出された1n細胞は精細胞あるいは精子と考えられる。以上のことから、2n-3nキメラでは、2nドナー由来のPGCから機能的な精子が作られたと判断できる。

Claims (23)
- a) 魚類卵又は魚類精子の一方を遺伝的に不活化した後に媒精させて半数体ドナー受精卵を得る工程と、
b) ホスト魚類の受精卵を得る工程と、
c) 半数体ドナー受精卵から発生した半数体ドナー胚から得たドナー始原生殖細胞を、前記ホスト受精卵由来のホスト胚に移植する工程を含む、半数体生殖細胞を有するキメラ魚類を得る方法。 - 前記魚類がドジョウである、請求項1に記載の方法。
- 前記魚類がキンギョである、請求項1に記載の方法。
- 前記魚類がゼブラフィッシュである、請求項1に記載の方法。
- 卵又は精子の一方の遺伝的不活化が、紫外線照射又は放射線照射によって行われる、請求項1~4のいずれかに記載の方法。
