Description
本発明は、多孔質構造体に関する。特に、本発明は、多糖の水への溶解性を向上させるための改良に関する。
外科手術において、手術操作により生体組織が損傷される場合がある。また、切開によって生体組織を空気中に露出させることにより、生体組織が乾燥または酸化され、その結果、生体組織が損傷することが知られている。そして、損傷した組織が術後に炎症などを起こすと、本来離れているべき組織同士が癒着する可能性がある。このような術後の組織の癒着は、例えば、腹腔領域においては、腸閉塞や不妊症などの深刻な合併症の原因となりうる。このため、生体組織の損傷箇所を被覆し、癒着を防ぐための癒着防止材が種々開発されている。
現在知られている癒着防止材は、生体に悪影響を及ぼしにくい多糖やポリペプチドなどの生体由来の高分子材料を主成分としており、その形態も粉末状、シート状、ゼリー状、または液体状など様々である。なかでも、液体状の癒着防止材は、生体組織の目的部位に噴霧することにより被覆できるという操作性のよさから、特に注目されている。
このような癒着防止材のうち、例えば、特許文献1では、多糖側鎖に活性水素含有基と反応しうる活性エステル基が導入された、架橋性多糖誘導体からなる癒着防止材が開示されている。該架橋性多糖誘導体は、アルカリ条件下での水との接触により、該活性エステル基と活性水素含有基との共有結合による架橋物を形成しうる。
さらに、該特許文献1では、好ましい形態として、上記架橋性多糖誘導体を含む水溶液を生体組織の目的部位に塗布した後、この上からアルカリ条件にするためのpH調整剤を含む水溶液を噴霧し、該架橋性多糖誘導体をゲル化する癒着防止方法が開示されている。
また、上述の架橋性多糖誘導体を含む水溶液およびpH調整剤を含む水溶液のような2液の薬液を、同時に噴霧することができる塗布具が開発されている(例えば、特許文献2)。かような塗布具を用いることによって、1回の噴霧で癒着防止材を目的部位に塗布することができる。なお、水に溶解した状態の多糖は乾燥固体状の多糖よりも劣化しやすいために、手術現場で乾燥固体状の多糖に水を加えて多糖水溶液を都度調製することが望ましい。
しかしながら、上述の癒着防止材として好ましく使用されうる多糖は、疎水性官能基を含むために、あるいは分子量が大きいために、一般に難水溶性である。よって、乾燥固体状の多糖を水に溶解するのに時間がかかり、迅速な多糖水溶液の調製が困難である。そのため、難水溶性の多糖を含む癒着防止材は、手術現場での急な適用に対応できないという問題点を有していた。
そこで本発明は、多糖の水への溶解性を向上させる手段を提供することを目的とする。
本発明者らは、上記の課題を解決すべく鋭意研究を行った。その結果、多糖を凍結乾燥させる際の条件を制御することにより、所定の孔径を有し、水への溶解性が著しく向上した多孔質構造体が得られることを見出し、本発明を完成させた。すなわち、本発明の上記課題は、以下の(1)~(5)により達成される。
(1)多糖を含み、多数の孔を有する、多孔質構造体であって、該孔の平均孔径が40μm以上であり、該孔の総数に対する、孔径が50μm以上の孔の数の割合が30%以上である、多孔質構造体。
(2)多糖を含む多糖水溶液を、下記(A)および(B)の工程により凍結乾燥してなる、多孔質構造体;(A)該多糖水溶液を冷却し、過冷却点を経た後、5分間以上の最大氷結晶生成帯通過時間を経て、該多糖水溶液の氷結晶を得る凍結工程、および(B)該氷結晶に含まれる水分を減圧下で昇華させることにより除去する、乾燥工程。
(3)該多糖が、カルボキシル基、活性エステル基、カルボン酸塩、アミノ基、およびアルデヒド基からなる群から選択される少なくとも1種を有する、(1)または(2)に記載の多孔質構造体。
(4)該孔径の標準偏差σが5~30μmである、(1)~(3)のいずれかに記載の多孔質構造体。
(5)多糖を含む多糖水溶液を、下記(A)および(B)の工程により凍結乾燥する、多孔質構造体の製造方法;(A)該多糖水溶液を冷却し、過冷却点を経た後、5分間以上の最大氷結晶生成帯通過時間を経て、該多糖水溶液の氷結晶を得る凍結工程、および(B)該氷結晶に含まれる水分を減圧下で昇華させることにより除去する、乾燥工程。
本発明によると、多糖の水への溶解性を向上することができる。よって、手術現場で迅速に多糖水溶液を調製することが可能となる。
以下、本発明の好ましい形態を説明する。本形態は、多糖を含み、多数の孔を有する多孔質構造体に関する。そして、該孔の平均孔径が40μm以上であり、該孔の総数に対する、孔径が50μm以上の孔の数の割合が30%以上である。
<多孔質構造体>
本形態の多孔質構造体は多糖を必須に含む。本明細書において、「多糖」とは、単糖がグリコシド結合によって結合してなる重合体であって、分子量が1000以上であるものをいう。

多糖を構成する単糖は、特に制限はないが、例えば、リボース、キシロース、アラビノース、グルコース、マンノース、ガラクトース、フルクトース、ソルボース、ラムノース、フコース、およびリボデオース、ならびにこれらに任意の官能基が導入された単糖誘導体が挙げられる。多糖は、これらの単糖のうち、1種のみから構成されてもよいし、2種以上が組み合わされて構成されてもよく、直鎖状であってもよいし、分岐を含んでいてもよい。また、天然に存在する多糖を用いてもよいし、合成したものであっても構わない。これらの多糖のうち、生体への投与実績の観点から、グルコースまたはグルコース誘導体を含む多糖であることが好ましく、具体的には、デキストリン、デキストラン、およびセルロース、ならびにこれらの誘導体からなる群から選択される少なくとも1種の多糖を含むことがより好ましい。
本形態に係る多糖は、好ましくは、カルボキシル基、活性エステル基、カルボン酸塩、アミノ基、およびアルデヒド基からなる群から選択される少なくとも1種を有することが好ましい。このような官能基は、エステル結合またはアミド結合を形成する架橋点となりうるので、癒着防止材の構成成分として好適である。癒着防止材としてこれらの官能基が含まれると、該多糖を含む水溶液をアルカリ条件下としたときに、多糖に含まれる上記官能基、およびヒドロキシ基、ならびに生体組織表面に存在するヒドロキシ基などがエステル結合またはアミド結合を形成して架橋する。これにより、癒着防止材がゲル状となり生体組織の表面を被覆することができる。
カルボキシル基を有する多糖の一例としては、多糖のヒドロキシ基にカルボキシアルキル基が導入されてなる形態が挙げられる。該カルボキシアルキル基としては、好ましくは、炭素原子数2~5のカルボキシアルキル基が挙げられ、具体的には、カルボキシメチル基、カルボキシエチル基、カルボキシプロピル基、カルボキシイソプロピル基、およびカルボキシブチル基などが挙げられる。このうち、カルボキシメチル基またはカルボキシエチル基であることが好ましく、カルボキシメチル基であることがより好ましい。
活性エステル基を有する多糖の一例としては、カルボキシル基を有する多糖に含まれるカルボキシル基と、N-ヒドロキシアミン系化合物とがエステル化されてなる形態が挙げられる。該N-ヒドロキシアミン系化合物としては、例えば、N-ヒドロキシスクシンイミド、N-ヒドロキシノルボルネン-2,3-ジカルボン酸イミド、2-ヒドロキシイミノ-2-シアノ酢酸エチルエステル、2-ヒドロキシイミノ-2-シアノ酢酸アミド、N-ヒドロキシピペリジンなどが挙げられ、このうち、N-ヒドロキシスクシンイミドを用いることが好ましい。
カルボン酸塩を有する多糖は、カルボキシル基を有する多糖に含まれるカルボキシル基から水素イオンが抜けたカルボン酸イオンが、水素イオン以外の陽イオンとイオン結合してなる多糖である。該陽イオンとしては、例えば、アルカリ金属イオン、アルカリ土類金属イオン、テトラ(n-ブチル)アンモニウム、およびテトラ(n-プロピルメチル)アンモニウムなどの四級アンモニウムイオンなどが挙げられる。
また、アミノ基を有する多糖としては、例えば、グルコサミンおよびグルコサミンなどのアミノ糖を含む多糖や、イソウレア中間体を含む多糖(下記化学式1の構造を含む多糖)などが挙げられる。
また、アルデヒド基を有する多糖としては、例えば、多糖の還元末端にアルドースを含む多糖が挙げられる。
上記官能基を有する多糖は、天然から得られるものであってもよいし、化学的または生物学的手法によりこれらの官能基を有する多糖を製造しても構わない。これらの官能基を含む多糖の製造方法は、従来公知の手段を適宜採用することができる。
なお、本形態の多孔質構造体は、上記多糖以外の構成成分を含んでもよいことはいうまでもない。多糖以外の構成成分としては、例えば、分子量1000未満のオリゴ糖などが挙げられるが、これらに制限されるものではない。
多糖の重量平均分子量は、特に制限はないが、好ましくは50000~200000であり、より好ましくは50000~100000である。このような範囲の分子量の多糖を用いることによって、本形態の多孔質構造体を癒着防止材の構成成分として用いた場合に、架橋後の多糖が良好なゲル硬度を有しうる。なお、本明細書において、重量平均分子量はGPCによって得られる値を採用する。
本形態の多孔質構造体は、多数の孔を有する。そして、該孔の平均孔径は40μm以上であり、好ましくは40μm~200μmである。本形態の多孔質構造体は、後述の通り、5分間以上の最大氷結晶生成帯通過時間を経て、多糖水溶液の氷結晶を得た後、当該氷結晶を減圧下で昇華させることにより製造する。このような製造方法によると、氷結晶が均一に成長するため、孔の平均孔径が200μmを超えることは実質的に困難である。本明細書において、「孔」とは、上記多糖を含む多孔質構造体の構成成分からなる壁に囲まれた空隙部分をいう。「多数の孔」とは、1cm四方あたり、500以上の孔があることを意味する。「孔径」とは、上記空隙部分の長さを意味する。「平均孔径」とは、上記孔径の平均値を意味する。なお、本明細書における「孔径」および「平均孔径」の値は、後述の実施例で用いる測定方法によって得られる値を採用するものとする。また、本形態の多孔質構造体は、孔の総数に対する、孔径が50μm以上の孔の数の割合が、30%以上であることが必須であり、好ましくは40~80%である。このように、本形態の多孔質構造体は、従来と比較して大きな孔を多数有するために、水と接触して多糖が溶解した際に孔内でのゲル化が抑制され、短時間での溶解が可能となる。
また、多孔質構造体の孔径の標準偏差σは、5~30μmであることが好ましく、5~20μmであることがより好ましい。孔径の標準偏差σがこのような範囲にあると、微細孔が少なく、水への溶解性が良好となる。
<多孔質構造体の製造方法>
本形態の多孔質構造体は、上記多糖を含む多糖水溶液を以下の凍結工程(A)および乾燥工程(B)の2つの工程を含む凍結乾燥によって容易に製造することができる。以下、本形態の多孔質構造体の好ましい製造方法を説明する。

まず、上記多糖を含む多糖水溶液を調製する。多糖水溶液を調製する際に使用する水は、特に制限はないが、好ましくは不純物が少ない、RO水、蒸留水、および注射用水などを用いる。また、多糖水溶液の濃度も特に制限はないが、多糖1gに対して一般的には6~20g、好ましくは8~15gの水を使用して多糖水溶液を調製する。多糖を水に溶解する際の溶解方法も特に制限はなく、一般的には攪拌によって行うが、多糖の化学的特性に著しい悪影響を及ぼさない限りにおいては、加温を伴う溶解であってもよい。
次に、凍結工程(A)において、上記で調製した多糖水溶液を冷却し、多糖水溶液の氷結晶を形成させる。この際、多糖水溶液から氷結晶になる過程において、多糖水溶液が過冷却点を経た後、5分間以上の最大氷結晶生成帯通過時間を経て氷結晶となることが必要である。該最大結晶生成帯通過時間は、好ましくは10分間以上であり、より好ましくは15分間以上である。なお、最大氷結晶生成帯通過時間の上限値は、多糖水溶液の量や冷却装置の冷却能力に左右されるので、特に制限はないが、より短時間で凍結工程を完結させる観点から、180分間未満が好ましく、より好ましくは60分間未満である。本明細書において「過冷却点」とは、液体を冷却する過程において、過冷却状態にある液体が固体化する時点を意味し、図1に示すように、冷却曲線の傾きがマイナスからプラスに転じる点である。また、本明細書において「最大氷結晶生成帯通過時間」とは、図1に示すように、冷却曲線において、過冷却点を経て傾きがプラスに転じた後に再びマイナスに転じた時点の温度から、-7~+2℃の温度領域に該曲線が存在する時間をいう。該温度領域において、溶液が固体化して氷結晶を形成する。最大氷結晶生成帯通過時間は、5分間以上であることが必須であるが、好ましくは、10分間以上である。このような冷却工程を経て多糖水溶液の氷結晶を形成することにより、多孔質構造体の孔径を制御することができる。
凍結工程における多糖水溶液の冷却条件は、多糖水溶液の量、冷却装置の特性などにより、一概にはいえないが、60分間程度の時間をかけて冷却温度を25~-30℃まで順次低くしていくことにより、過冷却点を有し、最大氷結晶生成帯通過時間が5分以上である凍結工程が達成されうる。
冷却に用いる冷却装置は特に制限はないが、上述の凍結工程を達成するために、冷却温度を調節できる冷却装置であることが好ましく、冷却温度および冷却時間をプログラムによって制御できるものであることがより好ましい。また、凍結工程と、続く乾燥工程とをスムーズに行うために、凍結乾燥装置を用いると尚好ましい。
凍結工程(A)で多糖水溶液の氷結晶を得た後、乾燥工程(B)によって該氷結晶に含まれる水分を減圧下で昇華させることにより除去する。この際の減圧条件は特に制限はないが、一般的には3~20Paである。また、当然のことであるが、氷結晶の水分を液化させることなく昇華させるために、氷結晶を-30~-20℃程度に保つことが必要である。なお、乾燥装置は、冷却下で減圧乾燥できるものであればよく、公知の装置を適宜採用することができる。
本発明の作用効果を、以下の実施例および比較例を用いて説明する。ただし、本発明の技術的範囲が以下の実施例のみに制限されるわけではない。
<多孔質構造体の製造>
[実施例1]
(多糖水溶液の調製)
16mlバイアルに安定剤としてのトレハロース0.75gを秤量し、RO水(逆浸透(Reverse Osmosis)膜で不純物を取り除いた水)10gを加えて溶解した。これにカルボキシメチルデキストリン(重量平均分子量70000)1.5gを加えて、ボルテックスミキサーを用いて、室温(25℃)にて1400rpmで10分間攪拌した。
[実施例1]
(多糖水溶液の調製)
16mlバイアルに安定剤としてのトレハロース0.75gを秤量し、RO水(逆浸透(Reverse Osmosis)膜で不純物を取り除いた水)10gを加えて溶解した。これにカルボキシメチルデキストリン(重量平均分子量70000)1.5gを加えて、ボルテックスミキサーを用いて、室温(25℃)にて1400rpmで10分間攪拌した。
(凍結乾燥)
アルミブロック(16穴)に上記バイアルを設置し、ゴム栓を半開の状態で取り付けた。これを、予め棚温度を-35℃に設定した凍結乾燥装置(DFR-5N-A、アルバック社製)に設置した。バイアル内には熱電対を差し込み、多糖水溶液の温度をモニタできるようにした。
アルミブロック(16穴)に上記バイアルを設置し、ゴム栓を半開の状態で取り付けた。これを、予め棚温度を-35℃に設定した凍結乾燥装置(DFR-5N-A、アルバック社製)に設置した。バイアル内には熱電対を差し込み、多糖水溶液の温度をモニタできるようにした。
棚温度を、-35℃に設定し、10時間冷却することによってバイアル内の多糖水溶液を凍結した[凍結工程]。なお、凍結開始から10時間後の最終凍結温度は-30℃であった。
続いて、下記表1のプログラムにしたがって、バイアル内の水を昇華し、乾燥することによって多孔質構造体を製造した[乾燥工程]。
[実施例2]
上記(多糖水溶液の調製)において、RO水を14gとしたことを除いては、実施例1と同様の方法で、多孔質構造体を製造した。なお、凍結工程における、凍結開始から10時間後の最終凍結温度は-30℃であった。

[比較例1]
(多糖水溶液の調製)
16mlバイアルにトレハロース0.25gを秤量し、RO水2.7gを加えて溶解した。これにカルボキシメチルデキストリン(重量平均分子量70000)0.5gを加えて、ボルテックスミキサーを用いて、室温(25℃)にて1400rpmで10分間攪拌した。
(多糖水溶液の調製)
16mlバイアルにトレハロース0.25gを秤量し、RO水2.7gを加えて溶解した。これにカルボキシメチルデキストリン(重量平均分子量70000)0.5gを加えて、ボルテックスミキサーを用いて、室温(25℃)にて1400rpmで10分間攪拌した。
(凍結乾燥)
予め-45℃に冷却したアルミブロック(16穴)に上記バイアルを設置し、ゴム栓を半開の状態で取り付けた。これを、予め棚温度を-35℃に設定した凍結乾燥装置(DFR-5N-A、アルバック社製)に設置した。バイアル内の多糖水溶液に熱電対を差し込み、多糖水溶液が凍結する際の温度をモニタリングできるようにした。
予め-45℃に冷却したアルミブロック(16穴)に上記バイアルを設置し、ゴム栓を半開の状態で取り付けた。これを、予め棚温度を-35℃に設定した凍結乾燥装置(DFR-5N-A、アルバック社製)に設置した。バイアル内の多糖水溶液に熱電対を差し込み、多糖水溶液が凍結する際の温度をモニタリングできるようにした。
棚温度を、-35℃に設定し、10時間冷却することによってバイアル内の多糖水溶液を凍結した[凍結工程]。なお、凍結開始から10時間後の最終凍結温度は-30℃であった。
続いて、上記表1のプログラムにしたがって、バイアル内の水を昇華し、乾燥することによって多孔質構造体を製造した[乾燥工程]。
[比較例2]
上記[比較例1](多糖水溶液の調製)において、トレハロースを0.75g、RO水を10g、カルボキシメチルデキストリンを1.5gとしたことを除いては、比較例1と同様の方法で、多孔質構造体を製造した。

<多糖水溶液が凍結する際の温度の測定>
上記実施例および比較例において、多糖水溶液が凍結する際の温度を、熱電対を用いてモニタリングした。また、該温度の経時変化をグラフ化した。実施例1および2のグラフを図2に、比較例1のグラフを図3に、比較例2のグラフを図4にそれぞれ示す。そして、該グラフより、過冷却点の有無、最大氷結晶生成帯通過時間を確認した。結果を下記表2に示す。
上記実施例および比較例において、多糖水溶液が凍結する際の温度を、熱電対を用いてモニタリングした。また、該温度の経時変化をグラフ化した。実施例1および2のグラフを図2に、比較例1のグラフを図3に、比較例2のグラフを図4にそれぞれ示す。そして、該グラフより、過冷却点の有無、最大氷結晶生成帯通過時間を確認した。結果を下記表2に示す。
<平均孔径の算出>
上記実施例および比較例で得られた多孔質構造体を1cm四方の大きさに切り出し、切断面の表面を、走査型電子顕微鏡(SEM)を用いて倍率100倍で観察した。観察したSEM画像を図5~8に示す。

そして、該SEM画像を紙面に印刷し、多孔質構造体の切断面0.5mm四方に相当する部分を2箇所選択した。そして、該箇所に存在する孔の孔径を全て測定した。孔が円形である場合はその直径を孔径とし、その他の形状の場合は、その孔を最小面積となるような平行四辺形で囲んだ際の、該平行四辺形の長辺と短辺とを足して2で割った値を孔径とした。そして、求めた全ての孔径の平均値を平均孔径とした。結果を表2に示す。
<孔径が50μm以上である孔の割合>
また、上記で選択した2箇所に存在する孔の総数に対する、孔径が50μm以上である孔の数の割合を算出した。結果を下記表2に示す。

<孔径の標準偏差σの算出>
上記で求めた全ての孔の孔径および平均孔径より標準偏差σを求めた。結果を下記表2に示す。

<密度の測定>
上記実施例および比較例で得られた円柱状の多孔質構造体の底面積および高さを測定し、多孔質構造体の体積を算出した。また、該多孔質構造体の質量を測定した。そして、求めた質量を体積で除して、構造体の密度を算出した。結果を下記表2に示す。

<空隙率の測定>
上記<密度の測定>で用いた多孔質構造体にエタノールを0.5cm3ずつ滴下し、該構造体に吸収されるエタノールの体積を求めた。求めた体積を下記式1に当てはめ、多孔質構造体の空隙率を算出した。結果を下記表2に示す。

<溶解性試験>
上記実施例および比較例で得られた多孔質構造体0.15g分を切り出し、ポリプロピレン製試験管に入れた。室温(25℃)において、この試験管にRO水0.35gを加え、直ちにボルテックを用いて攪拌し、溶解するまでの時間を測定した。また、溶解中のダマの生成の有無を確認した。結果を下記表2に示す。
上記実施例および比較例で得られた多孔質構造体0.15g分を切り出し、ポリプロピレン製試験管に入れた。室温(25℃)において、この試験管にRO水0.35gを加え、直ちにボルテックを用いて攪拌し、溶解するまでの時間を測定した。また、溶解中のダマの生成の有無を確認した。結果を下記表2に示す。
表2の結果より、過冷却点を有し、最大氷結晶生成帯通過時間が5分間以上となるように凍結工程を行った実施例1および2の多孔質構造体は、ともに、平均孔径が40μm以上であり、孔径が50μm以上である孔の割合が35%以上であった。これら実施例の多孔質構造体は、溶解時間が10秒間と顕著に短く、溶解中のダマの生成は認められなかった。
一方、比較例1、2では、均一な冷却を意図して、予め-45℃に冷却したアルミブロックにバイアルを設置したが、冷却速度が速すぎたためか、過冷却点および5分間以上の最大氷結晶生成帯通過時間を有する所望の冷却曲線が得られなかった。このため、多孔質構造体の溶解に長時間を要し、溶解中にダマの生成が認められた。
なお、所望の冷却曲線を得るための凍結条件は、多糖水溶液の量や冷却装置などによって異なることから、上記実施例の方法に限定されるものではなく、当業者が適宜調節可能である。

Claims (5)
- 多糖を含み、多数の孔を有する多孔質構造体であって、
前記孔の平均孔径が40μm以上であり、
前記孔の総数に対する、孔径が50μm以上の孔の数の割合が30%以上である、多孔質構造体。 - 多糖を含む多糖水溶液を、下記(A)および(B)の工程により凍結乾燥して得られる、請求項1に記載の多孔質構造体;
(A)前記多糖水溶液を冷却し、過冷却点を経た後、5分間以上の最大氷結晶生成帯通過時間を経て、前記多糖水溶液の氷結晶を得る凍結工程、および
(B)前記氷結晶に含まれる水分を減圧下で昇華させることにより除去する、乾燥工程。 - 前記多糖が、置換または非置換のカルボキシル基、アミノ基、またはアルデヒド基を少なくとも1つ有する、請求項1または2に記載の多孔質構造体。
- 前記孔径の標準偏差σが5~30μmである、請求項1~3のいずれか1項に記載の多孔質構造体。
- 多糖を含む多糖水溶液を、下記(A)および(B)の工程により凍結乾燥する、多孔質構造体の製造方法;
(A)前記多糖水溶液を冷却し、過冷却点を経た後、5分間以上の最大氷結晶生成帯通過時間を経て、前記多糖水溶液の氷結晶を得る凍結工程、および
(B)前記氷結晶に含まれる水分を減圧下で昇華させることにより除去する、乾燥工程。
